Abstract:A series of 3,3-arylidene bis (4-hydroxycoumarins) 2 were synthesized by the reaction of aromatic aldehydes with 4-hydroxycoumarin using dodecylbenzenesulfonic acid as Brønsted acid-surfactant catalyst in aqueous media and under microwave irradiation. The present method is operationally simple and the use of water as the reaction medium makes the process environmentally benign. The epoxydicoumarins 5 were then obtained with a good yield by heating 3,3'-arylidenebis-4-hydroxycoumarins 2 in acetic anhydride. Techniques such as elemental analysis, 1H, 13C-1H NMR, and infrared spectroscopy were employed to characterize these compounds. The synthesized compounds displayed good antibacterial potential against Escherichia coli (ATCC 25988), Pseudomonas aeruginosa (ATCC 27853), Klebsilla pneumonia (ATCC 700603), Staphylococcus aureus (ATCC 29213), methicillin-resistant Staphylococcus aureus (ATCC 43300) and Candida albicans (ATCC 14053). The MIC values of 23 mg/mL for compound 5e against Escherichia coli (ATCC 25988) and 17 mg/mL for 2a were observed. Furthemore, a molecular docking simulation has been performed to evaluate the antibacterial activities and the probable binding modes of the studied compounds 2a-f and 5a-g toward the active sites of a series of well known antibacterial targets. Among the investigated compounds, the binding modes and docking scores demonstrate that 2a has the most antibacterial and antifungal activities. Additionally, DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS has been tested for their ability to scavenge hydrogen peroxide and free radicals. According to our results, these compounds exhibit excellent radical scavenging properties. Furthermore, compounds 2-5 were evaluated for anti-inflammatory activity by indirect haemolytic and lipoxygenase inhibition assays and revealed good activity.

Abstract:A series of novel α-terpineol derivatives were designed and synthesized through structural derivatization of the tertiary hydroxyl moiety or reduction of the double bond. Of the resulting compounds, eight compounds enhanced relaxation of airway smooth muscle (ASM) compared to the α-terpineol precursor, and four compounds (4a, 4d, 4e, and 4i)were superior or comparable to aminophylline at a concentration of 0.75 mmol/L. Assays for 3'-5'-Cyclic adenosine monophpsphate (cAMP) activation revealed that some representative α-terpineol derivatives in this series were capable of upregulating the level of cAMP in ASM cells. Further in vivo investigation using the asthmatic rat model, illustrated that treatment with the compounds 4a and 4e resulted in significantly lowered lung resistance (RL) and enhanced dynamic lung compliance (Cldyn), two important parameters for lung fuction. Moreover, treatment with 4e downregulated the levels of both IL-4 and IL-17. Due to its several favorable physiological functions, including ASM relaxation activity, cAMP activation capability, and in vivo anti-asthmatic efficacy, 4e is a promising remedy for bronchial asthma, meriting extensive development.

Abstract:Effective therapeutic agents are highly desired for immune-mediated allergic diseases. Herein, we report the design, synthesis, and structure-activity relationship of an o-aminopyridinyl alkyne series as novel orally bioavailable antiallergic agents, which was identified through phenotypic screening. Compound optimization yielded a highly potent compound 36, which effectively suppressed mast cell degranulation in a dose-dependent manner (IC50, 2.54 nM for RBL-2H3 cells; 48.28 nM for peritoneal mast cells (PMCs)) with a good therapeutic index. It also regulated the activation of FcεRI-mediated downstream signaling proteins in IgE/Ag-stimulated RBL-2H3 cells. In addition, 36 exhibited excellent in vivo pharmacokinetic properties and antiallergic efficacy in both passive systemic anaphylaxis (PSA) and house dust mite (HDM)-induced murine models of pulmonary allergic inflammation. Furthermore, preliminary analysis of the kinases profile identified Src-family kinases as potential targets for 36. Compound 36 may serve as a new valuable lead compound for future antiallergic drug discovery.

Abstract:A new class of 2,5-disubstituted-dioxacycloalkanes were designed and synthesized via stereoselective synthetic method as cancer chemoprevention agents. The anti-inflammatory activities of these compounds were tested using the xylene-induced mouse ear edema model. Some of these compounds exhibited comparable or better anti-inflammatory activities than that of aspirin suggesting that they can be further developed as potential anti-inflammatory drug lead compounds. In addition, treatment of these anti-inflammatory agents did not prolong tail bleeding time in mice. The structure/activity relationships were also analyzed among these compounds.

Abstract:Pseudo-allergic reactions frequently occur following clinical drug use and sometimes even cause mortal danger. Mas-related G-protein-coupled receptor member X2 (MRGPRX2) is a novel receptor that mediates pseudo-allergy and is an important target in the treatment of allergies. However, to date, there are no synthetic small-molecule inhibitors that prevent anaphylactoid reactions through this pathway. Our preliminary research suggested that B10-S and mubritinib effectively inhibited LAD2 cells. Therefore, two novel derivatives were synthesized by integrating the active substructures of B10-S and mubritinib, according to the molecular docking results. The antiallergic inhibitory effects of the two compounds were preliminarily evaluated in vitro using β-hexosaminidase release, histamine release, and intracellular Ca2+ mobilization assays, and their binding sites on MRGPRX2 were analyzed by molecular docking. Both substances inhibited β-hexosaminidase and histamine release in LAD2 cells and decreased intracellular Ca2+ by inhibiting MRGPRX2 in MRGPRX2-HEK293 cells treated with C48/80 in a dose-dependent manner. The docking results suggested that the molecules could competitively bind to the active site on MRGPRX2 and Glu141, which were combined by C48/80. Our study indicated that the two compounds have potential anti-allergic properties, which may provide evidence that will facilitate the development of synthetic molecules with anti-pseudo-allergic activity for clinical use in the future.

Abstract:New pyranocoumarin and coumarin-sulfonamide derivatives were prepared and evaluated for their antioxidant, antimicrobial, and/or anti-inflammatory activities. Coumarin-sulfonamide compounds 8a–d demonstrated significant antioxidant activity, while 7c,d, 8c,d, and 9c,d exhibited antimicrobial activity equal to or higher than the standard antimicrobials against at least one tested microorganism. Regarding the anti-inflammatory testing, pyranocoumarins 2b, 3a,b and 5c and coumarin-sulfonamide compound 9a showed more potent antiproteinase activity than aspirin in vitro; however, five compounds were as potent as aspirin. The anti-inflammatory activity of the promising compounds was further assessed pharmacologically on formaldehyde-induced rat paw oedema and showed significant inhibition of oedema. For in vitro COX-inhibitory activity of coumarin derivatives, pyranocoumarin derivative 5a was the most selective (SI = 152) and coumarin-sulfonamide derivative 8d was most active toward COX-2 isozyme. The most active derivatives met the in silico criteria for orally active drugs; thus, they may serve as promising candidates to develop more potent and highly efficient antioxidant, antimicrobial, and/or anti-inflammatory agents.

Hydrotrope assisted green synthesis of dicoumarols and in silico and in vitro antibacterial, antioxidant and xanthine oxidase inhibition studies

Abstract:Aqueous hydrotrope has been employed for the first time to synthesize heteroaryl dicoumarols by condensation of 4-hydroxycoumarin and different heterocyclic aldehydes. This method is highly efficient and green, and the same aqueous hydrotropic solution can be used up to five times without any considerable loss of yield in the product. The synthesized compounds showed good antibacterial potential against Gram-positive (Staphylococcus aureus/NTCC 0997 and B. oceanisediminis) and Gram-negative (Escherichia coli/D0157:H7 and E. coli rosetta) bacterial strains using the Resazurin microtiter plate visual method. The MIC value of 312 µg/ml for compounds 3b, 3k and 3l for S. aureus while 39 µg/ml for compounds 3a, 3b and 3k for E. coli and 625 µg/ml for 3a and 3b for B. oceanisediminis was observed. The compounds were screened via computational methods like molecular docking studies and molecular dynamic simulations with PDB Id's 2W9S and 2EX6. Antioxidant activity was assessed using DPPH and H2O2 assays. Five compounds with the best binding score in molecular docking with XO (PDB ID: 1FIQ) have been tested in an in-vitro study using an enzyme inhibition assay. Novel compound 3b gave the IC50 value of 0.28 µg/ml, comparable to the standard drug Allopurinol.Communicated by Ramaswamy H. Sarma.

Abstract:In this study, several novel coumarin derivatives, 7-hydroxy-2-oxo-2H-chromene-3-carboxyl-Trp-Trp-AA-OBzl compounds, were designed and synthesized as potential anticancer agents. Their in vitro cytotoxic activities were evaluated using methylthiazoltetrazolium (MTT) assay. The anti-tumor activity of the newly coumarin derivatives was determined in a S180 bearing mouse model and some of the compounds demonstrated tumor growth inhibition similar to the positive control, doxorubicin. Compared to doxorubicin, most of the compounds exhibited enhanced immunologic function suggesting a relatively minor toxic effect. The intercalation of the coumarin derivatives synthesized with calf thymus (CT) DNA was also studied.

Abstract:MRGPRX2 antagonists possess the potential for the treatment of allergic rhinitis, atopic dermatitis, and chronic urticaria. Previously, we identified a class of diaryl urea (DPU) MRGPRX2 antagonists with sub-micromolar IC50 values in vitro. However, the structure–activity relationship remains unclear. Herein, we adopted a “relative symmetry with electronegativity of different key-groups” strategy for further modification of DPUs to achieve a promising MRGPRX2 antagonist with higher activity and safety. Electrostatic potential energy analysis and biological evaluation revealed that B-1023 and B-5023, that possess relatively symmetric electron-withdrawing substituents, remarkable inhibited mast cell degranulation at a sub-micromolar IC50 in vitro and alleviated anaphylactic symptoms. Furthermore, B-1023, mitigated antigen-induced pulmonary inflammation (AIPI) in mice and competitively bonded to MRGPRX2. In summary, the “relative symmetry with electronegativity of different key-groups” strategy provided a drug design pattern for MRGPRX2 antagonists and identified promising antiallergic precursors for AIPI treatment.

Abstract:Thirty-two novel DG F-spiroacetal ring-opening derivatives, including 24 acetylated derivatives and 8 nitrogenous derivatives, were designed and synthesized from diosgenin (DG). The cytotoxicity of the novel derivatives was evaluated by MTT assay, except for compounds 4a, 4e, 4i, 4 l, 5a and 5 h, which were potentially cytotoxic to RAW264.7 cells, all the other derivatives had no significant cytotoxicity. The NO release inhibitory activities of novel derivatives were screened by Griess method. The results showed that the anti-inflammatory activity of the DG acetylated derivatives was stronger than the nitrogenous derivatives, and 4a-4 m containing acetyl groups at the 3-position may have better anti-inflammatory effects than 5a-5 k containing free hydroxyl groups. In ELISA assay, compound 4 m exhibited potent anti-inflammatory activity by inhibiting the production of NO in RAW264.7 cells activated by LPS with IC50 values 0.449 ± 0.050 μM. The results of docking experiments showed that 4 m has a good affinity for p65 protein.

Abstract:Nonsteroidal anti-inflammatory durgs(NSAIDs) are among the most widely prescribed medications with their uses limited by significant toxicity.The studies of the cyclooxygenase(COX) show that the side effects of classical NSAIDs come from the inhibition of COX-1,while the therapeutic effects come from the inhibition of COX-2.Selective COX-2 inhibitors were developed as NSAIDs which display the profiles of good potent activities and low side effects. In order to search for the new NSAIDs,using the marketed selective COX-2 inhibitor celecoxib as a leading compound and based on the result of molecular modeling study of COX-2 inhibitors,a series of novel diaryl-substituted-1,2,4-trizole derivatives were designed and synthesized.All the target compounds were confirmed by IR,MS,~1H NMR spectra and elemental analysis.The results of preliminary pharmacological tests in vivo show that some compounds have certain anti-inflammatory effect.
Synthesis and pharmacological activities of 4-ary1-3,4-dihydrocoumarin derivatives

Abstract:4-Aryl-3,4-dihydrocoumarins(neoflavones) represent a minor class of natural compounds which based on the C6—C3—C6 skeleton with an characteristic aryl group in the 4 position.Structurally,4-aryl-3,4-dihydrocoumarins can also be regarded as the hybrids of isoflavones and coumarins,thus being expected to possess corresponding bioactivity.In this paper,ten 4-aryl-3,4-dihydrocoumarins were synthesized by two steps.The first condensation of substituted benzaldehydes and malonic acids in the presence of pyridine and piperidine at 70-90 ℃ gave the expected cinnamic acids in good yields.Then condensation of substituted cinnamic acids with appropriate phenols in the presence of BF3-Et2O and POCl3 gave the target molecule at room temperature.Their structures were confirmed by EI-MS,IR and 1H-NMR.The antioxidant and antitumor activity of these compounds were evaluated by DPPH and MTT assay,respectively.Some of 4-aryl-3,4-dihydrocoumarin derivatives showed strong scavenging activities against 1,1-diphenyl-2-picrylhydrazyl radical:(±)-7,8-dihydroxy-4-(3-hydroxy-4-methoxyphenyl)-3,4-dihydrocoumarin(3c) EC50=(2.69±0.09) μmol · L-1,(±)-7,8-dihydroxy-4-(4-methoxyphenyl)-3,4-dihydrocoumarin(3g) EC50=(2.32±0.10) μmol · L-1 and weak antitumor in vitro activity against BGC-823 cell line[(±)-7,8-dihydroxy-4-(3-hydroxy-4-methoxyphenyl)-3,4-dihydrocoumarin(3c) IC50=(136±21) μmol · L-1].The results showed that the 4-arylcoumarins which possessing the catechol moiety may be a beneficial scaffold for therapeutic purpose.

Abstract:A new series of 5-glycylamino-2-substituted-phenyl-1,3-dioxacycloalkanes were designed and synthesized. The anti-inflammatory activities of these compounds were tested using the xylene-induced mouse ear edema model. Sixteen of these new compounds exhibited comparable or better anti-inflammatory activities than aspirin suggesting that they can be further developed as potential anti-inflammatory drug leads. In addition, treatment with these anti-inflammatory agents did not prolong tail bleeding time in mice. The structure/activity relationships were also analyzed among these compounds. Considering their good efficacy and safety profiles, some 5-glycylamino-2-substituted-phenyl-1,3-dioxacycloalkanes are worthy to be explored further in assessing the possible link between anti-inflammation and cancer prevention.
An Effective, Green Synthesis Procedure for Obtaining Coumarin-Hydroxybenzohydrazide Derivatives and Assessment of Their Antioxidant Activity and Redox Status

Abstract:In this study, green synthesis of two derivatives of coumarin-hydroxybenzohydrazide, (E)-2,4-dioxo-3-(1-(2-(2,3,4-trihydroxybenzoyl)hydrazyl)ethylidene)-chroman-7-yl acetate (C-HB1), and (E)-2,4-dioxo-3-(1-(2-(3,4,5-trihydroxybenzoyl)hydrazyl)ethylidene)chroman-7-yl acetate (C-HB2) is reported. Using vinegar and ethanol as a catalyst and solvent, the reactions were carried out between 3-acetyl-4-hydroxy-coumarin acetate and corresponding trihydroxybenzoyl hydrazide. The antioxidant potential of these compounds was investigated using the DPPH and ABTS assays, as well as the FRAP test. The obtained results reveal that even at very low concentrations, these compounds show excellent radical scavenging potential. The IC50 values for C-HB1 and C-HB2 in relation to the DPPH radical are 6.4 and 2.5 μM, respectively, while they are 4.5 and 2.0 μM in relation to the ABTS radical. These compounds have antioxidant activity that is comparable to well-known antioxidants such as gallic acid, NDGA, and trolox. These results are in good correlation with theoretical parameters describing these reactions. Moreover, it was found that inhibition of DPPH● follows HAT, while inactivation of ABTS+● follows SET-PT and HAT mechanisms. Additionally, coumarin-hydroxybenzohydrazide derivatives induced moderate cytotoxic activity and show significant potential to modulate redox status in HCT-116 colorectal cancer cells. The cytotoxicity was achieved via their prooxidative activity and ability to induce oxidative stress in cancer cells by increasing O2˙- concentrations, indicated by increased MDA and GSH levels. Thus, ROS manipulation can be a potential target for cancer therapies by coumarins, as cancer cells possess an altered redox balance in comparison to normal cells. According to the ADMET analysis, the compounds investigated show good pharmacokinetic and toxicological profiles similar to vitamin C and gallic acid, which makes them good candidates for application in various fields of industry and medicine.

Abstract:A new series of optically pure 5-amino-2-substitutedphenyl-1,3-dioxacycloalkanes were designed and synthesized via a chemo-enzymatic combined method to develop new chemoprevention agents. Twenty-four of newly synthesized compounds significantly inhibited xylene-induced rat ear edema and exhibited comparable or better anti-inflammatory activities than the reference drug aspirin. Treatment of these anti-inflammatory agents did not prolong the tail bleeding time in rat. In addition, 5-amino-2-substitutedphenyl-1,3-dioxacycloalkanes exhibited good membrane permeability based on in vitro Caco-2 cell monolayer permeability assay. Furthermore, some preliminary structure-activity relationships were further analyzed among these compounds. Taken together, 5-amino-2-substitutedphenyl-1,3-dioxacycloalkanes may represent a new class of anti-inflammatory drugs with safer pharmacological profile.

Abstract:Licorice (Glycyrrhiza uralensis Fisch), a significant traditional Chinese herbal medicine, has been extensively utilized in China to treat various ailments. Natural bioactive coumarins, glycycoumarin, glycyrin, and 3-O-methylglycyrol, were isolated from licorice, and they exhibited various pharmacological properties. In this report, we have accomplished the total synthesis of glycycoumarin, glycyrin, and 3-O-methylglycyrol in 5–7 linear steps from commercially available 2,4,6-trihydroxybenzaldehyde with yields of 12.3–21.2%. Additionally, their anti-inflammatory activities were studied and compared. Glycycoumarin, glycyrin, and 3-O-methylglycyrol exhibited different levels of anti-inflammatory activities, with glycyrin being the most potent. Mechanistic studies indicated that glycyrin exerted its anti-inflammatory properties by inhibiting the activation of TNF-α, IL-6, and IL-1β, making it a potential anti-inflammatory lead compound for further optimization and discovery of new agents.

Abstract:Eucryphin, a natural product widely found in plants, has a variety of biological activities. In this work, a series of eucryphin analogs were designed, synthesized, characterized and the anti-inflammatory activity in vivo was evaluated comparing with eucryphin and its 7- O -glycosyl derivatives to study the structure- activity relationship. Eucryphin exhibited remarkable anti-inflammatory activity and minor side effects in 2,4-dinitrofluorobenzene (DNFB) induced mice auricle edema model. Glycosyl substitution at the position of C-3 was essential for the anti-inflammatory activity. Methyl substitution at C-2 and glycosylation at C-7 of eucryphin reduce anti-inflammatory activity.

Abstract:A class of 5-trifluoroacetylamino-1,3-dioxacycloalkanes, 5-benzoylamino-1,3-dioxacycloalkanes, and 5-amino-1,3-dioxacycloalkane compounds were stereoselectively synthesized as potential anti-inflammatory drug candidates. The anti-inflammatory activities of these compounds were tested using the xylene-induced mouse ear edema model, from which multiple compounds possessing anti-inflammatory properties which surpass aspirin were identified; these compounds were then compared to establish structure–activity relationships.
