Abstract:Porcine reproductive and respiratory syndrome virus (PRRSV) is one of the most economically important pathogens in the global swine industry over the past three decades. There is no licensed antiviral medication that can effectively control this infection. In the present study, the structure of SP-1 isolated and purified from Sargassum weizhouense was analyzed, and its antioxidant capacity and antiviral effect in MARC-145 cells against PRRSV were investigated. The results showed that SP-1 is a novel polysaccharide which mainly is composed of →4)-β-D-ManpA-(1→, →4)-α-L-GulpA-(1→ and a small amount of →4)-β-D-GalpA-(1→. PRRSV adsorption, replication, and release were all suppressed by SP-1. SP-1 therapy down-regulated mRNA expression of the CD163 receptor while increasing the antioxidant gene expression of Nrf2, TXNIP, and HO-1; increasing the protein expression of NQO1 and HO-1; and drastically reducing the protein expression of p-p65. The findings indicated that SP-1 reduces PRRSV adsorption, replication, and release through blocking the expression of the crucial CD163 receptor during infection. Meanwhile, SP-1 exerts antioxidant effects in PRRSV-infected cells through the activation of the Nrf2-HO1 signaling pathway.

Abstract:Brown algae possess a diverse array of acidic polysaccharides, including fucoidan. The present research intends to investigate the extraction and characterization of algal polysaccharides to explore their antiviral activity. A light brown sulfated polysaccharide was extracted (with a yield of 18% of dry weight) from Sargassum asperifolium algal powder. The results of fractionation of sulfated polysaccharide revealed the occurrence of two primary fractions: low-sulfated polysaccharides (SPF1) and high-sulfated polysaccharides (SPF2). The bioassays conducted on SPF2 demonstrated a greater level of antioxidant activity compared to SPF1, with respective IC50 values of 17 ± 1.3 μg/mL and 31 ± 1.1 μg/mL after a duration of 120 min. The cytotoxicity of SPF2 on Vero cells was determined, and the calculated half-maximal cytotoxic concentration (CC50) was found to be 178 ± 1.05 μg/mL. Based on these results, an antiviral activity assay was conducted on SPF2. The results demonstrated that SPF2 had greater efficacy against Hepatitis A Virus (HAV) compared to Herpes Simplex Virus Type 1 (HSV-1), with corresponding half-maximal inhibitory concentrations (IC50) of 48 ± 1.8 μg/mL and 123 ± 2.6 μg/mL, respectively. The active SPF2 was characterized by FT-IR, 1H, and 13C NMR spectroscopy. The extracted fucoidan can be used as a natural therapeutic agent in combating various viral infections.
Antiviral activity of sulfated polysaccharides from Sargassum ilicifolium against fish Betanodavirus infection

Abstract:Sulfated polysaccharides (SP) from seaweeds exhibit numerous biological activities and are less toxic than conventional antiviral drugs. However, little is known about their mode of action. This study aims at extracting the polysaccharides from brown seaweed <i>Sargassum ilicifolium</i> and then evaluating its antiviral activity against fish Betanodavirus. The extracted SP was characterized through FT-IR, <sup>1</sup>H NMR, and thermal gravimetric analysis (TGA). The antiviral activity of SP was tested on SGA cell line by syncytia reduction and the polysaccharide represented a prominent antiviral activity to Betanodavirus by reducing the cytopathic effect (CPE) formation in vitro. The study revealed that SP effectively blocked viral attachment and entry. Moreover, during the in vivo challenge experiment in <i>Gambusia affinis</i>, SP treatment showed a significant decrease in the viral titers. Hence, these results indicate that the polysaccharides obtained from <i>S. ilicifolium</i> exhibited the high antiviral potential to Betanodavirus infection in fishes both in vitro and in vivo.

In vitro antiviral effect of sulfated pectin from Mangifera indica against the infection of the viral agent of childhood bronchiolitis (Respiratory Syncytial Virus - RSV)

Abstract:The Human Respiratory Syncytial Virus (RSV) is the leading cause of acute respiratory infections in children. Currently, no safe, effective, or feasible option for pharmacological management of RSV exists. Hence, plant-derived natural compounds have been explored as promising antiviral agents. Mangifera indica is a globally distributed plant with reported anti-inflammatory, cardioprotective, and antiviral activities. Our study investigated the antiviral potential of a novel pectin from M. indica peels (PMi) and its chemically sulfated derivative (PSMi) against RSV in HEp-2 cells. The compounds were characterized using Fourier-transform infrared spectroscopy and nuclear magnetic resonance (NMR). NMR analysis revealed the presence of ester and carboxylic acid groups in PMi, and sulfation resulted in a sulfation degree of 0.5. PMi and PSMi showed no cytotoxic effects even at concentrations as high as 2000 μg/mL. PSMi completely inhibited RSV infectivity (100–1.56 μg/mL, 50 % inhibitory concentration of viral infectivity = 0.77 ± 0.11 μg/mL). The mechanism of action was investigated using the 50 % tissue culture infectious dose assay PSMi displayed virucidal activity at concentrations from 100 to 6.25 μg/mL, and a significant reduction in viral infection was observed at all treatment times. Overall, PSMi is antiviral, cell-safe, and exhibits promising potential as an RSV treatment.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Objective</h3><p>To investigate the antiviral effect of <em>Salvia plebeia</em> R. Br. polysaccharides (SPP) against RSV and underlying mechanisms.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Methods</h3><p>SPP was extracted <em>via</em> alcohol-precipitation method and extract was separated into various fractions using ultrafiltration method. The polysaccharide content was determined using UV-Vis. Antiviral effect of SPP and fractions was measured using MTT method and Reed-Muench method. Sixty Balb/c mice were randomly divided into 6 groups, and received either Ribavirin or SPP. Their body weight and food intake were recorded every day throughout the experiment period. The lung index inhibition ratio and pulmonary virus titer were determined followed by the histological analysis of lungs. Furthermore, time-of-addition and effective stage analysis were carried out to determine the mechanism of action. The TLR-3 and TLR-4 levels in the lungs were determined using qRT-PCR. The levels of IFN-γ, IL-2 and TNF-α in serum were determined using ELISA.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3><p>The SPP content is 4.396%. SPP has shown a good anti-RSV effect both <em>in vitro</em> (TI = 123.041) and <em>in vivo</em> models. The antiviral activity of fractions with molecular weight ≥ 10,000 is found to possess more potent antiviral activity than other fractions. SPP inhibits the RSV proliferation and reduces the lung lesions induced by RSV. The mechanism of action involves the inhibition of TLR-3 and TLR-4 in lungs, up-regulation of IFN-γ and IL-2, and down-regulation of TNF-α in serum. It is also shown to improve the body's immune function.</p><h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusion</h3><p>SPP has a potential to treat diseases caused by RSV.</p>

Abstract:Jerusalem artichoke (Helianthus tuberosus L.) polysaccharide (JAP) is a chain polysaccharide composed of D-fructose connected by β (1-2) glycosidic bonds, which is a kind of inulin. This study evaluated the anti-respiratory syncytial virus (RSV) activity of JAP in vivo and in vitro. To investigate its antiviral activity, an MTT assay, q-PCR, enzyme-linked immunosorbent assay (ELISA), and lung histological observation were performed. The results showed that JAP showed anti-RSV activity in vitro with a half maximal inhibitory concentration (IC50) of approximately 29.15 μg/mL. In vivo results suggested that JAP could effectively inhibit RSV proliferation in the lungs and improve lung tissue lesions in RSV-infected mice. Additionally, JAP could also reduce the expression of TLR3 and TLR4 in the lungs, increase serum anti-inflammatory factors IL-4 levels, and reduce pro-inflammatory factors TNF-α and TNF-β levels, which may be related to its anti-RSV activity. This study provides a new approach to anti-RSV therapy and enriches the potential applications of JAP.

Abstract:Respiratory viral infections have been a leading cause of morbidity and mortality worldwide. Despite massive advancements in the virology field, no specific treatment exists for most respiratory viral infections. Approved therapies against respiratory viruses rely almost exclusively on synthetic drugs that have potential side effects, restricting their use. This review aims to present natural marine sulfated polysaccharides possessing promising antiviral activity against respiratory viruses that could be a safe alternative to synthetic broad-spectrum antiviral drugs. The antiviral properties of marine sulfated polysaccharides are presented according to their mechanism of action on different types and strains of respiratory viruses, and the potential limits of their use are discussed.

Abstract:Taking the degree of substitution (DS) as the index, the carboxymethylation conditions of Sargassum fusiforme polysaccharide (SFP) were studied. According to the single factor experiment results, the optimum experimental conditions were obtained: sodium hydroxide concentration, 15% (20 mL); alkalization temperature, 50 °C; dosage of chloroacetic acid 1.5 g; etherification time, 2 h, and the Carboxymethyl Sargassum fusiforme polysaccharide (CSFP) with the highest DS (0.635) was obtained. And then, the physicochemical properties, structural information and bioactivity of SFP and CSFP were characterized. The SFP and CSFP were composed of four monosaccharides, with a small amount of protein, and their molecular weights to 780.2 kDa and 386.3 kDa respectively. The results of FTIR and NMR showed that the carboxymethyl was successfully grafted onto the C-4 and C-6 of sugar chain. The results of anti UVC experiment showed that SFP and CSFP had a certain negative effect on cell activity, and the degree of damage caused by UVC radiation was weakened, and the anti UVC performance of CSFP was better than that of SFP.

Abstract:In order to investigate the antioxidant properties of the polysaccharides from the brown alga Sargassum fusiforme , the crude polysaccharides from S. fusiforme (SFPS) were extracted in hot water, and the lipid peroxidation inhibition assay exhibited that SFPS possessed a potential antioxidant activity. Hence, two purely polymeric fractions, SFPS-1 and SFPS-2 were isolated by the column of DEAE (2-diethylaminoethanol)-Sepharose Fast Flow, with their molecular weights of 51.4 and 30.3 kDa determined by high performance gel permeation chromatography (HPGPC). They were preliminarily characterized using chemical analysis in combination of infrared (IR) and nuclear magnetic resonance (NMR) spectroscopies and found to contain large amounts of uronic acids and β-glycosidical linkages. The antioxidant activities of these two SFPS fractions were evaluated using superoxide and hydroxyl radical-scavenging assays. The results show that the antioxidant ability of SFPS-2 was higher than that of SFPS-1, probably correlating with the molecular weight and uronic acid content.

Abstract:To alleviate the adverse effects of chemotherapy and bolster immune function, a novel polysaccharide derived from Sargassum fusiforme named as SFP-αII. The structural composition of SFP-αII predominantly consisted of guluronic and mannuronic acids in a molar ratio of 33.8:66.2, with an average molecular weight of 16.5 kDa. Its structure was primarily characterized by →4)- α -GulA-(1 → and →4)- β -ManA-(1 → linkages confirmed by FT-IR, methylation, and NMR analyses. The absence of a triple-helix structure was in SFP-αII was confirmed using circular dichroism and Congo red dye assays. The dimensions varied with lengths ranging from 20 nm up to 3 μm revealed by atomic force microscopy (AFM). SFP-αII has been found to enhance immunomodulatory activity in cyclophosphamide (CTX)-induced immunosuppressed mice. This was evidenced by improvements in immune organ indices, cytokine levels, and the release of nitric oxide (NO). Specifically, SFP-αII mitigated immunosuppression by upregulating the secretion of IL-1β (167.3 %) and TNF-α (227.1 %) at a dose of 400 mg/kg, compared with the CTX group in macrophages. Ultimately, SFP-αII may serve as a mechanism for immune enhancement through modulation of TLR4-mediated NF-κB and MAPK signaling pathways. This integration of traditional Chinese and Western medicine, leveraging SFP-αII as a potential functional food could be pivotal in alleviating immunosuppressive side effects in CTX treatment.

Glycoprotein from Sargassum fusiforme exhibiting anti-inflammatory responses in vitro and in vivo via modulation of TLR4/MyD88 and NF-κB signaling

Abstract:This study focuses on the identification and characterization of a glycoprotein from Sargassum fusiforme (Harvey) Setchell (SFGP), as well as investigating its potential anti-inflammatory properties both in vitro and in vivo, along with the underlying mechanism. SDS-PAGE analysis revealed a prominent band with a molecular weight of <10 kDa, consisting of 58.39 % protein and 41.61 % carbohydrates, which was confirmed through glycoprotein staining and Coomassie blue staining. Various analytical techniques, including high-resolution mass spectrometry (HRMS), FTIR, amino acid analysis, and UV-visible spectrometry, provided evidence for the presence of monosaccharides (such as d-glucose and mannose) and 17 amino acids linked by an O-glycopeptide bond. In vitro and in vivo studies were conducted to assess the anti-inflammatory activities of SFGP. The results demonstrated that SFGP effectively attenuated nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) expressions in LPS-treated RAW264.7 cells. Moreover, SFGP administration significantly and dose-dependently suppressed TLR4/MyD88 signaling as well as the phosphorylation of MAPKs, IκB, and NF-κB, leading to a reduction in the production of TNF-α, IL-1β, and IL-6 in LPS-stimulated RAW264.7 cells. Furthermore, the anti-inflammatory efficacy of SFGP was validated in a carrageenan-induced inflammatory mouse model. These findings indicate that SFGP exhibits anti-inflammatory characteristics and has the potential to be utilized as a novel anti-inflammatory agent.
Chemically sulfated arabinoxylans from Plantago ovata seed husk: Synthesis, characterization and antiviral activity

Abstract:Limited options for the treatments of diseases triggered through viral infections revealed the quest for novel antiviral drugs. Polysaccharide sulfates owing to their unique mode of action are prominent antiviral drug candidates. Herein, the arabinoxylan of Plantago ovata seed husk was simultaneously extracted and chemically sulfated using sulphur trioxide-pyridine reagent in N,N-dimethylformamide solvent (SO3⋅Py/DMF). Thus, three arabinoxylan sulfates (IS1201-IS1203) holding variable degrees of sulfation (DS: 0.1-0.9), molar masses (18.4-31.3 kDa) and glycosyl makeup (Ara: Xyl::10-19:81-90; molar ratio) were produced and then characterized. According to the results, these polymers displayed anti-herpes simplex virus type 1 activity and their potency depends upon DS. The utmost effective compound (IS1203, IC50: 2.9 μg mL-1) was a 18.4 kDa arabinoxylan possessing sulfate groups at O-3 and O-2,3 positions of xylopyranosyl (Xylp), and O-5 of arabinofuranosyl (Araf) residues. Besides, this polymer showed no cytotoxicity at concentration up to 1000 μg mL-1. Given that polysaccharide sulfates have antiviral activities, synthesis of new molecules possessing diverse structures will be a useful addition to the arsenal of antivirals.
A sulfated glucuronorhamnan from the green seaweed Monostroma nitidum: Characteristics of its structure and antiviral activity

Abstract:A water-soluble polysaccharide from Monostroma nitidum, designated MWS, was isolated using water extraction, anion-exchange and size-exclusion chromatography. MWS was a sulfated glucuronorhamnan consisting of →3)-α-l-Rhap-(1→, →4)-β-d-GlcpA-(1→ and →2)-α-l-Rhap-(1→ units. Sulfate ester groups located at C-4/C-2 of →3)-α-l-Rhap-(1→ and C-4/C-3 of →2)-α-l-Rhap-(1→ units. In in vitro tests, it was proved that MWS possessed broad spectrum against different viruses, especially for enterovirus 71 (EV71) with nearly no toxicity in relation to cell lines used. MWS may largely inhibit EV71 infection before or during viral adsorption through binding to virus particles and block some early steps of virus life cycle by down-regulating host phosphoinositide 3-kinase /protein kinase B signaling pathway. Intramuscular injection of MWS markedly reduced viral titers in EV71-infected mice. The data demonstrated that MWS could have great promising to become an antiviral drug for prevention and therapy of EV71 infection.

Abstract:Objective:to investigate the structure and in vitro immunomodulatory activity of polysaccharides from bamboo shavings (BSP).Methods:the structure was analyzed by the methods of methylation and nuclear magnetic resonance spectroscopy;the in vitro immunomodulatory activity was evaluated by the assays of mouse splenic lymphocyte proliferation,the phagocytosis of neutral red and NO secretion of RAW264.7 cell.Results:BSP was O-acetyl-(4-O-methylglucurono)-arabinoxylans consisting of a linear (l→4)-β-D-xylopyranosyl backbone decorated with branches at O-2 of 4-O-methylglucuronic acid units and acetyl groups or O-3 of α-L-arabinofuranosyl and acetyl groups.The structural element could be expressed as below:1,4-β-D-[2-O-Ac][3-O-Ac][4-O-Me-D-GlcpA-(1→2)][α-L-Araf-(1→3)]-xylp;BSP could evidently strengthen concanavalin A or lipopolysaccharide-induced splenic lymphocyte proliferation at 10-100μg/mL;BSP could significantly enhance the phagocytosis activity and NO production of RAW264.7 cell at 10-1 000 μg/mL in a dose-dependent manner.Conclusion:BSP exhibits excellent immunostimulatory activity in vitro,and can be considered as an ideal source of bio-active polysaccharides due to the abundance of its raw material.

Abstract:The main purpose of this study was to analyze the structural properties and anti-inflammatory activity of the purified fractions derived from UV/H2O2-degraded polysaccharides from Sargassum fusiforme. Results indicated that twofractions with different monosaccharide compositions and morphological characteristics, PT-0.25 (yield 39.5%) and PT-0.5 (yield 23.9%), were obtained. The average molecular weights of PT-0.25 and PT-0.5 were 14.52 kDa and 22.89 kDa, respectively. In addition, PT-0.5 exhibited better anti-inflammatory activity with a clear dose dependence. The mechanism was associated with the inhibition of LPS-activated Toll-like receptor 4-mediated inflammatory pathways in RAW264.7 cells. The results showed that PT-0.5 was a complex polysaccharide mainly composed of 4-Fucp, t-Manp, 6-Galp, t-Fucp, and 3,4-GlcAp. These results would provide theoretical support for studying the structural properties and biological activities of UV/H2O2-degraded polysaccharides.

Physicochemical properties of polysaccharide fractions from Sargassum fusiforme and their hypoglycemic and hypolipidemic activities in type 2 diabetic rats

Abstract:<p>Two polysaccharide fractions (SFPs, designated as respectively SFP-1 and SFP-2) were acquired from <em>Sargassum fusiforme</em> by ultrasound-assisted enzymatic extraction, and their physicochemical properties and hypoglycemic and hypolipidemic effects were investigated. Structural analysis indicated that SFPs were obvious different in the zeta potential, molecular weight distribution, characteristic organic group, microstructure and the contents of total sugar, uronic acid, sulfate and moisture. SFPs consisted of fucose, mannose, rhamnose, glucose, galactose and glucuronic acid with different molar ratios. Congo red test explained that SFPs had no triple-helix structure. SFP-1 exhibited lower viscosity due to its lower molecular weight. Regarding to hypoglycemic and hypolipidemic effects, oral administration of SFPs prominently restrained loss of body weight and increase of water intake, and also significantly controlled the increase of levels of fasting blood glucose, triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), uric acid (UA), urea nitrogen (BUN), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) of diabetic rats, and SFP-2 showed better effects in controlling fasting blood glucose, ALT, UA and BUN levels. Intervention of SFP-2 reduced the levels of insulin, FFA and TBA of diabetic rats. Histomorphological observation further demonstrated that SFPs could attenuate liver and kidney damage caused by hyperglycemia and hyperlipidemia. Data indicated that SFPs, especially SFP-2, significantly improved hyperglycemia, hyperlipidemia and liver and kidney function of diabetic rats.</p>

Abstract:Natural polysaccharides derived from plants, fungi and animals are well known as ideal functional products with multiple biological activities and few side effects. Among them, natural occurring sulfated polysaccharides and those from synthetic origin are increasingly causing more attention worldwide, as they have been proved to possess broad-spectrum antiviral activities. The focus of this review is on analyzing the current state of knowledge about the origin of sulfated polysaccharides, more importantly, the potential connection between the structure and their antiviral mechanisms. Sulfated polysaccharide may interfere with a few steps in the virus life cycle (i.e. adsorption, invasion, transcription and replication) and/or improve the host antiviral immune response. Moreover, their antiviral activity was affected by degree of substitution, substitution position, molecular weight, and spatial conformation. This review may provide approach for the development of novel and potent therapeutic agents.

Hypolipidemic Activity and Mechanism of Action of Sargassum fusiforme Polysaccharides

Abstract:Sargassum fusiforme polysaccharide (SFP) is a kind of biologically active macromolecule with biological functions. In this study, oxidative stress and high-fat HepG2 cell models were established to investigate its lipid-lowering activity and mechanism of action. It was found that SFP and its two isolated fractions had antioxidant effects on the cells. It was also found the polysaccharides decreased the content of total cholesterol and total triglyceride in the high-fat cells. RT-qPCR assays revealed that the three polysaccharides down-regulated the mRNA expression level of ACC, PPARγ, and SREBP-2. It could be concluded that the hypolipidemic effect of SFPs is achieved via multiple pathways, including the regulation on the expression level of lipid metabolism-related key enzymes and factors, and binding with bile acids. The hypolipidemic effect of SFPs could be partially due to their antioxidant activity. SFPs developed in the present work have potential as ingredients of functional foods with hypolipidemic effect.
Marine Sulfated Polysaccharides as Promising Antiviral Agents: A Comprehensive Report and Modeling Study Focusing on SARS CoV-2

Abstract:SARS-CoV-2 (severe acute respiratory syndrome coronavirus-2) is a novel coronavirus strain that emerged at the end of 2019, causing millions of deaths so far. Despite enormous efforts being made through various drug discovery campaigns, there is still a desperate need for treatments with high efficacy and selectivity. Recently, marine sulfated polysaccharides (MSPs) have earned significant attention and are widely examined against many viral infections. This article attempted to produce a comprehensive report about MSPs from different marine sources alongside their antiviral effects against various viral species covering the last 25 years of research articles. Additionally, these reported MSPs were subjected to molecular docking and dynamic simulation experiments to ascertain potential interactions with both the receptor-binding domain (RBD) of SARS CoV-2’s spike protein (S-protein) and human angiotensin-converting enzyme-2 (ACE2). The possible binding sites on both S-protein’s RBD and ACE2 were determined based on how they bind to heparin, which has been reported to exhibit significant antiviral activity against SARS CoV-2 through binding to RBD, preventing the virus from affecting ACE2. Moreover, our modeling results illustrate that heparin can also bind to and block ACE2, acting as a competitor and protective agent against SARS CoV-2 infection. Nine of the investigated MSPs candidates exhibited promising results, taking into consideration the newly emerged SARS CoV-2 variants, of which five were not previously reported to exert antiviral activity against SARS CoV-2, including sulfated galactofucan (1), sulfated polymannuroguluronate (SPMG) (2), sulfated mannan (3), sulfated heterorhamnan (8), and chondroitin sulfate E (CS-E) (9). These results shed light on the importance of sulfated polysaccharides as potential SARS-CoV-2 inhibitors.
