Abstract:The utilization of biomass in the production of renewable bioenergy and biomaterials has been a popular topic since the past decades as they are rich in carbohydrates. Most biomasses, such as wood, monocotyledons, and agriculture residues, need to be pretreated before the conversion of carbohydrates in order to break down the recalcitrant cell wall structure and increase the fiber accessibility. To date, a variety of pretreatment methods have been developed that vary from physical to chemical and biological methods. Pretreatment processes affect the cell wall physical structure as well as the chemical structure of the cell wall constituents. Comparing to the studies of the cellulose and hemicelluloses structural changes during pretreatment, such studies on lignin are relatively limited. On the other hand, in order to utilize the part of lignin from biorefinery processes, the understanding of the lignin structural changes during the refining process becomes important. In this study, typical pretreatment methods such as hydrothermal pretreatment, alkaline pretreatment, biodegradation, and oxidative pretreatment are introduced and their corresponding impacts on the lignin structures are reviewed.
Sulfite Pretreatment Enhances Tobacco Stalk Deconstruction for Cellulose Saccharification and Lignin Pyrolysis

Abstract:Sulfite-catalyzed acid pretreatment to overcome the inherent recalcitrance of biomass offers a significant advantage in terms of obtaining high glucose conversion. However, the residual lignin after enzymatic hydrolysis has not been fully exploited. Herein, this study introduced a joint approach using sulfite-catalyzed acid pretreatment (SPROL) and pyrolysis to upgrade tobacco stalk to produce fermentable sugar, and the resulting lignin is used to produce bio-oil and bio-char. The results suggest that SPROL pretreated tobacco stalk yields a high cellulose-based glucose selectivity of 75.9% with 15 FPU/g substrate enzyme dosage at 50 °C after 72 h of enzymolysis. Lignin characterization reveals that sulfonation occurred during SPROL pretreatment, and as the dosage of sulfonating agent increased, the thermal stability of the residue lignin decreased. After sample pyrolysis at 600 °C for 30 min, approximately 22%, 33%, and 45% of the lignin undergoes conversion into bio-oil, bio-char, and gas products, respectively. The bio-oil analysis results demonstrated that acetic acid is the most abundant identified GC-MS component at around 69.91% at the optimal condition, which implied that it could be of high value when utilized for pyroligneous acid. This research provides a synthetic approach using the SPORL technique to process tobacco stalk into fermentable sugar, bio-oil, and bio-char, which is significant for the commercial utilization of agricultural waste into value-added products.

Abstract:The efficient utilization of crop waste is limited because of plant cell wall recalcitrance. Solving this problem through sustainable processing while producing value-added products is crucial. Here we report a rapid lignocellulose fractionation method applied to common reed using milling under mild alkaline conditions without involving any organic solvents or other reagents. Lignin-containing cellulose nanofiber (LCNF) and native-like lignins were obtained in high yield. LCNF films prepared from the LCNF suspension could have their properties, such as transparency and hydrophobicity, adjusted by changing the pretreatment conditions. Overall this biomass fractionation provides value-added products and is an alternative to simply burning the crop waste.
Mild three-stage alkali-oxygen treatment preserving the native macromolecular structure of lignin for effective disassembling of tobacco stalk

Abstract:Tobacco stalks, as one of the annual economic crops rich in biomacromolecules such as cellulose and hemicellulose, are more difficult to decompose into cellulose fibers due to their high degree of lignification compared to other ordinary straw feedstocks, resulting in their underutilization. In this study, we developed a mild three-stage alkali‑oxygen (AO) process to efficiently deconstruct the tobacco stalk cell walls. The process, involving alkaline dosages of 15 %, 10 %, and 3 % at each stage, effectively dissociated the cell walls and yielded cellulose fibers with high brightness (42.0 % ISO). The organics in the spent liquor, including lignin, hemicellulose, and small-molecular extracts, were isolated through acid/ethanol precipitation and organic solvent extraction. Lignin characterization by 2D HSQC NMR indicated that the majority of native β-aryl ether linkages were preserved after AO treatment, making it suitable for producing chemicals or biofuels via depolymerization. Additionally, the small-molecular extracts contained numerous depolymerized products from lignin and carbohydrates, as well as bioactive compounds derived from the tobacco stalk. Overall, this mild, efficient, and eco-friendly process offers a promising approach for the valorization of tobacco stalks and similar biomass resources.

Abstract:The value-added utilization of tobacco stalk lignin is the key to the development of tobacco stalk resources. However, the serious heterogeneity is the bottleneck for making full use of tobacco stalk lignin. Based on this, lignin was separated from tobacco stalk through hydrothermal assisted dilute alkali pretreatment. Subsequently, the tobacco stalk alkaline lignin was fractionated into five uniform lignin components by sequential solvent fractionation. Advanced spectral technologies (FT-IR, NMR, and GPC) were used to reveal the effects of hydrothermal assisted dilute alkali pretreatment and solvent fractionation on the structural features of tobacco stalk lignin. The lignin fractions extracted with n -butanol and ethanol had low molecular weight and high phenolic hydroxyl content, thus exhibiting superior chemical reactivity and antioxidant capacity. By contrast, the lignin fraction extracted with dioxane had high molecular weight and low reactivity, nevertheless, the high residual carbon rate made it suitable as a precursor for preparing carbon materials. In general, hydrothermal assisted dilute alkali pretreatment was proved to be an efficient method to separate lignin from tobacco stalk, and the application of sequential solvent fractionation to prepare lignin fractions with homogeneous structural features has specific application prospect.

Abstract:Lignocellulose, a kind of biological resource widely existing in nature, which can be transformed into value-added biochemical products through saccharification, fermentation or chemical catalysis. Pretreatments are the necessary step to increase the accessibility and digestibility of lignocellulose. This paper comprehensively reviewed different pretreatment progress of lignocellulose in recent year, including mechanical/thermal, biological, inorganic solvent, organic solvent and unconventional physical-chemical pretreatments, focusing on quantifying the influence of pretreatments on subsequent biomass conversion. In addition, related pretreatment techniques such as genetic engineering, reactor configurations, downstream process and visualization technology of pretreatment were discussed. Finally, this review presented the challenge of lignocellulose pretreatment in the future.

Abstract:The resourcilization technologies of stalks, typically as ethanol production technology, have caught much attention due to its feasibility and valuable production. Pretreatment and hydrolyzation are the key processes of ethanol production from stalks, and multifarious chemical and biological technologies have been developed for stalks conversion. Supercritical/subcritical technology demonstrate obvious advantages over traditional ways, such as much higher reaction rate, not requiring additional catalyst, and no inhibitory reaction of products. Progress in research and development of supercritical/subcritical technology for pretreatment and hydrolyzation is reviewed in this paper, especially the principle, process and interrelated research of combined supercritical/subcritical technology, which includes the primary treatment in supercritical water for separating the polymeric components and hydrolyzing of cellulose to oligosaccharides, and the secondary treatment in subcritical water for hydrolyzing of oligosaccharides to glucose. Additional information on the research of hydrothermal treatment is also provided. Furthermore, the prospect of research and application of supercritical/subcritical technology is discussed.

Abstract:Lignin polymer as a natural aromatic macromolecule presents significant prospects in producing functional and sustainable materials, and achieving a comprehensive characterization will facilitate their target valorization. In the present study, deep eutectic solvent (DES) and alkaline delignification were adopted to deconstruct tobacco stalk before and after hydrothermal pretreatment, obtaining diverse lignin fractions with fascinating characteristics. DES lignin exhibited a higher yield and homogenous molecular structure than MWL. A severe cleavage of the inter-unit linkages in lignin was also observed. This result mostly originated from the efficient delignification of the DES deconstruction system adopted. Moreover, all the recovered lignin fractions exhibited good micro-nanoparticle size that can enhance the valorization of lignin in nanomaterial production, in which the hydrothermal-assisted DES deconstruction promoted the formation of the smaller lignin nanoparticle size. Next, all the recovered lignin presented an excellent UV absorption and structure-related absorption performance or thermal properties. Overall, this work provides an important foundation for further exploiting DES/alkaline delignification lignin that can be applied as an ideal feedstock for producing sustainable functional or micro/nanomaterials.

Abstract:Exploration of lignocellulosic biomass provides a sustainable and eco-friendly route for producing liquid fuels, materials, and chemicals. However, direct utilization of lignocelluloses is limited by the stable and complicated cross-linking structure of the plant cell wall. Hydrothermal pretreatment (HTP) is a green and cost-effective technology because it can disrupt lignin-carbohydrate complex (LCC) linkages, dissolve hemicelluloses and lignin, and redistribute lignin in the cell wall layers without utilization of any chemicals. Thus, HTP is expected to achieve industrial scale in second-generation biorefineries and circular bioeconomies. This review analyzed the deconstruction of lignocelluloses by HTP, with particular emphasis on the formation mechanism of hemicellulose degradation products and the structural evolution of hemicelluloses and lignin accompanying HTP. Meanwhile, the formation mechanism of pseudolignin and its effect on the enzymatic hydrolysis of cellulose as well as strategies for inhibiting lignin recondensation were discussed.
Biodegradation of lignin and nicotine with white rot fungi for the delignification and detoxification of tobacco stalk

Abstract:BackgroundTobacco stalk is one kind of abundant crop residues in China. The high lignification of tobacco stalk increases its reusing cost and the existing of nicotine will cause serious pollution. The biodegradation of lignocellulosic biomass has been demonstrated to be an environmental and economical approach for the utilization of plant stalk. Meanwhile, many nicotine-degrading microorganisms were found in nature. However, microorganisms which could degraded both nicotine and lignin haven’t been reported. Therefore, it’s imperative to find some suitable microorganisms to break down lignin and simultaneously remove nicotine in tobacco stalk.ResultsThe nicotine in tobacco stalk could be degraded effectively by Trametes versicolor, Trametes hirsute and Phanerochaete chrysosporium. The nicotine content in tobacco stalk was lowered to below 500 mg/kg (a safe concentration to environment) after 10 days of fermentation with Phanerochaete chrysosporium and Trametes versicolor, and 15 days with Trametes hirsute. The degradation rate of lignin in the fermented tobacco stalk was 37.70, 51.56 and 53.75% with Trametes versicolor, Trametes hirsute and Phanerochaete chrysosporium, respectively. Meanwhile, 24.28% hemicellulose was degraded by Phanerochaete chrysosporium and 28.19% cellulose was removed by Trametes hirsute. Through the enzyme activity analysis, the main and highest ligninolytic enzymes produced by Phanerochaete chrysosporium, Trametes hirsute and Trametes versicolor were lignin peroxidase (88.62 U · L−1), manganese peroxidase (100.95 U · L−1) and laccase (745.65 U · L−1). Meanwhile, relatively high and stable cellulase activity was also detected during the fermentation with Phanerochaete chrysosporium, and the highest endoglucanase, exoglucanase and filter paper enzyme activities were 0.38 U · mL−1, 0.45 U · mL−1 and 0.35U · mL−1, respectively. Moreover, the products in the fermentation of tobacco stalk with P. chrysosporium were identified with GC-MS, besides the chemicals produced in the degradation of lignin and nicotine, some small molecular valuable chemicals and fatty acid were also detected.ConclusionsOur study developed a new method for the degradation and detoxification of tobacco stalk by fermentation with white rot fungi Phanerochaete chrysosporium and Trametes hirsute. The different oxidative enzymes and chemical products detected during the degradation indicated a possible pathway for the utilization of tobacco stalk.

Abstract:With the energy problem increasingly serious,to seek renewable clean energy has become the key problem to be solved. Transformation of agricultural and forestry wastes into new materials,calorific value of energy,chemical materials are becoming an important new tendency. Bioethanol is attacking more and more attentions since it is a sustained development model of environmental protection type. Pretreatment is an initial step in ethanol preparation process and traditional chemical-pretreatment methods consume a lot of energy, require highly on equipment, seriously degrade hemicellulose and pollute environment seriously. Besides, the traditional pretreatment methods were not focused on hemicellulose and lignin for high value applications, facing the low-coefficient utilization of whole biomass and the financial upside of single product model. As one of the pretreatment methods,biological process is an eco-friendly and low-cost process, but it also has disadvantages of hydrothermal pretreatment tries to keep the structure of natural macromolecules,and break plant cells at a certain degree,in which water is the only reagent. It has such advantages as lower chemical dosage,environmental friendly,low operation costs and the major effects of hydrothermal pretreatment on biomass cell wall are breaking the bonds of lignin-carbohydrate connection ( LCC ) ,removing parts of hemicellulose, modifying lignin and reducing cellulose crystallinity. Inhibitors ( furfural,5-hydroxymethyl furfural,formic acid,acetic acid) are generated in the hydrothermal pretreatment process and restrain the biological degradation process. The quantity of inhibitors varies indifferent pretreatment conditions and can be controlled by changing the conditions. Enzyme hydrolysis is a bioprocess transforming cellulose into monosaccharide,more time and lower ethanol content could be obtained by direct fermentation of pretreated substrate. Hemicellulose and lignin hinder enzyme contacting with cellulose. When hemicellulose and lignin are broken by pretreatment,the contacting efficiency of enzyme and cellulose could be increased. The yield of ethanol increased with the increasing temperature,however,the yield decreased when the temperature was too high. The chemical and structural changes of cellulose,lignin and hemicelluloses are summarized,in this paper,as well as the generated inhibition in the hydrothermal process. The effects of various pretreatment conditions on enzyme hydrolysis and ethanol fermentation are also discussed in detail.

Abstract:Alkaline pretreatment has great potential for the separation of biomass components. In this study, taking bamboo and cotton stalk as raw materials, two-stage alkaline pretreatment was designed for evaluating the components fractionation. The chemical structure of isolated xylan from bamboo and cotton stalk have a backbone of (1 → 4)-β- D -Xylp units attached with 4-O-Me-α- D -GlcpA and/or α-L-Araf units as the branched chain, characterized by Fourier transform infrared spectroscopy and Heteronuclear Single Quantum Coherence Nuclear Magnetic Resonance. Its excellent pretreatment performance was mainly due to the alkali solution weakening the hydrogen bond between cellulose and hemicellulose and saponifying the ester bond between hemicellulose and lignin, thus increasing the fiber porosity. The cellulose enzymatic hydrolysis yield of two-stage alkaline pretreatments cotton stalks and bamboo could reach to 74.2% and 82.1%. The recovery of xylan and lignin of two-stage alkaline pretreatments cotton stalks and bamboo was 85.4% and 84.5%, and 85.1% and 85.1%. This technology was a great potential to promote the efficiency of the component separation of nonwood biomass.

Abstract:Energy crops play a vital role in meeting future energy and chemical demands while addressing climate change. However, the idealization of low-carbon workflows and careful consideration of cost-benefit equations are crucial for their more sustainable implementation. Here, we propose tobacco as a promising energy crop because of its exceptional water solubility, mainly attributed to a high proportion of water-soluble carbohydrates and nitrogen, less lignocellulose, and the presence of acids. We then designed a strategy that maximizes biomass conversion into bio-based products while minimizing energy and material inputs. By autoclaving tobacco leaves in water, we obtained a nutrient-rich medium capable of supporting the growth of microorganisms and the production of bioproducts without the need for extensive pretreatment, hydrolysis, or additional supplements. Additionally, cultivating tobacco on barren lands can generate sufficient biomass to produce approximately 573 billion gallons of ethanol per year. This approach also leads to a reduction of greenhouse gas emissions by approximately 76% compared to traditional corn stover during biorefinery processes. Therefore, our study presents a novel and direct strategy that could significantly contribute to the goal of reducing carbon emissions and global sustainable development compared to traditional methods.

Abstract:Straw biomass is an inexpensive, sustainable, and abundant renewable feedstock for the production of valuable chemicals and biofuels, which can surmount the main drawbacks such as greenhouse gas emission and environmental pollution, aroused from the consumption of fossil fuels. It is rich in organic content but is not sufficient for extensive applications because of its natural recalcitrance. Therefore, suitable pretreatment is a prerequisite for the efficient production of fermentable sugars by enzymatic hydrolysis. Here, we provide an overview of various pretreatment methods to effectively separate the major components such as hemicellulose, cellulose, and lignin and enhance the accessibility and susceptibility of every single component. This review outlines the diverse approaches (e.g., chemical, physical, biological, and combined treatments) for the excellent conversion of straw biomass to fermentable sugars, summarizes the benefits and drawbacks of each pretreatment method, and proposes some investigation prospects for the future pretreatments.

Abstract:As a clean and renewable energy, bioenergy is one of the most promising alternatives to fossil fuels. Lignocellulose possesses great potential for bioenergy production, but the recalcitrant and heterogeneous structure limits its application. Pretreatment technology offers an effective solution to fractionate the main components of the lignocellulose and uncover the available cellulose. The obtained feedstock can be applied to bioconversion into energy, e.g., bioethanol, biogas, biohydrogen, etc. Here, the current state of lignocellulose pretreatment technologies was comprehensively reviewed, the advances in bioenergy production from pretreated lignocellulose was described, with particular attention to key challenges involved. Several new strategies for overcoming pretreatment barriers to realize highly efficient lignocellulose bioconversion were highlighted. The insights given in this review will facilitate further development on lignocellulosic bioenergy production, towards addressing the global energy crisis and climate change related to the use of fossil fuels.

Comprehensive insights of pretreatment strategies on the structures and bioactivities variation of lignin-carbohydrate complexes

Abstract:Introduction: Due to its unique structural features and bioactivities, the lignin-carbohydrate complex (LCC) displays great potential in vast industrial applications. However, the elucidation of how various pretreatment methods affect the structure and bioactivities remains unaddressed. Method: The three pretreatment methods were systematically studied on the variations of structures and bioactivities, and the Gramineae plant, i.e., wheat straw, was adopted in this study. The structures and bioactivities variation caused by different pretreatments were studied in detail. Result and Discussion: The results showed that compared to physical or chemical pretreatments, biological pretreatment was the most effective approach in improving the bioactivities of LCC. The LCC from biological pretreatment (enzymatic hydrolysis, ELCC4) had more functional groups while the lower weight-average molecular weight (Mw) and polydispersity index (PDI) were well-endowed. The highest antioxidant abilities against ABTS and DPPH of ELCC4 were high up to 95% and 84%, respectively. Furthermore, ELCC4 also showed the best ultraviolet (UV)-blocking rate of 96%, which was increased by 6% and 2% compared to LCC8 (physical pretreatment) and LLCC4 (chemical pretreatment). Synthesis of levulinic acid from tobacco stalk via steam explosion and oxidation–hydrolysis route

Abstract:BACKGROUND Tobacco stalk, the main waste from tobacco planting, was used as raw material to produce levulinic acid, one of the most important platform chemicals. It was reported that steam explosion was an efficient method to treat lignocellulose to hydrolyze the hemicellulose component. Oxidation treatment combined with catalysis of Al2(SO4)3 proved to be efficient for the conversion of cellulose to levulinic acid in our recent report. Here, tobacco stalk was first treated by steam explosion, and then the solid residue was oxidized and converted to levulinic acid in the presence of Al2(SO4)3. RESULTS A xylo‐oligosaccharide yield of 6.6 g 100 g−1 was obtained from tobacco stalk via steam explosion. 15.4 g 100 g−1 (39.6 mol%) yield of levulinic acid was obtained from oxidized tobacco stalk in water at 180 °C in 5 h using Al2(SO4)3 as a catalyst. As high as 20.8 g 100 g−1 (53.5 mol%) yield of levulinic acid can be obtained using NaCl as a co‐catalyst. CONCLUSION Steam explosion was efficient for the depolymerization of the hemicellulose component of tobacco stalk to xylo‐oligosaccharide. A simple salt of Al2(SO4)3 showed good catalytic activity for conversion of oxidized tobacco stalk to levulinic acid. Nicotine in tobacco stalk promoted the formation of lactic acid. Thus, the yield of levulinic acid decreased in the presence of nicotine. NaCl enhanced the catalytic performance of Al2(SO4)3. © 2024 Society of Chemical Industry (SCI).

Abstract:Lignocellulosic materials, such as forest, agriculture, and agroindustrial residues, are among the most important resources for biorefineries to provide fuels, chemicals, and materials in such a way to substitute for, at least in part, the role of petrochemistry in modern society. Most of these sustainable biorefinery products can be produced from plant polysaccharides (glucans, hemicelluloses, starch, and pectic materials) and lignin. In this scenario, cellulosic ethanol has been considered for decades as one of the most promising alternatives to mitigate fossil fuel dependence and carbon dioxide accumulation in the atmosphere. However, a pretreatment method is required to overcome the physical and chemical barriers that exist in the lignin-carbohydrate composite and to render most, if not all, of the plant cell wall components easily available for conversion into valuable products, including the fuel ethanol. Hence, pretreatment is a key step for an economically viable biorefinery. Successful pretreatment method must lead to partial or total separation of the lignocellulosic components, increasing the accessibility of holocellulose to enzymatic hydrolysis with the least inhibitory compounds being released for subsequent steps of enzymatic hydrolysis and fermentation. Each pretreatment technology has a different specificity against both carbohydrates and lignin and may or may not be efficient for different types of biomasses. Furthermore, it is also desirable to develop pretreatment methods with chemicals that are greener and effluent streams that have a lower impact on the environment. This paper provides an overview of the most important pretreatment methods available, including those that are based on the use of green solvents (supercritical fluids and ionic liquids).
A feruloyl esterase/cellulase integrated biological system for high-efficiency and toxic-chemical free isolation of tobacco based cellulose nanofibers

Abstract:Tobacco based cellulose nanofiber (TCNF) is a novel nanocellulose that has recently been used to replace undesirable wood pulp fibers in the preparation of reconstructed tobacco sheets (RTS). However, given the strict requirements for controlling toxic chemical content in tobacco products, there is a global interest in developing a green, efficient, and toxic-chemical free approach to isolate TCNF from tobacco stem as a bioresource. In this study, we propose a creative and environmentally friendly method to efficiently and safely isolate TCNF from tobacco stem pulp, which involves integrated biological pretreatment followed by a facile mechanical defibrillation process. Feruloyl esterase is used to pretreat the stem pulp by disrupting the ether and ester bonds between lignin and polysaccharide carbohydrates within the fiber wall, which effectively facilitates cellulase hydrolysis and swelling of the stem pulp fiber, as well as the following mechanical shearing treatment for TCNF isolation. The results demonstrate that TCNF obtained by the comprehensive feruloyl esterase/cellulase/mechanical process exhibit uniform and well-dispersed nanofiber morphology, higher crystallinity, and stronger mechanical properties than those of the control. The addition of 0.5 % TCNF can replace wood pulp by 18 wt% ~ 25 wt% in the production of RTS samples while maintaining their reasonable strength properties.
