Abstract:Phytosterol organic acid esters are important food resources and the components of biomembrane structure. Due to the lack of extraction and synthesis techniques, more research has been focused on phytosterols, and the research on phytosterol acid esters have encountered a bottleneck, but phytosterol acid esters confer substantial benefits to human health. In this study, stigmasteryl vanillate (VAN), stigmasteryl protocatechuate (PRO) and stigmasteryl sinapate (SIN) were prepared through the Steglich reaction. The processes are promotable and the products reach up to 95% purity. In addition, their stability was evaluated by differential scanning calorimetry and thermogravimetric analysis. HPLC analysis revealed an enhancement in water solubility after esterification with phenolic acid. In an in vitro digestion model, the bioaccessibility of stigmasteryl phenolates was significantly higher than that of stigmasterols (STIs). Regarding the anti-inflammatory properties, VAN, PRO, and SIN exhibit superior effects against TNF-α induced pro-inflammatory responses compared to STI. All stigmasteryl phenolates supplementation increased the ATP production, the basal, and maximal oxygen consumption rate in mitochondrial stress test. Overall, we present a synthesis method for stigmasteryl phenolates. It will contribute to the development and research of phytosterol acid ester analysis, functions and utilization in food. Moreover, the nutrient-stigmasterol hybrids tactic we have constructed is practical and can become a targeted mitochondrial delivery strategy with enhanced anti-inflammatory effects.
Prevention of Hypercholesterolemia with "Liposomes in Microspheres" Composite Carriers: A Promising Approach for Intestinal-Targeted Oral Delivery of Astaxanthin

Abstract:Cardiovascular diseases are caused by hypercholesterolemia. Astaxanthin (AST) has been reported to exhibit antioxidant and anti-inflammatory properties. However, its bioavailability is poor because of low solubility and instability. In order to improve the bioavailability of AST, we developed an intestinal-responsive composite carrier termed as "liposomes in micropheres" incorporating N-succinyl-chitosan (NSC)-poly(ethylene glycol) (PEG) liposomes that functionalized by neonatal Fc receptors (FcRn) into hydrogels of sodium alginate (SA) and carboxymethyl chitosan (CMCS). In the AST NSC/HSA-PEG liposomes@SA/CMCS microspheres, the AST's encapsulation efficiency (EE) was 96.26% (w/w) and its loading capacity (LC) was 6.47% (w/w). AST NSC/HSA-PEG liposomes had stability in the gastric conditions and achieved long-term release of AST in intestinal conditions. Then, AST NSC/HSA-PEG liposomes@SA/CMCS bind to intestinal epithelial cell targets by the neonatal Fc receptor. In vitro permeation studies show that there was a 4-fold increase of AST NSC/HSA-PEG liposomes@SA/CMCS in AST permeation across the intestinal epithelium. Subsequent in vivo experiments demonstrated that the composite carrier exhibited a remarkable mucoadhesive capacity, allowing for extended intestinal retention of up to 12 h, and it displayed deep penetration through the mucus layer, efficiently entering the intestinal villi epithelial cells, and enhancing the absorption of AST and its bioavailability in vivo. And oral administration of AST NSC/HSA-PEG liposomes@SA/CMCS could effectively prevent hypercholesterolemia caused by a high-fat, high-cholesterol diet (HFHCD). These advancements highlight the potential of NSC/HSA-PEG liposomes@SA/CMCS composite carriers for targeted and oral uptake of hydrophobic bioactives.
Surface modification by chitosan for improving stability and antioxidative activity of astaxanthin-loaded liposomes

Abstract:Astaxanthin (AST) is a compound renowned for its extraordinary antioxidant and anti-inflammatory properties. However, its application is limited due to low solubility, instability and bioavailability. To overcome these drawbacks, we encapsulated AST in liposomes (AST-loaded liposomes, AST-LIP) using chitosan (CS) as coating materials by thin film dispersion and high-pressure homogenization. We examined the physicochemical characteristics (particle size, polydispersity index, zeta potential, encapsulation efficiency, fourier transform infrared spectroscopy (FT-IR), and thermogravimetric analyzer (TGA)) of AST-LIP coated with various concentrations of CS, and confirmed the optimal concentration for the formulation of CS-coated AST-LIP was 1.5 mg/mL. Additionally, the storage stability and thermal stability analysis revealed that CS-AST-LIP exhibited significantly higher AST retention rates than AST-LIP under varying environmental conditions. Furthermore, CS-AST-LIP showed significantly higher DPPH·, OH· radical scavenging effects, and iron-reducing power than AST-LIP after storage for 4 weeks at 4 °C. For the cellular antioxidant assays, the CS-AST-LIP at the CS concentration of 1.5 mg/mL exhibited a satisfactory protective effect against oxidative damage induced by H 2 O 2 in Raw264.7 cells. In conclusion, these findings highlight the potential of chitosan as a promising coating material for improving the storage stability, thermal stability and antioxidant activity of AST-LIP.
Enhancement of Astaxanthin Bioaccessibility by Encapsulation in Liposomes: An In Vitro Study

Abstract:Astaxanthin was encapsulated in liposomes by a thin layer dispersion and ultrasound method using soybean phospholipid. The digestion properties of liposomes for encapsulating astaxanthin were investigated in light of particle size, size distribution, zeta potential, and microstructure during in vitro digestion as a function of time. These results exhibited that the average particle size increased gradually with liposomal vesicles retained round shapes and a fairly uniform distribution after passage through the simulated gastric fluid digestion. The result revealed that astaxanthin-loaded liposomes were stable in low pH conditions. It was also found that the mixed micelles formed in a simulated intestinal fluid. The zeta potential of astaxanthin-loaded liposomes had a decrease in negativity after digestion. In comparison with free astaxanthin, there was an appreciable increase in the bioaccessibility of astaxanthin after encapsulation in liposomes. This enhancement can be attributed to more soluble astaxanthin in the mixed micelles for astaxanthin-loaded liposomes. It indicated that the barrier of the liposomal bilayer could inhibit astaxanthin fading and leaking after encapsulation in liposomes. These results provide useful information for designing more stable delivery systems in the gastrointestinal tract and improving the bioaccessibility of lipophilic nutraceuticals.

Abstract:Astaxanthin-loaded liposomes were prepared by a thin-film ultrasonic method, and the effects of the different membrane surface modifiers chitosan hydrochloride (CH) and lactoferrin (LF) on the physicochemical stability of the liposomes and bioaccessibility of astaxanthin were studied. Based on the negative charge characteristics of egg yolk lecithin, LF and CH with positive charge were assembled on the surface of liposomes by an electrostatic deposition method. The optimal concentrations of modifiers were determined by particle size, zeta potential and encapsulation efficiency. The interaction between the liposomes and the coatings was characterized by Fourier Transform infrared spectroscopy. The stability of astaxanthin in different systems (suspension and liposomes) was investigated, and its antioxidant capacity and bioaccessibility were determined. The results showed that both membrane surface modifications could interact with liposomes and protect astaxanthin from oxidation or heat degradation and enhance the antioxidant activity of the liposome, therefore membrane surface modification played an important role in stabilizing the lipid bilayer. At the same time, the encapsulated astaxanthin exhibited higher in vitro bioaccessibility than the free astaxanthin. CH and LF modified liposomes can be developed as formulations for encapsulation and delivery of functional ingredients, providing a theoretical basis for the development of new astaxanthin series products.

Controlling release of astaxanthin in β-sitosterol oleogel-based emulsions via different self-assembled mechanisms and composition of the oleogelators

Abstract:In this study, three types of β-sitosterol-based oleogels (β-sitosterol + γ-oryzanol oleogels, β-sitosterol + lecithin, oleogels and β-sitosterol + monostearate oleogels), loaded with astaxanthin, were employed as the oil phase to create oleogel-based emulsions (SO, SL, and SM) using high-pressure homogenization. The microstructure revealed that fine-scale crystals were dispersed within the oil phase of the droplets in the β-sitosterol oleogel-based emulsion. The bioaccessibility of astaxanthin was found to be 58.13 %, 51.24 %, 36.57 %, and 45.72 % for SM, SL, SO, and the control group, respectively. Interestingly, the release of fatty acids was positively correlated with the availability of astaxanthin (P = 0.981). Further analysis of FFAs release and kinetics indicated that the structural strength of the oil-phase in the emulsions influenced the degree and rate of lipolysis. Additionally, the micellar fraction analysis suggested that the nature and composition of the oleogelators in SM and SL also impacted lipolysis and the bioaccessibility of astaxanthin. Furthermore, interfacial binding of lipase and isothermal titration calorimetry (ITC) measurements revealed that the oleogel network within the oil phase of the emulsion acted as a physical barrier, hindering the interaction between lipase and lipid. Overall, β-sitosterol oleogel-based emulsions offer a versatile platform for delivering hydrophobic molecules, enhancing the bioavailability of active compounds, and achieving sustained release.

Abstract:Astaxanthin, a hydrophobic carotenoid found in marine plants and animals, is claimed to exhibit various beneficial biological activities. Its use as a nutraceutical in foods, however, is currently limited by its low water-solubility and poor bioavailability. The goal of this paper was to fabricate astaxanthin-loaded colloidal particles to overcome these challenges. Astaxanthin was encapsulated in poly(lactic- co-glycolic acid) (PLGA) nanoparticles coated with chitosan oligosaccharides (COS). The properties of the loaded nanoparticles were characterized by transmission electron microscopy, scanning electron microscopy, and dynamic light scattering. The influence of PLGA properties on the loading capacity, water solubility, stability, and release of the astaxanthin were determined. The nanoparticles were smooth spheres with mean particle diameters around 150 nm and positive surface potentials (ζ = +30 mV). The encapsulation efficiency (>85%) and loading capacity (>15%) of the astaxanthin in the nanoparticles was relatively high. X-ray analysis suggested that the encapsulated astaxanthin was in an amorphous form. The nanoparticles had good dispersibility and stability in aqueous solutions, as well as high cytocompatibility. In vitro studies showed that the astaxanthin was released from the nanoparticles under simulated gastric and small intestinal conditions. Overall, our results suggest the core-shell nanoparticles developed in this study may be suitable for encapsulating this important nutraceutical in functional foods and cosmetics.

Abstract:Astaxanthin has 550 times more antioxidant activity than vitamin E, so it can scavenge free radicals in vivo and improve body immunity. However, the poor stability of astaxanthin becomes a bottleneck problem that limits its application. Herein, Haematococcus pluvialis (H. pluvialis) as a raw material was used to extract astaxanthin, and the optimal extraction conditions included the extraction solvent (EA:EtOH = 1:6, v/v), extraction temperature (60 °C), and extraction time (70 min). The extracted astaxanthin was then loaded using lecithin to form corresponding liposomes via the ethanol injection method. The results showed that the particle size and zeta potential of the prepared liposomes were 105.8 ± 1.2 nm and −38.0 ± 1.7 mV, respectively, and the encapsulation efficiency of astaxanthin in liposomes was 88.83%. More importantly, the stability of astaxanthin was significantly improved after being embedded in the prepared liposomes.

Abstract:In this study, a layer-by-layer (LBL) encapsulated astaxanthin (Ast) nanoemulsion delivery system based on chitosan (CS) and tremella polysaccharide (TP) was successfully developed. The system constructed an Ast-CS-TP emulsion with high encapsulation efficiency and an excellent stability profile by utilizing the opposite charge properties of CS and TP. This study evaluated the effects of different stresses (including temperature, salt addition, pH, UV irradiation, and centrifugal force) on the emulsion's stability. To further investigate the protective mechanism of the emulsions, we performed antioxidant activity experiments after UV treatment. Additionally, an in vitro digestion experiment was conducted to assess the behavior of Ast emulsion under simulated gastrointestinal conditions. The stability correlation coefficients were calculated using the Python database Pandas. The results showed that Ast-CS-TP emulsions exhibited turbidity and enhanced homogeneity with a small particle size of around 400 nm and a high absolute zeta potential of 35 mV and exhibited excellent stability under various stresses. The Ast-CS-TP emulsions also exhibited pH-responsive release at pH ≥ 7, consistent with pH changes in the gastrointestinal tract, and were stable in highly concentrated salt solutions. We found that the CS and TP layers significantly improved the photostability of Ast. CS and TP significantly enhanced Ast's oral bioavailability. The stability correlation coeffcients showed that pH and salt concentration were the largest factors that affected the stability of the emulsion. This study provided important insights into the encapsulation and targeting of Ast, providing a theoretical foundation and technical guidance for the comprehensive utilization of Ast.

Abstract:Oral administration of astaxanthin (AST), a potent antioxidant, is limited owing to its low solubility, physicochemical stability, and bioavailability. This study developed pH-responsive nanocarriers by the electrostatic self-assembly of 2,2,6,6-tetramethylpiperidine-1-oxyradical (TEMPO)-oxidized cellulose nanofibers (TCNFs) and chitosan (CS) to enhance the intestinal delivery of AST. The TCNF/CS@AST nanoparticles were optimized through single-factor experiments and Box-Behnken design, subsequently overcoming the hydrophobicity of AST and demonstrating improved stability against environmental stressors and controlled release in the intestinal environment. Transmission electron microscopy confirmed the near-spherical shape of these nanoparticles, with an average hydrodynamic diameter of 64 nm. TCNF/CS@AST enhanced the antioxidant effectiveness of AST after digestion and in lipopolysaccharide-stimulated RAW 264.7 cells while demonstrating good cellular compatibility. These nanoparticles present a promising strategy for the oral delivery of hydrophobic bioactive compounds orally, with potential applications in precision nutrition.
Preparation of astaxanthin/zeaxanthin-loaded nanostructured lipid carriers for enhanced bioavailability: Characterization-, stability-and permeability study

Abstract:Astaxanthin (ASTA) and zeaxanthin (ZEA) are xanthophyll carotenoids showing a wide spectrum of health-promoting properties. However, their utilization is limited, mostly due to poor water solubility, limited bioavailability, and a tendency to oxidate, as well as photo- and thermal instability. The aim of this work was to develop ASTA- and ZEA-loaded nano-structured lipid carriers (NLCs) that would protect them against degradation and improve their intestinal stability/permeability. Obtained NLCs were characterized by an effective diameter of 294 nm for ASTA-NLC and 280 nm for ZEA-NLC; polydispersity index (PDI) lower than 0.2; and zeta potential of -29.4 mV and -29.0 mV, respectively. Interestingly, despite similar physicochemical characteristics, our investigation revealed differences in the encapsulation efficiency of ASTA-NLC and ZEA-NLC (58.0 % vs. 75.5 %, respectively). Obtained NLCs were stable during a 21 day-storage period in the dark at room temperature or at 4 °C. Investigation of gastrointestinal stability showed no change in effective diameter and PDI under gastric conditions while both parameters significantly changed under intestinal conditions. Our results showed for the first time that both ASTA- and ZEA-NLCs intestinal absorption investigated in the in vitro model is significantly increased (in relation to pure compounds) and is affected by the presence of mucus. This study provides useful data about the advantages of using NLC as a delivery system for ASTA and ZEA that might facilitate their applications in the food and pharmaceutical industry.
Lyotropic Liquid Crystal System for Drug Delivery of Astaxanthin: Physical Characterization and Enhanced Antioxidant Potential

Abstract:Astaxanthin is a xanthophyll carotenoid, well known for its potent anti-inflammatory and antioxidant properties, owing to its unsaturated molecular structure. Aquatic plants and animals contain the hydrophobic carotenoid astaxanthin, which is thought to possess a number of advantageous biological traits. However, due to its weak bioavailability and low water solubility, its use as a nutraceutical in food is currently restricted. Cubosomal encapsulation has been considered an effective alternative for improving the bioavailability and solubility of hydrophobic bioactives. The current paper aimed to conquer these issues by encapsulating astaxanthin in lyotropic liquid crystal nano-formulations prepared via the fabrication method. The physicochemical properties of astaxanthin-loaded cubosomes (AST-LC) have also been analyzed, in order to know the morphology of the prepared formulations and their bioavailability in the biological system. The formulation has been tested for particle size, thermal behavior, zeta potential, crystallinity, encapsulation efficiency, and drug-polymer interactions. The observed experimental results showed the particle size and zeta potential of astaxanthin-loaded cubosomes (AST-LC) as 199 ± 0.23 nm −27.4 ± 4.67 mV, with a small polydispersity index (0.283 ± 1.01). The cubic structure and nano-range size of the ideal formulation were verified by a field emission scanning electron microscope (FESEM) and (HRTEM) high-resolution transmission electron microscopic examination. The formulation exhibits a higher encapsulation efficiency with good yield. Results from X-ray diffraction and diffraction scanning calorimetry demonstrated an amorphous state of astaxanthin incorporated into the formulation. Fourier transform infrared spectroscopy (FTIR) analysis of AST-LC showed the absence of astaxanthin main peaks, indicating its complete encapsulation inside the formulation. The drug–excipient interaction was carried out with diffraction scanning calorimetry DSC and FTIR, resulting in no interaction between them. These results offered important details about increasing astaxanthin bioavailability by incorporating it into cubosomes. Furthermore, the astaxanthin loaded into cubosomes has been evaluated for antioxidant potency, compared with astaxanthin extract using 2,2-diphenylpicrylhydrazylassay (DPPH assay).

Abstract:Background: Astaxanthin (Asta) is widely used in the nutraceutical and food industry because of its strong antioxidant properties. However, natural Asta mainly exists in the esterified form with various fatty acid chains, making it difficult to understand the particular molecular structure of astaxanthin esters (Asta-Es) that have better antioxidant capacity. In this study, Asta-Es with different molecular structures was systematically prepared, and identified by using thin-layer chromatography (TLC), ultraviolet-visible (UV-visible), high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), and proton nuclear magnetic resonance (1 H-NMR). In addition, their antioxidant properties were evaluated by 2,2'-diphenylpicrylhydrazyl (DPPH) and ABTS scavenging activity. Results: Fourteen Asta-Es with different molecular structures were systematically synthesized. This study used a simple and efficient method for the separation and purification of astaxanthin monoester (Asta-ME) and astaxanthin diester (Asta-DE) with high purity (86%-94%) by silica gel column chromatography. 13-cis-Asta-E and 9-cis-Asta-E were firstly identified from Asta-E. The DPPH clearance rates and ABTS scavenging rates of Asta-C4:0, Asta-C8:0, Asta-C12:0, and Asta-C18:0 were relatively close, but the DPPH and ABTS scavenging rates of Asta-C18:0, Asta-C18:1, Asta-C18:2, and Asta-C22:6 increased gradually. Among all Asta-Es, Asta-C22:6/C22:6 showed the highest antioxidant capacity, with the DPPH and ABTS scavenging rates of 77.22 ± 3.29% and 51.84 ± 1.65%, respectively. Conclusion: In this study, it was concluded that chemically synthesized Asta-Es contained cis-astaxanthin ester and polyunsaturated fatty acid chain increased the antioxidant activity of Asta, showing less effect of the length of fatty acid chain. These results provide useful information for the production and use of highly efficient Asta-E as functional food and pharmaceutical ingredients. © 2020 Society of Chemical Industry.

These illustrate the questionable role of supposed causes, the different modes of onset and course of sudden hearing loss, the audiometric findings and concomitant symptoms. The situation of both ears prior to the acute loss seems to be of great importance and offers a frame for classification, which is of clinical relevance. These features and others are discussed with regard to common theories on the etiology of sudden hearing loss. The difficulties in assessing therapeutic effects are pointed out considering the high rate of spontaneous recovery.
Emotional reactions and life changes of caregivers of demented patients when home caregiving ends

Abstract:Many studies indicate that the cessation of caregiving of demented patients has positive effects on caregivers' lives but contradictory findings have also been reported. The aim of this study was to investigate how the caregivers of demented patients experienced the cessation of caregiving, how caregivers' lives changed after this event, and which factors predicted emotional reactions and life changes. Data were collected from 64 caregivers of demented patients, recruited from a previous controlled intervention study. The semi-structured telephone interviews sought (1) sociodemographic data, (2) emotional reactions the caregivers recalled from the point of cessation of caregiving, and (3) life changes experienced after that time point. At the point of cessation of caregiving, spouse caregivers had higher risk for loneliness, depressive feelings and sorrow than non-spouse caregivers. Feelings of relief were associated with non-spouses. Community care support obtained during the previous intervention study had protected the caregivers from loneliness, depressive feelings and sorrow. After the end of caregiving, spouse caregivers tended to experience more negative life changes. The increased number of leisure activities was associated with the support during community care.
Preparation of Oxidized Starch/β-Lactoglobulin Complex Particles Using Microfluidic Chip for the Stabilization of Astaxanthin Emulsion

Abstract:Here, we designed an oxidized starch/β-lactoglobulin (OS/β-lg) complex colloidal particle using a dual-channel microfluidic chip for the stabilization of astaxanthin emulsion. The effect of the mixing ratio, pH, and the degree of substitution (DS) of the oxidized starch on the formation of OS/β-lg complex particles was investigated in detail. The optimal complexation occurred at a pH of 3.6, a mixing ratio of 2:10, and a DS of 0.72%, giving an ideal colloidal particle with near-neutral wettability. With this optimum agent, the astaxanthin-loaded oil-in-water emulsions were successfully prepared. The obtained emulsions showed the typical non-Newton fluid behavior, and the rheological data met the Herschel–Bulkley model. The microscopic images confirmed the dense adsorption of the particle on the oil/water interface. In vitro release and stability studies demonstrated this compact layer contributed to the controlled-release and excellent stability of astaxanthin emulsions facing heat, ultraviolet, and oxidative intervention. This work suggests the potential of microfluidics for the production of food-grade solid emulsifiers.

Abstract:Astaxanthin (AST), as natural hydrophobic nutrition, has exhibited health-promoting benefits for its outstanding antioxidant property. However, most studies tend to enhance its stability and solubility while the targeted delivery of AST is limited. In this study, liver-targeted nanocarriers were designed and prepared by lactobionic acidmodified (2-hydroxypropyl-beta-cyclodextrin) for efficient controlled delivery of AST. The minimum average size of AST nanoparticles was about 98 nm with a polydispersity index (PDI) of 0.41. The lactobionic acid-modified AST nanoparticles exhibited significant cellular uptake, and an admirable ability to scavenge free radicals for H2O2induced HepaRG cells in preventing mitochondrial depolarization. Moreover, accumulation of AST nanoparticles in liver was observed due to the modification of lactobionic acid (LA) of the nanocarriers through the specific binding of LA-asialoglycoprotein receptors. The results in this study provided a new idea for liver-specific nutrition delivery of AST in developing functional food for liver disease nutrition intervention.
Influence of oil matrixes on stability, antioxidant activity, bioaccessibility and bioavailability of astaxanthin ester

Abstract:Background: Astaxanthin ester (Asta-E) is used as functional nutraceuticals in many food products. Unfortunately, Asta-E utilization is currently limited owing to its chemical instability and low bioavailability. The purpose of this study is to investigate the promotion effect of oil matrixes on the stability, antioxidant activity, bioaccessibility and bioavailability of Asta-E. Results: The results showed that the stability of Asta-E in six oil matrixes was improved. Based on the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity experiment, the antioxidant activity of Asta-E was positively correlated with the degree of unsaturation of the oil matrixes, but not with the side chain length. The in vitro gastrointestinal tract (GIT) simulation model and in vivo experiment using mice were also employed to investigate the digestion and absorption characteristics of Asta-E in various oil matrixes. The results demonstrated that the bioaccessibility and bioavailability of Asta-E increased with the increase of fatty acid chain length of oil matrixes (triglyceride oleate > triglyceride caprylate > triglyceride butyrate), as well as with the decrease of unsaturation degree (olive oil > corn oil > fish oil). Conclusion: Monounsaturated fatty acids (MUFA) and long-chain triglyceride (LCT) in an oil matrix were the factors that could efficiently improve the bioavailability of Asta-E. Moreover, the size of the mixed micelles of Asta-E during digestion was the main factor influencing the bioaccessibility of Asta-E. This study provides references for the design of suitable oil matrixes for Asta-E. © 2020 Society of Chemical Industry.
