Abstract:Molecular data on a limited number of chromosomal loci has shown that the population of Neisseria meningitidis, a deadly human pathogen, is structured in lineages called clonal complexes and is influenced by a significant rate of homologous recombination. Mechanisms resulting in the emergence of these lineages, their persistence in time and the implications for the pathogenicity of the bacterium, are yet incompletely understood. Here we show with whole genome sequences that N. meningitidis is structured in phylogenetic clades which are made up of clonal complexes. Through acquisition of specific genes and via insertions and rearrangements, each clade has acquired and remodeled specific genomic-tracts with a potential impact on the host-microbial interaction We confirm high rates of detectable recombination throughout the whole chromosome, in contrast with the observed population structure. However, gene-conversion events are found to be significantly longer within clades than between clades, suggesting a DNA cleavage mechanism associated with the phylogeny of the species. We identify twenty-two Restriction-Modification systems, probably acquired by horizontal gene transfer from outside of the species/genus, whose presence in the different strains coincides with the phylogenetic clade structure. Results suggest that clades evolve from the acquisition of a specific set of restriction modification systems that generate a differential barrier to DNA exchange. These findings have general implications for the emergence of lineage structure and virulence in recombining bacterial populations, and could provide an evolutionary framework for the population biology of a number of other bacterial species that show contradictory population structure and

Abstract:The aim of this research was to study the effect of plant-growth-promoting bacteria (PGPB) isolated from the drought-tolerant plants camel thorn (Alhagi pseudoalhagi (M.Bieb.) Fisch) and white pigweed (Chenopodium album L.) on wheat (Triticum aestivum L.) plants cv. Lenigradskaya 6, growing under hydroponic conditions and osmotic stress (generated by 12% polyethylene glycol-6000 (PEG)). Based on the assumption that plants create a unique microbiome that helps them overcome various stresses, we hypothesized that bacteria isolated from drought-tolerant plants may assist cultivated wheat plants in coping with drought stress. PGPB were isolated from seeds and leaves of plants and identified as Bacillus spp. (strains Cap 07D, Cap 09D, and App 11D); Paenibacillus sp. (Cap 286); and Arthrobacter sp. (Cap 03D). All bacteria produced different phytohormones such as indole acetic acid (IAA), abscisic acid (ABA), and gibberellic acid (GAS3) and were capable of stimulating wheat growth under normal and osmotic stress conditions. All PGPB reduced the malondialdehyde (MDA) content, increased the total chlorophyll content by increasing chlorophyll a, and modulated wheat hormone homeostasis and CAT and POX activities under osmotic conditions. Selected strains can be promising candidates for the mitigating of the drought stress of wheat plants.

Assessment of Plant Growth Promoting and Abiotic Stress Tolerance Properties of Wheat Endophytic Fungi

Abstract:The aims of the present work were to isolate and characterize fungal endophytic communities associated with healthy wheat ( Triticum aestivum L. ) plants, collected from the North China. Segregated endophytes were screened for their PGP traits, abiotic stresses (heavy metals, salinity, drought, and temperature), and antibiotic sensitivity. A total of 16 endophytic fungi were isolated using the culture-dependent approach from different tissue parts of wheat plants. Based upon their internal transcribed spacer (ITS) rDNA gene sequencing, 15 out of 16 isolates were selected for further analysis. In the contemporary investigation, a number of the tested endophytes exhibited fairly good 1-aminocyclopropane-1-carboxylic acid deaminase (ACCD) (0.03±0.011 to 1.43±0.01 μ mol α -KB mg −1 protein hr −1 ), indole acetic acid (IAA) (1.125±0.04 to36.12±0.004 μ gml −1 ), and phosphate solubilizing index (PSI) (2.08±0.03to5.16±0.36) activities. More than 30% isolates gave positive result for siderophore and ammonia tests, whereas all exhibited catalase activity but only 2 (582PDA1 and 582PDA11) produced hydrogen cyanide. Trichoderma strains showed salt, heavy metals, and drought tolerance at high levels and also exhibited resistance to all the tested antibiotics. Strain 582PDA4 was found to be the most temperature (55°C) tolerant isolate. The findings of this study indicated that the microbial endophytes isolated from wheat plants possessing a crucial function to improve plant growth could be utilized as biofertilizers or bioagents to establish a sustainable crop production system.

Abstract:Water shortage limits agricultural productivity, so strategies to get higher yields in dry agricultural systems is vital to circumvent the effect of climate change and land-shortage. The plant rhizosphere harbors beneficial bacteria able to confer biotic/abiotic tolerance along with a positive impact on plant growth. Herein, three bacterial strains, Proteus mirabilis R2, Pseudomonas balearica RF-2 and Cronobacter sakazakii RF-4 (accessions: LS975374, LS975373, LS975370, respectively) isolated from native desert-weeds were investigated for their response to improve wheat growth under drought stress. The bacteria showed drought tolerance up to 20% polyethylene glycol (PEG; -0.6 MPa), and salt (65-97 g l-1 ), 1-aminocyclopropane-1-carboxylate (ACC)-deaminase activity, P/Zn/K-solubilization, calcite degradation, IAA, and siderophore production. The plant growth-promoting rhizobacteria (PGPR) were evaluated on wheat under water stress. The P. balearica strain RF-2 primed seeds showed a maximum promptness index and germination index under PEG-stress, that is, 68% and 100%, respectively. Inoculation significantly improved plant growth, leaf area, and biomass under water stress. P. mirabilis R2 inoculated plant leaves showed the highest water contents as compared to the plants inoculated with other strains. C. sakazakii RF-4 inoculated plants showed minimum cell injury, electrolyte leakage, and maximum cell membrane stability at PEG stress. After 13 days exposure to drought, C. sakazakii RF-4 treated plants showed an overall higher expression of cytosolic ascorbate peroxidase (cAPX) and ribulose-bisphosphate carboxylase (rbcL) genes. The activity of stress-induced catalase and polyphenol oxidase was reduced, while that of peroxidase and superoxide dismutase increased after inoculation but the response was temporal. Taken together, this data explains that different PGPR (especially C. sakazakii RF-4) modulate differential responses in wheat that eventually leads towards drought tolerance, hence, it has the potential to enhance crop production in arid regions.

Abstract:Wheat ( Triticum aestivum L.) yield and physiology are adversely affected due to limited water availability. However, desiccation-tolerant plant growth-promoting rhizobacteria (DT-PGPR) are potential candidates that can overcome the negative impacts of water stress. In the present study, a total of 164 rhizobacterial isolates were screened for desiccation tolerance up to −0.73 MPa osmotic pressure, of which five isolates exhibited growth and expression of plant growth properties under the influence of desiccation stress of −0.73 MPa. These five isolates were identified as Enterobacter cloacae BHUAS1, Bacillus cereus BHUAS2, Bacillus megaterium BHUIESDAS3, Bacillus megaterium BHUIESDAS4, and Bacillus megaterium BHUIESDAS5. All five isolates exhibited plant growth-promoting properties and production of exopolysaccharide (EPS) under the impact of desiccation stress. Furthermore, a pot experiment on wheat (variety HUW-234) inoculated with the isolates Enterobacter cloacae BHUAS1, Bacillus cereus BHUAS2, and Bacillus megaterium BHUIESDAS3 exhibited a positive influence on the growth of wheat under the condition of water stress. A significant improvement in plant height, root length, biomass, chlorophyll and carotenoid content, membrane stability index (MSI), leaf relative water content (RWC), total soluble sugar, total phenol, proline, and total soluble protein, were recorded under limited water-induced drought stress in treated plants as compared with non-treated plants. Moreover, plants treated with Enterobacter cloacae BHUAS1, Bacillus cereus BHUAS2, and Bacillus megaterium BHUIESDAS3 depicted improvement in enzymatic activities of several antioxidant enzymes such as guaiacol peroxidase (POD), catalase (CAT), and ascorbate peroxidase (APX). Beside this significant decrease in electrolyte leakage, H 2 O 2 and malondialdehyde (MDA) contents were also recorded in treated plants. From the results obtained, it is evident that E . cloacae BHUAS1, B . megaterium BHUIESDAS3, and B . cereus BHUAS2 are the potential DT-PGPR having the capability to sustain growth and yield, alleviating the deleterious effect of water stress in wheat.

Abstract:Plant growth-promoting bacteria (PGPB) are beneficial free-living soil, rhizospheric, epiphytic, and endophytic microorganisms capable to stimulate plant growth and increase host plant resistance and tolerance to a wide range of biotic and abiotic stresses. A number of PGPB associated with wheat and different cereals have been identified comprising bacterial strains belonging to such genera as Bacillus, Azospirillum, Arthrobacter, Acinetobacter, Azotobacter, Citricoccus, Lysinibacillus, Burkholderia, Paenibacillus, Serratia, Pseudomonas, etc. Several studies have confirmed that some species of bacteria associated with the rhizosphere of plants are useful for growth, development, and formation of yield and quality of agricultural crops. Furthermore, those bacteria which are capable to colonize internal plant tissues, namely, endophytes, may be more successful (compared to rhizospheric bacteria) in the promotion of plant growth and development under both normal and long-term stress conditions. PGPB-induced development of defense responses and the formation of tolerance under the exposure to various abiotic stresses have been demonstrated in numerous plants, including wheat. The mechanisms of such physiological effect of PGPB on host plants are believed to be varied, intertwined, and specific; PGPB positively affects on plants through biosynthesis of numerous biologically active compounds, for instance, substances with antibiotic and insecticidal activities, biosurfactants, siderophores, chelators, phytohormones, enzymes, and nitrogen fixation, regulating the level of ethylene in plants, and improving macro−/micronutrient bioavailability, development of systemic resistance to diseases, and tolerance to abiotic stresses with involving salicylate-dependent or jasmonate-dependent signaling pathways. In this review, the role of beneficial PGPB in ameliorating the many deleterious consequences during abiotic stresses has been considered. Besides, B. subtilis’ efficiency on abiotic stress tolerance induction in wheat according to their ecological groups (ecotypes) is discussed as well.

Abstract:Abstract Background The effectiveness of two PGPB; Azospirillum brasilense NO40 and Stenotrophomonas maltophilia B11 was investigated in enhancing the drought tolerance of wheat ( Triticum aestivum L.) seedlings cultivar Gemiza9. The inoculated or uninoculated grains were sown in unsterilized sandy soil and watered normally untill the 8 th day. Drought stress was initiated by completely withholding water for 7 days (until wilting). Samples were collected after 15 days from sowing to evaluate some growth criteria, damage and defense indicators and to analyze the roots’ protein pattern. Results The results showed that inoculating wheat seedlings with these strains significantly diminished the inhibitory effects of drought stress on the relative water content of roots, shoots and leaves; area of leaves; contents of pigments (chlorophyll a and b) and ascorbic acid; and on the protein patterns of roots. Moreover, the bacterial inoculation notably reduced the drought-induced damage indicated by lower leakage of electrolytes and less accumulation of Malondialdehyde and hydrogen peroxide, surprisingly with less enhanced production of proline and activities of catalase and peroxidase than their uninoculated counterparts. Under normal conditions, inoculating wheat plants with these PGPB resulted in significantly promoted growth and elevated contents of pigments and altered protein patterns of roots. Conclusion Overall, we can say that both Azospirillum brasilense NO40 and Stenotrophomonas maltophilia B11 were able to deactivate the growth inhibition in wheat seedlings to some extent, while maintaining a certain level of efficient protection against damage under drought stress.

Abstract:Plant growth-promoting rhizobacteria (PGPRs) represent a promising strategy for enhancing plant resilience and yields under drought-stress conditions. This study isolated and characterized PGPR from wheat rhizosphere soil in Egypt. Four PGPR strains were evaluated for an array of plant growth-promoting traits, including IAA production, biofilm formation, siderophore production, nitrogen fixation, ACC deaminase activity, phosphate solubilization, and antagonistic potential. Molecular identification via 16S rRNA sequencing classified three isolates (MMH101, MMH102, and MMH103) within the Bacillus genus and one isolate (MMH104) as Myroides sp. Greenhouse experiments examined the effects of PGPR inoculation on the drought-stressed Egyptian wheat cultivar, Gimmeza-9. Wheat plants inoculated with PGPR isolates showed dramatic improvements in growth parameters and stress tolerance indicators compared to non-inoculated controls when subjected to a 10-day drought period, with Bacillus rugosus (MMH101) inoculation resulting in increases of 61.8% in fresh biomass, 77.2% in dry biomass, 108.5% shoot length, and 134.9% root length. PGPR treatments also elevated the chlorophyll and proline content while reducing malondialdehyde levels. The findings demonstrate the effectiveness of PGPR inoculation in enhancing the morphology, physiology, and drought stress resilience of wheat. Isolated PGPR strains hold promise as biofertilizers for improving cereal productivity under water-deficit conditions.

Abstract:Soil salinity is one of the main yield-limiting factors in various crops. Under different environmental stresses, many rhizobacteria have demonstrated encouraging role in enhancing plant growth and tolerate stress conditions. In this study, three potential 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase- and exopolysaccharides (EPS)-secreting bacterial strains including Bacillus megaterium, B. tequilensis, and Pseudomonas putida have been assessed for their growth-promoting characteristics. These bacterial strains positively affected the physiology, biochemistry, and antioxidant enzymatic activities of wheat plant, under salinity stress. Results of this study depicted that the inoculation of PGPR positively invigorates growth attributes like relative water content and photosynthetic pigments of wheat seedling under saline conditions. Moreover, plants inoculated with PGPR also showed decreased concentration of malondialdehyde (MDA) and hydrogen peroxide (H2O2). Inoculation of PGPR reduced electrolytic leakage and enhanced enzymatic activity for the scavenging of reactive oxygen species (ROS). These PGPR also increased the production of proline and total soluble sugar. Expression analysis of selected genes by qPCR revealed higher expression of Salt Overly Sensitive (SOS1 and SOS4) genes and predicted their potential role in stress tolerance. These genes can be further overexpressed in wheat plant to tolerate salinity stress. On the basis of these findings, it can be concluded that the priming of seeds with aforementioned PGPR can decrease the adverse effects of salinity on wheat plant.

Abstract:Growth stimulating bacteria help remediate dry arid soil and plant stress. Here, Pseudomonas sp. and Pantoea sp. we used to study the stress ecology of Hordeum vulgare and the environmental impact of water deficit on soil characteristics, growth, photosynthesis apparatus, mineral acquisition and antioxidiant defense. Plants inoculated with Pseudomonas or Pantoea had significantly higher (about 2 folds) soil carbon flux (soil respiration), chlorophyll levels (18%), net photosynthetic rate (33% in Pantoea and 54% in Pseudomonas ), (44%) stomatal conductance than uninoculated plants in stressed conditions. Both bacterial strains improved leaf growth (23-29%) and root development under well-watered conditions but reduced around (25%) root biomass under drought. Plants inoculated with Pseudomonas or Pantoea under drought also increased of about 27% leaf respiration and transpiration (48%) but decreased water use efficiency, photoinhibition (91%), and the risk of oxidative stress (ETR/A) (49%). Drought stress increased most of the studied antioxidant enzymatic activities in the plants inoculated with Pseudomonas or Pantoea , which reduce the membrane damage and protect plants form oxidative defenses. Drought stress increased K + acquisition around 50% in both shoots inoculated with Pseudomonas or Pantoea relative to non-stressed plants. Plants inoculated with Pseudomonas or Pantoea increased shoot Na + while root Na + only increased in plants inoculated with Pseudomonas in stressed conditions. Drought stress increased shoot Mg 2+ in plants inoculated with Pseudomonas or Pantoea but did not affect Ca 2+ relative to non-stressed plants. Drought stress increased about 70% K + /Na + ratio only in plants inoculated with Pseudomonas relative to non-stressed plants. Our results indicate that inoculating barley with the studied bacterial strains increases plant biomass and can therefore play a role in the environmental remediation of drylands for food production.

Abstract:Drought stress (DS) is one of the severe abiotic stresses that cause reductions in agricultural production. This study was conducted to examine the efficiency of arbuscular mycorrhizal fungi (M: consortium) and plant growth-promoting rhizobacteria (R: Z2 and Z4 strains, B: CHIN14, AAP22, AAP2, and CHIN15 strains), individually or in combination, as an appropriate strategy to improve durum wheat productivity under two water regimes: well-watered (75% FC) and DS (25% FC). DS exhibition reduced biomass, stomatal conductance, stem water potential, and increased malondialdehyde and hydrogen peroxide content. Under DS, AMF or B promoted plant growth, such as shoot height by 49 and 17%, respectively; by activating the photosynthetic mechanism and increasing the synthesis of proline content by 369% in plants treated with AMF, and antioxidant enzyme activities, such as polyphenoloxydase (PPO), in leaves by 36% in plant inoculated with B. After the experiment, total organic matter and carbon of the soil were improved by single application of B by 99 and 317%, respectively. This finding implies that the combination of mycorrhiza and PGPR regulates the physiological and biochemical processes utilized by wheat to cope with drought and improves the comprehension of the soil-plant relationship.
The stimulatory effects of plant growth promoting rhizobacteria and plant growth regulators on wheat physiology grown in sandy soil

Abstract:The present study was aimed to investigate the effects of plant growth promoting rhizobacteria (PGPR) and plant growth regulators (PGRs) on the physiology and yield of wheat grown in less fertile sandy soil. The isolated PGPR strains were identified by 16S-rRNA gene sequencing as Planomicrobium chinense (P1), Bacillus cereus (P2) and Pseudomonas fluorescens (P3). Wheat varieties (Galaxy-13 and Pak-2013) differing in sensitivity to drought were soaked in fresh cultures of bacterial isolates and the PGRs (salicylic acid and putrescine) were sprayed at 150 mg/L on seedlings at three leaf stage. PGPR and PGRs treated plants showed significant increase in the contents of chlorophyll, sugar and protein even under harsh environmental conditions. Drought stress enhanced the production of proline, antioxidant enzymes and lipid peroxidation but a decrease was noted in the biochemical content (i.e. chlorophyll, protein and sugar) of inoculated plants. PGPR inoculation also significantly enhanced the yield parameters (i.e. plant height, spike length, grain yield and weight) and improved the fertility status of sandy soil. The accumulation of macronutrient, total NO3-N and P concentration and soil moisture content of rhizosphere soil was also enhanced by PGPRs inoculation. It is concluded that the combined effects of PGPR and PGRs have profound effects on the biochemical responses and drought tolerance of wheat grown in sandy soils.

Mitigation of water scarcity with sustained growth of Rice by plant growth promoting bacteria

Abstract:Climate change augments the risk to food security by inducing drought stress and a drastic decline in global rice production. Plant growth-promoting bacteria (PGPB) have been known to improve plant growth under drought stress. Here in the present study, we isolated, identified, and well-characterized eight drought-tolerant bacteria from the rice rhizosphere that are tolerant to 20% PEG-8000. These strains exhibited multiple plant growth-promoting traits, i.e., 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity, exopolysaccharide production, phosphate (P)-solubilizing activity (51–356 μg ml -1 ), indole-3 acetic acid (IAA) production (14.3–46.2 μg ml -1 ), and production of organic acids (72–178 μg ml -1 ). Inoculation of bacterial consortium ( Bacillus subtilis NM-2, Brucella haematophilum NM-4, and Bacillus cereus NM-6) significantly improved seedling growth and vigor index (1009.2-1100) as compared to non-inoculated stressed plants (630-957). Through rhizoscanning, efficiency of the consortium was validated by improved root parameters such as root length (17%), diameter, and surface area (18%) of all tested genotypes as compared with respective non-inoculated stressed treatments. Furthermore, the response of consortium inoculation on three rice genotypes was positively correlated with improved plant growth and drought stress ameliorating traits by the accumulation of osmoprotectant, i.e., proline (85.8%–122%), relative water content (51%), membrane stability index (64%), and production of antioxidant enzymes to reduce oxidative damage by reactive oxygen species. A decrease in temperature and improved chlorophyll content of inoculated plants were found using infrared thermal imaging and soil plant analyzer development (SPAD), respectively. The key supporting role of inoculation toward stress responses was validated using robust techniques like infrared thermal imaging and an infrared gas analyzer. Furthermore, principal component analysis depicts the contribution of inoculation on stress responses and yield of tested rice genotypes under water stress. The integration of drought-tolerant rice genotype (NIBGE-DT02) and potential bacterial strains, i.e., NM-2, NM-4, and NM-6, can serve as an effective bioinoculant to cope with water scarcity under current alarming issues related to food security in fluctuating climate.

Abstract:Soil salinity hampers durum wheat plant growth and development at various stages. The detrimental effects of salinity on plant cellular and physiological processes necessitate strategies to alleviate its negative impact and improve overall crop yield. This study investigates the efficacy of plant growth-promoting rhizobacteria (PGPR) bacteria inoculation in mitigating salinity stress on two durum wheat genotypes with contrasting degrees of salinity tolerance (Tamaroi, salt-sensitive and Line 5004, salt-tolerant) under greenhouse and field conditions. For this purpose, two halotolerant-PGPR strains, Pseudomonas jordanii strain G34 and Oceanobacillus jordanicus strain GSFE11, were utilized for the inoculation. For the greenhouse experiment, the two selected genotypes were subjected to salinity at the flag leaf stage with continuous irrigation with a Hoagland solution supplemented with 50 mM NaCl. Field experiments were conducted across two locations with contrasting salinity levels over two growing seasons. At the end of both experiments, various parameters including total weight, spike weight, grain weight, spike number, spikelet number, grains per spike and thousand kernel weight were measured. The halotolerant PGPRs, P. jordanii strain G34 and O. jordanicus strain GSFE11, proved effective in alleviating salinity-induced adverse effects and enhancing growth under greenhouse and field conditions. However, bacterial inoculation significantly improved growth in the salt-sensitive genotype and such effects were not observed in the tolerant genotype, emphasizing genotype-specific responses. Notably, inoculation with O. jordanicus increased Na+ and Ca2+ uptake in the salt-tolerant "Line 5004" without hindering growth, suggesting one of its potential mechanisms for salt tolerance. This research demonstrates the potential of halotolerant-PGPR inoculation in enhancing durum wheat production in saline environments, but also underscores the importance of understanding genotype-specific responses for tailored interventions.

Abstract:Wheat is an important crop, used as staple food in numerous countries around the world. However, wheat productivity is low in the developing world due to several biotic and abiotic stresses, particularly drought stress. Non-availability of drought-tolerant wheat genotypes at different growth stages is the major constraint in improving wheat productivity in the developing world. Therefore, screening/developing drought-tolerant genotypes at different growth stages could improve the productivity of wheat. This study assessed seed germination and seedling growth of eight wheat genotypes under polyethylene glycol (PEG)-induced stress. Two PEG-induced osmotic potentials (i.e., -0.6 and -1.2 MPa) were included in the study along with control (0 MPa). Wheat genotypes included in the study were ‘KLR-16’, ‘B6’, ‘J10’, ‘716’, ‘A12’, ‘Seher’, ‘KTDH-16’, and ‘J4’. Data relating to seed germination percentage, root and shoot length, fresh and dry weight of roots and shoot, root/shoot length ratio and chlorophyll content were recorded. The studied parameters were significantly altered by individual and interactive effects of genotypes and PEG-induced osmotic potentials. Seed germination and growth parameters were reduced by osmotic potentials; however, huge differences were noted among genotypes. A reduction of 32.83 to 53.50% was recorded in seed germination, 24.611 to 47.75% in root length, 37.83 to 53.72% in shoot length, and 53.35 to 65.16% in root fresh weight. The genotypes, ‘J4’, ‘KLR-16’ and ‘KTDH-16’, particularly ‘J4’ better tolerated increasing osmotic potentials compared to the rest of the genotypes included in the study. Principal component analysis segregated these genotypes from the rest of the genotypes included in the study indicated that these can be used in the future studies to improve the drought tolerance of wheat crop. The genotype ‘J4’ can be used as a breeding material to develop drought resistant wheat genotypes.

Abstract:Climate change has devastating effects on plant growth and yield. During ontogenesis, plants are subjected to a variety of abiotic stresses, including drought and salinity, affecting the crop loss (20–50%) and making them vulnerable in terms of survival. These stresses lead to the excessive production of reactive oxygen species (ROS) that damage nucleic acid, proteins, and lipids. Plant growth-promoting bacteria (PGPB) have remarkable capabilities in combating drought and salinity stress and improving plant growth, which enhances the crop productivity and contributes to food security. PGPB inoculation under abiotic stresses promotes plant growth through several modes of actions, such as the production of phytohormones, 1-aminocyclopropane-1-carboxylic acid deaminase, exopolysaccharide, siderophore, hydrogen cyanide, extracellular polymeric substances, volatile organic compounds, modulate antioxidants defense machinery, and abscisic acid, thereby preventing oxidative stress. These bacteria also provide osmotic balance; maintain ion homeostasis; and induce drought and salt-responsive genes, metabolic reprogramming, provide transcriptional changes in ion transporter genes, etc. Therefore, in this review, we summarize the effects of PGPB on drought and salinity stress to mitigate its detrimental effects. Furthermore, we also discuss the mechanistic insights of PGPB towards drought and salinity stress tolerance for sustainable agriculture.

Abstract:Plant growth-promoting bacteria play an essential role in enhancing the physical, chemical and biological characters of soils by facilitating nutrient uptake and water flow, especially under abiotic stress conditions, which are major constrains to agricultural development and production. Drought is one of the most harmful abiotic stress and perhaps the most severe problem facing agricultural sustainability, leading to a severe shortage in crop productivity. Drought affects plant growth by causing hormonal and membrane stability perturbations, nutrient imbalance and physiological disorders. Furthermore, drought causes a remarkable decrease in leaf numbers, relative water content, sugar yield, root yield, chlorophyll a and b and ascorbic acid concentrations. However, the concentrations of total phenolic compounds, electrolyte leakage, lipid peroxidation, amounts of proline, and reactive oxygen species are considerably increased because of drought stress. This negative impact of drought can be eliminated by using plant growth-promoting bacteria (PGPB). Under drought conditions, application of PGPB can improve plant growth by adjusting hormonal balance, maintaining nutrient status and producing plant growth regulators. This role of PGPB positively affects physiological and biochemical characteristics, resulting in increased leaf numbers, sugar yield, relative water content, amounts of photosynthetic pigments and ascorbic acid. Conversely, lipid peroxidation, electrolyte leakage and amounts of proline, total phenolic compounds and reactive oxygen species are decreased under drought in the presence of PGPB. The current review gives an overview on the impact of drought on plants and the pivotal role of PGPB in mitigating the negative effects of drought by enhancing antioxidant defense systems and increasing plant growth and yield to improve sustainable agriculture.

Abstract:Introduction: The escalating threat of drought poses a significant challenge to sustainable food production and human health, as water scarcity adversely impacts various aspects of plant physiology. Maize, a cornerstone in staple cereal crops, faces the formidable challenge of drought stress that triggers a series of transformative responses in the plant. Methods: The present study was carried out in two sets of experiments. In first experiment, drought stress was applied after maintaining growth for 45 days and then irrigation was skipped, and plant samples were collected at 1 st , 3 rd and 6 th day of drought interval for evaluation of changes in plant growth, water relation (relative water content) and antioxidants activity by inoculating indigenously isolated drought tolerant biofilm producing rhizobacterial isolates ( Bacillus subtilis SRJ4, Curtobacterium citreum MJ1). In the second experiment, glycine betaine was applied as osmoregulator in addition to drought tolerant PGPR to perceive modulation in photosynthetic pigments (Chlorophyll a and b) and plant growth under varying moisture stress levels (100, 75 and 50% FC). Results and discussion: Results of the study revealed upsurge in root and shoot length, fresh and dry biomass of root and shoot besides increasing chlorophyll contents in water stressed inoculated plants compared to uninoculated plants. Glycine betaine application resulted in an additional boost to plant growth and photosynthetic pigments, when applied in combination with bacterial inoculants. However, both bacterial inoculants behaved differently under drought stress as evident from their biochemical and physiological attributes. Isolate SRJ4 proved to be superior for its potential to express antioxidant activity, leaf water potential and relative water contents and drought responsive gene expression while isolate MJ1 showed exclusive increase in root dry biomass and plant P contents. Though it is quite difficult to isolate the bacterial isolates having both plant growth promoting traits and drought tolerance together yet, such biological resources could be an exceptional option to be applied for improving crop productivity and sustainable agriculture under abiotic stresses. By exploring the combined application of PGPR and glycine betaine, the study seeks to provide insights into potential strategies for developing sustainable agricultural practices aimed at improving crop resilience under challenging environmental conditions.

Abstract:This review is devoted to the analysis and systematization of modern data on the participation of endophytic plant growth-promoting ( PGP ) bacteria in the regulation of growth, development, yield formation, and stress resistance of cultivated plants, mainly spring wheat as the main bread crop. The present data on the interaction of plants with PGP bacteria under normal and drought conditions are described. Particular attention is paid to the molecular mechanisms of regulation of plant metabolism by PGP bacteria, as well as their role in reducing the negative effects of drought, achieved by modulating various processes in plants, for example, improving the supply of moisture and mineral nutrients, and activating the antioxidant and osmoprotective plant systems. A key role in the adaptation and resistance/tolerance of plants caused by PGP bacteria is played by their ability to produce various metabolites with the properties of biologically active substances, including substances with antimicrobial and hormonal activity, enzymes and other compounds. Information about the endophytic microbiome of wheat is given and the elucidation of its role and functions in the plant stress response and adaptation that are necessary for the development of effective and safe strategies for their practical application in order to maximize the adaptation and productive potential of wheat under changing environmental conditions.
