Abstract:The contamination of certain food crop with aflatoxin poses a real public health problem for consumers and causes many market losses for exporters. Thus, several research works are oriented in the direction of developing methods to combat aflatoxinogenic fungus. This study aimed at identifying and characterizing strains of Aspergillus flavus and A. parasiticus on groundnut, maize and rice seeds grown or imported into Senegal. Four species (A. niger, A. tamarii, A. flavus and A. parasiticus) were isolated from the seeds with incidences ranging from 0 to 100% depending on the samples and their provenance. Six strains of A. flavus and 3 strains of A. parasiticus have been identified and characterized on CYA and G25N culture media. The characterization focused on the morphological characteristics (color and appearance, mycelial growth) of the colonies on the different culture media, and on some microscopic characteristics such as the density of sporulation (production of conidia) and the appearance of the conidiophore. Strains of A. flavus were more frequent on groundnuts and isolates with the same traits were also identified from rice and maize samples, hence the 3 strains of A. parasiticus were isolated.

Abstract:Fungi are one of the main food spoilage agents. Numerous species when subjected to stress conditions produce secondary metabolites known as mycotoxins, which are mutagenic and carcinogenic substances. The fungus Aspergillus flavus is one of the main contaminants of grains and is known to produce Aflatoxin. Pesticides are used in agriculture to contain fungi and other pests, but they harm other species, the environment and the human health, in addition to the development of resistance to these substances in pest species. Natural alternatives have been sought to control these organisms. In this context, essential oils are a viable option against A. flavus. The aim of this study was to identify the main components and evaluate the effectiveness of lemongrass essential oil (Cymbopogon flexuosus) for controlling the fungus Aspergillus flavus. Initially, the effect of essential oil on mycelial growth of the fungus was assessed by in vitro tests at the doses: 0.05; 0.1; 0.2; 0.4; 0.6; 0.8; 1.6; 3.2; 6.4; and 12.8 μL mL-1. The minimum inhibitory concentration (MIC) was 0.8 μL mL-1. The in vivo test was performed at the following concentrations: 0.6; 0.8; and 1.6 μL mL-1. The results showed that the essential oil has fungicidal potential against A. flavus. The main component of the essential oil was citral.

Abstract:The antifungal and antimycotoxigenic activities of the essential oils (EO) from Cuminum cyminum and Laurus nobilis, and their respective principal compounds, cuminaldehyde and 1,8-cineole, were evaluated against fungi of the genus Aspergillus: A. carbonarius, A. niger, A. ochraceus, and A. westerdijkiae. The antifungal activity was determined by the contact method and the mycelial growth of the fungi was evaluated. Scanning electron microscopic (SEM) images were obtained to suggest modes of action of the compounds analysed. The antimycotoxigenic activity was determined by high-performance liquid chromatograph. Aspergillus carbonarius was completely inhibited by cumin EO (500 µl l-1), by laurel EO and by cuminaldehyde (5000 µl l-1). The cumin EO (500 µl l-1) completely inhibited the growth of A. niger. All the samples inhibited the mycelial growth of A. ochraceus, especially cumin EO and cuminaldehyde (250 µl l-1). Aspergillus westerdijkiae was completely inhibited by cumin EO and cuminaldehyde (1000 µl l-1), by laurel EO and 1,8-cineole (10 000 µl l-1). A decrease in the production of ochratoxin A (OTA) was observed post-treatment, except in A. ochraceus, only inhibited by laurel EO. SEM images showed morphological changes in fungal structures and spore inhibition post-treatment. The results confirmed the antifungal and antimycotoxigenic effect of EO and their principal constituents on fungi evaluated.
Antifungal Activity of Menthol, Eugenol and Their Combination against Aspergillus ochraceus and Aspergillus niger In Vitro and in Stored Cereals

Abstract:Aspergillus ochraceus and Aspergillus niger are spoilage and mycotoxin-producing fungi that can contaminate agricultural commodities and derived products. In the present study, menthol, eugenol, and their combination (mix 1:1) were tested to determine their contact and fumigation toxicity against the two fungi. Menthol, eugenol, and their mixture significantly reduced mycelial growth and spore germination at concentrations from 300 to 600 µg/mL, and the inhibitory effects showed clear dose dependence. The minimum inhibitory concentration (MIC) values against A. ochraceus were 500 µg/mL (menthol), 400 µg/mL (eugenol), and 300 µg/mL (mix 1:1), while the MIC values for A. niger were 500 µg/mL (menthol), 600 µg/mL (eugenol), and 400 µg/mL (mix 1:1). Additionally, the analyzed compounds exhibited more than 50% protection against A. ochraceus and A. niger by fumigation of stored cereal grains (maize, barley, and rice) in sealed containers. The binary mixture of menthol and eugenol showed synergistic effects against both fungi in both in vitro direct contact and stored grain fumigation trials. The results of the present study provide a scientific basis for the application of a combination of natural antifungals in food preservation.
Combinational inhibitory action of essential oils and gamma irradiation for controlling Aspergillus flavus and Aspergillus parasiticus growth and their aflatoxins biosynthesis in vitro and in situ conditions

Abstract:The purpose of this study was to investigate the effects of certain essential oils (star anise, lemon leaves, marjoram, fennel, and lavender) on the fungal growth of Aspergillus flavus and Aspergillus parasiticus and their production of aflatoxin B1 (AFB1). The degree of suppression of the aflatoxigenic strains' growth and their production of AFB1 is mainly affected by the kind and the concentration of the tested essential oils (EOs). Star anise essential oil had the lowest minimum inhibitory concentration (0.5 and 1.0 μL/mL) against A. flavus and A. parasiticus, respectively, so it was the best among the five different oils. The study of liquid chromatography with tandem mass spectrometry revealed that star anise EO resulted in a 98% reduction in AFB1 without a breakdown of AFB1 products after treatment thus the complete removal of AFB1 was done without any toxic residues. The combination showed a synergistic effect, the combinational treatment between γ-irradiation at a low dose (2 kGy) and star anise EO at concentrate 0.5 μL/g destroyed A. flavus and A. parasiticus inoculated (individually) in sorghum and peanut, respectively throughout the storage period (8 weeks).

Abstract:The antifungal and antiaflatoxigenic effects of four distinct plant species against Aspergillus flavus and Aspergillus parasiticus were investigated. Essential oils and methanolic extracts were prepared from aerial parts of Lippia javanica , Ocimum gratissimum , Satureja punctata , and stem barks of Toddalia asiatica by hydro-distillation and maceration, respectively. The poisoned food method was used to confirm the antifungal activity of essential oils and methanolic extracts from four different plant species against Aspergillus flavus and Aspergillus parasiticus , and high-performance liquid chromatography was used to quantify the antiaflatoxigenic activity. The essential oils of Satureja punctata and Lippia javanica showed the highest antiaflatoxigenic activity against the fungi strains tested at concentrations of 1.25, 2.5, and 5 μL/mL, followed by Ocimum gratissimum essential oil while Toddalia asiatica essential oil exerted moderate antiaflatoxigenic activity. Meanwhile, the methanolic extracts showed a wide spectrum of low to high antifungal and antiaflatoxigenic activities at concentrations of 125, 250, and 500 μg/mL against A. flavus and A. parasiticus . This study has indicated that the essential oils of Satureja punctate , Lippia javanica , and Ocimum gratissimum had substantial antifungal and antiaflatoxigenic activities compared to their methanolic extracts, while Toddalia asiatica methanolic extract had a moderate antifungal activity compared to its essential oil.

Abstract:The contamination of food with Aspergillus flavus has caused serious economic losses and posed a great threat to human and animal health. Cumin, a condiment and flavoring in many dishes, was showed to have great potential to prevent A. flavus contamination in the present study. By using cumin essential oil (CEO), the occurrence of A. flavus contamination on peanuts was prevented during the storage period even up to 9 months. We further identified cuminaldehyde as the predominant component in CEO with great inhibitory activity to both the growth of A. flavus and aflatoxin B1 (AFB1) biosynthesis. The data showed that the minimum inhibitory concentration (MIC) of cuminaldehyde to conidial growth was 0.64 μL/mL, and the mycelia exposed to 0.80 μL/mL cuminaldehyde reduced AFB1 production by more than 99% in comparison to the control. RNA-Seq profiles showed that some genes, involving the synthesis pathway of ergosterol and AFB1, were significantly down-regulated in the transcription by cuminaldehyde. Further investigation confirmed a significantly reduced cellular ergosterol content in the presence of cuminaldehyde, and the qRT-RCR results were also in agreement with that by RNA-Seq, indicating down-regulated expressions of those genes involving ergosterol and AFB1 synthesis. Taken together, our study strongly supported cuminaldehyde as a potential agent for the prevention of A. flavus contamination.

Abstract:Aspergillus flavus is a facultative parasite that contaminates several important food crops at both the pre- and post-harvest stages. Moreover, it is an opportunistic animal and human pathogen that causes aspergillosis diseases. A. flavus also produces the polyketide-derived carcinogenic and mutagenic secondary metabolite aflatoxin, which negatively impacts global food security and threatens human and livestock health. Recently, plant-derived natural compounds and essential oils (EOs) have shown great potential in combatting A. flavus spoilage and aflatoxin contamination. In this review, the in situ antifungal and antiaflatoxigenic properties of EOs are discussed. The mechanisms through which EOs affect A. flavus growth and aflatoxin biosynthesis are then reviewed. Indeed, several involve physical, chemical, or biochemical changes to the cell wall, cell membrane, mitochondria, and related metabolic enzymes and genes. Finally, the future perspectives towards the application of plant-derived natural compounds and EOs in food protection and novel antifungal agent development are discussed. The present review highlights the great potential of plant-derived natural compounds and EOs to protect agricultural commodities and food items from A. flavus spoilage and aflatoxin contamination, along with reducing the threat of aspergillosis diseases.
Potential fungicidal and antiaflatoxigenic effects of cinnamon essential oils on Aspergillus flavus inhabiting the stored wheat grains

Abstract:Wheat is one of the essential crops for the human and animal nutrition, however, contamination with aflatoxigenic fungi, due to the improper storage conditions and high humidity, was the main global threats. So, preventing the growth of aflatoxigenic fungi in stored wheat grains, by using different essential oils was the main objective of this work. Aspergillus flavus EFBL-MU12 PP087400, EFBL-MU23 PP087401 and EFBL-MU36 PP087403 isolates were the most potent aflatoxins producers inhabiting wheat grains. The effect of storage conditions of wheat grains "humidity, temperature, incubation period, and pH" on growth of A. flavus , was assessed by the response surface methodology using Plackett-Burman design and FCCD. The highest yield of aflatoxins EFBL-MU12 B 1 and B 2 by A. flavus grown on wheat grains were 145.3 and 7.6 μg/kg, respectively, at incubation temperature 35°C, 16% moisture contents, initial pH 5.0, and incubated for 14 days. The tested oils had a powerful antifungal activity for the growth and aflatoxins production by A. flavus in a concentration-dependent manner. Among these oils, cinnamon oil had the highest fungicidal activity for A. flavus at 0.125%, with about 85-90 % reduction to the aflatoxins B 1 and B 2 , conidial pigmentation and chitin contents on wheat grains. From the SEM analysis, cinnamon oils had the most deleterious effect on A. flavus with morphological aberrations to the conidial heads, vegetative mycelia, alteration in conidiophores identity, hyphae shrank, and winding. To emphasize the effect of the essential oils on the aflatoxins producing potency of A. flavus, the molecular expression of the aflatoxins biosynthetic genes was estimated by RT-qPCR. The molecular expression of nor-1, afLR , pKsA and afLJ genes was suppressed by 94-96%, due to cinnamon oil at 0.062% compared to the control. Conclusively, from the results, cinnamon oils followed by the peppermint oils displayed the most fungicidal activity for the growth and aflatoxins production by A. flavus grown on wheat grains.

Abstract:AIMS:To study the antifungal activities of a prepared food-grade dilution-stable microemulsion against Aspergillus niger.METHODS AND RESULTS:Results from the antifungal activity on solid medium by agar dilution method showed that the microemulsion caused complete growth inhibition at 2000 ppm, and at 1000 ppm, showed 55% growth inhibition after 4 days of incubation and a delay of conidiation by 1 day compared with controls. Results from the antifungal activity in liquid medium by broth dilution method showed that the growth of A. niger was completely inhibited when a liquid medium containing 10(6) spores per ml was treated with 500 ppm of microemulsion, which was determined by minimum fungicidal concentration. Study of fungicidal kinetics showed that more than 99% of viable spores were killed within 15 min. These antifungal activities were confirmed by scanning electron microscopy, light microscopy and increased Ca(+2), K(+) and Mg(+2) leakages.CONCLUSIONS:The results suggest that the prepared microemulsions are effective antifungal systems with excellent growth inhibition and sporicidal activities, and indicate that their antifungal activity may be to the result of the disruption and dysfunction of A. niger cell walls and biological membranes.SIGNIFICANCE AND IMPACT OF THE STUDY:This study suggests the potential use of food-grade dilution-stable microemulsions for antifungal use in the food and pharmaceutical industries.
Inhibitory mechanism and application of cinnamon essential oil against Aspergillus flavus

Abstract:Aspergillus flavus produces highly toxic and carcinogenic aflatoxins that cause severe economic losses and pose a serious threat to public health. Cinnamon essential oils (CEO) have recently attracted attention as promising alternative antifungal agents. In this study, the inhibition mechanism of CEO on A . flavus was investigated, and its application in the prevention of A. flavus in coix seeds was explored for the first time. The results showed that mycelial growth, spore germination, and aflatoxin synthesis of A. flavus were completely inhibited after treatment with 0.5 μL/mL of CEO. Morphological, physiological, and biochemical changes after CEO treatment were also analyzed to elucidate the inhibitory mechanism. Transcriptomic analysis showed that some genes involved in conidiophore development ( flbA and abaA ), ergosterol synthesis ( CYP51 , Erg1 , Erg3 , Erg7 , Erg9 , Erg24 , and Erg25 ), aflatoxin biosynthesis ( aflR , aflO , aflT , aflJ , aflW , and aflY ), and oxidative stress ( Pbs2 , Ste20 , CAT , atfB , mtfA , PacC , ppoC , and gprM ) were significantly regulated. Moreover, A. flavus infection in coix seeds was effectively inhibited by 20.0 μL/g of CEO. This study shed important light on the processes underlying the antifungal mechanism of CEO and affirmed the possibility of using CEO as a green anti-aflatoxigenic agent in the preservation of coix seed.
Antifungal compounds from Bacillus subtilis B-FS06 inhibiting the growth of Aspergillus flavus

Abstract:The cell-free culture filtrate (CCF) was prepared from a culture of an Aspergillus flavus antagonist, Bacillus subtilis B-FS06. The CCF inhibited the growth and spore germination of A. flavus at a series of concentrations (10, 25, 50%) (v/v). It still retained the activity after treatment at pH values ranging from 2 to 12 for 24 h or at 100 °C for 30 min. The antifungal activity, however, was reduced by 30% after treatment at 121 °C for 20 min. After purification by anion exchange chromatography, gel filtration chromatography and HPLC, the active compounds revealed six ion peaks: [M–H] m/z = 1006.78, 1020.71, 1034.74, 1049.54, 1056.78, and 1071.64 by using electrospray ionization mass spectrometry (ESI-MS) analysis. In the presence of the active compounds at 200 μg/g, the growth of A. flavus on peanuts was completely inhibited.

Abstract:The major objective of this study was to evaluate the inhibitory effect of essential oil (EO) from Zanthoxylum armatum DC. on Aspergillus flavus . The chemical composition of the EO was identified by gas chromatography–mass spectrometer. The minimum inhibitory concentration (MIC) of EO was investigated by liquid fermentation. The morphology, colony number, and aflatoxin content of A. flavus in platycladi semen were investigated by stereomicroscopy, scanning electron microscopy, plate counting, and high-performance liquid chromatography. The results indicated that the MIC of EO was 0.8 μL⋅mL –1 , and the main components were β-phellandrene (7.53%), D -limonene (13.24%), linalool (41.73%), terpinen-4-ol (5.33%), and trans -nerolidol (6.30%). After the EO fumigated the platycladi semen, the growth of A. flavus slowed, and the mycelium shrank considerably. The number of colonies after EO treatment at room temperature and cold storage was significantly reduced, the inhibition effect was better under cold storage, and the aflatoxin B1 content did not exceed the standard within 100 days. Therefore, this study demonstrated the good potential of A. flavus growth inhibition during the storage of platycladi semen.

Abstract:As a result of increased emergence of bacterial resistance to the conventionally used antibiotics, it is necessary to search for alternatives from other sources like plant materials. This study was aimed at evaluating the in vitro antibacterial activity of Citrullus lanatus seed extract against isolates of Klebsiella pneumoniae from urine and Staphylococcus aureus from wound. The seeds were extracted using distilled water and methanol via cold maceration and then analysed for the presence of phytochemicals using standard methods. Tannins, steroids, cardiac glycosides and carbohydrates were present in both extracts. Only the aqueous extract contained alkaloids. The antibacterial activity of the two extracts was determined by agar well diffusion method. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined using broth dilution method. The aqueous extract had better activity than the methanol extract, having inhibition zones of 20 mm and 23 mm at 200 mg/mL and, 10 mm and 11 mm at 25 mg/mL against Klebsiella pneumoniae isolates respectively, and mean zone of inhibition between 27.50 mm to 29.33 mm at 200 mg/mL and, 16.50 mm to 17.33 mm at 25 mg/mL against Staphylococcus aureus isolates with MIC and MBC values of 0.195 mg/mL and 0.39 mg/mL against Klebsiella pneumoniae and, mean MIC and MBC values ranging from 0.30 mg/mL to 0.39 mg/mL and 0.78 mg/mL to 2.73 mg/mL against Staphylococcus aureus isolates. The results obtained reveal the potential use of Citrullus lanatus seeds in the development of standard antibiotics.
Synergistic effects of combined cinnamaldehyde and nonanal vapors against Aspergillus flavus

Abstract:This study evaluated the synergistic antifungal effects of vapor-phase natural agents against Aspergillus flavus with an aim to prevent fungal contamination in agricultural products. Screening different combinations of natural antifungal vapor agents using the checkerboard assay revealed that the cinnamaldehyde and nonanal (S CAN ) blend could exert the strongest synergistic antifungal activities against A. flavus , with a minimum inhibitory concentration (MIC) of 0.03 μL/mL, which caused a 76 % decrease in fungal population compared to when each agent was used separately. Subsequent gas chromatography–mass spectrometry (GC/MS) analysis demonstrated that the cinnamaldehyde/nonanal combination was stable and no effects on their individual molecular structures. S CAN at 2 × MIC completely inhibited the fungal conidia production and mycelial growth. The calcofluor white (CFW) and dichloro-dihydro-fluorescein diacetate (DCFH-DA) staining assays showed that S CAN treatment could accelerate the destruction of cell wall integrity and accumulation of reactive oxygen species (ROS) in A. flavus . Moreover, pathogenicity assay indicated that in contrast to separate treatment with cinnamaldehyde or nonanal, S CAN could cause a decrease in the production of A. flavus asexual spores and AFB1 on peanuts, which verified its potential synergistic activity against fungal propagation. In addition, S CAN effectively preserves the organoleptic and nutritional properties of stored peanuts. Overall, our findings strongly indicated that the cinnamaldehyde/nonanal combination is a potentially significant antifungal agent against A. flavus contamination during the postharvest storage of peanuts.

Antifungal Activity of Essential Oil Compounds (Geraniol and Citral) and Inhibitory Mechanisms on Grain Pathogens (Aspergillus flavus and Aspergillus ochraceus)

Abstract:The grain contamination by Aspergillus spp. has been a serious issue. This study exhibited the excellent antifungal effects of the essential oil compounds (EOCs) geraniol and citral against common grain pathogens (A. flavus and A. ochraceus) in vitro and in situ. The inhibitory mechanisms were also evaluated from the perspective of cell membrane permeability, reactive oxygen species (ROS) generation, and Aspergillus spp. growth-related gene expression. Meanwhile, the combined effects of EOCs in the vapor phase and modified atmosphere packaging (MAP) were examined to find an alternative preservation method for controlling Aspergillus spp. The results indicated that citral exhibited the antifungal activity mainly by downregulating the sporulation- and growth-related genes for both pathogens. Geraniol displayed inhibitory effectiveness against A. flavus predominantly by inducing the intracellular ROS accumulation and showed toxicity against A. ochraceus principally by changing cell membrane permeability. Furthermore, the synthetic effects of EOCs and MAP (75% CO₂ and 25% N₂) induced better grain quality than the current commercial fumigant AlP. These findings reveal that EOCs have potential to be a novel grain preservative for further application.
Inhibition of Essential Oils on Growth of Aspergillus flavus and Aflatoxin B1 Production in Broth and Poultry Feed

Abstract:Aflatoxin B1 (AFB1), a common contaminant in food and feed during storage, does great harm to human and animal health. Five essential oils (thymol, carvacrol, cinnamaldehyde, eugenol, and citral) were tested for their inhibition effect against Aspergillus flavus (A. flavus) in broth and feed. Cinnamaldehyde and citral were proven to be most effective against A. flavus compared to others and have a synergistic effect when used simultaneously. The broth supplemented with cinnamaldehyde and citral was inoculated with A. flavus (106 CFU/mL) by using the checkerboard method, and mold counts and AFB1 production were tested on days 0, 1, 3, and 5. Similarly, 100 g poultry feed supplemented with the mixture of cinnamaldehyde and citral at the ratio 1:1 was also inoculated with A. flavus, and the same parameters were tested on days 0, 7, 14, and 21. In poultry feed, cinnamaldehyde and citral significantly reduced mold counts and AFB1 concentrations (p < 0.05). Results showed that cinnamaldehyde and citral have a positive synergy effect and could both inhibit at least 90% the fungal growth and aflatoxin B1 production at 40 μg/mL in broth and poultry feed, and could be an alternative to control aflatoxin contamination in food and feed in future.

Transcriptomic and Metabolomic Analyses of the Response of Resistant Peanut Seeds to Aspergillus flavus Infection

Abstract:Peanut seeds are susceptible to Aspergillus flavus infection, which has a severe impact on the peanut industry and human health. However, the molecular mechanism underlying this defense remains poorly understood. The aim of this study was to analyze the changes in differentially expressed genes (DEGs) and differential metabolites during A. flavus infection between Zhonghua 6 and Yuanza 9102 by transcriptomic and metabolomic analysis. A total of 5768 DEGs were detected in the transcriptomic study. Further functional analysis showed that some DEGs were significantly enriched in pectinase catabolism, hydrogen peroxide decomposition and cell wall tissues of resistant varieties at the early stage of infection, while these genes were differentially enriched in the middle and late stages of infection in the nonresponsive variety Yuanza 9102. Some DEGs, such as those encoding transcription factors, disease course-related proteins, peroxidase (POD), chitinase and phenylalanine ammonialyase (PAL), were highly expressed in the infection stage. Metabolomic analysis yielded 349 differential metabolites. Resveratrol, cinnamic acid, coumaric acid, ferulic acid in phenylalanine metabolism and 13S-HPODE in the linolenic acid metabolism pathway play major and active roles in peanut resistance to A. flavus. Combined analysis of the differential metabolites and DEGs showed that they were mainly enriched in phenylpropane metabolism and the linolenic acid metabolism pathway. Transcriptomic and metabolomic analyses further confirmed that peanuts infected with A. flavus activates various defense mechanisms, and the response to A. flavus is more rapid in resistant materials. These results can be used to further elucidate the molecular mechanism of peanut resistance to A. flavus infection and provide directions for early detection of infection and for breeding peanut varieties resistant to aflatoxin contamination.

Abstract:Plant extracts may represent an ecofriendly alternative to chemical fungicides to limit aflatoxin B1 (AFB1) contamination of foods and feeds. Mate (Ilex paraguariensis), rosemary (Romarinus officinalis) and green tea (Camellia sinensis) are well known for their beneficial properties, which are mainly related to their richness in bioactive phenolic compounds. AFB1 production is inhibited, with varying efficiency, by acetone/water extracts from these three plants. At 0.45 μg dry matter (DM)/mL of culture medium, mate and green tea extracts were able to completely inhibit AFB1 production in Aspergillus flavus, and rosemary extract completely blocked AFB1 biosynthesis at 3.6 μg DM/mL of culture medium. The anti-AFB1 capacity of the extracts correlated strongly with their phenolic content, but, surprisingly, no such correlation was evident with their antioxidative ability, which is consistent with the ineffectiveness of these extracts against fungal catalase activity. Anti-AFB1 activity correlated more strongly with the radical scavenging capacity of the extracts. This is consistent with the modulation of SOD induced by mate and green tea in Aspergillus flavus. Finally, rutin, a phenolic compound present in the three plants tested in this work, was shown to inhibit AFB1 synthesis and may be responsible for the anti-mycotoxin effect reported herein.

Abstract:During an experiment where we were cultivating aflatoxigenic Aspergillus flavus on peanuts, we accidentally discovered that a bacterium adhering to the peanut strongly inhibited aflatoxin (AF) production by A. flavus. The bacterium, isolated and identified as Klebsiella aerogenes, was found to produce an AF production inhibitor. Cyclo(l-Ala-Gly), isolated from the bacterial culture supernatant, was the main active component. The aflatoxin production-inhibitory activity of cyclo(l-Ala-Gly) has not been reported. Cyclo(l-Ala-Gly) inhibited AF production in A. flavus without affecting its fungal growth in a liquid medium with stronger potency than cyclo(l-Ala-l-Pro). Cyclo(l-Ala-Gly) has the strongest AF production-inhibitory activity among known AF production-inhibitory diketopiperazines. Related compounds in which the methyl moiety in cyclo(l-Ala-Gly) is replaced by ethyl, propyl, or isopropyl have shown much stronger activity than cyclo(l-Ala-Gly). Cyclo(l-Ala-Gly) did not inhibit recombinant glutathione-S-transferase (GST) in A. flavus, unlike (l-Ala-l-Pro), which showed that the inhibition of GST was not responsible for the AF production-inhibition of cyclo(l-Ala-Gly). When A. flavus was cultured on peanuts dipped for a short period of time in a dilution series bacterial culture broth, AF production in the peanuts was strongly inhibited, even at a 1 × 104-fold dilution. This strong inhibitory activity suggests that the bacterium is a candidate for an effective biocontrol agent for AF control.
