Extracellular HSP90a promotes pulmonary fibrosis by impairing autophagic flux in lung fibroblasts

Abstract:Activation of lung fibroblasts,which is the main source of extracellular matrix,plays a pivotal role in pulmonary fibrosis. Our previous study indicated that extracellular HSP90α is involved in the myofibroblast differentiation in pulmonary fibrosis. However extracellular HSP90α whether could modulate autophagy in pulmonary fibrosis remains unclearly. This study aimed to elucidate the extracellular HSP90α induced pulmonary fibrosis through regulating autophagy in lung fibroblasts. In vivo, we investigated the autophagy markers (LC3,P62 and Beclin1 ) as well as fibrosis-related markers through western blot and immunohistochemistry. In vitro, lung fibroblasts were treated with different concentrations of human recombinant HSP90α to examine the expression levels of α-SMA, collagen I and autophagy markers. What’s more, lung fibroblasts were transfected with siRNA to knock down P62 and Beclin1 and detected by western blot.We found that extracellular HSP90α could increase the expression of LC3, P62 and Beclin1. Consistently, 1G6-D7 could significantly decrease LC3,P62 and Beclin1 expression in vitro and in vivo. Furthermore, inhibiting P62 and Beclin1 by siRNA effectively reduced the expression of fibrosis-related markers induced by extracellular HSP90α.Taken together, this study indicated that extracellular HSP90α mediated the autophagic flux dysfunction in the lung fibroblasts through P62 and Beclin1. Our research highlights that extracellular HSP90α plays an important role in the development of pulmonary fibrosis, providing a new perspective for the diagnosis and treatment of pulmonary fibrosis.
