Abstract:Single-nanoparticle detection has received tremendous interest due to its significance in fundamental physics and biological applications. Here, we demonstrate an optical nanofibre-enabled microfluidic sensor for the detection and sizing of nanoparticles. Benefitting from the strong evanescent field outside the nanofibre, a nanoparticle close to the nanofibre can scatter a portion of the field energy to the environment, resulting in a decrease in the transmitted intensity of the nanofibre. On the other hand, the narrow and shallow microfluidic channel provides a femtoliter-scale detection region, making nanoparticles flow through the detection region one by one. By real-time monitoring of the transmitted intensity of the nanofibre, the detection of a single polystyrene (PS) nanoparticle as small as 100 nm in diameter and exosomes in solution is realised. Based on a statistical analysis, the mean scattering signal is related to the size of the nanoparticle. Experimentally, a mixture of nanoparticles of different diameters (200, 500, and 1000 nm) in solution is identified. To demonstrate its potential in biological applications, high-throughput counting of yeasts using a pair of microchannels and dual-wavelength detection of fluorescently labelled nanoparticles are realised. We believe that the developed nanoparticle sensor holds great potential for the multiplexed and rapid sensing of diverse viruses. In this study, we demonstrate that Raman imaging can be employed to visualise and identify microplastics and nanoplastics down to 100 nm, by distinguishing the laser spot, the pixel size/image resolution, the nanoplastics size/position (within a laser spot), the Raman signal intensity, and via the sample preparation etc. We also validate this technique for the analysis of paint-polishing dust samples that have been collected from a driveway when a vehicle's clear coating of poly-acrylic was polished by hand. Abstract:The rising incidence of nanoplastics contamination in environmental ecosystems has led to substantial health risks. Traditional analysis methods are suboptimal due to their inability to efficiently analyze nanoplastics at low concentrations and time-consuming operations. Herein, we developed an innovative strategy, employing luminescent metal–phenolic networks (L-MPNs) coupled with surface-enhanced Raman spectroscopy (SERS) to separate and label nanoplastics, thus facilitating rapid, sensitive and quantitative detection of nanoplastics. We used L-MPNs composed of zirconium ions, tannic acid and rhodamine B, to uniformly label diverse sizes (50-500 nm) and types of nanoplastics (i.e., polystyrene, polymethyl methacrylate, polylactic acid). Rhodamine B, serving as a Raman reporter in L-MPNs-based SERS tags can offer sufficient sensitivity for trace measurement of nanoplastics and L-MPNs labeling can also facilitate separation of nanoplastics from liquid medium. By using a portable Raman instrument, our method offers cost-effective, rapid, and field-deployable detection features with excellent sensitivity in nanoplastic analysis with a limit of detection of 0.1 ppm. Moreover, this study provides a highly promising strategy for the robust and sensitive analysis of a wide range of particle analytes through the effective labeling performance of L-MPNs when coupled with SERS techniques.

Abstract:Background: While the concept of microplastic (<5 mm) is well -established, emergence of nanoplastics (<1000 nm) as a new contaminant presents a recent and evolving challenge. The field of nanoplastic research remains in its early stages, and its progress is contingent upon the development of reliable and practical analytical methods, which are currently lacking. This review aims to address the intricacies of nanoplastic analysis by providing a comprehensive overview on the application of advanced imaging techniques, with a particular focus on Raman imaging, for nanoplastic identification and simultaneous visualisation towards quantification. Results: Although Raman imaging via hyper spectrum is a potentially powerful tool to analyse nanoplastics, several challenges should be overcome. The first challenge lies in the weak Raman signal of nanoplastics. To address this, effective sample preparation and signal enhancement techniques can be implemented, such as by analysing the hyper spectrum that contains hundred -to -thousand spectra, rather than a single spectrum. Second challenge is the complexity of Raman hyperspectral matrix with dataset size at megabyte (MB) or even bigger, which can be adopted using different algorithms ranging from image merging to multivariate analysis of chemometrics. Third challenge is the laser size that hinders the visualisation of small nanoplastics due to the laser diffraction (lambda/2NA, 300 nm), which can be solved with involving the use of super -resolution. Signal processing, such as colour off -setting, Gaussian fitting (via deconvolution), and re -focus or image re -construction, are reviewed herein, which show a great promise for breaking through the diffraction limit. Significance: Overall, current studies along with further validation are imperative to refine these approaches and enhance the reliability, not only for nanoplastics research but also for broader investigations in the realm of nanomaterials.

Abstract:Plastic pollution represents a pressing global environmental issue, with microplastics (MPs) and nanoplastics (NPs) being ubiquitously found in both food and the environment. However, the investigation of NPs has been hampered by limited detection technologies, necessitating the development of advanced techniques. This study introduces a sol-based surface-enhanced Raman spectroscopy (SERS) approach for the swift detection of MPs and NPs in aqueous environment. By leveraging the aggregation effect between silver nanoparticles (Ag nanoparticles) and plastic particles, the plastic Raman signals is significantly enhanced, effectively lowering the detection limit. Utilizing Ag nanoparticles, plastic particles as small as 20 nm were detected in liquid samples, with a detection limit of 0.0005%. With the developed method, nanoplastic particles in seafood packaging samples were successfully tested, with concentration found to be at μg/L level. This method offers a rapid, economical, and convenient means of detecting and identifying MPs and NPs. The sensitivity of the method allows for capturing plastic signals within 2 min, making it valuable for aquatic environment contamination detection. SERS technology also holds promise for rapid plastic solution detection, potentially becoming a fast detection method for food safety.

Abstract:Nanoplastics present a significant hazard to both the environment and human health. However, the development of rapid and sensitive analysis techniques for nanoplastics is limited by their small size, lack of specificity, and low concentrations. In this study, a surface-enhanced Raman scattering (SERS) chessboard substrate was introduced as a multi-channel platform for the pre-concentration and detection of nanoplastics, achieved by polydomain aggregating silver nanoparticles (PASN) on a hydrophilic and a punched hydrophobic PVDF combined filter membrane. Through a straightforward suction filtration process, nanoplastics were captured by the PASN gap in a single step for subsequent SERS detection, while excess moisture was promptly eliminated from the filter membrane. The PASN-based SERS chessboard substrate, benefiting from the enhanced electromagnetic (EM) field, effectively discriminated polystyrene (PS) nanoplastics ranging in size from 30 nm to 1000 nm. Furthermore, this substrate demonstrated favorable repeatability (RSD of 8.6 %), high sensitivity with a detection limit of 0.001 mg/mL for 100 nm of PS nanoplastics, and broad linear detection ranges spanning from 0.001 to 0.5 mg/mL (R2 = 0.9916). Additionally, the SERS chessboard substrate enabled quantitative analysis of nanoplastics spiked in tap and lake water samples. Notably, the entire pre-concentration and detection procedure required only 3 μL of sample and could be completed within 1 min. With the accessibility of portable detection instruments and the ability to prepare substrates on demand, the PASN-based SERS chessboard substrate is anticipated to facilitate the establishment of a comprehensive global nanoplastics map.
Cost-Effective and Wireless Portable Device for Rapid and Sensitive Quantification of Micro/Nanoplastics

Abstract:The accumulation of micro/nanoplastics (MNPs) in ecosystems poses tremendous environmental risks for terrestrial and aquat-ic organisms. Designing rapid, field-deployable, and sensitive devices for assessing the potential risks of MNPs pollution is critical. However, current techniques for MNPs detection have limited effectiveness. Here, we design a wireless portable de-vice that allows rapid, sensitive, and on-site detection of MNPs, followed by remote data processing via machine learn-ing algorithms for quantitative fluorescence imaging. We uti-lized a supramolecular labeling strategy, employing lumines-cent metal-phenolic networks composed of zirconium ions, tannic acid, and rhodamine B, to efficiently label various sizes of MNPs (e.g., 50 nm – 10 μm). Results showed that our de-vice can quantify MNPs as low as 330 microplastics and 3.08×106 nanoplastics in less than 20 min. We demonstrated the applicability of the device to real-world samples through determination of MNPs released from plastic cups after hot water and flow induction, and nanoplastics in tap water. More-over, the device is user-friendly and operative by untrained personnel to conduct data processing on APP remotely. The analytical platform integrating quantitative imaging, custom-ized data processing, decision tree model and low-cost analysis ($0.015 per assay) has great potential for high-throughput screening of MNPs in agrifood and environmental systems.

Abstract:Nanoplastic pollution represents an increasing concern for the scientific community. However, the identification of tiny nano-/microplastics (smaller than 5 μm) is still a huge challenge. Herein, we report rapid detecting and 3D imaging of nano-/microplastics at the single-particle level via a strategy based on stimulated Raman scattering microscopy. We demonstrate detection of nanoplastics as small as 100 nm and discriminate between different types of nanoplastics using high wavenumber scanning with low wavenumber verification. Nano-/microplastics, such as polyethylene and polypropylene, are detected in atmospheric and human lung tissue samples. This work provides a 3D, single-particle-level nano-/microplastics detection method, holding great potential to facilitate the characterization of nano-/microplastics in the environment and human tissues. By demonstrating the real-world utility of our methodology, we hope to foster wider adoption and understanding of this critical tool.

Abstract:Microplastic (MP) residues in marine have become an increasingly serious environmental pollution issue, and in recent years the detection of MPs in marine started to attract worldwide research interests. Optical-fiber-based environmental sensors have been extensively employed for their several merits such as high sensitivity, pressure resistance, compactness and ease of constructing communication networks. However, fiber-optic refractive index sensors are not specifically developed for distinguishing MPs from other inorganic particles suspended in water. In this paper, an metal-organic framework (MOF) ZIF-8 functionalized S-tapered fiber (STF) sensor is proposed for specific detection of polystyrene nanoplastics (PSNPs) in aqueous environment. ZIF-8 coordination nanoporous polymers with different film thickness were immobilized over the surface of the fabricated STF structure based on self-growth technique and yielding a large surface area over the sensor surface. High sensitivity detection can be achieved by converting the concentration perturbation of PSNPs into evanescent waves over the ZIF-8 functionalized STF surface through the strong electrostatic adsorption effect and π-π stacking, while the fabricated sensor is insensitive to gravels with silica as the primary component in water. It is found that the proposed detector with 18 film layers achieves a sensitivity up to 114.1353nm/%(w/v) for the PSNPs concentration range of 0.01 %(w/v) to 0.08 %(w/v).
Counting the Fingers of Birds and Dinosaurs

Abstract:Homologies of the three fingers in birds have been debated for more than a century. Paleontologists have traditionally identified avian digits as I-II-III largely on the basis of phalangeal counts, whereas embryologists number them II-III-IV on the basis of development in the egg. The report by Ann C. Burke and Alan Feduccia (1) is a synthesis of the embryological evidence. They observe that in most amniotes, the first digit to form in the pentadactyl manus is digit IV, which develops a “primary axis.” With the use of this developmental constraint, they identified the primary axis in birds as digit IV, and the surviving digits as II-III-IV. This conclusion is incompatible with theropod (dinosaur) ancestry of birds because theropod digits are identified as I-II-III, with vestigial fourth and fifth digits apparent in the fossil record. The origin of birds from dromaeosaurid-like theropods is supported by a large suite of synapomorphies, and thus has achieved wide acceptance (2–4). Burke and Feduccia apparently regard these similarities as convergence, not homology. The interpretation of Burke and Feduccia is based on the “ground-plan” of the hand of living tetrapods (alligators, for example) in which digit IV always appears first during development. In the case of digital reduction, however, the correspondence between primary axis and digit IV appears to break down. For example, in salamanders, the first digit to form is digit II, not digit IV (5). Burke and Feduccia acknowledge that the loss of digit IV in theropod evolution was unusual and does not follow the general rule of hand development. Furthermore, digit IV was absent in two-fingered Tyrannosaurus (I-II) and one-fingered Mononykus (I), which reveals the shortcoming of the rule. If birds are members of theropods (3–4), the presence of digit IV in Archaeopteryx and adult birds is doubtful. It would be difficult to argue that later birds re-evolved digit IV, while losing digit I, regardless of the pattern of development. It is more likely that, over eons, the primary axis shifted its position in concert with the reduction of digits. Shubin (6) proposed that the primary axis in birds may actually represent digit III because of developmental acceleration, which would favor the I-II-III hypothesis. The shift of the axis may be linked to the ossification of the distal carpal elements, which may have caused perturbation of the distal branching pattern by modification of the expression domains of the Hox D genes (5). That the ulnare is supposed to part of the primary axis, but is lost during avian ontogeny (7), is strong evidence for perturbation of the primary axis in birds. Developmental biologists have used other criteria to support the II-III-IV hypothesis, such as the topographic position of the pisiform and the sequence of chondrogenesis. Hinchliffe (7) identifies five precartilaginous elements in developing chicks: the radiale, ulnare, distal carpal 3 (semilunate bone), X, and pisiform. He regards two proximal carpals as the pisiform and the radiale. He states that the true ulnare regresses during development and is replaced by a new “element X” of uncertain affinity. Because the pisiform occurs laterally and adjacent to the fifth metacarpal in primitive archosaurs, the rudimentary metacarpal in the chick’s wing is identified as digit V, and the rest as digits IV, III, and II. Abstract:With the increasing use and release of plastic products, microplastics have rapidly accumulated in ecological environments. When microplastics enter the food chain, they cause serious harm to organisms and humans. Microplastics pollution has become a growing concern worldwide; however, there is still no standardized method for rapidly and accurately detecting microplastics. In this work, we used fluorescence lifetime imaging technology to detect four kinds of Nile red-stained and unstained microplastics, and the unique phasor fingerprints of different microplastics were obtained by phasor analysis. Tracing the corresponding pixels of the “fingerprint” in the fluorescence lifetime image allowed for the quick and intuitive identification of different microplastics and their location distributions in a mixed sample. In our work, compared with staining the four microplastics with a fluorescent dye, using the phasor “fingerprint library” formed by the autofluorescence lifetimes of the microplastics was more easily distinguished than microplastics in the mixed samples. The feasibility of this method was further tested by adding three single substances—SiO2, chitin and decabromodiphenyl ethane (DBDPE), and surface sediments to simulate interferent in the environment, and the results providing potential applications for the identification and analysis of microplastics in complex environments.

Abstract:The pervasive presence of nanoplastics (NPs) in environmental media has raised significant concerns regarding their implications for environmental safety and human health. However, owing to their tiny size and low level in the environment, there is still a lack of effective methods for measuring the amount of NPs. Leveraging the principles of Mie scattering, a novel approach for rapid in situ quantitative detection of small NPs in low concentrations in water has been developed. A limit of detection of 4.2 μg/L for in situ quantitative detection of polystyrene microspheres as small as 25 nm was achieved, and satisfactory recoveries and relative standard deviations were obtained. The results of three self-ground NPs showed that the method can quantitatively detect the concentration of NPs in a mixture of different particle sizes. The satisfactory recoveries (82.4% to 110.3%) of the self-ground NPs verified the good anti-interference ability of the method. The total concentrations of the NPs in the five brands of commercial bottled water were 0.07 to 0.39 μg/L, which were directly detected by the method. The proposed method presents a potential approach for conducting in situ and real-time environmental risk assessments of NPs on human and ecosystem health in actual water environments.

Abstract:The thorough investigation of nanoplastics (NPs) in aqueous environments requires efficient and expeditious quantitative analytical methods that are sensitive to environmentally relevant NP concentrations and convenient to employ. Optical analysis-based quantitative methods have been acknowledged as effective and rapid approaches for quantifying NP concentrations in laboratory-scale studies. Herein, we compared three commonly used optical response indicators, namely fluorescence intensity (FI), ultraviolet absorbance, and turbidity, to assess their performance in quantifying NPs. Furthermore, orthogonal experiments were conducted to evaluate the influence of various water quality parameters on the preferred indicator-based quantification method. The results revealed that FI exhibits the highest correlation coefficient (> 0.99) with NP concentration. Notably, the limit of quantification (LOQ) for various types of NPs is exceptionally low, ranging from 0.0089 to 0.0584 mg/L in ultrapure water, well below environmentally relevant concentrations. Despite variations in water quality parameters such as pH, salinity, suspended solids (SS), and humic acid, a robust relationship between detectable FI and NP concentration was identified. However, an increased matrix, especially SS in water samples, results in an enhanced LOQ for NPs. Nevertheless, the quantitative method remains applicable in real water bodies, especially in drinking water, with NP LOQ as low as 0.0157–0.0711 mg/L. This exceeds the previously reported detectable concentration for 100 nm NPs at 40 μg/mL using surface-enhanced Raman spectroscopy. This study confirms the potential of FI as a reliable indicator for the rapid quantification of NPs in aqueous environments, offering substantial advantages in terms of both convenience and cost-effectiveness.

Abstract:Nanoplastics (NPLs), of which polystyrene (PS) plastics are typical representatives, are of global concern as emerging contaminants, especially the leakage of NPLs into drinking water, which is directly related to human health. To onsite visually assess the extent of NPLs contamination in drinking water in a rapid and non-destructive manner, this work extends the application of the highly sensitive response to pH of mercaptoundecanoic acid (MUA)-functionalized gold nanoparticles (Au NPs). Further, a visual sensing method based on the binding of alkyl chains to PS NPLs with hydrophobic force to suppress pH-induced aggregation of functionalized nanoparticles was constructed. Particularly, the designed analytical strategy is a significant renewal of the first strategy for the visual detection of PS NPLs, featuring non-destructive, fast response (~30 s), high selectivity, and detection limit (26 μg/L). Importantly, it can reliably distinguish the more hazardous PS NPLs of smaller particle size (<100 nm). Moreover, visualization-based detection observes the color change of the reaction solution from dark purple to wine red. The employment of a smartphone installed App to read the color three-channel value R further ensures the credibility of the analysis results. The proposed visual analysis strategy will provide a promising new avenue for efficient detection of surface hydrophobic NPLs and further evaluation of their particle size or shape.

Abstract:The ubiquity of micro-/nanoplastics poses a visible threat to the environment, aquatic organisms, and human beings and has become a global concern. Here, we proposed a liquid interface-based strategy using surface-enhanced Raman spectroscopy to coassemble nanoplastics and gold nanoparticles into a dense and homogeneous plasmonic array, thereby enabling the rapid and sensitive detection of trace nanoplastics. In addition, due to the uniqueness of the oil-water immiscible two-phase interface, we achieved ideal results for the detection of nanoplastics in a complex matrix (e.g., aqueous environment and edible oil) with a detection limit of μg/mL. With the aid of the principal component analysis algorithm, the differentiation and identification of multiple nanoplastic components (e.g., polystyrene, polyethylene, and polyethylene terephthalate) in aqueous environments and common hazards (e.g., Bap and Phe) in edible oil were achieved. Therefore, our self-assembled plasmonic arrays are expected to be used for monitoring environmental pollution and food safety.

Abstract:Fast quantification is the primary challenge in monitoring microplastic fiber (MPF) pollution in water. The process of quantifying the number of MPFs in water typically involves filtration, imaging on a filter membrane, and manual counting. However, this routine workflow has limitations in terms of speed and accuracy. Here, we present an alternative analysis strategy based on our high-resolution lensless shadow microscope (LSM) for rapid imaging of MPFs on a chip and modified deep learning algorithms for automatic counting. Our LSM system was equipped with wide field-of-view submicron-pixel imaging sensors (>1 cm2; ∼500 nm/pixel) and could simultaneously capture the projection image of >3-μm microplastic spheres within 90 s. The algorithms enabled accurate classification and detection of the number and length of >10-μm linear and branched MPFs derived from melamine cleaning sponges in each image (∼0.4 gigapixels) within 60 s. Importantly, neither MPF morphology (dispersed or aggregated) nor environmental matrix had a notable impact on the automatic recognition of the MPFs by the algorithms. This new strategy had a detection limit of 10 particles/mL and significantly reduced the time of MPF imaging and counting from several hours with membrane-based methods to just a few minutes per sample. The strategy could be employed to monitor water pollution caused by microplastics if an efficient sample separation and a comprehensive sample image database were available.

Abstract:Nanoplastics (NPs) have been identified as an emerging concern for the environment and our food chains in recent years. Monitoring the concentration and size of nanoplastics is essential to assess the potential risks that nanoplastic particles may pose. In this study, we presented a multi-technique based analytical platform to identify, characterize and quantify nanoplastics in water samples through a combination of sample pre-concentration, asymmetric flow field-flow fractionation coupled with multi-angle light scattering (AF4-MALS) and pyrolysis-GC/MS (Py-GC/MS). Models for predicting NPs concentration and particle number in unknown samples were established and validated using NPs standards of known size and AF4-MALS response. Py-GC/MS was applied for further identification of polymer type and quantification of mass concentration. Filtration conditions for pre-concentration were optimized to ensure a high recovery rate with minimal effect on original particle size. The addition of 0.05% SDS prior to filtration, using controlled filtration procedures, effectively improved the recovery. Furthermore, this study demonstrates the application of the analytical platform for the characterization and quantification of different nanoparticles (e.g. spiked PMMA and PS NPs) in the size range 60 nm–350 nm with detection limits down to 0.01 ppm in water samples. The established analytical platform can fill an analytical gap by offering a solution for quantifying size-resolved mass concentrations of nanoplastics and providing comprehensive data on size distribution, particle number and mass quantification with high sensitivity for detection.

Abstract:Microplastics contaminating drinking water is a growing issue that has been the focus of a few recent studies, where a major bottleneck is the time-consuming analysis. In this work, a micro-optofluidic platform is proposed for fast quantification of microplastic particles, the identification of their chemical nature and size, especially in the 1-100 µm size range. Micro-reservoirs ahead of micro-filters are designed to accumulate all trapped solid particles in an ultra-compact area, which enables fast imaging and optical spectroscopy to determine the plastic nature and type. Furthermore, passive size sorting is implemented for splitting the particles according to their size range in different reservoirs. Besides, flow cytometry is used as a reference method for retrieving the size distribution of samples, where chemical nature information is lost. The proof of concept of the micro-optofluidic platform is validated using model samples where standard plastic particles of different size and chemical nature are mixed.
