Abstract:A novel antimicrobial composite material was prepared by encapsulating orange essential oil (OEO) in zein prolamine (ZP) via the coaxial electrospinning (ES) technique. By manipulating process parameters, the morphological features of ZP/OEO fibers were modulated. Fine fibers with diameters ranging from 0.7 to 2.3 μm were obtained by regulating ZP solution concentration and process parameters during the ES process. Optimal loading capacity (LC) and encapsulation efficiency (EE) of OEO in fibrous ZP mats were determined to be 22.28% and 53.68%, respectively, and were achieved using a 35 w/v% ZP ES solution. The encapsulation of OEO was found to be reliant on ZP solution concentration (the enveloping medium). SEM analysis indicates the surface morphology of ZP/OEO electrospun fibers is dependent on ZP solution loading volume, with lower ZP concentrations yielding defective fibrous structures (for example, beaded and spindled-string like morphologies). Furthermore, this loading volume also influences OEO LC, EE, mat water contact angle and oil retention. CCK-8 assay and cell morphology assessment (HEK293T cells) indicate no significant change with electrospun ZP and ZP/OEO fibrous membranes over an 8 h period. Antimicrobial activity assessment using Escherichia coli, suggests composite nonwovens possess sterilization properties; elucidating potential application in active food packaging, food preservation and therefore sustainability.

Abstract:Using smart nanopesticide formulations based on nanomaterials can offer promising potential applications for decreasing pesticide residues and their effects on human health and the environment. In this study, a novel nanoformulation (NF) of thiamethoxam (TMX) was fabricated using the solvent evaporation method through loading TMX on cellulose nanocrystals (CNCs) as the carrier. The synthesized TMX-CNCs was investigated through different techniques, such as Fourier transform infrared spectrometer (FT-IR), X-ray diffraction (XRD), transmission electron microscopy (TEM), dynamic light scattering (DLS), and thermogravimetric analysis (TGA). The results revealed that the loading efficiency and entrapment efficiency were 18.7% and 83.7 ± 1.8% for TMX, respectively. The prepared nanoformulation (TMX-CNCs) had a width of 7–14 nm and a length of 85–214 nm with a zeta potential of −23.6 ± 0.3 mV. The drug release behavior study exhibited that the release of TMX from TMX-loaded CNCs was good and sustained. Furthermore, bioassay results showed that the insecticidal activity of TMX-CNCs against Phenacoccus solenopsis was significantly superior to that of the technical and commercial formulation, as indicated by the lower LC50 value. The results indicate that the TMX nanoformulation has great potential for application in agriculture for pest control.

Abstract:Controlled release formulations (CRFs) are considered an effective way to solve the low bioavailability of traditional pesticides. However, CRFs prepared by coating or encapsulation has the disadvantage of explosive release of the ingredients. Sustained-release pesticides prepared by coupling with a carrier can overcome this shortcoming. In the present study, an emamectin-lignin sulfonic acid conjugate (EB-SL), in which emamectin was connected via sulfonamide bonds with lignin, was prepared using sodium lignosulfonate as the carrier. The structure of the conjugate was characterized by IR, 1HNMR, and elemental analysis. The sustained-release results showed that EB-SL maintained its original structure when released in pure water and soil columns, and the sulfamide bond did not break. The photolysis test displayed that the photolysis half-life T 0.5 of EB-SL was increased by 1.5 times compared with the emamectin suspending concentrate (EB-SC). Bioactivity tests in the greenhouse showed that EB-SL not only had similar insecticidal toxicity to emamectin emulsion concentrate (EB-EC) against Ostrinia nubilalis but also displayed a longer duration. The lethality of EB-SL on O. nubilalis was maintained at more than 70% across 19 days, whereas EB-EC as the control was less than 50% after 11 days of application.
Preparation, characterization, ex vivo transdermal properties and skin irritation evaluation of 1,8-cineole nanoemulsion gel

Abstract:1,8-cineole (1,8-CIN) is a monoterpene that has significant inhibitory effects on dermatophytes. However, its volatile and difficult to stay in the skin have been the major limitation against its use. The aim of this study was to increase the deposition amount of 1,8-CIN in the skin and enhance its targeting to the skin surface. In the present study, 1,8-cineole nanoemulsion (1,8-CIN-NE) was prepared by the Water Titration method. Then, 1,8-cineole nanoemulsion gel (1,8-CIN-NG) was prepared by mixing 1,8-CIN-NE with gel substrates. Finally, its characterization, stability, in vitro antifungal activity, skin irritation, and transdermal properties were evaluated. The optimal 1,8-CIN-NG was around 21 nm in size with a high degree of monodispersity and the nanoemulsion droplets were surrounded by gridded gel substrates. 1,8-CIN-NG maintained excellent stability under various conditions and had no skin irritation. 1,8-CIN-NG had a significant inhibitory effect on dermatophytes. Nanoemulsion gel (NG) increased the deposition of 1,8-CIN in the skin. The histopathological evaluation indicated that 1,8-CIN-NG treatment group showed less morphological changes in the skin than the 1,8-CIN-NE or 1,8-CIN alone groups. This result is consistent with the results of the skin irritation test slight, indicating that 1,8-CIN-NG is a safe topical preparation. These results suggested that 1,8-CIN-NG can be used as an efficient agent to manage dermatophytes infections.

Abstract:A new approach employing a combination of pyrethroid and repellent is proposed to improve the protective efficacy of conventional pyrethroid-treated fabrics against mosquito vectors. In this context, the insecticidal and repellent efficacies of commonly used pyrethroids and repellents were evaluated by cone tests and arm-in-cage tests against Stegomyia albopicta (=Aedes albopictus) (Diptera: Culicidae). At concentrations of LD50 (estimated for pyrethroid) or ED50 (estimated for repellent), respectively, the knock-down effects of the pyrethroids or repellents were further compared. The results obtained indicated that deltamethrin and DEET were relatively more effective and thus these were selected for further study. Synergistic interaction was observed between deltamethrin and DEET at the ratios of 5:1, 2:1, 1:1 and 1:2 (but not 1:5). An optimal mixing ratio of 7:5 was then microencapsulated and adhered to fabrics using a fixing agent. Fabrics impregnated by microencapsulated mixtures gained extended washing durability compared with those treated with a conventional dipping method. Results indicated that this approach represents a promising method for the future impregnation of bednet, curtain and combat uniform materials.

Abstract:The encapsulation of botanical compounds (such as citronellal) can be used to increase their efficiency and stability for functionalizing textile materials. In this study, the citronellal nanocapsules (CNC) with citronella oil as the core material and isophorone diisocyanate (IPDI) as the shell material were prepared and characterized. Factors, including core-to-shell ratio, single-phase ratios, emulsification time, as well as polymerization time and temperature, were investigated to determine the optimal process for the preparation of CNC. The average particle size of the prepared CNC was about several hundred nanometers, with uniform particle size distribution. Further, composite Lyocell fibers were prepared by the wet spinning process of co-blending CNC with Lyocell spinning stock. Evaluations were made of the biological activity of CNC towards mosquitoes, as well as E. coli and S. aureus, showing excellent mosquito repelling rate, of more than 90%, and an inhibition rate of 97.72% towards S. aureus.

Characterization of an antibacterial peptide from indian cobra (Naja naja) venom

Abstract:Due to the development of antibiotic resistance in microorganisms, antimicrobial peptides from natural sources have attracted attention in recent times. Several antimicrobial peptides have been isolated from a wide range of animal sources, particularly snake venoms. Naja naja venom showed antibacterial as well as direct and indirect hemolytic activities, and an antibacterial peptide was purified through gel permeation and ion exchange chromatography. Its molecular mass was 2491Da, which was determined using Matrix Assisted Laser Desorption/Ionization-Time-of-Flight (MALDI-TOF) mass spectrometry and the amino acids sequence of the N-terminus was DEQSTHGAYVWKL. The purified peptide showed potent antibacterial activity against Gram-negative and Gram-positive bacterial strains like Escherichia coli, Pseudomonas aeruginosa and Vibrio cholerae, and Staphylococcus aureus, Enterococcus faecalis, Streptococcus pneumoniae, Streptococcus pyogenes, Bacillus subtilis, respectively. The most potent activity was towards Gram-negative bacteria. Activity was retained at concentrations as low as 100µg/ml. Minimum inhibitory concentrations (MIC; in mg) of Naja Antibacterial Peptide (NAP) and known antibiotics against Gram-positive and Gram-negative bacteria were determined using microdilution susceptibility test in sterile 96-well microdilution plates. However, the peptide did not show direct or indirect hemolytic activity.

Abstract:Microbial infections due to bacteria, viruses, and molds are a serious threat to both human life and the health of other organisms. To develop inexpensive, easy-to-prepare, efficient, and portable nano-antibacterial materials, as well as to explore the antibacterial prospects of cationic antibacterial agents, in this work, six different membrane materials were prepared by the electrostatic spinning method and characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and Fourier transform infrared (FT-IR). The materials were tested for antimicrobial properties using a modified AATCC100-200 test method. Under the most suitable spinning conditions, the doping amount of the cationic antimicrobial agent, CTAB, had the greatest influence on the antimicrobial performance. The antimicrobial performance of PCL/PEO/CS/CTAB0.4 was the highest among the prepared materials, with 83.7% effectiveness against S. aureus and 99.9% against E. coli. The antimicrobial performance was found to be stable. In our study, we determined the most suitable spinning ratio to prepare an inexpensive and efficient cationic antimicrobial agent. Biodegradable, high-antimicrobial-activity antimicrobial materials can be applied as films, and this new nanofiber material has shown great potential in wound dressings and as a mask material due to its remarkable antimicrobial efficiency.

Abstract:New approaches to deal with the growing concern associated with antibiotic-resistant bacteria are emerging daily. Essential oils (EOs) are natural antimicrobial substances with great potential to mitigate this situation. However, their volatile nature, in their liquid-free form, has restricted their generalized application in biomedicine. Here, we propose the use of cellulose acetate (CA)/polycaprolactone (PCL) wet-spun fibers as potential delivery platforms of selected EOs to fight infections caused by Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). Twenty EOs were selected and screened for their minimal inhibitory concentration (MIC), using the antibiotic ampicillin as positive control. The cinnamon leaf oil (CLO), cajeput oil (CJO), and the clove oil (CO) were the most effective EOs, against the Gram-positive (MIC < 22.38 mg/mL) and the Gram-negative (MIC < 11.19 mg/mL) bacteria. Uniform microfibers were successfully wet-spun from CA/PCL with an averaged diameter of 53.9 ± 4.5 µm, and then modified by immersion with CLO, CJO and CO at 2 × MIC value. EOs incorporation was confirmed by UV-visible spectroscopy, Fourier-transformed infrared spectroscopy, and thermal gravimetric analysis. However, while microfibers contained ampicillin at MIC (control) after the 72 h modification, the CLO, CO and CJO-loaded fibers registered ≈ 14%, 66%, and 76% of their MIC value, respectively. Data showed that even at small amounts the EO-modified microfibers were effective against the tested bacteria, both by killing bacteria more quickly or by disrupting more easily their cytoplasmic membrane than ampicillin. Considering the amount immobilized, CLO-modified fibers were deemed the most effective from the EOs group. These results indicate that CA/PCL microfibers loaded with EOs can be easily produced with increased antibacterial action, envisioning their use as scaffolding materials for the treatment of infections.

Abstract:Bovine spermatozoa were separated into different density subpopulations utilizing water insoluble hydrocarbon and silicone gels of defined specific gravity. Sperm density profiles were generated for 13 bulls. The separations were found to be repeatable and characteristic of the bull examined. Considerable density variation among animals was demonstrated. Analysis of the separated spermatozoa, before and after freezing, demonstrated that good motility and acrosomal integrity of spermatozoa were maintained. When the least dense fraction of spermatozoa was used for insemination, conception rates were similar to those obtained routinely by artificial insemination with unfractionated spermatozoa. Therefore, this system may be useful in separating spermatozoa of various densities and for removing extraneous matter from semen. However, the sex ratio, among 51 60-day-old fetuses recovered from heifers inseminated with the lowest density fraction of spermatozoa, was 26 males:25 females.

Abstract:Controlled pesticide release in response to environmental stimuli by encapsulating pesticide in carrier is a feasible approach to improve the effective utilization rate. Here, a temperature-responsive release microcapsule loaded with chlorpyrifos () was prepared from n-hexadecane-in-water emulsions via interfacial polymerization. The microcapsule was consisted of nanofibrillated cellulose (NFC) as the shell wall material and isophorone diisocyanate (IPDI) as the crosslinker. The prepared had pesticide-loading efficiency (33.1 wt%) and favorable adhesion on the surface of cucumber and peanut foliage compared with conventional formulation. Additionally, could protect chlorpyrifos against photodegradation effectively. The in vitro release test showed that microcapsule had adjustable controlled-release characteristics with the change in temperature based on phase transition of the n-hexadecane core. Bioassay studies showed that control efficacy of microcapsule against P. xylostella was positively correlated with temperature because of temperature-induced changes in release rate. The acute toxicity of to zebrafish was reduced more than 5-fold compared with that of CPF technical. These results indicated that the microcapsule release system has great potential in the development of an effective and environmentally friendly pesticide formulation.

Abstract:Emamectin benzoate (EB), a widely used pesticide, is prone to decomposition by ultraviolet light and suffers from the corresponding loss of efficacy. The timed release of EB based on microspheres is one of the effective methods to solve this issue. As a non-toxic cellulose ester, cellulose acetate butyrate (CAB) is regarded as one of the best wall-forming materials for microcapsules with a good controlled release performance. Herein, two methods—mechanical activation (MA) technology and a conventional liquid phase (LP) method—were employed to synthesize different CABs, namely CAB-MA and CAB-LP, respectively. The molecular structure, rheological property, and thermal stability of these CABs were investigated. The two CABs were used to prepare microspheres for the loading and release of EB via an o/w (oil-in-water) solvent evaporation method. Moreover, the performances such as drug loading, drug entrapment, and anti-photolysis of the drug for these microspheres were studied. The results showed that both CABs were available as wall materials for loading and releasing EB. Compared with CAB-LP, CAB-MA presented a lower molecular weight and a narrower molecular weight distribution. Moreover, the MA method endowed the CAB with more ester substituent groups and less crystalline structure in comparison to the LP method, which had benefits including pelletizing and drug loading.

Abstract:Although the use of synthetic chemicals is the principal method for insect pest management, their widespread application has led to numerous side effects, including environmental pollution and threats to human and animal health. Plant essential oils have been introduced as promising natural substitutes for synthetic insecticides. However, high volatility and/or low durability are the main limiting factors for essential oil application for control of insect pests. Accordingly, along with an evaluation of the fumigant toxicity of Eucalyptus largiflorens essential oil against the cowpea weevil, Callosobruchus maculatus, essential oil was nanoencapsulated by two mesoporous silicates, MCM-41 and zeolite 3A, to enhance fumigant persistence and toxicity. The chemical profile of essential oil was also analyzed through gas chromatographic-mass spectrometry. E. largiflorens essential oil showed significant concentration-dependent toxicity against insect pests; a concentration of 5.16 μL/L resulted in 100% mortality after 48 h. The toxicity of essential oil could be attributed to the presence of various insecticidal terpenes, such as spathulenol (15.6%), cryptone (7.0%), and 1,8-cineole (5.8%). Fumigant persistence was increased from 6 days to 19 and 17 days for pure and capsulated essential oil with MCM-41 and Zeolite 3A, respectively. The insect mortality also increased from 99 insects in pure essential oil to 178 and 180 insects in MCM-41 and Zeolite 3A encapsulated formulations, respectively. Therefore, the encapsulation of E. largiflorens essential oil by MCM- 41 and Zeolite 3A is a beneficial method for enhancing its persistence and toxicity against C. maculatus.

Abstract:Using insect female pheromone to disrupt their mating process is a new technology, which has been applied as environment friendly pesticides in agricultural and forestry industries to control pest infestation. Dodecanol (C(12)OH), as one of the simple pheromone component, was chosen and encapsulated as core material using gelatin (GE) and acacia gum (AG) as wall materials via complex coacervation. Through variations in capsule preparations, particularly the crosslinking density of the wall materials, release controllability of C(12)OH was studied. A series of C(12)OH-containing capsules were prepared with different concentrations of GE and AG and different crosslinkings. Crosslinking and C(12)OH encapsulation were enhanced when more crosslinker, either formaldehyde or glutaraldehyde, were used. At same level of crosslinker, lower crosslinking and higher C(12)OH encapsulation were obtained in microcapsules done with formaldehyde than those with glutaraldehyde. Constant release of C(12)OH was achieved in capsules prepared with glutaraldehyde. Mechanisms of C(12)OH release were discussed based on the results. Sustainable and effective antimicrobial surface based on cellulose thin films

Abstract:In the present work, we developed a sustainable and effective antimicrobial surface film based on Micro-Fibrillated Cellulose. The resulting porous cellulose thin film is barely noticeable to human eyes due to its sub-micron thickness, of which the coverage, porosity and microstructure can be modulated by the formulations developed. Using goniometers and a quartz crystal microbalance (QCM), we observed a threefold reduction in water contact angles and accelerated (more than 50% faster) water evaporation kinetics on the cellulose film. The thin film exhibits not only a rapid inactivation effect against SARS-CoV-2 in 5 minutes, following deposition of the virus loaded droplets, but also an exceptional ability to reduce contact transfer of liquid, e.g. respiratory droplets, onto surfaces such as artificial skin by more than 90%. It also exhibits excellent antimicrobial performance in inhibiting the growth of both gram-negative and gram-positive bacteria (<a class="link-external link-http" href="http://E.coli" rel="external noopener nofollow">this http URL</a> and <a class="link-external link-http" href="http://S.epidermidis" rel="external noopener nofollow">this http URL</a>) due to the excellent porosity and hydrophilicity. Additionally, the cellulose film shows nearly 100% resistance to skin scraping in dry condition thanks to its strong attachment to the substrate, whilst good removability once wetted, suggesting its practical suitability for daily use. Importantly, the coating can be formed on solid substrates readily by spraying and requires solely a simple formulation of a plant-based cellulose material with no additives, rendering it a scalable, affordable and green solution for antimicrobial surfaces. Implementing such cellulose films could thus play a significant role in controlling future pan- and epidemics, in particularly during the first phase when appropriate medication needs to be developed.

Abstract:The self‐assembly of lignin and cationic surfactants into nanoparticles loaded with emamectin benzoate exhibits good sustained release effect, pH sensitive release attributes, and a better mortality rate against model pests compared to commercial emulsions. Nanoformulation was an important strategy to reduce the use of pesticides and the impacts on the environment. However, complex process and high cost hurdled the application in agriculture. In this work, sodium lignosulfonate (SL) and cetyltrimethyl ammonium chloride (CTAC) were combined into nanoparticles and utilized as carriers for emamectin benzoate (EB), using a one‐pot method at mild conditions. The structure of nanoparticle was investigated by dynamic light scattering, ζ‐potential measurement, Fourier transform infrared spectroscopy, and transmission electron microscopy, and related to the loading and release properties. The mass ratio of SL and CTAC had great impacts on the structure of nanoparticle. The release of EB from SL‐CTAC nanoparticles was pH‐triggered. The retarding efficacy was enhanced in the medium of higher pH, due to the increased interaction between the EB and carrier. The incorporation into the nanoparticles greatly increased the insecticidal toxicity of EB. The median lethal concentration, against Spodoptera litura Fabricius larvae, of typical nanoformulation was 55% and 63% of that of conventional formulation emulsifiable concentrates after feeding for 24 and 48 h, respectively. The results indicated SL‐CTAC nanoparticles were good candidates of nanovehicles with high efficiency and low‐cost, which was of great significance to prompt the nanopesticides from laboratory investigation to agricultural application.

Abstract:Efficient utilization of pesticides has presented various advantages in the ecological environment, food safety, and agricultural sustainability. Pesticide microcapsule could significantly improve the pesticide utilization ratio, and thus, we prepared a sustained‐release microcapsule from cellulose nanocrystal (CNCs) stabilized Pickering emulsion template. The NaCl was added to overcome the strong electrostatic repulsion among CNCs for forming a stable oil‐in‐water (O/W) CNCs‐based Pickering emulsion template, followed by crosslinking with hexamethylene diisocyanate on the interface. UV–vis and zeta‐potential were used to characterize the as‐prepared Pickering emulsion, and the stability and emulsion fraction were calculated. Results indicated that NaCl contributed to the stability of Pickering emulsion, even at a low concentration. The obtained CNCs‐based microcapsule was characterized by FT‐IR, XRD, SEM, TG‐DTG, and three‐phase contact angles. The microcapsule presented an encapsulation efficiency of 96.76% and sustained release of the imidacloprid for more than 82 h. Results indicated the potential for loading and sustained release of imidacloprid, which is beneficial to the environment, food, and agriculture.
