Abstract:Exosomes are small extracellular vesicles involved in many physiological activities of cells in the human body. Exosomes from cancer cells have great potential to be applied in clinical diagnosis, early cancer detection and target identification for molecular therapy. While this field is gaining increasing interests from both academia and industry, barriers such as supersensitive detection techniques and highly-efficient isolation methods remain. In the clinical settings, there is an urgent need for rapid analysis, reliable detection and point-of-care testing (POCT). With these challenges to be addressed, this article aims to review recent developments and technical breakthroughs including optical, electrochemical and electrical biosensors for exosomes detection in the field of cancer and other diseases and demonstrate how nanobiosensors could enhance the performance of conventional sensors. Working strategies, limit of detections, advantages and shortcomings of the studies are summarized. New trends, challenges and future perspectives of exosome-driven POCT in liquid biopsy have been discussed.

Abstract:Liver cancer is one of the deadliest types worldwide and early diagnosis is highly important for successful treatment. Therefore, it is necessary to develop rapid, sensitive, simple, and inexpensive analytical tools for its detection. MicroRNAs (miRNA) represent unique biomarkers whose expression in biofluids is strongly associated with cancer in general and miR-21, -31, -122, -145, -146a, -200c, -221, -222, and -223 in liver cancer, specifically. Various biosensors for miRNA detection have been developed. These include electrochemical biosensors based on amperometric, potentiometric, conductometric and impedimetric technology. Furthermore, the use of advanced nanomaterials with enhanced chemical stability, conductivity and electrocatalytic activity have greatly increased the sensitivity and specificity of these devices. The present review focuses on recent advances in electrochemical biosensors for miRNA detection in liver cancer.

Abstract:Ovarian cancer, known as "silent killer", is a gynocological cancer with high mortality that usually diagnosed in the late stages. Gold standard immunoassay technique is evaluation of CA-125 levels which is not merely specific to ovarian cancer. Therefore, there is a need for sensitive determination of more specific biomarkers. miR-200 family is RNA nucleic acids that known to be upregulated in the presence of ovarian cancer. Since diagnosis based on a single biomarker is prone to generate misleading results, it is important to develop point-of-care systems that allow diagnosis of multiple miRNAs. Herein, an electrochemical nanobiosensor platform was developed for the multiplexed and simultaneous detection of miR-200c and miR-141. Biorecognition part was constitutued of methylene blue and ferrocene labeled hairpin DNA probes immobilized onto carboxylated graphene oxide modified pencil graphite electrodes. Their hybridization with miRNAs were examined upon "signal-off" approach using Square Wave Voltammetry. The platform demonstrated a linear detection range of 0.1 pM to 10 nM for both miR-141 and miR-200c, with low detection limits of 0.029 pM and 0.026 pM, respectively. We assume that the developed biosensor platform may pave the way in early diagnosis of the disease and the development of more effective treatment strategies.

Abstract:Background: Cancer is known as one of the main non-communicable diseases and the leading cause of death in the new era. Early diagnosis of cancer requires the identification of special biomarkers. Currently, microRNAs (miRNAs) have attracted the attention of researchers as useful biomarkers for cancer early detection. Hence, various methods have been recently developed for detecting and monitoring miRNAs. Among all miRNAs, detection of miRNA-21 (miR-21) is important because it is abnormally overexpressed in most cancers. Here, a new biosensor based on silver nanoclusters (AgNCs) is introduced for detecting miR-21. Results: As a fluorescent probe, a rationally designed hairpin sequence containing a poly-cytosine motif was used to facilitate the formation of AgNCs. A guanine-rich sequence was also employed to enhance the sensing signal. It was found that in the absence of miR-21, adding a guanine-rich sequence to the detecting probe caused only a slight change in the fluorescence emission intensity of AgNCs. While in the presence of miR-21, the emission signal enhanced. A direct correlation was observed between the increase in the fluorescence of AgNCs and the concentration of miR-21. The performance of the proposed biosensor was characterized thoroughly and confirmed. The biosensor detected miR-21 in an applicable linear range from 9 pM to 1.55 nM (LOD: 2 pM). Significance: The designed biosensor was successfully applied for detecting miR-21 in human plasma samples and also in human normal and lung and ovarian cancer cells. This biosensing strategy can be used as a model for detecting other miRNAs. The designed nanobiosensor can measure miR-21 without using any enzymes, with fewer experimental steps, and at a low cost compared to the reported biosensors in this field.

Abstract:Ovarian cancer is one of the most aggressive and significant malignant tumor forms in the female reproductive system. It is the leading cause of death among gynecological cancers owing to its metastasis. Since its preliminary disease symptoms are lacking, it is imperative to develop early diagnostic biomarkers to aid in treatment optimization and personalization. In this vein, microRNAs, which are short sequence non-coding molecules, displayed great potential as highly specific and sensitive biomarker. miRNAs have been extensively advocated and proven to serve an instrumental part in the clinical management of cancer, especially ovarian cancer, by promoting the cancer cell progression, invasion, delayed apoptosis, epithelial-mesenchymal transition, metastasis of cancer cells, chemosensitivity and resistance and disease therapy. Here, we cover our present comprehension of the most up-to-date microRNA-based approaches to detect ovarian cancer, as well as current diagnostic and treatment strategies, the role of microRNAs as oncogenes or tumor suppressor genes, and their significance in ovarian cancer progression, prognosis, and therapy.

Abstract:Ovarian cancer is a significant health concern among the female vulnerable populations due to delayed diagnosis. Point-of-care devices that can be utilized for mass screening of the population may improve patient survival through early diagnosis. In this context, we report an ultrasensitive DNA-based nano-biosensor for the quantification of cancer biomarker miRNA-21 using a label-free sensing technique. Magnetite selenium ( ) nanocomposite was prepared by co-precipitation method followed by characterization using TEM, EDX, FTIR, VSM, and XRD. The average size of the nanomaterial was found to be 20 nm with a low polydispersity index of 0.2. The biosensor was fabricated by immobilizing a single-stranded DNA probe specific to miRNA-21 onto the nanocomposite modified electrode surface. At the working electrode/electrolyte interface, the binding of the DNA probe results in decreased charge transfer impedance. The developed magnetite-based impedimetric biosensor demonstrated a linear variation of resistance to charge transfer with miRNA-21 concentration (75 zM to 0.751 pM). The ultrasensitive low limits of detection (75 zM), the sensitivity of 1774.34 / g/ L/ and high selectivity over other biomarkers (miRNAs–miRNA-30e, miRNA-143, miRNA-204, DNA, protein, and cocktail of all makes it suitable for early diagnosis. Furthermore, owing to miRNA-21 prognostic as well as diagnostic value, the magnetite-based miRNA-21 impedimetric biosensor offers the promise of a sensitive consumer-centric device for mass screening of ovarian cancer.

Abstract:M are a class of non-protein-coding small RNA molecules with 17-25 nucleotides. The expression of microRNAs is related to some biological processes including cell differentiation, apoptosis, proliferation, and immunological response. Recently, microRNAs have emerged as new candidate diagnostic and prognostic biomarkers for the detection of a wide variety of cancers such as hematopoietic cancers, breast, lung, and colon cancers. Due to short sequences and high sequence homology among family members, the quantitative microRNAs analysis is still a challenge. Electrochemical biosensors have attracted increasing attention because of its low cost, convenient operation, rapid detection, and good sensitivity. To further improve electrochemical signals, labeling methods integrated with signal amplification strategies such as nanoparticles-based amplification, enzymatic signal amplification, and rolling circle amplification etc. have been developed. In our researches, a novel electrochemical biosensor with triple signal amplification for the ultrasensitive detection of microRNA was developed based on phosphatase, redox-cycling amplification, bimetallic metal alloy supported graphene functionalized gold electrode, and two stem-loop structured DNA were used as target capturers. In addition, another ultrasensitive electrochemical genosensor based on two DNA as the target capturers and metal functionalized nanoparticles as a label is employed for the detection of microRNA at an attomole level. These proposed biosensors are sufficiently selective to discriminate the target microRNA from homologous microRNAs and attractive candidates for the development of accurate, selective, and ultrasensitive methods for microRNA expression profiling and clinical diagnostics.

Abstract:Currently, early diagnosis of breast cancer with novel biomarkers of microRNAs has developed as promising approach in medicine and preventing unnecessary surgery. In this work, we designed a colorimetric biosensor based on the catalytic and peroxidase-like properties of copper oxide nanoparticles (CuO) in the interaction with DNA. The mimic enzymatic activity with combination to no PCR amplification strategy of HCR can be easily use to detect multiple microRNAs, simultaneously. The biosensing method was confirmed using UV spectroscopy, XRD, FESEM, and electrophoresis analysis. The results showed a high specificity and sensitivity for dual targets (miR-21 and miR-155) with a detection limit of 0.31 nM and a linear response range of 0.1-1 μM. It was found that the approach could provide a cheap, fast, and reliable multiple detection technique to diagnose and monitor breast cancer in patients, with high accuracy and repeatability.

Abstract:OBJECTIVE:This review aimed to update the progress of microRNA (miRNA) in early detection of ovarian cancer. We discussed the current clinical diagnosis methods and biomarkers of ovarian cancer, especially the methods of miRNA in early detection of ovarian cancer.DATA SOURCES:We collected all relevant studies about miRNA and ovarian cancer in PubMed and CNKI from 1995 to 2015.STUDY SELECTION:We included all relevant studies concerning miRNA in early detection of ovarian cancer, and excluded the duplicated articles.RESULTS:miRNAs play a key role in various biological processes of ovarian cancer, such as development, proliferation, differentiation, apoptosis and metastasis, and these phenomena appear in the early-stage. Therefore, miRNA can be used as a new biomarker for early diagnosis of ovarian cancer, intervention on miRNA expression of known target genes, and potential target genes can achieve the effect of early prevention. With the development of nanoscience and technology, analysis methods of miRNA are also quickly developed, which may provide better characterization of early detection of ovarian cancer.CONCLUSIONS:In the near future, miRNA therapy could be a powerful tool for ovarian cancer prevention and treatment, and combining with the new analysis technology and new nanomaterials, point-of-care tests for miRNA with high throughput, high sensitivity, and strong specificity are developed to achieve the application of diagnostic kits in screening of early ovarian cancer.

Abstract:Early detection of ovarian cancer is crucial for improving treatment outcomes and increasing survival rates, as this malignancy is often diagnosed at an advanced stage due to the lack of specific early symptoms. In this study, we present a novel diagnostic platform integrating platinum nanozyme-induced cascade signal amplification with a gas-driven visual amplification chip (VAC) for the sensitive and specific detection of the ovarian cancer biomarkers CA125 and HE4. This system leverages a sandwich complex formed by capture antibody-functionalized magnetic silica nanoparticles (Ab1-MSNPs) and detection antibody-functionalized platinum nanoparticles (Ab2-PtNPs) in the presence of target antigens. The PtNPs’ peroxidase-like activity catalyzes dopamine polymerization, leading to polydopamine (PDA) deposition and subsequent capture of 4-mercaptophenylboronic acid-modified PtNPs, amplifying the detection signal. The VAC achieved exceptional sensitivity, with detection limits as low as 0.001 U mL−1 for CA125 and 0.1 pg mL−1 for HE4, surpassing traditional methods. Finally, the platform demonstrated a strong correlation with electrochemiluminescence assays, making it a promising tool for point-of-care testing and early ovarian cancer detection in clinical settings.

Abstract:Breast cancer continues to be a significant contributor to global cancer deaths, particularly among women. This highlights the critical role of early detection and treatment in boosting survival rates. While conventional diagnostic methods like mammograms, biopsies, ultrasounds, and MRIs are valuable tools, limitations exist in terms of cost, invasiveness, and the requirement for specialized equipment and trained personnel. Recent shifts towards biosensor technologies offer a promising alternative for monitoring biological processes and providing accurate health diagnostics in a cost-effective, non-invasive manner. These biosensors are particularly advantageous for early detection of primary tumors, metastases, and recurrent diseases, contributing to more effective breast cancer management. The integration of biosensor technology into medical devices has led to the development of low-cost, adaptable, and efficient diagnostic tools. In this framework, electrochemical screening platforms have garnered significant attention due to their selectivity, affordability, and ease of result interpretation. High Fidelity Detection of miRNAs from Complex Physiological Samples Through Electrochemical Nanosensors Empowered by Proximity Catalysis and Magnetic Separation

Abstract:Electrochemical detection of miRNA biomarkers in complex physiological samples holds great promise for accurate evaluation of tumor burden in the perioperative period, yet limited by reproducibility and bias issues. Here, nanosensors installed with hybrid probes that responsively release catalytic DNAzymes (G-quadruplexes/hemin) were developed to solve the fidelity challenge in an immobilization-free detection. miRNA targets triggered toehold-mediated strand displacement reactions on the sensor surface and resulted in amplified shedding of DNAzymes. Subsequently, the interference background was removed by Fe 3 O 4 core-facilitated magnetic separation. Binding aptamers of the electrochemical reporter (dopamine) were tethered closely to the catalytic units for boosting H 2 O 2 -mediated oxidation through proximity catalysis. The one-to-many conversion by dual amplification from biological-chemical catalysis facilitated sufficient homogeneous sensing signals on electrodes. Thereby, the nanosensor exhibited a low detection limit (2.08 fM), and high reproducibility (relative standard deviation of 1.99%). Most importantly, smaller variations (RSD of 0.51-1.04%) of quantified miRNAs were observed for detection from cell lysates, multiplexed detection from unprocessed serum, and successful discrimination of small upregulations in lysates of tumor tissue samples. The nanosensor showed superior diagnostic performance with an area under curve (AUC) of 0.97 and 94% accuracy in classifying breast cancer patients and healthy donors. These findings demonstrated the synergy of signal amplification and interference removal in achieving high-fidelity miRNA detection for practical clinical applications.

Three-Way Electrochemical Sensing of Ultra-Low MicroRNA Levels

Abstract:MicroRNAs (miRNAs) are an emerging class of biomarkers that are frequently deregulated in cancer cells and have shown a great promise for cancer classification and prognosis. In this work, we developed a three-way electrochemical sensor for detection and quantitation of ultra-low amounts of miRNAs in a wide dynamic range of measured concentrations. The sensor facilitates three detection modes based on hybridization (H-SENS), p19 protein binding (P-SENS) and protein displacement (D-SENS). The combined HPD sensor (HPD-SENS) identifies as low as 5 aM or 90 molecules of miRNA per 30\muL of sample without PCR amplification, and can be operated within the dynamic range from 10 aM to 1 \muM. The HPD senor is made on a commercially-available gold nanoparticles-modified electrode and suitable for analysing multiple miRNAs on a single electrode. This three-way sensor exhibits high selectivity and specificity and was successfully used for sequential analysis of miR-32 and miR-122 on one electrode. Furthermore, the P-SENS was employed for profiling of three endogenous microRNAs (miR-21, miR-32 and miR-122) in human blood serum.

Abstract:Background MicroRNAs (miRNAs) are important non-coding RNA entities that affect gene expression and function by binding to target mRNAs, leading to degradation of the mRNAs or inhibiting their translation. miRNAs are widely involved in a variety of biological processes, such as cell differentiation, development, metabolism, and apoptosis. In addition, miRNAs are associated with many diseases, including cancer. However, conventional detection techniques often suffer from shortcomings such as low sensitivity, so we need to develop a rapid and efficient detection strategy for accurate detection of miRNAs. Results We have developed an innovative homogeneous electrochemiluminescence (ECL) biosensor. This biosensor employs CRISPR/Cas12a gene editing technology for accurate and efficient detection of microRNA (miRNA). Compared to conventional technologies, this biosensor employs a unique homogeneous detection format that eliminates laborious probe fixation steps and greatly simplifies the detection process. By using two amplification techniques - isothermal amplification and T7 RNA polymerase amplification - the biosensor improves the sensitivity and specificity of the assay, providing excellent detection performance in the assay. This makes it possible to evaluate miRNA directly from a variety of biological samples such as cell lysates and diluted human serum. Experimental results convincingly demonstrate the extraordinary performance of this biosensor, including its extremely low detection limit of 1.27 aM, high sensitivity, reproducibility and stability. Significance The application of our constructed sensor in distinguishing between cancerous and non-cancerous cell lines highlights its potential for early cancer detection and monitoring. This innovative approach represents a major advancement in the field of miRNA detection, providing a user-friendly, cost-effective, and sensitive solution with broad implications for clinical diagnosis and patient care, especially in point-of-care settings.

Abstract:The most important reason for death from ovarian cancer is the late diagnosis of this disease. The standard treatment of ovarian cancer includes surgery and chemotherapy based on platinum, which is associated with side effects for the body. Due to the nonspecific nature of clinical symptoms, developing a platform for early detection of this disease is needed. In recent decades, the advancements of microfluidic devices and systems have provided several advantages for diagnosing ovarian cancer. Designing and manufacturing new platforms using specialized technologies can be a big step toward improving the prevention, diagnosis, and treatment of this group of diseases. Organ-on-a-chip microfluidic devices are increasingly used as a promising platform in cancer research, with a focus on specific biological aspects of the disease. This review focusing on ovarian cancer and microfluidic application technologies in its diagnosis. Additionally, it discusses microfluidic platforms and their potential future perspectives in advancing ovarian cancer diagnosis.

OCaMIR—A Noninvasive, Diagnostic Signature for Early-Stage Ovarian Cancer: A Multi-cohort Retrospective and Prospective Study

Abstract:Abstract Purpose: Due to the lack of effective screening approaches and early detection biomarkers, ovarian cancer has the highest mortality rates among gynecologic cancers. Herein, we undertook a systematic biomarker discovery and validation approach to identify microRNA (miRNA) biomarkers for the early detection of ovarian cancer. Experimental Design: During the discovery phase, we performed small RNA sequencing in stage I high-grade serous ovarian cancer (n = 31), which was subsequently validated in multiple, independent data sets (TCGA, n = 543; GSE65819, n = 87). Subsequently, we performed multivariate logistic regression-based training in a serum data set (GSE106817, n = 640), followed by its independent validation in three retrospective data sets (GSE31568, n = 85; GSE113486, n = 140; Czech Republic cohort, n = 192) and one prospective serum cohort (n = 95). In addition, we evaluated the specificity of OCaMIR, by comparing its performance in several other cancers (GSE31568 cohort, n = 369). Results: The OCaMIR demonstrated a robust diagnostic accuracy in the stage I high-grade serous ovarian cancer patients in the discovery cohort (AUC = 0.99), which was consistently reproducible in both stage I (AUC = 0.96) and all stage patients (AUC = 0.89) in the TCGA cohort. Logistic regression-based training and validation of OCaMIR achieved AUC values of 0.89 (GSE106817), 0.85 (GSE31568), 0.86 (GSE113486), and 0.82 (Czech Republic cohort) in the retrospective serum validation cohorts, as well as prospective validation cohort (AUC = 0.92). More importantly, OCaMIR demonstrated a significantly superior diagnostic performance compared with CA125 levels, even in stage I patients, and was more cost-effective, highlighting its potential role for screening and early detection of ovarian cancer. Conclusions: Small RNA sequencing identified a robust noninvasive miRNA signature for early-stage serous ovarian cancer detection.

Abstract:MicroRNAs (miRNAs) have emerged as a promising class of biomarkers for early detection of various cancers, including ovarian cancer. However, quantifying miRNAs in human blood samples is challenging owning to issues of sensitivity and specificity. In this study, hsa-miR-200a-3p of the miR-200a sub-family, which is a biomarker of ovarian cancer, was used as the analyte to demonstrate the analytical capability of an integrated biosensing platform using an extremely sensitive surface enhanced Raman scattering (SERS) nanotag – nanoaggregate-embedded beads (NAEBs), magnetic nanoparticles (MNPs), a pair of highly specific locked nucleic acid (LNA) probes, and a semi-automated paper-based electrowetting-on-dielectric (pEWOD) device to provide the labor-less and thorough sample cleanup and recovery. A sandwich approach where NAEBs modified by one LNA-1 probe and MNPs modified by another LNA-2 probe was applied. Then the target analyte miRNA-200a-3p was introduced to form a sandwich nanocomplex through hybridization with the pair of LNA probes. The pEWOD device was used to achieve short cleanup time and good recovery of the nanocomplex, bringing the total analysis time to be less than 30 min. The detection limit of this approach can reach 0.26 fM through SERS detection. The versatility of this method needless of RNA extraction for clinical samples is expected to have a good potential in detecting other miRNAs.
