Abstract:Phytosomes are referred to as novel compounds containing lipophiliccomplexes of components of different plants which are composed of glycerophospholipids. In the novel drug delivery system, it is a need of the hour to search for such a novel delivery system that can provide more bioavailability and efficacy for any extract containing phytoconstituents or Phyto extracts. The present study data were collected from extensive literature surveys from different esteemed scientific repositories such as Pubmed, Science direct, Research gate, Cochrane, Google scholar, Willey, Core, 1Library, etc. Many phytoconstituents that are obtained from nature have high biological activities in many chronic diseases and has the best therapeutic efficacy. The main advantage of using phytoconstituents is that it does not have most adverse side effects. Phytosome is used for phytopharmaceutical formulations, herbal formulations, and in Ayurveda. It improves the bioavailability of medicaments and absorption of drug molecules. It has a better pharmacokinetic and pharmacodynamic profile as compared to the normal traditional medicinal plant extracts. Abstract:Phytosomes (phytophospholipid complex) are dosage forms that have recently been introduced to increase the stability and therapeutic effect of herbal medicine. Currently, bioactive herbs and the phytochemicals they contain are considered to be the best remedies for chronic diseases. One promising approach to increase the efficacy of plant-based therapies is to improve the stability and bioavailability of their bio-active ingredients. Phytosomes employ phospholipids as their active ingredients, and use their amphiphilic properties to solubilize and protect herbal extracts. The unique properties of phospholipids in drug delivery and their use in herbal medicines to improve bioavailability results in significantly enhanced health benefits. Review on Niosome-A Future of Targeted Drug Delivery System

Abstract:Niosome are non-ionic surfactant vesicles obtained on hydration of synthetic nonionic surfactants, with or without incorporation of cholesterol or their lipids. They are vesicular systems similar to liposomes that can be used as carriers of amphiphilic and lipophilic drugs. Niosome appears to be a well preferred drug delivery system over liposome as niosome being stable and economic. Also niosomes have great drug delivery potential for targeted delivery of anti-cancer, anti-infective agents. Niosomes can entrap both hydrophilic and lipophilic drugs and can prolong the circulation of the entrapped drug in body. Encapsulation of drug in vesicular system can be predicted to prolong the existence of drug in the systemic circulation and enhance penetration into target tissue, perhaps reduce toxicity if selective uptake can be achieved. This review article focuses on the advantages, disadvantages, preparation methods, factors affecting, characterizations, mechanism of action, and applications of noisome.

Abstract:Abstract: Phytoconstituents have been widely used since ancient times to form a complex with phospholipids due to their various therapeutic actions. Despite having strong pharmacodynamic efficiency, numerous phytoconstituents have shown lower in vivo bioavailability and few adverse effects. Phytochemicals soluble in water exhibit poor absorption, leading to a limited therapeutic impact. Phytosome nanotechnology overcomes this limitation by creating a bound of phytochemicals with phospholipids. This method exhibits improved absorption because phytosomes inhibit significant herbal extract components from being degraded by gastric juices and gut flora. This improves bioavailability, increases clinical benefit, and ensures delivery to tissues without compromising nutritional stability. This review also aims to highlight those vesicular systems that could be used in phytosome technology. Additionally, this review highlights the preparation, advantage, characterization, applications, and recent development of phytosome and ethosome with a list of recent patents and marketed formulations and their uses.

Abstract:Phytopharmaceuticals are the newly termed herbal medicine, which includes standardized extract, bioactive fraction, and phytoconstituent. They have been practiced to cure, treat, and mitigate diseases. Phytopharmaceuticals have many health benefits, but their therapeutic efficacy is limited due to poor absorption, low bioavailability, and early elimination profile. A novel phospholipid complex is a newly introduced patented technology developed to incorporate the standardized plant extracts/fractions or water-soluble phytoconstituents into phospholipids to produce lipid compatible molecular complex, called phytosome, which improves their absorption and bioavailability. In herbal formulations, phytosome is the most advanced dosage form that has an upgraded absorption rate and enhanced pharmacokinetics compared with conventional products. The phospholipid complex results from hydrogen bonding between phospholipids and phytoconstituents, offering the maximum incorporation of herbal active ingredients into the lipidic layer and core. The increased therapeutic efficacy is due to the formation of amphiphilic phospholipid-complex of herbal medicine. This review highlights the role of phospholipids on delivery of herbal bioactives and natural extracts with particular emphasis on phytosomes. Moreover, the status of bioavailabilities, commercial products, patents, and clinical trials of phytosomal systems of phytopharmaceuticals were addressed.

Abstract:The herbal drug are used extensively for the treatment of the various diseases by the globe. The minimum side effects of medicinal plants attract people for its medicinal uses. The conventional dosage form of herbal medicine has certain limitation like low absorption, reduced bioavailability and lower penetration across biological membrane, it decrease their uses. Hence the introduction of novel drug delivery system namely Phyto-Phospholipid Complexes techniques resolve all these issues of herbal extract or plant actives. This advance technology technique reduced dose to produce desired therapeutic effect, improved stability due to chemical linkage, ability to permeate through skin systematic targeting to transit from hydrophilic to lipophilic environment and improved pharmacokinetic parameters. This review summarizes the latest investigations regarding the possible application of Phytosomes complexes for therapy of different dieseases, their marketed formulation, mechanism of transportation through phytosome and future prospect. The prospectus of Phytosomes technique can suggest new directions and endless frontier as novel drug treatment.

Abstract:Phytochemicals are compounds found in plants that have various biological activities and health benefits. Although phytochemicals have diverse therapeutic applications, they confront several challenges, such as poor solubility, instability, and low bioavailability. Phytosomes are used to overcome those challenges. The phytosome is a complex of phytochemicals and phospholipids that transports the drug to the target site, thereby increasing phytochemical absorption and bioavailability. The present study focuses on phytosome preparation methods and evaluation parameters, as well as the role of phytosomes in various ailments such as COVID-19, pulmonary fibrosis, asthma, migraine, arthritis, obesity, neuroprotective, antioxidant, anti-inflammatory, cancer, diabetes, metabolic syndrome, hyperlipidemic, and antimicrobial, which demonstrates phytosome complexes are more potent when compared to free extracts. Due to poor absorption and metabolism, phytoconstituents may not be effective in their free form. Phytosomes make phytoconstituents more bioavailable, stable, and effective. It also discusses recent formulations of phytosomes that can act as an effective or alternative regimen for various health conditions.

Nevertheless, the lipid core of phytosomes enhances their permeability across these barriers, thereby augmenting the bioavailability and efficacy of enclosed plant components. Cancer remains a prominent global cause of mortality, posing significant challenges for existing treatments, which often entail serious side effects associated with conventional approaches like surgery, radiation therapy, and chemotherapy. In exploring alternative strategies, the utilization of herbal active compounds in cancer treatment shows considerable promise in synergizing with conventional methods. While natural plant-derived active components demonstrate robust in vitro pharmacological activity, their in vivo absorption is often limited. Addressing this constraint, phytosomes emerge as a burgeoning nanotechnology offering a solution by enhancing the adaptability of bioactive phytoconstituents within lipid-rich barriers. This study delves into a meta-analysis of the physicochemical attributes of phytosomes, elucidating their structural intricacies. Abstract:Herbal medicine and phytochemicals have always been great therapeutic options to treat various diseases due to their compatibility, safety, easy production, low cost and side effects. However, low absorption rate, and poor bioavailability are notable challenges that have limited the use of most phytoconstituents. To address these challenges, different drug delivery methods have been proposed for generating operative carrier systems for phytochemicals. Among them, transdermal drug delivery (TDD) using nanophytosome has been introduced as a promising candidate for the delivery of insoluble phytoconstituents. Phytosomes can overcome skin barriers, and are therefore effective carriers for herbal medicines. They are generally produced by combining phytoconstituent agents in medicinal plant extracts with phospholipid molecules. They have improved the clinical applications of phytoconstituents such as polyphenols by enhancing their bioavailability and uptake. In this review, the main benefits of transdermal delivery systems, advantages of nanophytosome over other lipid nanoparticles in TDD of phytochemicals, and methods of phytosome preparation and characterization are briefly reviewed.

Abstract:Phenylethanoid glycosides (PhGs) are generally water-soluble phenolic compounds that occur in many medicinal plants. Until June 2020, more than 572 PhGs have been isolated and identified. PhGs possess antibacterial, anticancer, antidiabetic, anti-inflammatory, antiobesity, antioxidant, antiviral, and neuroprotective properties. Despite these promising benefits, PhGs have failed to fulfill their therapeutic applications due to their poor bioavailability. The attempts to understand their metabolic pathways to improve their bioavailability are investigated. In this review article, we will first summarize the number of PhGs compounds which is not accurate in the literature. The latest information on the biological activities, structure-activity relationships, mechanisms, and especially the clinical applications of PhGs will be reviewed. The bioavailability of PhGs will be summarized and factors leading to the low bioavailability will be analyzed. Recent advances in methods such as bioenhancers and nanotechnology to improve the bioavailability of PhGs are also summarized. The existing scientific gaps of PhGs in knowledge are also discussed, highlighting research directions in the future.

Abstract:In recent years, due to their distinctive and adaptable therapeutic effects, many natural bioactive compounds have been commonly used to treat diseases. Their limited solubility, low bioavailability, inadequate gastrointestinal tract stability, high metabolic rate, and shorter duration of action limited their pharmaceutical applications. However, those can be improved using nanotechnology to create various drug delivery systems, including lipid-based nanoparticles, to adjust the compounds' physicochemical properties and pharmacokinetic profile. Because of the enormous technical advancements made in the fundamental sciences and the physical and chemical manipulation of individual atoms and molecules, the subject of nanotechnology has experienced revolutionary growth. By fabricating certain functionalized particles, nanotechnology opens an innovative horizon in research and development for overcoming restrictions, including traditional medication administration systems. Nanotechnology-driven bioactive compounds are certain to have a high impact and clinical value for current and future uses. Lipid-based nanotechnologies were shown to deliver a range of naturally occurring bioactive compounds with decent entrapment potential and stability, a successfully controlled release, increased bioavailability, and intriguing therapeutic activity. This review outlines bioactive compounds such as paclitaxel, curcumin, rhodomyrtone, quercetin, kaempferol, resveratrol, epigallocatechin-3-gallate, silymarin, and oridonin, fortified within either a natural or synthetic lipid-based drug delivery system based on nanotechnology and their evaluation and clinical considerations.

Abstract:The utilization of medicinal plant extracts in therapeutics has been hindered by various challenges, including poor bioavailability and stability issues. Nanovesicular delivery systems have emerged as promising tools to overcome these limitations by enhancing the solubility, bioavailability, and targeted delivery of bioactive compounds from medicinal plants. This review explores the applications of nanovesicular delivery systems in antibacterial and anticancer therapeutics using medicinal plant extracts. We provide an overview of the bioactive compounds present in medicinal plants and their therapeutic properties, emphasizing the challenges associated with their utilization. Various types of nanovesicular delivery systems, including liposomes, niosomes, ethosomes, and solid lipid nanoparticles, among others, are discussed in detail, along with their potential applications in combating bacterial infections and cancer. The review highlights specific examples of antibacterial and anticancer activities demonstrated by these delivery systems against a range of pathogens and cancer types. Furthermore, we address the challenges and limitations associated with the scale-up, stability, toxicity, and regulatory considerations of nanovesicular delivery systems. Finally, future perspectives are outlined, focusing on emerging technologies, integration with personalized medicine, and potential collaborations to drive forward research in this field. Overall, this review underscores the potential of nanovesicular delivery systems for enhancing the therapeutic efficacy of medicinal plant extracts in antibacterial and anticancer applications, while identifying avenues for further research and development.

Abstract:Over thousands of years, natural bioactive compounds derived from plants (bioactive phytocompounds, BPCs) have been used worldwide to address human health issues. Today, they are a significant resource for drug discovery in the development of modern medicines. Although many BPCs have promising biological activities, most of them cannot be effectively utilized in drugs for therapeutic applications because of their inherent limitations of low solubility, structural instability, short half-life, poor bioavailability, and non-specific distribution to organs. Researchers have utilized emerging nanoformulation (NF) technologies to overcome these limitations as they have demonstrated great potential to improve the solubility, stability, and pharmacokinetic and pharmacodynamic characteristics of BPCs. This review exemplifies NF strategies for resolving the issues associated with BPCs and summarizes recent advances in their preclinical and clinical applications for imaging and therapy. This review also highlights how innovative NF technologies play a leading role in next-generation BPC-based drug development for extended therapeutic applications. Finally, this review discusses the opportunities to take BPCs with meaningful clinical impact from bench to bedside and extend the patent life of BPC-based medicines with new formulations or application to new adjacent diseases beyond the primary drug indications.

Abstract:In order to cure a range of ailments, scientists have investigated a number of bioactive antioxidant compounds produced from natural sources. Mangiferin, a C-glycosyl xanthone-structured yellow polyphenol, is abundant in mangoes and other dietary sources. In-depth examinations found that it is effective in the treatment of a variety of disorders due to its antiviral, anti-inflammatory, antiproliferative, antigenotoxic, antiatherogenic, radioprotective, nephroprotective, antihyperlipidemic, and antidiabetic properties. However, it is recognised that mangiferin's poor bioavailability, volatility, and limited solubility restrict its therapeutic usefulness. Over time, effective solutions to these problems have arisen in the shape of effective delivery methods. The current articles present a summary of the several researches that have updated Mangiferin's biopharmaceutical characteristics. Additionally, strategies for enhancing the bioavailability, stability, and solubility of this phytodrug have been discussed. This review provides detailed information on the development of innovative Mangiferin delivery methods such as nanoparticles, liposomes, micelles, niosomes, microspheres, metal nanoparticles, and complexation, as well as its therapeutic applications in a variety of sectors. This article provides effective guidance for researchers who desire to work on the formulation and development of an effective delivery method for improved magniferin therapeutic effectiveness.

Abstract:Plant-based medicines have received a lot of attention in recent years. Such medicines have been employed to treat medical conditions since ancient times, and in those times only the observed symptoms were used to determine dose accuracy, dose efficacy, and therapy. Rather than novel formulations, the current research work on plant-based medicines has mostly concentrated on medicinal active phytoconstituents. In the past recent decades, however, researchers have made significant progress in developing "new drug delivery systems" (NDDS) to enhance therapeutic efficacy and reduce unwanted effects of bioactive compounds. Nanocapsules, polymer micelles, liposomes, nanogels, phytosomes, nano-emulsions, transferosomes, microspheres, ethosomes, injectable hydrogels, polymeric nanoparticles, dendrimers, and other innovative therapeutic formulations have all been created using bioactive compounds and plant extracts. The novel formulations can improve solubility, therapeutic efficacy, bioavailability, stability, tissue distribution, protection from physical and chemical damage, and prolonged and targeted administration, to name a few. The current study summarizes existing research and the development of new formulations, with a focus on herbal bioactive components.

Abstract:Secondary metabolites, also known as phytochemicals, natural products or plant constituents are responsible for medicinal properties of plants to which they belong. The role they play in the plant is not, to date, well known or understood, but it may be beyond the protection. Their classification is based on chemical structure, composition, their solubility in various solvents, or the pathway by which they are synthesized. The main classification system includes three major groups: terpenoids, alkaloids and phenolics. For each one, we find subclasses with complexity in structure. In this review, we deal with the description of second metabolites, their biosynthesis, function, and the current pharmacological findings. Natural products are an important source of drug candidates in pharmaceutical industry, more deeply we understand them, the easier it is for scientists to intervene in alleviating different kind of diseases. The recent references have been consulted for presenting updated information, but also showing the new potentialities of plant second metabolites in drug research and development.

Abstract:Several biologically active secondary metabolites from aquatic plants have been extracted and identified using modern instrumental BioTechniques and used in various ways as flavors, food, additives, coloring agents, nutraceuticals, cosmetics, and also as unique source of pharma industries for the discovery or development of new drugs. From algae to aquatic macrophytes belonging to various categories, aquatic plants produce a variety of compounds such as polyketides, peptides, alkaloids, flavonoids, phenolic compounds, terpenes, steroids, quinones, tannins, coumarins, and essential oils commercially involving in antibiotic, antiviral, antioxidant, antifouling, anti-inflammatory, anticancer, cytotoxic, and antimitotic activities; thus making them a rich source of medicinal compounds. Moreover, they are comprehensively used in human therapy, veterinary, agriculture, scientific research, and in countless areas. Importantly these chemicals are exercised for developing new antimicrobial and cancer drugs. Furthermore, antioxidant molecules in aquatic plants and seaweeds have recently been acknowledged. This review contains a consolidated contemporary document consisting of entire knowledge available on pharmaceutical products of aquatic plants and highlights major differences among secondary metabolites found in aquatic (algae) and terrestrial plants.
