Abstract:Multi-isotope and multi-elemental analyses were performed on 600 red wine samples imported into China from 7 different countries and compared with Chinese wine. Carbon and oxygen isotopes and 16 elements were used to determine origin traceability. Our goal was to build a classification tool using data modeling that can verify the geographic origin of wines imported into China. Multivariate analyses of the isotopic and elemental data revealed that it is possible to determine the geographical origin for most imported wines with a high level of confidence (>90%). The results show that Artificial Neural Network method had a high discrimination accuracy and is more suitable than Discrimination Analysis and Random Forest methods when it comes to classifying wine origin on a global scale. In conclusion, stable isotope and trace element analyses followed by multivariate processing of the data is a fast and efficient technique suitable for global wine traceability.

Abstract:This work was undertaken to evaluate whether it is possible to determine the variety of a Chinese wine on the basis of its volatile compounds, and to investigate if discrimination models could be developed with the experimental wines that could be used for the commercial ones. A headspace solid-phase microextraction gas chromatographic (HS-SPME-GC) procedure was used to determine the volatile compounds and a blind analysis based on Ac/Ais (peak area of volatile compound/peak area of internal standard) was carried out for statistical purposes. One way analysis of variance (ANOVA), principal component analysis (PCA) and stepwise linear discriminant analysis (SLDA) were used to process data and to develop discriminant models. Only 11 peaks enabled to differentiate and classify the experimental wines. SLDA allowed 100% recognition ability for three grape varieties, 100% prediction ability for Cabernet Sauvignon and Cabernet Gernischt wines, but only 92.31% for Merlot wines. A more valid and robust way was to use the PCA scores to do the discriminant analysis. When we performed SLDA this way, 100% recognition ability and 100% prediction ability were obtained. At last, 11 peaks which selected by SLDA from raw analysis set had been identified. When we demonstrated the models using commercial wines, the models showed 100% recognition ability for the wines collected directly from winery and without ageing, but only 65% for the others. Therefore, the varietal factor was currently discredited as a differentiating parameter for commercial wines in China. Nevertheless, this method could be applied as a screening tool and as a complement to other methods for grape base liquors which do not need ageing and blending procedures.
Hydrogen and Oxygen Isotope Fractionation Effects in Different Organ Tissues of Grapes under Drought Conditions

Abstract:A study of different grapevine tissues and organs (root, stem, leaf, fruit) water isotope fractionation models from high-quality wine grapes produced in the Helan Mountains, a key wine-producing area in northwestern China, was undertaken. Results showed that δ2H values of local groundwater sources were more negative than rivers and precipitation. Soil water δ2H and δ18O values were significantly higher than those of other environmental water sources. Water from the soil surface layer (0-30 cm, δ2H and δ18O values) was more positive than the deeper layer (30-60 cm), indicating that soil water has undergone a positive fractionation effect. δ2H and δ18O values of tissues and organs from different grape varieties followed a similar pattern but were more negative than the local atmospheric precipitation line (slope between 4.1 to 5.2). The 2H and 18O fractionation relationship in grapevine organs was similar, and 18O has a higher fractionation effect than 2H. δ2H and δ18O values showed a strong fractionation effect during the transportation of water to different grape organs (trend of stem > fruit > leaf). This study showed that 18/16O fractionation in grapes is more likely to occur under drought conditions and provides a theoretical basis to improve traceability accuracy and origin protection of wine production areas.

Abstract:A method was developed for analyzing the stable carbon isotope ratio of five volatile components (Ethanol, Glycerol, Acetic acid, Ethyl lactate, 2-methyl-butanol) in wine using gas chromatography- combustion. isotope ratio mass spectrometer (GC-C-IRMS). The sample injection volume was less than 0. 5 mu L, and the analytical time of each run was less than 14 min. The precision of this method was 0. 08 parts per thousand-0. 25 parts per thousand for analyzing standards, while 0. 09 parts per thousand-0. 36 parts per thousand for wine samples. Compared to element analysis-isotope ratio mass spectrometry (EA-IRMS) results, the deviations were lower than 0. 5 parts per thousand. Fifty-four wine samples from France, Australia, America and China were considered. The delta C-13 of five volatile components were measured using GC-C-IRMS. Discriminant analysis (DA) was employed for analyzing the geographical origin traceability of selected wine. The result indicated that delta C-13 of volatile components could be used to distinguish the origin of wines. The method was shown to be effective in improving detection of the origin traceability of wine.
Geographical identification of Chinese wine based on chemometrics combined with mineral elements, volatile components and untargeted metabonomics

Abstract:Identifying the geographic origin of a wine is of great importance, as origin fakery is commonplace in the wine industry. This study analyzed the mineral elements, volatile components, and metabolites in wine using inductively coupled plasma-mass spectrometry, headspace solid phase microextraction gas chromatography-mass spectrometry, and ultra-high-performance liquid chromatography-quadrupole-exactive orbitrap mass spectrometry. The most critical variables (5 mineral elements, 13 volatile components, and 51 metabolites) for wine origin classification were selected via principal component analysis and orthogonal partial least squares discriminant analysis. Subsequently, three algorithms-K-nearest neighbors, support vector machine, and random forest -were used to model single and fused datasets for origin identification. These results indicated that fused datasets, based on feature variables (mineral elements, volatile components, and metabolites), achieved the best performance, with predictive rates of 100% for all three algorithms. This study demonstrates the effectiveness of a multi-source data fusion strategy for authenticity identification of Chinese wine.

Abstract:The demand for the effective traceability of hazardous chemicals is crucial for preventing and controlling chemical spills and other accidents involving hazardous chemicals. The aim of the study was to investigate the correlation between the geographical location of ethanol-producing industrial sites and the carbon, hydrogen, and oxygen stable isotope ratios of the Chinese-manufactured ethanol using statistical classification analysis to enable the traceability of the ethanol. The isotopic data of 54 ethanol samples obtained from 18 different ethanol manufacturing plants in China between 2019 and 2020. The results of the statistical analysis demonstrated that the δ18O values of the ethanol positively correlated with latitudes of the production plants but negatively correlated with the δ13C values of the ethanol. A small number of samples derived from sites that were geographically close to each other could not be visually distinguished by PCA and HCA. However, by applying and comparing the results of classification by LDA, K-NN and Ensemble, an optimal classification model was obtained. Upon application of these models, 96.3% of the ethanol samples were correctly classified based on their geographical origin, indicating that the combination of isotopic ratios and latitude data is practical and effective for measuring the traceability of ethanol.

Abstract:Discrimination of Chinese rice wines from three well-known wineries ("Guyuelongshan", "Kuaijishan", and "Pagoda") in China has been carried out according to mineral element contents in this study. Nineteen macro and trace mineral elements (Na, Mg, Al, K, Ca, Mn, Fe, Cu, Zn, V, Cr, Co, Ni, As, Se, Mo, Cd, Ba and Pb) were determined by inductively coupled plasma mass spectrometry (ICP-MS) in 117 samples. Then the experimental data were subjected to analysis of variance (ANOVA) and principal component analysis (PCA) to reveal significant differences and potential patterns between samples. Stepwise linear discriminant analysis (LDA) and partial least square discriminant analysis (PLS-DA) were applied to develop classification models and achieved correct classified rates of 100% and 97.4% for the prediction sample set, respectively. The discrimination could be attributed to different raw materials (mainly water) and elaboration processes employed. The results indicate that the element compositions combined with multivariate analysis can be used as fingerprinting techniques to protect prestigious wineries and enable the authenticity of Chinese rice wine.

Abstract:The objective of this study was to investigate the geographical origin of Chinese teas using carbon and nitrogen stable isotope ratio technology. The results showed that inter-provincial dispersion of teas in Guangdong (GD), Guangxi (GX), Hainan (HA), Fujian (FJ), Shandong (SD), Sichuan (SC), Chongqing (CQ), and Henan (HN) provinces was high, while in Zhejiang (ZJ), Hubei (HB), Yunnan (YN), and Anhui (AH) provinces, it was low. Tea samples from GD, GX, HA, and FJ provinces were clustered in one group and separated from those from AH and HB provinces. Thus, carbon and nitrogen stable isotope ratio technology could discriminate teas from among some provinces of China, but not from among others. Better separation might be obtained with a combination of isotopic ratios and other indexes, such as elemental data and organic components.

Abstract:Authentication of geographical traceability is essential to the safety of traditional Chinese medicines (TCMs). Aconiti Lateralis Radix Praeparata (Chinese: Fuzi), a well-known TCMs, the therapeutic efficacy and toxicity are clearly correlated with its geographical origin, and a rapid and effective method of origin tracing must be established. Herein, an elemental analysis-isotope ratio mass spectrometry (EA-IRMS) combined with a machine learning model is constructed for the origin traceability of Fuzi. We analyzed stable isotope information (δ13C‰, δ15N‰, δ2H‰, δ18O‰, C% and N%) for a total of 243 samples from eight major producing areas. Classification models were built using seven machine learning techniques, including random forest (RF), support vector machine (SVM), AdaBoost, etc., to investigate the feasibility of stable isotopes in Fuzi geographic identification. Results show that stable isotopes exhibit an overwhelming origin discrimination advantage. Among them, the RF model showed the best recognition performance with 94.2 % accuracy, 95.45 % sensitivity, and 99.15 % specificity in the independent test set, respectively. The results show that stable isotopes combined with machine learning techniques have great potential for the origin traceability of TCMs.

Abstract:Verification of the origin of 116 concentrated apple juice (CAJ) samples from three production areas (BHB, SCH and LP) of China was investigated in a trial using stable isotopic (δ 13C, δ 18O and δD) and the contents of 26 mineral elements. Our aim was to identify and differentiate the geographical origin of these CAJ samples, and more importantly to establish a model that can be used to verify geographical origin of Chinese CAJ samples, as to date there is no such study. Combining with the multivariate statistical analyses including principal component analysis (PCA) and partial least squared-discriminant analysis (PLS-DA), the predictive models were successfully established to discriminate diverse CAJ producing areas with an accuracy rate of 93.2 %. The most important variables to discriminate CAJ of Chinese origin were turned out to be δD, Mg, Tl, δ 18O, Mo, Sr, V, As, and δ 13C. Therefore, the results of this first exploratory work highlight the possibility of discriminating CAJ of different provenance. This work proposes the use of combined analyses related to stable isotopes and minerals elements as markers for CAJ certification. Our study helps to fill in the gap of origin identification of CAJ in China.

Differentiation between Organic and Non-Organic Wines from Helan Mountain East Region Based on Elemental Contents and Stable Isotope Ratios

Abstract:In this study, we analyzed elemental contents and stable isotope ratios of carbon (C) and nitrogen (N) in organic and non-organic wines from Helan mountain east region in Ningxia of China and then used the obtained data to distinguish between the two wines. Our results showed that there were significant differences in the contents of several metal elements including Na, Mg, K, Al and Cu between the organic and non-organic wines and that the δ13C ratios of ethyl alcohol and glycerol in the organic wine were much lower than those in the non-organic one, but the N stable isotope ratios were significantly higher than those in the non-organic one. Thus, C and N stable isotopes and the contents of selected metal elements could be used to effectively distinguish between organic and non-organic wines by principal component analysis. This method has a great potential to be applied for organic wine authentication and therefore can provide a technical support for the development of the wine industry in this region. © 2017, China Food Publishing Company. Abstract:Discrimination of Chinese rice wines from four geographical origins ('Zhejiang', 'Jiangsu', 'Shanghai' and Fujian) in China was investigated according to their UV-vis spectra. The UV-vis absorption spectra of 112 samples in the wavelength range of 190-800 nm were collected. Then the absorption data was subjected to principal component analysis to reveal differences between samples and the potential possibility of using multivariate methods to distinguish differences. Classification models were developed by soft independent modelling of class analogy, linear discriminate analysis (LDA), discriminant partial least squares and support vector machine. Seven mathematical pre-treatments were applied to improve the performance of the multivariate classification models. Among the seven pre-treatments, standard normal transformation (SNV) was superior to the other six methods. The results showed that, compared with other models, SNV-treated LDA models achieved better performances with an average correct classification rate of 98.96% in the training set and 100% in the testing set. The results demonstrate that UV spectroscopy combined with chemometric data analysis, as a rapid method to classify Chinese rice wines according to their geographical origins, is feasible. Abstract:The authenticity of the geographical origin of grapes and wines is vital for ensuring food safety in the wine market. To authenticate the geographical origin of Ecolly grapes and wines from different Chinese regions, forty-six elements were determined by inductively coupled plasma mass spectrometry (ICP-MS). Moreover, the relationships of element content between grapes and wines and regional differences in element profile were analyzed by multivariate statistics. K, Na, Cs, V, Li, Sc, In, Mn, Mg, Al, and Li, Na, Mg, K, Ca, Mn, Fe, Ni, Cu, Zn were important characteristic variables to distinguish grape and wine origins based on chi-squared tests, respectively. A feed-forward neural network (FNN), random forest (RF), and support vector machine (SVM) exhibited 98.33 %, 96.67 %, and 100 % accuracy for distinguishing the geographical origin of Ecolly grapes using these variables, respectively. All models obtained 100 % accuracy for distinguishing wines. FNN exhibited superior performance compared with RF and SVM according to a comprehensive evaluation based on the overall accuracy, the receiver operating characteristic curve (ROC), an area under the curve (AUC), and tenfold cross-validation. Consequently, this study provides powerful evidence of the potential application of ICP-MS coupled with machine learning models to discriminate the geographical origin of Chinese grapes and wines.

Preliminary study on the geographical origin of Chinese 'Cuiguan' pears using integrated stable isotope and multi-element analyses

Abstract:Distinguish the geographical origin of the pear is important due to the increasingly valued brand protection and reducing the potential food safety risks. In this study, the profiles of stable isotopes (δ13C, δ15N, δ2H, δ18O) and the contents of 16 elements in pear peer from four production areas were analyzed. The δ13C, δ15N, δ2H, δ18O and 12 elements were significantly different (p < 0.05) in the four production areas. Chemometrics analysis including principal component analysis (PCA), orthogonal partial least squares discriminant analysis (OPLS-DA) and linear discriminant analysis (LDA) were exploited for geographical origin classification of samples. OPLS-DA analysis showed that crucial variables (δ13C, δ18O, δ2H, Ni, Cd, Ca, δ15N, Sr and Ga) are more relevant for the discrimination of the samples. OPLS-DA achieved pear origin accuracy rates of 87.76 % by combining stable isotope ratios and elemental contents. LDA had a higher accuracy rate than OPLS-DA, and the LDA analysis showed that the original discrimination rate reached to 100 %, while the cross-validated rate reached to 95.7 %. These studies indicated that this method could be used to assess the geographical discrimination of pear from different producing areas and could potentially control the fair trade of pear in fruit markets.
Discrimination between Chinese rice wines of different geographical origins by NIRS and AAS

Abstract:The feasibility of near infrared spectroscopy (NIRS) for discrimination between Chinese rice wine of different geographical origins (Shaoxing and Jiashan, China) is presented in this research. NIR spectra were collected in transmission mode in the wavelength range of 800–2500 nm. Qualitative analysis models were developed based on partial least squares regression (PLSR). The prediction performance of calibration models in different wavelength range was also investigated. The best models gave a 100% classification of wines of the two geographical origins in the range of 1300–1650 nm. The content of trace metals (potassium, magnesium, zinc, and iron) was also investigated to classify wines of the two categories by atomic absorption spectroscopy (AAS). The AAS results were in agreement with NIRS, with 100% classification for wines of the two categories. In addition, the correlation between NIRS and AAS was also investigated by PLSR. Potassium and magnesium were well predicted by quantitative models based on NIR spectra and AAS data. The correlation coefficient of calibration ( R cal ) for potassium and magnesium were 0.958 and 0.885, respectively, and the correlation coefficient of validation ( R val ) were 0.861 and 0.700, respectively. The results demonstrated that NIRS technique could be used as a rapid method for classification of geographical origin of Chinese rice wine, and AAS could be used as an alternative technique or to validate the discrimination results.

Abstract:Huangjing (Polygonati Rhizoma) is a high-value homology of medicine and food in China, for which the geographical origins (i.e., geoherbalism) and farming patterns (natural or farmed) are decisive in its functions and selling price. In this study, five stable isotope ratios (δ13C, δ15N, δ18O, δ2H, and 87Sr/86Sr) and 48 multi-element signatures of 238 Huangjing samples collected from seven main production areas in China were analyzed together for origin traceability and farming authentication purposes. An analysis of variance (ANOVA) for each variable revealed that δ18O, δ2H, 87Sr/86Sr, Fe, and rare earth elements were significantly different among origins. The orthogonal partial least squares discriminant analysis (OPLS-DA) modeling of all variables indicated that K, Ca, Mg, Fe, and Mn were the most important variables (VIP > 1) for identifying Qingyang Huangjing protected geographical indication (PGI) from those of other origins, and discriminant accuracy reached 100%. Moreover, ANOVA and OPLS-DA demonstrated that δ15N, K, Ca, and Mg were the elements that effectively distinguished natural and farmed Huangjing. Therefore, the strategy may be an important alternative tool for authenticity management, which is significant for combating origin mislabeling and the fraudulent conducts of Huangjing products.

[Determination of wine original regions using information fusion of NIR and MIR spectroscopy]

Abstract:Geographical origins of wine grapes are significant factors affecting wine quality and wine prices. Tasters' evaluation is a good method but has some limitations. It is important to discriminate different wine original regions quickly and accurately. The present paper proposed a method to determine wine original regions based on Bayesian information fusion that fused near-infrared (NIR) transmission spectra information and mid-infrared (MIR) ATR spectra information of wines. This method improved the determination results by expanding the sources of analysis information. NIR spectra and MIR spectra of 153 wine samples from four different regions of grape growing were collected by near-infrared and mid-infrared Fourier transform spe trometer separately. These four different regions are Huailai, Yantai, Gansu and Changli, which areall typical geographical originals for Chinese wines. NIR and MIR discriminant models for wine regions were established using partial least squares discriminant analysis (PLS-DA) based on NIR spectra and MIR spectra separately. In PLS-DA, the regions of wine samples are presented in group of binary code. There are four wine regions in this paper, thereby using four nodes standing for categorical variables. The output nodes values for each sample in NIR and MIR models were normalized first. These values stand for the probabilities of each sample belonging to each category. They seemed as the input to the Bayesian discriminant formula as a priori probability value. The probabilities were substituteed into the Bayesian formula to get posterior probabilities, by which we can judge the new class characteristics of these samples. Considering the stability of PLS-DA models, all the wine samples were divided into calibration sets and validation sets randomly for ten times. The results of NIR and MIR discriminant models of four wine regions were as follows: the average accuracy rates of calibration sets were 78.21% (NIR) and 82.57% (MIR), and the average accuracy rates of validation sets were 82.50% (NIR) and 81.98% (MIR). After using the method proposed in this paper, the accuracy rates of calibration and validation changed to 87.11% and 90.87% separately, which all achieved better results of determination than individual spectroscopy. These results suggest that Bayesian information fusion of NIR and MIR spectra is feasible for fast identification of wine original regions.
Validation and analysis of the geographical origin of Angelica sinensis (Oliv.) Diels using multi-element and stable isotopes

Abstract:Background Place of origin is an important factor when determining the quality and authenticity of Angelica sinensis for medicinal use. It is important to trace the origin and confirm the regional characteristics of medicinal products for sustainable industrial development. Effectively tracing and confirming the material’s origin may be accomplished by detecting stable isotopes and mineral elements. Methods We studied 25 A. sinensis samples collected from three main producing areas (Linxia, Gannan, and Dingxi) in southeastern Gansu Province, China, to better identify its origin. We used inductively coupled plasma mass spectrometry (ICP-MS) and stable isotope ratio mass spectrometry (IRMS) to determine eight mineral elements (K, Mg, Ca, Zn, Cu, Mn, Cr, Al) and three stable isotopes (δ 13 C, δ 15 N, δ 18 O). Principal component analysis (PCA), partial least square discriminant analysis (PLS-DA) and linear discriminant analysis (LDA) were used to verify the validity of its geographical origin. Results K, Ca/Al, δ 13 C, δ 15 N and δ 18 O are important elements to distinguish A. sinensis sampled from Linxia, Gannan and Dingxi. We used an unsupervised PCA model to determine the dimensionality reduction of mineral elements and stable isotopes, which could distinguish the A. sinensis from Linxia. However, it could not easily distinguish A. sinensis sampled from Gannan and Dingxi. The supervised PLS-DA and LDA models could effectively distinguish samples taken from all three regions and perform cross-validation. The cross-validation accuracy of PLS-DA using mineral elements and stable isotopes was 84%, which was higher than LDA using mineral elements and stable isotopes. Conclusions The PLS-DA and LDA models provide a theoretical basis for tracing the origin of A. sinensis in three regions (Linxia, Gannan and Dingxi). This is significant for protecting consumers’ health, rights and interests.

Abstract:The objective of this study was to select the elements in tea leaves correlated with provenance soils, and further build a robust discriminant model of tea according to the geographical origin. Fresh tea leaves along with corresponding topsoil and subsoil samples were collected from three different tea producing regions in China. The concentrations of 20 elements were determined in tea leaf and soil samples. Analysis of variance, Pearson correlation analysis and linear discriminant analysis were used to analyze the obtained data. The results indicated that elemental fingerprinting profiles were different for tea leaves and provenance soils from different regions. The accumulation of some elements in tea leaves was closely correlated with their concentrations in soils. And this study revealed that Na, Mg, Ca, Ni, Rb, Sr and Pb were good descriptors to build discriminant models related to geology for tracing tea.
