Abstract:The brown planthopper, Nilaparvata lugens Stål, has become a major threat in tropical Asian and China since the rice green revolution of the 1960s. Currently, insecticide application remains the primary choice for controlling this rice insect pest, but heavy use of insecticides poses dangerous risks to beneficial natural enemies and pollinators, and stimulates N. lugens reproductivity, and has caused a resurgence of the pest in the major rice-planting regions throughout Asia. Achieving the long-lasting goal of sustainable management of N. lugens requires understanding of the molecular basis of outbreaks of the pest and the development of environment-friendly pest-control strategies. Here, we review the recent molecular advances in N. lugens research on the aspects of its endosymbionts, virus transmission, insecticide resistance, and interaction between N. lugens and rice plants. We also put forward further research directions that may shed some lights on management of the rice pest.

Abstract:To meet the World’s food demand, there is a growing need for sustainable pest management practices. This study describes the results from complementary laboratory and field studies of a “banker plant system” for sustainable management of the rice brown planthopper (BPH) ( Nilaparvata lugens Stål) – the economically most important rice pest in Asian rice growing areas. The banker plant system consisted of planting a grass species, Leersia sayanuka , adjacent to rice fields. L. sayanuka is the host plant of a planthopper, Nilaparvata muiri . An egg parasitoid, Anagrus nilaparvatae , parasitizes eggs of both BPH and N. muiri , and its establishment and persistence are improved through plantings of L. sayanuka and thereby attraction of N. muiri . Laboratory results showed that BPH was unable to complete its life cycle on L. sayanuka , and N. muiri could not complete its life cycle on rice. Thus, planting L. sayanuka did not increase the risk of planthopper damage to rice fields. Field studies showed that BPH densities were significantly lower in rice fields with banker plant system compared to control rice fields without banker plant system.

Abstract:In this study,the brown planthopper( BPH),Nilaparvata lugens( Hemiptera: Delphacidae),cultured on Bt rice lines( T1C-19 with cry1 C gene and T2A-1 with cry2 A gene) and their parental rice MH63,were treated with a series of concentrations of triazophos and deltamethrin when they developed up to the third instar for the evaluation of the effects of Bt rice on BPH resurgence induced by pesticides. The results indicated that BPH nymphal duration decreased with the increasing concentration of deltamethrin,while no significant differences of BPH nymphal duration were observed on the same rice variety treated with the different concentrations of triazophos. Furthermore,the survival rate of BPH significantly declined and their fecundity significantly increased with the increasing concentration of insecticides. However,ecological fitness parameters of BPH including nymphal duration,survival rate,female adult mass,fecundity and egg hatchability were not significantly different among Bt and non-Bt rice varieties when they treated with the same concentration of insecticide. These results implied that no significant difference in the BPH resurgence induced by pesticides was observed among the two tested Bt rice varieties and their non-Bt parental rice variety.

Abstract:The brown planthopper, Nilaparvata lugens (Stal), is one of the most important insect pests on paddy rice in tropical and temperate Asia. Overuse and misuse of insecticides have resulted in the development of high resistance to many different insecticides in this pest. Studies were conducted to evaluate the change of resistance level to four insecticides over 15 generations without any exposure to insecticides in brown planthopper. After 15 generations' rearing without exposure to insecticide, brown planthopper could reverse the resistance to imidacloprid, chlorpyrifos, fipronil, and fenobucarb. The range and style of resistance reversal of brown planthopper differed when treated with four different insecticides. To monitor potential changes in insect physiological responses, we measured the activity of each of the three selected enzymes, including acetylcholinesterases (AChE), general esterases (EST), and glutathione S-transferases. After multiple generations' rearing without exposure to insecticide, AChE and EST activities of brown planthopper declined with the increased generations, suggesting that the brown planthopper population adjusted activities of EST and AChE to adapt to the non-insecticide environment. These findings suggest that the reducing, temporary stop, or rotation of insecticide application could be incorporated into the brown planthopper management.
Effects of Bt Rice with Cry1c and Cry2a on the Ecological Generation Fitness of Rice Brown Planthoppers (nilaparvata Lugens ) and Whitebacked Planthoppers (sogatella Furcifera)at Various Nitrogen Rates

Abstract:In order to evaluate the effects of Bt rice on the generation fitness of non-target pest at different nitrogen rates,as wall as the potential impact of Bt rice and nitrogen fertilizer levels on non-target pest,the brown plantthopper (BPH)(Nilaparvata lugens)and the whitebacked planthopper (WBPH)(Sogatella furcifera),were successively cultured for four generations on Bt rice,T1C-19 with cry1C and T2A-1 with cry2A ,and their parental rice Minghui 63,at nitrogen rates of 0,100 and 250 kg/hm2 .The results indicated that the ecological fitness of the BPH and WBPH was increased with the increasing nitrogen fertilizer level, including shortened nymph developmental duration, enhanced fecundity, prolonged longevity, increased nymph survival rate, female weight, egg hatchability and population growth rate.There were significant differences in rice planthopper nymphal development duration,body weight of female adult,fecundity and population growth rate for four successive generations under different nitrogen rates.Transgenic Bt rice,T1C-19 and T2A-1,had no significant effect on the fitness of BPH and WBPH at the same generations and nitrogen rates,except the BPH fecundity.BPH fecundity on transgenic Bt rice T1C-19 at the nitrogen levels of 100 kg/hm2 and 250 kg/hm2 (N)was lower compared with the transgenic Bt rice T2A-1 and its parent Minghui 63 from the first generation to the third generation,but no significant difference was found among varieties at the fourth generation at three nitrogen rates.No significant difference was found on the fitness of BPH and WBPH with the generation increase, between non-transgenic Bt rice and transgenic Bt rice.Therefore, the present study demonstrated that nitrogen fertilizer levels exerted a significant influence on the ecological fitness of the rice planthoppers with the generation increase,while transgenic Bt rice didn′t.

Abstract:Biochemical analysis of healthy and brown planthopper (BPH), Nilaparvata lugens damaged rice plants across three varieties, viz., Pusa 1509 and Pusa 1121 (Basmati varieties prevalent in cultivation in North India) and the susceptible variety TN-1 revealed substantial variations in the chlorophyll, carotenoid, protein, flavonoid contents and relative water content in infested plants. These alterations serve as vital indicators of pest severity, aiding in cost-effective pest management. Assessing the impact of BPH over 20 and 40 days after infestation (DAI) highlighted significant reductions in chlorophyll, carotenoid, and protein levels. Flavonoid content initially increased after infestation but decreased with high BPH stress after 40 DAI. Relative water content decreased, indicating sustained adverse effects of BPH on rice plants. These findings underscore the susceptibility of physiological traits to BPH stress, urging further research for sustainable pest management to ensure global food security.

Abstract:Background: Nanoparticles have been employed as nanopesticides for pest control in agriculture. However, the harmful effects of their chemical synthesis on human and environmental health have resulted in increased use of green synthetic approaches, including the use of plant extracts. The brown planthopper, Nilaparvata lugens (Stal) (BPH), is a severe pest of rice plants (Oryza sativa L.), especially in Asia. It is usually controlled chemically but has developed resistance against many insecticides.Results: In this study, we synthesized metallic silver (Ag-NPs) and copper-oxide (CuO-NPs) nanoparticles using the exogenous phytohormone, gibberellic acid (GA3), as a reducing agent. We then sprayed them separately on rice plants and BPH together and evaluated their effects on the plants and insects. SEM and TEM images showed that the synthesis was successful, indicated by the sizes (25-60 nm), uniform shape and spherical and cubical structures of Ag-NPs, as well as by the rugby sheet-like of CuO-NPs with lateral sizes of 150-340 nm and thickness of 30-70 nm. Independent applications of the nanoparticles and GA3 on rice plants induced different volatile profiles, of which the highest number emitted was under Ag-NPs, including the highest emission of linalool. Transcriptome analysis showed that Ag-NPs-treated rice plants showed different transcriptome profiles compared to the control, 24 h after treatment, including the upregulation of the linalool synthase gene, genes of plants transcription factors such as WRKY, bHLH and NAC and other genes involved in plant defense responses. In all treatments, the mortality rate of BPH increased with an increase in NPs concentrations over time but was prominent under Ag-NPs treatment. The LC50 values for Ag-NPs and CuO-NPs decreased with an increase in time. Also, the nanoparticles increased the activities of protective enzymes (POD, SOD and CAT), inhibited that of detoxification enzymes (A-CHE, ACP and AKP), and reduced total protein concentrations in the BPH.Conclusions: These results show that synthesizing nanoparticles using phytohormones may be a safer and envi-ronmentally friendly option, which also holds promise for controlling the BPH in rice production.

Abstract:The study was carried out to assess the reaction of different rice genotypes in response to brown planthopper (BPH)Nilaparvata lugens (Stål.) attack and the possible use of these genotypes in BPH management. The resistant genotypes PTB 33, ADT 45 and ASD 7 and the moderately resistant genotypes CO 43 and KAU 1661 recorded the lowest nymphal preference, fecundity, feeding rate, survival, growth index, population buildup and plant dry weight loss per mg of insect dry weight produced, and more unhatched eggs, longer nymphal development period, days to wilt and higher Functional Plant Loss Index compared with the susceptible genotype TN1. In resistant and moderately resistant genotypes, a greater accumulation of defense enzymes such as peroxidase and polyphenol oxidase in response toN. lugens infestation was recorded one day after infestation, and more pathogenesis-related protein and chitinase activity was noted 3 days after infestation. The activity was sustained for more than a week after infestation compared with the susceptible genotype TN1.

Selection of Global Metarhizium Isolates for the Control of the Rice Pest Nilaparvata Lugens (homoptera : Delphacidae)

Abstract:BACKGROUND: This study was initiated to search for fungal candidates for microbial control of brown planthopper (BPH) Nilaparvata lugens Stal, to which little attention has been paid in the past two decades.RESULTS: Thirty-five isolates of Metarhizium anisopliae (Metschnikoff) Sorokin and M. flavoviride Gams & Rozsypal from different host insects worldwide were bioassayed for their lethal effects against third-instar BPH nymphs at 25 degrees C and a 14:10 h light:dark photoperiod at ca 1000 conidia mm(-2). On day 9 post-treatment, mortality attributable to mycosis ranged from 6.5 to 64.2% and differed significantly among the tested isolates with no apparent relationship to their host origin. Only two BPH-derived M. anisopliae isolates from the Philippines (ARSEF456) and Indonesia (ARSEF576) killed > 50% of the nymphs. Both isolates were further bioassayed for time-concentration-mortality responses of the nymphs to the sprays of 19-29, 118-164 and 978-1088 conidia mm(-2) in repeated bioassays. The resultant data fitted a time-concentration-mortality model very well. Their LC50 values were estimated as 731 and 1124 conidia mm(-2) on day 7 and fell to 284 and 306 conidia mm(-2), respectively, on day 10.CONCLUSION: The two M. anisopliae isolates are potential biocontrol agents of BPH for further research. This is the first report of the lethal effects of global Metarhizium isolates on the rice pest. (C) 2008 Society of Chemical Industry.
Transcriptome and metabolome analyses reveal the responses of brown planthoppers to RH resistant rice cultivar

Abstract:The brown planthopper (BPH) Nilaparvata lugens (Stål) (Hemiptera: Delphacidae) is one of the most destructive rice pests in Asia. The application of insect-resistant rice cultivars is currently one of the principal means of controlling BPH. Understanding the physiological response mechanisms of BPH feeding on insect-resistant rice is the key for maintaining rice yield. Here, we measured the ecological fitness and analyzed the whole-body transcriptome and metabolome of BPH reared on susceptible cultivar Taichung Native 1 (TN1) and resistant cultivar Rathu Heenati (RH). Our results showed that RH significantly decreased the survival rate, female adult weight, honeydew secretion, the number of eggs laid per female and fat content of BPH. We identified 333 upregulated and 486 downregulated genes in BPH feeding on RH. These genes were mainly involved in energy metabolism, amino acid metabolism, hormone synthesis and vitamin metabolism pathways. We also detected 145 differentially accumulated metabolites in BPH reared on RH plants compared to BPH reared on TN1 plants, including multiple carbohydrates, amino acids, lipids, and some nucleosides. Combined analyses of transcriptome and metabolome showed that five pathways, including starch, sucrose, and galactose metabolism, were altered. The network for these pathways was subsequently visualized. Our results provide insights into the mechanisms of metabolite accumulation in BPH feeding on the RH rice variety. The results could help us better understand how insect-resistant rice cultivars combat BPH infestation, which is important for the comprehensive management of BPH.

Abstract:Screening of 50 rice genotypes against brown planthopper infestation was conducted in open field conditions over two consecutive years (Kharif 2018-19 and 2019-20). Based on pest population per hill, rice genotypes IR82475-110-2-2-1-2, Akshyadhan, and MTU-1010 had the least brown planthoppers. TN1, Swarna, MTU 7029, Rajendra Kasturi, Baranideep, and Sahbhagidhan had the highest population and were classified as pest-prone. Rice leaf biochemical characteristics examined in selected genotypes, revealed that the pest population was significantly and positively correlated with total sugar (r = 0.608), crude protein (r = 0.306) and total free amino acid (r = 0.358), but significantly negatively correlated with phenol (r = -0.429), crude silica (r = -0.401), and tannin (r = -0.301). Correlation analysis revealed that susceptible entries contained more total sugar, crude protein, and total free amino acids, whereas resistant genotypes contained significantly more phenol, crude silica, and tannins. This study highlighted the significance of antixenotic properties in rice genotypes against brown planthoppers.

Abstract:The brown planthopper (BPH), Nilaparvata lugens , is a serious migratory rice pest, which is distributed in the broad area of the tropical and temperate Asian-Pacific region. However, we know little about key aspects regarding its evolution such as how they diverged and dispersed worldwide. By resequencing and analyzing 358 BPH genomes from 92 populations across the world, we uncover the genetic relationships among their worldwide populations and the history of their global dispersal. We recovered five genetic groups representing the major population structures. Of these, Australian BPHs were shown large genetic divergence with Asian BPHs; two distinct groups have formed in South and Southeast/East Asia that show strong genetic admixture in the southwest border regions of China and west Thailand with Myanmar; two local populations in Bangladesh and Fujian province of China, respectively, unexpectedly separated with surrounding populations. We also find the genetic similarity and closely phylogenetic relationships between majority of East Asian BPHs and Indo-china peninsula BPHs, indicating that Southeast Asia mainland is the major insect sources and overwintering sites for East Asia. Our study provides important molecular evidence to address BPH evolution and other key aspects of its biology such as insecticides resistance and rice varieties virulence.
Dynamic metabolic responses of brown planthoppers towards susceptible and resistant rice plants

Abstract:Brown planthopper (Nilaparvata lugens Stål, BPH) causes huge economic losses in rice-growing regions, and new strategies for combating BPH are required. To understand how BPHs respond towards BPH-resistant plants, we systematically analysed the metabolic differences between BPHs feeding on the resistant and susceptible plants using NMR and GC-FID/MS. We also measured the expression of some related genes involving glycolysis and biosyntheses of trehalose, amino acids, chitin and fatty acids using real-time PCR. BPH metabonome was dominated by more than 60 metabolites including fatty acids, amino acids, carbohydrates, nucleosides/nucleotides and TCA cycle intermediates. After initial 12 h, BPHs feeding on the resistant plants had lower levels of amino acids, glucose, fatty acids and TCA cycle intermediates than on the susceptible ones. The levels of these metabolites recovered after 24 h feeding. This accompanied with increased level in trehalose, choline metabolites and nucleosides/nucleotides compared with BPH feeding on the susceptible plants. Decreased levels of BPH metabolites at the early feeding probably resulted from less BPH uptakes of sap from resistant plants and recovery of BPH metabolites at the later stage probably resulted from their adaptation to the adverse environment with their increased hopping frequency to ingest more sap together with contributions from yeast-like symbionts in BPHs. Throughout 96 h, BPH feeding on the resistant plants showed significant up-regulation of chitin synthase catalysing biosynthesis of chitin for insect exoskeleton, peritrophic membrane lining gut and tracheae. These findings provided useful metabolic information for understanding the BPH–rice interactions and perhaps for developing new BPH-combating strategies.

Abstract:Rice (Oryza sativa L.) is the staple food of more than half of Earth's population. Brown planthopper (Nilaparvata lugens Stål, BPH) is a host-specific pest of rice responsible for inducing major losses in rice production. Utilizing host resistance to control N. lugens is considered to be the most cost-effective method. Therefore, the exploration of resistance genes and resistance mechanisms has become the focus of breeders' attention. During the long-term co-evolution process, rice has evolved multiple mechanisms to defend against BPH infection, and BPHs have evolved various mechanisms to overcome the defenses of rice plants. More than 49 BPH-resistance genes/QTLs have been reported to date, and the responses of rice to BPH feeding activity involve various processes, including MAPK activation, plant hormone production, Ca2+ flux, etc. Several secretory proteins of BPHs have been identified and are involved in activating or suppressing a series of defense responses in rice. Here, we review some recent advances in our understanding of rice–BPH interactions. We also discuss research progress in controlling methods of brown planthoppers, including cultural management, trap cropping, and biological control. These studies contribute to the establishment of green integrated management systems for brown planthoppers.

Host Plant Salinity Stress Affects the Development and Population Parameters of Nilaparvata lugens (Hemiptera: Delphacidae).

Abstract:Salinization is one of the most critical abiotic stress factors for crops and a rising setback in agro-ecosystems. Changes in weather, land usage, and the salinization of irrigation water are increasing soil salinity of many farmlands. Increased soil salinity alters the plant quality, which subsequently may trigger bottom-up effects on herbivorous insect. We examined the bottom-up effect of salinity stress on population parameters of the brown planthopper (BPH), Nilaparvata lugens through rice (Oryza sativa L.) plant. The results revealed that salinity interfered with egg hatching of BPH. The nymphal development period, adult longevity, and oviposition were also influenced by salinity. Notable differences appeared in the intrinsic growth rate (r), the finite increase rate (λ) and the net reproduction rate (R0) of BPH, and a concentration-dependent effect was detected. Although salinity adversely affected BPH development, population projection predicted a successful growth of the BPH population in a relatively short time under the treatment of low and medium levels of salinity (6, 8, and 10 dS/m of NaCl), whereas higher salt concentrations (12 and 14 dS/m) lead to significant fitness costs in BPH populations. Our study predicts that BPH could become a problem in areas with lower and medium salinity and that those planthoppers may exacerbate the negative effects of salinity for rice production. This study will provide valuable information for understanding the field abundance and distribution of BPH on saline rice field, thus contributing to the development of eco-friendly strategies to manage this pest in saline ecosystems.
Rice: Nilaparvata lugens Stal interaction—current status and future prospects of brown planthopper management

Abstract:Pragmatically, Oryza sativa L. is the most considerable food in the world and about 3 billion people rely on rice for their living sustenance and rice serves as a staple food across South East Asia. Unfortunately, the diseases and insect pests remain as the key biotic stresses limiting rice production significantly. The great dissatisfaction that about 52% of rice productivity is declined due to various biotic stress factors. Among the biotic stress factors, brown planthoppers (BPHs) are the major issues, and this destructive insect pests lead to extensive yield loss. As a matter of fact, significant efforts have been taken in the past to develop resistant varieties against different pests, particularly brown planthopper with identified resistant genes. In such an atmosphere, most of the developed varieties have lost their resistance resulting in more frequent and severe pest outbreaks. Consequently, again the search has begun for new resistant donors of different pest's resistance cultivars from the existing vast germplasm collection. In this context, molecular marker-aided gene selection will provide information about the presence and absence of resistance genes. It can be utilized in various biotic stress resistance breeding programs for the improvement of the broad spectrum of insect pest's resistant and susceptible varieties. The study has found that identifying and stacking broad-spectrum resistant genes/QTLs in the popular cultivar is the foremost idea of any rice breeding program which will benefit rice farmers.

Abstract:Salinity stress triggers changes in plant morphology, physiology and molecular responses which can subsequently influence plant-insect interactions; however, these consequences remain poorly understood. We analyzed plant biomass, insect population growth rates, feeding behaviors and plant gene expression to characterize the mechanisms of the underlying interactions between the rice plant and brown planthopper (BPH) under salinity stress. Plant bioassays showed that plant growth and vigor losses were higher in control and low salinity conditions compared to high salinity stressed TN1 (salt-planthopper susceptible cultivar) in response to BPH feeding. In contrast, the losses were higher in the high salinity treated TPX (salt-planthopper resistant cultivar). BPH population growth was reduced on TN1, but increased on TPX under high salinity condition compared to the control. This cultivar-specific effect was reflected in BPH feeding behaviors on the corresponding plants. Quantification of abscisic acid (ABA) and salicylic acid (SA) signaling transcripts indicated that salinity-induced down-regulation of ABA signaling increased SA-dependent defense in TN1. While, up-regulation of ABA related genes in salinity stressed TPX resulted in the decrease in SA-signaling genes. Thus, ABA and SA antagonism might be a key element in the interaction between BPH and salinity stress. Taken together, we concluded that plant-planthopper interactions are markedly shaped by salinity and might be cultivar specific.
Effects of water-stressed rice on the brown planthopper transcriptome

Abstract:To explain the rule of outbreak and catastrophe of Nilaparvata lugens (St(a)l) (brown planthopper,BPH),the insects used for the sequencing were reared on the rice variety TN1 that were subjected to two levels of water stress using polyethylene glycol (PEG6000) at concentrations of 0 and 15％ in 48 h,respectively.Insects reared on plants without water stress was the control.Illumina technology de novo transcriptome sequencing was used to study the effect of water stress on BPH.Transcriptome analysis obtained 47 335 functional unigenes by sequence assemblage,with a mean length of 1 234 bp.Compared with the control,there were 1 331 unigenes showing significantly different expression in water-stressed BPH.Among them,up-regulated and down-regulated transcripts were 712 and 619,respectively.Twenty of the significant differentially expressed genes were randomly selected for verifying the sequencing results by using the real-time qPCR.The results showed that the expression change trends of 13 genes were consistent with the results of transcriptome sequencing.The Gene Ontology (GO) annotation and KEGG pathway analysis of significant difference expressed genes revealed that energy and water metabolism pathways were enriched in the water-stressed BPH,suggesting these genes might play crucial roles in the water-stress response in BPH.
Evaluation of Alternative Rice Planthopper Control by the Combined Action of Oil-Formulated Metarhizium Anisopliae and Low-Rate Buprofezin

Abstract:BACKGROUND: High resistance of brown planthopper (BPH) Nilaparvata lugens Stal to common insecticides is a challenge for control of the pest. An alternative control strategy based on the combined application of fungal and chemical agents has been evaluated.RESULTS: Three gradient spore concentrations of oil-formulated Metarhizium anisopliae (Metschnikoff) Sorokin (Ma456) were sprayed onto third-instar nymphs in five bioassays comprising the low buprofezin rates of 0, 10, 20, 30 and 40 mu g mL(-1) respectively. Fungal LC50 after 1 week at 25 degrees C and 14 : 10 h light : dark photoperiod decreased from 386 conidia mm(-2) in the buprofezin-free bioassay to 40 at the highest chemical rate. Buprofezin (LC50: 1647, 486 and 233 mu g m(L-1) on days 2 to 4) had no significant effect on the fungal outgrowths of mycosis-killed cadavers at the low application rates. The fungal infection was found to cause 81% reduction in reproductive potential of BPH adults. In two 40 day field trials, significant planthopper (mainly BPH) control (54-60%) was achieved by biweekly sprays of two fungal candidates (Ma456 and Ma576) at 1.5 x 10(13) conidia ha(-1) and elevated to 80-83% by incorporating 30.8 g buprofezin ha(-1) into the fungal sprays.CONCLUSION: The combined application of the fungal and chemical agents is a promising alternative strategy for BPH control.
