Abstract:Abstract Sensory inputs enter a constantly active brain, whose state is always changing from one moment to the next. Currently, little is known about how ongoing, spontaneous brain activity participates in online task processing. We employed 7 Tesla fMRI and a threshold-level visual perception task to probe the effects of prestimulus ongoing brain activity on perceptual decision-making and conscious recognition. Prestimulus activity originating from distributed brain regions, including visual cortices and regions of the default-mode and cingulo-opercular networks, exerted a diverse set of effects on the sensitivity and criterion of conscious recognition, and categorization performance. We further elucidate the mechanisms underlying these behavioral effects, revealing how prestimulus activity modulates multiple aspects of stimulus processing in highly specific and network-dependent manners. These findings reveal heretofore unknown network mechanisms underlying ongoing brain activity’s influence on conscious perception, and may hold implications for understanding the precise roles of spontaneous activity in other brain functions.

Spontaneous perception: a framework for task-free, self-paced perception

Abstract:Flipping through social media feeds, viewing exhibitions in a museum, or walking through the botanical gardens, people consistently choose to engage with and disengage from visual content. Yet, in most laboratory settings, the visual stimuli, their presentation duration, and the task at hand are all controlled by the researcher. Such settings largely overlook the spontaneous nature of human visual experience, in which perception takes place independently from specific task constraints and its time course is determined by the observer as a self-governing agent. Currently, much remains unknown about how spontaneous perceptual experiences unfold in the brain. Are all perceptual categories extracted during spontaneous perception? Does spontaneous perception inherently involve volition? Is spontaneous perception segmented into discrete episodes? How do different neural networks interact over time during spontaneous perception? These questions are imperative to understand our conscious visual experience in daily life. In this article we propose a framework for spontaneous perception. We first define spontaneous perception as a task-free and self-paced experience. We propose that spontaneous perception is guided by four organizing principles that grant it temporal and spatial structures. These principles include coarse-to-fine processing, continuity and segmentation, agency and volition, and associative processing. We provide key suggestions illustrating how these principles may interact with one another in guiding the multifaceted experience of spontaneous perception. We point to testable predictions derived from this framework, including (but not limited to) the roles of the default-mode network and slow cortical potentials in underlying spontaneous perception. We conclude by suggesting several outstanding questions for future research, extending the relevance of this framework to consciousness and spontaneous brain activity. In conclusion, the spontaneous perception framework proposed herein integrates components in human perception and cognition, which have been traditionally studied in isolation, and opens the door to understand how visual perception unfolds in its most natural context.

Abstract:Sensory perception is essential for transforming incoming information in the brain into targeted behavior. Our brains are everlastingly active, and variations in perception are ubiquitously associated with human behavioral performance. Previous studies indicate that changes in spontaneous neural activity within local sensory areas correlate with the perception of ambiguous stimuli. However, the contribution of whole brain spontaneous networks to perception is not well understood. Using an ambiguous tactile temporal discrimination task, we demonstrate that the interaction between whole-brain networks in the seconds of the spontaneous pre-stimulus period also contributes to perception during the task. Transitions to a frontal and a multi-frequency network across the brain are essential for the correct percept. Conversely, incorrect percepts are mainly preceded by transitions to an alpha-parietal network. Brain transitions occur faster during the period before stimulus presentation for correct stimuli detection, suggesting the need for enhanced network flexibility during this phase.

Abstract:The neuroscience of consciousness aims to identify neural markers that distinguish brain dynamics in healthy individuals from those in unconscious conditions. Recent research has revealed that specific brain connectivity patterns correlate with conscious states and diminish with loss of consciousness. However, the contribution of these patterns to shaping conscious processing remains unclear. Our study investigates the functional significance of these neural dynamics by examining their impact on participants' ability to process external information during wakefulness. Using fMRI recordings during an auditory detection task and rest, we show that ongoing dynamics are underpinned by brain patterns consistent with those identified in previous research. Detection of auditory stimuli at threshold is specifically improved when the connectivity pattern at stimulus presentation corresponds to patterns characteristic of conscious states. Conversely, the occurrence of these conscious state-associated patterns increases after detection, indicating a mutual influence between ongoing brain dynamics and conscious perception. Our findings suggest that certain brain configurations are more favorable to the conscious processing of external stimuli. Targeting these favorable patterns in patients with consciousness disorders may help identify windows of greater receptivity to the external world, guiding personalized treatments.

Abstract:Despite intense recent research, the neural correlates of conscious visual perception remain elusive. The most established paradigm for studying brain mechanisms underlying conscious perception is to keep the physical sensory inputs constant and identify brain activities that correlate with the changing content of conscious awareness. However, such a contrast based on conscious content alone would not only reveal brain activities directly contributing to conscious perception, but also include brain activities that precede or follow it. To address this issue, we devised a paradigm whereby we collected, trial-by-trial, measures of objective performance, subjective awareness, and the confidence level of subjective awareness. Using magnetoencephalography recordings in healthy human volunteers, we dissociated brain activities underlying these different cognitive phenomena. Our results provide strong evidence that widely distributed slow cortical potentials (SCPs) correlate with subjective awareness, even after the effects of objective performance and confidence were both removed. The SCP correlate of conscious perception manifests strongly in its waveform, phase, and power. In contrast, objective performance and confidence were both contributed by relatively transient brain activity. These results shed new light on the brain mechanisms of conscious, unconscious, and metacognitive processing.
Spontaneous Brain Activity in the Default Mode Network is Sensitive to Different Resting-State Conditions with Limited Cognitive Load.

Abstract:BACKGROUND:Recent functional MRI (fMRI) studies have demonstrated that there is an intrinsically organized default mode network (DMN) in the resting brain, primarily made up of the posterior cingulate cortex (PCC) and the medial prefrontal cortex (MPFC). Several previous studies have found that the DMN is minimally disturbed during different resting-state conditions with limited cognitive demand. However, this conclusion was drawn from the visual inspection of the functional connectivity patterns within the DMN and no statistical comparison was performed.METHODOLOGY/PRINCIPAL FINDINGS:Four resting-state fMRI sessions were acquired: 1) eyes-closed (EC) (used to generate the DMN mask); 2) EC; 3) eyes-open with no fixation (EO); and 4) eyes-open with a fixation (EO-F). The 2-4 sessions were counterbalanced across participants (n = 20, 10 males). We examined the statistical differences in both functional connectivity and regional amplitude of low frequency fluctuation (ALFF) within the DMN among the 2-4 resting-state conditions (i.e., EC, EO, and EO-F). Although the connectivity patterns of the DMN were visually similar across these three different conditions, we observed significantly higher functional connectivity and ALFF in both the EO and the EO-F conditions as compared to the EC condition. In addition, the first and second resting EC conditions showed significant differences within the DMN, suggesting an order effect on the DMN activity.CONCLUSIONS/SIGNIFICANCE:Our findings of the higher DMN connectivity and regional spontaneous activities in the resting state with the eyes open suggest that the participants might have more non-specific or non-goal-directed visual information gathering and evaluation, and mind wandering or daydreaming during the resting state with the eyes open as compared to that with the eyes closed, thus providing insights into the understanding of unconstrained mental activity within the DMN. Our results also suggest that it should be cautious when choosing the type of a resting condition and designating the order of the resting condition in multiple scanning sessions in experimental design.

Abstract:Research on the neural basis of conscious perception has almost exclusively shown that becoming aware of a stimulus leads to increased neural responses. By designing a novel form of perceptual filling-in (PFI) overlaid with a dynamic texture display, we frequency-tagged multiple disappearing targets as well as their surroundings. We show that in a PFI paradigm the disappearance of a stimulus and subjective invisibility are associated with increases in neural activity, as measured with steady-state visually evoked potentials (SSVEP), in electroencephalography (EEG). We also find that this increase correlates with alpha-band activity, a well-established neural measure of attention. These findings cast doubt on the direct relationship previously reported between the strength of neural activity and conscious perception, at least when measured with current tools, such as the SSVEP. Instead we conclude that SSVEP strength more closely measures changes in attention.

Abstract:One of the goals of the neuroscience of consciousness is to identify neural markers capable of distinguishing brain dynamics in awake, healthy individuals from unconscious conditions. This problem also has a clinical diagnostic interest in disorders of consciousness. Recent research has shown that brain connectivity patterns characterized by long-range interactions and anticorrelations are associated with conscious states and diminish with loss of consciousness in human and non-human primates. However, the precise contribution of these patterns to conscious processing and subjective experience formation remains unclear. In this study, we investigated the functional role of these brain patterns in shaping conscious content by examining their influence on participants’ ability to process external information during wakefulness. Participants underwent fMRI recordings during an auditory detection task. Phase coherence-based functional connectivity and k-means clustering confirmed that the ongoing dynamics were underpinned by brain patterns consistent with those identified in previous research, including the “high pattern” characteristic of conscious states. We found that the detection of auditory stimuli at threshold was specifically improved when the connectivity pattern at the time of presentation corresponded to this high-pattern. In return, the occurrence of the high-pattern increased after detection, indicating that participants were more likely to transition to a high-pattern following stimulus detection. Our findings suggest that ongoing brain dynamics and conscious perception mutually influence each other and that certain brain configurations are more favorable for conscious processing of external stimuli. In the future, targeting these moments of favorable patterns in patients with disorders of consciousness may help us identify windows of greater receptivity to the external world, paving the way for developing individualized patient care protocols.

Abstract:After more than 30 years of extensive investigation, impressive progress has been made in identifying the neural correlates of consciousness (NCC). However, the functional role of spatiotemporally distinct consciousness-related neural activity in conscious perception is debated. Based on empirical EEG findings, e.g., of the enhanced early negative wave and late positive wave under conscious conditions, an influential framework proposed that consciousness-related neural activities could be dissociated into two distinct processes: phenomenal and access consciousness. This framework has been supported mainly by comparison of neural activity between report and no-report paradigms; however, though hotly debated, its authenticity has not been examined in a single paradigm with more informative intracranial recordings. In the present study, we employed a novel visual awareness task and recorded the local field potential (LFP) of epilepsy patients with electrodes implanted in cortical and subcortical regions. Overall, we found that the latency of visual awareness-related activity exhibited a bimodal distribution, and the recording sites with short and long latencies were largely separated in location, except in the lateral prefrontal cortex (lPFC). The mixture of short and long latencies in the lPFC indicates that it plays a critical role in linking phenomenal and access consciousness. However, the division between the two is not as simple as the central sulcus, as proposed previously. Moreover, in 4 patients with electrodes implanted in the bilateral prefrontal cortex, early awareness-related activity was confined to the contralateral side, while late awareness-related activity appeared on both sides. Finally, Granger causality analysis showed that awareness-related information flowed from the early sites to the late sites. These results provide the first LFP evidence of neural correlates of phenomenal and access consciousness, which sheds light on the spatiotemporal dynamics of NCC in the human brain.

Abstract:Most people can experience things that are not there: when dreaming, remembering the past or imagining the future. Contemporary theories of brain function propose that such internally generated perceptual experience relies on similar neural machinery to that engaged by externally driven perception (Bastos et al., Citation 2012 ; Friston et al., Citation 2006 ). In line with this, several lines of research have demonstrated overlapping patterns of activity in sensory cortices during imagination and perception. For example, activity in early visual cortices can be used to decode the content of dreams (Horikawa et al., Citation 2013 ; Siclari et al., Citation 2017 ); sensory patterns are reinstated in early visual regions during memory (Bosch et al., Citation 2014 ; Harrison & Tong, Citation 2009 ); and decoders trained on visual cortex activity during perception can predict what people imagine (Albers et al., Citation 2013 ; Lee et al., Citation 2012 ; Ragni et al., Citation 2020 ), especially when they imagine very vividly (Dijkstra et al., Citation 2017 ). Given this overlap in neural activation patterns, how does our brain keep apart sensory activity representing reality from that representing imagination?

Abstract:Spontaneous activity of the human brain provides a window to explore intrinsic principles of functional organization. However, most studies have focused on interregional functional connectivity. The principles underlying rich repertoires of instantaneous activity remain largely unknown. We apply a recently proposed eigen-microstate analysis to three resting-state functional MRI datasets to identify basic modes that represent fundamental activity patterns that coexist over time. We identify five leading basic modes that dominate activity fluctuations. Each mode exhibits a distinct functional system-dependent coactivation pattern and corresponds to specific cognitive profiles. In particular, the spatial pattern of the first leading basis mode shows the separation of activity between the default-mode and primary and attention regions. Based on theoretical modelling, we further reconstruct individual functional connectivity as the weighted superposition of coactivation patterns corresponding to these leading basic modes. Moreover, these leading basic modes capture sleep deprivation-induced changes in brain activity and interregional connectivity, primarily involving the default-mode and task-positive regions. Our findings reveal a dominant set of basic modes of spontaneous activity that reflect multiplexed interregional coordination and drive conventional functional connectivity, furthering the understanding of the functional significance of spontaneous brain activity.

Abstract:If conscious perception requires global information integration across active distant brain networks, how does the loss of conscious perception affect neural processing in these distant networks? Pioneering studies on perceptual suppression (PS) described specific local neural network responses in primary visual cortex, thalamus and lateral prefrontal cortex of the macaque brain. Yet the neural effects of PS have rarely been studied with intracerebral recordings outside these cortices and simultaneously across distant brain areas. Here, we combined (1) a novel experimental paradigm in which we produced a similar perceptual disappearance and also re-appearance by using visual adaptation with transient contrast changes, with (2) electrophysiological observations from human intracranial electrodes sampling wide brain areas. We focused on broadband high-frequency (50-150 Hz, i.e., gamma) and low-frequency (8-24 Hz) neural activity amplitude modulations related to target visibility and invisibility. We report that low-frequency amplitude modulations reflected stimulus visibility in a larger ensemble of recording sites as compared to broadband gamma responses, across distinct brain regions including occipital, temporal and frontal cortices. Moreover, the dynamics of the broadband gamma response distinguished stimulus visibility from stimulus invisibility earlier in anterior insula and inferior frontal gyrus than in temporal regions, suggesting a possible role of fronto-insular cortices in top-down processing for conscious perception. Finally, we report that in primary visual cortex only low-frequency amplitude modulations correlated directly with perceptual status. Interestingly, in this sensory area broadband gamma was not modulated during PS but became positively modulated after 300 ms when stimuli were rendered visible again, suggesting that local networks could be ignited by top-down influences during conscious perception.

Abstract:The characterization of topological architecture of complex brain networks is one of the most challenging issues in neuroscience. Slow (<0.1 Hz), spontaneous fluctuations of the blood oxygen level dependent (BOLD) signal in functional magnetic resonance imaging are thought to be potentially important for the reflection of spontaneous neuronal activity. Many studies have shown that these fluctuations are highly coherent within anatomically or functionally linked areas of the brain. However, the underlying topological mechanisms responsible for these coherent intrinsic or spontaneous fluctuations are still poorly understood. Here, we apply modern network analysis techniques to investigate how spontaneous neuronal activities in the human brain derived from the resting-state BOLD signals are topologically organized at both the temporal and spatial scales. We first show that the spontaneous brain functional networks have an intrinsically cohesive modular structure in which the connections between regions are much denser within modules than between them. These identified modules are found to be closely associated with several well known functionally interconnected subsystems such as the somatosensory/motor, auditory, attention, visual, subcortical, and the "default" system. Specifically, we demonstrate that the module-specific topological features can not be captured by means of computing the corresponding global network parameters, suggesting a unique organization within each module. Finally, we identify several pivotal network connectors and paths (predominantly associated with the association and limbic/paralimbic cortex regions) that are vital for the global coordination of information flow over the whole network, and we find that their lesions (deletions) critically affect the stability and robustness of the brain functional system. Together, our results demonstrate the highly organized modular architecture and associated topological properties in the temporal and spatial brain functional networks of the human brain that underlie spontaneous neuronal dynamics, which provides important implications for our understanding of how intrinsically coherent spontaneous brain activity has evolved into an optimal neuronal architecture to support global computation and information integration in the absence of specific stimuli or behaviors.

Ultrahigh field fMRI reveals different roles of temporal and fronto-parietal cortices in subjective awareness

Abstract:A central question in consciousness theories is whether one is dealing with a dichotomous (“all-or-none”) or a gradual phenomenon. In this 7T fMRI study we investigated whether dichotomy or gradualness in fact depends on the brain region associated with perceptual awareness reports. Both male and female human subjects performed an emotion discrimination task (fear vs neutral bodies) presented under continuous flash suppression with trial-based perceptual awareness measures. Behaviorally, recognition sensitivity increased linearly with increased stimuli awareness and was at chance level during perceptual unawareness. Physiologically, threat stimuli triggered a slower heart rate than neutral ones during ‘almost clear’ stimulus experience, indicating freezing behavior. Brain results showed that activity in occipito-temporal, parietal and frontal regions as well as in amygdala increased with increased stimulus awareness while early visual areas showed the opposite pattern. The relationship between temporal area activity and perceptual awareness best fitted a gradual model while the activity in fronto-parietal areas fitted a dichotomous model. Furthermore, our findings demonstrate that specific experimental decisions, such as stimulus type or the approach used to evaluate awareness, play pivotal roles in consciousness studies and warrant careful consideration. Significance Statement The neural basis of consciousness is still highly debated as major theoretical and methodological questions on perceptual awareness and its assessment remain unsolved. Most studies have employed a dichotomous measure (yes/no, seen/unseen) to assess awareness and consequently found that consciousness is an “all-or-none” phenomenon. However, finer measures have revealed intermediate levels of awareness. It is still an open question whether affective signal processing, such as for body expressions, presents a gradual or a dichotomous relationship to perceptual awareness. We take up this challenge and show that perceptual awareness is gradual or dichotomous depending on the brain region considered, possibly reflecting different area functionality.

Abstract:The role of the prefrontal cortex (PFC) in consciousness is hotly debated. Frontal theories argue that the PFC is necessary for consciousness, while sensory theories propose that consciousness arises from recurrent activity in the posterior cortex alone, with activity in the PFC resulting from the mere act of reporting. To resolve this dispute, we re-analysed an EEG dataset of 30 participants from a no-report inattentional blindness paradigm where faces are (un)consciously perceived. Dynamic causal modelling was used to estimate the effective connectivity between the key contended brain regions, the prefrontal and the posterior cortices. Then, a second-level parametric empirical Bayesian model was conducted to determine how connectivity was modulated by awareness and task-relevance. While an initial data-driven search could not corroborate neither sensory nor frontal theories of consciousness, a more directed hypothesis-driven analysis revealed strong evidence that both theories could explain the data, with a very slight preference for frontal theories. Specifically, a model with backward connections switched off within the posterior cortex explained awareness better (53%) than a model without backward connections from the PFC to sensory regions. Our findings provide some support for a subtle, yet crucial, contribution of the frontal cortex in consciousness, and highlight the need to revise current theories of consciousness.

Propagating cortical waves coordinate sensory encoding and memory retrieval in the human brain

Abstract:Complex behavior entails a balance between taking in sensory information from the environment and utilizing previously learned internal information. Experiments in behaving mice have demonstrated that the brain continually alternates between outward and inward modes of cognition, switching its mode of operation every few seconds. Further, each state transition is marked by a stereotyped cascade of neuronal spiking that pervades most forebrain structures. Here we analyzed large fMRI datasets to demonstrate that a similar switching mechanism governs the operation of the human brain. We found that human brain activity was punctuated every several seconds by coherent, propagating waves emerging in the exteroceptive sensorimotor regions and terminating in the interoceptive default mode network. As in the mouse, the issuance of such events coincided with fluctuations in pupil size, indicating a tight relationship with arousal fluctuations, and this phenomenon occurred across behavioral states. Strikingly, concurrent measurement of human performance in a visual memory task indicated that each cycle of propagating fMRI waves sequentially promoted the encoding of semantic information and self-directed retrieval of memories. Together, these findings indicate that human cognitive performance is governed by autonomous switching between exteroceptive and interoceptive states. This apparently conserved feature of mammalian brain physiology bears directly on the integration of sensory and mnemonic information during everyday behavior.

Abstract:Our brain processes the different timescales of our environment's temporal input stochastics. Is such a temporal input processing mechanism key for consciousness? To address this research question, we calculated measures of input processing on shorter (alpha peak frequency, APF) and longer (autocorrelation window, ACW) timescales on resting-state high-density EEG (256 channels) recordings and compared them across different consciousness levels (awake/conscious, ketamine and sevoflurane anaesthesia, unresponsive wakefulness, minimally conscious state). We replicate and extend previous findings of: (i) significantly longer ACW values, consistently over all states of unconsciousness, as measured with ACW-0 (an unprecedented longer version of the well-know ACW-50); (ii) significantly slower APF values, as measured with frequency sliding, in all four unconscious states. Most importantly, we report a highly significant correlation of ACW-0 and APF in the conscious state, while their relationship is disrupted in the unconscious states. In sum, we demonstrate the relevance of the brain's capacity for input processing on shorter (APF) and longer (ACW) timescales - including their relationship - for consciousness. Albeit indirectly, e.g., through the analysis of electrophysiological activity at rest, this supports the mechanism of temporo-spatial alignment to the environment's temporal input stochastics, through relating different neural timescales, as one key predisposing factor of consciousness.

Abstract:Abstract Competing theories of consciousness disagree on the role of frontoparietal networks. Here we recorded neural activity from 727 intracerebral contacts in 13 epileptic patients, while they detected near-threshold targets preceded by supra-threshold exogenous cues. Trajectory k -means clustering revealed three patterns: (1) Validly cued seen targets elicit sustained activity in right-hemisphere fronto-temporal regions, connected by superior longitudinal fasciculus (SLF) II/III, and late accumulation activity in bilateral dorsolateral prefrontal cortex and in right-hemisphere orbitofrontal cortex (SLF I/III). (2) Invalidly cued seen targets elicit early, sustained activity in the right-hemisphere reorienting network (SLF III). (3) Seen targets elicit late sustained left dorsolateral prefrontal activity. Task modeling with recurrent neural networks supported the causal contribution of these networks to conscious perception, and elucidated their interplay through excitatory-inhibitory mechanisms. We provide new, compelling evidence on the role of hemisphere-asymmetric frontoparietal networks associated with attentional enhancement and reorienting in conscious perception.
