Abstract:Abstract Objectives To evaluate the antimicrobial efficacy of white vinegar, acetic acid, and peracetic acid on photostimulable phosphor (PSP) plates disinfection, and to assess the disinfectant influence on the radiographic quality. Methods Eight PSP plates (Express system) were contaminated with Streptococcus mutans and Candida albicans. These plates were wiped with tissues without any substance, with white vinegar, acetic acid, and peracetic acid, followed by an agar imprint. Number of microbial colonies formed was recorded. Afterwards, the quality of radiographs was tested using the more efficient disinfectant. Before disinfection and after every five disinfections, two radiographs of an acrylic-block and two radiographs of an aluminium step-wedge were acquired for each plate. Density, noise, uniformity, and contrast were analysed. Three oral radiologists evaluated the images for the presence of artefacts. One-way analysis of variance compared changes on grey values among the disinfections (α = .05). Intra- and inter-examiner agreement for the presence of artefacts was calculated by weighted Kappa. Results Peracetic acid was the only one that eliminated both microorganisms. Density and uniformity decreased after 100 disinfections, and contrast changed without a pattern in the course of disinfections (P ≤ .05). Small artefacts were observed after 30 disinfections. Intra- and inter-examiner agreements were almost perfect. Conclusions Disinfection with peracetic acid eliminated both microorganisms. However, it also affected density, uniformity, and contrast of radiographs, and led to the formation of small artefacts.

Antimicrobial Photodynamic Therapy as a Technique for Decontamination of Acrylic Resin Devices Provided by Different Dental Laboratories

Abstract:Introduction: Dentures, occlusal splints, surgical guides and orthodontic appliances are examples of acrylic resin devices made in dental laboratories, which must be disinfected and even sterilized before insertion into the oral cavity. This study evaluated the antimicrobial effect of photodynamic therapy (PDT) applied to acrylic resin specimens received from different laboratories. Methods: Three hundred standardized specimens were ordered from six randomly selected laboratories registered in the Council of Dentistry of Ceará (n=50). The PDT consisted in the association of 22 µM erythrosine, as a photosensitizer (P), and a 520-nm LED at 38 J/cm2 (L). The specimens of each laboratory were randomly distributed into five groups: positive control, sterilized with ethylene oxide; negative control, untreated (P-L-); erythrosine control, only stained (P+L-); LED control, only irradiated (P-L+); PDT (P+L+). Then, the specimens were individually sonicated in saline solution; the suspension was diluted, plated on culture mediums (blood agar, sabouraud dextrose agar and a non-selective chromogenic agar), and incubated for 48 hours at 37°C. Colony-forming-unit (CFU) counts were done and statistical tests of Kruskal-Wallis/Dunn were carried out. Results: The specimens from all laboratories were contaminated with bacteria and yeasts. Staphylococcus aureus, Staphylococcus saprophyticus, Escherichia coli, Enterococcus spp., Klebsiella and Pseudomonas spp. were identified. The PDT significantly reduced CFU counts (P<0.0001), compared to P-L-. Conclusion: PDT was able to effectively decontaminate the acrylic resin specimens provided from dental laboratories.
In Vitro Comparative Evaluation of the Efficacy of Antimicrobial Photodynamic Therapy, Chlorhexidine, Sodium Fluoride, and Hydrogen Peroxide for Acrylic Resin Disinfection

Abstract:Introduction: Plaque accumulation on the surface of removable orthodontic appliances could lead to dental caries, periodontitis, and fungal infections. This study evaluated the effectiveness of antimicrobial photodynamic therapy (aPDT), chlorhexidine (CHX), sodium fluoride (NaF), and hydrogen peroxide (H2O2) for the disinfection of acrylic resin. Methods: In this in vitro experimental study, 100 acrylic resin specimens were randomly divided into five groups (n=20 each): Enterococcus faecalis, Streptococcus mutans, Streptococcus salivarius, Streptococcus sanguinis, and Lactobacillus acidophilus. Each group was immersed separately in 5 mL of microbial suspension. They were then incubated until biofilm formation on their surface. Of each microorganism, one biofilm sample in phosphate-buffered saline was considered as negative control, and other biofilm samples (n=80) were subjected to aPDT with curcumin, 0.12% CHX (positive control), 1% H2O2, and 0.2% NaF. Finally, the number of colonies was counted. Data were analyzed by the Kruskal-Wallis and Mann-Whitney tests, two-way ANOVA, and Bonferroni adjustment at a significance level of 0.05. Results: The interaction effect of the treatment modality and type of microorganism was significant on the microbial count (effect size: 0.91, P<0.05). Maximum bacterial proliferation was noted in the following combinations: NaF/E. faecalis, H2O2/E. faecalis, and H2O2/S. salivarius. Microorganisms had no or insignificant growth and proliferation in the aPDT and CHX groups. Conclusion: The results supported the optimal antimicrobial efficacy of PDT which was comparable to that of CHX. aPDT showed superior antimicrobial efficacy to NaF and H2O2 for the disinfection of acrylic resin.

Abstract:INTRODUCTION: This randomized clinical trial assessed, by using microbial culture and scanning electron microscopy (SEM), the contamination by mutans streptococci (MS) colonies/biofilms on acrylic baseplates of removable orthodontic appliances and evaluated the efficacy of antimicrobial sprays (Periogard [Colgate-Palmolive Ind. Brasileira, Osasco, SP, Brazil], Cepacol [Merrell Lepetit Farmacêutica e Industrial Ltda, Santo Amaro, SP, Brazil], and sterile tap water [control]) on their disinfection.METHODS: Seventeen children were randomly enrolled in a 3-stage changeover system with a 1-week interval between each stage. All solutions were used in all stages by a different group of children. The acrylic baseplates were worn full time except at meals. At the end of each week of the trial, the baseplates were submitted to a randomized disinfection protocol and were sent for microbiologic analysis. New baseplates were constructed, and the same sequence of procedures was repeated 2 more times. Acrylic baseplates representing each test solution were examined by SEM. The Friedman test assessed differences at the 5% significance level among the solutions for MS biofilm formation on acrylic surface.RESULTS: Cepacol and Periogard reduced the formation of MS colonies/biofilms, and both solutions differed statistically from sterile tap water (P <.001). However, Periogard was significantly more effective against MS than Cepacol (P <.001). When MS colonies/biofilms were detected on acrylic surfaces under stereomicroscopy, this was confirmed with SEM.CONCLUSIONS: Acrylic baseplates of removable orthodontic appliances worn by children were contaminated by MS colonies/biofilms in all cases after 1 week. Although Cepacol had better results than sterile tap water, spraying with Periogard showed significantly greater efficacy in reducing MS colonies/biofilms on acrylic surfaces.

Abstract:A representation formalism for N-ary relations, quantification, and definition of concepts is described. Three types of conditions are associated with the concepts: (1) necessary and sufficient properties, (2) contingent properties, and (3) necessary properties. Also explained is how complex chains of inferences can be accomplished by representing existentially quantified sentences, and concepts denoted by restrictive relative clauses as classification hierarchies. Abstract:In a case control study, 70 children consecutively hospitalized for acute ingestion of poisons were compared with 140 other hospitalized children matched for age. Children aged less than 3 years and boys were most often the victims. Univariate analysis identified Indian race, having a parent younger than 21 years, residing at present address for less than 1 year and living in a household with more than five occupants as significant risk factors. Experience of a recent stressful event in the family barely failed to reach the level of significance. Logistic regression analysis confirmed that Indian race, having a parent younger than 21 years and residing less than 1 year at the present address were independent predictors of acute poisoning. Positive interactions were noted between Indian race and duration of residence and between parental age and duration of residence. Knowledge of risk factors and their interactions may be useful in planning preventive measures against childhood poisoning.

The STERIS SYSTEM 1 Processor and the STERIS 20 Sterilant Concentrate have met or exceed the rigorous requirements of the Environmental Protection Agency and Food and Drug Administration for making claims related to sterilization. With this system, medical devices including endoscopes are sterilized and ready for use in less than 30 min.

Abstract:Antimicrobial resistance (AMR) poses a global health threat to aquatic environments and its propagation is a hot topic. Therefore, deactivating antibiotic-resistant bacteria (ARB) and removing antibiotic resistance genes (ARGs) from water is crucial for controlling AMR transmission. Peracetic acid (PAA), which is known for its potent oxidizing properties and limited by-product formation, is emerging as a favorable disinfectant for water treatment. In this study, we aimed to assess the efficacy of pre-exposure to PAA followed by UV treatment (PAA-UV/PAA) compared with the simultaneous application of UV and PAA (UV/PAA). The focus was on deactivating vancomycin-resistant Enterococcus faecalis (VREfs), a typical ARB in water. Pre-exposure to PAA significantly enhanced the efficacy of subsequent UV/PAA treatment. At a UV fluence of 7.2 mJ cm-2, the PAA-UV/PAA method achieved a 6.21 log reduction in VREfs, surpassing the 1.29 log reduction observed with UV/PAA. Moreover, compared to UV/PAA, PAA-UV/PAA showed increased efficacy with longer pre-exposure times and higher PAA concentrations, maintaining superior performance across a broad pH range and in the presence of humic acid. Flow cytometry analysis indicated minimal cellular membrane damage using both methods. However, the assessments of superoxide dismutase (SOD) activity and adenosine triphosphate content revealed that PAA-UV/PAA induced greater oxidative stress under similar UV irradiation conditions, leading to slower bacterial regrowth. Specifically, SOD activity in PAA-UV/PAA surged to 3.06 times its baseline, exceeding the 1.73-fold increase under UV/PAA conditions. Additionally, pre-exposure to PAA amplified ARGs degradation and reduced resistance gene leakage, effectively mitigating the spread of AMR. Pre-exposure to 200 μM PAA for 10 and 20 min enhanced vanB gene removal efficiency by 0.14 log and 1.29 log, respectively. Our study provides a feasible approach for optimizing UV/PAA disinfection for efficient removal of ARB and ARGs.

Abstract:Poorly-treated wastewater harbors harmful microorganisms, posing risks to both the environment and public health. To mitigate this, it is essential to implement robust disinfection techniques in wastewater treatment plants. The use of performic acid (PFA) oxidation has emerged as a promising alternative, due to its powerful disinfection properties and minimal environmental footprint. While PFA has been used to inactivate certain microbial indicators, its potential to tackle the entire microbial community in effluents, particularly resistant bacterial strains, remains largely unexplored. The present study evaluates the efficacy of PFA disinfection on the microbial communities of a WWTP effluent, through microbial resistance mechanisms due to their membrane structure. The effluent microbiome was quantified and identified. The results showed that the number of damaged cells increases with CT, reaching a maximum for CT = 240 mg/L•min and plateauing around 60 mg/L•min, highlighting the optimal conditions for PFA-disinfection against microbial viability. A low PFA level with a 10-min contact time significantly affected the microbial composition. It is worth noting the sensitivity of several bacterial genera such as Flavobacterium, Pedobacter, Massilia, Exiguobacterium, and Sphingorhabdus to PFA, while others, Acinetobacter, Leucobacter, Thiothrix, Paracoccus, and Cloacibacterium, showed resistance. The results detail the resistance and sensitivity of bacterial groups to PFA, correlated with their Gram-positive or Gram-negative membrane structure. These results underline PFA effectiveness in reducing microbial levels and remodeling bacterial composition, even with minimal concentrations and short contact times, demonstrating its suitability for widespread application in WWTPs.

Abstract:Peracetic acid is a strong disinfectant with a wide spectrum of antimicrobial activity. Due to its bactericidal, virucidal, fungicidal, and sporicidal effectiveness as demonstrated in various industries, the use of peracetic acid as a disinfectant for wastewater effluents has been drawing more attention in recent years. The desirable attributes of peracetic acid for wastewater disinfection are the ease of implementing treatment (without the need for expensive capital investment), broad spectrum of activity even in the presence of heterogeneous organic matter, absence of persistent toxic or mutagenic residuals or by-products, no quenching requirement (i.e., no dechlorination), small dependence on pH, short contact time, and effectiveness for primary and secondary effluents. Major disadvantages associated with peracetic acid disinfection are the increases of organic content in the effluent due to acetic acid (AA) and thus in the potential microbial regrowth (acetic acid is already present in the mixture and is also formed after peracetic acid decomposition). Another drawback to the use of peracetic acid is its high cost, which is partly due to limited production capacity worldwide. However, if the demand for peracetic acid increases, especially from the wastewater industry, the future mass production capacity might also be increased, thus lowering the cost. In such a case, in addition to having environmental advantages, peracetic acid may also become cost-competitive with chlorine.

Abstract:Wastewater disinfection is a key treatment for the protection of the environment and public health from pathogenic infections. Peracetic acid (PAA) is an organic peroxyacid with a high oxidation potential that can be an alternative to conventional disinfectants. The aim of this pilot-scale study was to assess six disinfection conditions using PAA as a disinfectant agent of wastewater. To this end, a 6-month study was performed, and six disinfection conditions were studied: 15 mg/L of PAA with 5, 10, and 15 min of contact time; 10 mg/L of PAA with 10 and 15 min of contact time; and 5 mg/L of PAA with 10 min of contact time. After the disinfection, an increase in the 5-day biochemical oxygen demand (BOD5) and the chemical oxygen demand (COD) was observed; however, this was not proportional to the PAA dose because the higher doses tested did not result in a higher increment. In addition, PAA resulted in a slight decrease in pH. Nevertheless, the PAA impact on pH was not dependent on the PAA dose or the contact time. The PAA disinfection performance showed that doses of 10 and 15 mg/L achieved Escherichia coli log reduction higher than 2 logs, and even higher than 3 logs when 15 mg/L were tested with 10 and 15 min of contact time. The results showed also that the contact time was not a limiting factor for the PAA performance, at least for the contact times used in this study. This pilot-scale study proved that PAA can be an alternative disinfectant for wastewater.

Abstract:PURPOSE: Dental prosthetic and orthodontic appliances are transported from the clinic to the laboratory for additions and repairs. These appliances, containing microbes from the oral flora, are a high risk for cross-contamination. The aim of this study was to evaluate the effect of chemical and ultrasound disinfection against two in vitro biofilms and an in vivo formed biofilm grown on unprepared and polished polymethyl methacrylate (PMMA) surfaces.MATERIALS AND METHODS: Rough and polished self-curing PMMA surfaces were infected with strains of both Candida albicans and Streptococcus oralis. After incubation, the samples were treated with different disinfection methods, including ultrasound treatment for both 15 and 30 seconds, and immersion in glutaraldehyde and alcohol-based chemical disinfectants (MD520 and Minuten, respectively). The disinfecting efficacy was assessed by colony forming units (CFU) analysis and by scanning electron microscopy (SEM). Furthermore the adequacy of bacterial elimination of application of 30-second ultrasound and MD520 was assessed on PMMA retrieved from ten volunteers by CFU analyses. ANOVA with p = 0.05 followed by the Tukey post hoc test and the Student t-test was used to analyze the data.RESULTS: The ultrasound treatment for 30 seconds, MD520, and Minuten were the most effective disinfectant methods as they reduced the microbial counts compared to the control (p < 0.05) as shown in the in vitro analyses. S. oralis adhered more to rough acrylic resin surfaces (p < 0.05). Ultrasound treatment was the most effective way to reduce microbial counts on PMMA exposed to oral flora (p = 0.043).CONCLUSION: Ultrasound treatment for 30 seconds was effective against C. albicans, S. oralis, and the oral flora as shown by testing microbial growth on agar plates and SEM.
Antimicrobial efficacy, optical properties and flexural strength following antimicrobial photodynamic therapy over vacuum-formed orthodontic retainers

Abstract:Background and aims: Vacuum-formed retainers (VFRs) are widely used in dental practices because of their superior esthetics. However, simultaneous maintenance of their hygiene, mechanical and optical properties is challenging. This study aimed to evaluate and compare the disinfection efficacy of antimicrobial photodynamic therapy (aPDT), chlorhexidine (CHX), sodium hypochlorite (NaOCl) and water over VFRs specimens, along with their effect on flexural strength and color stability. Materials and methods: Fabricated VFRs of thicknesses 1 mm and 2 mm were contaminated with S. mutans and C. albicans. Brain Heart Infusion agar was used to seed Streptococcus Mutans whereas Candida Albicans was seeded in Sabourand Dextrose Agar. The contaminated retainers were divided into four groups and decontaminated using four different treatment protocols: aPDT, CHX, NaOCl and water, with 10 specimens per group. Microbial viability following decontamination, was assayed using MTT. To assess the color stability, pre and post decontamination color difference of 10 specimens was recorded using a spectrophotometer by blinding technique. The flexural strength of 10 specimens was measured using a universal testing machine by a standardized 3-point flexural strength (FS) test. Result: aPDT showed the highest reduction in the viability of S. mutans (28%) and C. albicans (20%) species in 1 mm thickness group while the second highest reduction following disinfection with CHX (40%) in followed by NaOCl disinfection (50%). On comparison between 1 mm and 2 mm, a statistically significant difference was noted for S. mutans in aPDT (p = 0.033) and NaOCl (p = 0.028) groups, and for C. albicans, a significant difference was noted within NaOCl (p = 0.001) and CHX (p = 0.043) groups. For FS, a statistically significant difference (p < 0.05) was observed between aPDT and the rest of the groups, whereas there was no statistically significant difference in the flexural strength between the CHX, NaOCl and water groups (p > 0.05) in both 1 mm and 2 mm specimens. Between 1 mm and 2 mm specimens, no statistically significant difference in the mean color difference was noted after any disinfection protocol (p > 0.05). Conclusion: Antimicrobial photodynamic therapy showed a remarkable reduction in candidal and bacterial viability over VFRs. aPDT treated VFR specimens showed increased flexural strength and a higher color difference post treatment compared to other reagents.

Abstract:AIMS: Evaluation of the efficiency of peracetic acid in the disinfection of wastewater in a large treatment plant.METHODS AND RESULTS: Over a period of 18 months 30 sample collections were made, each consisting of three samples taken from: raw incoming sewage, secondary effluent (after 10-12 h) and secondary effluent disinfected with 1.5-2 mg l(-1) of peracetic acid (contact time: 20 min). Total coliforms and Escherichia coli declined from 10(7) MPN 100 ml(-1) in the raw sewage to 10(2) in the disinfected effluent and the enterococci fell from 10(6) MPN 100 ml(-1) to 702 MPN 100 ml(-1). The reduction of bacteria increased with the rise in temperature and decreased with the rise in BOD5.CONCLUSIONS: Disinfection with peracetic acid reduced levels of faecal contamination by 97%, thus attaining the limit recommended by current Italian law (Escherichia coli <or= 5000 MPN 100 ml(-1)) for discharge into surface waters.SIGNIFICANCE AND IMPACT OF THE STUDY: The process of disinfection with peracetic acid is easier to manage than other more common methods and the tests performed confirm that from the bacteriological point of view good results can be obtained for urban effluents.

Abstract:The ever-increasing need for access to safe water has meant that alternative water sources and innovative water reclamation approaches are often required to meet the global water demand. As a result, many wastewater treatment facilities have faced regulatory pressure to seek alternative disinfection methods that ensure public health safety, while adhering to regulations that set limits on carcinogenic disinfection by-products (DBPs). Peracetic acid (PAA) is an emerging wastewater disinfectant in the United States that has been widely used in other industries such as food sanitization and does not produce carcinogenic DBPs. However, several factors such as transport, storage, and physical and chemical effects have stymied its widespread use in wastewater markets. Therefore, the purpose of this study was to examine the antimicrobial efficacy of an on-site generated PAA compared against a commercially available PAA. Antimicrobial efficacy was assessed using standard fecal contamination indicators (i.e., total coliforms and Escherichia coli) in six urban wastewater treatment facilities ranging in size and treatment processes. Overall, few statistical differences were found between the antimicrobial efficacies of on-site generated PAA and commercially available PAA; however, before becoming more widely utilized, the on-site PAA should be tested against emerging fecal contamination indicators (e.g., human norovirus and enterovirus) and be assessed in terms of economic and sustainability impacts. PRACTITIONER POINTS: Alternative Ct approaches should be considered when using disinfectants like PAA. On-site generated PAA can achieve the same level of disinfection as commercial PAA. On-site generation of PAA may help further its use as a wastewater disinfectant.

Evaluation of home disinfection protocols for acrylic baseplates of removable orthodontic appliances: A randomized clinical investigation

Abstract:INTRODUCTION: In this randomized clinical trial, we investigated, using the microbial culture technique and scanning electron microscopy, the contamination of acrylic baseplates of removable orthodontic appliances by mutans streptococci (MS) and evaluated the efficacy of different home disinfection protocols with a 0.12% chlorhexidine gluconate spray (Periogard, Colgate-Palmolive, São Bernardo do Campo, Brazil).METHODS: Fifteen dental students were randomly enrolled in a 3-stage changeover system with a 1-week interval between each stage. The acrylic baseplates were worn full time except at meals to simulate the routine use of removable appliances under clinical conditions. Three 1-week home disinfection protocols were tested in all stages by a different group of students: protocol I, toothbrushing + baseplate brushing + sterile tap water spraying once a day; protocol II, toothbrushing + baseplate brushing + Periogard spraying on the seventh day after appliance placement; and protocol III, toothbrushing + baseplate brushing + Periogard spraying on the fourth and seventh days after appliance placement. After the first week, the volunteers received new baseplates, toothbrushes, and dentifrices, and the regimens were repeated 2 more times. At the end of each week, the baseplates had a randomized disinfection protocol and were sent for microbiologic analysis. A scanning electron microscope was used to examine 3 acrylic baseplates representing each home protocol. The Friedman test (α = 0.05) compared the home protocols for the formation of MS colonies or biofilms on the acrylic surfaces.RESULTS: MS colonies or biofilms were found on all acrylic baseplates after protocol I. Protocols II and III reduced significantly (P <0.05) the number of MS colonies and biofilms on the acrylic surfaces. No significant difference (P >0.05) was observed between protocols II and III. The scanning electron microscope analysis confirmed the results of the microbiologic cultures.CONCLUSIONS: Disinfection of baseplates of removable orthodontic appliances by using 0.12% chlorhexidine spray once or twice a week reduced the contamination by MS on the acrylic surface in vivo.

Abstract:Objectives: To subjectively assess radiographs obtained with photostimulable phosphor (PSP) plates exposed to clinical levels of ambient light prior to read-out to potentially set a safe limit for acceptable image quality. Methods and materials: Six dental regions of a dry human skull were X-rayed using PSP plates from VistaScan and Express under four exposure times: 0.1, 0.2, 0.32, and 0.4 s. Before read-out, the PSP plates were exposed to ambient light for 0, 5, 10, 30, 60, and 90 s. Six observers were asked to classify the 288 resulting radiographs as acceptable or unacceptable based on the identification of anatomical structures and global image quality. The number of answers classifying radiographs as unacceptable was used to calculate a rejection rate; a pairwise comparison for better image quality was further conducted among radiographs considered acceptable. Reproducibility was tested by having 25% of all experimental groups reassessed. Results: Intra- and inter observer agreement ranged from 0.87 to 1.00 and from 0.81 to 0.92, respectively. Exposure of PSP plates to ambient light increased rejection rates mostly as of 10 s. In the pairwise comparison, subtle differences were observed between radiographs obtained with PSP plates not exposed and those exposed to ambient light for 5 s. Conclusions: Ambient light exposure of PSP plates impairs the image quality of radiographs. A safe limit of ambient light exposure of 5 s for VistaScan and Express should be considered. Ambient light exposure of PSP plates within safe limits can avoid retakes and reduce unnecessary patient exposure to X-rays.

Abstract:The species of the Bacillus cereus group have the ability to adhere to and form biofilms on solid surfaces, including stainless steel, a material widely used in food industries. Biofilms allow for recontamination during food processing, and the "clean-in-place" (CIP) system is largely used by industries to control them. This study thus proposes to evaluate the efficacy of peracetic acid and sodium hypochlorite against biofilms induced on stainless-steel surfaces. The SAMN07414939 isolate (BioProject PRJNA390851), a recognized biofilm producer, was selected for biofilm induction on AISI 304 stainless steel. Biofilm induction was performed and classified into three categories: TCP (Tindalized, Contaminated, and Pasteurized milk), TCS (Tindalized milk Contaminated with Spores), and TCV (Tindalized milk Contaminated with Vegetative cells). Subsequently, the coupons were sanitized simulating a CIP procedure, on a pilot scale, using alkaline and acid solutions followed by disinfectants (peracetic acid and sodium hypochlorite). Microorganism adhesion on the surfaces reached 6.3 × 105 to 3.1 × 107 CFU/cm-2. Results did not show significant differences (p > 0.05) for surface adhesion between the three tested categories (TCP, TCS, and TCV) or (p > 0.05) between the two disinfectants (peracetic acid and sodium hypochlorite). Microbial populations adhered to the stainless-steel coupons are equally reduced after treatment with peracetic acid and sodium hypochlorite, with no differences in the control of B. cereus s.s. biofilms on AISI 304 stainless-steel surfaces.
Efficiency of Pathogenic Bacteria Inactivation by Non-Thermal Plasma Activated Water

Abstract:Non-thermal plasma is a promising bactericidal method due to the existence of ultraviolet, high energy electrons, reactive oxygen species, and reactive nitrogen species in plasma. Recently, the plasma activated water (PAW) has attracted more and more attention. In our study, PAW was applied to four different bacteria: Streptococcus mutans (S. mutans), Staphylococcus aureus (S. aureus), Escherichia coli (E. coli) and Candida albicans (C. albicans). The inactivation effect of PAW was observed through colony counting method and fluorescence staining. The reactive oxygen species in the PAW was investigated by optic emission spectrometer and Hydrogen Peroxide Test Kit. Effective inactivation under PAW treatment of S. mutans, S. aureus, E. coli was achieved within 20 min, while weak inactivation effect to C. albicans. In addition, reactive oxygen species such as atomic oxygen and hydrogen peroxide were detected in the PAW. Our study provides an experimental basis in the prevention of oral diseases and sterilization of medical tubular equipment by PAW.

Abstract:Performic acid (PFA) has been increasingly used in wastewater disinfection due to its strong oxidizing ability and few disinfection byproducts. However, its disinfection pathways and mechanisms towards pathogenic bacteria disinfection are poorly understood. In this study, E. coli , S. aureus , and B. subtilis were inactivated using sodium hypochlorite (NaClO), PFA, and peracetic acid (PAA) in simulated turbid water and municipal secondary effluent. Cell culture-based plate counting showed that E. coli and S. aureus were extremely susceptible to NaClO and PFA and achieved a 4-log inactivation at CT s ≤ 1 mg/L·min with an initial disinfectant concentration of 0.3 mg/L. B. subtilis was much more resistant. At the initial disinfectant dose of 7.5 mg/L, PFA required CT s of 3–13 mg/L·min to achieve a 4-log inactivation. Turbidity negatively affected the disinfection. In the secondary effluent, the CT s required for PFA to achieve a 4-log inactivation of E. coli and B. subtilis were 6–12 times higher than those required in simulated turbid water, and a 4-log inactivation of S. aureus could not be achieved. PAA showed a much weaker disinfection ability than the other two disinfectants. The reaction pathways of E. coli inactivation by PFA included both direct and indirect reactions, in which the PFA molecule accounted for 73 %, and ·OH and peroxide radicals accounted for 20 % and 6 %, respectively. During PFA disinfection, E. coli cells were severely disintegrated, while the S. aureus cell exteriors remained mostly intact. B. subtilis was the least affected. Compared with cell culture-based analysis, the inactivation detected by flow cytometry was significantly lower. Viable but non-culturable bacteria after disinfection were believed to be primarily responsible for this inconsistency. This study suggested that PFA was able to control regular bacteria in wastewater, but it should be used with caution when treating recalcitrant pathogens.
