Abstract:The effects of prednisolon, vincristine and Mitoxantrone on glucocorticoid receptor (GCR) content and affinity were examined in HL-60 cell line with a whole-cell assay in vitro. The fall in GCR number was observed following a short time exposure to prednisolon, vincristine or Mitoxantrone alone. The significant decrease occurred in a dose-dependent (10(-8)-10(-6) mol/L) and time-dependent manner. The treatment of HL-60 cells with 10(-6) mol/L led to a decrease in the affinity of whole cell GCR for [3H]Dex, but vincristine had no that effect. Synergia in GCR content was found during the treatment of HL-60 cells with prednisolon plus Mitoxantrone or prednisolon plus vincristine. The significance of these effects is discussed.

Abstract:Rapid activation of JNK and p38 and their translocation to the cell nucleus by glucocorticoids, corticosterone (Cort), and bovine serum-conjugated corticosterone (Cort-BSA) were studied in primary cultured hippocampal cells by using immunobloting and immunofluorescence confocal microscopy. The rapid activation occurred 5 min after stimulation and was maintained at plateau for as long as 2-4 hr; i.e., the response persisted for 2 hr after washing out the 15-min application of Cort-BSA. The activation occurred at a minimal concentration of 10(-9) M for Cort and 10(-8) M for Cort-BSA. GDP beta S blocked the activation, but RU38486, a nuclear glucocorticoid receptor antagonist, could not block the activation, indicating the involvement of the membrane-delineated receptor in this reaction. The protein kinase C (PKC) inhibitor Go6976 blocked the response, whereas the protein kinase A inhibitor H89 could not, implying the involvement of PKC in the intracellular signal transduction pathway. The nongenomic nature of the responses and the transduction pathway and the significance of persistent action and biological significance are discussed. (c) 2005 Wiley-Liss, Inc.

Abstract:OBJECTIVES:To investigate whether CRHR1 and CRHR2 are colocalized in CRH-specific neurons in rat brain.METHODS:Double/triple immunofluorescence, and combined in situ hybridization were performed in the PVN, amygdala and hippocampus, and triple immunofluorescence was applied to the median eminence (ME), dorsal raphe (DR) and locus coeruleus (LC).RESULTS:Both CRHR1 and CRHR2 immunoreactivity were highly coexpressed in the PVN, central nucleus of the amygdala (CeA) and hippocampus. Triple immunofluorescence under confocal microscopy confirmed that CRHR1 and CRHR2 are coexpressed in CRH-producing neurons in these regions. The results of in situ hybridization combined with double immunofluorescence further strengthened the finding that CRHR1 and CRHR2 were coexpressed in CRH-specific neurons in the PVN, CeA and hippocampus. In addition, CRH immunoreactivity signals were evidently distributed in the ME, DR and LC, and were coexpressed with both receptors.CONCLUSION:CRH receptors colocalize in CRH-containing neurons in the PVN, CeA and hippocampus, and CRH, CRHR1, and CRHR2 coexist in the DR and LC. Our results implicate CRHR1 and CRHR2 in coordinating the regulation of CRH neuronal activity in stress and behavioral responses.

Abstract:Traditionally, steroid hormone action has been described as the modulation of nuclear transcription, thus triggering genomic events that are responsible for physiological effects. Despite early observation of rapid steroid effects that were incompatible with this theory, nongenomic steroid action has been widely recognized only recently. More and more evidence for these rapid effects has been reported in recent years, but the signal transduction pathways of steroid hormones are not clear yet. Based on the study of the effect of glucocorticoids on the secretion of catecholamine induced by high K+, nicotine and bradykinin and the signal transduction pathway as well in PC12 cells, a new model of corticosterone’s rapid nongenomic action in PC12 cells is proposed. Glucocorticoid might act through the PTX-sensitive-G protein-PKC pathway in PC12 cells.

Abstract:Objective To evaluate expression of HDAC2 in peripheral blood mononuclear cells(PBMCs)from glucocorticoid-resistant versus glucocorticoid-sensitive patients with sudden sensorineural hearing loss and identi-ty the relationship between the level of HDAC2 and glucocorticoid insensitivity.Methods PBMCs were collected from10 patients with deviation of nasal septum (control group)and 20 sudden sensorineural hearing loss patients be-fore and after intratympanic methylprednisolone for 10 days.We divided the SSNHL patients into 2 groups (GC sensitive group and GC insensitive group)according to their hearing recovery.Real time PCR and HDAC2 Assay Kit were used to detect the expression level of HDAC2 mRNA and HDAC2 activity in PBMCs.The data were analyzed with SPSS 17.0 software.ResuIts Before intratympanic methylprednisolone,the level of HDAC2 activity were sig-nificantly depressed in SSNHL patients,while the HDAC2 mRNA expressing much higher than the control group. The expression level of HDAC2 mRNA increased significantly after intratympanic methylprednisolone.The HDAC2 activity in GC sensitive group increased significantly.ConcIusion Knockdown of HDAC2 expression induces corti-costeroid in-sensitivity.Glucocorticoids can increase the expression of HDAC2 mRNA.HDAC2 activity can be down-regulated by post-translational modifications.

Abstract:Glucocorticoid (GC) plays a key role in controlling numerous cellular processes during embryogenesis and fetal development. The actions of glucocorticoids are mediated by interaction with their receptors. We previously reported that hippocampal neurons from embryonic day 18 (E18) rats showed silence of glucocorticoid receptor (GR) expression when cultured in serum-free condition. In this study, using western blot, immunofluorescence staining and real-time RT-PCR, we found that while this silence occurred in hippocampal neurons isolated from E16 and E18 rats, it did not happen in those from E20 and neonatal (P0) rats. And when cultured under serum-containing condition, none of them showed GR silence anymore. Corticosterone could not rescue the expression of GR in E16 and E18 neurons in serum-free condition, whereas adding of serum could induce the re-expression of the silenced GR. The absence of GR silence in P0 neurons was not due to the perturbation during parturition. Moreover, the unique expression profile of GR in protein and mRNA level was well reflected in the changes of GR function. These results suggested that under in vitro condition, serum was critical for the maintaining of GR expression in hippocampal neurons of early embryonic stages but less important in later developmental stages. Thus, our data implied that at different developmental stages, the expression of GR in hippocampal neurons might have different susceptibilities to environment changes and there might be a critical time window for the switching of such characteristics during development.
A Novel Method for Monitoring Glucocorticoid-Induced Changes of the Glucocorticoid Receptor in Kidney Transplant Recipients

Abstract:INTRODUCTION:Initial high dose glucocorticosteroid (GC) treatment of transplant recipients induces changes in the glucocorticoid receptor (GR) that may affect the mode of action of the drug. We developed a TaqMan one-step RT-PCR quantification method to investigate the effect of bolus GC treatment on the structure of the GR complex by measuring the levels of GR isoform expression and FK binding protein 5 (FKBP5) transcription.METHODS:Peripheral blood mononuclear cells (PBMCs) were obtained before renal transplantation and immediately after the initial methylprednisolone treatment. Gene expression of the GR-alpha, -beta, -P isoforms and FKBP5 were quantified using a simplex TaqMan relative quantification assay. Genotyping was performed using the TaqMan allele discrimination assay.RESULTS:The gene expression assay was validated and is efficient to detect extremely low quantity of GR-beta expression. Increased expression of FKBP5 (14.01+11.52 folds), GR-alpha (1.47+1.00 folds), GR-beta (6.21+5.71 folds) and GR-P (2.72+1.90 folds) were observed after steroid bolus. The GR haplotype A-T-G is significantly related to elevation of GRbeta transcription (p=0.002). Patients' hospitalization time after transplantation correlated with increment of GR-alpha (p=0.01) and FKBP5 (p=0.007) gene expression. The GR-beta expression level is extremely low compared with the GR-alpha isoform. By contrast, the GR-P isoform is relatively abundantly expressed in PBMCs.CONCLUSION:The method to measure GR isoform enabled us to determine that acute high-dose GC treatment alters the structure of the GR. The initial therapy with high dose steroid may induce a GC resistance phenotype. The modulatory role of the GR-P isoform is not fully defined but may provide another dimension in understanding the function of GR and the role of steroid in transplant immunosuppression.

Abstract:Choosing 8~10 days cultured hippocampal neurons of SD rat, using Calcium Orange AM and DAF-FM diacetate as the fluorescent indicators of intracellular Ca2+ and nitric oxide(NO), simultaneous detection of intracellular Ca2+ and NO was proposed by double-label method on laser scanning confocal microscope (LSCM). The dyeing process includes two steps and "Two Track" mode of LSCM is applied to realize simultaneous detection of intracellular Ca2+ and NO through quickly switching excitation wavelengths. The experiment results show that there is no cross talk between two dyes and the double-label method can reveal the changes of intracellular Ca2+ and NO concentrations under the stimulation of N-methyl-D-aspartate (NMDA), quite consistent with the results of respective single-label experiments. The analysis of slicing image sequences of double-labeled neurons shows that both Ca2+ and NO are mainly located in the center area of cell bodies, while their distribution details are different. The results suggest that the double-label method can simultaneously detect the intracellular Ca2+ and NO in cultured hippocampal neurons and thus provide an approach to investigate the roles of Ca2+ and NO in neurons as well as the interaction between them.
Rapid Enhancement of High Affinity Glutamate Uptake by Glucocorticoids in Rats Cerebral Cortex Synaptosomes

Abstract:The effects of glucocorticoids on the Na+-dependent, high affinity uptake of (3)[H]-L-glutamate in synaptosomes from cerebral cortex of rats were investigated. Glucocorticoids, including corticosterone (B), corticosterone-sulfate (Bs), hydrocortisone-hemisuccinate (F) and dexamethasone-phosphate (DEX) were found stimulating glutamate uptake in synaptosomes after 4 min incubation. The uptake was increased up to 121.4%, 125.5%, 120.8% and 113.6% of the control uptake for 10(-6) mol/L of Bs, B, F and DEX respectively. The stimulation was dose-dependent in the case of Bs, and steroid-specific while deoxycorticosterone and 17 beta-estradiol-sulfate were ineffective, and progesterone and testosterone enhanced the uptake to a lesser extent than B. The rapid stimulations of glucocorticoids on glutamate uptake in synaptsomes, detached from perikaryon, are undoubtedly attributed to the nongenomic mechanisms. The inhibitor of G-protein activation, guanosine 5-O-(2-thiodiphosphate) could at least partially block the stimulation of Bs, suggesting that the G-protein on membranes of synaptosomes might be involved in the effect of glucocorticoids.

Abstract:The traditional genomic theory of steroid action does not fully explain the rapid effects of hormonal steroids, and it is thought that the nongenomic actions mediated by a putative membrane receptor may provide a plausible explanation. Although there is a rich body of evidence to substantiate the rapid, nongenomic effects of steroid hormones, the signal transduction pathways involved have proved to be complex and pleiotropic. Based on previous studies on the rapid, nongenomic actions of glucocorticoid (GC) and the G-protein–protein kinase pathways involved, including our own studies on PC12, SK-N-SH, BT-325 cells, and synaptosomes, in this review we will discuss the issue of multiple signal transduction pathways involved in the rapid, nongenomic effects of GC.
Binding Characteristics of Glucocorticoid Receptors on Synaptic Plasma Membrane from Rats

Abstract:Objective To develop Binding characteristics of glucocorticoid receptors on synaptic plasma membrane from rats. Methods The synaptic plasma membrane(SPM) was prepared from rat brains and there was no cytosolic contamination.The marked enzyme-LDH was used for monitoring the SPM purity. Results(The radioligand binding) assay showed that the radio labeled ligand corticosterone to SPM was in a saturable manner.The saturation binding curve was S shaped and the convex curve in scatchard plot revealed a positive cooperativity.The binding capacity was(450.20±28.20) nmol·L~(-1).Various steroids bound with corticosterone specific binding sites on SPM were in the following order: aldosterone corticosterone ≈ cortisol progesterone testosterone ≈ estradiol desamethasone. Conclusion There were glucocorticoid receptors on SPM.

Abstract:A fine-tuned balance of glucocorticoid receptor (GR) activation is essential for organ formation, with disturbances influencing health outcomes. Excess GR-activation has been linked to brain-related negative outcomes, with unclear underlying mechanisms, especially regarding cell-type specific effects. To address this, we used an model of fetal human brain, induced pluripotent-stem-cell-derived cerebral organoids, and mapped GR-activation effects using single-cell transcriptomics across development. Interestingly, neurons showed targeted regulation of differentiation- and maturation-related transcripts, suggesting a delay of these processes upon GR-activation. Uniquely in neurons, differentially-expressed transcripts were significantly enriched for genes associated with behavior-related phenotypes and disorders. This suggests that aberrant GR-activation could impact proper neuronal maturation, leading to increased disease susceptibility, through neurodevelopmental processes at the interface of genetic susceptibility and environmental exposure.

Abstract:Hormone-activated nuclear receptors (NRs) control myriad cellular processes. The classical paradigm for hormone delivery is secretion from endocrine organs and blood-borne distribution to responding cells. However, many hormones can also be synthesized in the same tissues in which responding cells are found (paracrine signaling). In both endocrine and paracrine signaling, numerous factors affect hormone availability to target cell NRs, including hormone access to and sequestration by carrier proteins, transport across cell membranes, metabolism, and receptor availability. These factors can differ dramatically during development, between anatomical locations, and across cell types, and may cause highly variable responses to the same hormone signal. This has been difficult to study because current approaches are unable to quantify cell-intrinsic exposure to NR hormone ligands, precluding assessment of cell-specific hormone access and signaling. We have used the ligand-dependent interaction of the endogenous glucocorticoid (GC) receptor with chromatin as a biosensor that quantifies systemic access of GCs to cells within tissues at the single cell level, showing that tissues are buffered against circulating GCs. This approach also showed highly targeted paracrine GC signaling within the thymus, where GCs promote the positive selection of thymocytes with moderate affinity for self-antigens and the development of a safe and effective T-cell repertoire. We believe that this and complementary biosensor approaches will be useful to identify endocrine and paracrine target cells in situ and quantify their exposure to hormones regardless of the mode of delivery.

Abstract:Abstract(1) This study investigated the functional genomics of glucocorticoid and opioid receptor stimulation in cellular adaptations using a cultured neuronal cell model. (2) Human SH-SY5Y neuroblastoma cells grown in hormone-depleted serum were treated for 2-days with the glucocorticoid receptor-II agonist dexamethasone (30 nM); the μ-opioid receptor agonist [D-Ala2, N-Me-Phe4, Gly5-ol]-Enkephalin acetate (DAMGO; 1 nM); or dexamethasone (30 nM) plus DAMGO (1 nM). RNA was extracted; purified, reverse transcribed, and labeled cDNA was hybridized to a 10,000-oliogonucleotide-array human gene chip. Gene expression changes that were significantly different between treatment groups and were of interest due to biological function were verified by real-time reverse transcription polymerase chain reaction (RT-PCR). Five relevant genes were identified for which the combination of dexamethasone plus DAMGO, but neither one alone, significantly up-regulated gene expression (ANOVA, P < 0.05). (3) Proteins coded by the identified genes: FRS2 (fibroblast growth factor receptor substrate-2; CTNNB1 (β1-catenin); PRCP (prolyl-carboxypeptidase); MPHOSPH9 (M-phase phosphoprotein 9); and ZFP95 (zinc finger protein 95) serve important neuronal functions in signal transduction, synapse formation, neuronal growth and development, or transcription regulation. Neither opioid, glucocorticoid nor combined treatments significantly altered the cell growth rate determined by cell counts and protein. (4) We conclude that sustained μ-opioid receptor stimulation accompanied by glucocorticoids can synergistically regulate genes that influence neuronal function. Future studies are warranted to determine if combined influences of glucocorticoid fluctuations and opioid receptor stimulation in vivo can orchestrate exagerated neuroadaptation to reinforcing drugs under chronic mild stress conditions.

Radioimmunocytochemical localization of corticotropin-releasing factor and adrenocorticotropin in the hypothalamo-hypophyseal system of the rat: effects of adrenalectomy.

Abstract:Various radioimmunocytochemical approaches have been utilized to localize primary antibody-antigen complexes. Here we examined the binding properties of three different radioiodinated compounds for their ability to label the antibody-antigen complex, including: donkey anti-rabbit immunoglobulin, donkey anti-rabbit F(ab')2-IgG, and a biotinylated goat anti-rabbit secondary antibody followed by [125I]-avidin. These probes were used to localize rabbit primary antisera against corticotropin-releasing factor (CRF) and adrenocorticotropin-releasing hormone (ACTH) in the hypothalamo-hypophyseal system of the rat. The pattern of labeling with each radiolabeled probe was consistent with the light microscopic immunocytochemical staining for CRF and ACTH. The utility of the radioimmunocytochemical method for quantitative analyses was further tested by studying the effects of adrenalectomy (ADX) on the levels of immunoreactive CRF and ACTH in the hypothalamo-hypophyseal system. Computer-assisted microdensitometric analysis of immunoreactive CRF levels in the median eminence indicated that there was a 33% decrease 24 h after ADX. Immunoreactive ACTH levels in the anterior pituitary were significantly decreased from 1 day (38%) to 1 week (36%) after ADX and were increased at 2 weeks (89%). The changes in CRF and ACTH levels, as measured radioimmunocytochemically after ADX, were consistent with previous biochemical studies. These results indicate that computer-assisted radioimmunocytochemical analysis can be used quantitatively to measure immunoreactivity in tissue sections. The high resolution and high sensitivity provided by this method should make it widely applicable.

Abstract:Introduction: Corticotropin-releasing hormone (CRH) signaling through its cognate receptors, CRHR1 and CRHR2, contributes to diverse stress-related functions in the mammalian brain. Whereas CRHR2 is predominantly expressed in choroid plexus and blood vessels, CRHR1 is abundantly expressed in neurons in discrete brain regions, including the neocortex, hippocampus and nucleus accumbens. Activation of CRHR1 influences motivated behaviors, emotional states, and learning and memory. However, it is unknown whether alterations in CRHR1 signaling contribute to aberrant motivated behaviors observed, for example, in stressful contexts. These questions require tools to manipulate CRHR1 selectively. Here we describe and validate a novel Crhr1-FlpO mouse. Methods: Using bacterial artificial chromosome (BAC) transgenesis, we engineered a transgenic mouse that expresses FlpO recombinase in CRHR1-expressing cells. We used two independent methods to assess the specificity of FlpO to CRHR1-expressing cells. First, we injected Crhr1-FlpO mice with Flp-dependent viruses expressing fluorescent reporter molecules. Additionally, we crossed the Crhr1-FlpO mouse with a transgenic Flp-dependent reporter mouse. CRHR1 and reporter molecules were identified using immunocytochemistry and visualized via confocal microscopy in several brain regions in which CRHR1 expression and function is established. Results: Expression of Flp-dependent viral constructs was highly specific to CRHR1-expressing cells in all regions examined (over 90% co-localization). In accord, robust and specific expression of the Flp-dependent transgenic reporter was observed in a reporter mouse, recapitulating endogenous CRHR1 expression. Conclusions: The Crhr1-FlpO mouse enables selective genetic access to CRHR1-expressing cells within the mouse brain. When combined with Cre-lox or site-specific recombinases, the mouse facilitates intersectional manipulations of CRHR1-expressing neurons.

A Quantitative in Situ Hybridization Protocol for Formalin-Fixed Paraffin-Embedded Archival Post-Mortem Human Brain Tissue

Methods

Abstract:The use of radioactive in situ hybridization (ISH) to quantitatively determine low-to-moderate abundant mRNA expression in formalin-fixed, paraffin-embedded archival post-mortem human brain tissue is often limited by non-specific-deposits, visible as speckles. In the present study, optimal hybridization conditions were achieved for quantifying the mRNA expression of histidine decarboxylase (HDC) by a number of alterations in a routine protocol, which included (1) during purification of the oligo-probes, glycogen was omitted as a carrier for precipitation, (2) after precipitation, the labeled probe contained within the pellet was first dissolved in water instead of in hybridization buffer (HBF), (3) during hybridization, the dithiothreitol (DTT) concentration was increased from 200 to 800mM in HBF, and (4) stringencies during hybridization and post-hybridization washes were increased by increasing the temperature. The effect of the adjustment was quantified on adjacent sections from 18 subjects (9 with Parkinson’s disease and 9 controls), by comparing the data from the standard and new protocol. The results showed that the improved protocol brought about significantly clearer background with higher signal-to-noise ratios (p=0.001). We propose that this protocol is also applicable for detection of other lower-abundant genes in human brain tissue and probably in other tissues as well. In the present study, this is not only illustrated for HDC ISH, but also for corticotrophin-releasing hormone mRNA expression in the hypothalamic paraventricular nucleus.
Establishment of reporter cells that respond to glucocorticoids by a transposon-mediated promoter-trapping system

Abstract:Previously, we had established a highly sensitive trap vector system for the efficient isolation of reporter cells for a certain condition of interest. In this study, we used this system to screen reporter cells that express the luciferase and enhanced green fluorescent protein genes in response to dexamethasone, a glucocorticoid receptor agonist to facilitate glucocorticoid signaling research. In total, 10 clones were isolated. The insertion sites of the trap vector were analyzed using 5' rapid amplification of cDNA ends (5' RACE), whereupon LPIN1, PKP2, and FKBP5 were identified as genes that were upregulated by the dexamethasone treatment. Specifically, PKP2 has not previously been focused as a gene that responds to glucocorticoids. The PKP2 mRNA was analyzed and induction of the endogenous gene was confirmed by real-time polymerase chain reaction. Given that PKP2 does not appear to have a consensus glucocorticoid response element (GRE) sequence, this reporter clone could supplement the current GRE-based reporter systems that are prevalently used. Because different clones showed different responses to glucocorticoids, these clones should provide more information than analysis with a single reporter clone. This paper demonstrates that the previously developed trap vector technology can contribute to the rapid construction of drug evaluation systems.
