Abstract:The purpose of this paper is to utilize near-infrared (NIR) spectroscopy to characterize powder blending in-line. A multivariate model-based approach was used to determine end-point and variability at the end-point of blending processes. Two monitoring positions for NIR spectrometers were evaluated; one was located on the top of the Bin-blender and the other was on the rotation axis. A ternary powder mixture including acetaminophen (APAP, fine and coarse powder), lactose (LAC) and microcrystalline cellulose (MCC, Avicel 101 and 200) was used as a test system. A Plackett-Burman design of experiments (DOE) for different blending parameters and compositions was utilized to compare the robustness of end-point determination between the multivariate model-based algorithm and reference algorithms. The end-point determination algorithm, including root mean square from nominal value (RMSNV) and two-tailed Student's t-test, was developed based on PLS predicted concentrations of all three constituents. Mean and standard deviation of RMSNV after end-point were used to characterize blending variability at the end-point. The blending end-point and variability of two sensors were also compared. The multivariate model-based algorithm proved to be more robust on end-point determination compared to the reference algorithms. Blending behavior at the two sensor locations demonstrated a significant difference in terms of end-point and blending variability, indicating the advantage to employ process monitoring via NIR spectroscopy on more than one location on the Bin-blender.

Abstract:Objective: To establish a simple and rapid method based on diffuse reflectance NIR spectra to in-line monitor the blending process of Zhongsheng pill powder. Method: Zhongsheng pill powder consisted of Zhongsheng spray powder, artificial cowbezoar powder and bile arisaema powder. By means of a revolutionary miniature near-infrared analyzer mounted onto the blender, the powder homogeneity was determined in real time with noninterference. The moving block standard deviation (MBSD) method was used to identify the end point of blending process, providing real-time data and instant feedback about the blending course. Evaluation of blend uniformity was carried out by stratified in-process sampling, and the content of baicalin in samples was determined by high performance liquid chromatography (HPLC). Result: The results calculated through MBSD method were generally coincided with HPLC analysis. Blend homogeneity was assumed to be reached when the standard deviations between spectra approached 0.002, and RSD values of baicalin content in samples from six positions were lower than 5%. Moreover, stratified in-process sampling has significant influence on blending process when the powder loading volume was small. Conclusion; Based on this study, NIR spectroscopy can be possibly extended to be applied in blending process of Zhongsheng pill powder, ensuring a remarkable saving of time and manpower employed and creating a reliable and efficient process measurement technology.
Blend uniformity monitoring in a continuous manufacturing mixing process for a low-dosage formulation using a stream sampler and near infrared spectroscopy

Abstract:Continuous manufacturing has the potential to offer several benefits for the production of oral solid dosage forms, including reduced costs, low-scale equipment, and the application of process analytical technology (PAT) for real-time process control. This study focuses on the implementation of a stream sampler to develop a near infrared (NIR) calibration model for blend uniformity monitoring in a continuous manufacturing mixing process. Feeding and mixing characterizations were performed for three loss-in-weight feeders and a commercial continuous mixer to prepare powder blends of 2.5-7.5 % w/w ibuprofen DC 85 W with a total throughput of 33 kg/h. The NIR spectral acquisition was performed after the mixing stage using a stream sampler for flowing powders. A continuous mixer shaft speed of 250 RPM was selected to operate the mixing process based on a variability analysis developed with in-line spectral data acquired using the stream sampler at 6 RPM. A partial least squares regression (PLS-R) model was performed and evaluated, yielding a root-mean-square error of prediction (RMSEP) of 0.39 % w/w and a bias of 0.05 % w/w. An independent experimental run conducted two days later revealed that the continuous mixing process and the NIR calibration model presented low day-to-day variation. The minimum practical error (MPE) and sill values through variographic analysis showed low variance associated with the sampling process using the stream sampler. Results demonstrated the promising capacity of the stream sampler coupled to an NIR probe to be implemented within continuous manufacturing processes for the real-time determination of API concentration.
Assessment of blend uniformity in a continuous tablet manufacturing process

Abstract:Blend uniformity was monitored throughout a continuous manufacturing (CM) process by near infrared (NIR) spectroscopic measurements of flowing blends and compared to the drug concentration in the tablets. The NIR spectra were obtained through the chute after the blender and within the feed frame, while transmission spectra were obtained for the tablets. The CM process was performed with semi-fine acetaminophen blends at 10.0% (w/w). The blender was operated at 250 RPM, for best performance, and 106 and 495 rpm where a lower mixing efficiency was expected. The variation in blender RPM increased the variation in drug concentration at the chute but not at the feed frame. Statistical results show that the drug concentration of tablets can be predicted, with great accuracy, from blends within the feed frame. This study demonstrated a mixing effect within the feed frame, which contribute to a 60% decrease in the relative standard deviation of the drug concentration, when compared to the chute. Variographic analysis showed that the minimum sampling and analytical error was five times less in the feed frame than the chute. This study demonstrates that the feed frame is an ideal location for monitoring the drug concentration of powder blends for CM processes.
On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.

Abstract:A rapid and simultaneous quantification of the main active compounds of the in-process extract solutions is beneficial to the process monitoring as well as ensuring quality consistency of the end products during the production of traditional Chinese medicine. Here we present a near infrared (NIR) spectroscopy-based method for rapid analysis of extract solutions in the production of compound E Jiao oral liquid. The partial least-squares regression (PLSR) models for four quality indicators (viz. total flavonoids, total saponins, total saccharides and soluble solid contents) were established and validated. The results showed that all of the four models exhibited satisfactory fitting and predictive capacity. The root mean squares error of prediction (RMSEP) was 0.0384 mg mL−1, 0.0154 mg mL−1, 3.80 mg mL−1 and 0.199% for total flavonoids, total saponins, total saccharides and soluble solid contents, respectively. This work here demonstrated that NIR spectroscopy coupled with PLSR calibration can offer a reliable and non-destructive alternative in the routine monitoring of the extraction process in the production of compound E Jiao oral liquid. The presented approach is expected to be equally applicable to the mixed decoction of other herbal medicines.

Abstract:Blending is a critical intermediate unit operation for all solid oral formulations. For blend uniformity testing, API content in the blend must be quantified precisely. A detailed study was conducted to demonstrate the suitability of inline NIR (near-infrared) spectroscopy for blend uniformity testing of two solid oral formulations: existing direct compression (DC) product with a multistep blending process and granulation-based product with API granules. Both qualitative and quantitative methods were developed at a laboratory scale using statistical moving block standard deviation (MBSD) and multivariate data analysis such as principal component analysis (PCA) and partial least squares (PLS) regression. The qualitative MBSD method demonstrated that there was no need for multiple steps for the existing DC product. Hence, a simplified single-step process was developed for blending. Quantitative PLS models for blending processes of both the products were developed, validated, and successfully implemented at a commercial scale for the real-time release of blends. Results obtained from the validated model were in good agreement with the current method of sampling and chromatography.

Abstract:In-line monitoring of continuous powder flow is an integral part of the continuous manufacturing process of solid oral dosage forms in the pharmaceutical industry. Specifically, monitoring downstream from loss-in-weight (LIW) feeders and/or continuous mixers provides important data about the state of the process. Such measurements support control of the process and thereby enhance product quality. Near Infrared Spectroscopy (NIRS) is a potential PAT tool to monitor the homogeneity of a continuous powder flow stream in pharmaceutical manufacturing. However, the association of analytical results from NIR sampling of the powder stream and the homogeneity (content uniformity) of the resulting tablets provides several challenges; appropriate sampling strategies, adequately robust modeling techniques and poor sensitivities (for low dose APIs) are amongst them. Information from reflectance-based NIRS sampling is limited. The region of the powder bed that is interrogated is confined to the surface where the measurement is made. This potential bias in sampling may, in turn, limit the ability to predict the homogeneity of the finished dosage form. Further, changes to the processing parameters (e.g., rate of powder flow) often have a significant effect on the resulting data. Sample representation, interdependence between process parameters and their effects on powder flow behavior are critical factors for NIRS monitoring of continuous powder flow system. A transmission NIR method was developed as an alternative technique to monitor continuous powder flow and quantify API in the powder stream. Transmission NIRS was used to determine the thickness of the powder stream flowing from a loss-in-weight feeder. The thickness measurement of the powder stream provided an in-depth understanding about the effects of process parameters such as tube angles and powder flow rates on powder flow behaviors. This knowledge based approach helped to define an analytical design space that was specific to flow properties and to determine the optimum process parameters for successful quantitation of powder stream. A PAT method based on transmission NIRS was developed to monitor the homogeneity of API in a continuously flowing powder stream.

Abstract:Fluid bed is an important unit operation in pharmaceutical industry for granulation and drying. To improve our understanding of fluid bed granulation, in-line near infrared spectroscopy (NIRS) and novel environmental temperature and RH data logger called a PyroButton® were used in conjunction with partial least square (PLS) and principal component analysis (PCA) to develop multivariate statistical process control charts (MSPC). These control charts were constructed using real-time moisture, temperature and humidity data obtained from batch experiments. To demonstrate their application, statistical control charts such as Scores, Distance to model (DModX), and Hotelling's T2 were used to monitor the batch evolution process during the granulation and subsequent drying phase; moisture levels were predicted using a validated PLS model. Two data loggers were placed one near the bottom of the granulator bowl plenum where air enters the granulator and another inside the granulator in contact with the product in the fluid bed helped to monitor the humidity and temperature levels during the granulation and drying phase. The control charts were used for real time fault analysis, and were tested on normal batches and on three batches which deviated from normal processing conditions. This study demonstrated the use of NIRS and the use of humidity and temperature data loggers in conjunction with multivariate batch modeling as an effective tool in process understanding and fault determining method to effective process control in fluid bed granulation.
Interaction between thiopentone and subhypnotic doses of ketamine.

Abstract:The hypnotic effects of thiopentone and ketamine were studied in ASA Grade I female patients aged 20-30 without premedication. The dose-response curves of ketamine 0.4 mg kg-1 and thiopentone following ketamine 0.4 mg kg-1 were determined by probit analysis. The interaction between ketamine and thiopentone was found to be an antagonistic one. This effect can be interpreted as a consequence of the different neurophysiological spectra of action of the two drugs.

Abstract:Monitoring granule property is essential for fluidization maintenance and product quality control in fluidized bed granulation (FBG). In this study, two non-invasive techniques, near-infrared (NIR) spectroscopy and acoustic emission (AE), were applied for quantitative analysis of moisture content (MC) and median particle size (D50) in a FBG process, combined with chemometrics and data fusion strategies. Partial least squares (PLS) and support vector machine (SVM) regression models were established based on NIR and AE spectral data. The optimal quantitative models were identified considering the effect of spectra preprocessing and variable selection. In the comparison study, the best separate models for MC and D50 quantification were based on NIR and AE, respectively. The NIR model exhibited the better prediction ability with the determination coefficient of validation set (R2v) of 0.9815, root mean square error of validation set (RMSEv) of 0.2226 %, and residual predictive deviation (RPD) of 7.4674 for MC. Meanwhile, the AE model presented the better prediction performance with R2v of 0.9710, RMSEv of 18.2643 mu m, and RPD of 5.9740 for D50. Furthermore, among three data fusion strategies, the high-level fusion model achieved the best overall performance on D50 quantification with R2v of 0.9863, RMSEv of 12.5707 mu m, and RPD of 8.6798. The results indicated that both NIR and AE are effective monitoring tools for MC and D50 analysis in fluidized bed granulation process. In addition, a more accurate and reliable analysis of particle size can be achieved by combining NIR and AE technology with high-level data fusion.
Development of a Near-Infrared Spectroscopy (nirs)–based Characterization Approach for Inherent Powder Blend Heterogeneity in Direct Compression Formulations

Abstract:With the advent of continuous direct compression (CDC) process, it becomes increasingly desirable to characterize inherent powder blend heterogeneity at a small batch scale for a robust and CDC-amenable formulation. To accomplish this goal, a near infrared spectroscopy (NIRS)-based characterization approach was developed and implemented on multiple direct compression (DC) blends in this study, with the intended purpose of complementing existing formulation development tools and enabling to build an early CMC data package for late-phased process analytical technology (PAT) method development. Three fumaric acid DC blends, designed to harbor varied degrees of inherent blend heterogeneity, were employed. Near infrared spectral data were collected on a kg-scale batch blender via both time- and angle-based triggering modes. The time-triggered data were used to investigate the blending heterogeneity with respect to rotation angles, while the angle-triggered data were used to provide blending variability characterization and compare against off-line HPLC-based results. The time-triggered data revealed that the greatest blend variability was observed between revolutions, while the blending variability within a single revolution stayed relatively low with respect to rotation angles. This confirmed earlier literature findings that the bottom layer of powder blends tends to move with the blender within each revolution, and the most intense powder mixing takes place across revolutions. This also indicates the use of blending speed and the number of co-adds are not able to increase sampling volume to improve signal-to-noise ratio under a tumble-bin blender as what were typically done in a feedframe application. The angle-triggered data showed that there is a consistent trend between NIRS and HPLC-based methods on characterizing blend heterogeneity across the blends at a given sample size. This study contributes to establishing NIRS as a potential characterization approach for inherent powder blend heterogeneity for early R D. It also highlights the promise of continuous characterization of inherent powder blend heterogeneity from gram scale to mini-batch CDC scale.
Development of a Near Infrared Spectroscopy method for the in-line quantitative bilastine drug determination during pharmaceutical powders blending

Abstract:The Food and Drug Administration (FDA)'s guidelines and the Process Analytical Technology (PAT) approach conceptualize the idea of real time monitoring of a process, with the primary objective of improvement of quality and also of time and resources saving. New instruments are needed to perform an efficient PAT process control and Near Infrared Spectroscopy (NIRS), thanks to its rapid and drastic development of last years, could be a very good choice, in virtue of its high versatility, speed of analysis, non-destructiveness and absence of sample chemical treatment. This work was aimed to develop a NIR analytical method for bilastine assay in powder mixtures for direct compression. In particular, the use of NIR instrumentation should allow to control the bilastine concentration and the whole blending process, assuring the achievement of a homogeneous blend. The commercial tablet formulation of bilastine was particularly suitable for this purpose, due to its simple composition (four excipients) and direct compression manufacturing process. Calibration and validation set were prepared according to a Placket-Burman experimental design and acquired with a miniaturized NIR in-line instrument (MicroNIR by Viavi Solution Inc.). Chemometric was applied to optimize information extraction from spectra, by subjecting them to a Standard Normal Variate (SNV) and a Savitzky-Golay second derivative pre-treatment. This spectra pre-treatment, combined with the most suitable wavelength interval (resulted between 1087 and 1217 nm), enabled to obtain a Partial Least Square (PLS) model with a good predictive ability. The selected model, tried on laboratory and production batches, provided in both cases good assay predictions. Results were confirmed by traditional HPLC (High Performance Liquid Chromatography) API (Active Pharmaceutical Ingredient) content uniformity test on the final product.

Abstract:As a rapid and accurate technique, near infrared (NIR) spectroscopy has been widely used in process monitoring to improve efficiency and ensure quality consistency. In the present work, the application of NIR spectroscopy for on-line quantitative monitoring of the concentration process of Wangbi tablets was investigated. Partial least squares regression (PLSR) models based on the full-range spectra or the key wavelengths selected by the competitive adaptive reweighted sampling (CARS) method were established. The accuracy of the CARS-PLSR method was greater than that of full-spectrum PLSR for three quality parameters (soluble solid contents, paeoniflorin and icariin). For the prediction set samples using the CARS-PLSR models the coefficients of determination (r(2)) and root mean square error [RMSEP (%)] were 0.97 and 1.29 for soluble solid contents, 0.93 and 0.014 for paeoniflorin and 0.85 and 0.009 for icariin, respectively. Overall the results indicated that NIR spectroscopy coupled with CARS-PLSR calibration is a reliable and non-destructive alternative method for on-line monitoring of the concentration process of Wangbi tablets on an industrial scale.
Characterization of Herbal Powder Blends Homogeneity Using Near-Infrared Spectroscopy

Abstract:Homogeneity of powder blend is essential to obtain uniform contents for the tablets and capsules. Near-infrared (NIR) spectroscopy with fiber-optic probe was used as an on-line technique for monitoring the homogeneity of pharmaceutical blend during the blending process instead of the traditional techniques, such as high performance liquid chromatograph (HPLC) method. In this paper NIRS with a SabIR diffuse reflectance fiber-optic probe was used to monitor the blending process of coptis powder and lactose (excipient) with different contents, and further qualitative methods, like similarity, moving block of standard deviation and mean square were used for calculation purposes with the collected spectra after the pretreatment of multiplicative signal correction (MSC) and second derivative. Correlation spectrum was used for the wavelength selection. Four different coptis were blended with lactose separately to validate the proposed method, and the blending process of "liu wei di huang" pill was also simulated in bottles to verify this method on multiple herbal blends. The overall results suggest that NIRS is a simple, effective and noninvasive technique can be successfully applied to the determination of homogeneity in the herbal blend.

Abstract:The application of near-infrared (NIR) spectroscopy for on-line monitoring of the extraction process of red paeony root was investigated. For NIR measurements, two fiber optic probes designed to transmit NIR radiation through a 2mm pathlength flow cell were employed to collect spectra in real-time. Partial least squares regression (PLSR) calibration models were developed for the parameters of interest: total solid, paeoniflorin, and benzoic acid. The influences of flow rate and air bubble on the NIR spectra and calibration models were also investigated. The established models were used for on-line and real-time monitoring of extraction process, and a model updating method was proposed for the long-term usage of the developed models. Furthermore, both the moving block of standard deviation (MBSD) and relative concentration changing rate (RCCR) methods were used to identify the end point of extraction process. The results of this particular application of implementing NIR spectroscopy to monitor extraction process are very encouraging. Successful models have been built and applied on-line, which proffers real-time data and instant feedback about the extraction course, and in turn, provides improved control.

The study utilized robust experimental design and multiple sensors to investigate multivariate data acquisition, model development, and model scale-up from lab to manufacturing. The impact of the selection of various blend endpoint algorithms on predicted blend endpoint (i.e., mixing time) was explored. Spectral data collected at two process scales using two NIR spectrometers was incorporated in a single (global) calibration model. Unique endpoints were obtained with different algorithms based on standard deviation, average, and distributions of concentration prediction for major components of the formulation. Control over phenytoin sodium's distribution was considered critical due to its narrow therapeutic index nature. It was found that algorithms sensitive to deviation from target concentration offered the simplest interpretation and consistent trends. In contrast, algorithms sensitive to global homogeneity of active and excipients yielded the longest mixing time to achieve blending endpoint. However, they were potentially more sensitive to subtle uniformity variations. Qualitative algorithms using principal component analysis (PCA) of spectral data yielded the prediction of shortest mixing time for blending endpoint. The hybrid approach of combining NIR data from different scales presents several advantages. It enables simplifying the chemometrics model building process and reduces the cost of model building compared to the approach of using data solely from commercial scale. Success of such a hybrid approach depends on the spectroscopic variability captured at different scales and their relative contributions in the final NIR model.

Abstract:Objective:A rapid method was established to judge the end point of blending process of Zhengtian pills by near-infrared spectroscopy,which was in order to improve blending efficiency of this preparation.Method:Near-infrared spectroscopy of blending samples were collected,then blending uniformity was calculated at different time.Principal component analysis (PCA) combined with Mahalanobis distance was used to identify the end point of blending process.HPLC was employed to determine the content of paeoniflorin in order to verify judgment.Result:Blending uniformity of powders increased with blending time extended.After 20 min,blending uniformity reached more than 97％.PCA-Mahalanobis distance results showed that distance remained below 1.0 and kept unchanged basically,suggesting that blending process reached the end point.Quantitative validation results showed that RSD of paeoniflorin content tended to stable after 20 min.Conclusion:Near-infrared spectroscopy technology can be used to determine the end point of blending process of Zhengtian pills.

Abstract:In the processing of polymer blends and composites, in-line near-infrared (NIR) spectroscopy enables monitoring of the composition and its composite uniformity and contributes to rapid process development and quality control. However, in the injection molding process, the study of the composition of polymer materials has been delayed due to high-pressure conditions. Our research group developed NIR probes for transmission and diffuse reflectance measurements that can withstand high-pressure and temperature conditions up to 130 MPa and 200 °C. In this research, transmission and diffuse reflectance spectra were measured inline during the injection molding process of polymer blends of poly(lactic acid) and polybutylene succinate adipate. The intensity of each polymer band in the second-derivative spectra exhibited a monotonic increase or decrease in response to changes in the blend ratio. Using transmission and diffuse reflectance spectra as explanatory variables of the partial least squares regression model simultaneously, the model showed high estimation accuracy for the entire region of the blend ratio. Finally, this model was applied to monitor the polymer changeover operation, and the change in the blend ratio in the molded product was successfully estimated in line.
