Abstract:This paper discussed the preparation methods and material properties of silk fibroin fiber porous materials,regenerative silk fibroin sponge porous materials and silk fibroin non-woven nanofiber porous materials.It is believed that these materials have appropriate porous structure and excellent biocompatibility,a variety of cells from different apparatus or tissue cultured on these materials and showed attachment and proliferation.Application states of silk fibroin porous materials used in tissue engineering of artificial skin,bone,cartilage,artificial ligament and muscle tendon were summarized as a kind of nice tissue engineering scaffold material.

Abstract:Electrospinning is a very effective method to fabricate nanofibers. This technique has been achieved more attention due to its cheap cost, simple device and obtaining nanofibers similar to extracellular matrix.Silk fibroin(SF) is a natural protein with good biodegradability and low immunogenicity, Therefore, it can be widely applied in tissue engineering. In this paper, we summarize the research of electrospun silk fibrion nanofibers in regenerative medicine. Furthermore, the future research work of SF nanofibers was suggested.

Abstract:Silk fibroin (SF) is a structural protein derived from natural silkworm silks. Materials fabricated based on SF usually inherit extraordinary physical and biological properties, including high mechanical strength, toughness, optical transparency, tailorable biodegradability, and biocompatibility. Therefore, SF has attracted interest in the development of sustainable biodevices, especially for emergent bio-electronic technologies. To expand the function of current silk devices, the SF characteristic sequence has been used to synthesize recombinant silk proteins that benefit from SF and other functional peptides, such as stimuli-responsive elastin peptides. In addition to genetic engineering methods, innovated chemistry modification approaches and improved material processing techniques have also been developed for fabricating advanced silk materials with tailored chemical features and nanostructures. Herein, this review summarizes various methods to synthesize functional silk-based materials from different perspectives. This review also highlights the recent advances in the applications of natural and recombinant silks in tissue regeneration, soft robotics, and biosensors, using B. mori SF and silk-elastin-like proteins (SELPs) as examples.

Abstract:The unique amino acid composition and spatial structure of silk fibroin make it an excellent biomaterial with perfect biocompatibility and biodegradability.Silk fibroin nanofibers and composite fibroin nanofibers prepared via high voltage electrostatic spinning technology are conferred further superior property for application as biomedical materials.This review systematically introduces advances in application and modification of the electrospun silk fibroin nanofibers and composite fibroin nanofibers in bone tissue engineering,in repair of cartilage,wound,nerve and blood vessel,and in controlled release of drugs in recent years,based on which key innovations for future development of electrospun silk fibroin nanofiber materials are briefly discussed.

Abstract:Silk fibroin (SF), which offers the benefits of biosafety, biocompatibility, and mechanical strength, has potential for use as a good biomedical material, especially in the tissue engineering field. This study investigated the use of SF biomaterials as a wound dressing compared to commercially available collagen materials. After human fibroblasts (WI-38) were cultured on both films and sponges, their cell motilities and gene expressions related to wound repair and tissue reconstruction were evaluated. Compared to the collagen film (Col film), the SF film induced higher cell motility; higher expressions of genes were observed on the SF film. Extracellular matrix production-related genes were up-regulated in WI-38 fibroblasts cultured on the SF sponges. These results suggest that SF-based biomaterials can accelerate wound healing and tissue reconstruction. They can be useful biomaterials for functional wound dressings.

Abstract:Regeneration of large bone defects is challenging in the next few years because the age of the population is growing. Bone grafting as a gold standard of bone repairing is limited as a result of many disadvantages. In recent years, many novel materials and processing methods have been introduced to construct the native bone extracellular matrix (ECM) and restore the functions of degenerated bone. Silk fibroin (SF) as a natural polymer, has established a good reputation for bone tissue engineering applications due to its many unique properties, including exceptional biocompatibility, biodegradability, mechanical behavior, and ease of processability. In this review, recent advances in the design and application of SF-based scaffolds for bone regeneration are discussed.

Abstract:Despite advances in the development of silk fibroin (SF)‐based hydrogels, current methods for SF gelation show significant limitations such as lack of reversible crosslinking, use of nonphysiological conditions, and difficulties in controlling gelation time. In the present study, a strategy based on dynamic metal‐ligand coordination chemistry is developed to assemble SF‐based hydrogel under physiological conditions between SF microfibers (mSF) and a polysaccharide binder. The presented SF‐based hydrogel exhibits shear‐thinning and autonomous self‐healing properties, thereby enabling the filling of irregularly shaped tissue defects without gel fragmentation. A biomineralization approach is used to generate calcium phosphate‐coated mSF, which is chelated by bisphosphonate ligands of the binder to form reversible crosslinkages. Robust dually crosslinked (DC) hydrogel is obtained through photopolymerization of acrylamide groups of the binder. DC SF‐based hydrogel supports stem cell proliferation in vitro and accelerates bone regeneration in cranial critical size defects without any additional morphogenes delivered. The developed self‐healing and photopolymerizable SF‐based hydrogel possesses significant potential for bone regeneration application with the advantages of injectability and fit‐to‐shape molding.

Abstract:Regeneration Bombyx mori(B.mori) silk fibroin(SF) nanofibers prepared by electrospinning were widely applied in in the fields of biomaterials and tissue engineering, with the advantages of high specific surface area, high porosity, and good biocompatibility. Pure regenerated SF nanofibers were fragile and poor in mechanical performance, however the composite nanofibers synthesized by blending different inorganic particles or synthetic polymers into SF has much more advantages for it can greatly improve the suitability compared with which of a single material. This paper summarized preparation of SF and inorganic particles or synthetic polymers composite nanofibers in the domestic and at abroad in recent years, which aims to improve the comprehensive performance of SF nanofibers and expand its application.

Abstract:Silk fibroin (SF) is an excellent protein-based biomaterial produced by the degumming and purification of silk from cocoons of the Bombyx mori through alkali or enzymatic treatments. SF exhibits excellent biological properties, such as mechanical properties, biocompatibility, biodegradability, bioabsorbability, low immunogenicity, and tunability, making it a versatile material widely applied in biological fields, particularly in tissue engineering. In tissue engineering, SF is often fabricated into hydrogel form, with the advantages of added materials. SF hydrogels have mostly been studied for their use in tissue regeneration by enhancing cell activity at the tissue defect site or counteracting tissue-damage-related factors. This review focuses on SF hydrogels, firstly summarizing the fabrication and properties of SF and SF hydrogels and then detailing the regenerative effects of SF hydrogels as scaffolds in cartilage, bone, skin, cornea, teeth, and eardrum in recent years.

Abstract:Silk fibroin (SF) has been utilized in various applications, including as, in bone, cartilage, nerve, skin regeneration, owing to its biocompatibility, controllable biodegradability, minimal inflammation, and tunable mechanical properties. Excellent neovascularization of silk matrices is prerequisite for the success of promoting wound healing in different tissues. Neovascularization of SF can provide a nutrient supply to the newly developed granulation tissue at the wound site and accelerate vessel formation and remodeling. In this review, we summarize the neovascularization of SF in different forms and dissolution by recombining different composites, modifying active peptides, and adding growth factors in the treatment of various wound healing processes.

Abstract:The application progress and multifunctional modification strategies of silk fibroin (SF) hydrogel to promote the regeneration of body tissues, and the mechanism of SF to bone and skin regeneration and repair is emphasized. Created with BioRender.com. The creation of functional hydrogels with robust load‐bearing capacity adaptable to complex tissue regeneration remains challenging. Silk fibroin (SF) is a natural biomaterial with excellent mechanical strength and cell adhesion capacity, possessing tremendous potential to solve the aforementioned dilemma. The excellent biodegradability and biocompatibility and the molecular structure with multiple modifiable moieties provide opportunities for the injectability and multifunctionality of hydrogels. Furthermore, the incorporation of other polymers or active ingredients can improve the basic properties, confer biological activities and pharmacological effects, and provide the prerequisites for hydrogels to fulfill specific requirements. Therefore, SF‐based hydrogels are widely applied in tissue repair and regeneration, especially in bone, skin, nerve, liver, myocardium, and cornea, which have effectively addressed the challenges of scaffolds' lack of cell adhesion sites, excessive degradation, as well as the lack of efficient, long‐lasting antimicrobial properties. This review outlines the properties of SF, discusses the types of materials commonly used in the preparation of hydrogels and their characteristics, and describes the current construction methods of SF hydrogels. Simultaneously, recent advancements in applying SF‐based multifunctional hydrogels within biological tissues are explored, with a focus on their role in tissue repair, highlighting the repair mechanism of SF based on bone and skin.

Abstract:The golden rule in tissue engineering is the creation of a synthetic device that simulates the native tissue, thus leading to the proper restoration of its anatomical and functional integrity, avoiding the limitations related to approaches based on autografts and allografts. The emergence of synthetic biocompatible materials has led to the production of innovative scaffolds that, if combined with cells and/or bioactive molecules, can improve tissue regeneration. In the last decade, silk fibroin (SF) has gained attention as a promising biomaterial in regenerative medicine due to its enhanced bio/cytocompatibility, chemical stability, and mechanical properties. Moreover, the possibility to produce advanced medical tools such as films, fibers, hydrogels, 3D porous scaffolds, non-woven scaffolds, particles or composite materials from a raw aqueous solution emphasizes the versatility of SF. Such devices are capable of meeting the most diverse tissue needs; hence, they represent an innovative clinical solution for the treatment of bone/cartilage, the cardiovascular system, neural, skin, and pancreatic tissue regeneration, as well as for many other biomedical applications. The present narrative review encompasses topics such as (i) the most interesting features of SF-based biomaterials, bare SF’s biological nature and structural features, and comprehending the related chemo-physical properties and techniques used to produce the desired formulations of SF; (ii) the different applications of SF-based biomaterials and their related composite structures, discussing their biocompatibility and effectiveness in the medical field. Particularly, applications in regenerative medicine are also analyzed herein to highlight the different therapeutic strategies applied to various body sectors.

Abstract:Silk fibroin is an important natural fibrous protein with excellent prospects for tissue engineering applications. With profound studies in recent years, its potential in tissue repair has been developed. A growing body of literature has investigated various fabricating methods of silk fibroin and their application in tissue repair. The purpose of this paper is to trace the latest developments of SF-based scaffolds for tissue engineering. In this review, we first presented the primary and secondary structures of silk fibroin. The processing methods of SF scaffolds were then summarized. Lastly, we examined the contribution of new studies applying SF as scaffolds in tissue regeneration applications. Overall, this review showed the latest progress in the fabrication and utilization of silk fibroin-based scaffolds.

Abstract:Silk fibroin (SF), a pivotal biomaterial, holds immense promise for diverse applications within the realm of bone tissue engineering. SF is an ideal scaffold material with exceptional biocompatibility, mechanical robustness, biodegradability, and bioactivity. A plethora of investigations have corroborated SF's efficacy in supporting bone tissue repair and regeneration. This comprehensive review delves into the structural attributes, physicochemical characteristics, and extraction methodologies of SF. Moreover, it elucidates the strides taken in harnessing SF across a spectrum of forms, including films, hydrogels, scaffolds, electrospun fibers, and composites for bone tissue engineering applications. Moreover, the application bottleneck of SF as a bone repair material is highlighted, and its development prospects and potential biomedical applications are also presented in this review. We expect that this review can inspire the broad interest of a wide range of readers working in the fields of materials science, tissue engineering, biomaterials, bioengineering, and biomedicine.

Abstract:Tissue engineering (TE) is the approach to combine cells with scaffold materials and appropriate growth factors to regenerate or replace damaged or degenerated tissue or organs. The scaffold material as a template for tissue formation plays the most important role in TE. Among scaffold materials, silk fibroin (SF), a natural protein with outstanding mechanical properties, biodegradability, biocompatibility, and bioresorbability has attracted significant attention for TE applications. SF is commonly dissolved into an aqueous solution and can be easily reconstructed into different material formats, including films, mats, hydrogels, and sponges via various fabrication techniques. These include spin coating, electrospinning, freeze drying, physical, and chemical crosslinking techniques. Furthermore, to facilitate fabrication of more complex SF-based scaffolds with high precision techniques including micro-patterning and bio-printing have recently been explored. This review introduces the physicochemical and mechanical properties of SF and looks into a range of SF-based scaffolds that have been recently developed. The typical TE applications of SF-based scaffolds including bone, cartilage, ligament, tendon, skin, wound healing, and tympanic membrane, will be highlighted and discussed, followed by future prospects and challenges needing to be addressed.

This review introduces the physicochemical and mechanical properties of SF and looks into a range of SF-based scaffolds that have recently been developed. The typical applications of SF-based scaffolds for TE of bone, cartilage, teeth and mandible tissue, cartilage, skeletal muscle, and vascular tissue are highlighted and discussed followed by a discussion of issues to be addressed in future studies.
