Abstract:Resistant starch (RS) is widely used as a new type of dietary fiber, but it is still a limitation of controllable formation of microstructure and polymeric architecture for preparing single-component RS (pure-phase). In this study, RS was prepared using maize starch with two ratios of amylose to amylopectin (RAA1:1 and RAA1:2) and four pullulanase concentrations (5, 15, 25, 35 U/g) in a high-solid dynamic reaction environment of bioextrusion. Based on the long-and short-range orders reflected by the glycosidic bonds (C-13 NMR), chain length distributions (SEC/FACE), single-/double-helices (FTIR), lamellas (SAXS) and crystalline pattern (XRD), the rearranged supramolecular structure of starch and their relationships with physicochemical properties (DSC and RVA) was illuminated. There were two morphologies of RS were created: 1) disordered fusion-like structure displayed by RAA1:1-P15 and RAA1:2-P35, which had the high contents of RS (54.95 and 54.19%) and short amylopectin (DP <= 24) (70.52% and 60.20%). The short (A chains) and extremely short (DP < 6) amylopectin could contribute to form submicro-crystallite with B1 chains in the extruded matrix at a low moisture level; 2) thinner, scalier and layered-like structure displayed by RAA1:2-P15 that was the transition state from thick layered strips of large granules. Its SDS content was high (33.61%) and owned the larger rearranged dLorentz (16.97 nm). Overall, the entangled regime of amylose and amylopectin in the modifications of starch with different structural units or micro-domains in this study will provide a new idea and reference to explore the preparation of novel RS.

Abstract:Debranching enzymes contribute to the enzymatic production of resistant starch (RS) by reducing substrate molecular weight and increasing amylose yield. In the present study, the action pattern of a thermostable isoamylase-type debranching enzyme on different types of starch was investigated. The molecular weight distribution, glycosidic bond composition and contents of oligosaccharides released were monitored by various liquid chromatography techniques and nuclear magnetic resonance spectroscopy (NMR). These analyses showed that the isoamylase could specifically and efficiently attack a1,6-glucosidic linkages at branch points, leaving the amylose favored by other amylolytic enzymes. Its ability to attack side chains composed of 1-3 glucose residues differentiates it from other isoamylases, a property which is also ideal for the RS preparation process. The enzyme was used as an auxiliary enzyme in the hydrolytic stage. The highest RS yield (53.8%) was achieved under the optimized conditions of 70 C and pH 5.0, using 7 U isoamylase per g starch and 2 NU amylase per g starch. These data also help us better understand the application of isoamylase for preparation of other products from highly branched starch materials. (C) 2017 Elsevier Ltd. All rights reserved.

Abstract:Resistant starch (RS) is a non-digestible carbohydrate that has numerous biological benefits. However, the amounts of RS are limited in most native cereal starches, thus developing RS-fortified starch sources and food products is necessary. This review summarizes the effectiveness of RS improvement by chemical, enzymatic, and physical modification approaches in starches from various origins. The functions of RS in regulating the metabolisms and the influences of RS on the nutritional and textural properties of various foods are also elucidated. After modification, the molecular structures are altered and the starch granules became more resistant to enzymatic digestion. However, the comparison of the RS contents between raw and cooked starch is absent in many studies. In addition, RS plays an essential role in attenuating postprandial glycemia, inhibiting glucose and lipid synthesis and promoting glucose tolerance, insulin sensitivity, and lipid degradation, which are controlled by the enriched diversity and abundance of the beneficial microbiota in the colon, together with more short-chain fatty acids. The incorporation of RS in food products significantly increases nutritional value without unacceptable quality loss, which provides a new direction for manufacturing functional foods.

Abstract:Physicochemical modification methods have been widely applied to increase resistant starch (RS) content of waxy starch, but their effects on the functional properties of non-waxy starches are not fully understood. In this study, five non-waxy rice starches with intermediate to very high apparent amylose contents (AAC) were modified by pullulanase (PUL), heat-moisture treatment (HMT) and citric acid (CA). RS contents, AAC, crystalline structures, pasting properties and thermal properties were analyzed. The three modification treatments were effective in increasing the RS contents, especially CA modification. PUL modified starches had relatively higher AAC and relative crystallinity (RC), and their crystalline patterns changed from A-type to mixed B + V type. Both HMT and CA increased gelatinization temperature significantly but had opposite effects on changing paste viscosity of different rice starches. Compared with other non-waxy rice varieties, high-amylose rice starch exhibited the highest RS, AAC, and RC after modification. This study provides practical information for improving the nutritional benefits of rice.

Abstract:The molecular structure of amylopectin and its ϕ,β-limit dextrins from starch of 13 amaranth cultivars was determined by HPAEC-PAD after debranching. Chain length profiles of amylopectins showed bimodal distributions. The molar-based ratios of the average chain lengths of amylopectins (CLap) ranged from 17.41 to 18.22. The molar-based average chain lengths (CLld) and average B-chain lengths (BCLld) of ϕ,β-limit dextrins varied from 7.68 to 8.05, and from 14.10 to 14.73, respectively. Correlation analysis indicated that most structural parameters were positively correlated with thermal properties with few exceptions, whereas the content of fraction fa′ (“′” stands for molar-based chain length ratio) was negatively correlated with the thermal properties. Pasting properties of cold paste viscosity (CPV) and setback were also correlated with amylopectin structural parameters.

Abstract:Resistant starch (RS) plays a key role in providing metabolic and colonic health benefits. In particular, RS type III (RS3) is of great interest because of its thermal stability and its preserved nutritional functionality. RS3 can be prepared by physical treatment, including high hydrostatic pressure, ultrasound, extrusion, autoclaving, microwave cooking, and heat-moisture treatment. The acid and enzymatic hydrolysis can also be applied to facilitate the generation of small molecules, which increases the inherent crystallinity of RS3 upon retrogradation. Depending on processing conditions, RS3 with diversified structural characteristics can be formed. These structures play a key role in determining the physiological behavior of RS3. Therefore, a deep understanding of the structural rearrangement pattern during different processing treatment is of great importance for regulating the molecular structure of RS3 and thereby its corresponding physiological properties. This review thus focuses on the past and current status of research into the in-depth study of RS3 formation mechanism and the changes to RS3 structural characteristics under different processing conditions. The objective was to provide a theoretical guidance for the rational selection of preparation methods for RS3 and for designing RS3 structures with specific physiological functionalities for relevant industrial applications.

An Innovative Approach for Significantly Increasing Enzyme Resistant Starch Type 3 Content in High Amylose Starches by Using Extrusion Cooking

Abstract:In this study effects of extrusion cooking on enzyme resistant starch (RS) formation in high amylose corn starches (Hylon V and VII) and the functional properties of RS preparations were investigated. Native starches were extruded at 50, 60, 70% feed moisture contents, at constant screw speed (100 rpm) and barrel temperature (140 degrees C). Among these samples, the highest RS contents were observed at 60% feed moisture. Therefore, feed moisture in the second and third extrusion cycles was set at 60%. There were significant increases in RS contents of both Hylon V and Hylon VII after the second extrusion cycle (p < 0.05). After the third extrusion, the RS levels reached to 40.0 and 45.1% for Hylon V and Hylon VII, respectively. Substantial loss of birefringence in these samples indicated that the increases in RS were mainly due to RS3 formation. The RS samples produced by extrusion did not have high emulsion capacity, but the ones produced from Hylon VII had high emulsion stability. Although, decreases in L* and increases in b* values of extruded samples were significant as compared to respective native starches, the changes were not substantial. Characterisation of Starch During Germination and Seedling Development of a Rice Mutant with a High Content of Resistant Starch

Abstract:To acquire a better understanding of whether RS influences the dynamics of in vivo starch digestion and seed vigour, the high-RS rice mutant RS4 (RS ca. 10%) and the wild type R7954 were used to investigate total amylase activity seedling vigour, starch content and starch granule structure during germination. RS4 exhibited similar seed vigour to R7954. Amylose and amylopectin in R7954 showed synchronous degradation throughout the whole process, while amylopectin was hydrolysed significantly faster than amylose in RS4 during the earlier germination stages. The starch residues of RS4 after germination (GD) lost endotherm peaks and showed a special X-ray diffraction pattern with only two peaks at around 16.90 degrees and 21.62 degrees, probably due to remnants of amylopectin and its tight crosslinking with the cell wall. The remaining starch after 10 GD, primarily amylopectin may make a critical contribution to total resistant starch content. These results indicated that RS had-no negative impact on seed vigour in rice lines, although RS cannot be hydrolysed by alpha-amylase from human and animal in vitro. By appropriately increasing the special amylopectin fraction, a new breeding programme of high RS crops and improvement in the eating quality of high RS rice varieties might be achieved. Abstract:Resistant starch (RS) is a special kind of starch with beneficial effects on obesity, type 2 diabetes and other chronic complications. Breeding high-RS rice varieties is considered a valuable way to improve public health. However, most rice cultivars only contain an RS level lower than 2% in cooked rice, and cloning of RS genes is critical to improve RS levels in rice. The loss of function of Starch Synthases IIIa (SSIIIa) and SSIIIb, two amylopectin biosynthetic genes, could elevate RS levels up to 10%. Here, we performed a systematic genetic study of 14 amylopectin biosynthetic genes in the ssIIIa ssIIIb double mutant via genome editing, and investigated their effects on RS formation, the eating quality and grain yield. The results showed that deficiency in SSIIa, SSIVb or ISA2 under the ssIIIa ssIIIb background could each elevate RS content to above 14%, and the quadruple mutants of sbeI sbeIIb ssIIIa ssIIIb and sbeI ssIVb ssIIIa ssIIIb could further increase RS levels to over 18%. Furthermore, the eating quality of cooked rice and grain yield decreased along with the elevated RS contents, showing a trade-off among these traits. In these mutants, ssIIIa ssIIIb showed the balanced performance of RS and grain yield. This study provides insights into RS biosynthesis with a series of RS genes in the amylopectin biosynthesis pathway and practical strategy to breed high-RS rice varieties with balanced performance.
Improving the resistance to enzyme digestion of rice debranched starch via narrowing chain length distribution combined with oven drying

Abstract:In this study, the effects of chain length distribution and drying method on the structural properties and in‐vitro digestibility of resistant starch (RS) were investigated. Rice starch was debranched at 10% w/w solid concentrations, incubated at 60°C, and further subjected to freeze drying and oven drying treatment to obtained debranched starch (DBS). The chain length distribution showed that DBS (The degree of polymerization (DP) = 19.52±0.13) had a high proportion of B1 and B2 chains (DP13‐36) and a narrow chain length distribution. In addition, the results of scanning electron microscopy (SEM) showed that the oven‐dried samples had denser structures and larger particle morphology compared to samples prepared by freeze‐drying. In FTIR, LCM‐Raman and X‐ray diffraction patterns, debranched starch supernatant fraction 1 (DBSS1) exhibited higher short‐range molecular order and crystallinity than other samples. Digestibility analysis showed that DBSS1 with narrow chain length distribution (DP13‐36) promoted the high content of RS formation and oven drying was more favorable for RS formation than freeze drying (RSDBSFS1 = 79.69±5.09% and RSDBSOS1 = 84.17±0.17%). This suggested that the proper narrow chain length distribution was conducive to the formation of ordered structure and an increase in resistance. Abstract:Starch digestibility and starch structure of a high-resistant starch (RS) rice (RS111) was compared to that of a wild rice type (R7954). RS111 exhibited high RS content and incomplete starch hydrolysis in both cooked rice and retrograded rice flour. High IRS rice RS111 had a higher lambda(max) of absorbance and blue value of iodine-binding starch complex and contained a higher percentage of intermediate chains of amylopectin than R/954. X-ray diffraction pattern of IRS isolated from cooked high IRS rice displayed a mixture of B- and V-type that was more resistant to starch hydrolysis by alpha-amylase. Resistant starch in RS111 is composed mainly of linear amylose and some low-molecular-weight amylopectin.

Abstract:RS2 and RS3 are two of the main resistant starch types in food, and the primary types found in raw and cooked rice respectively. To elucidate the physiochemical determinants for RS2 and RS3 content in rice, this study investigated the fine amylopectin structure, starch granules morphologies and starch pasting properties of nine different rice accessions. The results revealed that not all the accessions showed a decrease in resistant starch after cooking and cooling. Granular size, paste properties, fb1 and fb3 showed a significant negative correlation with RS3, whereas fa showed a significant positive correlation with RS3. RS2 had no significant correlation with granular size, but did have a significant positive correlation with HPV, CPV and fb2. Different rice types could be distinguished by the fine amylopectin structure of DP6-12 and DP13-24 chains. Taken together fa, fb2 and HPV and CPV values may have the potential to be used as indices for distinguishing or characterising rice with different RS2 and RS3 contents.
Human cytomegalovirus-mediated enhancement of human immunodeficiency virus type-1 production in monocyte-derived macrophages.

Abstract:Monocyte-derived macrophages were co-infected with human immunodeficiency virus (HIV), strain HIVBa-L, and human cytomegalovirus (HCMV) strain AD 169. HCMV enhanced the production of p24 antigen and infectious HIV, but HIV had no effect on HCMV production. In dually infected cultures HIV p24 antigen levels were increased 5- to 15-fold during the first 3 weeks of culture. This enhancement was observed both for cell-associated and extracellular p24 antigen from co-infected cells. The replication of macrophage-tropic clinical isolates of HIV were also enhanced by HCMV, but HCMV did not enable T cell-tropic viruses to replicate in macrophages. Conversely, HCMV clinical isolates from blood or urine were able to upregulate HIV production. Similar enhancement of HIV p24 antigen levels was observed with noninfectious, heat-inactivated HCMV, indicating that replication of HCMV in the macrophages was not necessary. Although replication was not required, the enhancing ability of HCMV was associated with the virus. Enhancement of HIV segregated with the HCMV pelleted virus particles and not the supernatant in stock virus. The fact that HCMV and HIV are detected in macrophages in persons with HIV infection highlights the importance of this system for studying interactions between HIV and HCMV in persons with HIV infection.
Structural Features and Digestibility of Corn Starch With Different Amylose Content

Abstract:In this study, the in vitro digestibility of corn starch with different amylose content was determined. The results showed that waxy corn starch (WCS) and corn starch (CS) have the highest digestibility, while high amylose corn starch (HACS) has a higher content of resistant starch (RS). Besides being related to amylose content, RS content is also closely related to particle shape, debranched fine structure, molecular structure, and semi-crystalline structure. HACS can maintain a complete particle structure after gelatinization and enzymolysis; differential scanning calorimetry showed a positive correlation between gelatinization enthalpy and RS content. As the amylose content increased, the content of fa (DP 6–12) decreased, while the content of fb2 (DP 25–36) and fb3 (DP ≥ 37 ) increased, which in-turn decreased the cluster polymer formed by short branch chains, and the formation of more hydrogen bonds between long chain branches improved starch stability. D, which characterizes the compactness of starch semi-crystalline structure, increased with the increase of RS content. HACS 60 with the highest RS content had a unique surface fractal structure between 7.41 < d (2π /q ) < 10.58 nm, indicating that the dense structure is effective in maintaining the RS content.

Abstract:The content of resistant starch (RS) was considered positively correlated with the apparent amylose content (AAC). Here, we analyzed two Indica rice mutants, RS111 and Zhedagaozhi 1B, similar in high AAC and found that their RS content differed remarkably. RS111 had higher RS3 content but lower RS2 content than Zhedagaozhi 1B; correspondingly, cooked RS111 showed slower digestibility. RS111 had smaller irregular and oval starch granules when compared with Zhedagaozhi 1B and the wild type. Zhedagaozhi 1B showed a B-type starch pattern, different from RS111 and the wild type, which showed A-type starch. Meantime, RS111 had more fa and fb1 but less fb3 than Zhedagaozhi 1B. Both mutants showed decreased viscosity and swelling power when compared with the parents. RS111 had the lowest viscosity, and Zhedagaozhi 1B had the smallest swelling power. The different fine structures of amylopectin between RS111 and Zhedagaozhi 1B led to different starch types, gelatinization properties, paste viscosity, and digestibility. In addition to enhancing amylose content, modifications on amylopectin structure showed great potent in breeding rice with different RS2 and RS3 content, which could meet the increasing needs for various rice germplasms.

Abstract:Starch properties and structure of a wheat mutant (WRS-1) high in resistant starch (RS) was compared to that of the wild type (Sumai No.6). In cooked hot flour and retrograded flour, the amounts of RS in mutant WRS-1 were 7.8 and 9.4 times of wild type (0.5 in hot flour and 0.6 in retrograded flour), respectively, and correspondingly the slower and incomplete starch hydrolysis was also in both types of samples. WRS-1 had a higher λmax of absorbance, blue value (BV), apparent amylose content (AAC), and contained the increased intermediate and long chains with 18≤ degree of polymerization (DP) and decreased short chains with DP≤17. Two types of starch granules, large A in lenticular shape and small B in spherical shape, were observed in both mutant and wild types. However, the starch granules in WRS-1 were relatively larger and some were irregularly-shaped. High content of total starch (TS), decreased contents of lipid (LC) and protein (PC), lower onset temperature (To), peak temperature (Tp), final temperature (Tc), and the enthalpy of gelatinization (ΔHgel) were observed in WRS-1.

Abstract:High-amylose cereal starch has a great benefit on human health through its resistant starch (RS) content. Enzyme hydrolysis of native starch is very helpful in understanding the structure of starch granules and utilizing them. In this paper, native starch granules were isolated from a transgenic rice line (TRS) enriched with amylose and RS and hydrolyzed by α-amylase. Structural properties of hydrolyzed TRS starches were studied by X-ray powder diffraction, Fourier transform infrared, and differential scanning calorimetry. The A-type polymorph of TRS C-type starch was hydrolyzed faster than the B-type polymorph, but the crystallinity did not significantly change during enzyme hydrolysis. The degree of order in the external region of starch granule increased with increasing enzyme hydrolysis time. The amylose content decreased at first and then went back up during enzyme hydrolysis. The hydrolyzed starches exhibited increased onset and peak gelatinization temperatures and decreased gelatinization enthalpy on hydrolysis. These results suggested that the B-type polymorph and high amylose that formed the double helices and amylose-lipid complex increased the resistance to BAA hydrolysis. Furthermore, the spectrum results of RS from TRS native starch digested by pancreatic α-amylase and amyloglucosidase also supported the above conclusion.

Abstract:This study focused on constructing a high-solid reaction system to prepare type 3 resistant starch (RS3) with high-amylose maize starch as raw material by mechanical activation (MA) pretreatment combined with thermal and freeze-thaw treatments. MA pretreatment effectively destroyed the crystal structure and molecular structure of native starch. MA damaged starch with a certain viscosity could form dough with a small amount of water to construct a starch continuous phase system. RS content increased with the damage levels of starch as the formation of double helix structure, attributed to that the molecules of MA damaged starch could be easy to move and form recrystallization structure. Thermal and freeze-thaw treatments contributed to strong interaction of starch-water and the re-formation of internal crystal structure of MA damaged starch to form RS3. This study provides insight into the development of a highly effective approach for large scale production of resistant starch.

Abstract:Changes in the structural, morphological, and thermal properties of four types of resistant starch (RS), including native (RS2), retrograded (RS3), chemically modified with octenyl succinic anhydride (RS4), and complexed with stearic acid (RS5), were investigated and compared in salivary, salivary-gastric, and salivary-gastric-intestinal digestion. In the process of digestion, the crystal types of RS2, RS3, RS4, and RS5 remained unchanged, maintaining a B-type structure, except for the missing V-type structure of RS5. The total short-chain fatty acid yield of RS2 was significantly higher than that of RS3, RS4, and RS5 at 6 and 12 h of fermentation. RS2 and RS4 had a better proliferation effect on Bifidobacterium in late fermentation than RS3 and RS5, while RS3 could promote proliferation to a larger extent in early fermentation. RS2, RS3, and RS5 had more obvious proliferation effects on Lactobacillus in the late fermentation period. This work suggests that the type of RS plays an important role in influencing the structural and digestive properties of RS during gastrointestinal digestion and can be used to develop formulations of healthy starch-based products with desired functionalities.
