Abstract:With the wide use of nanomaterials in daily life, nano-titanium dioxide (nano-TiO2) presents potential ecological risks to marine ecosystems, which can be exacerbated by ocean warming (OW). However, most previous studies have only centered around waterborne exposure, while there is a scarcity of studies concentrating on the impact of trophic transfer exposure on organisms. We investigated the differences in toxic effects of 100 μg/L nano-TiO2 on mussels via two pathways (waterborne and foodborne) under normal (24 °C) and warming (28 °C) conditions. Single nano-TiO2 exposure (waterborne and foodborne) elevated the superoxide dismutase (SOD) and catalase (CAT) activities as well as the content of glutathione (GSH), indicating activated antioxidatant response in the intestine. However, depressed antioxidant enzymes and accumulated peroxide products (LPO and protein carbonyl content, PCC) demonstrated that warming in combination with nano-TiO2 broke the prooxidant-antioxidant homeostasis of mussels. Our findings also indicated that nano-TiO2 and high temperature exhibited adverse impacts on amylase (AMS), trypsin (PS), and trehalase (THL). Additionally, activated immune function (lysozyme) comes at the cost of energy expenditure of protein (decreased protein concentration). The hydrodynamic diameter of nano-TiO2 at 24 °C (1693-2261 nm) was lower than that at 28 °C (2666-3086 nm). Bioaccumulation results (range from 0.022 to 0.432 μg/g) suggested that foodborne induced higher Ti contents in intestine than waterborne. In general, the combined effects of nano-TiO2 and warming demonstrated a more pronounced extent of interactive effects and severe damage to antioxidant, digestive, and immune parameters in mussel intestine. The toxicological impact of nano-TiO2 was intensified through trophic transfer. The toxic effects of nano-TiO2 are non-negligible and can be exerted together through both water- and foodborne exposure routes, which deserves further investigation.

Abstract:Biochemical responses of the mussel Mytilus coruscus exposed to different concentrations of titanium dioxide nanoparticles (nano-TiO2) (0, 2.5, 10 mg L−1) and two pH levels (pH 8.1 and pH 7.3) for 14 days. Mussel responses were also investigated after a 7 days recovery period (pH 8.1 and no nanoparticle). Exposure to nano-TiO2 led changes in antioxidant indexes (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione (GSH)), biotransformation enzyme activity (GST) and malondialdehyde level (MDA) in gills and digestive glands. An increase in MDA level and a decrease in SOD and GSH activities were observed in gill of mussels exposed to 10 mg L−1 nano-TiO2. This effect was more severe in mussels kept at pH 7.3 as compared to pH 8.1. A different response was observed in the digestive gland as SOD, CAT and GSH levels increased in mussels exposed to nano-TiO2. These contrasting results in digestive glands and gills were only evident at high concentration of nano-TiO2 and low pH. A 7 days recovery period was not sufficient to fully restore SOD, GPx, GST, GSH and MDA levels to levels before exposure to nano-TiO2 and low pH. Overall, our results confirmed that seawater acidification modulates effects of nanoparticles in mussels, and that gills are more sensitive to these stressors as compared with digestive glands.
Dietary Exposure to Ntio2 Reduces Byssus Performance of Mussels under Ocean Warming.

Abstract:Nano-titanium dioxide (nTiO2) is a widely used nanomaterial posing potential ecological risk for marine ecosystems that might be enhanced by elevated temperatures such as expected during climate change. nTiO2 may affect benthic filter feeders like mussels through waterborne exposures and via food chain due to the adsorption on/in algae. Mussel byssus are proteinaceous fibers secreted by byssal glands of the mussels for attachment. Byssus production and me-chanical properties are sensitive to environmental stressors but the combined effects of warming and nTiO2 on byssus performance of mussels are unclear hampering our understanding of the predation and dislodgement risk of mussels under the multiple stressor scenarios. We explored the effects of a short-term (14-day) single and combined exposures to warming (28 & DEG;C) and 100 & mu;g L-1 nTiO2 (including food co-exposure) on the byssus performance of the thick shell mussel Mytilus coruscus. The mechanical strength (measured as the breaking force) of the byssal threads was impaired by warming and nTiO2 (including food co-exposure), but the number and length of the byssal threads were increased. The mRNA expression levels of mussel foot proteins (mfp-3, mfp-5) and pre-collagens (preCOL-D, preCOL-P, preCOL-NG) were up-regulated to varying degrees, with the strongest effects induced by warming. This indicates that the phys-iological and molecular mechanisms of byssus secretion are plastic. However, downregulation of the mRNA expression of preCOL-D and preCOL-P under the combined warming and nTiO2 exposures indicate the limits of these plasticity mechanisms and suggest that the attachment ability and survival of the mussels may be impaired if the pollution or temperature conditions further deteriorate.

Abstract:Coastal habitats are exposed to increasing pressure of nanopollutants commonly combined with warming due to the seasonal temperature cycles and global climate change. To investigate the toxicological effects of TiO 2 nanoparticles (TiO 2 NPs) and elevated temperature on the intestinal health of the mussels ( Mytilus coruscus ), the mussels were exposed to 0.1 mg/L TiO 2 NPs with different crystal structures for 14 days at 20 °C and 28 °C, respectively. Compared to 20 °C, the agglomeration of TiO 2 NPs was more serious at 28 °C. Exposure to TiO 2 NPs led to elevated mortality of M. coruscus and modified the intestinal microbial community as shown by 16S rRNA sequence analysis. Exposure to TiO 2 NPs changed the relative abundance of Bacteroidetes , Proteobacteria and Firmicutes. The relative abundances of putative mutualistic symbionts Tenericutes and Fusobacteria increased in the gut of M. coruscus exposed to anatase , which have contributed to the lower mortality in this group. LEfSe showed the combined stress of warming and TiO 2 NPs increased the risk of M. coruscus being infected with potential pathogenic bacteria. This study emphasizes the toxicity differences between crystal structures of TiO 2 NPs, and will provides an important reference for analyzing the physiological and ecological effects of nanomaterial pollution on bivalves under the background of global climate change.

Hemocyte Responses of the Thick Shell Mussel Mytilus Coruscus Exposed to Nano-Tio2 and Seawater Acidification.

Abstract:With increasing production from nanotechnology industries, nanomaterials are inevitably released into the aquatic environment, thereby posing a potential risk to aquatic organisms. Thus, concerns have been raised on the potential ecotoxicological effect of nanoparticle. Furthermore, the ecotoxicological consequences caused by the interaction of nanoparticles with other environmental stresses, such as seawater acidification on marine animals, have not been evaluated. In particular, whether acidification enhances the susceptibility to nanoparticles in bivalves needs to be evaluated. In the present study, we investigated the combined effects of low pH and nanoscale titanium dioxide (nano-TiO2) on some immune parameters of hemocytes in the mussel Mytilus coruscus by flow cytometry under six combinations of two pH values (7.3 and 8.1) and three nano-TiO2 concentrations (0, 2.5, and 10mgL-1) for 14 d. Afterward, the mussels were shifted to normal conditions without nano-TiO2 at pH 8.1 for 7 d further to test their recovery from the multiple stresses. Total hemocyte count (THC), phagocytosis (Pha), esterase (Est), and lysosomal content (Lyso) decreased under low pH and high nano-TiO2 concentration conditions, whereas hemocyte mortality (HM) and reactive oxygen species (ROS) increased with nano-TiO2 concentrations under low pH conditions. The interactive effects between pH and nano-TiO2 were observed at the latter part of the exposure experiment (7 and 14 d) in most hemocyte parameters. Nano-TiO2 influenced the immune functions of mussel more severely than low pH. Slight recovery from the combined stresses was observed for HM, THC, Pha, and Lyso, but significant carry-over effects of nano-TiO2 and low pH were still observed. This study demonstrated that both low pH and high concentration of nano-TiO2 had negative effects on mussels, and these effects still acted for some time even though the mussels were already out of such stressors.

Abstract:Nano-titanium dioxide (nano-TiO 2 ) is a ubiquitous contaminant in the marine environment that accumulates in sediments and biological tissues. Coupled with global warming, these challenges can enhance the deleterious properties of nano-TiO 2 , leading to compounded pollution effects on marine life and ecosystems. This study investigated the effects of nano-TiO 2 and increased temperatures on the Japanese swimming crab's gut microbiota and digestive system, Charybdis japonica , through different scenarios. We employed three exposure scenarios: direct exposure (DE) of the crabs to warming and nano-TiO 2 , indirect exposure (IE) through consumption of mussels Mytilus coruscus subjected to the same conditions, and combined exposure (CE), where crabs were directly exposed to warming and nano-TiO 2 while consuming affected mussels. Additionally, a control group was established, comprising Japanese crab C. japonica and thick-shelled mussel M. coruscus that were reared under standard temperature (22 °C, the average annual temperature in the region where the mussels and crabs were sampled) and 0 mg L −1 nano-TiO 2 concentration conditions. The findings indicated that warming and nano-TiO 2 disrupted the crabs' ATP production, digestive responses, and body chemical composition, leading to intestinal flora dysfunction. Notably, nano-TiO 2 exerted a stronger impact on the crabs' digestive enzymes and intestinal flora than warming alone; however, the concurrent presence of warming and nano-TiO 2 , especially under the direct exposure (DE) conditions, generally exacerbated the negative effects of nano-TiO 2 . This research provides valuable insights into the implications of nano-TiO 2 and elevated temperature on the digestive responses of marine crabs.

Abstract:Organic and nanoparticle pollutants are the main environmental problems affecting marine species, which have received great attention. However, the combined effect of pollutants on marine life in the presence of predators needs to be clarified. In this study, the effects of pentachlorophenol (PCP) and titanium dioxide nanoparticles (nano-TiO 2 ) on the energy metabolism of mussels ( Mytilus coruscus ) in the presence of predators were assessed through cellular energy allocation (CEA) approach. Mussels were exposed to PCP (0, 1, and 10 μg/L), nano-TiO 2 (1 mg/L, 25 and 100 nm), and predators ( Portunus trituberculatus presence/absence) for 14 days. Exposure to high concentrations of PCP (10 μg/L) with small particle size nano-TiO 2 (25 nm) decreased cellular energy stores (carbohydrates, lipids, and proteins) and increased cellular energy demand (measured as the activity of the mitochondrial electron transport system, ETS). During the first 7 days, energy was supplied mainly through the consumption of carbohydrates, while lipids are mobilized to participate after 7 days. The presence of predators caused a further decrease in energy stores. These findings demonstrate that PCP, nano-TiO 2 and predators have a negative impact on energy metabolism at the cellular level. Carbohydrates are not able to meet the metabolic demand, lipids need to be consumed, and energy metabolism was also mediated by the involvement of proteins. Overall, our results suggest that PCP, nano-TiO 2 and predators disrupt the cellular energy metabolism of mussels through reduced cellular energy allocation, small particles and predators drive mussels to exert energetic metabolic adjustments for detoxification reactions when toxic contaminants are present.

Abstract:The increasing usage of nanoparticles has caused their considerable release into the aquatic environment. Meanwhile, anthropogenic CO 2 emissions have caused a reduction of seawater pH. However, their combined effects on marine species have not been experimentally evaluated. This study estimated the physiological toxicity of nano-TiO 2 in the mussel Mytilus coruscus under high pCO 2 (2500–2600 μatm). We found that respiration rate (RR), food absorption efficiency (AE), clearance rate (CR), scope for growth (SFG) and O:N ratio were significantly reduced by nano-TiO 2 , whereas faecal organic weight rate and ammonia excretion rate (ER) were increased under nano-TiO 2 conditions. High pCO 2 exerted lower effects on CR, RR, ER and O:N ratio than nano-TiO 2 . Despite this, significant interactions of CO 2 -induced pH change and nano-TiO 2 were found in RR, ER and O:N ratio. PCA showed close relationships among most test parameters, i.e., RR, CR, AE, SFG and O:N ratio. The normal physiological responses were strongly correlated to a positive SFG with normal pH and no/low nano-TiO 2 conditions. Our results indicate that physiological functions of M. coruscus are more severely impaired by the combination of nano-TiO 2 and high pCO 2 .

Abstract:With the development of industry, agriculture and intensification of human activities, a large amount of nano-TiO2 dioxide and pentachlorophenol have entered aquatic environment, causing potential impacts on the health of aquatic animals and ecosystems. We investigated the effects of predators, pentachlorophenol (PCP) and nano titanium dioxide (nano-TiO2) on the gut health (microbiota and digestive enzymes) of the thick-shelled mussel Mytilus coruscus. Nano-TiO2, as the photocatalyst for PCP, enhanced to toxic effects of PCP on the intestinal health of mussels, and they made the mussels more vulnerable to the stress from predators. Nano-TiO2 particles with smaller size exerted a larger negative effect on digestive enzymes, whereas the size effect on gut bacteria was insignificant. The presence of every two of the three factors significantly affected the population richness and diversity of gut microbiota. Our findings revealed that the presence of predators, PCP, and nano-TiO2 promoted the proliferation of pathogenic bacteria and inhibited digestive enzyme activity. This research investigated the combined stress on marine mussels caused by nanoparticles and pesticides in the presence of predators and established a theoretical framework for explaining the adaptive mechanisms in gut microbes and the link between digestive enzymes and gut microbiota.
The combined effects of phenanthrene and micro-/nanoplastics mixtures on the cellular stress responses of the thick-shell mussel Mytilus coruscus

Abstract:Pollution with complex mixtures of contaminants including micro- and nano-plastics (MNPs) and organic pollutants like polycyclic aromatic hydrocarbons (PAH) poses a major threat to coastal marine ecosystems. Toxic mechanisms of contaminant mixtures are not well understood in marine organisms. We studied the effects of single and combined exposures to polycyclic aromatic hydrocarbon phenanthrene (Phe) and MNPs mixture with sizes of 70 nm, 5 μm and 100 μm on the immune health and oxidative stress parameters in the thick-shell mussel Mytilus coruscus . Immune cells (hemocytes) were more sensitive to the pollutant-induced oxidative stress than the gills. In hemocytes of co-exposed mussels, elevated mortality, lower lysosomal content, high production of reactive oxygen species (ROS) and decrease mitochondrial were found. Disparate responses of antioxidant enzymes in the hemolymph (e.g. increased superoxide dismutase (SOD) activity without a corresponding increase in catalase (CAT) in Phe exposures and an increase in CAT without a change in SOD in MNPs exposures) suggests misbalance of the antioxidant defense in the pollutant-exposed mussels. Gill lacked pronounced oxidative stress response showing a decline in ROS and antioxidant levels. Tissue-specific single and combined effects of Phe and MNPs suggest variation in bioavailability and/or different sensitivity to these pollutants in the studied tissues. Notably, the combined effects of MNPs and Phe were additive or antagonistic, showing that MNPs do not enhance and occasionally mitigate the toxic effects of Phe on the hemocytes and the gills of the mussels. Overall, our study sheds light on the impact of long-term exposure to MNPs and Phe mixtures on mussels, showing high sensitivity of the immune system and modulation of the Phe toxicity by MNPs co-exposure. These findings that may have implications for understanding the impacts of combined PAH and MNPs pollution on the health of mussel populations from polluted coastal habitats.

Metabolic dysfunctions in pearl oysters following recurrent marine heatwaves

Abstract:Marine heatwaves (MHWs) have become more frequent, intense and extreme in oceanic systems in the past decade, resulting in mass mortality events of marine invertebrates and devastating coastal marine ecosystems. While metabolic homeostasis is a fundamental requirement in stress tolerance, little is known about its role under intensifying MHWs conditions. Here, we investigated impacts of MHWs on the metabolism in pearl oysters (Pinctada maxima) - an ecologically and economically significant bivalve species in tropical ecosystems. Activities of digestive enzymes (gastric proteases, lipases, and amylases) did not significantly respond to various scenario of recurrent MHWs varying from 24 °C to 28 °C (moderate) and 32 °C (severe). The metabolomics analysis revealed nine and five key metabolism pathways under both MHWs scenarios. Specifically, pathways associated with energy metabolism were impaired by moderate MHWs, manifesting in downregulation of differential metabolite (The nicotinic acid and N-acetyl-glutamic acid). The content of CDP-ethanolamine was significantly decrease, and the perturbations of oxidative stress caused by the decreased of content of D-glutamine. Metabolites related to a suite of body functions (e.g., the lipid metabolism, biomineralization, and antioxidant defenses) showed significantly negative responses by severe MHWs. These findings reveal the metabolic impairments of marine bivalves when subjected to MHWs varying in intensity and frequency, implying cascading consequences which deserve further investigation.
Size-dependent effects of plastic particles on antioxidant and immune responses of the thick-shelled mussel Mytilus coruscus

Abstract:Micro-/nano-plastic particles (MNPs) are present in the ocean with potential detrimental impacts on marine ecosystems. Bivalves are often used as marine bioindicators and are ideal to evaluate the threat posed by various-sized MNPs. We exposed the mussel Mytilus coruscus to MNPs with different particle sizes (70 and 500 nm, 5, 10 and 100 μm) for 3, 72 h and 30 days. The antioxidant responses in digestive gland and the hemolymph were then evaluated. The time of exposure played a strong modulating role in the biological response. A 3-hour exposure had no significant impact on the digestive gland. After 72 h, an increase in oxidative stress was observed in the digestive gland, including increased hydrogen peroxide (H 2 O 2 ) level, catalase (CAT), glutathione peroxidase (GPx) activities and malondialdehyde (MDA) production. After a 30-day exposure, the oxidative stress decreased while lipid peroxidation increased. A 30-day exposure increased hemocyte mortality (HM) and reactive oxygen species (ROS) levels in the hemolymph, while phagocytosis (PA), lysosome content (LC), mitochondrial number (MN) and mitochondrial membrane potential (MMP) significantly decreased. Longer-term exposure to MNPs caused oxidative stress in the digestive gland as well as impaired viability and immunity of hemocytes. Particle size also influenced the response with smaller particles having more severe effects. A depuration for 7 days was enough to reverse the negative effects observed on the digestive gland and hemolymph. This study provides new insights on the effects of small-sized MNPs, especially nanoplastic particles (NPs), on aquatic organisms, and provides a solid theoretical knowledge background for future studies on toxic effects of MNPs.

Abstract:Long-term exposure of Mytilus galloprovincialis to temperatures beyond 26°C triggers mussel mortality. The present study aimed to integratively illustrate the correlation between intermediary metabolism, hsp gene expression, and oxidative stress-related proteins in long-term thermally stressed Mytilus galloprovincialis and whether they are affected by thermal stress magnitude and duration. We accordingly evaluated the gene expression profiles, in the posterior adductor muscle (PAM) and the mantle, concerning heat shock protein 70 and 90 ( hsp70 and hsp90), and the antioxidant defense indicators Mn-SOD, Cu/Zn-SOD, catalase, glutathione S-transferase, and the metallothioneins mt-10 and mt-20. Moreover, we determined antioxidant enzyme activities, oxidative stress through lipid peroxidation, and activities of intermediary metabolism enzymes. The pattern of changes in relative mRNA expression levels indicate that mussels are able to sense thermal stress even when exposed to 22°C and before mussel mortality is initiated. Data indicate a close correlation between the magnitude and duration of thermal stress with lipid peroxidation levels and changes in the activity of antioxidant enzymes and the enzymes of intermediary metabolism. The gene expression and increase in the activities of antioxidant enzymes support a scenario, according to which exposure to 24°C might trigger reactive oxygen species (ROS) production, which is closely correlated with anaerobic metabolism under hypometabolic conditions. Increase and maintenance of oxidative stress in conjunction with energy balance disturbance seem to trigger mussel mortality after long-term exposure at temperatures beyond 26°C. Eventually, in the context of preparation for oxidative stress, certain hypotheses and models are suggested, integrating the several steps of cellular stress response.
Synergistic impacts of nanopollutants (nZnO) and hypoxia on bioenergetics and metabolic homeostasis in a marine bivalve Mytilus edulis

Abstract:Coastal ecosystems face increasing threats from anthropogenic pollution and environmental stressors like hypoxia and nanoparticle exposure. The Baltic Sea exemplifies these challenges due to nutrient pollution and hypoxia. We investigated the combined effects of zinc oxide nanoparticles (nZnO), which possess unique properties such as high reactivity and bioavailability, and hypoxia on bioenergetics and metabolite homeostasis of the blue mussel Mytilus edulis from the Baltic Sea. Mussels were first exposed to environmentally relevant concentrations of nZnO (100 μg Zn L −1 ) and subsequently subjected to short-term (24 h) or long-term (7 d) hypoxia (<0.1% air saturation) followed by recovery periods (1 h and 24 h). Our findings reveal complex effects of nZnO on mussel metabolism under normoxic and hypoxic conditions. Under normoxic conditions, nZnO alters mussel metabolism without causing energy deficit. Prolonged severe hypoxia induces anaerobic metabolism and glycogen depletion. Under hypoxic conditions, nZnO disrupts mussels' metabolic response to anaerobic conditions, threatening their anaerobic survival capacity. Control mussels swiftly recover metabolic homeostasis upon reoxygenation, whereas nZnO-exposed mussels show delayed recovery, with ongoing energy disturbances. Overall, these findings underscore the metabolic impacts of nZnO and hypoxia in keystone marine mussels and emphasize the importance of considering oxygen levels in assessments of nanoparticle toxicity in coastal ecosystems.

Abstract:Marine heatwaves (MHW) threaten marine organisms and tend to increase in frequency and intensity. We exposed the blue mussel Mytilus edulis to a MHW lasting 23 days, including two 10-d periods of thermal intensity increase of +5 °C (20 °C–25 °C) interspersed by 1 day back to 20 °C, followed by a 4-d recovery period. We investigated behaviour responses of mussels and gene expression changes relative to the circadian rhythm ( Per ), oxidative stress ( SOD ), cellular apoptosis ( CASP3 ), energy production ( ATPs ), and general stress response ( hsp70 ). Results showed that the MHW disturbed the valve activity of mussels. Particularly, mussels increased the number of valve micro-closures, showing a stressful state of organisms. Mussels also decreased Per , CASP3 , ATPs , and Hsp70 gene expression. Some behavioural and molecular effects persisted after the MHW, suggesting a limited recovery capacity of individuals. This work highlighted the vulnerability of M. edulis to a realistic MHW.

Abstract:Harmful algal blooms (HABs) and thermal stress as climate changes become more common in global water ecosystem, especially under eutrophic habitats. Here our study examined the combined impacts of bloom forming cyanobacteria Microcystis aeruginosa and thermal stress on the antioxidant responses of the ecologically important species triangle sail mussel Hyriopsis cumingii. The differential responses of a series of enzymes, e.g. superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione-S-transferase (GST), as well as signal metabolites including reactive oxygen species (ROS), malondialdehyde (MDA) and glutathione (GSH) involved in antioxidant defense mechanisms were analyzed during 14 d exposure to toxic cyanobacterium M. aeruginosa and 7 d depuration period. The activities of SOD and GPx as well as the content of ROS and MDA in H. cumingii increased, while CAT activity reduced due to M. aeruginosa exposure. Thermal stress resulted in decrease of CAT, the accumulation of GSH and the enhance of GST and SOD. Meanwhile, the interactive effects among M. aeruginosa, thermal stress and time were also observed on most parameters except for GST activity. The total amount of microcystins (MC) in sail mussels increased with concentrations of exposed M. aeruginosa, independently of the presence or absence of thermal stress Although around 50% of MC in mussels dropped in the depuration period, most parameters showed alterations because of cyanobacteria exposure and thermal stress. Overall, these findings suggested that toxic cyanobacteria or thermal stress induces oxidative stress and severely affects the enzymes activities and intermediates level associated with antioxidant defense mechanisms in sail mussels respectively. More importantly, the toxic impacts on sail mussels could be intensified by their combination.

Abstract:The increasing use of nanoscale TiO2 particles (nTiO2) and their subsequent leakage into aquatic environments poses a threat to the ecosystem. One major concern is that nTiO2 may alter the environmental behaviors of arsenic (As) and disrupt the equilibrium of As accumulation and speciation in organisms. In this study, we investigated the effects of nTiO2 on the bioaccumulation and biotransformation of As(V) in the mussel Perna viridis. Exposure to nTiO2 significantly increased As accumulation in mussels. Our As speciation analysis demonstrated that nTiO2 treatment increased the proportion of inorganic As and reduced that of organic As, displaying inhibitory effects on the methylation and detoxification of inorganic As in mussels. Analysis of enzyme systems related to As metabolism in mussels demonstrated that nTiO2 might limit the methylation of inorganic As by suppressing the GST activity and GSH content. The strong adsorption capacity and weak desorption rate of As by nTiO2, which could result in the disruption of As distribution and decrease of the amount of As involved in biotransformation, might serve as another mechanism to the limition on As methylation in mussels. Moreover, exposure to nTiO2 disturbed the osmotic adjustment system in mussels by reducing arsenobetaine and Na+-K+-ATPase activity, resulting in enhanced toxicity of As after coexposure. Immune Toxicity of TiO2 under Hypoxia in the Green-Lipped Mussel Perna Viridis Based on Flow Cytometric Analysis of Hemocyte Parameters

Abstract:The combined effects of DO and TiO2 (mixed rutile/anatase phase, 7/3) on immune responses in Perna viridis were examined. Mussels were exposed to six combinations of oxygen levels (hypoxia: 1.5mgO2l−1, normoxia: 6.0mgO2l−1) and TiO2 concentrations (0, 2.5mgl−1 and 10mgl−1) for 216h. Mussels were sampled after 24h, 48h, 120h and 216h, and immune parameters of hemocytes, including mortality, phagocytosis, non-specific esterase, ROS production, lysosomal content and total hemocyte count were investigated using flow cytometric assay. Hemocyte mortality was higher under hypoxia than normoxia, and increased with TiO2 concentrations, but no interaction was found between DO and TiO2. Phagocytosis was reduced under hypoxia and decreased with TiO2 exposure, and the interactive effect between time and TiO2 was observed. The percentage of hemocytes showing non-specific esterase activity was lower under hypoxia, and decreased as TiO2 concentration increased with the significant interactive effect of DO and TiO2. ROS production and lysosomal content were lower under hypoxia and reduced as concentration of TiO2 increased, and interactive effect of DO and TiO2 on ROS was evident. THC was significantly affected by the interactive effect between TiO2 and DO, with higher values under normoxia in the presence of TiO2. The present study demonstrated that immune functions of P. viridis were influenced by both nano-TiO2 and hypoxia with some synergistic effects between the two stressors. This implies that DO has to be considered in the evaluation of the toxicity of nano-materials to bivalves.

Abstract:Titanium dioxide nanoparticle (nTiO2) pollution of marine environments is rapidly increasing with potentially deleterious effects on wildlife. Yet, the impacts of nTiO2 on reproduction remain poorly understood. This is especially the case for broadcast spawners, who are likely to be more severely impacted by environmental disturbances because their gametes are directly exposed to the environment during fertilisation. In addition, it is unclear whether rising water temperatures will further exacerbate the impact of nTiO2 toxicity. Here, in a series of fertilisation trials, we systematically examine the main and interactive effects of nTiO2 exposure and seawater temperature on fertilisation success in the Mediterranean mussel Mytilus galloprovincialis. Specifically, our fertilisation trials explored whether nTiO2 exposure influences fertilisation rates when (i) eggs alone are exposed, (ii) both sperm and eggs are exposed simultaneously, and (iii) whether increases in seawater temperature interact with nTiO2 exposure to influence fertilisation rates. We also ask whether changes in nTiO2 concentrations influence key sperm motility traits using computer-assisted sperm analysis (CASA). In fertilisation trials for treatment groups (i) and (ii), we found no main effects of nTiO2 at environmentally relevant concentrations of 5, 10 and 50 μg L-1 on fertilisation capacity relative to the control. Consistent with these findings, we found no effect of nTiO2 exposure on sperm motility. However, in treatment group (iii), when fertilisation trials were conducted at higher temperatures (+6 °C), exposure of gametes from both sexes to 10 μg L-1 nTiO2 led to a reduction in fertilisation rates that was significantly greater than when gametes were exposed to elevated temperature alone. These interacting effects of nTiO2 exposure and seawater temperature demonstrate the toxic potential of nTiO2 for fertilisation processes in a system that is likely to be impacted heavily by predicted future increases in sea surface temperatures.
