Abstract:In the last decades, silk fibroin and wool keratin have been considered functional materials for biomedical applications. In this study, fabrics containing silk fibers from Bombyx mori and Tussah silk fibers from Antheraea pernyi, as well as wool keratin fabrics, were grafted with phosmer CL and phosmer M (commercial names, i.e., methacrylate monomers containing phosphate groups in the molecular side chain) with different weight gains. Both phosmers were recently proposed as flame retarding agents, and their chemical composition suggested a possible application in bone tissue engineering. IR and Raman spectroscopy were used to disclose the possible structural changes induced by grafting and identify the most reactive amino acids towards the phosmers. The same techniques were used to investigate the nucleation of a calcium phosphate phase on the surface of the samples (i.e., bioactivity) after ageing in simulated body fluid (SBF). The phosmers were found to polymerize onto the biopolymers efficiently, and tyrosine and serine underwent phosphorylation (monitored through the strengthening of the Raman band at 1600 cm-1 and the weakening of the Raman band at 1400 cm-1, respectively). In grafted wool keratin, cysteic acid and other oxidation products of disulphide bridges were detected together with sulphated residues. Only slight conformational changes were observed upon grafting, generally towards an enrichment in ordered domains, suggesting that the amorphous regions were more prone to react (and, sometimes, degrade). All samples were shown to be bioactive, with a weight gain of up to 8%. The most bioactive samples contained the highest phosmers amounts, i.e., the highest amounts of phosphate nucleating sites. The sulphate/sulphonate groups present in grafted wool samples appeared to increase bioactivity, as shown by the five-fold increase of the IR phosphate band at 1040 cm-1.
Tyrosinase-catalyzed modification of Bombyx mori silk fibroin: Grafting of chitosan under heterogeneous reaction conditions

Abstract:The capability of mushroom tyrosinase to catalyze the oxidation of tyrosine residues of Bombyx mori silk fibroin was studied under heterogeneous reaction conditions, by using a series of silk substrates differing in surface and bulk morphology and structure, i.e. hydrated and insoluble gels, mechanically generated powder and fibre. Tyrosinase was able to oxidize 10-11% of the tyrosine residues of silk gels. The yield of the reaction was very low for the powder and undetectable for fibres. FT-Raman spectroscopy gave evidence of the oxidation reaction. New bands attributable to vibrations of oxidized tyrosine species (o-quinone) appeared, and the value of the I853/I829 intensity ratio of the tyrosine doublet changed following oxidation of tyrosine. The thermal behaviour of SF substrates was not affected by enzymatic oxidation. o-Quinones formed by tyrosinase onto gels and powder were able to undergo non-enzymatic coupling with chitosan. FT-IR and FT-Raman spectroscopy provided clear evidence of the formation of silk-chitosan bioconjugates under heterogeneous reaction conditions. Chitosan grafting caused a beta-sheet --> random coil conformational transition of silk fibroin and significant changes in the thermal behaviour. Chitosan grafting did not occur, or occurred at an undetectable level on silk fibres. The results reported in this study show the potential of the enzymatically initiated protein-polysaccharide grafting for the production of a new range of bio-based, environmentally friendly polymers.

Abstract:Silk fibroin (SF) has the characteristics of non-toxicity, good biocompatibility and low immunogenicity, and exhibits many potential applications in biomaterial fields. In the present work, chemical phosphorylation of SF was carried out via the Maillard reaction using D-glucose-6-phosphate (GP), aiming at increasing the number of binding-sites for calcium ion and promoting the biomimetic mineralization of SF films. Changes in the molecular weight of SF before and after GP grafting were analyzed by SDS-PAGE and SEC chromatogram. Structures, morphologies, and elemental compositions for the fibroin-based films before and after mineralization were evaluated by the means of EDS, SEM, FTIR, and XRD. The results indicated that GP was successfully grafted onto SF surfaces, companying with a slight decrease in the molecular weight. Biomimetic mineralization led to a noticeable deposition of hydroxyapatite (HAp) on the film of SF-g-GP, and higher contents of calcium and phosphorous were detected on the mineralized material, revealing that phosphorylation promoted the mineralization processing. Meanwhile, biocompatibility of the obtained SF-g-GP/HAp was above the acceptable level according to ISO 10993-5-2009. The present work provides an alternative to prepare the fibroin-based bone repair materials.

Abstract:Enzymatic crosslinking of tyrosine is a simple and modular method for adding functional peptides to silk fibroin (SF) hydrogels. Silk fibroin is a naturally derived polymer notable for its robust mechanical properties, biological compatibility, and versatility. Hydrogels fabricated from SF are elastic, optically clear, and have tunable moduli, however, they do not contain native biological epitopes for cell interactions. In this work we demonstrate the attachment of peptides to SF hydrogels through crosslinking of tyrosine with horseradish peroxidase (HRP) and hydrogen peroxide (H2O2). The goal was to understand the utility of this approach and to study how the addition of peptides affects the SF material properties. SF hydrogels conjugated to model peptides with different molecular weights and hydrophobic properties were studied by liquid chromatography/tandem mass spectroscopy (LC–MS/MS) (bond formation), fluorescent imaging (spatial distribution), Fourier transform infrared spectroscopy (FTIR) (protein secondary structure), and rheology (gelation time, modulus). As a proof of concept using a biologically relevant peptide, a peptide containing the cell binding domain Arg-Gly-Asp (RGD) was conjugated to SF, and the density and morphology of primary human fibroblasts were assessed. This work demonstrates a facile method for adding peptides to silk fibroin that can be adopted for a variety of biomaterials applications.

Abstract:The identification of appropriate mouse models could be useful in carefully evaluating the actual role of the in vivo development of antigen‐loss variants during antigen‐specific vaccine therapy of human tumors. In this study we investigated the level of efficacy of a MART‐1/Melan‐A‐specific CD8+ T cell clone against its autologous melanoma in a severe combined immunodeficiency (SCID) mouse model, in which the tumor cells expressed in vivo heterogeneous and suboptimal levels of MART‐1. The subcutaneous co‐injection of the MART‐1/Melan‐A‐reactive T cell clone A42 with MART‐1/Melan‐A+ autologous human melanoma cells into SCID mice caused a total inhibition of tumor growth. However, the systemic treatment with A42 clone lymphocytes resulted inonly 50–60% inhibition of tumor growth, although the T cell clone targeted the tumors and the MART‐1+ cells virtually disappeared from the tumors. This study suggests that an immunotherapybased on the expansion of an antigen‐specific T cell clone generated in vitro is highly efficient in abolishing tumor growth when the target antigen is fully expressed, but leads to in vivoimmunoselection of antigen‐loss variants in the presence of suboptimal levels of antigen expression. Furthermore, this work shows that human tumors/SCID mouse models may be useful in evaluating thein vivo efficacy of adoptive immunotherapies.

Abstract:Protein-polymer conjugates combine properties of biopolymers and synthetic polymers, such as specific bioactivity and increased stability, with great benefits for various applications from catalysis to biomedicine. Furthermore, polymer conjugation can mimic important posttranslational modifications of proteins such as glycosylation. There are typically two approaches to create protein-polymer conjugates: the protein is functionalized in advance with an initiator for a grafting-from method or a previously produced polymer is conjugated to the protein via a grafting-to method. In this study, we present a new approach that uses native proteins and allows for direct grafting-from using a thiol-induced, light-activated controlled radical polymerization (TIRP) that is initiated at thiols from specific cysteine residues of the protein. This straightforward method is employed to introduce polymers onto proteins and enzymes without any prior protein modifications, it works in aqueous buffer and maintains the protein's native structure and activity. The resulting protein-polymer conjugates exhibit high molar masses and low dispersities. We demonstrate the versatility of this approach by introducing different types of polymers such as hydrophilic poly(2-hydroxyethyl acrylate) (pHEAA), temperature-responsive poly(N-isopropylacrylamide) (pNIPAM) as well as glycopolymers mimicking the natural protein glycosylation and enabling selective interactions. We present successful combinations of the protein and polymer functions e.g., temperature-induced aggregation leading to an increase in enzyme activity and the introduction of artificial glycosylation inducing specific protein-protein cluster formation and giving straightforward access to glycosurfaces. Based on this straightforward, potentially scalable yet highly controlled synthesis of protein-polymer conjugates, various areas of applications are envisioned ranging from biomedicine to material sciences.

Functionalization of Silk Fibroin Electrospun Scaffolds via BMSC Affinity Peptide Grafting through Oxidative Self-Polymerization of Dopamine for Bone Regeneration

Abstract:Electrospun scaffolds have been broadly studied to enhance bone regeneration because of the ability to simulate the structure and biological functions of the extracellular matrix. Polydopamine (PDA) is used to coat various surfaces at a slightly basic pH (8-8.5) and spontaneously reacts with nucleophilic functional groups. It is suitable for surface modifications of scaffolds correlated with bone formation. E7 is a newly discovered peptide with specific affinity for bone marrow mesenchymal stem cells (BMSCs). It can be useful for recruiting stem cells. Here, electrospun silk fibroin (SF) scaffolds were fabricated, and PDA was used for surface modification followed by grafting E7 (SF-PDA-E7). These composite SF-PDA-E7 electrospun scaffolds improved hydrophilicity, facilitated cell proliferation and adhesion, and boosted the osteogenic differentiation of BMSCs by creating osteoinduction conditions under the synergistic effects of PDA and E7. Moreover, the scaffolds showed high efficiency for recruiting BMSCs induced by E7 both in vitro and in vivo, which was associated with the SDF-1α/CXCR4 axis and the p38, extracellular signal-related kinase, and Akt signal transduction pathways. These functionalized electrospun scaffolds promoted regeneration of bone in the rat calvarial bone defect model. In general, this study verified that PDA could be a simple and efficient method for surface modification, and E7-grafted PDA-modified SF electrospun scaffolds were suitable for bone tissue engineering.

Abstract:Chitosan is considered as a green additive with broad application prospects due to its advantages like biodegradability and antibacterial ability. Herein, we proposed an effective chitosan grafting approach via "one-enzyme double catalysis" strategy which aimed at functionalizing wool fibers to achieve bidirectionally multiple covalent crosslinking between chitosan and wool by laccase/2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) mediated oxidization. The mechanism was studied using models of wool and chitosan in terms of UV, FTIR, GPC and MALDI-TOF MS. Meanwhile, the structure and morphology of wool fiber grafted with chitosan were characterized by ATR-FTIR and SEM. Compared with untreated wool, this efficient method can significantly improve the dimensional stability to felting (2.53 %), wettability and dyeability of wool fabric, and can also compensate for the strength loss caused by the pretreatment. The present work provides a useful path for the enzymatic modification of keratin-containing fibers like wool using chitosan and other natural biopolymers with similar structure.

Abstract:Two biopolymers, silk fibroin (SF) and chitosan, were conjugated by tyrosinase (EC 1.14.18.1), a polyphenolic oxidase, to improve their physicochemical properties, such as their thermal properties and morphological stabilities in organic solvents. The crosslinking between SF and chitosan took place mainly through Michael addition reactions. A main reaction between the amino groups in chitosan ando-quinone, the oxidation product of the tyrosyl residue in SF, was confirmed by UV spectroscopy. Measurements of viscosity and light scattering indicated that the crosslinked SF/chitosan conjugate was compact: it had a smaller particle size because of tight bonding forces between the SF and chitosan molecular chains. Thermal decomposition of SF/chitosan conjugates crosslinked by tyrosinase occurred at higher temperatures. The adhesiveness of the SF/chitosan conjugates decreased steadily as the crosslinking reaction progressed. We propose that this new crosslinking method be used for the preparation of silk fibroin/chitosan conjugates using tyrosinase. We expect that SF/chitosan conjugates crosslinked by tyrosinase can be used preferentially in biomedical applications because of its unique properties and non-toxicity.

Abstract:Here we demonstrate a reaction sequence that can be used to quantitatively modify the tyrosine residues in silk protein from B. mori silkworms. A primary amine is installed ortho to the hydroxyl group on the tyrosine ring using a diazonium coupling reaction followed by reduction of the azo bond. The resulting amine is then acylated using carboxylic acid or NHS‐ester derivatives at room temperature and neutral pH conditions. The silk derivatives are characterized using 1H NMR, UV/vis spectroscopy, ATR‐FTIR, and a unique method to follow this reaction sequence using isotopically‐labeled reagents and 2D NMR spectroscopy is also employed. We further demonstrate that this sequence can be used to install alkyne or azide functional groups which can undergo further bio‐orthogonal cycloaddition reactions under mild conditions. Abstract:Small-diameter vascular grafts made of biomedical polytetrafluoroethylene (PTFE) suffer from the poor long-term patency rate originating from thrombosis and intimal hyperplasia, which can be ascribed to the insufficient endothelialization and chronic inflammation of the materials. Hence, bio-functionalization of PTFE grafts is highly desirable to circumvent these disadvantages. In this study, a versatile "implantation-incubation" approach in which the biomedical PTFE is initially modified by plasma immersion ion implantation (PIII) is described. After the N2 PIII treatment, the surface of biomedical PTFE is roughened with nanostructures and more importantly, the abundant free radicals generated underneath the surface continuously migrate to the surface and react with environmental molecules. Taking advantage of this mechanism, various biomolecules with different functions can be steadily immobilized on the surface of PTFE by simple solution immersion. As examples, three typical biomolecules, heparin, SDF-1α, and CD47, are covalently grafted onto the PTFE. In addition to retaining the bioactivity, the surface-functionalized PTFE exhibits reduced thrombogenicity, facilitates the recruitment of endothelial progenitor cells, and even alleviates the inflammatory immune responses of monocytes-macrophages and is thus promising to the development of small-diameter prosthetic vascular grafts with good long-term patency.

An environment-friendly chemical modification method for thiol groups on polypeptide macromolecules to improve the performance of regenerated keratin materials

Abstract:The R&D and application of natural biomass resources is one of the important ways to solve the world's energy shortage and environmental pollution. However, the complex processing technology and poor mechanical properties seriously hinder the development of the biomass polymers. A kind of natural antioxidant, alpha-lipoic acid, was used to modify wool keratin by grafting thiols in this study, so as to ameliorated the physical properties of regenerated materials. The optimal process parameters and modification mechanism were explored. Research results indicated that grafting ratio of lipoic acid can reach more than 13% and the polypeptide chains were more stretched in solution, when 30% lipoic acid was added into keratin solution at 40℃ with a reaction time of 2h. In addition, lipoic acid grafted on thiol groups can effectively weaken the intra- and intermolecular forces of keratin, this change made the arrangement of macromolecular chains more orderly. The physical structure of modified keratin became uniform and compact, and the materials had obtained better thermal and mechanical properties. This research provided a novel and environment-friendly method for the modification technology of keratin, and it was beneficial to expand the application fields of biomass materials.

Facile Fabrication of Cross-Linked Fluorescent Organic Nanoparticles with Aggregation-Induced Emission Characteristic Via the Thiol-Ene Click Reaction and Their Potential for Biological Imaging.

Abstract:Over the past several years, the biomedical applications of fluorescent organic nanoparticles (FONs) with aggregation-induced emission (AIE) feature have been extensively explored because the AIE-active FONs could effectively overcome the aggregation caused quenching (ACQ) effect of FONs based on conventional organic dyes. The development of novel methods for synthesis of AIE-active FONs plays a centre role for their biomedical applications. In this work, we reported a facile one-step thiol-ene click reaction for fabrication of AIE-active FONs through conjugation of acrylated PEG and AIE-active tetraphenylethylene (TPE) with two ene bonds using pentaerythritol tetra(3-mercaptopropionate) as the linkage. The successful synthesis of TPE containing AIE-active copolymers was evidenced by various characterization techniques. The particle size and fluorescence properties of the resultant TPE-S-PEG copolymers were evaluated by transmission electronic microscopy and fluorescence spectroscopy. Moreover, the cell viability and cell uptake behavior was also examined to evaluate their potential for biological imaging. We demonstrated that the cross-linked TPE-S-PEG show small size, high water dispersibility, low cytotoxicity and strong fluorescence for tracing. All of these advantages endow the TPE-S-PEG FONs great potential for biological imaging applications. Furthermore, this novel click reaction can take place under mild experimental conditions with high efficiency. It could be also further expanded for preparation of multifunctional AIE-active materials due to the universality of the thiol-ene click reaction and good precursor applicapability. Taken together, we have developed a novel and effective thiol-ene click reaction to fabricate the cross-linked AIE-active FONs, which display excellent physicochemical and biological properties and are promising for biomedical applications.

Abstract:Synthetic methods inspired by Nature often offer unique advantages including mild conditions and biocompatibility with aqueous media. Inspired by an ergothioneine biosynthesis protein EgtB, a mononuclear non-haem iron enzyme capable of catalysing the C-S bond formation and sulfoxidation, herein, we discovered a mild and metal-free C-H sulfenylation/intramolecular rearrangement cascade reaction employing an internally oxidizing O-N bond as a directing group. Our strategy accommodates a variety of oxyamines with good site selectivity and intrinsic oxidative properties. Combining an O-N bond with an X-S bond generates a C-S bond and an S=N bond rapidly. The newly discovered cascade reaction showed excellent chemoselectivity and a wide substrate scope for both oxyamines and sulfenylation reagents. We demonstrated the biocompatibility of the C-S bond coupling reaction by applying a coumarin-based fluorogenic probe in bacterial lysates. Finally, the C-S bond coupling reaction enabled the first fluorogenic formation of phospholipids, which self-assembled to fluorescent vesicles in situ.

Abstract:Silk fibroin (SF) is a natural protein polymer and promising biomaterial. Chemical modifications have attracted growing interest in expanding SF applications. However, the majority of amino acid residues in SF are non-reactive and most of the reactive ones are in the crystalline region. Herein, a modification was conducted to investigate the possibility of direct modification on the surface of natural SF by a reagent with a mild reactivity, the type and quantity of the residues involved in the reactions, and the structural changes upon modification. Infrared spectrum, 1H NMR, titration and amino acid analyses, X-ray diffraction, and hemolysis test were used to analyze the materials. The results showed that sulfonic acid groups were grafted onto SF and the reaction occurred mainly at serine residues through hydroxyl groups. In total, 0.0958 mmol/g of residues participated in the modification with a modification efficiency of 7.6%. Moreover, the crystallinity and the content of β-sheet structure in SF increased upon modification. The modified material had good blood-compatibility. In conclusion, surface modification on native SF through serine residues was practicable and had the advantage of increased β-sheet structure. This will provide an alternative way for the modification of fibroin for the desired application in the biomedical field.

Eco-friendly Grafting of Chitosan as a Biopolymer onto Wool Fabrics Using Horseradish Peroxidase

Abstract:Chitosan and enzymes have been extensively used in modification of wool, due to their non-toxic and eco-friendly characteristics, as well as to reduce pollution in textile production. In the present work, chitosan was grafted onto wool fabrics using horseradish peroxidase (HRP) in order to endow wool with outstanding properties. The reaction mechanism, grafting percentage, and properties of chitosan-grafted pretreated wool fabric including wettability, dyeability, shrink resistance, and antibacterial activity were studied. P-hydroxyphenylacetamide (PHAD), as a model compound for tyrosine residues in wool, was used to study the mechanism of HRP-mediated grafting of chitosan onto wool. UV and FTIR analyses indicated that quinones, which are intermediate of HRP/H2O2-catalyzed PHAD, reacted with the amino groups of chitosan by Schiff base or Michael addition reactions. Scanning electron microscopy (SEM) showed that dichloroisocyanuric acid pretreatment had an etching effect and some substances were deposited onto the wool fiber. SEM and Fourier transform infrared (FTIR) spectroscopy further confirmed the covalent grafting of chitosan onto wool. Compared with chitosan-grafted unpretreated wool, there was an obvious improvement in grafting efficiency of chitosan onto wool fabric, physical and mechanical properties after pretreatment and chitosan grafting on the wool. For chitosan-grafted pretreated wool fabrics, there was no observable change in the thermal behavior, while the hydrophilicity, dyeability, shrinkage resistance, and antibacterial activity were remarkably improved compared to that of untreated wool. The present work offers a new ecologically acceptable process of grafting chitosan on the wool.

Bioorthogonal Functionalization of Material Surfaces with Bioactive Molecules

Abstract:The functionalization of material surfaces with biologically active molecules is crucial for enabling technologies in life sciences, biotechnology, and medicine. However, achieving biocompatibility and bioorthogonality with current synthetic methods remains a challenge. We report herein a novel surface functionalization method that proceeds chemoselectively and without a free transition metal catalyst. In this method, a coating is first formed via the tyrosinase-catalyzed putative polymerization of a tetrazine-containing catecholamine (DOPA-Tet). One or more types of molecule of interest containing trans-cyclooctene are then grafted onto the coating via tetrazine ligation. The entire process proceeds under physiological conditions and is suitable for grafting bioactive molecules with diverse functions and structural complexities. Utilizing this method, we functionalized material surfaces with enzymes (alkaline phosphatase, glucose oxidase, and horseradish peroxidase), a cyclic peptide (cyclo[Arg-Gly-Asp-D-Phe-Lys], or c(RGDfK)), and an antibiotic (vancomycin). Colorimetric assays confirmed the maintenance of the biocatalytic activities of the grafted enzymes on the surface. We established the mammalian cytocompatibility of the functionalized materials with fibroblasts. Surface functionalization with c(RGDfK) showed improved fibroblast cell morphology and cytoskeletal organization. Microbiological studies with Staphylococcus aureus indicated that surfaces coated using DOPA-Tet inhibit the formation of biofilms. Vancomycin-grafted surfaces additionally display significant inhibition of planktonic S. aureus growth.

Abstract:Glycoproteins and their mimics are challenging to produce because of their large number of polysaccharide side chains that form a densely grafted protein-polysaccharide brush architecture. Herein a new approach to protein bioconjugate synthesis is demonstrated that can approach the functionalization densities of natural glycoproteins through oligosaccharide grafting. Global amino acid substitution is used to replace the methionine residues in a methionine-enriched elastin-like polypeptide with homopropargylglycine (HPG); the substitution was found to replace 93% of the 41 methionines in the protein sequence as well as broaden and increase the thermoresponsive transition. A series of saccharides were conjugated to the recombinant protein backbones through copper(I)-catalyzed alkyne-azide cycloaddition to determine reactivity trends, with 83-100% glycosylation of HPGs. Only an acetyl-protected sialyllactose moiety showed a lower level of 42% HPG glycosylation that is attributed to steric hindrance. The recombinant glycoproteins reproduced the key biofunctional properties of their natural counterparts such as viral inhibition and lectin binding.
