Abstract:Objective. Genetic factors substantially contribute to the development and duration of arterial hypertension. The study of the A1166C polymorphism of the angiotensin II type 1 receptor gene (AGTR1) in arterial hypertension is an auspicious area for assessing the relationship between heredity, hypertension development, and adipokines, but it still remains debatable. The purpose of the current study was to investigate serum adipokines levels depending on the AGTR1 A1166C polymorphism. Methods. A total of 86 patients with arterial hypertension were examined, who underwent the evaluation of the allelic A1166C polymorphism of AGTR1 by polymerase chain reaction with electrophoretic detection and determination of serum adipokines levels using enzyme-linked immunosorbent assay. Results. In the group of patients with arterial hypertension, a significant increase in serum adipokines (resistin, adiponectin, and leptin) levels was found against the background of a decrease in the antianorexic hormone ghrelin with a predominance of CC genotype carriers compared with AA genotype carriers of the AGTR1. A statistically significant decrease in ghrelin and an increase in serum adipokines (resistin, adiponectin, and leptin) in CC genotype carriers compared with AA genotype carriers of the AGTR1 were found suggesting that CC genotype carriers may be predictors of the development of arterial hypertension in our patients. Conclusions. Statistically significant decrease in ghrelin and increase in serum adipokines (resistin, adiponectin, and leptin) were found in CC genotype carriers compared with AA genotype carriers of the AGTR1, which suggests that carriers of the CC genotype are predictors of the arterial hypertension development in our patients.
Association of Single Nucleotide Polymorphisms in ADIPOQ Gene with Risk of Hypertension: a Systematic Review and Meta-Analysis.

Abstract:BACKGROUND:Hypertension has been continuing to be a major contributor to the global burden of disease and to the global mortality, leading to over 10 million deaths each year. The purpose of this study was to investigate the association between Adiponectin gene polymorphism with Essential hypertension (EH).METHODS:PubMed, EMbase, the Cochrane Library, and China National Knowledge Infrastructure (CNKI) were searched independently by two investigators. Pooled odds ratios and 95% confidence intervals were calculated to estimate the associations of Adiponectin polymorphism with EH.RESULTS:Thirteen studies with 3198 cases and 3076 controls for meta-analysis (MA) were included in present study. Pooled results showed that rs2241766 polymorphism is associated with the risk of EH in the allelic model (G vs. T: OR=1.10; 95% CI, 1.01-1.21). In the <40 years subgroup, rs2241766 polymorphism is associated with the risk of EH in allele model (G vs. T: OR=1.43; 95% CI, 1.06-1.94), recessive model (GG vs. GT + TT: OR=5.26, 95% CI=1.47-18.76), homozygous model of GG (GG vs.TT: OR=5.27, 95% CI=1.47-18.95), and rs266729 in recessive model (GG vs. GT + TT: OR=2.33, 95% CI=1.33-4.08).CONCLUSIONS:Our meta-analysis results show that the rs2241766 polymorphism is associated with the risk of hypertension. There still need a larger sample with better design to verify.
Relationship Between Bradykinin B1 Receptor Gene Polymorphism And Antihypertensive Therapy

Abstract:Objective To investigate the 1098A/G gene polymorphism of the human bradykinin B1 receptor in essential hypertensive female patients and the relationship between 1098A/G gene polymorphism and antihypertensive response to ACEI. Methods One hundred fifty female patients with essential hypertensive were received ACEI for four weeks and compare with reduction of BP in different genomic . The 1098A/G was genotyped by the polymerase chain reaction combined with restriction enzyme digestion methods. Results the genotype frequencies were 78.5% ,19.6%,1.9% for AA ,GA,GG, respectively; The reduction of SBP in patients carrying AA and GA + GG were 25. 21±13. 69 mmHg and 34. 50±12. 56 mm-Hg, respectively. A significant difference greater in patients among the two groups was found (P<0.05) . Conclusion The reduction of SBP was significantly greater in patients with the G allele, suggesting that the 1098 A/G polymorphism of human bradykinin B1 receptor extron was possibly related to the response to ACEI in essential hypertensive patients.
A1166c Genetic Variation of the Angiotensin Ii Type I Receptor Gene and Susceptibility to Coronary Heart Disease: Collaborative of 53 Studies with 20,435 Cases and 23,674 Controls

Abstract:Objective: Angiotensin II induces vasoconstriction and vascular smooth muscle growth via stimulation of the angiotensin II type I receptor (AGTR1). Some studies have reported an association between a genetic variant (A1166C) in the 3' un-translated region of AGTR1 and increased risk of coronary heart disease (CHD), but other have yielded apparently conflicting results.Methods: Literature-based meta-analyses were performed on 48 papers including 53 studies published before June 2008 in relation to the A1166C polymorphism (NCBI, dbSNP: rs5186) of the AGTR1, involving a total of 20,435 CHD cases and 23,674 controls. We also explored potential sources of heterogeneity and conducted appropriate stratified analyses.Results: In a combined analysis, the per-allele odds ratio (OR) for CHD of the A1166C polymorphism was 1.11 (95% confidence interval: 1.03-1.19), but there is an indication of publication bias and heterogeneity among the 53 studies. Sample size and study quality were significant sources of heterogeneity among studies of the A1166C polymorphism with possibly overestimates in studies of smaller sample-size and poor-quality. When the analyses were restricted to 11 larger studies (>= 500 cases), and to 8 high-quality studies (quality score: >= 11 points), the summary per-allele odds ratios were 0.992 (95% confidence interval, 0.944-1.042) and 0.990 (95% confidence interval, 0.915-1.072), respectively.Conclusions: An overall weak association between the A1166C polymorphism and CHD is observed but this is likely to be due to publication bias and heterogeneity between studies. There were no significant associations among the larger sample-size and high-quality studies which are less prone to selective publication and have greater power to detect a true association. Abstract:Objective: To determine the peculiarities of lipid metabolism indicators’ changes in healthy individuals with a genetic predisposition to arterial hypertension (AH) and patients with essential arterial hypertension (EAH). Design and method: 231 patients with second stage of EAH (105 men and 126 women; average age was 52.3±1.2 years) were examined. The duration of EAH was 9.7±0.45 years. 121 of them had a concomitant stable coronary artery disease (CAD). The control group included 30 healthy people of similar age and gender. A comparison group consisted of 23 healthy individuals with a AH family history. Additionally to lipid spectrum, levels of lipoprotein (a) (Lp(a)), apolipoprotein B100 (apoB100) and apolipoprotein A-1 (apoA-1) were determined. Statistical calculations were performed using Statistica for Windows 10.0. Results: Patients with genetic predisposition to AH had similar to indicators in patients with II EAH stage proatherogenic changes in lipid fractions. Their Lp(a) levels increased to 29.9 (24.5; 32.1) mg/dl, which is 67.0% higher than the levels in the control group (p=0.0005). Levels of apolipoproteins (apoB100 and apoA1) in the serum of healthy persons with AH predisposition did not differ from the control group. Patients with stage II EAH showed an increase in proatherogenic fractions of lipoproteins, apoB100, Lp(a), and a decrease in high-density lipoprotein cholesterol (HDL-C) and apoA-1 compared to control (p<0.0001). Analysis of lipid indicators in patients with concomitant CAD compared to patients without CAD revealed no significant differences between these two groups, except of increasing in the Lp(a) level by 24.4% (p<0.05). A significant positive correlations were found between the Lp(a) level with both the sum of scores on the SCORE scale and the CAD presence (r=0.25 and r=0.35, p=0.002), respectively. Conclusions: The lipid metabolism disorders in people with determinism for AH begin even before an increase in blood pressure and are genetically determined. The addition of hypertension contributes to the further progression of lipid metabolism disorders. The obtained data indicate the high prognostic significance of Lp(a) in patients with EAH in the development and establishment of both hypertension itself and CAD and increasing of patients’ cardiovascular risk.

Abstract:OBJECTIVE:To investigate whether the angiotensin II type I receptor gene (AGTR1) A1166C polymorphism is associated with a high risk of diabetic nephropathy.METHODS:The allele frequency and the genotype distribution of the AGTR1 A1166C polymorphism were studied in normal controls (157 cases), simple diabetes (141 cases, duration of diabetes >10 years), and diabetic nephropathy (152 cases) by means of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).RESULTS:Patients with diabetic nephropathy had a higher frequency of C allele of the AGTR1 A1166C polymorphism than that of normal controls and simple diabetes (P<0.05); but there was no significant difference in frequency of C allele between the normal controls and patients with simple diabetes.CONCLUSION:The diabetic patients with AGTR1 C allele may be more susceptible to diabetic nephropathy than diabetic patients with A allele.

Abstract:Objective — to reveal an association of I/D polymorphism of ACE gene with metabolic factors of cardiovascular risk (CVR) such as visceral obesity (VO), insulin resistance (IR), dyslipidemia (DLP) as well as with indices characterized structural and functional state of left cardiac chambers in patients (pts) with diabetic nephropathy (DN) and essential hypertension (EH). Materials and methods. The investigation was conducted at the Clinical Department of Arterial Hypertensions and Kidney Diseases in L. T. Mala National Therapy Institute of NAMS of Ukraine (Kharkiv). Clinical examinations involved 82 pts, 42 (51.2 %) females and 40 (48.8 %) males with DN of I — IV stage and EH of II — III stage, aged 31 to 82 years old (an average age is (61.65 ± 1.28) years old). Three genotypes of I/D polymorphism of ACE gene included II — 18 (24.7 %), ID — 32 (43.8 %) and DD — 23 (31.5 %) of cases correspondingly were investigated in 73 (89.0 %) pts. Anthropometric measurements with calculations of body mass index (BMI), body fat percentage (BFP), body fat mass (BFM), fat mass index (FMI) were conducted by known formulas in all pts. Blood lipids were detected by enzyme method with measuring of total cholesterol, high‐density lipoprotein cholesterol (HDL‐C) and triglycerides (TG). Levels of cholesterol of very low density and low‐density lipoprotein (VLDL‐C, LDL‐C) were calculated by standard formulas. Blood glucose level was measured by glucose oxidase method and serum insulin was detected by immunoassay method. IR indices such as HOMA‐IR, triglyceride glucose index (TGGI) and METS‐IR (metabolic score for insulin resistance) were obtained by known formulas. I/D polymorphism of ACE gene (rs 4646994) was investigated with a usage of polymerase chain reaction. Structural and functional state of left cardiac chambers was estimated using transthoracic echocardiography. Systolic and diastolic blood pressure (SBP and DBP) were measured by Korotkov’s method. The results were statistically obtained with computer programs Microsoft Office Excel 2003 and Statistica 23.0. Results. In pts with II genotype, LDL‐C levels positively correlated with BFP (r = 0.490; p = 0.046) and BFM (r = 0.484; p = 0.049) in conditions with hypertriglyceridemia due to activation of visceral fat lipolysis in which a reverse correlation between relative thickness of left ventricle wall (RTLVW) and serum TG concentration (r = –0.540; p = 0.02) could be explained. In pts with ID genotype, presence of D allele in genotype was associated with an increase in RTLVW due to SBP influence (r = 0.358; p = 0.045). In genotype ID there was a relationship between IR index HOMA‐IR and BMI (r = 0.399; p = 0.029) and HOMA‐IR and FMI (r = 0.402; p = 0.025) that demonstrated VO participation of in the development of IR in pts with DN and EH and genotype ID. In pts with DN and EH, who had DD genotype, SBP level reversely correlated with HDL‐C (r = –0.498; p = 0.018) that could be explained by influence of IR on elevation of SBP and reduction of HDL‐C. In pts with DD genotype both SBP and DBP almost equally influenced on the left atrial diameter (r = 0.460; p = 0.036 and r = 0.453; p = 0.034 correspondingly). In pts with DN and EH whose had DD genotype IR index METS‐IR positively correlated with RTLVW (r = 0.419; p = 0.047) and left ventricle myocardial mass index (r = 0.518; p = 0.011) that confirmed an association of D allele of ACE gene with left ventricle hypertrophy caused both by EH and IR. Conclusions. It has been established that in patients with diabetic nephropathy and essential hypertension, the I/D polymorphism of ACE gene was associated with such metabolic factors of cardiovascular risk as visceral obesity, insulin resistance, dyslipidemia, and pathological left ventricle remodeling.
Association of endothelial lipase gene Thr111Ile and Gly26Ser polymorphism with lipoprotein in patients with coronary heart disease

Abstract:AIM:To investigate the association of endothelial lipase gene(LIPG)Thr111Ile and Gly26Ser polymorphism with lipoprotein in patients with coronary heart disease(CHD)in Chinese.METHODS:438 patients were classified as 242 CHD group and 196 controls group by selective coronary angiography.Plasma level of lipoprotein was determined and the Thr111Ile and Gly26Ser polymorphism was screened by PCR-RELP.RESULTS:The frequencies of Thr111Ile genotype in Chinese were CC 76.7%,CT 23.3%,TT 0.0%.The frequencies of allele were C 88.3%,T 11.7%.The plasma level of HDL-c in CT group was significantly higher than that in CC group(P<0.05)on logistic regression analysis.However,logistic regression analysis revealed that there was no significant difference between CHD group and control group for Thr111Ile polymorphism(P>0.05).No Gly26Ser mutation was observed in this study.CONCLUSION:The polymorphism of Thr111Ile is present in patients with CHD in Chinese,and T allele is related to high HDL-c level.There is no significant association between the polymorphism of Thr111Ile and CHD.The Gly26Ser mutation has not found in this study.

Abstract:Metabolic violations, obesity and other related comorbidities are obvious in arterial hypertension. It is known that Hepatic Steatosis (HS) and Arterial hypertension (AH) has to some extend common pathogenesis realized through metabolic and immune mechanisms involving vascular and digestive system injury, and forming Gut-Liver axis, which remain mostly untouched in recent studies. We aimed on evaluating the endothelial function and mesenteric vessels remodeling depending on I/D polymorphism of angiotensin-converting enzyme (ACE) gene and A1166C polymorphism of angiotensin II type 1 receptor (AGTR1) gene in hypertensive patients with HS. Study included 104 AH patients with HS (50 females, 54 males, age 53.2±8.7). Intimae-media thickness (IMT) of abdominal aorta (AO) and other flow mediated parameters of mesenteric vessels state evaluated by sonography. NO (nitrite/nitrate) plasma concentration, vascular adhesive molecule (sVCAM-1) level were defined by IEA. ACE (I/D) and AGTR1 (A1166C) genes polymorphisms assessed with PCR. High risk of endothelial dysfunction (ED) is: D-allele of ACE gene (p<.001); C-allele of AGTR1 gene (p = .02). In DD-genotype were higher flow-mediated parameters of mesenteric vessels. IMT of AO was thicker in CC-genotype of AGTR1 gene (p = .039-.01) and independent on I/D genotypes of ACE gene. sVCAM-1 level was higher (p<.01) in D-allele of ACE gene, than in II-genotype and not depending on A1166C polymorphism of AGTR1 gene. NO plasma level doesn’t depend on ACE (I/D) and AGTR1 (A1166C) genes polymorphisms. Vascular remodeling in HS patients (AO IMT enlargement for more than 0.9 mm) is associated with dysbiosis III-IV grades’ and sVCAM-1 plasma level growth by 1,3-1,6 times (p<.05-.001). AO IMT correlates with dysbiosis severity, presence of HS and sVCAM-1 level and is independent from AH severity. Risk contingents for endothelial dysfunction and mesenteric vessels remodeling in HS patients are: by sVCAM-1 plasma level and diameter of mesenteric vessels increasing – DD-genotype carriers of ACE gene; by IMT of abdominal aorta enlargement – CC-genotype carriers of AGTR1 gene (p<0.02-0.001).

Alpha-adducin 1 (rs4961) gene and its expression associated with sodium sensitivity in hypertensive patients: a cohort study in the western Ukrainian population

Abstract:Objective. The aim of this study was to evaluate the association of the α-adducin-1 gene (ADD1) (Gly460Trp [rs4961]) polymorphism and its expression in association with renal dysfunction and sodium sensitivity in hypertensive patients in western Ukrainian population. Methods. One-hundred patients with essential arterial hypertension (EAH) and hypertensive-mediated target organ damage (stage 2), moderate, high, and very high cardiovascular risk were enrolled in case-control study. Sixty healthy individuals were assigned as controls. Sodium sensitivity and sodium resistance were determined by salt load reaction. The ADD1 (rs4961) genotyping was performed in RT-PCR. Results. The expression of the quantitative trait loci (eQTL) of ADD1 gene (rs4961) (chr4:2906707 [hg19]) was confirmed in 37 tissues and organs with 23 phenotypic traits. Two hundred eQTL associations revealed - all cis-variants (cis-QTL); 73 methylation QTL (mQTL), 34 splicing QTL (sQTL), 14 histone modification QTL (hQTL), 2 protein QTL (pQTL), 23 transcript utilization QTL (tuQTL), and 4 loci of incorporated long noncoding areas of RNA (lncRNA). GG-genotype unreliably enhances EAH risk (OR=1.92; 95%CI: 0.90-4.10; p=0.066). Sodium sensitivity was observed in 54.0% of patients and in 20.0% of controls (c2=17.89; p<0.001). Sodium sensitivity in T-allele carriers of the ADD1 gene (1378G>T; rs4961) dominated 12-fold in general (OR 95%CI: 2.24-64.29; p=0.001), in women - 4.71 times (OR 95%CI: 1.92-11.56; p<0.001), and in men - 4.09 times (OR 95%CI: 1.03-16.28; p=0.041). Sodium sensitivity elevated the likelihood of severe EAH twice (OR=2.19; OR 95%CI: 1.00-5.05; p=0.049). Conclusion. T-allele associates with sodium sensitivity in essential arterial hypertension patients and increases the risk of hypertension regardless the gender. Sodium sensitivity enhances the probability of severe essential arterial hypertension in observed population.

Abstract:Objective: No clear consensus has been reached on the β2-adrenergic receptor polymorphisms (A46G and C79G) and essential hypertension risk. We performed a meta-analysis to summarize the possible association. Methods From 301 reports, we included 18 articles (20 studies) at the inclusion criteria. The fixed-effects model and the random-effects model were applied for dichotomous outcomes to combine the results of the individual studies. Results: There was no statistical association between A46G and hypertension risk in all subjects, Asians or Caucasians. However, the association was observed in dominant genetic model (AA vs. (AG + GG)) (P = 0.04, OR = 1.38, 95% CI [1.01–1.87], Pheterogeneity = 0.98) in the subgroup of the mixed Africans. No statistical association could be found between C79G and hypertension risk in overall or any ethnic subgroup. In the research conducted on the severe hypertension (SBP ≥160mmHg and/or DBP≥95mmHg hypertensive population), significant association could be found in dominant genetic model (CC vs. (CG + GG)) (P = 0.04, OR = 1.38, 95% CI [1.02 - 1.86], Pheterogeneity = 0.03), and there was also a boardline significance between the C79 allele and severe hypertension (P = 0.05, OR = 1.26, 95% CI [1.00–1.57], Pheterogeneity = 0.04). No association could be found in the study between these two polymorphisms and stage 2 hypertension. Conclusion Significant association between ADRB2 A46G polymorphism and hypertension was found in the population of the mixed African. The ADRB2 C79G polymorphism has significant association with the SBP ≥160 mmHg and/or DBP ≥95 mmHg hypertensive population.

There was no statistical association between A46G and hypertension risk in all subjects, Asians or Caucasians. However, an association was observed in the dominant genetic model (AA vs . (AG+GG)) ( P =0.04, odds ratio (OR)=1.38, 95% confidence interval (CI) 1.01–1.87, P heterogeneity =0.98, fixed-effects model) in the subgroup of mixed Africans. No overall statistical association could be found between C79G and hypertension risk or any ethnic subgroup. In the research conducted on severe hypertension (systolic blood pressure ⩾160 mm Hg and/or diastolic blood pressure ⩾95 mm Hg hypertensive population), significant association was found in the dominant genetic model (CC vs. (CG+GG)) ( P =0.04, OR=1.38, 95% CI 1.02–1.86, P heterogeneity =0.03, random-effects model), and there was also a borderline significance between the C79 allele and severe hypertension ( P =0.05, OR=1.26, 95% CI 1.00–1.57, P heterogeneity =0.04, random-effects model). No association could be found in this study between the two polymorphisms and stage 2 hypertension. More studies stratified for different ethnicities and different stages of hypertension should be performed in the future.

Abstract:Objective: Arterial hypertension (AHT) is a multifactorial pathology in which environmental and genetic factors interact leading to the development of this pathology. The genetic influence on the onset of AHT has been addressed in candidate gene studies. Polymorphisms that interfere with sodium and water balance are described in the literature as associated with increased risk of AHT. However, there are few studies that study the synergism between them in the risk of AHT. Objective: Evaluate whether there is a synergistic effect between two polymorphisms that interfere with sodium and water balance, alpha adducin (ADD1 G460W rs4961) and sodium epithelial channel (SCNN1G G(-173)A rs5718), in the increased risk of AHT. Design and method: A case-control study was conducted with a population of 1712 individuals (mean age of 51.0±7.9), depending on whether or not they had AHT. We obtained two groups: 860 hypertensive patients and 852 controls. All subjects collected blood for analytical tests and DNA for genetic analysis. The polymorphisms rs4961 and rs5718 were evaluated in both groups. The Odds Ratio (OR) with 95% confidence intervals were calculated. Data analysis was performed using SPSS for Windows, version 25.0. The significance threshold was set at p<0.05. Results: The WW genotype of ADD1 was more frequent in the hypertensive group than in the control group (OR 2.76; 95%CI:1.38-5.55; p=0.004). The GG genotype of SCNN1G was not statistically significant between both groups (OR 0.95; 95%CI: 0.78-1.16; p=0.597). However, when we associate the two genetic variants, there is an increased risk of AHT, with an OR of 7.68 (95% CI 1.75-33.73) with statistical significance (p=0.007). Conclusions: With our results, we can conclude that the alpha adducin polymorphism (ADD1 G460W) is a statistically significant risk factor for AHT, and the sodium epithelial channel polymorphism (SCNN1G G(-173)A) alone is not associated with AHT onset. However, when together, these two polymorphisms increase the risk of AHT to na OR of 7.7. These results lead us to conclude that AHT is not explained in a simple way with a single polymorphism, since there are synergistic effects between genes that favor the development of AHT.

[Association Between AvaII Exon 13 Polymorphism at the LDL Receptor Gene Different and Serum Lipid Levels in Normotensives and Essential Hypertensives in Shanghai].

Abstract:OBJECTIVE:To investigate the AvaII polymorphism of low density lipoprotein receptor gene in both health and essential hypertension populations, and to evaluate the association of AvaII polymorphism with level of blood lipid. METHODS:Using polymerase chain reaction (PCR), AvaII polymorphism was studied in 109 health individuals and 319 patients with essential hypertension. RESULTS:There were three kinds of genotype: (+/+), (+/-), (-/-). The frequencies of the three genotypes were shown as follows: (+/+) 0.9%, (+/-) 27.5%, (-/-) 71.6% in health population and (+/+) 1.2%, (+/-) 33.9%, (-/-) 64.9% in essential hypertension population, respectively. The frequencies of the two alleles were shown as follows: (+) 14.7%, (-) 85.3% in health population, (+) 18.2%, (-) 81.8% in essential hypertension population and (+) 17.3%, (-) 82.7% in the community, respectively. In male essential hypertension cases, the genotypes were associated with serum TC and LDL-C level in the following order: (-/-) < (+/-) (P < 0.05). In women and healthy population, there was a similar trend but not statistically significant (P > 0.05). CONCLUSIONS:A significant association was found between the common variation of LDL-R gene and serum TC and LDL-C levels. (+) Allele was associated with elevated level of serum TC and LDL-C, but (-) allele was associated with a low level of serum TC and LDL-C. The frequencies of (-) allele in both group were related to serum low level TC while LDL-C was much higher than that reported in the western countries. These data indicated that genetic factors which resistant to hypercholesterolemia in Chinese people were different from those findings in West while might be one of the reasons to explain why that serum TC level in Chinese was lower than people in the western countries.

Abstract:Background: There is considerable variability in individual response to antihypertensive agents. The reason for this is not known, but may be related to individual genetic variability. This study examined whether the therapeutic efficacy of benazepril on essential hypertension is modified by β2 adrenergic receptor gene (ADRB2) Arg16Gly (R16G) polymorphism. Methods and Results: We conducted a family‐based study of 321 and 610 hypertensive subjects from Yuexi and Huoqiu Counties of Anhui, China, respectively. Both systolic and diastolic blood pressures (SBP and DBP) before and after a 15‐day benazepril treatment were measured. ADRB2 R16G genotypes were determined for all subjects. ADRB2 G16 allele frequency was found to be 41.0% and. 47.4% in Huoqiu and Yuexi, respectively. In Yuexi family‐based association test (FBAT) revealed that the G16 allele was associated with a greater DBP decrease in response to a 15‐day benazepril treatment (Z = 2.12, P = 0.03), and the data were consistent with a dominant inheritance model. A similar trend was observed in Huoqiu Chinese, but the magnitudes of effects were smaller and did not reach statistical significance. The FBAT results were further confirmed by using a generalized estimating equation model. Conclusion: Our family‐based study provided the first evidence that ADRB2 R16G polymorphism may play an important role in DBP response to benazepril treatment, although the magnitude of the effect appears to be modified by other risk factors such as plasma lipid and glucose profiles.

Abstract:Objective — to search an association of rs1799983 endothelial nitric oxide synthase (eNOS) gene polymorphism with dyslipidemia (DLP) and the efficacy of atorvastatin used in correction of lipid exchange disorders in patients with diabetic nephropathy (DN) and essential hypertension (EH). Materials and methods. The study had been conducted in the clinical Department of Arterial Hypertension and Kidney Disease at L. T. Mala National Therapy Institute of NAMS of Ukraine (Kharkiv). Clinical examinations involved 89 patients (pts) (43(48.3%) females and 46(51.7%) males) with DN of II—IV stages and EH of II—III stages aged 34 to 78 years old (an average age is 57.11±1.89 years old). They were also genotyped for rs1799983 eNOS gene polymorphism (G894T) with a use of polymerase chain reaction. GG genotype was revealed in 29(32.6%) of pts, GT genotype was found in 33(37.1%) of cases and 27(30.3%) participants of the study had TT genotype. With the use of immune enzyme method, serum levels of total cholesterol (TC), high‐density lipoprotein cholesterol (HDL‐C) and triglycerides (TG) were measured in all included pts. Very low density lipoprotein cholesterol (VLDL‐C), low‐density lipoprotein cholesterol (LDL‐C), non‐HDL‐C and coefficient of atherogenicity were calculated by known formulas. Atorvastatin was prescribed to all the pts included in study for correction of lipid exchange disorders in daily doses of 10—20 mg (an average daily dose was 16.7±0.91 mg) for 24 weeks. Results. Rs1799983 eNOS gene polymorphism in pts with DN and EH was found to be associated with lipid parameters characterized a phenotype of diabetic DLP with its peculiar disorders in the systems of reverse cholesterol transport and lipoprotein lipolysis of TG‐containing VLDL. In this case a dominant G allele was associated with less significant lipid disorders than a recessive T allele. A homozygous state with allele G (genotype GG) in pts with DN and EH was associated with abnormalities in the system of TG‐containing VLDL lipolysis and an appearance of T allele in genotype (genotypes GT and TT) made an additional negative contribution to the state of lipid exchange and transport. In pts with genotype GT abnormalities in reverse cholesterol transport added to disorders in the system of TG‐containing VLDL lipolysis and in pts with genotype TT there was a combination of disorders in the systems of direct, reverse cholesterol transport and lipoprotein lipolysis. The study of hypolipidemic effects of atorvastatin depended on genotype of rs1799983 eNOS gene polymorphism showed that in GG genotype atorvastatin demonstrated mainly a hypocholesterolemic action while in genotypes contained T allele (GT and TT) we observed a reduction of hypocholesterolemic effect against the background of significant hypotriglyceridemic action and elevation of HDL‐C. Conclusions. In pts with DN and EH rs1799983 eNOS gene polymorphism was associated with diabetic DLP. A recessive allele T was associated with significant lipid disorders compared with a dominant G allele. A significant hypocholesterolemic effect of atorvastatin was observed in case of a presence of G allele in pts genotype while a presence of T allele in pts genotype (GT and TT) was associated with a reduction of atorvastatin cholesterol lowering action.

Abstract:Objective: To investigate whether the genetic polymorphisms in the angiotensin I converting enzyme ( ACE ) (insertion/deletion, or I/D), angiotensin II type 1 receptor ( AT1R ) (rs5186), and ACE2 (rs2285666) could be associated with dyslipidemia in Type 2 diabetic (T2D) patients of Chinese Han origin. Design and methods: The above 3 polymorphisms were genotyped in a total of 282 patients with T2D and dyslipidemia (Group A), 182 patients with T2D but without dyslipidemia (Group B), and 324 healthy controls. The association between a certain polymorphism and each group was assessed by an odds ratio (OR). Results: The D allele of the ACE (I/D) was significantly associated with the risk of T2D accompanying dyslipidemia between group A and controls [OR=1.37, 95% confidence interval (CI)=1.08–1.74; p=0.010], and significant association of the D allele with dyslipidemia was also observed in diabetic patients (OR=1.88, 95% CI=1.40-2.54; p <0.001). Furthermore, the ID genotype had a decreased risk of developing T2D without dyslipidemia as compared with controls (OR=0.52, 95% CI=0.32–0.82; p =0.0060). The distributions of the AT1R (rs5186) and ACE2 (rs2285666) genotypes and alleles did not differ between T2D patients with or without dyslipidemia and the controls. Conclusions: This study demonstrates that the ACE (I/D) polymorphism is associated with T2D, regardless of the absence or presence of dyslipidemia. The polymorphisms in the AT1R (rs5186) and ACE2 (rs2285666) seem to play lesser roles in the development of T2D.

Abstract:Objectives Peroxisome proliferator-activated receptor(PPAR)-γcoactivator 1α(PGC-1α)was the first member identified as a cofactor for the nuclear hormone receptor PPAR_γ.It plays a critical role in the control of energy thermogenesis,including glucose uptake in muscle,and hepatic gluconeogenesis in liver.In view of the close relationship between type 2 diabetes and hypertension in clinical elements and etiology,the present study focused on the association of polymorphisms in PGC-1αgene with lipid levels in diabetic patients with hypertension in northern Chinese population.Method We selected 427 cases with type 2 diabetes and hypertension and 549 age-and sex-matched normal controls.All participants were unrelated Han population from the northern area of China.All subjects have not taken medication to regulate lipids.The polymerase chain reaction-restriction fragment length polymorphism was used to genotype the polymorphisms of 1302G/A(rs2970847)and G482S(rs8192678)in the PGC-1αgene.Results In case group,the 1302G/A polymorphism was associated with the triglycerides(TG)level adjusted by age, gender and body mass index(P=0.0296 and 0.0147 in the additive and recessive model,respectively). Patients carried with 1302A allele have a lower low-density lipoprotein cholesterol(LDLc)level than homozygous GG in the dominant model(P=0.0412).For the G482S polymorphism,patients with GG genotype have a lower LDLc level than patients with 482S allele(P=0.0467).Compared with patients with 482S allele, patients with 482GG genotype had a lower level of total cholesterol,although not significantly(P= 0.0699).Among patients with 6 common diplotypes composed by these two polymorphisms there were borderline difference in the level of TG(P=0.0502).Patients with 1302AA-482GG had the lowest TG level which confirmed the results of single SNP analysis.No similar findings were in control group.Conclusion The 1302G/A and G482S polymorphisms were associated with the lipid phenotypes among patients with type 2 diabetes and hypertension.These results suggested that 1302A and 482G alleles might be protective factors of abnormal level of lipids in Northern Han Chinese population.

Abstract:This study was conducted to explore whether the renin C‐5312T, angiotensin II type 1 receptor A1166C, and angiotensin‐converting enzyme I/D polymorphisms were associated with ambulatory blood pressure (BP) and central hemodynamics in an untreated hypertensive population. A total of 471 participants with no previous treatment for raised BP were eligible for the study. Ambulatory and central BP were measured. DD carriers had the highest daytime systolic/diastolic BP, nighttime systolic BP, 24‐hour systolic BP, and 24‐hour diastolic BP values, whereas carriers of DD had higher central systolic BP and augmentation index compared with those with the II genotype. Multivariate regression analysis demonstrated that DD genotype was independently associated with 24‐hour systolic BP, 24‐hour diastolic BP, central systolic BP, and central augmentation index. There was an independent association of the angiotensin‐converting enzyme polymorphism with central and ambulatory BP in Chinese patients with hypertension.

Abstract:Background: CYP4A11 is a member of the cytochrome P450 enzymes and is responsible for metabolizing arachidonic acid to 20-hydroxyeicosatetraenoic acid, a metabolite involved in the regulation of blood pressure. This study aimed to evaluate whether or not the CYP4A11 gene polymorphism T8590C (rs1126742) is involved in essential hypertension in the western Chinese Han population.Methods: In a case-control study, the participants included 864 (523 males and 341 females) patients with essential hypertension and 661 (422 males and 239 females) healthy subjects. The T8590C polymorphism of the CYP4A11 gene was analyzed by using the TaqMan (R) SNP Genotyping Assay.Results: For men, the frequencies of the CC genotype and the C allele were higher in essential hypertension than in the control group (p = 0.022 and p = 0.016, respectively). After adjustment of confounding factor such as diabetes, smoking, BMI, TG and TC, the significant difference was observed in CC genotype (OR 1.897, 95% confidence interval [CI] 1.026-3.508; p = 0.041). No difference was found in all participants and females.Conclusions: The CC genotype and C allele were associated with essential hypertension in the male western Chinese Han population.
