Abstract:N-nitrosodiphenylamine (NDPhA) is a disinfection byproduct (DBP) in drinking water, However, it is not known what governs the formation of NDPhA and which precursor(s) in the raw water is responsible for its formation. We report here diphenylamine (DPhA) as a key precursor of NDPhA, and we describe the effect of water pH and chloramination conditions on the formation of NDPhA. To identify precursors of NDPhA, raw water samples were collected from the same drinking water system in which NDPhA was previously detected. Analysis of the raw water samples showed the presence of 1.3 ng/L of DPhA and no detectable NDPhA. Seven hours after the treatment of the raw water with chloramines, the concentration of DPhA decreased to 0.4 ng/L with corresponding formation of NDPhA (0.4 ng/L). Controlled experiments involving chloramination of DPhA in water showed that chloramines were essential to the formation of NDPhA, and that increasing the pH from 4 to 10 resulted in 64-fold enhancement in NDPhA formation. Removal of DPhA and formation of NDPhA was found by mass imbalance, which led to the identification of two new DBPs, phenazine (MW 180 Da) and a chlorinated phenazine derivative (MW 216 Da), using liquid chromatography tandem mass spectrometry and gas chromatography mass spectrometry. Both new DBPs were detected only in the treated water and not in the raw water. Phenazine and N-chlorophenazine have never been reported as DBPs and neither their occurrence in drinking water nor their health effects are known.
Formation of Haloacetonitriles, Haloacetamides, and Nitrogenous Heterocyclic Byproducts by Chloramination of Phenolic Compounds

Abstract:The potential formation of nitrogenous disinfection byproducts (N-DBPs) was investigated from the chloramination of nitrogenous and non-nitrogenous aromatic compounds. All molecules led to the formation of known N-DBPs (e.g., dichloroacetonitrile, dichloroacetamide) with various production yields. Resorcinol, a major precursor of chloroform, also formed di/trichloroacetonitrile, di/trichloroacetamide, and haloacetic acids, indicating that it is a precursor of both N-DBPs and carbonaceous DBPs (C-DBPs) upon chloramination. More detailed experiments were conducted on resorcinol to understand N-DBPs formation mechanisms and to identify reaction intermediates. Based on the accurate mass from high resolution Quadrupole Time-of-Flight GC-MS (GC-QTOF) and fragmentation patterns from electronic impact and positive chemical ionization modes, several products were tentatively identified as nitrogenous heterocyclic compounds (e.g., 3-chloro-5-hydroxy-1H-pyrrole-2-one with dichloromethyl group, 3-chloro-2,5-pyrroledione). These products were structurally similar to the heterocyclic compounds formed during chlorination, such as the highly mutagenic MX (3-chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone) or halogenated pyrroles. To our knowledge, this is the first time that the formation of halogenated nitrogenous heterocyclic compounds is reported from chloramination process. The formation of these nitrogenous byproducts during chloramination might be of concern considering their potential toxicity.
Identification of N-Nitrosamines and Nitrogenous Heterocyclic Byproducts during Chloramination of Aromatic Secondary Amine Precursors

Abstract:With diminishing pristine water, wastewater-affected waters that contain complex anthropogenic compounds are becoming important sources of drinking water and the compounds will inevitably react with disinfectants to form disinfection byproducts (DBPs). Secondary amines such as diphenylamine (DPA) analogues are considered as potential precursors of N-nitrosamines. In this study, an in situ 14N/15N-labeling and screening workflow was used to systematically investigate the formation of nitrogenous DBPs (N-DBPs) and putative reaction pathways. Twenty-four pairs of N-DBPs were generated and identified from chloramination of DPA through two main pathways, in which chloramines reacted with the amino and phenyl functional groups to form N-nitrosodiphenylamine and monochlorinated 5,10-dihydro-phenazine (Cl-DiH-Phe), respectively. Cl-DiH-Phe could further produce phenazine and the coupling products with another DPA molecule. Selective N-DBP formation was pH and dose-dependent, and the same reactions were observed for additional two aromatic DPA analogues. Effects of alkyl substituents on the formation pathways were investigated using a series of dialkyl and N-alkyl aromatic analogues. Only the amino pathway to form nitrosamines was noticed for dialkyl amines, nevertheless, both the main reactions occurred for N-alkyl aromatic amines. These findings suggested that the reaction with chloramines through a phenyl pathway was likely to be crucial for novel nitrogenous heterocyclic byproducts.
Formation of iodinated trihalomethanes and haloacetic acids from aromatic iodinated disinfection byproducts during chloramination

Abstract:Iodinated disinfection byproducts (DBPs) are widely present in disinfected drinking waters and wastewater effluents, and they have drawn increasing concern owing to their high toxicity. To date, the reported iodinated DBPs mainly include aliphatic and aromatic ones, and iodinated trihalomethanes (THMs) and haloacetic acids (HAAs) are the most commonly detected aliphatic iodinated DBPs in disinfected waters. In this study, the formation of iodinated THMs and HAAs from aromatic iodinated DBPs during chloramination was investigated. The decomposition kinetics of the aromatic iodinated DBPs and the formation of iodinated THMs and HAAs were studied, the formation pathways of iodinated THMs and HAAs from the aromatic iodinated DBPs were explored, the factors affecting the formation were examined, and the toxicity change was evaluated. The results revealed that four aromatic iodinated DBPs (2,4,6-triiodophenol, 3,5-diiodo-4-hydroxybenzaldehyde, 3,5-diiodosalicylic acid, and 2,6-diiodo-4-nitrophenol) all underwent transformation to form triiodomethane (TIM), monoiodoacetic acid (MIAA), and diiodoacetic acid (DIAA) during chloramination. The decomposition of the aromatic iodinated DBPs all followed a pseudo-first-order decay during chloramination, and the rank order of the decomposition rate constants was as follows: 2,4,6-triiodophenol > 3,5-diiodo-4-hydroxybenzaldehyde ≥ 3,5-diiodosalicylic acid > 2,6-diiodo-4-nitrophenol. Several polar iodinated intermediates were detected and identified (e.g., 2,6-diiodo-1,4-benzoquinone and iodobutenedioic acid) during chloramination of 2,4,6-triiodophenol, based on which the formation pathways of TIM, MIAA, and DIAA from 2,4,6-triiodophenol during chloramination were proposed and further validated. The results also revealed that monochloramine dose, pH, temperature, and short free chlorine contact time all affected the formation of TIM, MIAA, and DIAA from 2,4,6-triiodophenol during chloramination. The cytotoxicity order of the eight iodinated DBPs was MIAA > 2,6-diiodo-4-nitrophenol > 2,4,6-triiodophenol > 2,6-diiodo-1,4-benzoquinone > DIAA ≥ 3,5-diiodosalicylic acid >3,5-diiodo-4-hydroxybenzaldehyde > TIM. The toxicity of the chloraminated 2,4,6-triiiodophenol sample first decreased and then increased over time due to the transformation.
Derivates variation of phenylalanine as a model disinfection by-product precursor during long term chlorination and chloramination

Abstract:Dissolved nitrogenous organic matter in water can contain precursors of disinfection by-products (DBPs), especially nitrogenous DBPs (N-DBPs). Amino acids are ubiquitous as dissolved nitrogenous organic matter in source water and can pass through drinking water treatment processes to react with disinfectants in finished water and in the distribution system. Phenylalanine (Phe) was selected as a model amino acid precursor to investigate its derived DBPs and their variations during a chlorination regime that simulated water distribution with residue chlorine. The 7-day DBPs formation potential (DBPsFP) test with chlorine revealed chlorination by-products of phenylalanine including trihalomethanes (THMs), haloacetic acids (HAAs), haloacetonitriles (HANs), and halonitromethanes (HNMs), but not trichloronitromethane (TCNM) which was a significant N-DBP detected during the first 48 h of chlorine contact. The formation of most carbonaceous DBPs (C-DBPs) increased with chlorination time; however N-DBPs and non-chlorinated byproducts of phenylacetonitrile and phenylacetaldehyde reached their highest concentration after 2 h of reaction, and then gradually decreased until below detection after 7 days. The chlorination influencing factors indicated that light enhanced the peak yield of DBPs; the pH value showed different influences associated with corresponding DBPs; and the presence of bromide ions (Br-) generated a variety of bromine-containing DBPs. The DBPsFP test with chloramine reduced C-DBPs generation to about 1/3 of the level observed for chlorine disinfection and caused an increase in dichloroacetonitrile. Surveillance of DBPs during drinking water distribution to consumers should consider the varying contact times with disinfectants to accurately profile the types and concentrations of C-DBPs and N-DBPs present in drinking water.

Abstract:Disinfection byproducts (DBPs) in drinking water exhibit considerable adverse health effects; recent focus is on the brominated disinfection byproducts (Br-DBPs). The chlorination and chloramination of bromide ion containing water produce reactive bromo species, which subsequently react with natural organic matter (NOM) to yield Br-DBPs. The possible reactions involved in generating DBPs are presented. Identified Br-DBPs include bromomethanes, bromoacetic acid, bromoacetamides, bromoacetonitriles, and bromophenols. Mixed chloro- and bromo-species have also been identified. Pathways of the formation of Br-DBPs have been described. The concentration of Br- ion, pH, reaction time, and the presence of Cu(II) influence the yield of DBPs. The effects of water conditions on the production of Br-DBPs are presented. The epidemiological studies to understand the potential toxic effects of DBPs including Br-DBPs are summarized. Brominated DBPs may have higher health risks than their corresponding chlorinated DBPs. A potential role of an emerging alternate disinfectant, ferrate (FeV)O(2-)4), in minimizing DBPs is briefly discussed.

Abstract:Developments in the electricity sector, related to integration of sustainable energy, not only affects the medium voltage but also the low voltage infrastructure. Condition monitoring of Low Voltage (LV) underground power cables becomes of increasing interest to safeguard reliable power delivery. Since the start of phasing out oil-paper insulated cables, polymeric insulation is most widely applied. As phenomena associated with high electric field stresses are not expected, little is known about the degradation mechanism of LV cables with polymeric insulation. External damage is known to be a major cause of defects, which directly causes a failure or initiates degradation resulting in a failure later on. To study the degradation mechanisms, short cable sections with artificial damage have been subjected to periodic water exposure in the laboratory. This shows that dry-band arcing is a major mode of degradation, with accumulation of pollutants due to chemical decomposition. Several factors such as water conductivity, presence of oxygen, soil type and insulation material have been investigated. Strong dependencies upon these parameters are observed. However, it may be hard to quantify the degradation process in practical situations. The presence of a water film that can sustain dry-band arcing will occur only in certain situations and for limited amount of time. However, the accumulation of these events, mainly caused by periods of precipitation, will eventually lead to breakdown at the damaged spot.
Formation of known and unknown disinfection by-products from natural organic matter fractions during chlorination, chloramination, and ozonation

Abstract:Natural organic matter (NOM) is the main precursor of disinfection by-products (DBPs) formed during drinking water treatment processes. Previous studies of the relationships between DBP formation and NOM fractionation have mainly been focused on currently regulated DBPs and a few certain emerging DBPs. In this work, the Suwannee River NOM solution was fractionated into groups with different hydrophobicities using DAX-8 resins, and volatile and semi-volatile DBPs formed during the chlorination, chloramination and ozonation of the NOM fractions were examined by a nontargeted screening of comprehensive two-dimensional gas chromatography-quadrupole mass spectrometry procedure. The results showed that a total of 302 DBPs representing nine chemical classes were detected, of which 266 were possibly newly detected, based on library searching with NIST 08 library (using similarity and reverse values of at least 600 and 700, respectively) and linear retention indices. The characterization of DBP precursors suggests that hydrophobic (HPO) NOM contains the major fraction of precursor for the formation of nitrogenous DBPs (contributing about 60% of the total nitrogenous DBPs) during all three disinfection processes. Much larger amounts of heterocyclic DBPs were formed from the HPO fraction than from the hydrophilic fraction during chlorination. During chloramination and ozonation, 5-15 times more ketones were formed from the hydrophilic fraction than from the HPO fraction. During ozonation, more than twice the amounts of esters and alcohols were formed from the hydrophilic fraction than from the HPO fraction. Three-dimensional excitation-emission matrix spectra suggest that similar to the formation of regulated DBPs, humic acid-like substances are probably the precursors of halogen-containing DBPs. Relatively higher nitrogenous DBPs formation from the HPO fraction might be because of the existence of protein-like materials.

Abstract:Chloramination was commonly used as disinfectant for killing pathogens in water. However, in this process, nitrogen-containing disinfection by-products (N-DBPs) would accidently form and subsequently rise toxicity. Here, we investigated acute toxicity variation and by-products formation during chloramination treatment on UV filter 2-hydroxy-4-methoxy-5-sulfonic acid benzophenone (BP-4). Under alkaline conditions, the acute toxicity of this system had significant increase. A total of 17 transformation products were tentatively identified, and for them, plausible transformation pathways were proposed. Noticeably, numerous aniline and nitrosobenzene analogs were detected, and the dramatic increase of acute toxicity in this system might be primarily attributed to the formation of benzoquinone and aniline analogs. Besides, bromophenol, iodophenol and iodobenzoquinone analogs exhibiting high toxicity were generated in the presence of bromine and iodide ions. This study indicates that chloramination treatment may significantly increase potential health risk, further management on disinfection system is reasonable.

Abstract:Many studies have demonstrated the different trends of disinfection by-products (DBPs) formation between chlorination and chloramination. However, the reactions between precursors and disinfectants are widely assumed to be “black box” and the reasons for abovementioned difference are not well illustrated. This study focused on source water with high levels of natural organic matter (NOM) and bromide, and compared the transformation of NOM specific characteristics and the ratios of specific DBPs as an equivalent of chlorine to total organic halogen (TOX) among three disinfection scenarios of chlorination, chloramination and chlorine–chloramine sequential treatment (Cl2–NH2Cl process). A three-reaction-phrases model was proposed thereafter to illustrate the major reactions involved in, i.e., stage-I: rapid consumption of fast reactive sites (DOC1), which transformed to slow reactive sites (DOC2) and measured DBPs, i.e., trihalomethanes, haloacetic acids, etc; stage-II: oxidation and/or halogenation of DOC2 into unknown TOX (UTOX) intermediates; stage-III: oxidation of UTOX intermediates into measured DBPs. The effect of ammonia was also quantified. Ammonia is observed to inhibit the formation of measured DBPs by 68–92%, 94–99%, and 92–95% of that in chlorination in Stage-I, II, and III, respectively, and the formation of UTOX is reduced by 2–80%, 60–94%, and 82–93% accordingly. These effects lead to the steady accumulation of DBPs intermediates such as UTOX, and to the elevated UTOX/TOX during chloramination and Cl2–NH2Cl process thereafter. The results illustrate the mechanism of ammonia participating in DBPs formation, and are valuable to fill in the gap between the transformation of precursors and the formation of different DBPs.

Abstract:Colony‐stimulating factor‐1 (CSF‐1) is a cytokine required for proliferation, differentiation, activity, and survival of cells of the mononuclear phagocytic system. The growth factor is synthesized as a soluble, matrix, or membrane associated molecule. The specific functions of these forms are not clear. However, some data suggest a dependence of the development of various populations of tissue macrophages on the locally expressed and presented cytokine. Deficiency in CSF‐1, as is the case in the murine mutant strain op/op, results in low numbers of macrophages and monocytes and, most striking, leads to osteopetrosis due to a virtual absence of osteoclasts. Using the op/op mutation as a model, CSF‐1 was established as one of the growth factors for osteoclasts. The expression of CSF‐1 receptors, encoded by the proto‐oncogene c‐fms, by osteoclast precursors and osteoclasts, suggested an effect of this cytokine not only during osteoclast formation but also on the mature cells. In fact, CSF‐1 was shown to inhibit the resorbing activity, to stimulate migration, and to support survival of isolated osteoclasts in vitro. By these actions on cells of the osteoclast lineage, CSF‐1 induces recruitment of new osteoclasts, leading to a net increase of bone resorption, and might govern the spatial distribution of resorption sites within the bone. During these processes, locally expressed and presented forms of the growth factor may play a crucial role, as will be discussed in this article. © 1994 Wiley‐Liss, Inc.
Halogenated semivolatile acetonitriles as chloramination disinfection by-products in water treatment: a new formation pathway from activated aromatic compounds

Abstract:The use of monochloramine as an alternative disinfectant to chlorine in drinking water treatment can lead to increased formation of emerging nitrogenous halogenated disinfection by-products (DBPs), even when the formation of regulated halogenated DBPs has decreased. In this study, we investigated formation of the semivolatile haloacetonitriles (HANs) from model nitrogen-containing compounds (6 amines, 1 amide, 6 amino acids, and 2 nitrogen-containing aromatic chemicals) and natural organic matter (NOM) reference materials after chloramination. In agreement with previous studies, most amino acids formed dichloroacetonitrile (DCAN). Additionally, DCAN formed from two amines containing aromatic rings (N,N-dimethylaniline and 3-(dimethylamino-methyl)indole) and the two nitrogen-containing aromatic chemicals (cotinine and phenytoin). This is the first report of DCAN formation from these precursors. DCAN also formed after chloramination of NOM reference materials, with the highest formation from the NOM material with the highest aromaticity. The results provide new evidence of a DCAN formation pathway from cleavage of activated aromatic structures after electrophilic substitution of chlorine and addition of monochloramine to the ring system. In particular, the results suggest that the previously proposed aldehyde pathway from the amino acid group is not responsible for the majority of DCAN formation from amino acids with an activated aromatic ring system. This newly proposed formation pathway for DCAN from activated aromatic organic matter has significant implications for NOM removal during water treatment to minimise DBP formation. Studies using 15N-labelled monochloramine showed that there was significant incorporation of nitrogen from monochloramine into DCAN, demonstrating that monochloramine disinfection promotes the formation of HANs.

Abstract:Drinking water disinfectants react with natural organic material (NOM) present in source waters used for drinking water to produce a wide variety of by-products. Several hundred disinfections by-products (DBPs) have been identified, but none have been identified with sufficient carcinogenic potency to account for the cancer risks projected from epidemiological studies. In a search for DBPs that might fill this risk gap, the present study projected reactions of chlorine and chloramine that could occur with substructures present in NOM to produce novel by-products. A review of toxicological data on related compounds, supplemented by use of a quantitative structure toxicity relationship (QSTR) program TOPKAT®) identified chemicals with a high probability of being chronically toxic and/or carcinogenic among 489 established and novel DBPs. Classes of DBPs that were specifically examined were haloquinones (HQs), related halo-cyclopentene and cyclohexene (HCP&H) derivatives, halonitriles (HNs), organic N-chloramines (NCls), haloacetamides (HAMs), and nitrosamines (NAs). A review of toxicological data available for quinones suggested that HQs and HCP&H derivatives appeared likely to be of health concern and were predicted to have chronic lowest observed adverse effect levels (LOAELs) in the low μg/kg day range. Several HQs were predicted to be carcinogenic. Some have now been identified in drinking water. The broader class of HNs was explored by considering current toxicological data on haloacetonitriles and extending this to halopropionitriles. 2,2-dichloropropionitrile has been identified in drinking water at low concentrations, as well as the more widely recognized haloacetonitriles. The occurrence of HAMs has been previously documented. The very limited toxicological data on HAMs suggests that this class would have toxicological potencies similar to the dihaloacetic acids. Organic N-halamines are also known to be produced in drinking water treatment and have biological properties of concern, but no member has ever been characterized toxicologically beyond bacterial or in vitro studies of genotoxicity. The documented formation of several nitrosamines from secondary amines from both natural and industrial sources prompted exploration of the formation of additional nitrosamines. N-diphenylnitrosamine was identified in drinking waters. Of more interest, however, was the formation of phenazine (and subsequently N-chorophenazine) in a competing reaction. These are the first heterocyclic amines that have been identified as chlorination by-products. Consideration of the amounts detected of members of these by-product classes and their probable toxicological potency suggest a prioritization for obtaining more detailed toxicological data of HQs>HCP&H derivatives>NCls>HNs. Based upon a ubiquitous occurrence and virtual lack of in vivo toxicological data, NCls are the most difficult group to assign a priority as potential carcinogenic risks. This analysis indicates that research on the general problem of DBPs requires a more systematic approach than has been pursued in the past. Utilization of predictive chemical tools to guide further research can help bring resolution to the DBP issue by identifying likely DBPs with high toxicological potency.

Abstract:The changes in dissolved organic matter (DOM) throughout the treatment processes in a drinking water treatment plant in Sweden and the formation of disinfection byproducts (DBPs) were evaluated by using ultra-high-resolution mass spectrometry (resolution of ∼500,000 at m/z 400) and nuclear magnetic resonance (NMR). Mass spectrometric results revealed that flocculation induced substantial changes in the DOM and caused quantitative removal of DOM constituents that usually are associated with DBP formation. While half of the chromophoric DOM (CDOM) was removed by flocculation, ∼4-5 mg L(-1) total organic carbon remained in the finished water. A conservative approach revealed the formation of ∼800 mass spectrometry ions with unambiguous molecular formula assignments that contained at least one halogen atom. These molecules likely represented new DBPs, which could not be prevented by the flocculation process. The most abundant m/z peaks, associated with formed DBPs, could be assigned to C5HO3Cl3, C5HO3Cl2Br, and C5HO3ClBr2 using isotope simulation patterns. Other halogen-containing formulas suggested the presence of halogenated polyphenolic and aromatic acid-type structures, which was supported by possible structures that matched the lower molecular mass range (maximum of 10 carbon atoms) of these DBPs. 1H NMR before and after disinfection revealed an ∼2% change in the overall 1H NMR signals supporting a significant change in the DOM caused by disinfection. This study underlines the fact that a large and increasing number of people are exposed to a very diverse pool of organohalogens through water, by both drinking and uptake through the skin upon contact. Nontarget analytical approaches are indispensable for revealing the magnitude of this exposure and to test alternative ways to reduce it.

Nonhalogenated Aromatic DBPs in Drinking Water Chlorination: A Gap between NOM and Halogenated Aromatic DBPs

Abstract:Halogenated disinfection byproducts (DBPs) are generated via reactions with natural organic matter (NOM) in chlorine disinfection of drinking water. How large NOM molecules are converted to halogenated aliphatic DBPs during chlorination remains a fascinating yet largely unresolved issue. Recently, many relatively toxic halogenated aromatic DBPs have been identified in chlorinated drinking waters, and they behave as intermediate DBPs to decompose to halogenated aliphatic DBPs. There is still one gap between NOM and halogenated aromatic DBPs. In this study, nine nonhalogenated aromatic compounds were identified as new intermediate DBPs in chlorination, including 4-hydroxybenzaldehyde, 4-hydroxybenzoic acid, 3-formyl-4-hydroxybenzoic acid, salicylic acid, 5-formyl-2-hydroxybenzoic acid, 4-hydroxyphthalic acid, 4'-hydroxyacetophenone, 4-methylbenzoic acid, and 4-hydroxy-3-methylbenzaldehyde. These nonhalogenated aromatic DBPs formed quickly and reached the maximum levels at relatively low chlorine doses within a short contact time, and their formation pathways were proposed. The formation kinetics of three nonhalogenated aromatic DBPs and their corresponding monochloro-/dichloro-substitutes during chlorination were then modeled. The nonhalogenated aromatic DBPs contributed up to 84% of the formed monochloro-substitutes and 22% of the formed dichloro-substitutes, demonstrating that they somewhat acted as intermediates between NOM and halogenated aromatic DBPs. Furthermore, the formed nonhalogenated aromatic DBPs were found to be removed by >50% by granular activated carbon adsorption.

Abstract:Disinfection by-products (DBP) formed from natural organic matter and disinfectants like chlorine and chloramine may cause adverse health effects. Here, we evaluate how the quantity and quality of natural organic matter and other precursors influence the formation of DBPs during chlorination and chloramination using a comprehensive approach including chemical analysis of regulated and emerging DBPs, total organic halogen quantification, organic matter characterisation and bioanalytical tools. In vitro bioassays allow us to assess the hazard potential of DBPs early in the chain of cellular events, when the DBPs react with their molecular target(s) and activate stress response and defence mechanisms. Given the reactive properties of known DBPs, a suite of bioassays targeting reactive modes of toxic action including genotoxicity and sensitive early warning endpoints such as protein damage and oxidative stress were evaluated in addition to cytotoxicity. Coagulated surface water was collected from three different drinking water treatment plants, along with reverse osmosis permeate from a desalination plant, and DBP formation potential was assessed after chlorination and chloramination. While effects were low or below the limit of detection before disinfection, the observed effects and DBP levels increased after disinfection and were generally higher after chlorination than after chloramination, indicating that chlorination forms higher concentrations of DBPs or more potent DBPs in the studied waters. Bacterial cytotoxicity, assessed using the bioluminescence inhibition assay, and induction of the oxidative stress response were the most sensitive endpoints, followed by genotoxicity. Source waters with higher dissolved organic carbon levels induced increased DBP formation and caused greater effects in the endpoints related to DNA damage repair, glutathione conjugation/protein damage and the Nrf2 oxidative stress response pathway after disinfection. Fractionation studies indicated that all molecular weight fractions of organic carbon contributed to the DBP formation potential, with the humic rich fractions forming the greatest amount of DBPs, while the low molecular weight fractions formed more brominated DBPs due to the high bromide to organic carbon ratio. The presence of higher bromide concentrations also led to a higher fraction of brominated DBPs as well as proportionally higher effects. This study demonstrates how a suite of analytical and bioanalytical tools can be used to effectively characterise the precursors and formation potential of DBPs.

Abstract:When drinking water treatment plants disinfect water, a wide range of disinfection by-products (DBPs) of health and regulatory concern are formed. Recent studies have identified emerging DBPs (e.g. iodinated trihalomethanes (THMs) and acids, haloacetonitriles, halonitromethanes (HNMs), haloacetaldehydes, nitrosamines) that may be more toxic than some of the regulated ones (e.g. chlorine- and bromine-containing THMs and haloacetic acids). Some of these emerging DBPs are associated with impaired drinking water supplies (e.g. impacted by treated wastewater, algae, iodide). In some cases, alternative primary or secondary disinfectants to chlorine (e.g. chloramines, chlorine dioxide, ozone, ultraviolet) that minimize the formation of some of the regulated DBPs may increase the formation of some of the emerging by-products. However, optimization of the various treatment processes and disinfection scenarios can allow plants to control to varying degrees the formation of regulated and emerging DBPs. For example, pre-disinfection with chlorine, chlorine dioxide or ozone can destroy precursors for N -nitrosodimethylamine, which is a chloramine by-product, whereas pre-oxidation with chlorine or ozone can oxidize iodide to iodate and minimize iodinated DBP formation during post-chloramination. Although pre-ozonation may increase the formation of trihaloacetaldehydes or selected HNMs during post-chlorination or chloramination, biofiltration may reduce the formation potential of these by-products.

Abstract:While there is extensive literature covering byproducts formed during various water treatment processes, there is limited research focusing on the generation of brominated disinfection byproducts (Br-DBPs) specifically during ozonation in the presence of bromide (Br − ) and ammonium (NH 4 + ). This study used Orbitrap mass spectrometry and a custom halogen extraction code to selectively pinpoint 109 Br-DBPs generated during the ozonation of Suwannee River natural organic matter (SRNOM) containing Br − and NH 4 + . The Br-DBPs are predominantly distributed ranging from m / z 190 to 320, with elemental compositions consisting of CHOBr and CHONBr. They mainly fell within the H/C (0.5–1.5) and O/C (0.2–1.0) zones, with 16 % being aromatic compounds. NH 4 + suppressed Br-DBP formation in CHOBr 1-2 subgroups but promoted it in CHONBr 1-2 subgroups compared to Br − -containing SRNOM ozonation. Kendrick mass defect diagram revealed 64 identical Br-DBP formulas in the CHOBr 1-2 subgroups in both systems, and among them, 13 brominated carboxylic acids were identified, whose formation was suppressed by the addition of NH 4 + . Moreover, bromonitrophenol and bromobenzonitrile were identified in CHONBr 1-2 subgroups, and their formation pathways were proposed. Nitrogen-containing Br-DBPs, especially the aromatic nitrogen-containing ones, exhibited higher toxicity. Caution is necessary to minimize toxic Br-DBPs formation during Br − and NH 4 + -involved ozonation. This study offers insights into Br-DBP characterization, formation mechanisms and toxicity, guiding enhanced water treatment strategies.

Abstract:Recently, 13 new phenolic halogenated disinfection by-products (DBPs) have been reported in chlorinated drinking water and have been classified into four groups: dihalo-4-hydroxybenzaldehydes, dihalo-4-hydroxybenzoic acid, dihalo-salicylic acids, and trihalo-phenols. In this work, the four fully brominated species (3,5-dibromo-4-hydroxybenzoic acid, 3,5-dibromosalicylic acid, 2,4,6-tribromophenol, and 3,5-dibromo-4-hydroxybenzaldehyde) were selected as representatives, and the decomposition mechanism of these new DBPs during chlorination was studied with the aid of ultra performance liquid chromatography/electrospray ionization triple-quadrupole mass spectrometry (precursor ion scan, multiple reaction monitoring, and product ion scan). Except for 3,5-dibromosalicylic acid, the new DBPs were not stable and could be finally decomposed to haloacetic acids through multistep substitution, hydrolysis, and oxidation. Various decomposition intermediate DBPs were detected, including a new group of halogenated DBPs with cyclic structures (trihalo-hydroxyl-cyclopetene-diones).

Abstract:Algal blooms and wastewater effluents can introduce algal organic matter (AOM) and effluent organic matter (EfOM) into surface waters, respectively. In this study, the impact of bromide and iodide on the formation of halogenated disinfection byproducts (DBPs) during chlorination and chloramination from various types of dissolved organic matter (DOM, e.g., natural organic matter (NOM), AOM, and EfOM) were investigated based on the data collected from literature. In general, higher formation of trihalomethanes (THMs) and haloacetic acids (HAAs) was observed in NOM than AOM and EfOM, indicating high reactivities of phenolic moieties with both chlorine and monochloramine. The formation of haloacetaldehydes (HALs), haloacetonitriles (HANs) and haloacetamides (HAMs) was much lower than THMs and HAAs. Increasing initial bromide concentrations increased the formation of THMs, HAAs, HANs, and HAMs, but not HALs. Bromine substitution factor (BSF) values of DBPs formed in chlorination decreased as specific ultraviolet absorbance (SUVA) increased. AOM favored the formation of iodinated THMs (I-THMs) during chloramination using preformed chloramines and chlorination-chloramination processes. Increasing prechlorination time can reduce the I-THM concentrations because of the conversion of iodide to iodate, but this increased the formation of chlorinated and brominated DBPs. In an analogous way, iodine substitution factor (ISF) values of I-THMs formed in chloramination decreased as SUVA values of DOM increased. Compared to chlorination, the formation of noniodinated DBPs is low in chloramination.
