Abstract:Sensory neuron membrane proteins (SNMPs), which are located on the dendritic membrane of olfactory sensory neurons (OSNs), are proposed to be associated with odor reception in insects. Recent studies have demonstrated that SNMP1 is essential for electrophysiological responses of OSNs to the sex pheromone, cis-vaccenyl acetate (cVA) in Drosophila melanogaster. To investigate the function of Lepidoptera SNMPs, we cloned two SNMP genes, SlituSNMP1 and SltiuSNMP2, from Spodoptera litura (Lepidoptera: Noctuidae). Sequence alignment and phylogenetic analysis showed that both genes bear the general characteristics of SNMPs, including six conserved cysteine residues and two transmembrane domains. Further expression profile experiments showed that SlituSNMP1 is mainly expressed in the antenna, while SlituSNMP2 is broadly expressed in various tissues. By in situ hybridization experiments, it was found that SlituSNMP1 expressing cells are surrounded by the SlituSNMP2 expressing cells in the pheromone sensitive sensilla, suggesting different functions of the two SNMPs in insect olfaction.
Identification and Characterization of Chemosensory Genes in the Antennal Transcriptome of Spodoptera Exigua

Abstract:Chemical senses are crucial for insect behaviors such as host preference, mate choice and oviposition site selection. Various protein families are involved in these processes, including odorant receptors (ORs), ionotropic receptors (IRs), gustatory receptors (GRs), chemosensory proteins (CSPs), odorant binding proteins (OBPs) and sensory neuron membrane proteins (SNMPs). To better understand the olfactory mechanism in Spodoptera exigua, we conducted transcriptome analysis of adult antennae and identified a total of 157 candidate chemosensory genes encoding 51 ORs, 20 IRs, 7 GRs, 32 CSPs, 45 OBPs and 2 SNMPs. Quantitative real time PCR (qPCR) analysis of the tissue- and sex-specific expression profiles of ORs, GRs and IRs revealed that these genes could be detected in at least one tissue tested. SexiOR6, 11, 13 and 16, which were predicted to be pheromone receptors based on phylogenetic analysis, exhibited male-specific antennae expression. SexiOR18, 26, 28, 30, 34, 39, and 40 exhibited female-biased expression. SexiGR1, SexiGR2 and SexiGR3, are predicted carbon dioxide receptors, and the former was expressed specifically in antennae, and the latter two were expressed both in antennae and labial palps. SexiIRs had diverse expression profiles. SexiIR8a and SexiIR25a were quite conserved and expressed at high levels. This work will greatly facilitate the understanding of olfactory system in S. exigua and provides valuable information for further functional studies of the chemoreception mechanism in Lepidopteran moths.

Abstract:Sensory neuron membrane proteins (SNMPs) are olfactory-specific, two-transmembrane proteins. Previous publications reported that SNMP1 is expressed on the dendrite membrane of pheromone-sensitive neurons in Heliothis virescens and is an essential cofactor for pheromone detection in Drosophila. In this study, we cloned two SNMP genes (GenBank accession nos. JX469106 and JX469107) from the antenna of the beet armyworm moth Spodoptera exigua (Lepidoptera: Hübner). These SNMP genes are classified into two highly conserved subclades, indicating their importance in physiological activity of lepidopteran insects. SexiSNMP1 is antenna-specific in male and female adults, while SexiSNMP2 is antenna-abundant but also expressed in other chemosensory tissues, particularly proboscises and maxillary palps of adults both sexes. In situ hybridization revealed that both SNMPs are broadly expressed in long and short trichoid and basiconic sensilla. We infer that SNMP1 and SNMP2 act in the detection of the sex pheromone and general odorants.

Abstract:The diffusion (${\mathit{D}}_{\mathit{s}}$) of entangled and unentangled symmetric block copolymers is compared, above and below the ordering transition. The lamellar samples were not macroscopically oriented, and thus ${\mathit{D}}_{\mathit{s}}$ reflects a combination of mobilities parallel and perpendicular to the lamellar planes. For unentangled (Rouse) chains, ${\mathit{D}}_{\mathit{s}}$ is unaffected by the microstructure, whereas entangled (reptating) chains are retarded significantly; this difference reflects an interesting combination of thermodynamic and entanglement constraints. Furthermore, this retardation persists into the nominally disordered state.

Abstract:In Drosophila, olfactory sensory neurons (OSNs) rely primarily on two types of chemoreceptors, odorant receptors (Ors) and ionotropic receptors (Irs), to convert odor stimuli into neural activity. The cellular signaling of these receptors in their native OSNs remains unclear because of the difficulty of obtaining intracellular recordings from Drosophila OSNs. Here, we developed an antennal preparation that enabled the first recordings (to our knowledge) from targeted Drosophila OSNs through a patch-clamp technique. We found that brief odor pulses triggered graded inward receptor currents with distinct response kinetics and current-voltage relationships between Or- and Ir-driven responses. When stimulated with long-step odors, the receptor current of Ir-expressing OSNs did not adapt. In contrast, Or-expressing OSNs showed a strong Ca(2+)-dependent adaptation. The adaptation-induced changes in odor sensitivity obeyed the Weber-Fechner relation; however, surprisingly, the incremental sensitivity was reduced at low odor backgrounds but increased at high odor backgrounds. Our model for odor adaptation revealed two opposing effects of adaptation, desensitization and prevention of saturation, in dynamically adjusting odor sensitivity and extending the sensory operating range.

Abstract:Local interneurons in the Drosophila antennal lobe are thought to play important roles in shaping odor responses. However, the physiological properties of excitatory local interneurons (eLNs) and their connectivity in the antennal lobe remain unclear. We first characterized the firing patterns of krasavietz-Gal4-labeled eLNs (krasavietz eLNs) in response to depolarizing currents. Paired recordings of krasavietz eLNs and PNs showed reciprocal excitatory connections mediated by dendrodendritic cholinergic synapses and gap junctions. Reciprocal connections were also found between two krasavietz eLNs but were rare between krasavietz eLNs and inhibitory LNs. Analysis of response onset latencies showed that krasavietz eLNs received monosynaptic inputs from ORNs. Furthermore, each eLN responded with distinct patterns to different odors, and each odor elicited distinct responses in different eLNs, with specific temporal patterns of spiking, indicating that eLNs serve specific coding functions in addition to global excitation in Drosophila olfactory processing.

Abstract:Background: The nose of most animals comprises multiple sensory subsystems, which are defined by the expression of different olfactory receptor families. Drosophila melanogaster antennae contain two morphologically and functionally distinct subsystems that express odorant receptors (Ors) or ionotropic receptors (Irs). Although these receptors have been thoroughly characterized in this species, the subsystem-specific expression and roles of other genes are much less well-understood. Results: Here we generate subsystem-specific transcriptomic datasets to identify hundreds of genes, encoding diverse protein classes, that are selectively enriched in either Or or Ir subsystems. Using single-cell antennal transcriptomic data and RNA in situ hybridization, we find that most neuronal genes-other than sensory receptor genes-are broadly expressed within the subsystems. By contrast, we identify many non-neuronal genes that exhibit highly selective expression, revealing substantial molecular heterogeneity in the non-neuronal cellular components of the olfactory subsystems. We characterize one Or subsystem-specific non-neuronal molecule, Osiris 8 (Osi8), a conserved member of a large, insect-specific family of transmembrane proteins. Osi8 is expressed in the membranes of tormogen support cells of pheromone-sensing trichoid sensilla. Loss of Osi8 does not have obvious impact on trichoid sensillar development or basal neuronal activity, but abolishes high sensitivity responses to pheromone ligands. Conclusions: This work identifies a new protein required for insect pheromone detection, emphasizes the importance of support cells in neuronal sensory functions, and provides a resource for future characterization of other olfactory subsystem-specific genes.

Abstract:Serotonin plays different roles across networks within the same sensory modality. Previously, we used whole-cell electrophysiology in Drosophila to show that serotonergic neurons innervating the first olfactory relay are inhibited by odorants (Zhang and Gaudry, 2016). Here we show that network-spanning serotonergic neurons segregate information about stimulus features, odor intensity and identity, by using opposing coding schemes in different olfactory neuropil. A pair of serotonergic neurons (the CSDns) innervate the antennal lobe and lateral horn, which are first and second order neuropils. CSDn processes in the antennal lobe are inhibited by odors in an identity independent manner. In the lateral horn, CSDn processes are excited in an odor identity dependent manner. Using functional imaging, modeling, and EM reconstruction, we demonstrate that antennal lobe derived inhibition arises from local GABAergic inputs and acts as a means of gain control on branch-specific inputs that the CSDns receive within the lateral horn.

Abstract:In insect olfaction, sensitization refers to the amplification of a weak olfactory signal when the stimulus is repeated within a specific time window. In the vinegar fly, Drosophila melanogaster, this occurs already at the periphery, at the level of olfactory sensory neurons (OSNs) located in the antenna. In our study, we investigate whether sensitization is a widespread property in a set of seven types of OSNs, as well as the mechanisms involved. First, we characterize and compare the differences in spontaneous activity, response velocity and response dynamics, among the selected OSN types. These express different receptors with distinct tuning properties and behavioral relevance. Second, we show that sensitization is not a general property. Among our selected OSN types, it occurs in those responding to more general food odors, while OSNs involved in very specific detection of highly specific ecological cues like pheromones and warning signals show no sensitization. Moreover, we show that mitochondria play an active role in sensitization by contributing to the increase in intracellular Ca2+ upon weak receptor activation. Thus, by using a combination of single sensillum recordings (SSRs), calcium imaging and pharmacology, we widen the understanding of how the olfactory signal is processed at the periphery.
A spatial map of antennal-expressed ionotropic receptors in the malaria mosquito

Abstract:The mosquito's antenna represents its main olfactory appendage for detecting volatile chemical cues from the environment. Whole-mount fluorescence in situ hybridization of ionotropic receptors (IRs) expressed in the antennae reveals that the antenna might be divisible into proximal and distal functional domains. The number of IR-positive cells appear stereotyped within each antennal segment (flagellomere). Highly expressed odor-tuning IRs exhibit distinct co-localization patterns with the IR coreceptors Ir8a, Ir25a, and Ir76b that might predict their functional properties. Genetic knockin and in vivo functional imaging of IR41c-expressing neurons indicate both odor-induced activation and inhibition in response to select amine compounds. Targeted mutagenesis of IR41c does not abolish behavioral responses to the amine compounds. Our study provides a comprehensive map of IR-expressing neurons in the main olfactory appendage of mosquitoes. These findings show organizing principles of Anopheles IR-expressing neurons, which might underlie their functional contribution to the detection of behaviorally relevant odors.

Abstract:Insect olfactory receptor neurons are compartmentalized in sensilla. In a sensillum, typically two receptor neurons are in close contact and can influence each other through electrical interaction during stimulation. This interaction is passive, non-synaptic and a consequence of the electrical structure of the sensillum. It is analysed in a sensillum model and its effects on the neuron receptor potentials are investigated. The neurons in a sensillum can be both sensitive to a given odorant compound with the same sensory threshold or with different thresholds, or only one neuron be sensitive to the odorant. These three types of sensilla are compared with respect to maximum amplitude, threshold and dynamic range of the potentials. It is found that gathering neurons in the same sensillum is disadvantageous if they are identical, but can be advantageous if their thresholds differ. Application of these results to actual recordings from pheromone and food-odour olfactory sensilla is discussed.

Abstract:Insect odorant receptorsIn many organisms, from worms to humans, olfactory cues are detected by large families of seven transmembrane-spanning receptors, which have until now been classified as G protein-coupled receptors. Insects, however, have evolved a surprisingly simple and efficient sense of smell in which the odorant receptors require a second component — the ion-channel-forming chaperone protein Or83b — for correct function. In the first of two related papers, Sato et al. show that these heteromeric receptors form ligand-gated cation channels that are not dependent on G protein-coupled second messengers, and speculate that other seven transmembrane-spanning proteins may show similar ion channel activity. Wicher et al. show that, in addition to direct channel activation, ligand binding to odorant receptors causes G protein-coupled channel activation. This work has implications for the search for insect odorant receptor inhibitors for possible use in controlling host seeking behaviour of disease carrying insects such as the mosquito.

Abstract:Analysis of covariance (ANCOVA) techniques are often employed in the analysis of clinical trials to try to account for the effects of varying pretreatment baseline values of an outcome variable on posttreatment measurements of the same variable. Baseline measurements of outcome variables are typically random variables, which violates the usual ANCOVA assumption that covariate values are fixed. Therefore, the usual ANCOVA hypothesis tests of treatment effects may be invalid, and the ANCOVA slope parameter estimator biased, for this application. We show, however, that if the pretreatment - posttreatment measurements have a bivariate normal distribution, then (i) the ANCOVA model with residual error independent of the covariate is a valid expression of the relationship between pretreatment and posttreatment measurements; (ii) the usual (fixed-covariate analysis) ANCOVA estimates of the slope parameter and treatment effect contrasts are unbiased; and (iii) the usual ANCOVA treatment effect contrast t-tests are valid significance tests for treatment effects. Moreover, as long as the magnitudes of the treatment effects do not depend on the "true" pretreatment value of the outcome variable, the true slope parameter must lie in the interval (0, 1) and the ANCOVA model has a clear interpretation as an adjustment (based on between- and within-subject variability) to an analysis of variance model applied to the posttreatment-pretreatment differences.

Abstract:PURPOSE To determine whether frontal white matter diffusion abnormalities can help predict acute executive function impairment after mild traumatic brain injury (mTBI). MATERIALS AND METHODS This study had institutional review board approval, included written informed consent, and complied with HIPAA. Diffusion-tensor imaging and standardized neuropsychologic assessments were performed in 20 patients with mTBI within 2 weeks of injury and 20 matched control subjects. Fractional anisotropy (FA) and mean diffusivity (MD) images (imaging parameters: 3.0 T, 25 directions, b = 1000 sec/mm(2)) were compared by using whole-brain voxelwise analysis. Spearman correlation analyses were performed to evaluate associations between diffusion measures and executive function. RESULTS Multiple clusters of lower frontal white matter FA, including the dorsolateral prefrontal cortex (DLPFC), were present in patients (P < .005), with several clusters also demonstrating higher MD (P < .005). Patients performed worse on tests of executive function. Lower DLPFC FA was significantly correlated with worse executive function performance in patients (P < .05). CONCLUSION Impaired executive function following mTBI is associated with axonal injury involving the DLPFC.
Differential modulation of sensory response dynamics by cilia structure and intraflagellar transport within and across chemosensory neurons

Abstract:Sensory neurons contain morphologically diverse primary cilia that are built by intraflagellar transport (IFT) and house sensory signaling molecules. Since both ciliary structural and signaling proteins are trafficked via IFT, it has been challenging to decouple the contributions of IFT and cilia structure to neuronal responses. By acutely inhibiting IFT without altering cilia structure and vice versa, here we describe the differential roles of ciliary trafficking and sensory ending morphology in shaping chemosensory responses in C. elegans. We show that a minimum cilium length but not continuous IFT is necessary for a subset of responses in the ASH nociceptive neurons. In contrast, neither cilia nor continuous IFT are necessary for odorant responses in the AWA olfactory neurons. Instead, continuous IFT differentially modulates response dynamics in AWA. Upon acute inhibition of IFT, cilia-destined odorant receptors are shunted to ectopic branches emanating from the cilia base. Spatial segregation of receptors in these branches from a cilia-restricted regulatory kinase results in odorant desensitization defects, highlighting the importance of precise organization of signaling molecules at sensory endings in regulating response dynamics. We also find that adaptation of AWA responses upon repeated exposure to an odorant is mediated by IFT-driven removal of its cognate receptor, whereas adaptation to a second odorant is regulated via IFT-independent mechanisms. Our results reveal unexpected complexity in the contribution of IFT and cilia organization to the regulation of responses even within a single chemosensory neuron type, and establish a critical role for these processes in the precise modulation of olfactory behaviors.

Abstract:Insects use olfaction to detect ecologically relevant chemicals in their environment. To maintain useful responses over a variety of stimuli, olfactory receptor neurons are desensitized to prolonged or high concentrations of stimuli. Depending on the timescale, the desensitization is classified as short-term, which typically spans a few seconds; or long-term, which spans from minutes to hours. Compared with the well-studied mechanisms of desensitization in vertebrate olfactory neurons, the mechanisms underlying invertebrate olfactory sensitivity regulation remain poorly understood. Recently, using a large-scale functional screen, a conserved critical receptor phosphorylation site has been identified in the model insect Drosophila melanogaster, providing new insight into the molecular basis of desensitization in insects. Here, we summarize the progress in this area and provide perspectives on future directions to determine the molecular mechanisms that orchestrate the desensitization in insect olfaction.
