Abstract:The osteoinductive properties of beta tricalcium phosphate(β-TCP)/collagen scaffold loaded with morphogenetic protein 2(BMP-2) plasmid(100μg) were evaluated in vivo.BMP-2 gene-modified β-TCP/collagen scaffolds(n=8) were used to repair critically-sized defects(5mm in diameter) in the calvaria of rats,and pure β-TCP/collagen scaffolds(n=8) and empty defects(n=8) served as controls.The quality of newly regenerated bone was assessed by histological/histomorphometric analysis.The histological analysis revealed that porous β-TCP/collagen scaffold degraded completely at 12 weeks.Defects in BMP-2 DNA group healed completely whereas a significantly less bone ingrowth was observed in pure scaffolds by 12 weeks(p0.05).Immunohistochemistry analysis indicated that the expression of BMP-2 was significantly stronger in BMP-2DNA group than the controls(p0.05).In addition,compared to pure scaffold groups or blank groups,rats in BMP-2 gene-modified scaffold group showed significantly more bone generation by histomorphometric analysis(P 0.05).We conclude that porous β-TCP/collagen scaffold loaded with BMP-2 DNA is an appropriate candidate for bone engineering.

Abstract:The bone morphogenetic protein (BMP) signaling pathway is highly conserved across many species, and its importance for the patterning of the skeletal system has been demonstrated. A disrupted BMP signaling pathway results in severe skeletal defects. Murine calvaria has been identified to have dual-tissue lineages, namely, the cranial neural-crest cells and the paraxial mesoderm. Modulations of the BMP signaling pathway have been demonstrated to be significant in determining calvarial osteogenic potentials and ossification in vitro and in vivo. More importantly, the BMP signaling pathway plays a role in the maintenance of the homeostasis of the calvarial stem cells, indicating a potential clinic significance in calvarial bone and in expediting regeneration. Following the inherent evidence of BMP signaling in craniofacial biology, we summarize recent discoveries relating to BMP signaling in the development of calvarial structures, functions of the suture stem cells and their niche and regeneration. This review will not only provide a better understanding of BMP signaling in cranial biology, but also exhibit the molecular targets of BMP signaling that possess clinical potential for tissue regeneration.

Abstract:Transforming growth factor-beta (TGF-beta) and bone morphogenic protein (BMP) signaling has fundamental roles in both embryonic skeletal development and postnatal bone homeostasis. TGF-beta s and BMPs, acting on a tetrameric receptor complex, transduce signals to both the canonical Smad-dependent signaling pathway (that is, TGF-beta/BMP ligands, receptors, and Smads) and the non-canonical-Smad-independent signaling pathway (that is, p38 mitogen-activated protein kinase/p38 MAPK) to regulate mesenchymal stem cell differentiation during skeletal development, bone formation and bone homeostasis. Both the Smad and p38 MAPK signaling pathways converge at transcription factors, for example, Runx2 to promote osteoblast differentiation and chondrocyte differentiation from mesenchymal precursor cells. TGF-beta and BMP signaling is controlled by multiple factors, including the ubiquitin-proteasome system, epigenetic factors, and microRNA. Dysregulated TGF-beta and BMP signaling result in a number of bone disorders in humans. Knockout or mutation of TGF-beta and BMP signaling-related genes in mice leads to bone abnormalities of varying severity, which enable a better understanding of TGF-beta/BMP signaling in bone and the signaling networks underlying osteoblast differentiation and bone formation. There is also crosstalk between TGF-beta/BMP signaling and several critical cytokines' signaling pathways (for example, Wnt, Hedgehog, Notch, PTHrP, and FGF) to coordinate osteogenesis, skeletal development, and bone homeostasis. This review summarizes the recent advances in our understanding of TGF-beta/BMP signaling in osteoblast differentiation, chondrocyte differentiation, skeletal development, cartilage formation, bone formation, bone homeostasis, and related human bone diseases caused by the disruption of TGF-beta/BMP signaling.
SMAD EXPRESSION IN FRACTURE HEALING

Abstract:Fracture healing involves many local and systemic regulatory factors. Progress in identifying signaling events downstream has been made with the discovery of a novel family of proteins, the Smad, as TGF-s/activins/BMPs signal transducers. Smads are the vertebrate homologs of Mad (Mothers against decapentaplegic) gene from Drosophila and Sma genes from Caenorhabditis elegans. Smad-1, -2, -3, -5, -8 and -9 belong to the receptor-regulated class (R-Smad) which are activated by the TGF-s type I and II receptors, forming heteromers with the common-mediator class (Co-Smad): Smad-4. Smad-6 and -7 (Anti-Smad) perform a negative regulatory or balancing role. Smad-2 and -3 regulate TGF-s/activin effects, whilst Smad-1 and -5 work with BMPs. This study investigated the expression and localization of Smad proteins (Smad 1–6) and BMP-4 and -7 during fracture healing. Eighteen 3-month old female CD-COB rats were used. A standard closed fracture was made in the mid-shaft of right femur using a 3-point bending device. The left limb served as the non-fracture control. The rats were divided into 3 groups (6 per group) and sacrificed at day 3, 10 and 28 after fracture. The femurs were harvested, fixed in buffered formalin for 48 hours and decalcified with 10% formic acid-formalin solution. The decalcified tissues were embedded in paraffin and 5μm sections were cut onto silane-coated slides. Representative slides from each block were stained with routine haematoxylin and eosin (H&E). Sections were cut for immunohistochemistry for protein marker expression by a standard procedure for Smads and BMP 4 and 7. Sections were viewed and analysed by colour video image analysis using a 40x objective, a 10x eyepiece, and a fixed frame of 128 × 128 pixels (49152.0 μm 2 ). Ten fields per slide were examined. Smad proteins (Smads 1, 4, and 6) were expressed during the early stages (day 3) of fracture healing by bone marrow stromal cells, osteoblasts, fibroblasts and chondrocytes located in the intramembranous and endochondral ossification regions around the fracture site. Differential expressions of individual Smads, particularly Smad 1 and Smad 6, at different time-points (Smad-1 was higher than Smad-6 at day 3, whilst Smad-6 was much higher than Smad-1 at day 10) suggest that Smad proteins are not simply BMP signal transducers. Smads may also be responsible for up- and/or down-regulation of transcriptional events during the intramembranous and endochondral ossification. Smad-4, a Co-SMAD, expression newly formed bone and cartilage suggests an additional function beyond the signal transduction in rat fracture healing. BMP-4 and BMP-7 were highly expressed at day 3 and 10. BMP-7 expression was greater than BMP-4 at day 3 but switched by day 10 (BMP-4 > BMP-7). Smads represent a new level where specific therapeutic strategies can be targeted considering the interactions with a number of BMPs.
Epigenetically Modified Bone Marrow Stromal Cells in Silk Scaffolds Promote Craniofacial Bone Repair and Wound Healing

Abstract:Epigenetic regulation of gene expression is a central mechanism that governs cell stemness, determination, commitment, and differentiation. It has been recently found that PHF8, a major H4K20/H3K9 demethylase, plays a critical role in craniofacial and bone development. In this study, we hypothesize that PHF8 promotes osteoblastogenesis by epigenetically regulating the expression of a nuclear matrix protein, special AT-rich sequence-binding protein 2 (SATB2) that plays pivotal roles in skeletal patterning and osteoblast differentiation. Our results showed that expression levels of PHF8 and SATB2 in preosteoblasts and bone marrow stromal cells (BMSCs) increased simultaneously during osteogenic induction. Overexpressing PHF8 in these cells upregulated the expression of SATB2, Runx2, osterix, and bone matrix proteins. Conversely, knockdown of PHF8 reduced the expression of these genes. Furthermore, ChIP assays confirmed that PHF8 specifically bound to the transcription start site (TSS) of the SATB2 promoter, and the expression of H3K9me1 at the TSS region of SATB2 decreased in PHF8 overexpressed group. Implantation of the BMSCs overexpressing PHF8 with silk protein scaffolds promoted bone regeneration in critical-sized defects in mouse calvaria. Taken together, our results demonstrated that PHF8 epigenetically modulates SATB2 activity, triggering BMSCs osteogenic differentiation and facilitating bone formation and regeneration in biodegradable silk scaffolds.
Local application of rhTGF-beta2 modulates dynamic gene expression in a rat implant model

Abstract:Various anabolic agents, including transforming growth factor-beta (TGF-beta), have been shown to enhance intramembranous bone regeneration and strengthen the mechanical connection between implant and host skeleton, a prerequisite for clinical success with orthopedic and dental implants. Mechanisms underlying these observations at the level of the gene have received little attention. A rat model was used to examine levels of gene transcription for 21 "osteogenic" genes by real-time polymerase chain reaction at days 1, 3, 5, 7, 10, 14, and 28 in a control group and a group in which the implant was treated with 1 microg recombinant human TGF-beta2 (n = 42, equally divided among the 2 groups and 7 time points). Genes were chosen to represent three functional categories: (1) growth factors, their receptors and antagonists; (2) bone differentiation markers; and (3) inflammation markers. Examination of the transcription profiles showed that nine genes had up-regulated or down-regulated expression levels without a change in timing and 12 genes had accelerated or delayed expression profiles with or without a concomitant change in maximal or minimal expression. The earliest changes (days 1-3) involved accelerated expression profiles for IGF-1R and VEGF and up-regulation of TGF-beta2, TbetaRI, BMP-2, BMP-7, and Cbfa1. Furthermore, principal components analyses showed that some subsets of genes were co-expressed in both groups, although the temporal relationship of these subsets was altered following growth factor treatment. Thus, in addition to changes in individual transcription profiles, the regulatory connections between sets of co-expressed genes may also be affected by exogenously delivered anabolic agents during bone regeneration.
Gene expression of four adhesive proteins in the early healing of bone defect and bone-implant interface

Abstract:The objective of this study was to evaluate the gene expression of four bone-related adhesive proteins during the early healing of bone defect and bone-implant interface in animal experiments. T-shaped hollow pure titanium implants with dual acid-etched surfaces were placed into femurs of 17 Sprague-Dawley rats, and bone defects with the same size were made in the same site in 15 rats. Newly formed bone was harvested at 5 days, 8 days and 16 days respectively. The gene expression of fibronectin (FN), collagen I (COL I), bone sialoprotein II (BSP II) and osteopontin (OPN) in non-implant and bone-implant defects were examined using semi-quantity reverse transcription-polymerase chain reaction. The gene expression of OPN in the non-implant defect was slightly higher than that in the bone-implant interface. At 8 days postoperation, FN, COL I and BSP II expression were significantly up-regulated in the bone-implant group. All four proteins peaked at 8 days. The results indicate that the gene expression of the four adhesive proteins is different between bone defect and bone-implant interface. Intracellular synthesis of FN, COL I and BSP II was accelerated in the early healing stages of the bone-implant interface.

Abstract:The periosteum plays a critical role in bone repair and is significantly influenced by the surrounding immune microenvironment. In this study, we employed 10× single-cell RNA sequencing to create a detailed cellular atlas of the swine cranial periosteum, highlighting the cellular dynamics and interactions essential for cranial bone injury repair. We noted that such injuries lead to an increase in M2 macrophages, which are key in modulating the periosteum's immune response and driving the bone regeneration process. These macrophages actively recruit periosteal stromal cells (PSCs) by secreting Neuregulin 1 (NRG1), a crucial factor in initiating bone regeneration. This recruitment process emphasizes the critical role of PSCs in effective bone repair, positioning them as primary targets for therapeutic interventions. Our results indicate that enhancing the interaction between M2 macrophages and PSCs could significantly improve the outcomes of treatments aimed at cranial bone repair and regeneration.

Abstract:The immune response of the host towards foreign body, especially in the form of giant cell reaction against the implanted materials, is a vital factor for determining the repair outcome of bone defect. Biomaterials with good immunomodulatory capacity and suitable degradation may promote the regeneration of bone defect and osteointegration with the host. Small intestine submucosa (SIS), a decellularized material, has been shown to possess a property for inducing the M2 phenotype of macrophages. Our findings suggested that the SIS hydrogel could induce the macrophages to polarize into mixed M1/M2 phenotypes, and such phenotypes have enhanced the migration and tube formation of angiogenesis-associated cells in vitro, whilst having a mild effect on osteoclastogenesis and osteogenesis. Experiment with a rat model for critical-sized bone defect regeneration further demonstrated that the composite BMP-2@SIS hydrogel could sequentially activate M1 and M2 macrophages and promote early-stage angiogenesis, which altogether expedited final bone defect regeneration.
Localization of Smads, the TGF-beta family intracellular signaling components during endochondral ossification.

Abstract:Members of the transforming growth factor-beta (TGF-beta) family transduce signals from the cell membrane to the nucleus via specific type I and type II receptors and Smad proteins. Smad1 and Smad5 mediate intracellular signaling of bone morphogenetic protein (BMP), whereas Smad2 and Smad3 transduce TGF-beta signaling. Smad4 is a common mediator required for both pathways. Smad6 and Smad7 inhibit signaling by members of the TGF-beta superfamily, Here, we examined the expression of Smad1 to Smad7 proteins during endochondral ossification of epiphyseal plate of growing rats using immunohistochemical techniques. The expression of Smad proteins was correlated with the expression of TGF-beta 1 and its receptors, and BMP-2/4 and BMP receptors, The results show that TGF-beta 1 and BMP-2/4 were actively expressed in chondrocytes that are undergoing proliferation and maturation, which overlaps with expression of their corresponding type I and type II receptors, The Smads, however, exhibited a distinct expression pattern, respectively. For example, Smad1 and Smad5 mere highly expressed in proliferating chondrocytes and in those chondrocytes that are undergoing maturation. The TGF-beta/activin-restricted Smads were also expressed in a nearly complementary fashion; Smad2 was strongly expressed in proliferating chondrocytes, whereas Smad3 was strongly observed in maturing chondrocytes. Smad3 was broadly expressed in all zones of epiphyseal plate. Inhibitory Smads, Smad6 and Smad7, were strongly expressed in the zone of cartilage that contained mature chondrocytes, Our findings show a colocalization of the pathway-restricted and inhibitory Smads with activating Ligands or ligands whose action they antagonize and their receptors in various zones of epiphyseal growth plate, suggesting that TGF-beta superfamily Smad signaling pathways plays a morphogenic role during endochondral bone formation.

Abstract:BackgroundThe use of the Masquelet technique in the repair of large bone defects has gained increased acceptance in recent years. The core of this technique is the induction of granulation tissue membrane formation and the implantation of an autologous cancellous bone to reconstruct bone defects in the membrane. In this study, we purpose to explore the structure of induced membrane and the content of growth factors as well to compare between the structure and the effects on osteogenesis of induced membranes and the periosteum in animal models.MethodsBilateral radial bone defects were generated in 32 healthy adult rabbits. The defects were implanted with bone cement. The induced membranes and periosteum were removed after 2, 4, 6, and 8 weeks. Thereafter, hematoxylin-eosin staining (HE) and an enzyme-linked immunosorbent assay (ELISA) were performed to detect vascular endothelial growth factor (VEGF), angiotensin II (ANG-II), bone morphogenetic protein 2 (BMP2), fibroblast growth factor 2 (FGF2), and prostaglandin E2 (PGE2). Proteins isolated from total cell lysates were cultured with mesenchymal stem cells to test the cell proliferation and alkaline phosphatase activity using epimysium as a control.ResultsThe induced membrane and periosteum exhibited similar structures and growth factor levels after 4 and 6 weeks. The highest concentration of BMP-2 and VEGF in the induced membranes occurred in week 6, and FGF-2 and ANG-II concentrations peaked in week 4. The thickness and vascular density of induced membranes gradually decreased with time.ConclusionInduced membrane matured between the 4th and the 6th week and secreted growth factors to promote osteogenesis. The matured induced membrane and periosteum had similar structures and abilities to promote the osteogenesis of mesenchymal stem cells. However, the induced membrane was thicker than the periosteum.

Abstract:Myeloid cells are pivotal in the inflammatory and remodeling phases of fracture repair. Here, we investigate the effect of periostin expressed by myeloid cells on bone regeneration in a monocortical tibial defect (MTD) model. In this study, we show that periostin is expressed by periosteal myeloid cells, primarily the M2 macrophages during bone regeneration. Knockout of periostin in myeloid cells reduces cortical bone thickness, disrupts trabecular bone connectivity, impairs repair impairment, and hinders M2 macrophage polarization. Mechanical stimulation is a regulator of periostin in macrophages. By activating transforming growth factor-β (TGF-β), it increases periostin expression in macrophages and induces M2 polarization. This mechanosensitive effect also reverses the delayed bone repair induced by periostin deficiency in myeloid cells by strengthening the angiogenesis-osteogenesis coupling. In addition, transplantation of mechanically conditioned macrophages into the periosteum over a bone defect results in substantially enhanced repair, confirming the critical role of macrophage-secreted periostin in bone repair. In summary, our findings suggest that mechanical stimulation regulates periostin expression and promotes M2 macrophage polarization, highlighting the potential of mechanically conditioned macrophages as a therapeutic strategy for enhancing bone repair.

The mechanism research of non-Smad dependent TAK1 signaling pathway in the treatment of bone defects by recombination BMP-2-loaded hollow hydroxyapatite microspheres/chitosan composite

Abstract:<h3 class="Heading">Aims</h3><p class="Para">The present study aimed to evaluate whether the non-Smad dependent TAK1 signaling pathway (BMP-2-TAK1-p38-Osx signaling pathway) played an important role in bone repair mediated by hollow hydroxyapatite (HA) microspheres/chitosan (CS) composite.</p><h3 class="Heading">Methods</h3><p class="Para">Firstly, the biological activity of rhBMP-2 released from the complex was investigated. Then, differentiation test of osteoblasts including ALP activity and calcium deposition, X-ray scoring and three-point bending test were performed. Finally, the mRNAs expression of TAK1, p38, Osx and osteogenic markers was tested by reverse transcription-polymerase chain reaction (RT-PCR).</p><h3 class="Heading">Results</h3><p class="Para">RhBMP-2 could be loaded and released from the complex in bioactive form. Additionally, the complex provided a prolonged period of time compared with HA/CS scaffolds. Serum ALP activity was significantly decreased in the TAK1 inhibitor group and p38 inhibitor group. In the X-ray radiography, bone callus was observed in rhBMP-2-loaded hollow HA microspheres/CS composite group. In the three-point bending test, load values in p38 inhibitor group decreased. In the animal model, the mRNA expression of BSP on day 90 was significantly decreased in the p38 inhibitor group and TAK1 inhibitor group. In MC3T3-E1 cells, the mRNA expression of OSX was remarkably up-regulated in both rhBMP-2 group or rhBMP-2-loaded hollow HA microspheres/CS composite group; while the mRNA expression of OSX was significantly down-regulated in TAK1 inhibitor group and p38 inhibitor group.</p><h3 class="Heading">Conclusion</h3><p class="Para">The BMP-2-TAK1-p38-OSX signaling pathway may play an important role in bone formation and repair mediated by rhBMP-2-loaded hollow HA microspheres/CS composite.</p>

Studies on trans-sutural distraction osteogenesis-related genes based on transcriptome sequencing

Abstract:Trans-sutural distraction osteogenesis (TSDO) is an important approach to improve mid-face hypoplasia. In recent years, many studies have been carried out on physical mechanisms of TSDO; however, it’s specific cytological and molecular mechanisms are still unclear. In this study, we performed transcriptome sequencing analysis in Sprague Dawley rats at 1 and 2 weeks after suture osteogenesis and compared RNA expression levels between experimental and control groups. At one week, enrichment pathways were mainly up-regulated in muscle- and bone-related pathways. By contrast, pathways of the immune system showed a state of inhibition and down-regulation, especially for B cells; the main immune pathways showed significant down-regulation. However, two weeks later, the experimental group showed positive up-regulation of the pathways related to DNA synthesis and replication, cell cycle, and chromosome replication. At the same time, the immune pathways that were down-regulated in the first week were up-regulated in the second week. In other words, the up-regulated muscle- and bone-related pathways show opposite trends. The expression of bone- and myogenesis-related transcriptome was up-regulated and the immune-related pathways were down-regulated in the experimental group at 1 week. At 2 weeks, the pathways related to bone- and muscle were down-regulated, while those related to cell cycle regulation and DNA replication were up-regulated. These results suggest that musculoskeletal-related molecules may play an important role during suture osteogenesis at 1 week, and immune regulation may be involved in this process; however, at 2 weeks, molecules related to cell proliferation and replication may be a major role.
Spatiotemporal regulation of the bone immune microenvironment via dam‐like biphasic bionic periosteum for bone regeneration

Abstract:The bone immune microenvironment (BIM) regulates bone regeneration and affects the prognosis of fractures. However, there is currently no effective strategy that can precisely modulate macrophage polarization to improve BIM for bone regeneration. Herein, a hybridized biphasic bionic periosteum, inspired by the BIM and functional structure of the natural periosteum, was presented. The gel phase was composed of genipin‐crosslinked carboxymethyl chitosan and collagen self‐assembled hybrid hydrogels, which acted as the "dam" to intercept IL‐4 released during the initial burst from the bionic periosteum fiber phase, thus maintaining the moderate inflammatory response of M1 macrophages for mesenchymal stem cell recruitment and vascular sprouting at the acute fracture. With degradation of the gel phase, released IL‐4 cooperated with collagen to promote the polarization towards M2 macrophages, which reconfigured the local microenvironment by secreting PDGF‐BB and BMP‐2 to improve vascular maturation and osteogenesis twofold. In rat cranial defect models, the controlled regulation of the BIM was validated with the temporal transition of the inflammatory/anti‐inflammatory process to achieve faster and better bone defect repair. This strategy provided a drug delivery system that constructed a coordinated BIM, so as to break through the predicament of the contradiction between immune response and bone tissue regeneration. Abstract:Various studies have been investigated the phenotypic and functional distinctions of craniofacial and long bone cells involved in bone regeneration. However, the process of bone tissue regeneration after bone grafting involves complicated interactions between different cell types at the donor-recipient site. Additionally, differences in alterations of the immune microenvironment at the recipient site remained to be explored. Osteoblasts (OBs) and macrophages (MØ) play essential roles in the bone restoration and regeneration processes in the bone and immune systems, respectively. The modulation of MØ on OBs has been extensively explored in the literature, whereas limited research has been conducted on the influence of OBs on the MØ phenotype and function. In the present study, OBs from the mandible and femur (MOBs and FOBs, respectively) promoted cranial defect regeneration in rats, with better outcomes noted in the MOBs-treated group. After MOBs transplantation, a significant inflammatory response was induced, accompanied by an early increase in IL-10 secretion. And then, there was an upregulation in M2-MØ-related cell markers and inflammatory factor expression. Condition media (CM) of OBs mildly inhibited apoptosis in MØ, enhanced their migration and phagocytic functions, and concurrently increased iNOS and Arg1 expression, with MOB-CM demonstrating more pronounced effects compared to FOB-CM. In conclusion, our investigation showed that MOBs and FOBs have the ability to modulate MØ phenotype and function, with MOBs exhibiting a stronger regulatory potential. These findings provide a new direction for improving therapeutic strategies for bone regeneration in autologous bone grafts from the perspective of the immune microenvironment.

[EXPERIMENTAL STUDY ON OSTEOGENESIS OF SYNOVIUM-DERIVED MESENCHYMAL STEM CELLS IN VITRO AND IN VIVO]

Abstract:Objective: To investigate the osteogenic differentiation potential and the biological features of synovium-derived mesenchymal stem cells (SMSCs) in vitro and to observe the osteogenic capability of the composite scaffolds constructed with SMSCs and hydroxylapatite/chitosan/poly L-latic acid (HA/CS/PLLA) in vivo. Methods: SMSCs were separated and cultured with adherent method and enzymatic digestion method. Specific phenotypes of SMSCs were detected by flow cytometry after purification. Then, SMSCs were identified by oil red O staining, alkaline phosphatase (ALP) staining, and alizarin red staining after adipogenic and osteogenic induction, respectively. In vitro experiments: the expressions of osteogenic related genes [osteocalcin (OCN), collagen type I, ALP, and Runx-2] were detected by real-time fluorescent quantitative PCR at 1, 7, 14, 21, and 28 days after osteogenic induction; ALP activities were also determined by ELISA at 1, 3, 5, 7, 9, and 11 days after osteogenic induction; meanwhile, extracellular matrix calcium mineralization was detected by alizarin red S method at 7, 14, 21, and 28 days after osteogenic induction; the normal SMSCs were harvested as control group. In vivo experiments: Twenty-four Sprague Dawley (SD) rats were randomly divided into experimental group (n = 12) and control group (n = 12). The 3rd passage SMSCs were seeded on HA/CS/PLLA to construct composite scaffolds, after adhesion for 72 hours in vitro, the composite scaffolds were implanted into the right thigh muscle of 12 SD rats as experimental group; HA/CS/PLLA was implanted into the right thigh muscle of the other 12 SD rats as control group. At 4 and 8 weeks after implantation, the scaffolds were harvested for X-ray film and histological examination to observe ectopic bone formation. Results: The positive rates of CD147, CD90, CD105, and CD44 were more than 95%, while the positive rates of CD117, CD34, CD14, and CD45 were less than 10%. Oil red O staining demonstrated red lipid droplets in the cytoplasm, and alizarin red staining showed flaky red calcifications, and cytoplasm was dyed brown by the ALP staining. The mRNA expressions of collagen type I, ALP, and Runx-2 were significantly increased at 7 days after osteogenic induction, and OCN mRNA expression was significantly increased at 14 days after osteogenic induction; ALP activity was significantly higher at 5, 7, 9, 11 days after osteogenic induction in the SMSC-induced group than control group and reached a maximum at 7 days (P < 0.05). Calcium mineralization was significantly enhanced at 14 days after osteogenic induction, and gradually increased with time (P < 0.05); moreover, it was significantly higher in the SMSC-induced group than control group (P < 0.05). X-ray and histological examination demonstrated that the new bone tissues formed in 2 groups, but bone formation content of the experimental group was significantly more than that of the control group at 4 and 8 weeks after implantation (P < 0.05). Conclusion: SMSCs can be induced into osteoblasts both in vitro and in vivo, so SMSCs might be a promising seed cells for bone tissue engineering.

Abstract:Bone morphogenetic proteins play a key role in the regulation of the three major phases of fracture healing; the inflammatory response, the chondrogenic stage and the osteogenic stage. BMP-2 and 4 show a rapid response during the initial inflammatory stage, and BMP-2 has been shown to be essential for fracture healing to occur. BMP-2 and 4 are also present in the chondrogenic stage, although expression levels of BMP-3b (GDF10) and BMP-6 seemingly have specific peaks at day 7, suggesting a crucial role in this process. BMP-3, 4 and 5 are also abundantly present during this stage. In the osteogenic stage studies have shown that BMPs 1, 2, 3, 3b, 4, 5, 6, 7 and 8A have high levels of expression. It is important to note that of these, BMP-3 and 4 have a peak in their expression and BMP-7 and 8A are almost exclusively expressed during day 14 and 21. The expression of the different BMPs in these stages is believed to be orchestrated by autoregulatory activation between the BMPs, negative feed back loops and external antagonists. Alterations of the BMP signaling pathway, either by direct targeting of the BMPs or by their inhibitors has been shown to interfere with embryologic skeletal development and a post natal regenerative repair following skeletal trauma.

Abstract:Osteogenesis and angiogenesis are two integrated components in bone repair and regeneration. A deeper understanding of osteogenesis and angiogenesis has been hampered by technical difficulties of analyzing bone and neovasculature simultaneously in spatiotemporal scales and in 3D formats. To overcome these barriers, a cranial defect window chamber model was established that enabled high-resolution, longitudinal, and real-time tracking of angiogenesis and bone defect healing via multiphoton laser scanning microscopy (MPLSM). By simultaneously probing new bone matrix via second harmonic generation (SHG), neovascular networks via intravenous perfusion of fluorophore, and osteoblast differentiation via 2.3-kb collagen type I promoter-driven GFP (Col2.3GFP), we examined the morphogenetic sequence of cranial bone defect healing and further established the spatiotemporal analyses of osteogenesis and angiogenesis coupling in repair and regeneration. We showed that bone defect closure was initiated in the residual bone around the edge of the defect. The expansion and migration of osteoprogenitors into the bone defect occurred during the first 3 weeks of healing, coupled with vigorous microvessel angiogenesis at the leading edge of the defect. Subsequent bone repair was marked by matrix deposition and active vascular network remodeling within new bone. Implantation of bone marrow stromal cells (BMSCs) isolated from Col2.3GFP mice further showed that donor-dependent bone formation occurred rapidly within the first 3 weeks of implantation, in concert with early angiogenesis. The subsequent bone wound closure was largely host-dependent, associated with localized modest induction of angiogenesis. The establishment of a live imaging platform via cranial window provides a unique tool to understand osteogenesis and angiogenesis in repair and regeneration, enabling further elucidation of the spatiotemporal regulatory mechanisms of osteoprogenitor cell interactions with host bone healing microenvironment.
