Abstract:Purpose: To determine the role of glucose metabolism on cartilage development in postnatal developing bone in normal and IGF-II knockout mice. Methods: All animal care and experimental procedures were approved by the Tufts IACUC. Histological analysis. Samples were fixed in PFA, paraffin sectioned, and stained. In vitro glucose metabolism assays. The following kits were used: Glucose (HK) assay kit (Sigma), L-lactate assay kit I (Eton Biosciences), ATP Determination Kit (Invitrogen), Oxygen consumption (OCR) kit (Cayman Chemical), and Image-iT Live Green Reactive Oxygen Species Detection Kit (Invitrogen). Metatarsal bones ex vivo culture. Isolated bones were cultured in DMEM supplemented with 0.25% FBS, 50μg/mL ascorbic acid, and 1.25mM sodium pyruvate for 7 days in the absence or presence of 10mg/mL IGF-II or 1μM 3-bromopyruvate (3-BrPA). Microscopy. Two-photon excited fluorescence microscopy. NADH fluorescence images were acquired with an excitation=755nm and an emission=460 (± 20) nm filter. FAD images were acquired with an excitation=860 nm and an emission=525 (±25) nm filter. Statistical analysis. Data were reported as mean ± standard deviation, and analyzed using an unpaired t-test or one-way analysis of variance followed by post-hoc Dunnett's test. p <0.05 is considered significant. Results: 1. IGF-II regulates glucose metabolism in growth plate chondrocytes. The growth plate of Igf-II null was generally well formed, but its hypertrophic zone (HZ) was proportionally larger. In addition, there was a clear delay in secondary ossification center formation in the mutant, which contributed to the shortened cartilage template (Fig. A). We assessed glucose metabolism in postnatal epiphyseal chondrocytes cultured in alginate beads. Igf-II null chondrocytes had twice the rate of glucose consumption compared to WT cells, which was restored to basal levels upon exogenous IGF-II treatment of the mutant cells (Fig. B). Consistently, Igf-II null chondrocytes exhibited an increased intracellular ATP level, the lactic acid level, OCR and ROS levels, which were restored to WT levels by exogenous IGF-II (Fig. B). These results indicate that there is an overall increase in glucose metabolism in the Igf-II null cells, including both anaerobic glycolysis and oxidative phosphorylation. To investigate glucose metabolism in vivo, we measured redox levels through two-photon fluorescence imaging. This imaging method capitalizes on the endogenous fluorescence emitted by two key coenzymes in glucose metabolism: the oxidized form of flavin adenine dinucleotide (FAD) and reduced form of nicotinamide dinucleotide (NADH). The ratio of their fluorescence intensities FAD+/(FAD++NADH) thus constitutes the optical redox ratio, and reflects the balance of glycolysis and oxidative phosphorylation. We found that Igf-II null chondrocytes had a lower redox ratio, which is suggestive of increased oxidative phosphorylation in comparison with anaerobic glycolysis (Fig. C). Exogenously added IGF-II elevated the optical redox ratios of the mutant to the WT levels in metatarsal bones in ex vivo cultures (Fig. D). 2. Attenuating glycolysis restores normal longitudinal growth rate and chondrocyte maturation in Igf-II null bones. To address whether increased glucose metabolism is the cause of cartilage phenotype, we applied a widely used glycolysis inhibitor, 3-bromopyruvate (3-BrPA), to Igf-II null bones in ex vivo cultures. Significantly, 3-BrPA treatment restored the rate of longitudinal growth of the Igf-II null bone to WT bone levels, leading to a decrease in the length of the hypertrophic zone and an increase in the ratio of the columnar zone over the hypertrophic zone (Fig. E). Conclusions: 1. Igf-II is critical for regulating postnatal bone growth, which is consistent with a recent finding connecting IGF-II mutation to growth restriction. 2. A novel link between glucose metabolism and cartilage development was discovered. By keeping the balance of anaerobic glycolysis and oxidative phosphorylation under control, IGF-II regulates the pace of endochondral ossification (Fig. F).
