Abstract:α‐Indol‐3‐yl α,β‐unsaturated carbonyl compounds are synthesized from 3‐propargyl indoles, obtained by direct propargylation of indoles, via a gold‐catalyzed tandem oxidation–1,2‐indole migration reaction in the presence of pyridine N‐oxides. Fine‐tuning of the catalyst and oxidant enables the reaction of 3‐propargyl indoles bearing different substituents. The order of oxidation and indole migration is determined by the terminal or internal nature of the alkyne moiety. In addition, the process can be coupled with additional reactions, allowing a rapid increase in molecular complexity or the design of more elaborated tandem reactions. In this sense, indole derivatives bearing an alkenyl substituent at the alkyne position evolve through a gold‐catalyzed tandem oxidation–1,2‐indole migration–Nazarov cyclization producing α‐indolyl cyclopentenones.

Abstract:An efficient new method has been developed for the synthesis of ketone‐integrated cyclopentene‐fused indoline derivatives with a high regioselectivity through gold‐catalyzed oxidative cyclo‐coupling of 1,3‐enynes with indoles by using 2,6‐dichloropyridine N‐oxide as the terminal oxidant. A plausible mechanism has been proposed, in which, a cascade process of nucleophilic trapping of the vinyl gold carbene surrogate with indole followed by the subsequent intramolecular cyclization provides the dearomative formal [3+2] cycloaddition product. In addition, a scale‐up reaction and further derivation via cyclopentene's epoxidation & hydrazone derivative's 1,5‐ electrocyclization have been performed.

Abstract:The Fischer indole synthesis is perhaps the most powerful method for indole preparation, but it often suffers from low regioselectivities with unsymmetric aliphatic ketone substrates and strong acidic conditions and is not suitable for α,β-unsaturated ketones. In this article, we disclose an efficient synthesis of N-protected indoles from N-arylhydroxamic acids/N-aryl-N-hydroxycarbamates and a variety of alkynes via a cooperative gold and zinc catalysis. The zinc catalysis is similar to the related zinc ion catalysis in metalloenzymes such as human carbonic anhydrase II and substantially enhances the O-nucleophilicity of N-acylated hydroxamines by forming the corresponding Zn chelates. The Zn chelates can attack gold-activated alkynes to form O-alkenyl-N-arylhydroxamates, which can undergo facile 3,3-sigmatropic rearrangements and subsequent cyclodehydrations to yield N-protected indole products. This new chemistry offers several important improvements over the Fischer indole synthesis: a) the reaction conditions are mildly acidic and can tolerate sensitive groups such as Boc; b) broader substrate scopes including substrates with pendant carbonyl groups (reactive in the Fischer chemistry) and alkyl chlorides (e.g., 3f); c) better regioselectivities for the formation of 2-substituted indoles under much milder conditions; d) 2-alkenylindoles can be prepared readily in good to excellent yields, but the Fischer chemistry could not; e) with internal alkynes both steric and electronic controls are available for achieving good regioselectivities, while the Fischer chemistry is in general problematic.

Abstract:Gold-catalyzed and NaH-supported intramolecular cyclization of N-propargyl indole derivatives with pyrazole and pyrrole units attached to indole is described. An efficient route to the synthesis of pyrazolodiazepinoindole, pyrazolopyrazinoindole, and pyrrolopyrazinoindole has been established. First, N-propargyl 2-(1H-pyrazol-5-yl)-1H-indole and 2-(1H-pyrrol-2-yl)-1H-indole were synthesized. Introduction of various substituents into the alkyne functionality was accomplished by Sonogashira cross-coupling reaction. Gold-catalyzed cyclization of pyrazoles having a terminal alkyne afforded the 6-exo-dig cyclization product. However, exclusive formation of 7-endo-dig cyclization products was observed with internal alkynes. On the other hand, cyclization with NaH only resulted in the formation of 6-exo-dig cyclization products regardless of the substitution of the alkyne functionality. Allenic intermediates were postulated for this outcome.

Abstract:The synthesis of diverse products from the same starting materials is always attractive in organic chemistry. Here, a palladium-catalyzed substrate-controlled regioselective functionalization of unactivated alkenes with trifluoroacetimidoyl chlorides has been developed, which provides a direct but controllable access to a variety of structurally diverse trifluoromethyl-containing indoles and indolines. In more detail, with respect to γ,δ-alkenes, 1,1-geminal difunctionalization of unactivated alkenes with trifluoroacetimidoyl chloride enables the [4 + 1] annulation to produce indoles; as for β,γ-alkenes, a [3 + 2] heteroannulation with the hydrolysis product of trifluoroacetimidoyl chloride through 1,2-vicinal difunctionalization of alkenes occurs to deliver indoline products. The structure of alkene substrates differentiates the regioselectivity of the reaction.

Abstract:We have disclosed silver(I)-induced switching of regioselectivity in rhodium-catalyzed C-H alkynylation of indole derivatives with the help of a pivaloyl directing group by tuning C-H metalation modes. The judicious choice of AgOAc, Ag(2)O, and Ag(2)CO(3) affords an array of C2-alkynylated indoles, C4-alkynylated indoles, and C2,C4-dialkynylated indoles, respectively. The synthetic utility of the alkyne fragment is demonstrated by derivatization into valuable indole-based compounds.

Abstract:The functionalized 3-vinylindoles were obtained through the cyclization/nucleophilic addition of ynamides and o-alkynylanilines catalyzed by gold catalyst with high stereoselectivity and site-selectivity. The theoretical researches suggested that Cα of ynamides activated by gold complex was favorable to the addition at the C3 position of indoles. Additionally, JohnphosAu(MeCN)SbF6 could efficiently reduce the activation energy barrier of the addition process compared to PicAuCl2 and Au(PPh3)NTf2, and the steric hindrance effect of JohnphosAu(MeCN)SbF6 facilitates the formation of 3-vinylindoles with E configuration.

Abstract:We report on intermolecular gold‐catalyzed nitrene transfer, utilizing o‐functionalized aryl azides as nitrene transfer reagents. This catalytic reaction provides a modular route to 7‐functionalized 2‐aminoindoles. Due to the optimized reaction conditions (toluene, 80–100 °С), a variety of functional substituents proved compatible (36 examples; yields up to 98%). The reaction can be performed using both homogeneous and heterogeneous catalytic variants with a low catalyst loading (1 mol %). The synthetic potential of the obtained products was illustrated by post‐modifications of the indole backbone and peripheral substituents. A plausible reaction mechanism includes the generation of intermediate α‐imino carbenes directly from the o‐functionalized aryl azides or other nitrene sources, such as anthranils or benzofuroxans, which form during the reaction in the cases of the specific aryls.

Abstract:A novel gold (I)‐catalyzed synthetic strategy has been achieved for an efficient construction of indenylidene derivatives from substituted 1,6‐diynes. This reaction describes the unique reactivity of gold catalysis in facilitating the intramolecular [3,3]‐sigmatropic rearrangement, 5‐exo dig cyclization followed by 1,5‐migration of acryl group, resulting in the formation of substituted indenylidenes. Various substituted indenylidenes were successfully synthesized with up to 92% yields. In this protocol, two new C‐C bonds were sequentially formed atom economically in one pot.

Abstract:A highly efficient gold-catalyzed cycloisomerization of 1,5-diynes was developed. Various functional groups are tolerated under the mild reaction conditions, which provides an alternative approach for the synthesis of indeno[1,2-c]furans. On the basis of mechanistic studies, including crossover experiments, deuterium labeling, and computational chemistry, the product formation proceeds via a formal [5,5]-sigmatropic rearrangement, a yet unknown reactivity pattern in gold catalysis. Abstract:We report a switchable synthesis of acylindoles and quinoline derivatives via gold‐catalyzed annulations of anthranils and ynamides. With an N,O‐ligand on a gold(III) catalyst α‐imino gold carbenes, in situ generated from anthranils, could undergo electrophilic attack to the aryl π‐bond followed by unexpected and highly selective 1,4‐ or 1,3‐acyl migrations to form 6‐acylindoles or 5‐acylindoles. With the JohnPhos ligand gold(I) carbenes experienced carbene/carbonyl additions to deliver quinoline oxides. Some of these epoxides represent valuable substrates for the preparation of 3‐hydroxylquinolines, quinolin‐3(4H)‐ones and polycyclic compounds via facile in situ rearrangements. The reaction can be efficiently conducted on gram scale and the obtained products are valuable substrates for preparing other potential useful compounds. A computational study explained the unexpected selectivities and the dependency of the reaction pathway on the oxidation state and ligand of gold. With gold(III) the barrier for the formation of the strained oxirane ring is too high, with gold(I) this transition state becomes accessible. Also, the barriers for the migrations of the substituents on the intermediate sigma‐complexes perfectly explain the observed substitution pattern in the final product.

Abstract:Nitrogen-containing heterocyclic compounds are important motifs of pharmaceuticals and functional materials, and there has been a growing interest in new synthetic methods for their preparations. In this paper, we report a direct cross-coupling reaction of indole with N-aryl tetrahydroisoquinolines in the presence of gold catalyst. The reaction is compatible with a wide range of substituted indoles, to enable the formation of various alkylated heteroarenes under very mild reaction conditions.

Abstract:A cooperative N-heterocyclic carbene (NHC)/iridium catalysis has been developed to achieve highly stereoselective and regiodivergent [3 + 2] and [3 + 3] annulation reactions of 2-indolyl allyl carbonates with enals. The use of the NHC catalyst has introduced switchable homoenolate and enolate intermediates from the common enal precursor via a simple adjustment of reaction conditions in a predictable manner. This protocol furnishes two types of biologically important products, pyrrolo[1,2-a]indoles and pyridine[1,2-a]indoles, with high diastereo- and enantioselectivities (up to >20:1 dr and >99% ee). Notably, all four stereoisomers of these products with two vicinal stereocenters could be afforded through permutations of the enantiomers of the two chiral catalysts. Mechanistic investigations and further computational density functional theory calculations give an explanation of the origin of the regioselectivity. Abstract:A palladium-catalyzed direct C-H bond sulfonylation of indoles with the insertion of sulfur dioxide is achieved through a three-component reaction of 1-(pyridin-2-yl)indoles, DABCO·(SO2)2, and aryldiazonium tetrafluoroborates under mild conditions. Diverse 2-sulfonated indoles are generated by using 10 mol % of palladium(II) bromide as the catalyst at room temperature. This synthetic approach is efficient by merging palladium catalysis and insertion of sulfur dioxide via a radical process. 2-Pyrimidinyl can be used as the directing group well as for the C-H bond sulfonylation. Additionally, the directing group can be easily removed.
