Abstract:Antarctic krill ( Euphausia superba ) is an important source of biomass and high-quality protein. However, heat treatment negatively impacts the quality of Antarctic krill and compromises its utilization in the food industry. This study aimed to investigate the mechanisms underlying changes in Antarctic krill meat characteristics and physicochemical properties treated at different temperatures and holding times. Our findings indicated that hardness and cooking loss of Antarctic krill meat increased dramatically at higher temperatures and holding times. The low-field nuclear magnetic resonance (LF-NMR) analysis revealed that the loss of immobilized water increased, whereas the SDS-PAGE analysis showed that the content of myosin heavy chain decreased significantly and that protein degradation occurred. Fourier transform infrared spectroscopy (FT-IR) and intrinsic fluorescence spectra suggested that α-helix motifs were transformed into β-sheets and that more hydrophobic groups were exposed. The scanning electron microscopy (SEM) results showed that Antarctic krill meat formed corrugated folded regions after heat treatment without forming a three-dimensional water-entrapped structure, which led to significant water loss, resulting in rapid deterioration of Antarctic krill meat. These results provide the basis for a deeper understanding of the processing characteristics of Antarctic krill meat.

Abstract:Sous-vide cooking is a highly praised method used to cook muscle foods because of its desired effect of providing better sensory properties by maintaining texture. In this study, we further explored the effect of water on texture by revealing the mechanisms of moisture migration. Low field nuclear magnetic resonance (LF-NMR) showed that the nonflowing water in sous-vide cooking hairtail was 2.36 ± 0.33% higher than that in traditional cooking. Magnetic resonance imaging (MRI) was used to clarify the law of moisture migration induced by temperature, and the moisture migration of the sous-vide cooking hairtail was slower during the holding heating stage. The microstructure explained the change rules of the texture. The degree of change was consistent with the moisture migration level. Digitalizing analysis quantitatively verified the effect of sous-vide cooking on the hairtail microstructure. The low moisture migration rate of sous-vide cooking resulted in a less damaged microstructure of the hairtail, manifesting as a desirable texture. PRACTICAL APPLICATION: LF-NMR and MRI showed that sous-vide hairtails exhibited a lower moisture migration rate. The holding heating stage only slightly changed the microstructure of the hairtail. The digitalizing analysis confirmed the moisture migration mechanisms. Heat-induced protein denaturation was closely related to the water state.

Abstract:We have developed a methodology using advanced thermal analysis to characterize the role of water in a specially synthesized family of recombinant spider silk-like block copolymers. These proteins were inspired by the genetic sequences found in the dragline silk of Nephila clavipes, comprising an alanine-rich hydrophobic block, A; a glycine-rich hydrophilic block, B; and a C-terminus or a His-tag, H. This family of proteins serves as a model system in which the hydrophobicity is controlled by A and B block lengths, allowing systematic comparison of water effects within the family. Temperature-modulated differential scanning calorimetry and thermogravimetric analyses were employed to capture the glass to rubber transition, T-g, in water-cast protein films. Modeling of the solid and liquid state heat capacity baselines allows us to determine the critical role played by bound water which plasticizes and stabilizes the protein through interchain bonding. In samples containing bound water, two sequential glass transitions, T-g(1) and T-g(2), were observed during heating. The lower temperature glass transition, T-g(1), is related to conformational change induced by bound water removal, the hydrophobicity of the protein sequences, and the crystallinity of the protein. The higher temperature glass transition, T-g(2), is characteristic of the dry protein. The binding energy of water to protein compares favorably to ligandwater binding affinities. The energy absorbed by evaporating water depends upon the volume fraction of the hydrophilic B-block.

Abstract:Fibrous protein secondary structural transitions are affected by bound water. A family of specially designed recombinant spider silk-like block copolymers with a gradient of block length, hydrophobilicity, and degree of crystallinity was biosynthesized and characterized to demonstrate the effect of water on the protein structural transitions. These proteins were inspired by the genetic sequences found in the dragline silk of Nephila clavipes, comprising an alanine-rich hydrophobic block, A, a glycine-rich hydrophilic block, B, and a C-terminus or a His-tag, H. Because the A-block is hydrophobic and the B-block is hydrophilic, the spider silk-like block copolymers behave as amphiphilic molecules and self-assemble into various structures in water solution. We employ time-resolved Fourier transform infrared (FTIR) spectroscopy to assign the origin of specific secondary structural transitions during heating. A transition from random coils to beta-turns dominates during a lower temperature glass transition (of plasticized protein) mediated by the removal of bound water. Once the protein is in the dry solid state, further heating causes the now-dry protein to undergo the glass transition to the liquid state through conversion of alpha-helices into beta-turns. The structural transitions during protein glass transitions are intrinsic to the amorphous region of protein and are hardly affected by protein hydrophobicity, block length, or crystallinity. The self-assembly morphology of the spider silk-like block copolymers, investigated by scanning electron microscopy, indicates that the large-scale morphology is stable during heating through both the lower and upper temperature glass transitions.

Abstract:Control on the moisture distribution, protein structure changes, and protein degradation of Antarctic krill meat during freeze-thaw (F-T) cycles by presoaking with antifreeze protein (AFP) was investigated. The results from the thawing loss rate and cooking loss rate indicated that 0.1% was the optimal AFP concentration. Magnetic resonance imaging and low-field nuclear magnetic resonance results showed that AFP inhibited the changes in moisture distribution and maintained the moisture in Antarctic krill meat. The contents of nonprotein nitrogen and trichloroacetic acid-soluble peptides indicated that AFP reduced protein degradation. Further, SDS-PAGE showed that AFP reduced the degradation of actin, troponin T, and myosin light chain. The results of fluorescence spectra, circular dichroism, and chemical bond contents indicated that AFP reduced the damage of the protein tertiary and secondary structures of Antarctic krill meat by holding it in a weak polar environment. This study supplied basic theory for the quality control of Antarctic krill meat. PRACTICAL APPLICATION: Protein degradation, moisture distribution, and protein structure changes occurred to Antarctic krill meat during freeze-thaw cycles due to ice crystal growth and recrystallization, which leads to the decrease in quality. Antifreeze protein has been proven to avoid ice crystals' growth and inhibit ice recrystallization. During freeze-thaw cycles, the moisture distribution of Antarctic krill meat treated with antifreeze protein was more uniform, the degree of protein degradation was lower, and the protein structure was protected. This study demonstrated the potential of antifreeze protein as a water and protein protectant of Antarctic krill meat during freeze-thaw cycles.

Abstract:Antarctic krill is a potential and attractive resource for consumption. However, most Antarctic krill meat is used to produce primary products with low commercial value, with few highly processed products. This study aimed to evaluate and improve the gelling properties of Antarctic krill surimi, with Pacific white shrimp surimi as control. Compared with Pacific white shrimp surimi, the lower β-sheet content and protein aggregation degree had a severe impact on the formation of the gel network of Antarctic krill surimi, which resulted in weaker breaking force, gel strength, and viscoelasticity (p 0.05). However, the addition of ι-carrageenan (2%) or κ-carrageenan (1~2%) is an effective way to improve the texture properties of Antarctic krill surimi (p < 0.05). These findings will contribute to the development of reconstituted Antarctic krill surimi products with high nutritional quality and the promotion of deep-processing products of Antarctic krill meat.

Abstract:The high-emulation meat with 60% similar to 80% moisture content made use of high humidity extrusion technology was prepared with soybean protein isolate, defatted soybean meal and wheat gluten as main raw materials. The effect of extrusion system parameters (extrusion temperature, screw rpm, moisture content and soybean protein isolate content) on product textured degree was studied. Results showed that: the influence of the moisture content on the product textured degree was the most important, and screw speed was less influential. The textured degree of high moisture content fibriform imitated meat increased first and then decreased with the increasing of extrusion temperature, screw rpm, moisture content and soybean protein isolate content. So, better textured degree could be received by increasing the parameters to advisable numerical value, and the product would have an obvious fibroid structure. The conclusion could also be used as a value reference in the production of high-emulation meat of livestock and poultry. At present, we are committed to produced emulation meat with soy protein. High moisture extraction[1] is jointly developed by Clextral Co. France and Pro. Co.Ltd was one way of producing simulation meat. This technology, with special cooling die head twin-screw extruder for main equipment, prepared textured soy protein production at the moisture content of material higher than 45%. It had been discovered that with the increase of moisture content of textured soy protein the textured rate of production would be increased. The elastic, resiliency and fibroid structure would be much more like the animal meat. At present, extrusion of material moisture higher than 60% could not be achieved in the domestic. The twin-screw extruding technology on soy protein with moisture content 60%similar to 80% was studied in this subject. Production condition of high moisture content fibrous imitated meat would be established and technical features would be supplied to domestic extruding equipment through the influence of extruding temperature, material moisture content, screw speed and material composition on soy protein molecular recombination and fibrosis.

Abstract:<p>A novel protein extraction method of ultrasound-assisted basic electrolyzed water (BEW) was proposed, and its effects on the structural and functional properties of Antarctic krill proteins were investigated. Results showed that BEW reduced 30.9% (w/w) NaOH consumption for the extraction of krill proteins, and its negative redox potential (-800∼-900mV) protected the active groups (carbonyl, free sulfhydryl, etc.) of the proteins from oxidation compared to deionized water (DW). Moreover, the ultrasound-assisted BEW increased the extraction yield (9.4%), improved the solubility (8.5%), reduced the particle size (57nm), favored the transition of α-helix and β-turn to β-sheet, promoted the surface hydrophobicity and disulfide bonds formation of krill proteins when compared to BEW without ultrasound. These changes contributed to the enhanced foam capacity, foam stability and emulsifying capacity of the krill proteins. Notably, all the physicochemical, structural and functional properties of the krill proteins were comparable to those extracted by the traditional ultrasound-assisted DW. This study suggests that the ultrasound-assisted BEW can be a potential candidate to extract proteins, especially offering an alternative way to produce marine proteins with high nutritional quality.</p>

Abstract:Antarctic krill (Euphausia superba) has been widely studied and extensively recognized as a target for commercial fishing. In this study, Antarctic krill chitin was extracted from defatted Antarctic krill shell, and its crystalline structure and thermal properties were characterized by employing Fourier transform infrared spectroscopy, X-ray diffractometry, scanning electron microscopy, thermogravimetry, and differential scanning calorimetry. Results showed that Antarctic krill chitin corresponded to the α-polymorph, and was composed of small, stable, and uniform microcrystals. The degree of N-deacetylation was 11.28 ± 0.86%. The d-spacings of Antarctic krill chitin were 9.78 Å and 4.63 Å at (020) and (110) planes. The crystalline sizes were 6.07 nm and 5.16 nm at (020) and (110) planes, respectively. The activation energy of the polysaccharide chain decomposition was 123.35 kJ/mol and the glass transition (T(g)) of Antarctic krill chitin was 164.96 °C.

Abstract:The Maillard conjugates of cod skin collagen peptide (CSCP) and polydextrose (PDX) were used to stabilize the Antarctic krill oil (AKO) emulsion. First, CSCP–PDX glycoconjugate was prepared using microwave heating (MH) for 20 min, which had a higher grafting rate, wider structure, and better functional characteristics than CSCP–PDX glycoconjugate, which was prepared by traditional wet-heating (WH) after 10 h. Then, Maillard conjugates as an emulsifier improved the stability of AKO emulsion, as shown by the zeta potential, rheology, and so forth. In vitro digestibility indicated that the free fatty acid (FFA) release of MH (20 min)-stabilized emulsion was significantly increased by 5.22% (P < 0.05) compared with that of CSCP-stabilized emulsion, displaying a sustained release in simulated intestinal fluid (SIF). The results of this study highlighted the potential of Maillard conjugates to serve as a useful emulsifier for utilization in the food industry.

Abstract:Blending two or more materials to create better high-moisture meat analogues has been actively studied in the food science and technology field. Walnut protein is a high-quality plant-based protein resource, yet its full potential remains underexploited. Thus, this study focused on exploring the quality characteristics and fibrous structure formation mechanism of walnut protein (WP) and wheat gluten (WG) meat analogues during high-moisture extrusion cooking process. Results showed that the optimized WP and WG-blended high-moisture meat analogues exhibited a more pronounced anisotropic and oriented fibrous structure. The blending of WP and WG can protect the molecular chains from the thermal transition, and promote the aggregation of protein molecules mainly by enhancing the interaction between hydrophobic interactions and hydrogen bonds, increasing the apparent viscosity and forming protein subunits with larger molecular weights (>100 kDa) to stabilize the newly formed conformation. Additionally, the content of α-helix was the highest among the secondary structures. This study provides a theoretical basis for the application of WG and WP to produce HMMAs with rich fibrous structures.

Abstract:Defatted krill powder, rich in protein, has emerged as a potential, novel food raw material. The present study aimed to prepare Antarctic krill protein from the supernatant (AKP-S) and precipitation (AKP-P) using the ultrasound-assisted alkali dissolution and acid precipitation method, and analyze their composition, structure, and functional properties were analyzed. The protein contents of AKP-P and AKP-S were more than 85% (dry weight), and their total amino acid content was 48.75 and 43.59 g/100 g of protein, respectively. The SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and surface morphology analysis (SEM) results showed that the AKP-S was degraded during acid treatment and had a smaller particle size than AKP-P. The α-helix and random curled structures of AKP-P and AKP-S were few or even disappeared. The solubility of AKP-S was more than 70% in the pH range of 3–13, and the change in AKP-P solubility with pH was consistent with the trend of β-sheets. Except for pH 7.0–9.0, the solubility of AKP-P and AKP-S with pH was positively correlated with surface-hydrophobicity (H0). The emulsifying activity of AKP-P and AKP-S at different pH was approximately positively correlated with the solubility. The minimum gel-forming concentration of AKP-P was 8%, while AKP-S could not form gel at 14%. The gel water holding capacity reached more than 90% when the AKP-P concentration was 12%. The rheological results showed that the storage modulus (G′) predominated over the loss modulus (G″) of AKP-P in the range of 8%–14% concentration.

Abstract:Antarctic krill are a consumption resource with great exploitation potential. However, the poor gel properties of Antarctic krill meat seriously limit its high-value application. In the present study, the quality characteristics and proteome changes of the κ-/ι-carrageenan-Antarctic krill surimi gel were systematically analyzed and compared. In addition, the transcriptome sequencing of Antarctic krill was carried out, which filled the gap in the Antarctic krill database. Higher molecular forces (disulfide bond and hydrophobic interaction) and the degree of network cross-linking significantly promoted the formation of κ/ι-carrageenan-Antarctic krill surimi compared to that of Antarctic krill surimi. This is the first study to investigate and map potential protein markers for quality characteristics of Antarctic krill surimi based on mass spectrometry-based label-free quantitative proteomics. The results could provide a theoretical reference for the quality control of Antarctic krill during application.

Abstract:Intensive search of all cases of MS occurring in the Sardinian commune of Macomer since 1912 indicated that MS was absent up to the early 1950s. All 13 ascertained cases had clinical onset in the years 1952–1981. During this period, the average annual incidence was 4.8 per 100,000; the highest incidence was found in the period 1957–1961, slowly decreasing up to 1981. MS was probably introduced after 1945, when the centuries-old isolation of Macomer ended and the native population came into contact with individuals from high-and medium-risk areas.

Abstract:Summary The object of this study was a high‐protein Antarctic krill ball which was made of mixed shrimp surimi (Antarctic krill and white shrimp). The effects of four different exogenous additives on gel properties and spatial structure of mixed shrimp surimi systems during processing were investigated by texture profile analysis, low‐field nuclear magnetic resonance, magnetic resonance imaging, Fourier transformed infrared spectroscopy, scanning electron microscopy and rheology analysis, according to product formulation and processing conditions. Different exogenous additives improved distinctly the mixed shrimp surimi system. The use of the four exogenous additives combined had a synergistic promotional effect. Acetylated distarch adipate, egg white powder, and soy protein isolate played a vital role in producing high‐protein Antarctic krill balls. The relatively limited improvement of transglutaminase could be attributed to processing temperature and the presence of autolytic enzymes originated intrinsically from Antarctic krill surimi.

Gelation of protein recovered from whole Antarctic krill (Euphausia superba) by isoelectric solubilization/precipitation as affected by functional additives

Abstract:This study demonstrated that the novel isoelectric solubilization/precipitation can be applied to recover functional muscle protein in a continuous mode from whole Antarctic krill. Protein recovered from whole krill had a much lower ash content than whole krill, suggesting good removal of inedible impurities (shell, appendages, etc.). Lipids were retained to a higher degree with krill protein solubilized at acidic rather than basic pH. The viscoelastic modulus (G') showed that recovered krill protein failed to form heat-induced gel unless beef plasma protein (BPP) was added. Therefore, protease inhibitors are suggested for development of krill-derived products. Even with BPP, the G' decreased between 45 and 55 degrees C. However, krill protein solubilized at acidic pH had a higher decrease of the G' than the protein solubilized at basic pH, likely due to krill endogenous cathepsin L. Krill protein-based gels developed from protein solubilized at basic pH, especially pH 12.0, had better texture (torsion and Kramer tests and texture profile analysis) than acidic counterparts, possibly due to higher proteolysis and denaturation at acidic pH. Gels made from protein solubilized at acidic pH were brighter and whiter likely due to a higher lipid content.

The results revealed that the addition of AKP led to a reduction in the freezable water content, T2 relaxation time, and free sulfhydryl content of frozen dough. After 6 weeks of freezing, the viscoelastic properties of the dough with AKP were higher than those of the control. Additionally, the fermentation height of dough containing 1.5% AKP increased by 25.85% compared with the control, resulting in increased springiness and specific volume, and decreased hardness in the resulting steamed bread. Pearson correlation analysis showed a positive correlation between freezable water content and free sulfhydryl groups, while a negative correlation was observed with springiness. Furthermore, springiness showed a positive correlation with the DPPH free radical scavenging rate. These findings highlight the potential of AKP as a novel food cryoprotectant capable of significantly enhancing the quality and nutritional value of frozen wheat products.
