Abstract:A combination of chemotherapy and phototherapy has emerged as a promising strategy for cancer treatment. To achieve effective combinational therapy of cancer with reduced toxicity, it is highly desirable to improve the targeting of chemotherapeutic and near-infrared photosensitizers to enhance their accumulation in tumor. Here we report a novel tumor targeting cell membrane capsule (CMC), originate from living cells, to load doxorubicin hydrochloride (DOX) and indocyanine green (ICG), for combinational photo-chemotherapy against cancer. As a result, folic acid modified CMC (CMC-FA, with a diameter about 200 nm and a FA density of 0.4 molecule/nm(2)) showed 3-4 fold higher cell uptake by cancer cells in vitro and 2.3 times higher accumulation in mouse cancer xenografts in vivo than pristine CMC. DOX and ICG with therapeutically significant concentrations can be sequentially encapsulated into CMC-FA by temporary permeating the plasma membranes with high efficiency. The systematic administration of cancer targeting CMC-FA loaded with DOX and ICG could significantly inhibit tumor growth in mouse xenografts in the presence of a near-infrared light at 808 nm, without noticeable toxicity. These findings suggest that cancer targeting CMC may have considerable benefits in drug delivery and combinational cancer therapy. Statement of Significance A combination of chemotherapy and photothermal/photodynamic therapy has emerged as a promising strategy for cancer therapy. One-pot Synthesis of Hollow PDA@DOX Nanoparticles for Ultrasound Imaging and Chemo-Thermal Therapy in Breast Cancer

Abstract:Constructing nanocarriers with high drug loading capacity is a challenge, which limits the effective delivery of drugs to solid tumors. Here, we reported a one-pot synthesis of hollow nanoparticles (NPs) encapsulated by doxorubicin (DOX) and modified with polydopamine (PDA) to form PDA@DOX NPs for breast cancer treatment. PDA@DOX NPs demonstrated exceptionally high capacity (53.16%) for loading DOX. In addition, when PDA@DOX NPs were administered systemically, they exhibited responsive aggregation in the tumor sites and demonstrated a good controlled release effect for DOX due to the weak acidic environment of the tumor sites and targeting near-infrared (NIR) light irradiation. The PDA outer layer absorbed the near-infrared (NIR) light and facilitated simultaneous generation of heat energy for destroying the tumor cells to release the drug upon NIR irradiation. Moreover, this NIR-activated combined/synergistic therapy exhibited remarkably complete tumor growth suppression in a breast cancer mouse model. Importantly, NPs exhibited a good ultrasound performance both in vitro and in vivo, which could monitor the treatment process. In conclusion, this NIR-activated PDA@DOX NP system is demonstrated as a good US-guided combination (chemotherapy + PTT) therapy platform with high loading capacity and controlled drug release characteristics, which is promising for the treatment of breast cancer.

Abstract:Ultrasonic activation of nanoparticles provides the opportunity to deliver a large fraction of the injected dose to insonified tumors and produce a complete local response. Here, we evaluate whether the local and systemic response to chemotherapy can be enhanced by combining such a therapy with locally-administered CpG as an immune adjuvant. In order to create stable, activatable particles, a complex between copper and doxorubicin (CuDox) was created within temperature-sensitive liposomes. Whereas insonation of the CuDox liposomes alone has been shown to produce a complete response in murine breast cancer after 8 treatments of 6 mg/kg delivered over 4 weeks, combining this treatment with CpG resolved local cancers within 3 treatments delivered over 7 days. Further, contralateral tumors regressed as a result of the combined treatment, and survival was extended in systemic disease. In both the treated and contralateral tumor site, the combined treatment increased leukocytes and CD4+ and CD8+ T-effector cells and reduced myeloid-derived suppressor cells (MDSCs). Taken together, the results suggest that this combinatorial treatment significantly enhances the systemic efficacy of locally-activated nanotherapy.
Carboxymethyl-hexanoyl Chitosan Nanodroplets for Ultrasonic Imaging and Drug Delivery to Tumor

Abstract:Introduction: Although a great many strategies have been proposed for tumor-targeted chemotherapy, current delivery methods of anticancer drugs present limited success with inevitable systemic toxicity. The aim of this study was to develop a new kind of theranostic carrier for targeted tumor therapy. Methods: Prior to prepare CHC-PFP-DOX, carboxymethyl-hexanoyl chitosan (CHC) was synthesized by acylation of carboxymethyl chitosan. To develop CHC-PFP, perfluoropentane (PFP), an ultrasound gas precursor, was simultaneously encapsulated into the hydrophobic inner cores of pre-formulated CHC micelle in aqueous phase via using the oil in water (O/W) emulsion method. The size distribution and surface charges of these nanodroplets were measured and the morphology was observed by transmission electron microscopy (TEM). For ultrasound imaging application, in vitro model was established to evaluate the imaging of CHC-PFP-DOX under different concentration and mechanical index. After that, the anti-tumor effect of ultrasound combined with CHC-PFPDOX on ovarian cancer cells was investigated. Results: The resulting CHC-PFP-DOX had a nano-sized particle structure, with hydrophobic anticancer DOX/PFP inner cores and a hydrophilic carboxymethyl chitosan polymer outer shell. The favorable nano-scaled size offers the potential to extravagate from veins and accumulate in tumor tissues via enhanced permeation and retention (EPR) effect. Additionally, CHC-PFP-DOX showed the ability to serve as ultrasound imaging agent at body temperature. Notably, it exhibited an ultrasound-triggered drug release profile through the external ultrasound irradiation. Further study demonstrated that ultrasound combined with CHC-PFP-DOX can improve the killing effect of chemotherapy for tumor. Conclusion: CHC-PFP-DOX holds great promise in simultaneous cancer-targeting ultrasound imaging and ultrasound- mediated delivery for cancer chemotherapy.
Multifunctional mesoporous nanoellipsoids for biological bimodal imaging and magnetically targeted delivery of anticancer drugs

Abstract:A general polyelectrolyte-mediated self-assembly technique is adopted to prepare multifunctional mesoporous nanostructures as an effective biological bimodal imaging probe and magnetically targeted anticancer drug (doxorubicin) delivery systems (DDSs). A positively charged polyelectrolyte (PAH) and negatively charged fluorescent quantum dots (QDs) are successfully assembled onto the surface of ellipsoidal Fe(3)O(4)@SiO(2)@mSiO(2) composite nanostructures to combine the merits of tunable fluorescent/magnetic properties, mesoporous nanostructures for drug loading, and the uniform ellipsoidal morphology for enhanced uptake by cancer cells. The resultant nanoellipsoids are homogeneously coated with four layers of PAH/QDs, with an additional PAH layer to make the ellipsoidal surface positively charged. This acts to enhance cellular uptake, which is driven by electrostatic interactions between the positive nanoparticle surface and the negative cell surface. The high biocompatibility of the achieved multifunctional nanoellipsoids is demonstrated by a cell-cytotoxicity assay, hemolyticity against human red blood cells, and coagulation evaluation of fresh human blood plasma after exposure to the nanoparticles. Moreover, confocal microscopy and bio-TEM observations show that the cell uptake of nanocarriers is dose-dependent, and the nanoparticles accumulate mostly in the cytoplasm. The excellent capability of the nanocarriers as contrast agents for MRI is demonstrated by the relatively high r(2) value (143 mM(-1)s(-1)) and preliminary in vivo characterization. More importantly, the doxorubicin-loaded DDSs show higher cytotoxicity than the free doxorubicin drug as contributed by the intracellular release pathway of doxorubicin from the DDSs, indicating the potential application of the obtained multifunctional mesoporous nanoellipsoids as highly effective bimodal imaging probes and DDSs for cancer diagnosis and chemotherapy, simultaneously.

Abstract:BACKGROUND: Drug delivery in polymeric micelles combined with tumor irradiation by ultrasound results in effective drug targeting, but this technique requires prior tumor imaging. A technology that combined ultrasound imaging with ultrasound-mediated nanoparticle-based targeted chemotherapy could therefore have important applications in cancer treatment.METHODS: Mixtures of drug-loaded polymeric micelles and perfluoropentane (PFP) nano/microbubbles stabilized by the same biodegradable block copolymer were prepared. Size distribution of nanoparticles was measured by dynamic light scattering. Cavitation activity (oscillation, growth, and collapse of microbubbles) under ultrasound was assessed based on the changes in micelle/microbubble volume ratios. The effect of the nano/microbubbles on the ultrasound-mediated cellular uptake of doxorubicin (Dox) in MDA MB231 breast tumors in vitro and in vivo (in mice bearing xenograft tumors) was determined by flow cytometry. Statistical tests were two-sided.RESULTS: Phase state and nanoparticle sizes were sensitive to the copolymer/perfluorocarbon volume ratio. At physiologic temperatures, nanodroplets converted into nano/microbubbles. Doxorubicin was localized in the microbubble walls formed by the block copolymer. Upon intravenous injection into mice, Dox-loaded micelles and nanobubbles extravasated selectively into the tumor interstitium, where the nanobubbles coalesced to produce microbubbles with a strong, durable ultrasound contrast. Doxorubicin was strongly retained in the microbubbles but released in response to therapeutic ultrasound. Microbubbles cavitated under the action of tumor-directed ultrasound, which enhanced intracellular Dox uptake by tumor cells in vitro to a statistically significant extent relative to that observed with unsonicated microbubbles (drug uptake ratio = 4.60; 95% confidence interval [CI] = 1.70 to 12.47; P = .017) and unsonicated micelles (drug uptake ratio = 7.97; 95% CI = 3.72 to 17.08; P = .0032) and resulted in tumor regression in the mouse model.CONCLUSIONS: Multifunctional nanoparticles that are tumor-targeted drug carriers, long-lasting ultrasound contrast agents, and enhancers of ultrasound-mediated drug delivery have been developed and deserve further exploration as cancer therapeutics.

Echogenic Gold Nanorod Incorporated Hybrid Poly(2-oxazoline) Nanocapsules for Real-Time Ultrasound/Fluorescent Imaging and Targeted Cancer Theranostics

Abstract:In recent years, various nanocarrier systems have been explored to enhance the targeting of cancer cells by improving the ligand-receptor interactions between the nanocarrier and cancer cells for selective cancer cell imaging and targeted delivery of anticancer drugs. Herein, we report multifunctional hydrogen-bonded multilayer nanocapsules functionalized with both folic acid-derived quantum dots (FAQDs) and gold nanorods (AuNRs) for targeted cancer therapy and cancer cell imaging using fluorescence microscopy and medical-range ultrasound imaging systems. The encapsulation efficiency of nanocapsules was found to be 49% for 5-fluorouracil (5-FU). The release percentage reached a plateau at 37% after 1 h at pH 7.4 and increased to 57% after 3 h when the release pH was decreased to pH 5.5 (i.e., the pH of the tumor environment). Under ultrasound irradiation, the release was significantly accelerated, with a total release of 52% and 68% after only 6 min at pH 7.4 and pH 5.5, respectively. While the sonoporation process plays an important role in anticancer activity experiments under ultrasound exposure by generating temporary pores, the targeting ability of FAQDs brings the capsules closer to the cell membrane and improves the cellular uptake of the released drug, thereby increasing local drug concentration. In vitro cytotoxicity experiments with HCT-116 and HEp-2 cells demonstrated anticancer activities of 96% and 98%, respectively. The nanocapsules showed enhanced ultrasound scattering signal intensity and bright spots under ultrasound exposure, most likely caused by high scattering ability and internal reflections of preloaded AuNRs in the interior structure of the nanocapsules. Hence, the demonstrated nanocapsule system not only has the potential to be used as an integrated system for early- stage detection and treatment of cancer cells but also has the ability for live tracking and imaging of cancer cells while undergoing treatment with chemotherapy and radiation therapy.

Abstract:Despite the advancements, cancer has remained one of the appalling diseases, with numerous casualties due to delayed prognosis and poor survival rate. To this end, combinatorial strategies based on various therapeutic modalities have garnered enormous interest from researchers towards improved therapeutic performances. This study aims to investigate the fabrication of transition metal-based hollow nanoarchitectures for synergistic augmented photothermal (PTT), chemodynamic (CDT), and chemotherapeutic (CT) modalities. Initially, copper sulfide nanoparticles with hollow cavities (hCuS) are synthesized using the facile sacrificial templating-based approach and subsequently loaded with the CT agent (Doxorubicin, DOX), referred to shortly as hCuS-DOX. These hCuS-DOX nanoarchitectures are systematically characterized using various techniques to successfully explore the physicochemical properties. The DOX molecules are encapsulated in hCuS containers through the electrostatic interactions due to their contrary charges and coordination interactions between copper in CuS and nitrogen in DOX molecules. These innovative nanoarchitectures showed pH-responsive release of DOX molecules from the hollow cavities, specifically in the tumor microenvironment, which could be through the protonation of DOX and progressive degradation of hCuS. In addition, these architectures exhibited substantial PTT properties, on the one hand, by raising the temperature to over 60 °C. On the other hand, the released DOX molecules supported the CDT effect by generating enormous levels of deadly free radical species intracellularly, significantly reducing the viability of 4T1 cells. Finally, the apoptosis assay based on calcein AM/propidium iodide staining using various combinations validated the synergistic therapeutic modalities. In summary, these nanoarchitectures potentiated the synergistic therapeutic efficacy against breast carcinoma towards their ability in pharmaceutics. Abstract:A novel versatile system combining chemotherapy and photothermal therapy for cancer cells using CuS NCs is reported. CuS NCs were synthesized by a solid-liquid reaction between Cu2O nanocubes and thiourea. While the hollow nanocage can be used to load anticancer drugs for chemotherapy, the CuS can convert NIR light into heat for photothermal therapy. More importantly, CuS NCs display exceptionally high drug loading capacity of DOX with 500 wt% and release of DOX is pH-controllable, which is a spontaneous process under intracellular conditions. Compared with chemotherapy or photothermal therapy alone, the combination of chemotherapy and phototherapy can significantly improve the therapeutic efficacy. This improvement strategy using multifunctional nanomaterials may be particularly useful in biomedical applications.
Improving Chemotherapy Effectiveness: Utilizing CuS Nanoparticles Coated with AS1411 Aptamer and Chitosan for Targeted Delivery of Doxorubicin to Cancerous Cells

Abstract:Here, a novel targeted nanostructure complex was designed as an alternative to the traditional treatment approaches for breast cancer. A delivery system utilizing CuS nanoparticles (CuS NPs) was developed for the purpose of targeted administration of doxorubicin (Dox), an anticancer agent. To regulate Dox release, chitosan (CS), a biodegradable and hydrophilic polymer with biocompatible properties, was applied to coat the Dox-loaded CuS NPs. Furthermore, AS1411 aptamer, served as a targeting agent for breast cancer cells (MCF-7 and 4T1 cells), was conjugated with CS-Dox-CuS NPs effectively. To assess the effectiveness of APT-CS-CuS NPs, various methods such as flow cytometry analysis, MTT assay, fluorescence imaging, and in vivo antitumor efficacy were employed. The hollow core and porous surface of CuS NPs improved the Dox loading capacity and entrapment efficiency (almost 100%). The rate of drug release at the tumor site (citrate buffer with pH 5.6) exhibited a marked increase in comparison to that observed within the physiological environment (phosphate buffer with pH 7.4). The targeted formulation (APT-CS-Dox-CuS NPs) significantly increased cytotoxicity of the Dox payload in target cells, including 4T1 (p ≤ 0.0001 (****)) and MCF7 (p ≤ 0.01 (**)) cells compared to CHO cells. Moreover, the ability of tumor growth inhibition of the targeted system was significantly (p ≤ 0.05 (*)) more than free Dox in tumor-bearing mice. The findings indicate that the targeted formulation augmented effectiveness and specificity while minimizing harm to non-targeted cells, signifying its potential as a sophisticated cancer drug delivery system.

Abstract:The combination of chemotherapy and photothermal therapy (PTT) into a single formulation has attracted increasing attention as a strategy for enhancing cancer treatment. Here, hollow mesoporous silica nanoparticles (HMSNs) were used as a base carrier material, loaded with the anti-cancer drug doxorubicin (DOX), and surface functionalized with chitosan (CS) and copper sulfide (CuS) nanodots to give In this formulation, the CuS dots act as gatekeepers to seal the surface pores of the HMSNs, preventing a burst release of DOX into the systemic circulation. S-S bonds connect the CuS dots to the HMSNs; these are selectively cleaved under the reducing microenvironment of the tumor, permitting targeted drug release. This, coupled with the PTT properties of CuS, results in a potent chemo/PTT platform. The nanoparticles have a uniform size (150 ± 13 nm), potent photothermal properties (η= 36.4 %), and tumor-targeted and near infrared (NIR) laser irradiation-triggered DOX release. In vitro and in vivo experimental results confirmed that the material has good biocompatibility, but is effectively taken up by cancer cells. Moreover, the CuS nanodots permit, simultaneous thermal/photoacoustic dual-modality imaging. Treatment with and NIR irradiation caused extensive apoptosis in cancer cells both in vitro and in vivo, and could dramatically extend the lifetimes of animals in a murine breast cancer model. The system developed in this work therefore merits further investigation as a potential nanotheranostic platform for cancer treatment.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Statement of Significance</h3>: Conventional cancer chemotherapy is accompanied by unavoidable off-target toxicity. Combination therapies, which can ameliorate these issues, are attracting significant attention. Here, the anticancer drug doxorubicin (DOX) was encapsulated in the central cavity of chitosan (CS)-modified hollow mesoporous silica nanoparticles (HMSNs). The prepared system can target drug release to the tumor microenvironment. When exposed to near infrared laser (NIR) irradiation, CuS nanodots located at the surface pores of the HMSNs generate energy, accelerating drug release. In addition, a systematic in vitro and in vivo evaluation confirmed the platform to give highly effective synergistic chemotherapeutic-photothermal therapy and have effective thermal/photoacoustic dual-imaging properties. This work may open up a new avenue for NIR-enhanced synergistic therapy with simultaneous thermal/photoacoustic dual imaging.

Abstract:Developing nanomedicines with high-performance is a promising therapeutic strategy for cancer treatment. Herein, a multifunctional nanoplatform (CuS@COFs-BSA-FA/DOX) based on copper sulfide nanoparticles engineered covalent organic frameworks (CuS@COFs) has been established for synergistic photothermal therapy (PTT), chemotherapy and chemodynamic therapy (CDT). The high photothermal effect and excellent Fenton-like catalytic activity enabled CuS@COFs to generate a new minimally invasive PTT/CDT synergistic therapy. Moreover, with the pH-dependent fluorescence behavior and the inherited mesoporous structure, the nanocomposites can also be utilized as fluorescent probes for cancer cell imaging and drug-delivery carriers to encapsulate the chemotherapeutic agent, DOX. The densely coated PEI and BSA-FA layers on the surface of CuS@COFs not only increased the cancer-targeting efficiency but also prevented the nonspecific release of DOX from CuS@COFs. The acid tumor microenvironment and near-infrared light triggered the release of DOX for chemotherapy, which simultaneously augmented the CDT efficiency of CuS@COFs. Notably, the localized hyperthermia induced by PTT can further improve the CDT efficiency of the nanoplatform, leading to a synergistic PTT/chemotherapy/CDT effect. The nanoplatform possessed the capability of cancer cell-targeted imaging and achieved better therapeutic efficacy with negligible systemic toxicity both in vitro and in vivo. Our work opens up a powerful avenue for developing multifunctional COFs-based theranostic platform and shows strong potential in practical applications.

Multifunctional Gd-CuS loaded UCST polymeric micelles for MR/PA imaging-guided chemo-photothermal tumor treatment

Abstract:Hepatocellular carcinoma (HCC) is a life-threatening disease for which there is no effective treatment currently. Novel theranostics simultaneously having excellent imaging and therapeutic functions are highly desired in cancer therapy. Herein, we develop the sialic acid (SA) modified polymeric micelles at an upper critical solution temperature (UCST) of 43 °C (sialic acid-polyethylene glycol)-poly(acrylamide-co-acrylonitrile), SA-PEG-p(AAm-co-AN)), which further encapsulated with doxorubicin (DOX) and Gd-CuS nanoparticles (Gd-CuS NPs) for chemo-photothermal treatment of HCC guided by magnetic resonance (MR)/photoacoustic (PA) dual-mode imaging. The resultant SA-PEG-p(AAm-co-AN)/DOX/Gd-CuS (SPDG) had an excellent photothermal conversion efficiency, enabling SPDG with an instantaneous release behavior of DOX under near-infrared (NIR) irradiation. This study also revealed that SPDG could actively target to HCC, which was due to that SA had a high affinity with E-selectin overexpressed at the tumor site. Moreover, benefiting from the HCC-targeted ability and NIR light-controlled on-demand delivery of DOX, SPDG showed a superior potential in MR/PA dual-mode imaging-guided chemo-photothermal treatment. Overall, our study reveals that the designed SPDG may be used as an ideal multifunctional nanoplatform for cancer theranostics.

Abstract:The superior properties of metal organic frameworks (MOF) can provide great opportunities for merging functional nanoparticles to construct smart and versatile cancer theranostic agents. In this study, on the basis of non-mesoporous nanoparticles (molybdenum disulfide, MoS2), the structure of the MOF shell layer with an adjustable structure can be constructed through the natural coordination interaction between polydopamine (PDA) and iron ion, and the tumor cell target ligand was modified on the surface of the nanocomposite after loading the anticancer drug doxorubicin hydrochloride (DOX) to form a multifunctional cancer theranostics nanoplatform (DOX@MoS2-PMA). Benefiting from the excellent properties of MoS2 and MOF, the favorable photothermal properties and pH/near-infrared (NIR) laser-triggered DOX release behavior of composite nanoparticles were demonstrated. Its well-defined nanostructure, adequate colloidal stability, and satisfactory biocompatibility were further evidenced. Furthermore, the selective tumor cell targeting ability of DOX@MoS2-PMA can improve the cellular uptake efficacy and the photothermal-chemotherapy combination therapy can significantly enhance the killing effect on cancer cells both in vitro and in vivo. In addition, fluorescence imaging results show that nanoparticles can efficiently accumulate inside tumors. The photoacoustic (PA) and magnetic resonance (MR) imaging capabilities derived from different components of nanoparticles can perform better imaging effects. To the best of our knowledge, this is the first attempt to merge the performance of MoS2 with MOF for PA/MR dual-modality imaging-guided photothermal-chemotherapy combination therapy. Our work presented herein proves that MOF can be combined with non-mesoporous nanoparticles and exhibits excellent performance, thus opening a new avenue for endowing non-mesoporous nanoparticles with an efficient drug loading capacity and practical applications of MOFs in nanomedicine.
Cell membrane camouflaged Cu-doped mesoporous polydopamine for combined CT/PTT/CDT synergistic treatment of breast cancer

Abstract:Currently, traditional monotherapy for cancer often results in indiscriminate attacks on the body, leading to the emergence of new health problems. To confront these challenges, multimodal combination therapy has become necessary. However, how to develop new smart nanomaterials through green synthesis methods, delivering drugs while simultaneously synergizing multimodal combination therapies for tumor treatment, remains a topic of great significance. In this study, a biomimetic composite nanomaterial (RM-Cu/P) composed of mesoporous polydopamine (MPDA) as the core and red blood cell membranes (RBCMs) as the shell was synthesized as a drug carrier to deliver doxorubicin (DOX) while achieving synergistic chemotherapy, photothermal and chemodynamic therapy (CT/PTT/CDT). Herein, the nanoparticles were extensively characterized to examine their morphological characteristics, elemental composition, and drug-carrying capacity. Notably, the coating of RBCM reduced the toxicity of the RM-Cu/P@DOX nanoparticles, improved their targeting ability and prolonged their circulation time in vivo. The Cu-doped nanoparticles were capable of initiating a Fenton-like reaction to generate reactive oxygen species (ROS) for CDT, while the photothermal conversion efficiency (η) reached 45.20 % under NIR laser irradiation. Subsequently, the particles were examined by in vivo and in vitro experimental studies in cytotoxicity, cellular uptake, ROS levels, lysosomal escape, and mouse tumor model to evaluate their potential application in antitumor. Compared with monotherapy, the RM-Cu/P@DOX nanoparticles had multiple-stimulation response properties under redox, pH, and NIR, which exhibited the advantage of combined trimodal therapy, resulting in remarkable synergistic antitumor efficacy. In conclusion, this innovative platform exhibited promising applications in smart drug delivery and synergistic treatment of cancer.
Near-Infrared Upconversion Mesoporous Tin Dioxide Theranostic Nanocapsules for Synergetic Cancer Chemophototherapy

Abstract:Smart nanotheranostic systems (SNSs) have attracted extensive attention in antitumor therapy. Nevertheless, constructing SNSs with disease diagnosis ability, improved drug delivery efficiency, inherent imaging performance, and high treatment efficiency remains a scientific challenge. Herein, ultrasmall tin dioxide (SnO2) was assembled with upconversion nanoparticles (UCNPs) to form mesoporous nanocapsules by an in situ hydrothermal deposition method, followed by loading with doxorubicin (DOX) and modification with bovine serum albumin (BSA). pH/near-infrared dual-responsive nanotheranostics was constructed for computed tomography (CT) and magnetic resonance (MR) imaging-induced collaborative cancer treatment. The mesoporous channel of SnO2 was utilized as a reservoir to encapsulate DOX, an antineoplastic drug, for chemotherapy and as a semiconductor photosensitizer for photodynamic therapy (PDT). Furthermore, the DOX-loaded UCNPs@SnO2-BSA nanocapsules combined PDT, Nd3+-doped UCNP-triggered hyperthermia effect, and DOX-triggered chemotherapy simultaneously and demonstrated prominently enhanced treatment efficiency compared to the monotherapy model. Moreover, tin, as one of the trace elements in the human body, has a similar X-ray attenuation coefficient to iodine and therefore can act as a contrast agent for CT imaging to monitor the treatment process. Such an orchestrated synergistic anticancer treatment exhibited apparent inhibition of tumor growth in tumor-bearing mice with negligible side effects. Our study demonstrates nanocapsules with excellent biocompatibility and great potential for cancer treatment.

Abstract:Doxorubicin (Dox), a chemotherapeutic agent, encounters challenges such as a short half-life, dose-dependent toxicity, and low solubility. In this context, the present study involved the fabrication of N -(2-hydroxypropyl)methacrylamide (HPMA) and N -(3-aminopropyl)methacrylamide (APMA) bearing P(HPMA- s -APMA) copolymeric nanoparticles (P(HPMA- s -APMA) NPs) and their investigation for efficient delivery of Dox. Furthermore, the synthesized nanoparticles (NPs) were coated with chitosan (Cht) to generate positively charged nanoformulations. The prepared formulations were evaluated for particle size, morphology, surface charge analysis, percentage encapsulation efficiency (EE%), and drug release studies. The anticancer activity of Cht-P(HPMA- s -APMA)-Dox NPs was assessed in the HeLa cancer cell line. The prepared P(HPMA- s -APMA)-Dox NPs exhibited an average particle size of 240–250 nm. Chitosan decorated P(HPMA- s -APMA)-Dox NPs displayed a significant increase in particle size, and the zeta potential shifted from negative to positive. The EE% for Cht-P(HPMA-s-APMA)-Dox NPs was calculated to be 68.06 %. The drug release studies revealed a rapid release of drug from Cht-P(HPMA- s -APMA)-Dox NPs at pH 4.8 than pH 7.4, demonstrating the pH-responsiveness of nanoformulation. Furthermore, the cell viability assay and internalization studies revealed that Cht-P(HPMA-s-APMA)-Dox NPs had a high cytotoxic response and significant cellular uptake. Hence, the Cht-P(HPMA- s -APMA)-Dox NPs appeared to be a suitable nanocarrier for effective, and safe chemotherapy.

Abstract:Contrast-enhanced ultrasound (CEUS) is widely applied in cancer diagnosis clinically. However, the gas-filled contrast agents are unstable in the blood and exhibit shorter imaging time, which limit their clinical use. In this study, a diagnostic nanoparticle system was developed for dual-mode imaging (ultrasound and fluorescence), which after encapsulation with doxorubicin (DOX) demonstrated simultaneous therapeutic function towards cancer treatment. Thus, calcium carbonate (CaCO3) nanoparticles were encapsulated with doxorubicin (DOX) to obtain CaCO3-DOX. Under acidic conditions, it produced carbon dioxide (CO2) to enhance ultrasound imaging and increase the release of DOX. After intravenously injecting CaCO3-DOX to tumor-bearing mice, in the presence of an ultrasound field, CO2 bubbles were sufficiently generated at the tumor tissues for echogenic reflectivity. Also, the indocyanine green (ICG) was encapsulated into CaCO3 nanoparticles, to further detect the tumor with fluorescence. The resultant theranostic nanoparticle system exhibited therapeutic efficacy towards tumour-bearing mice. Overall, this investigation provides an attractive strategy for dual-mode cancer diagnostics.

Abstract:Inspired by the limitations of nanoparticles in cancer treatment caused by their low therapeutic effects and biotoxicity, biocompatible and photothermal enhanced copper oxide-decorated carbon nanospheres (CuO@CNSs) with doxorubicin hydrochloride (DOX) loading were constructed. CNSs as photothermal agents were synthesized by a hydrothermal reaction. CuO was adsorbed on the surface of CNSs, which improved the photothermal conversion efficiency due to the electron transitions between C-2p and Cu-3d. In addition, CuO would release Cu2+ ions in the tumor microenvironment, which could produce hydroxyl radical (·OH) to induce cancer cells apoptosis via Haber-Weiss and Fenton-like reactions. DOX as a chemotherapeutic agent was located on the surface of CuO@CNSs by electrostatic adsorption and released quickly in the tumor microenvironment to kill cancer cells. The CuO@CNSs-DOX nanoplatforms realized the combination therapy of photothermal therapy (PTT), chemodynamic therapy (CDT), and chemotherapy (CT), which have strong potential for cancer treatment.
