Abstract:As plastic pollution continues to increase and plastic waste is shredded to form smaller plastic particles, there is growing concern about the potential impact of nanoplastics (NPs) on freshwater ecosystems. In this work, the effects of three surface-modified NPs, including polystyrene (PS), PS-NH2, and PS-COOH, on the growth, photosynthetic activity, oxidative damage, and microcystins (MCs) production/release of Microcystis aeruginosa (M. aeruginosa) were investigated. Results indicated that all three NPs significantly inhibited the growth of M. aeruginosa after a 96 h exposure, and the growth inhibition followed the order of PS-NH2 > PS > PS-COOH (p < 0.05). Meanwhile, all three NPs at the concentration of 100 mg/L significantly increased the content of intra-MCs (115 %, 147 %, and 121 % higher than the control, respectively) and extra-MCs (142 %, 175 %, and 151 % higher than the control, respectively) after a 96 h exposure (p < 0.05). Moreover, our findings also suggested that the potential mechanisms of surface-modified PS NPs on M. aeruginosa growth and MCs production/release were associated with physical constraints, photosynthetic activity obstruct, and oxidative damage. Our findings provided direct evidence for different kinds of surface modifications of PS NPs on freshwater algae and improve the understanding of the potential risk of NPs in aquatic ecosystems.

Abstract:The ubiquitous nature of plastics, particularly nanoplastics, raises concern about their potential effects on primary producer microalgae. Currently, the impacts and potential mechanisms of nanoplastics on microalgae are not fully understood. In this study, the effects of two plain commercial polystyrene nanoplastics (PS-NPs) with different sizes (50 nm and 70 nm) on C. vulgaris were assessed in a concentration range of 0–50 mg/L during 72 h exposure periods. Results revealed that both PS-NPs have dose-dependent toxicity effects on C. vulgaris, as confirmed by the decrease of growth rates, chlorophyll a and esterase activities, and the increase of ROS, MDA, and membrane damage. The membrane damage was caused by the agglomeration of PS-NPs on microalgae and may be the key reason for the toxicity. Compared with 70 nm PS-NPs (72 h EC50 >50 mg/L), 50 nm PS-NPs posed greater adverse effects on algae, with an EC50–72h of 19.89 mg/L. FTIR results also proved the stronger variation of macromolecules in the 50 nm PS-NPs treatment group. This phenomenon might be related to the properties of PS-NPs in exposure medium. The lower absolute zeta potential value of 50 nm PS-NPs induced the stronger interaction between PS-NPs and algae as compared to 70 nm PS-NPs, leading to severe membrane damage and the loss of esterase activity as well as settlement. These findings emphasized the importance of considering the impacts of commercial PS-NPs properties in toxicity evaluation.

Abstract:Purpose: The presence of microplastics (MPs) and nanoplastics (NPs) in aquatic ecosystems has raised serious environmental and health concerns. Polystyrene is one of the most abundant plastic polymers found in the environment. Polystyrene MPs/NPs have harmful implications for human health and their removal from the environment has become a serious challenge. Methods: In this study, we investigated the adsorptive uptake of polystyrene nanoplastics (PS NPs) from aqueous solutions using fly ash-loaded magnetic nanoparticles (FAMNPs) as the magnetic nano-adsorbent. During the factor screening study, the adsorption process was studied as a function of four variables namely pH (5-10), adsorption time (30-120 min), amount of FAMNPs (0.01-0.04 g), and stirring speed (50-200 rpm). Central composite design (CCD) and response surface methodology (RSM) were employed to establish the relationship between the variables. Furthermore, toxicity assessments of PS NPs were checked on a zebrafish model, shedding light on its potential ecological effects. Results: Two variables namely the pH and amount of FAMNPs significantly influenced the adsorption capacity of FAMNPs and were further optimized for subsequent analysis. The optimum operational readings proposed by the model were pH (8.5), and the amount of FAMNPs (0.03 g), resulting in a good adsorption capacity of 29.12 mg/g for PS NPs. The adequacy of the proposed model was evaluated by analysis of variance (ANOVA). Zebrafish embryos exposed to PS NPs revealed physical deformations such as pericardial edema and malformed notochord. Conclusion: The study demonstrates the effectiveness of FAMNPs in the adsorption of PS NPs from aqueous solutions, with optimal conditions identified at pH 8.5 and 0.03 g of FAMNPs using RSM. The adequacy of the model was confirmed through ANOVA analysis. Toxicity assessments on zebrafish embryos exposed to PS NPs revealed significant mortality and physical deformations, highlighting the importance of PS NPs removal for environmental health.

Abstract:The contamination of marine and freshwater environments by nanoplastics is considered a global threat for aquatic biota. Taking into account the most recent concentration range estimates reported globally and recognizing a knowledge gap in polystyrene nanoplastics (PS-NPs) ecotoxicology, the present work investigated the harmful effects of 20 nm and 80 nm PS-NPs, at increasing biological complexity, on the rainbow trout Oncorhynchus mykiss RTG-2 and gilthead seabream Sparus aurata SAF-1 cell lines. Twenty nm PS-NPs exerted a greater cytotoxicity than 80 nm ones and SAF-1 were approximately 4-fold more vulnerable to PS-NPs than RTG-2. The engagement of PS-NPs with plasma membranes was accompanied by discernible uptake patterns and morphological alterations along with a nuclear translocation already within a 30-min exposure. Cells were structurally damaged only by the 20 nm PS-NPs in a time-dependent manner as indicated by distinctive features of the execution phase of the apoptotic cell death mechanism such as cell shrinkage, plasma membrane blebbing, translocation of phosphatidylserine to the outer leaflet of the cell membrane and DNA fragmentation. At last, functional analyses unveiled marked transcriptional impairment at both sublethal and lethal doses of 20 nm PS-NPs, with the latter impacting the "Steroid biosynthesis", "TGF-beta signaling pathway", "ECM-receptor interaction", "Focal adhesion", "Regulation of actin cytoskeleton" and "Protein processing in endoplasmic reticulum" pathways. Overall, a distinct ecotoxicological hazard of PS-NPs at environmentally relevant concentrations was thoroughly characterized on two piscine cell lines. The effects were demonstrated to depend on size, exposure time and model, emphasizing the need for a comparative evaluation of endpoints between freshwater and marine ecosystems.

Abstract:Plastic pollution has become a pressing issue due to its persistence in the environment. Smaller plastics are more easily ingested, potentially exerting greater influences on organisms. In this study, the effects of polystyrene nanoplastics (NP) on the toxic effects, bioaccumulation, biodegradation and enantioselectivity of ibuprofen (IBU) in algae Chlorella pyrenoidosa were explored. The influences on the growth rate, chlorophyll a, total antioxidant capacity (T-AOC), reactive oxygen species (ROS) and lipid peroxidation (MDA) were evaluated after 96 h of exposure to a combination of polystryene NP (1 mg L-1) and IBU (5-100 mg L-1). The results indicated that the inhibitory effect of IBU on C. pyrenoidosa growth was alleviated in the presence of NP. For instance, the 96 h-IC50 value for rac-IBU in the treatment lacking NP was 45.7 mg L-1, and the corresponding value in the treatment containing NP was 63.9 mg L-1. The co-exposure of NP led to a significant enhancement of T-AOC and slight reduction of ROS and MDA compared with the individual exposure (IBU) group, suggesting a decreased oxidative stress. In addition, treatment with NP led to a decreased bioaccumulation and accelerated biodegradation of IBU in C. pyrenoidosa and enhanced removal in the medium. The enantioselective toxicity, bioaccumulation and biodegradation of IBU were observed both in the absence and presence of NP. S-IBU exhibited a greater toxicity, and R-IBU was preferentially accumulated and degraded in C. pyrenoidosa. No interconversion of the two enantiomers occurred regardless of the presence of NP. This consequence implied that the influence of coexistent NP should be considered in the environmental risk assessment of pharmaceuticals and personal care products in aquatic environments. Abstract:Recently, the effects of nanoplastics (NPs) on aquatic organisms have attracted much attention; however, research on the toxicity of NPs to microalgae has been insufficient. In the present study, the effects of polystyrene nanoplastics (nano-PS, 50 nm) on growth inhibition, chlorophyll content, oxidative stress, and algal toxin production of the marine toxigenic dinoflagellate Amphidinium carterae Hulburt were investigated. Chlorophyll synthesis was promoted by nano-PS on day 2 but was inhibited on day 4; high concentrations of nano-PS (≥50 mg/L) significantly inhibited the growth of A. carterae. Moreover, despite the combined effect of superoxide dismutase (SOD) and glutathione (GSH), high reactive oxygen species (ROS) level and malondialdehyde (MDA) content were still induced by nano-PS (≥50 mg/L), indicating severe lipid peroxidation. In addition, the contents of extracellular and intracellular hemolytic toxins in nano-PS groups were significantly higher than those in control groups on days 2 and 8, except that those of extracellular hemolytic toxins in the 100 mg/L nano-PS group decreased on day 8 because of severe adsorption of hemolytic toxins to the nano-PS. Hence, the effects of nano-PS on A. carterae are closely linked to nano-PS concentration and surface properties and exposure time. These findings provide a deep understanding of the complex effects of NPs on toxigenic microalgae and present valuable data for assessing their environmental risks.

Abstract:Given their large production and widespread application, polystyrene nanoplastic (Nano-PS) and ibuprofen (IBU), as particularly relevant compounds, are found to coexist in wastewater treatment plants. In this study, the effect of combined Nano-PS and IBU on the activated sludge processes was investigated through short-term exposure in pure cultures and long-term stress in mixed communities. This study found that the inhibition of SO 4 2− removal by sulfate-reducing bacteria was more pronounced for small-sized Nano-plastics in motion state. Compared to individual pollutants, the toxicity of Nano-PS + IBU mixture showed antagonistic effects at low dosage and synergistic effect at high dosage. In mixed communities, COD and TN removal of sequencing batch reactor decreased from 93.3 % and 90.7 % to 80.1 % and 64.5 % with increasing Nano-PS/IBU. The membrane integrity was destroyed by excess reactive oxygen species, further reducing the protein of extracellular polymeric substances. Furthermore, Nano-PS/IBU altered the community structure, decreased microbial diversity and affected the distribution of abundance and rare taxa. These adverse effects, accompanied by a decline in the relative abundance of nitrifiers (e.g., Nitrosomonas and Ellin6067 ) and genes (e.g., AmoCAB and Hao), directly accounted for the strong deterioration observed in ammonia oxidation. This study provided important information on the effects of nanoplastic and pharmaceutical compounds on activated sludge system, which is a hidden danger to nitrogen removal in biological treatment and should give more attention.

Abstract:Nanoplastics are ubiquitous in marine environments, exhibiting high bioavailability and potential toxicity to marine organisms. However, the impacts of nanoplastics with various surface modifications on marine microalgae remain largely unexplored. This study explored the toxicity mechanisms of two nanoplastic types-polystyrene (PS) and polymethyl methacrylate (PMMA)-with distinct surface modifications on Skeletonema costatum at cellular and molecular levels. Results showed that nanoplastics significantly impaired the growth of microalgae, particularly PS-NH2, which caused the most pronounced growth inhibition, reaching 56.99 % after a 96-h exposure at 50 mg/L. Transcriptomic profiling revealed that nanoplastics disrupted the expression of genes predominantly involved in ribosome biogenesis, aminoacyl-tRNA biosynthesis, amino acid metabolism, and carbohydrate metabolism pathways. The integrated biochemical and transcriptomic evidence highlighted that PS-NH2 nanoplastics had the most adverse impact on microalgae, affecting fundamental pathways such as ribosome biogenesis, energy metabolism, photosynthesis, and oxidative stress. Our findings underscore the influence of surface-modified nanoplastics on algal growth and contribute new understanding to the toxicity mechanisms of these nanoplastics in marine microalgae, offering critical information for assessing the risks of emerging pollutants.
Nanoplastics enhanced the developmental toxicity of aromatic disinfection byproducts to a marine polychaete at non-feeding early life stage

Abstract:Micro/nanoplastics can act as vectors for organic pollutants and enhance their toxicity, which has been attributed to the ingestion by organisms and the "Trojan horse effect". In this study, we disclosed a non-ingestion pathway for the toxicity enhancement effect of nanoplastics. Initially, the combined toxicity of polystyrene microplastics (40 μm) or nanoplastics (50 nm) with three disinfection byproducts (DBPs) to a marine polychaete, Platynereis dumerilii, was investigated. No toxic effect was observed for the micro/nanoplastics alone. The microplastics showed no effect on the toxicity of the three DBPs, whereas the nanoplastics significantly enhanced the toxicity of two aromatic DBPs when the polychaete was in its non-feeding early life stage throughout the exposure period. The microplastics showed no interaction with the P. dumerilii embryos, whereas the nanoplastics agglomerated strongly on the embryonic chorion and fully encapsulated the embryos. This could contribute to higher actual exposure concentrations in the microenvironment around the embryos, as the concentrations of the two aromatic DBPs on the nanoplastics were 1200 and 120 times higher than those in bulk solution. Our findings highlight an important and previously overlooked mechanism by which nanoplastics and organic pollutants, such as DBPs, pose a higher risk to marine species at their vulnerable early life stages. This study may contribute to a broader understanding of the environmental impacts of plastic pollution and underscore the necessity to mitigate their risks associated with DBPs.
Micro(nano)plastics from synthetic oligomers persisting in Mediterranean seawater: Comprehensive NMR analysis, concerns and origins

Abstract:The presence in seawater of low-molecular-weight polyethylene (PE) and polydimethylsiloxane (PDMS), synthetic polymers with high chemical resistance, has been demonstrated in this study for the first time by developing a novel methodology for their recovery and quantification from surface seawater. These synthetic polymer debris (SPD) with very low molecular weights and sizes in the nano- and micro-metre range have escaped conventional analytical methods. SPD have been easily recovered from water samples (2 L) through filtration with a nitrocellulose membrane filter with a pore size of 0.45 μm. Dissolving the filter in acetone allowed the isolation of the particulates by centrifugation followed by drying. The isolated SPD were analysed by 1 H nuclear magnetic resonance spectroscopy ( 1 H NMR), identifying PE and PDMS. These polymers are thus persisting on seawater because of their low density and the ponderal concentrations were quantified in mg/m 3 . This method was used in an actual case study in which 120 surface seawater samples were collected during two sampling campaigns in the Mediterranean Sea (from the Gulf of Salerno to the Gulf of Policastro in South Italy). The developed analytical protocol allowed for achieving unprecedented simplicity, rapidity and sensitivity. The 1 H and 13 C NMR structural analysis of the PE debris indicates the presence of oxidised polymer chains with very low molecular weights. Additionally, the origin of those low molecular weight polymers was investigated by analysing influents and effluents from a wastewater treatment plant (WWTP) in Salerno as a hot spot for the release of SPD: the analysis indicates the presence of low molecular weight polymers compatible with wax PE, widely used for coating applications, food industry, cosmetics and detergents. Moreover, the origin of PDMS debris found in surface seawater can be ascribed to silicone-based antifoamers and emulsifiers.

Abstract:Currents, wave motion, solar radiation, and abrasion are mechanisms responsible for the degradation of large plastic artifacts and contribute to the dispersion of micro and nanoplastics into aquatic ecosystems, which are, currently, the most dangerous threats due to their invisibility and persistence. The present work evaluated the possible lethal and sublethal effects of amino-modified polystyrene nanoplastics (nPS-NH2) with diameters of 50 nm and 100 nm on Artemia salina (A. salina), an organism at the base of the trophic chain of the aquatic system, using a widely used model for the analysis of embryotoxicity from environmental pollutants. For this purpose, after evaluating the biodistribution of nanoplastics in the body of the tested animals, several endpoints such as anomalies, apoptosis, and ROS production were assessed. In addition, particular attention was dedicated to evaluating the correlation between toxicity and the particle size tested. The results reported that, despite the absence of a lethal impact, several sublethal effects involving gut and body size malformations, as well as the enhancement of apoptosis and oxidative stress in relation to an increase in tested concentration and a decrease in nanoparticle size.

Abstract:Micro- and nanoplastics have been detected in environmental compartments from the highest mountains to the deepest seas. They have been shown to be present at almost all trophic levels, and within humans they have been detected in numerous organs and human stool. Whilst their ubiquitous nature is indisputable, little is known about the health risks they may present. Much current research is focussed on the production of test materials with which to perform the necessary health studies. An important aspect of this is the correct storage and suspension of the materials to ensure they remain stable both chemically and with regards to size and shape. In this review, we look at the chemical stability of nine common polymers in a range of liquids; first with the use of commercial compatibility charts and then with a more quantitative approach using Hansen solubility parameters. We then look at stability with regards to particle agglomeration, whether and how stable compositions can be predicted, and which dispersants can be added to increase stability. Finally, we discuss the role of bio-surfactants and the eco-corona and how these may offer a route to both better stability and environmental relevance.

Abstract:The plastic value chain, central part of modern living, caused environmental pollution and bioaccumulation of plastic nanoparticles (PNPs). Their ubiquitous presence in different environmental and biological compartments has become a serious threat to human health and ecosystems. Frequently used plastic materials such as polypropylene (PP), polystyrene (PS) and polyethylene (PE) have been detected in the form of PNPs in the food chain, soil, water and air, as well as in human feces and blood. In this study, we aimed to provide novel insights in endocrine disrupting properties of PNPs using in vitro estrogen receptor (ER) transactivation assay. The effects of PP-NPs, PE-NPs and PS-NPs and their mixture on T47D-KBluc cell line stably transfected with luciferase as reporter enzyme was evaluated by means of cytotoxicity, cellular uptake and ER activation. Tested dose range for PNPs was 0.001 – 10 mg/L. Both cellular uptake and cytotoxicity for all PNPs was found to be dose-dependent. Only the highest dose of PP-NPs and PE-NPs induced apoptosis and cell death, while PS-NPs were not cytotoxic in tested dose range. For tested concentrations, PP-NPs and PE-NPs showed significant agonistic activity on ER, while PS-NPs cannot be considered ER active. When, applied as mixture, PNP demonstrated additive toxicity effects compared to the effect of each individual PNPs. Additivity was also observed for ER agonistic effect of PNPs mixture according to the benchmark dose-addition modelling approach. This study provides missing science-based evidence on endocrine disrupting effects of PE-NPs, PP-NPs, PS-NPs and their mixtures and highlights the importance of considering unintentional, aggregate and combined exposure to different PNPs in risk management.

Preparation of Nanoplastic Particles as Potential Standards for the Study of Nanoplastics

Abstract:Nanoplastics (NPs) are plastic fragments that are small enough to be absorbed by organisms through ingestion or inhalation. Recent studies indicate that nanoplastics can be ubiquitous in the environment, and there are growing concerns regarding the impacts of nanoplastics on the health of humans and other organisms. However, quantitative information on nanoplastics in the environment is still very limited, and most previous toxicity studies have used only polystyrene (PS) particles because of a lack of appropriate model particles of other plastics. We developed a nanoprecipitation-based method for the preparation of nanoplastic particles of five major polymers: low-density polyethylene (LDPE), high-density polyethylene (HDPE), polypropylene (PP), polyvinyl chloride (PVC), and polystyrene. A major advantage of our method is that the nanoplastic particles are prepared without using reagents that can remain in the particles as impurities. Analysis of the prepared particles' molecular weight (M<sub>w</sub>) distributions, crystallinities, and thermal properties revealed that their compositions and constitutions were within the general ranges for commercial products. The mechanisms underlying the formation of low-density polyethylene particles via our method were investigated by means of a simple population balance model, and particle diameter was found to be linearly correlated with the suspension density of the nanoplastic dispersion up to 0.4 mg·mL<sup>-1</sup>. Future studies should focus on improving our method to allow for precise, scale-independent production of nanoplastic particles. Methods for the preparation of labeled particles are also needed so that such particles can be used in nanoplastic risk assessments.

Abstract:The problem of microplastics (MPs) contamination in food has gradually come to the fore. MPs can be transmitted through the food chain and accumulate within various organisms, ultimately posing a threat to human health. The concentration of nanoplastics (NPs) exposed to humans may be higher than that of MPs. For the first time, we studied the differences in toxicity, and potential toxic effects of different polymer types of NPs, namely, polyethylene terephthalate (PET), polyvinyl chloride (PVC), and polystyrene (PS) on HepG2 cells. In this study, PET-NPs, PVC-NPs, and PS-NPs, which had similar particle size, surface charge, and shape, were prepared using nanoprecipitation and emulsion polymerization. The results of the CCK-8 assay showed that the PET-NPs and PVC-NPs induced a decrease in cell viability in a concentration-dependent manner, and their lowest concentrations causing significant cytotoxicity were 100 and 150 μg/mL, respectively. Moreover, the major cytotoxic effects of PET-NPs and PVC-NPs at high concentrations may be to induce an increase in intracellular ROS, which in turn induces cellular damage and other toxic effects. Notably, our study suggested that PET-NPs and PVC-NPs may induce apoptosis in HepG2 cells through the mitochondrial apoptotic pathway. However, no relevant cytotoxicity, oxidative damage, and apoptotic toxic effects were detected in HepG2 cells with exposure to PS-NPs. Furthermore, the analysis of transcriptomics data suggested that PET-NPs and PVC-NPs could significantly inhibit the expression of DNA repair-related genes in the p53 signaling pathway. Compared to PS-NPs, the expression levels of lipid metabolism-related genes were down-regulated to a greater extent by PET-NPs and PVC-NPs. In conclusion, PET-NPs and PVC-NPs were able to induce higher cytotoxic effects than PS-NPs, in which the density and chemical structure of NPs of different polymer types may be the key factors causing the differences in toxicity.

Abstract:Plastics pose a health hazard to living beings and the environment. Plastic degradation produces nano-sized plastic particles (NPs) that end up in terrestrial and aquatic ecosystems, including oceans, rivers, and lakes. Their presence in air, drinking water, sediments, food, and personal care products leads to a variety of exposure routes for living beings, including humans. The toxicity mechanisms of these nanomaterials (NMs) in living organisms and ecosystems are currently unknown, making it a priority to understand their effects at the molecular and cellular levels. The zebrafish (Zf) (Danio rerio) is a model organism which has a high homology with humans and has been widely used to assess the hazard of different xenobiotics. In this study, the expression changes of different genes in 120 hpf Zf embryos (Zfe) after exposure to polystyrene (PS) NPs (30 nm) at concentrations of 0.1, 0.5 and 3 ppm were investigated. The results showed that the gene encoding heat shock protein (hsp70) was down-regulated in a dose-dependent manner. The genes encoding superoxide dismutase (SOD 1 and SOD 2), apoptotic genes (cas 1 and cas 8) and interleukin 1-β (il1β) were activated at the concentration of 3 ppm PS NP, while the anti-apoptotic gene Bcl2α was inhibited at 0.5 and 3 ppm. In addition, the neurotransmitter-related gene Acetyl-Cholinesterase (ache) was significantly inhibited and the DNA repair genes (gadd45α and rad51) were also down-regulated. In contrast, the mitochondrial metabolism-related gene cox1 did not alter its expression in any of the treatments. Most of the changes in gene expression occurred at the highest concentration of NPs. Overall, the results indicated that NPs generated cellular stress that caused certain alterations in normal gene expression (oxidative stress, apoptotic and inflammatory processes, neurotoxicity and anti-apoptotic proteins), but did not cause any mortality after 120 hpf exposure at the three concentrations assayed. These results highlight the need for further studies investigating the effects, at the molecular level, of these materials in humans and other living organisms.
