Abstract:SENSITIVE TO PROTON RHIZOTOXICITY 1 (STOP1) is a core transcription factor that regulates the expression of aluminum (Al) resistance genes to manage Al toxicity in plants. However, the genome-wide roles of SlSTOP1 in the Al stress response of tomato (Solanum lycopersicum) remain largely unknown. Here, we report that SlSTOP1 is crucial for Al tolerance in tomato, as loss-of-function mutants of SlSTOP1 displayed hypersensitivity to Al stress. Aluminum stress had no effect on SlSTOP1 mRNA expression, but promoted accumulation of SlSTOP1 protein in the nucleus. Through integrated DNA affinity purification sequencing and RNA sequencing analysis, we identified 39 SlSTOP1-targeted Al-responsive genes, some of which are homologous to known Al resistance genes in other plant species, suggesting that these SlSTOP1-targeted genes play essential roles in Al resistance in tomato. Furthermore, using peak enrichment analysis of SlSTOP1-targeted sequences, we identified a cis-acting element bound by SlSTOP1 and validated this finding via dual-luciferase reporter and electrophoretic mobility shift assay (EMSA). Additionally, we demonstrated SlHAK5 is one of direct targets of SlSTOP1 and functionally characterized it in terms of Al stress tolerance. Compared with wild-type plants, Slhak5 mutants developed by CRISPR/Cas9 technology presented increased sensitivity to Al stress, which was associated with reduced citrate secretion from the roots. Together, our findings demonstrate that SlSTOP1 directly interacts with cis-acting elements located in the promoters of target genes involved in diverse pathways contributing to Al resistance in tomato.
Genome-Wide Analysis and Expression of MYC Family Genes in Tomato and the Functional Identification of slmyc1 in Response to Salt and Drought Stress

Abstract:Myelocytomatosis (MYC) transcription factors are crucial mediators of the jasmonate signaling pathway, which mediates the growth and developmental processes of plants. However, the function of MYC genes in tomato, Solanum lycopersicum (SlMYC), remains poorly understood. In this study, we have identified 14 non-redundant SlMYC genes across the genome of tomatoes. Six of the twelve chromosomes included these genes, and four syntenic pairs of SlMYC were identified. According to the results of phylogenetic analysis, 14 SlMYC genes were clustered into classes I, II, III, and IV, and their functional domains were predicted. The SlMYC upstream promoter region contained a variety of light-, stress-, and hormone-response regulatory elements. The expression of the 14 SlMYC genes differed significantly across organs. SlMYCs primarily showed an upregulation trend after methyl jasmonate (MeJA) treatment. In contrast, after treatment with sodium chloride (NaCl), SlMYCs showed a trend of downregulation. However, there were differences in the expression patterns of SlMYCs after mannitol treatment. Using clustered regularly interspaced short palindromic repeats/Cas 9 (CRISPR/Cas 9) technology, the loss-of-function of SlMYC1 (slmyc1) was obtained. The slmyc1 tomato plants demonstrated reduced resistance to NaCl and mannitol stress compared to wild-type plants due to their shorter root length and higher reactive oxygen species (ROS) content. In brief, this study provides valuable information about the taxonomy of the SlMYC genes in tomato. It establishes a foundation for future research on the mechanism by which SlMYC influences plant development and stress response.

Abstract:Mitochondrial transcription termination factor (mTERF) is a DNA-binding protein that is encoded by nuclear genes, ultimately functions in mitochondria and can affect gene expression. By combining with mitochondrial nucleic acids, mTERF regulates the replication, transcription and translation of mitochondrial genes and plays an important role in the response of plants to abiotic stress. However, there are few studies on mTERF genes in tomato, which limits the in-depth study and utilization of mTERF family genes in tomato stress resistance regulation. In this study, a total of 28 mTERF gene family members were obtained through genome-wide mining and identification of the tomato mTERF gene family. Bioinformatics analysis showed that all members of the family contained environmental stress or hormone response elements. Gene expression pattern analysis showed that the selected genes had different responses to drought, high salt and low temperature stress. Most of the genes played key roles under drought and salt stress, and the response patterns were more similar. The VIGS method was used to silence the SLmTERF13 gene, which was significantly upregulated under drought and salt stress, and it was found that the resistance ability of silenced plants was decreased under both kinds of stress, indicating that the SLmTERF13 gene was involved in the regulation of the tomato abiotic stress response. These results provide important insights for further evolutionary studies and contribute to a better understanding of the role of the mTERF genes in tomato growth and development and abiotic stress response, which will ultimately play a role in future studies of tomato gene function.

Abstract:Hypertrophy development induced by the overexpression of SlbHLH22 (also called SlUPA-like) was susceptible to Xanthomonas in tomatoes. Transcriptome and metabolome analyses were performed on the hypertrophy leaves of a SlbHLH22-overexpressed line (OE) and wild type (WT) to investigate the molecular mechanism. Metabolome analysis revealed that six key metabolites were over-accumulated in the OE, including Acetylserine/O-Acetyl-L-serine, Glucono-1,5-lactone, Gluconate, 2-Oxoglutarate, and Loganate, implying that the OE plants increased salt or oxidant resistance under normal growth conditions. The RNA-seq analysis showed the changed expressions of downstream genes involved in high-energy consumption, photosynthesis, and transcription regulation in OE lines, and we hypothesized that these biological processes were related to the GTgamma subfamily of trihelix factors. The RT-PCR results showed that the expressions of the GTgamma genes in tomatoes, i.e., SlGT-7 and SlGT-36, were suppressed in the hypertrophy development. The expression of the GTgamma gene was downregulated by salinity, indicating a coordinated role of GTgamma in hypertrophy development and salt stress. Further research showed that both SlGT-7 and SlGT-36 were highly expressed in leaves and could be significantly induced by abscisic acid (ABA). The GTgamma protein had a putative phosphorylation site at S96. These results suggested GTgamma's role in hypertrophy development by increasing the salt resistance.
Downregulation of the mitogen-activated protein kinase SlMAPK7 gene results in pollen abortion in tomato

Abstract:Mitogen-activated protein kinases (MAPKs) play important roles in various aspects including plant development and stress response. The biological role of MAPKs in tomato development remains unclear. Here, multiple stable transgenetic lines were developed using either gene overexpression or RNA interference (RNAi) technology to investigate the role of SlMAPK7 in tomato reproductive development. Either upregulation or downregulation of SlMAPK7 did not drastically disturb the vegetative growth of tomato. However, in RNAi-based gene silencing plants, approximately 90 % of the pollen grains were identified to be abnormal and lack viability, which were malformed or collapsed. Cytological observations showed that anthers could develop normal microspore mother cells, but the uninucleate microspores failed to form normal mature pollen grains. Transmission electron microscopy examination demonstrated that accelerated degradation of tapetum occurred in the anther and finally the intine layer of pollen appears abnormal. In addition, although inhibition of SlMAPK7 gene did not alter fruit morphology, seed number in RNAi fruits was markedly reduced. Thus, the results demonstrated that SlMAPK7 is involved in tomato pollen development, possibly via regulation the degradation of tapetum.

Abstract:Abstract Background Leaves, which are the most important organs of plants, can not only fix carbon sources through photosynthesis, but also absorb nutrients through transpiration. Leaf development directly determines the growth, flowering and fruiting of plants. There are many factors that affect leaf development, such as the growth environment, gene expression, and hormone synthesis. In this study, tomatoes were used to study the role of the transcription factor Solanum lycopersicum salt-related MYB1-like (SlSRM1-like) in the development of tomato leaves. Results Loss-of-function of the SlSRM1-like gene mediated by clustered, regularly interspaced, short palindromic repeat (CRISPR)/CRISPR-associated 9 (Cas9) resulted in abnormal tomato leaf morphology, including thinner leaves, wrinkled edges, raised veins, disordered edge veins, and left and right asymmetry. An analysis of the transcription levels of genes related to leaf development revealed that the expression of these genes was significantly altered in the SlSRM1-like mutants ( SlSRM1-like -Ms). Moreover, the SlSRM1-like gene was expressed at higher transcription levels in young tissues than in old tissues, and its expression was also induced in response to auxin. In addition, the transcription levels of genes related to the auxin pathway, which regulates tomato growth and development, were severely affected in the SlSRM1-like -Ms. Therefore, it is hypothesized that the SlSRM1-like gene functions in the regulation of tomato leaf development through the auxin-related pathway. Conclusions In this study, we successfully knocked out the SlSRM1-like gene in the tomato variety Ailsa Craig using CRISPR technology and found that knockout of the SlSRM1-like gene resulted in abnormal development of tomato leaves. Further research indicated that SlSRM1-like regulated tomato leaf development through auxin-related pathways. The results provide an important reference for the functional study of other SRM1-like genes in plants and provide new insights into the regulation of leaf development in tomato and other plants.

Abstract:Calmodulin-like (CML) proteins are primary calcium sensors and function in plant growth and response to stress stimuli. However, so far, the function of plant CML proteins, including tomato, is still unclear. Previously, it was found that a tomato (Solanum lycopersicum) CML, here named SlCML39, was significantly induced by high temperature (HT) at transcription level, but its biological function is scarce. In this study, the characteristics of SlCML39 and its role in HT tolerance were studied. SlCML39 encodes a protein of 201 amino acids containing four EF hand motifs. Many cis-acting elements related to plant stress and hormone response appear in the promoter regions of SlCML39. SlCML39 is mainly expressed in the root, stem, and leaf and can be regulated by HT, cold, drought, and salt stresses as well as ABA and H2O2. Furthermore, heterologous overexpression of SlCML39 reduces HT tolerance in Arabidopsis thaliana at the germination and seedling growth stages. To better understand the molecular mechanism of SlCML39, the downstream gene network regulated by SlCML39 under HT was analyzed by RNA-Seq. Interestingly, we found that many genes involved in stress responses as well as ABA signal pathway are down-regulated in the transgenic seedlings under HT stress, such as KIN1, RD29B, RD26, and MAP3K18. Collectively, these data indicate that SlCML39 acts as an important negative regulator in response to HT stress, which might be mediated by the ABA signal pathway.

Down-regulation of SlGT-26 gene confers dwarf plants and enhances drought and salt stress resistance in tomato

Abstract:Plant architecture, an important agronomic trait closely associated with yield, is governed by a highly intricate molecular network. Despite extensive research, many mysteries surrounding this regulation remain unresolved. Trihelix transcription factor family plays a crucial role in the development of plant morphology and abiotic stresses. Here, we identified a novel trihelix transcription factor named SlGT-26, and its down-regulation led to significant alterations in plant architecture, including dwarfing, reduced internode length, smaller leaves, and shorter petioles. The dwarf phenotype of SlGT-26 silenced transgenic plants could be recovered after spraying exogenous GA3, and the GA3 content were decreased in the RNAi plants. Additionally, the expression levels of gibberellin-related genes were affected in the RNAi lines. These results indicate that the dwarf of SlGT-26-RNAi plants may be a kind of GA3-sensitive dwarf. SlGT-26 was response to drought and salt stress treatments. SlGT-26-RNAi transgenic plants demonstrated significantly enhanced drought resistance and salt tolerance in comparison to their wild-type tomato counterparts. SlGT-26-RNAi transgenic plants grew better, had higher relative water content and lower MDA and H2O2 contents. The expression of multiple stress-related genes was also up-regulated. In summary, we have discovered a novel gene, SlGT-26, which plays a crucial role in regulating plant architecture and in respond to drought and salt stress.
A glutathione S-transferase regulates lignin biosynthesis and enhances salt tolerance in tomato

Abstract:Salt stress adversely affects the growth and yield of crops. Glutathione S-transferases (GSTs) are involved in plant growth and responses to biotic and abiotic stresses. In this study, 400 mM NaCl stress significantly induced the expression of Glutathione S-transferase U43 (SlGSTU43) in the roots of the wild-type tomato (Solanum lycopersicum L.) plants. Overexpressing SlGSTU43 enhanced the ability of scavenging reactive oxygen species (ROS) in tomato leaves and roots under NaCl stress, while SlGSTU43 knock-out mutants showed the opposite performance. RNA sequencing analysis revealed that overexpressing SlGSTU43 affected the expression of genes related to lignin biosynthesis. We demonstrated that SlGSTU43 can regulate the lignin content in tomato through its interaction with SlCOMT2, a key enzyme involved in lignin biosynthesis, and promote the growth of tomato plants under NaCl stress. In addition, SlMYB71 and SlWRKY8 interact each other, and can directly bind to the promoter of SlGSTU43 to transcriptionally activate its expression separately or in combination. When SlMYB71 and SlWRKY8 were silenced in tomato plants individually or collectively, the plants were sensitive to NaCl stress, and their GST activities and lignin contents decreased. Our research indicates that SlGSTU43 can enhance salt stress tolerance in tomato by regulating lignin biosynthesis, which is regulated by interacting with SlCOMT2, as well as SlMYB71 and SlWRKY8. This finding broadens our understanding of GST functions.

Abstract:Salt stress is a major abiotic stress that severely hinders crop production. However, the regulatory network controlling tomato resistance to salt remains unclear. Here, we found that the tomato WRKY transcription factor WRKY57 acted as a negative regulator in the salt stress response by directly attenuating the transcription of salt‐responsive genes (SlRD29B and SlDREB2) and an ion homeostasis gene (SlSOS1). We further identified two VQ‐motif containing proteins SlVQ16 and SlVQ21 as SlWRKY57‐interacting proteins. SlVQ16 positively, while SlVQ21 negatively modulated tomato resistance to salt stress. SlVQ16 and SlVQ21 competitively interacted with SlWRKY57 and antagonistically regulated the transcriptional repression activity of SlWRKY57. Additionally, the SlWRKY57‐SlVQ21/SlVQ16 module was involved in the pathway of phytohormone jasmonates (JAs) by interacting with JA repressors JA‐ZIM domain (JAZ) proteins. Silencing of SlMYB78-like Reduces the Tolerance to Drought and Salt Stress via the ABA Pathway in Tomato

Abstract:The MYB transcription factor family plays a crucial regulatory role in plant growth, development, biological progress, and stress responses. Here, we identified a R2R3-MYB transcription factor gene, SlMYB78-like, from tomato and characterized its function by gene silencing via RNA interference (RNAi). The results exhibited that the silencing of SlMYB78-like reduced the sensitivity of tomato seedlings to exogenous ABA. In addition, when exposed to drought and salt stresses, the RNAi lines grown in soil showed decreased tolerance, with lower ABA accumulation, relative water content, and chlorophyll content while displaying higher relative conductivity and malondialdehyde (MDA) content than the wild type. Moreover, the expression of genes related to chlorophyll biosynthesis, photosynthesis, and ABA biosynthesis/response were down-regulated in SlMYB78-like-silenced lines. Notably, the transcript level of SlCYP707-A2, which encodes a protein involved in ABA degradation, was up-regulated significantly after stresses. The transient expression assay Dual-luciferase (Dual-LUC) and a yeast one-hybrid (Y1H) assay demonstrated that SlMYB78-like bound to the promoter of SlCYP707-A2. Additionally, the physical interaction between SlMYB78-like and SlDREB3, which functioned in ABA signaling transduction, was identified through yeast two-hybrid (Y2H) and bimolecular fluorescence complementation (BiFC) assays. Collectively, our study illustrates that SlMYB78-like participates in the abiotic stress response via the ABA pathway.

Abstract:Salt stress can seriously reduce crop yield and quality, causing huge economic losses. Identification and functional of salt stress regulator could provide us a way to improve plants tolerance to salt stress. The HD-ZIP II subfamily is plant-specific transcription factors, plays a crucial role in plant development and abiotic stress. In this study, we analyzed the expression profiling of all HD-ZIP II members response to salt, drought, high temperature and cold stress using publicly accessible transcriptome data, and identified a novel salt stress negative regulator SlABIG1 in tomato. Under salt stress, knockout of SlABIG1 significantly improved the salt tolerance of tomato, with higher chlorophyll content and photosynthetic capacity, root dry weight and proline; decreased the accumulation of ROS, MDA and Na+. Collectively, our results revealed that SlABIG1 plays a key role in tomato tolerance to salinity stress and provide a candidate novel gene to create salt-tolerant tomato varieties in future.

Abstract:Choline monooxygenase (CMO) is a key enzyme involved in betaine synthesis and our preliminary work has shown that the SlCMO gene promoter (pC5: − 267 to + 128 base pair), cloned from Suaeda liaotungensis, is salt-inducible. In the present study, pC5-SlCMO was transferred into tomato (Solanum lycopersicon L. ‘Micro-Tom’) plants via Agrobacterium mediation. Homozygous transgenic plants were selected using quantitative real-time polymerase chain reaction. The expression of SlCMO in pC5-SlCMO transgenic plants was induced by salinity. Under salt tolerance, betaine content, chlorophyll content, and net photosynthetic rate were higher in transgenic plants than in wild-type (WT) plants. Proline content was lower in transgenic plants than in WT plants. Under normal conditions, seed germination, length of the whole plant, dry weight, and fruit products of transgenic plants were the same as in WT plants. These results demonstrated that the pC5 promoter can drive increased expression of SlCMO in transgenic tomato plants under salt stress and increase salt tolerance without affecting plant growth and yield.

Abstract:Auxin controls multiple aspects of plant growth and development. However, its role in stress responses remains poorly understood. Auxin acts on the transcriptional regulation of target genes, mainly through Auxin Response Factors (ARF). This study focuses on the involvement of SlARF4 in tomato tolerance to salinity and osmotic stress. Using a reverse genetic approach, we found that the antisense down-regulation of SlARF4 promotes root development and density, increases soluble sugars content and maintains chlorophyll content at high levels under stress conditions. Furthermore, ARF4-as displayed higher tolerance to salt and osmotic stress through reduced stomatal conductance coupled with increased leaf relative water content and Abscisic acid (ABA) content under normal and stressful conditions. This increase in ABA content was correlated with the activation of ABA biosynthesis genes and the repression of ABA catabolism genes. Cu/ZnSOD and mdhar genes were up-regulated in ARF4-as plants which can result in a better tolerance to salt and osmotic stress. A CRISPR/Cas9 induced SlARF4 mutant showed similar growth and stomatal responses as ARF4-as plants, which suggest that arf4-cr can tolerate salt and osmotic stresses. Our data support the involvement of ARF4 as a key factor in tomato tolerance to salt and osmotic stresses and confirm the use of CRISPR technology as an efficient tool for functional reverse genetics studies.

Mitogen-Activated Protein Kinase Is Involved in Salt Stress Response in Tomato (Solanum lycopersicum) Seedlings

Abstract:Salt stress impairs plant growth and development, thereby causing low yield and inferior quality of crops. In this study, tomato (Solanum lycopersicum L. 'Micro-Tom') seedlings treated with different concentrations of sodium chloride (NaCl) were investigated in terms of decreased plant height, stem diameter, dry weight, fresh weight, leaves relative water content and root activity. To reveal the response mechanism of tomato seedlings to salt stress, the transcriptome of tomato leaves was conducted. A total of 6589 differentially expressed genes (DEGs) were identified and classified into different metabolic pathways, especially photosynthesis, carbon metabolism, biosynthesis of amino acids and mitogen-activated protein kinase (MAPK) signaling pathway. Of these, approximately 42 DEGs were enriched in the MAPK signaling pathway, most of which mainly included plant hormone, hydrogen peroxide (H2O2), wounding and pathogen infection signaling pathways. To further explore the roles of MAPK under salt stress, MAPK phosphorylation inhibitor SB203580 (SB) was applied. We found that SB further decreased endogenous jasmonic acid, abscisic acid and ethylene levels under salt stress condition. Additionally, in comparison with NaCl treatment alone, SB + NaCl treatment reduced the content of O2- and H2O2 and the activities of antioxidant enzyme and downregulated the expression levels of genes related to pathogen infection. Together, the results revealed that MAPK might be involved in the salinity response of tomato seedlings by regulating hormone balance, ROS metabolism, antioxidant capacity and plant immunity.
Tomato SR/CAMTA Transcription Factors SlSR1 and SlSR3L Negatively Regulate Disease Resistance Response and SlSR1L Positively Modulates Drought Stress Tolerance.

Abstract:BACKGROUND:The SR/CAMTA proteins represent a small family of transcription activators that play important roles in plant responses to biotic and abiotic stresses. Seven SlSR/CAMTA genes were identified in tomato as tomato counterparts of SR/CAMTA; however, the involvement of SlSRs/CAMTAs in biotic and abiotic stress responses is not clear. In this study, we performed functional analysis of the SlSR/CAMTA family for their possible functions in defense response against pathogens and tolerance to drought stress.RESULTS:Expression of SlSRs was induced with distinct patterns by Botrytis cinerea and Pseudomonas syringae pv. tomato (Pst) DC3000. Virus-induced gene silencing (VIGS)-based knockdown of either SlSR1 or SlSR3L in tomato resulted in enhanced resistance to B. cinerea and Pst DC3000 and led to constitutive accumulation of H2O2, elevated expression of defense genes, marker genes for pathogen-associated molecular pattern-triggered immunity, and regulatory genes involved in the salicylic acid- and ethylene-mediated signaling pathways. Furthermore, the expression of SlSR1L and SlSR2L in detached leaves and whole plants was significantly induced by drought stress. Silencing of SlSR1L led to decreased drought stress tolerance, accelerated water loss in leaves, reduced root biomass and attenuated expression of drought stress responsive genes in tomato. The SlSR1 and SlSR3L proteins were localized in the nucleus of plant cells when transiently expressed in Nicotiana benthamiana and had transcriptional activation activity in yeast.CONCLUSIONS:VIGS-based functional analyses demonstrate that both SlSR1 and SlSR3L in the tomato SlSR/CAMTA family are negative regulators of defense response against B. cinerea and Pst DC3000 while SlSR1L is a positive regulator of drought stress tolerance in tomato.

Abstract:DNA methylation plays an important role in regulating plant growth, development and gene expression. However, less is known about the response of DNA methyltransferase and demethylase genes to various stresses. In this study, the effects of abiotic stresses on DNA methylation gene expression patterns in tomato seedlings were investigated. Results showed that most tomato DNA methyltransferase and demethylase genes contained stress-related elements. The expression of SlDML1 was significantly induced by cadmium (Cd) and sodium chloride (NaCl) stresses. SlDML2 was more sensitive and reached its maximum value under polyethylene (PEG) stress at 24 h. The expression of SlMET3L was repressed to varying degrees under Cd, NaCl and PEG stresses at 48 h. However, 5-aza-2'-deoxycytidine (5-azadC) treatment decreased the Cd and PEG stress tolerance by down-regulating the expression of DNA methyltransferase except for the SlMET3L , and up-regulating the expression levels of SlDML2 , SlDML3 and SlDML4 , cadmium transporters ( SlHMA5 , SlCAX3 , and SlACC3 ) and osmoregulators ( SlDREB , SlLEA and SlHSP70 ). Whereas 5-azadC treatment alleviated the salt stress through up-regulating DNA methyltransferase gene expression, and down-regulating the expression level of SlDML1 , SlDML3 , and SlDML4 , SlHKT1 , SlNHX1 , and SlSOS1 . Collectively, 5-azadC impaired Cd and PEG stress tolerance and enhanced salt stress tolerance by regulating the expression of methylation-related and stress-related genes in tomato seedlings. These results may provide useful information for further analysing function and evolution of DNA methylation methyltransferase and demethylase genes in tomato under stress conditions.

Abstract:Potassium deficiency confines root growth and decreases root-to-shoot ratio, thereby limiting root K+ acquisition. This study aimed to identify the regulation network of microRNA319 involved in low-K+ stress tolerance in tomato (Solanum lycopersicum). SlmiR319b-OE roots demonstrated a smaller root system, a lower number of root hairs and lower K+ content under low-K+ stress. We identified SlTCP10 as the target of miR319b using a modified RLM-RACE procedure from some SlTCPs’ predictive complementarity to miR319b. Then, SlTCP10-regulated SlJA2 (an NAC transcription factor) influenced the response to low-K+ stress. CR-SlJA2 (CRISPR-Cas9-SlJA2) lines showed the same root phenotype to SlmiR319-OE compared with WT lines. OE-SlJA2(Overexpression-SlJA2) lines showed higher root biomass, root hair number and K+ concentration in the roots under low-K+ conditions. Furthermore, SlJA2 has been reported to promote abscisic acid (ABA) biosynthesis. Therefore, SlJA2 increases low-K+ tolerance via ABA. In conclusion, enlarging root growth and K+ absorption by the expression of SlmiR319b-regulated SlTCP10, mediating SlJA2 in roots, could provide a new regulation mechanism for increasing K+ acquisition efficiency under low-K+ stress.

Abstract:The synthesis of the phytoprostane B1 types I and II is achieved in high overall yield (35-53%) by only two principal transformations starting from 1,3-cyclopentanedione. The first side chain is attached via O-acylation of the 1,3-dione followed by rearrangement and reduction to give the 2-alkyl-1,3-diones 4a-c. After conversion into the corresponding vinylic iodides 5a-c, the second side chain is introduced by transition metal catalysis following Heck- or Sonogashira-type protocols. The whole spectrum of the phytoprostane B1 types I, II, and the dinor isoprostane B1 type III and some structural analogs are rapidly accessible along the same general protocol.
