Abstract:Human scrum albumin (HSA) is a major protein component of blood plasma, and its assay is of obvious value to biological research. We, herein, present a readily accessible fluorescent bioprobe for HSA detection and quantitation. A nonemissive tetraphenylethene derivative named sodium 1,2-bis[4-(3-sulfonatopropoxyl)phenyl]-1,2-diphenylethene (BSPOTPE) is induced to emit by IBA, showing a novel phenomenon of aggregation-induced emission (AIE). The AIE bioprobe enjoys a broad working range (0-100 nM), a low detection limit (down to 1 nM), and a superior selectivity to albumins. The fluorescent bioassay is unperturbed by the miscellaneous bioelectrolytes in the artificial urine. The AIE luminogen can also be used as a rapid and sensitive protein stain in gel electrophoresis for HSA visualization. Utilizing the AIE feature of BSPOTPE and the Forster resonance energy transfer from HSA to BSPOTPE, the unfolding process of HSA induced by guanidine hydrochloride is monitored, which reveals a multistep transition with the involvement of molten globule intermediates. Computational modeling suggests that the AIE luminogens dock in the hydrophobic cleft between subdomains IIA and IIIA of HSA with the aid of hydrophobic effect, charge neutralization, and hydrogen bonding interactions, offering mechanistic insight into the microenvironment inside the hydrophobic cavity.

Abstract:Quantitative recognition of human serum albumin (HSA) is of great importance in the fields of biology and biomedicine. In biological fluids, HSA has been used as an important biomarker for disease early diagnosis. For example, microalbuminuria has been regarded as an early indicator of diabetic nephropathy. Therefore, it is of great significance to monitor HSA in urine. In this work, a BODIPY-based fluorescence probe, BDY-OH, was constructed for HSA detection, which showed a high signal to noise ratio and high selectivity. In addition, plot of fluorescence intensity exhibited a 90-fold enhancement and a good linear correlation from 0 mg/mL to 0.4 mg/mL. Encouraged by the outstanding properties, the ability of BDY-OH to distinguish proteinuria from normal urine was tested among 39 clinical urine samples and the excellent recognition rate of 89.7% suggested a great significance for the early diagnosis of kidney related diseases. We look forward to using the BDY-OH method as a new tool for preclinical diagnosis of HSA in urine in the future.

Abstract:Human serum albumin (HSA) has emerged as a pivotal biomarker and prognostic indicator for various human diseases. Real-time sensing and visual tracking of HSA in plasma or other biological systems will immensely facilitate the basic researchers and clinicians to better understand HSA-associated biological processes. Herein, a novel near-infrared (NIR) fluorescent probe (7-HTCF) was rationally constructed for light-up sensing and in-situ imaging of HSA in real samples, based on the principle of twisted intramolecular charge transfer (TICT). Under physiological conditions, 7-HTCF could be efficiently trapped by HSA to form a stable complex via binding on a non-drug binding site, while the complex emitted strong fluoresce signals around 670 nm. Further investigations demonstrated that 7-HTCF displayed a great combination of excellent selectivity and good chemical stability, as well as rapid fluorescent response and ultra-high sensitivity for HSA detection. Particularly, the newly developed light-up probe has been successfully utilized for quantitative detection of HSA in diluted plasma samples, while its readouts are hardly affected by the addition of therapeutic agents and herbal medicines. 7-HTCF is also successfully used for in-situ imaging of the reabsorbed HSA in living renal cells, while this dye exhibits good cell permeability and high resolution for in-situ imaging in living cells. Collectively, a novel TICT-based near-infrared fluorescent probe was devised for highly selective and ultra-sensitive sensing of HSA in plasma samples or imaging HSA in living cells, which offered a practical tool for clinical tests and for exploring HSA-associated biological processes.

Abstract:The tertiary structures of HSA and BSA show a 76 % similarity, and the abnormal levels of HSA in body fluids can cause many diseases. Therefore, accurate quantification of HSA and effectively distinguishing HSA from BSA have significant application value in various fields. In this paper, a deep-red fluorescent probe, NITH , based on 1,8-naphthalimide, is demonstrated as a potential tool for accurately quantifying HSA and distinguishing HSA from BSA. The probe not only exhibits large stokes shift (100 nm) and low detection limit (4.61 μg/L, 0.0692 nM), but also effectively detects HSA by entering its hydrophobic cavity and emitting red fluorescence. Furthermore, NITH is capable of tracking the distribution levels of HSA within the cells through endogenous and exogenous cellular imaging. The application of NITH spans various environments such as cells, zebrafish, urine, and test strips, solidifying it as an effective tool for HSA detection.

Abstract:Human serum albumin (HSA) is an important biomarker that can be used for the early diagnosis of many diseases. In this work, a TICT probe bearing fused naphtho‐furan scaffold (NPNF) was developed and employed in the selective turn‐on sensing of HSA. The probe's selectivity towards HSA was observed using steady‐state fluorescence experiments, with limit of quantitation in micromolar levels. NPNF also displayed its capability exclusively towards HSA over bovine serum albumin (BSA), which was studied/rationalized using anisotropy and time‐resolved studies. Molecular docking was used to shed light on the location of NPNF in the domain I of HSA. The practical application of the probe was also demonstrated by the detection of HSA in urine and the HSA‐assisted detection of cerium.

Abstract:In this study, an amphiphilic organic fluorescent molecule (denoted by BCPB) was employed as a ratiometric fluorescent probe for the detection of human serum albumin (HSA). In an aqueous solution, the micellar assemblies of BCPB showed a strong excimer emission at 660 nm. The presence of HSA can induce the disassembly of BCPB aggregates and reassemble with HSA to form BCPB-HSA complexes, resulting in a ratio change in the fluorescence emission of BCPB at 540 nm (monomer) and 660 nm (aggregate). Accordingly, the fluorescence color also undergoes a visually recognizable change from red to green. As a ratiometric fluorescence probe of HSA, BCPB has a fast response (<5 s), low limit of detection (8.3 nmol L −1 , S / N = 3), good selectivity, and strong anti-interference ability. Finally, BCPB was loaded into the hydrogel to prepare a portable hydrogel kit, which successfully realized the in situ visual detection of HSA in an aqueous solution and human serum.

Abstract:The human serum albumin (HSA) level is a valuable indicator of an individual's health status. Therefore, its detection/estimation can be used to diagnose several diseases. In this work, we have developed a series of donor–π–acceptor probes, which were found to selectively detect HSA over BSA (bovine serum albumin). Among these probes, A4 , which bears the trifluoroacetyl group, showed the highest selectivity for HSA, with limits of detection and quantification being 1.36 nM and 2.59 nM, respectively. CD spectroscopy of the HSA– A4 ensemble indicated an increase in the α-helicity of the protein, while the displacement assays revealed the localization of the probe in the hemin site of HSA. The probe works on the principle of excited state intramolecular charge transfer (ICT). Its selectivity was also validated computationally. Docking experiments confirmed the preference of the probe for the hemin binding IB site of HSA, as observed from the fluorescence displacement assay results, and a comparison of docking scores demonstrated the greater preference of A4 for HSA compared to BSA. Computational experiments also showed a change in preference for HSA amino acid residues exhibited by the excited state of probe A4 (Tyr161, Met123, Pro118, and Leu115) when compared to its ground state (Arg186 and His146). Hydrophobic interactions dominated the excited state protein–probe ensemble, whereas there was significant involvement of the water bridges along with the hydrophobic interactions in the ground state ensemble. Probe A4 was also assessed for its practical utility and found to successfully sense HSA in urine at extremely low concentrations. Moreover, the A4 –HSA ensemble was employed for hemin sensing with a detection limit of 0.23 μM.

Abstract:Human serum albumin (HSA) is an important biomarker for early disease diagnosis. Therefore, the detection of HSA in biological samples is important. In this study, for the sensitive detection of HSA, a fluorescent probe based on Eu(III)-doped yttrium hydroxide nanosheets was designed and sensitized by α-thiophenformyl acetone trifluoride as an antenna. The morphology and structure of the as-prepared nanosheet fluorescent probe were studied by transmission electron microscopy and atomic force microscopy. A detailed investigation of the fluorescence properties of the as-obtained nanosheet probe revealed that the Eu(III) emission intensity was linearly and selectively enhanced by the consecutive addition of HSA. Furthermore, the lifetime signal of this probe was enhanced with increasing concentration. The sensitivity of the nanosheet probe to HSA is discussed based on the results of ultraviolet-visible, fluorescence, and infrared spectral analyses, the findings demonstrate that the nanosheet fluorescent probe we prepared is a highly sensitive and selective tool for detecting HSA concentration with a high intensity and a large lifetime change.

Abstract:BACKGROUND:C-reactive protein (CRP), one of the classic biomarkers of inflammation, is closely related to infectious inflammation, cardiovascular disease, cancer, and other diseases. Therefore, timely and accurate detection of CRP in human blood is crucial for the discovery, diagnosis, and treatment of the aforementioned diseases. Herein, a novel label-free NIR fluorescence aptasensor with a large Stokes shift based on an AIEE anthracene derivative B and a molybdenum disulfide (MoS2) platform was developed and used for the high sensitivity and specificity detection of CRP. RESULTS:Compound B could emit near-infrared (NIR) fluorescence with a large Stokes shift (190 nm). Notably, this compound could bind with the aptamer of CRP (CRP-Apt) through electrostatic attraction to form a B/CRP-Apt complex, generating an aggregation-induced emission enhancement effect and enhancing the fluorescent intensity of B. B/CRP-Apt could be adsorbed on the surface of MoS2 with the addition of MoS2 to its solution, and the fluorescence of Compound B was quenched. CRP was then added to the above solution. CRP-Apt had a substantially higher affinity for CRP than MoS2. Therefore, B/CRP-Apt detached from the surface of MoS2 and bound to CRP, thereby restoring the fluorescence of B. Experimental results showed a good linear relationship between the fluorescent recovery intensity of B and the concentration of CRP in the concentration range of 0.3-70 ng mL-1, with a limit of detection as low as 0.1 ng mL-1. SIGNIFICANCE AND NOVELTY:The aptasensor integrates the advantages of high sensitivity of NIR fluorescence, high specificity of aptamers, good water-solubility and AIEE effect of Compound B. And it could be applied to the determination of CRP in human serum samples, while most of the reported methods can only determine CRP in spiked human serum samples.
Development of D-π-A organic dyes for discriminating HSA from BSA and study on dye-HSA interaction

Abstract:HSA (human serum albumin), a most abundant protein in blood serum, plays a key role in maintaining human health. Abnormal HSA level is correlated with many diseases, and thus has been used as an essential biomarker for therapeutic monitoring and biomedical diagnosis. Development of small-molecule fluorescent probes allowing the selective and sensitive recognition of HSA in in vitro and in vivo is of fundamental importance in basic biological research as well as medical diagnosis. Herein, we reported a series of new synthesized fluorescent dyes containing D-π-A constitution, which exhibited different optical properties in solution and solid state. Among them, dye M-H-SO3 with a hydrophilic sulfonate group at electron-acceptor part displayed selectivity for discrimination of HSA from BSA and other enzymes. Upon binding of dye M-H-SO3 with HSA, a significant fluorescence enhancement with a turn-on ratio about 96-fold was triggered. The detection limit was estimated to be ∼ 40 nM. Studies on the interaction mechanism revealed that dye M-H-SO3 could bind to site III of HSA with a 1:1 binding stoichiometry. Furthermore, dye M-H-SO3 has been applied to determine HSA in real urine samples with good recoveries, which provided a useful method for HSA analysis in biological fluids.

Abstract:In this work, a fluorescent probe, TPABF-HS, was developed for detecting hydrogen sulfide (H 2 S), accompanied by a human serum albumin (HSA)-binding-based approach for amplifying the fluorescence signal and extending the linear correlation range. Compared with the most recent probes for H 2 S, the most interesting feature of the detecting system here was the especially wide linear range of 0-1000 μM (0-100 eq), which covered the physiological and pathological level of H 2 S. TPABF-HS could serve for the application with a relatively high sensitivity and selectivity with the LOD value of 0.42 μM. Further site-competition experiment and molecular docking simulation indicated that the signal amplification was realized by binding the fluorophore TPABF into the naproxen-binding site of HSA. Moreover, the extension of the measurement span could achieve the applications in living cells and Caenorhabditis elegans for imaging both the exogenous and endogenous H 2 S. This work brought new information to the strategy of signal processing in exploiting fluorescent probes.

Abstract:Abnormal concentrations of human serum albumin (HSA) are closely related to the occurrence of various diseases, such as liver cirrhosis and kidney disease. Thus, it is of great importance to monitor HSA in body fluids, especially microalbumin in urine. However, due to the interference of other proteins and the low-polar microenvironment, high signal-to-background (S/B) ratio and selective detection of microalbumin are still a great challenge. To this end, we propose an innovative strategy based on the two-pronged effect of the twisted intramolecular charge transfer (TICT) and the excited state intramolecular proton transfer (ESIPT) mechanisms of flavonoid fluorophores, which is used to significantly improve the signal-to-background ratio of HSA selective fluorescent sensor. As a proof of strategy demonstration, an HSA specific binding flavonoid molecule (FNE3) was developed by combining chemical design and screening approaches. Due to binding to the new binding site (subdomain IB) of HSA in molecular simulations, FNE3 exhibits a remarkable 1042-fold fluorescence response for HAS but only a 1.8-fold for BSA, which is the highest S/B ratio among HSA probes so far, enabling highly sensitive differential detection of HSA from other proteins. Moreover, FNE3 is superior to the commercial BCG method in the rapid and sensitive detection of microalbumin in clinical urine samples, and the quantitative detection results were good agreement with the clinical data. With the excellent performance of FNE3, we further applied it to the selective detection of HSA-specifically bound drugs via a competitive displacement mechanism, and successfully realized the selective visualization of anti-inflammatory drugs diflunisal (DIF) in urine and living cells for the first time. This study indicating that FNE3 may offer a promising tool for the future HSA associated clinical testing and noninvasive pharmaceutical screening.

Abstract:Human serum albumin (HSA) is considered to be a diagnostic index of numerous diseases, and detecting HSA in subcellular environments is of great importance. In this work, we successfully synthesized a fluorescent probe NDIC based on hydrazide Schiff base, which can self-assemble into nanospheres in aqueous solution and produce distinct fluorescence signal in the presence of HSA. NDIC showed advantages including good sensitivity (LOD = 8.44 nM), high selectivity and anti-interference (over different coexisting proteins, amino acids, cations and anions). Job's plot, CD spectra, HSA destruction, site binding replacement and molecular docking provided reliable evidence for the fact that NDIC could combine at drug site I of HSA. Non-toxic probe NDIC could be applied to quantitatively detect HSA contents in urine, and achieve cancer identification by HSA imaging in cancer cells. More importantly, successful confocal experiment proved that NDIC possessed the lysosome selective imaging ability with the aid of HSA. These results showed the probe NDIC met the requirements of biological applications and was expected to explore HSA at the subcellular levels.

Abstract:Pre-clinical diagnosis of many diseases required quantitative detection of Human Serum Albumin (HSA). Herein a high-selective HSA sensor RhHSA was picked through a two-round selectivity evolution from the typical "Effector-π-Trigger" style. RhHSA suggested advantages including high selective (∼6 fold for HSA:BSA = 1:10), sensitive (LOD ∼ 5 nM, over 700-fold enhancement), steady (over 24 h) and wide linear range (0-0.5 mg/mL, applicative for conventional HSA measurement). The detecting system was free from media polarity or viscosity. HSA destruction, site competition and molecular docking provided reliable evidence for the fact that RhHSA could be embedded into both ibuprofen and phenylbutazone sites of HSA. These hints also supported the discrimination of HSA from BSA. Stepwisely fluid replacement in living cells and measuring in urine system both inferred the potential of RhHSA in biological applications.

Abstract:A novel time-resolved fluorescence (TRF) pobe is constructed to detect human serum albumin (HSA) by exploiting ZnGeO:Mn persistent luminescence nanorods (ZnGeO:Mn PLNRs) and polydopamine nanoparticles (PDA NPs). HSA-induced dynamic quenching leads to the fluorescence decrease of ZnGeO:Mn PLNRs, providing the basis for quantitative analysis of HSA. The excellent photo-thermal conversion performance of PDA NPs is helpful to the collision process between ZnGeO:Mn PLNRs and HSA, inducing significant improvement of sensitivity. HSA is quantified by measuring time-resolved fluorescence at 540 nm under excitation of 250-nm light. Under optimal conditions, HSA in the linear range 0.1–100 ng mL−1 are detected by this PDA-mediated ZnGeO:Mn probe with high sensitivity and selectivity, and the detection limit is 36 pg mL−1 (3σ/s). The RSD for the quantification of HSA (5 ng mL−1, n = 11) is 5.2%. The practicability of this TRF probe is confirmed by accurate monitoring HSA contents in urine samples, giving rise to satisfactory spiking recoveries of 96.2–106.0%.Graphical abstract

OTB exhibited favorable selective HSA sensitivity and strong HSA binding ability. As a cationic molecule, OTB could form a stable amphipathic complex with water-soluble pillar [5]arene (WP5). More interestingly, the WP5-OTB complex possessed better HSA responsiveness and stronger HSA binding ability than OTB. The detection limit (3σ/k) of WP5-OTB towards HSA was determined to be 0.33 μM. WP5-OTB complex possessed excellent anti-interference ability in the selective recognition of HSA. Common commercial drugs, fluorescent amino acids, and fatty acids had almost negligible effect on the HSA responsiveness of the complex. The WP5-OTB complex self-assembled into supramolecular fluorescent probe in aqueous solution, and the supramolecular probe possessed favorable HSA-induced bioimaging property, pH-sensitive drug delivery property, and photodynamic activity.

Innovative rural and remote primary health care models: what do we know and what are the research priorities?

Abstract:This paper examines the literature pertaining to 'innovative' primary health care models in rural and remote areas in order to identify areas where knowledge is lacking and describes future research priorities. Although a number of reviews have identified successful primary health care models and synthesised principles that help to understand why they are successful, there is generally a dearth of rigorously collected information regarding rural and remote health service delivery. The evidence base that supports the superiority of any one model or models in a given context is thin because of the lack of systematic, policy-informing evaluation of primary care innovations. The paper identifies the need for more rigorous health services evaluation information, including examination of optimal financing systems, the optimal range and mix of providers, and supports for team practice, appropriate community participation mechanisms, improved health information systems and relevant performance indicators.

Abstract:Human serum albumin (HSA) is an important biomarker for chronic kidney disease, cardiovascular disease, and liver disease, yet its measurement is subject to the routine clinical laboratories. Pioneering studies have demonstrated that few of the ratiometric fluorescent probes enable the point-of-care (POC) testing of HSA with the aid of affordable domestic apparatuses. Still, the lack of choices of fluorophores restricts the development of such assay. In the present work, we propose two design strategies on the typical donor-acceptor fluorescent probe via the structural engineering. The results suggest that replacement of the dimethylamino group in probe by the weaker electron donating thiophene group can effectively tune the fluorescence response to HSA from off-on to ratiometric, ideally suiting for the POC application. The screened fluorescent probe is capable of detecting HSA with the features of fast response (< 10 s), stable output, high sensitivity (limit of detection = 54 nM), tunable detection range, and great anti-interference. Moreover, the ratiometric fluorescence response to HSA can be recorded and analyzed by the portable miniaturized paper-based or 3D-printed devices at the point of need. The success of structural engineering in the present study provides an efficient way to rationally regulate the fluorescence response mode of the donor-acceptor probes.
