Abstract:A hydroponics trial was employed to study the effects of Pseudomonas putida CZ1 (CZ1), a heavy-metal-resistant bacterial strain isolated from the rhizosphere of Elsholtzia splendens (E. splendens), on the uptake and translocation of copper (Cu) in E. splendens. Significant promotion of plant growth coupled with the obvious plant-growth-promoting (PGP) characters of the bacteria suggested that CZ1 would be a plant-growth-promoting rhizobacterium (PGPR) to E. splendens under Cu stress condition. The results of inductively coupled plasma optical emission spectrometry (ICP-OES) showed that CZ1 increased the concentration of Cu in the shoots (up to 211.6 % compared to non-inoculation treatment) and translocation factor (TF) (from 0.56 to 1.83 %) of those exposed to Cu. The distribution of Cu in root cross section measured by synchrotron-based X-ray fluorescence microscopy (SRXRF) indicated that CZ1 promoted the transport of Cu from cortex to xylem in roots, which contributed to the accumulation of Cu in shoots. Furthermore, CZ1 improved the uptake of nutrient elements by plants to oppose to the toxicity of Cu. In summary, P. putida CZ1 acted as a PGPR in resistance to Cu and promoted the accumulation and translocation of Cu from root to shoot by element redistribution in plant root; hence, CZ1 is a promising assistance to phytoremediation.

Abstract:Waste Management &Research, Ahead of Print. The low efficiency and selectivity limitations of biohydrometallurgy technique compel the researchers to explore novel microbial strains acclimated to metal existence site with higher toxicity tolerance and bioleaching capability in order to improve the role of bioleaching process for e-waste management. The current study aimed to explore bioleaching potential of indigenous Bacillus sporothermodurans ISO1; isolated from metal habituated site. The statistical approach was utilized to optimize a variety of culture variables including temperature, pH, glycine concentration and pulp density that impact bio-cyanide production and leaching efficiency. The highest dissolution of Cu and Ag, 78% and 37% respectively, was obtained at 40 °C, pH 8, glycine concentration 5 g L−1, and pulp density 10 g L−1 through One Factor at a Time (OFAT), which was further increased up to 95% Cu and 44% Ag recovery through the interactive effect of key factors in the Response Surface Methodology (RSM) approach. Furthermore, Chemo-biohydrometallurgy approach was utilized to overwhelm the specificity limitation; as higher concentration of Cu in computer printed circuit boards (CPCBs) causes interference to recover other metals. The sequential leaching through ferric chloride (FeCl3), recovered Cu prior to bio-cyanidation by B. sporothermodurans ISO1 and resulted in the improved leaching of Ag (57%), Au (67%), Pt (60%), etc. The current work reports on B. sporothermodurans ISO1, a new Bacillus strain that exhibits highest toxicity tolerance (EC50 = 425 g L−1) than earlier reported stains and has higher leaching potential that can be implemented to large-scale biometallurgical process for e-waste treatment to achieve the agenda of sustainable development goal (SDG) under the strategies of urban mining.

Abstract:Cadmium (Cd) is a highly toxic and cumulative environmental pollutant. Siderophores are heavy metal chelators with high affinity to heavy metals, such as Cd. Ryegrass (Lolium perenne L.) has a potential remediation capacity for soils contaminated by heavy metals. Consequently, using ryegrass alongside beneficial soil microorganisms that produce siderophores may be an effective means to remediate soils contaminated with Cd. In this study, the Bacillus strains WL1210 and CD303, which were previously isolated from the rhizospheres of Nitraria tangutorum in Wulan and Peganum harmala L. in Dachaidan, Qinghai, China, respectively, both arid and sandy environments, were evaluated for heavy metal pollution mitigation. Our quantitative analyses have discerned that the two bacterial strains possess commendable attributes of phosphorus (P) solubilization and potassium (K) dissolution, coupled with the capacity to produce phytohormones. To assess the heavy metal stress resilience of these strains, they were subjected to a cadmium concentration gradient, revealing their incremental growth despite cadmium presence, indicative of a pronounced tolerance threshold. The subsequent phylogenetic analysis, bolstered by robust genomic data from conserved housekeeping genes, including 16S rDNA, gyr B gene sequencing, as well as dnaK and recA, delineated a species-level phylogenetic tree, thereby confirming the strains as Bacillus atrophaeus. Additionally, we identified the types of iron-carrier-producing strains as catechol (WL1210) and carboxylic acid ferrophilin (CD303). A genomic analysis uncovered functional genes in strain CD303 associated with plant growth and iron carrier biosynthesis, such as fnr and iscA. Ryegrass seed germination assays, alongside morphological and physiological evaluations under diverse heavy metal stress, underscored the strains' potential to enhance ryegrass growth under high cadmium stress when treated with bacterial suspensions. This insight probes the strains' utility in leveraging alpine microbial resources and promoting ryegrass proliferation.
Effect of combined microbes on plant tolerance to Zn-Pb contaminations

Abstract:The presence and composition of soil microbial communities has been shown to have a large impact on plant-plant interactions and consequently plant diversity and composition. The goal of the present study was to evaluate impact of arbuscular mycorrhizal fungi (AMF) and nitrogen-fixing bacteria, which constitutes an essential link between the soil and the plant's roots. A greenhouse pot experiment was conducted to evaluate the feasibility of using selected microbes to improve Hieracium pilosella and Medicago sativa growth on Zn-Pb-rich site. Results of studies revealed that biomass, the dry mass of shoots and roots, increased significantly when plants were inoculated with mycorrhizal fungi and nitrogen-fixing bacteria. The addition of Azospirillum sp. and Nostoc edaphicum without mycorrhiza suppressed plant growth. Single bacterial inoculation alone does not have a positive effect on M. sativa growth, while co-inoculation with AMF improved plant growth. Plant vitality (expressed by the performance index) was improved by the addition of microbes. However, our results indicated that even dry heat sterilization of the substratum created imbalanced relationships between soil-plant and plants and associated microorganisms. The studies indicated that AMF and N2-fixers can improve revegetation of heavy metal-rich industrial sites, if the selection of interacting symbionts is properly conducted.
Plant growth-promoting and heavy metal-resistant Priestia and Bacillus strains associated with pioneer plants from mine tailings

Abstract:Open mine tailings dams are extreme artificial environments containing sizeable potentially toxic elements (PTEs), including heavy metals (HMs), transition metals, and metalloids. Furthermore, these tailings have nutritional deficiencies, including assimilable phosphorus sources, organic carbon, and combined nitrogen, preventing plant colonization. Bacteria, that colonize these environments, have mechanisms to tolerate the selective pressures of PTEs. In this work, several Priestia megaterium (formerly Bacillus megaterium), Bacillus mojavensis, and Bacillus subtilis strains were isolated from bulk tailings, anthills, rhizosphere, and endosphere of pioneer plants from abandoned mine tailings in Zacatecas, Mexico. Bacillus spp. tolerated moderate HMs concentrations, produced siderophores and indole-3-acetic acid (IAA), solubilized phosphates, and reduced acetylene in the presence of HMs. The strains harbored different PIB-type ATPase genes encoding for efflux pumps and Cation Diffusion Facilitator (CDF) genes. Moreover, nifH and nifD nitrogenase genes were detected in P. megaterium and B. mojavensis genomic DNA. They showed similarity with sequences of the beta-Proteobacteria species, which may represent likely horizontal transfer events. These Bacillus species precede the colonization of mine tailings by plants. Their phenotypic and genotypic features could be essential in the natural recovery of the sites by reducing the oxidative stress of HMs, fixing nitrogen, solubilizing phosphate, and accumulating organic carbon. These traits of the strains reflect the adaptations of Bacillus species to the mine tailings environment and could contribute to the success of phytoremediation efforts.
Role of Bacillus cereus in Improving the Growth and Phytoextractability of Brassica nigra (L.) K. Koch in Chromium Contaminated Soil

Abstract:Plant growth-promoting rhizobacteria (PGPR) mediate heavy metal tolerance and improve phytoextraction potential in plants. The present research was conducted to find the potential of bacterial strains in improving the growth and phytoextraction abilities of Brassica nigra (L.) K. Koch. in chromium contaminated soil. In this study, a total of 15 bacterial strains were isolated from heavy metal polluted soil and were screened for their heavy metal tolerance and plant growth promotion potential. The most efficient strain was identified by 16S rRNA gene sequencing and was identified as Bacillus cereus. The isolate also showed the potential to solubilize phosphate and synthesize siderophore, phytohormones (indole acetic acid, cytokinin, and abscisic acid), and osmolyte (proline and sugar) in chromium (Cr+3) supplemented medium. The results of the present study showed that chromium stress has negative effects on seed germination and plant growth in B. nigra while inoculation of B. cereus improved plant growth and reduced chromium toxicity. The increase in seed germination percentage, shoot length, and root length was 28.07%, 35.86%, 19.11% while the fresh and dry biomass of the plant increased by 48.00% and 62.16%, respectively, as compared to the uninoculated/control plants. The photosynthetic pigments were also improved by bacterial inoculation as compared to untreated stress-exposed plants, i.e., increase in chlorophyll a, chlorophyll b, chlorophyll a + b, and carotenoid was d 25.94%, 10.65%, 20.35%, and 44.30%, respectively. Bacterial inoculation also resulted in osmotic adjustment (proline 8.76% and sugar 28.71%) and maintained the membrane stability (51.39%) which was also indicated by reduced malondialdehyde content (59.53% decrease). The antioxidant enzyme activities were also improved to 35.90% (superoxide dismutase), 59.61% (peroxide), and 33.33% (catalase) in inoculated stress-exposed plants as compared to the control plants. B. cereus inoculation also improved the uptake, bioaccumulation, and translocation of Cr in the plant. Data showed that B. cereus also increased Cr content in the root (2.71-fold) and shoot (4.01-fold), its bioaccumulation (2.71-fold in root and 4.03-fold in the shoot) and translocation (40%) was also high in B. nigra. The data revealed that B. cereus is a multifarious PGPR that efficiently tolerates heavy metal ions (Cr+3) and it can be used to enhance the growth and phytoextraction potential of B. nigra in heavy metal contaminated soil.

Abstract:Plant growth–promoting rhizobacteria (PGPR) can promote plant growth and development with several beneficial effects, especially in challenging environmental conditions, such as the presence of toxic contaminants. In this study, 49 isolates obtained from Trifolium sp. nodules growing on a Pb/Zn mine site were characterized for PGP traits including siderophores production, phosphate solubilization, extracellular enzymes production, and antifungal activity. The isolates were also screened for their ability to grow at increasing concentrations of NaCl and heavy metals, including lead, zinc, cobalt, copper, nickel, cadmium, and chromium. The findings of our study indicated that isolates Cupriavidus paucula RSCup01-RSCup08, Providencia rettgeri RSPro01, Pseudomonas putida RSPs01, Pseudomonas thivervalensis RSPs03-RSPs09, and Acinetobacter beijerinckii RSAci01 showed several key traits crucial for promoting plant growth, thus demonstrating the greatest potential. Most isolates displayed resistance to salt and heavy metals. Notably, Staphylococcus xylosus RSSta01, Pseudomonas sp. RSPs02, Micrococcus yunnanensis RSMicc01, and Kocuria dechangensis RSKoc01 demonstrated a significant capacity to grow at salt concentrations ranging from 10 to 20%, and isolates including Cupravidus paucula RSCup01-RSCup08 exhibited resistance to high levels of heavy metals, up to 1300 mg/L Pb ++ , 1200 mg/L Zn ++ , 1000 mg/L Ni ++ , 1000 mg/L Cd ++ , 500 mg/L Cu ++ , 400 mg/L Co ++ , and 50 mg/L Cr VI+ . Additionally, the analysis revealed that metal-resistant genes pbr A, czc D, and ncc A were exclusively detected in the Cupriavidus paucula RSCup01 strain. The results of this study provide insights into the potential of plant growth-promoting rhizobacteria strains that might be used as inoculants to improve phytoremediation in heavy metal-contaminated soils.

Abstract:Microorganisms are cost-effective and eco-friendly alternative methods for removing heavy metals (HM) from contaminated agricultural soils. Therefore, this study aims to identify and characterize HM-tolerant (HMT) plant growth-promoting rhizobacteria (PGPR) isolated from industry-contaminated soils to determine their impact as bioremediators on HM-stressed pepper plants. Four isolates [ Pseudomonas azotoformans ( Pa ), Serratia rubidaea ( Sr ), Paenibacillus pabuli ( Pp ) and Bacillus velezensis ( Bv )] were identified based on their remarkable levels of HM tolerance in vitro . Field studies were conducted to evaluate the growth promotion and tolerance to HM toxicity of pepper plants grown in HM-polluted soils. Plants exposed to HM stress showed improved growth, physio-biochemistry, and antioxidant defense system components when treated with any of the individual isolates, in contrast to the control group that did not receive PGPR. The combined treatment of the tested HMT PGPR was, however, relatively superior to other treatments. Compared to no or single PGPR treatment, the consortia ( Pa+Sr+Pp+Bv ) increased the photosynthetic pigment contents, relative water content, and membrane stability index but lowered the electrolyte leakage and contents of malondialdehyde and hydrogen peroxide by suppressing the (non) enzymatic antioxidants in plant tissues. In pepper, Cd, Cu, Pb, and Ni contents decreased by 88.0-88.5, 63.8-66.5, 66.2-67.0, and 90.2-90.9% in leaves, and 87.2-88.1, 69.4-70.0%, 80.0-81.3, and 92.3%% in fruits, respectively. Thus, these PGPR are highly effective at immobilizing HM and reducing translocation in planta . These findings indicate that the application of HMT PGPR could be a promising "bioremediation" strategy to enhance growth and productivity of crops cultivated in soils contaminated with HM for sustainable agricultural practices.

Abstract:Microorganisms affect cadmium (Cd) extraction by hyperaccumulators to varying degrees, but the potential mechanism has not been completely studied. Here, two plant growth-promoting bacteria (PGPB, Bacillus paranthracis NT1 and Bacillus megaterium NCT-2) were assessed for their influence on Cd uptake by Solanum nigrum L. and their influence mechanisms. The results showed that both two strains could regulate phytohormones secretion, alleviate oxidative stress and promote S. nigrum growth when exposed to Cd (dry weight was significantly increased by 21.51% (strain NCT-2) and 21.23% (strain NT1) compared with the control, respectively). Additionally, strain NCT-2 significantly elevated the translocation factor (TF) and bioconcentration factor (BCF), and thus significantly facilitated total Cd uptake by 41.80% of S. nigrum , whereas strain NT1 significantly reduced the BCF and TF, resulting in insignificant effect on total Cd uptake of S. nigrum compared with the control. Results of qPCR illustrated that the two strains influenced the detoxification of Cd in S. nigrum by affecting the expression of antioxidant enzyme genes and gene PDR2 . Moreover, the differential expression of heavy metal transport genes IRT1 and HMA may lead to the difference of Cd accumulation in S. nigrum . Principal component analysis and Pearson correlation coefficient analysis further verified the positive roles of salicylic acid and indole-3-acetic acid on Cd detoxification of S. nigrum , and the positive correlation relationship between transportation of Cd from underground to shoot, plant biomass and Cd uptake. Altogether, our results demonstrated that these two PGPB have great potential in helping plants detoxify Cd and could provide insights into the mechanism of PGPB-assisted phytoremediation of Cd-contaminated soil.

Abstract:The quality of soil containing heavy metals (HMs) around nonferrous metal mining areas is often not favorable for plant growth . Three types of plant growth promoting rhizobacteria (PGPR)-assisted ryegrass were examined here to treat Cd, Pb, and Zn contaminated soil collected from a nonferrous metal smelting facility. The effects of PGPR-assisted plants on soil quality, plant growth, and the migration and transformation of HMs were evaluated. Results showed that inter-root inoculation of PGPR to ryegrass increased soil redox potential , urease, sucrase and acid phosphatase activities, microbial calorimetry, and bioavailable P, Si, and K content. Inoculation with PGPR also increased aboveground parts and root length, P, Si, and K contents, and antioxidant enzyme activities. The most significant effect was that the simultaneous inoculation of all three PGPRs increased the ryegrass extraction (%) of Cd (59.04–79.02), Pb (105.56–157.13), and Zn (27.71–40.79), compared to CK control (without fungi). Correspondingly, the inter-root soil contents (%) of total Cd (39.94–57.52), Pb (37.59–42.17), and Zn (34.05–37.28) were decreased compared to the CK1 control (without fungi and plants), whereas their bioavailability was increased. Results suggest that PGPR can improve soil quality in mining areas, promote plant growth, transform the fraction of HMs in soil, and increase the extraction of Cd, Pb, and Zn by ryegrass. PGPR is a promising microbe-assisted phytoremediation strategy that can promote the re-greening of vegetation in the mining area while remediating HMs pollution.

Assessment of PGP traits of Bacillus cereus NDRMN001 and its influence on Cajanus cajan (L.) Millsp. phytoremediation potential on metal-polluted soil under controlled conditions

Abstract:The current study looked at the plant growth-promoting (PGP) traits of the pre-isolated and metal-tolerant Bacillus cereus NDRMN001 as well as their stimulatory effect on the physiology, biomolecule content, and phytoremediation potential of Cajanus cajan (L.) Millsp. on metal-polluted soil. The bauxite mine, which is surrounded by farmland (1 km away), has been severely polluted by metals such as Cd (31.24 ± 1.68), Zn (769.57 ± 3.46), Pb (326.85 ± 3.43), Mn (2519.6 ± 5.71), and Cr (302.34 ± 1.62 mg kg-1) that exceeded Indian standards. The metal-tolerant B. cereus NDRMN001 had excellent PGP activities such as synthesis of hydrogen cyanide (HCN), siderophore, indole acetic acid (IAA), N2 fixation, and P solubilization. Furthermore, the optimal growth conditions (temperature of 30°C, pH 6.5, 6% glucose, 9% tryptophan, and 1.5% tricalcium phosphate) for effective synthesis and expression of PGP traits in B. cereus NDRMN001 were determined. Such metal-tolerant B. cereus NDRMN001 traits can significantly reduce metals in polluted soil, and their PGP traits significantly improve plant growth in polluted soil. Hence, this strain (B. cereus NDRMN001) significantly improved the growth and phytoremediation potential of C. cajan (L.) Millsp on metal-polluted soil without [study I: 2 kg of sieved and autoclaved metal-polluted soil seeded with bacterium-free C. cajan (L.) Millsp. seeds] and with [study II: 2 kg of sieved and autoclaved metal-polluted soil seeded with B. cereus NDRMN001-coated C. cajan (L.) Millsp. seeds] B. cereus NDRMN001 amalgamation. Fertile soil was used as control. The physiological parameters, biomolecule contents, and the phytoremediation (Cr: 7.74, Cd: 12.15, Zn: 16.72, Pb: 11.47, and Mn: 14.52 mg g-1) potential of C. cajan (L.) Millsp. were significantly effective in study II due to the metal-solubilizing and PGP traits of B. cereus NDRMN001. These results conclude that the test bacteria B. cereus NDRMN001 considerably improved the phytoremediation competence of C. cajan (L.) Millsp. on metal-polluted soil in a greenhouse study.

Abstract:Heavy metal-resistant plant growth-promoting bacteria may help in reducing the toxic metal accumulation within the plants grown in metal-contaminated soils and thus be effective against biomagnification, enhancing food safety. In the present study, the most potential multi-metal resistant bacterium Pseudomonas aeruginosa JCM 5962 (obtained from arable soil of Uttar Dinajpur) was utilized for Cd biosorption analysis and also to observe the in vivo growth effect on Brassica plant including the estimation of Cd content accumulated within the plant parts of Brassica grown in Cd-contaminated soil. The maximum sorption of Cd ions by the dead bacterial biomass of Pseudomonas aeruginosa JCM 5962 was estimated to be 149.25 mg/g. The adsorption data provided an outstanding fit to the Freundlich isotherm model, indicating multilayer adsorption at heterogeneous surfaces. RL value (0.76, i.e.; <1) implied favourable adsorption. The in vivo experiment revealed the increased length of both roots and shoots in Brassica seedlings grown in Cd-contaminated soil under the effect of bacterial inoculants, which was also statistically significant (p < 0.001). In addition, the studied bacterium was effective in lowering Cd accumulation within the plant parts of Brassica. Approximately 62.76% reduction in Cd content was observed in Brassica seedlings grown in Cd-contaminated soil under the effect of bacterial inoculants than in the non-inoculated control set. This is a pioneering study, which could be further employed to develop strategies for microbe-assisted phytoremediation.
Effect of Plant Growth-Promoting Bacteria on Biometrical Parameters and Antioxidant Enzymatic Activities of Lupinus albus var. Orden Dorado Under Mercury Stress

Abstract:Heavy metal contamination of soils is a large-scale environmental problem. It leads to significant disqualification of the territory, in addition to being a source of the potential risk to human health. The exposure of plants to mercury (Hg) generates responses in its growth and their oxidative metabolism. The impact of increasing concentrations of Hg on the development of Lupinus albus var. Orden Dorado seedlings has been studied, as well as the plant's response to the maximum concentration of Hg that allows its development (16 μg ml –1 ). The result shows that only the inoculum with plant growth promoting bacteria (PGPB) allows the biometric development of the seedling (root length, weight, and number of secondary roots) and prevents the toxic effects of the heavy metal from aborting the seedlings. Specifically, treatments with strains 11, 20 ( Bacillus toyonensis ), 48 (not determined), and 76 ( Pseudomonas syringae ) are interesting candidates for further PGPB-assisted phytoremediation trials as they promote root biomass development, through their PGPB activities. The plant antioxidant response has been analyzed by quantifying the catalase (CAT), superoxide dismutase (SOD), ascorbate peroxidase (APX), and glutathione reductase (GR) enzyme activity in the root, under 16 μg ml –1 of HgCl 2 and different PGPB treatments. Results show that, although Hg stress generally induces enzyme activity, strains 31 and 69I ( Pseudomonas corrugata ) and 18 and 43 ( Bacillus toyonensis ) can keep SOD and APX levels close to those found in control without Hg ( p < 0.01). Strain 18 also shows a significant reduction of GR to control levels without Hg. The present work demonstrates the benefit of PGPB treatments in situations of high Hg stress. These findings may be a good starting point to justify the role of PGPB naturally isolated from bulk soil and the rhizosphere of plants subjected to high Hg pressure in plant tolerance to such abiotic stress conditions. More studies will be needed to discover the molecular mechanisms behind the phytoprotective role of the strains with the best results, to understand the complex plant-microorganism relationships and to find effective and lasting symbioses useful in bioremediation processes.

Abstract:Plant-growth-promoting bacteria (PGPB) help plants thrive in polluted environments and increase crops yield using fewer inputs. Therefore, the design of tailored biofertilizers is of the utmost importance. The purpose of this work was to test two different bacterial synthetic communities (SynComs) from the microbiome of Mesembryanthemum crystallinum, a moderate halophyte with cosmetic, pharmaceutical, and nutraceutical applications. The SynComs were composed of specific metal-resistant plant-growth-promoting rhizobacteria and endophytes. In addition, the possibility of modulating the accumulation of nutraceutical substances by the synergetic effect of metal stress and inoculation with selected bacteria was tested. One of the SynComs was isolated on standard tryptone soy agar (TSA), whereas the other was isolated following a culturomics approach. For that, a culture medium based on M. crystallinum biomass, called Mesem Agar (MA), was elaborated. Bacteria of three compartments (rhizosphere soil, root endophytes, and shoot endophytes) were isolated on standard TSA and MA media, stablishing two independent collections. All bacteria were tested for PGP properties, secreted enzymatic activities, and resistance towards As, Cd, Cu, and Zn. The three best bacteria from each collection were selected in order to produce two different consortiums (denominated TSA- and MA-SynComs, respectively), whose effect on plant growth and physiology, metal accumulation, and metabolomics was evaluated. Both SynComs, particularly MA, improved plant growth and physiological parameters under stress by a mixture of As, Cd, Cu, and Zn. Regarding metal accumulation, the concentrations of all metals/metalloids in plant tissues were below the threshold for plant metal toxicity, indicating that this plant is able to thrive in polluted soils when assisted by metal/metalloid-resistant SynComs and could be safely used for pharmaceutical purposes. Initial metabolomics analyses depict changes in plant metabolome upon exposure to metal stress and inoculation, suggesting the possibility of modulating the concentration of high-value metabolites. In addition, the usefulness of both SynComs was tested in a crop plant, namely Medicago sativa (alfalfa). The results demonstrate the effectiveness of these biofertilizers in alfalfa, improving plant growth, physiology, and metal accumulation.

Abstract:Four plant growth-promoting bacteria (PGPB) were used as study materials, among them two heavy metal-tolerant rhizosphere strains SrN1 (Arthrobacter sp.) and SrN9 (Bacillus altitudinis) were isolated from rhizosphere soil, while two endophytic strains SaN1 (Bacillus megaterium) and SaMR12 (Sphingomonas) were identified from roots of the cadmium (Cd)/zinc (Zn) hyperaccumulator Sedum alfredii Hance. A pot experiment was carried out to investigate the effects of these PGPB on plant growth and Cd accumulation of oilseed rape (Brassica napus) plants grown on aged Cd-spiked soil. The results showed that the four PGPB significantly boosted oilseed rape shoot biomass production, improved soil and plant analyzer development (SPAD) value, enhanced Cd uptake of plant and Cd translocation to the leaves. By fluorescent in situ hybridization (FISH) and green fluorescent protein (GFP), we demonstrated the studied S. alfredii endophytic bacterium SaMR12 were able to colonize successfully in the B. napus roots. However, all four PGPB could increase seed Cd accumulation. Due to its potential to enhance Cd uptake by the plant and to restrict Cd accumulation in the seeds, SaMR12 was selected as the most promising microbial partner of B. napus when setting up a plant-microbe fortified remediation system.

Abstract:Phytoremediation is considered as a novel environmental friendly technology, which uses plants to remove or immobilize heavy metals. The use of metal-resistant plant growth-promoting bacteria (PGPB) constitutes an important technology for enhancing biomass production as well as tolerance of the plants to heavy metals. In this study, we isolated twenty seven (NF1-NF27) chromium resistant bacteria. The bacteria were tested for heavy metals (Cr, Zn, Cu, Ni, Pb and Co) resistance, Cr(VI) reduction and PGPB characters (phosphate solubilization, production of IAA and siderophores). The results showed that the bacterial isolates resist to heavy metals and reduce Cr(VI), with varying capabilities. 37.14% of the isolates have the capacity of solubilizing phosphate, 28.57% are able to produce siderophores and all isolates have the ability to produce IAA. Isolate NF2 that showed high heavy metal resistance and plant growth promotion characteristics was identified by 16S rDNA sequence analysis as a strain of Cellulosimicrobium sp.. Pot culture experiments conducted under greenhouse conditions showed that this strain was able to promote plant growth of alfalfa in control and in heavy metals (Cr, Zn and Cu) spiked soils and increased metal uptake by the plants. Thus, the potential of Cellulosimicrobium sp. for both bioremediation and plant growth promotion has significance in the management of environmental pollution.

Abstract:Microorganisms showed unique mechanisms to resist and detoxify harmful metals in response to pollution. This study shows the relationship between presence of heavy metals and plant growth regulator compounds. Additionally, the responses of Rhodotorula mucilaginosa YR29 isolated from the rhizosphere of Prosopis sp. growing in a polluted mine jal in Mexico are presented. This research carries out a phenotypic characterization of R . mucilaginosa to identify response mechanisms to metals and confirm its potential as a bioremediation agent. Firstly, Plant Growth-Promoting (PGP) compounds were assayed using the Chrome Azurol S (CAS) medium and the Salkowski method. In addition, to clarify its heavy metal tolerance mechanisms, several techniques were performed, such as optical microscopy, scanning electron microscopy (SEM) and transmission electron microscopy (TEM) supplemented with assorted detectors. Scanning transmission electron microscopy (STEM) was used for elementary mapping of the cell. Finally, yeast viability after all treatments was confirmed by confocal laser scanning microscopy (CLSM). The results have suggested that R. mucilaginosa could be a PGP yeast capable of triggering Pb 2+ biosorption (representing 22.93% of the total cell surface area, the heavy metal is encapsulated between the cell wall and the microcapsule), and Pb 2+ bioaccumulation (representing 11% of the total weight located in the vacuole). Based on these results, R. mucilaginosa as a bioremediation agent and its wide range of useful mechanisms for ecological purposes are highlighted.

Abstract:Plant growth promoting bacteria (PGPB) have been reported to have the ability to promote plant growth, development and increase heavy metals (HMs) uptake. Therefore, PGPB inoculation as soil remediation agents into plants with larger biomass and potential of phytoextraction is of great importance to increase bioremediation efficiency. In this study, 12 PGPB strains isolated from a cadmium (Cd)/zinc hyperaccumulator Sedum alfredii Hance were inoculated into non-host plant Brassica juncea and their effects on plant growth and Cd uptake were determined. The results showed that inoculation of most PGPB strains promoted plant growth, boosted root development and improved chlorophyll content in the absence of Cd. Inoculation of PGPB strains promoted plant growth up to 111% in shoot and 358% in root when treated with 2 μM Cd. In addition, PGPB inoculation not only ameliorated plant root morphology including the total root length (RL), total surface area (SA), total root volume (RV) and number of root tips (RT), but also facilitated Cd uptake up to 126%. Furthermore, inoculation of PGPB strains promoted plant Cd accumulation up to 261% in shoot and up to 8.93-fold increase in root. Among all the 12 PGPB strains, Burkholdria SaMR10 and Sphingomonas SaMR12 were identified as the promising microbes for improving phytoremediation efficiency of Cd contaminated soils. These results not only provided useful findings for further investigation of interacting mechanisms between different bacterial strains and plants, but also facilitated the development of microbe-assisted phytoremediation application for HM contaminated soil.

Abstract:Bioremediation of Cd contaminated environments can be assisted by plant-growth-promoting bacteria (PGPB) enabling plant growth in these sites. Here a gram-negative Burkholderia contaminans ZCC was isolated from mining soil at a copper-gold mine. When exposed to Cd(II), ZCC displayed high Cd resistance and the minimal inhibitory concentration was 7 mM in LB medium. Complete genome analysis uncovered B. contaminans ZCC contained 3 chromosomes and 2 plasmids. One of these plasmids was shown to contain a multitude of heavy metal resistance determinants including genes encoding a putative Cd-translocating PIB-type ATPase and an RND-type related to the Czc-system. These additional heavy metal resistance determinants are likely responsible for the increased resistance to Cd(II) and other heavy metals in comparison to other strains of B. contaminans. B. contaminans ZCC also displayed PGPB traits such as 1-aminocyclopropane-1-carboxylate deaminase activity, siderophore production, organic and inorganic phosphate solubilization and indole acetic acid production. Moreover, the properties and Cd(II) binding characteristics of extracellular polymeric substances was investigated. ZCC was able to induce extracellular polymeric substances production in response to Cd and was shown to be chemically coordinated to Cd(II). It could promote the growth of soybean in the presence of elevated concentrations of Cd(II). This work will help to better understand processes important in bioremediation of Cd-contaminated environment.

Abstract:Due to a variety of natural and anthropogenic processes, heavy metal toxicity of soil constitutes a substantial hazard to all living beings in the environment. The heavy metals alter the soil properties, which directly or indirectly influence the agriculture systems. Thus, plant growth–promoting rhizobacteria (PGPR)–assisted bioremediation is a promising, eco-friendly, and sustainable method for eradicating heavy metals. PGPR cleans up the heavy metal–contaminated environment using various approaches including efflux systems, siderophores and chelation, biotransformation, biosorption, bioaccumulation, precipitation, ACC deaminase activity, biodegradation, and biomineralization methods. These PGPRs have been found effective to bioremediate the heavy metal–contaminated soil through increased plant tolerance to metal stress, improved nutrient availability in soil, alteration of heavy metal pathways, and by producing some chemical compounds like siderophores and chelating ions. Many heavy metals are non-degradable; hence, another remediation approach with a broader scope of contamination removal is needed. This article also briefly emphasized the role of genetically modified PGPR strains which improve the soil's degradation rate of heavy metals. In this regard, genetic engineering, a molecular approach, could improve bioremediation efficiency and be helpful. Thus, the ability of PGPRs can aid in heavy metal bioremediation and promote a sustainable agricultural soil system.
