Abstract:Resistance to grape powdery mildew ( Uncinula necator BURR.) of 13 known Vitis species and 5 unclassified grapes native to China was evaluated. 88 clones were tested with natural infection and a subset of 34 were artificially inoculated during the years of 1991-1992. 68 clones showed resistance to U. necator . In general, we found clones of V. bryoniifolia, V. davidii , and V. piasezkii resistant to U. necator . However, intraspecific variations were observed. Results from this study indicate that Chinese native wild grapes may provide valuable germplasm resources for powdery mildew resistance.
An Evaluation System for Rice Black-Streaked Dwarf Virus Disease and Screening for Resistant Rice Germplasm

Abstract:Rice black-streaked dwarf virus disease (RBSDVD) is one of the most serious viral diseases. The absence of efficient resistance evaluation system for RBSDVD restricts the excavation of the resistance resource, thereby the resistance breeding and basal researches are limited. Here, we proposed that the appropriate density of small brown planthopper (SBPH) should be 8′104 ha-1 and the rate of RBSDV-carrying SBPH should be over 5%for RBSDVD evaluation in field condition. Further, we optimized the artificial inoculation evaluation system including circulative period, number of SBPH per plant, infestation time and age of SBPH. We evaluated 1240 varieties from 21 countries for RBSDV resistance by natural inoculation in 2010, 34 cultivars had the disease incidence less than 10%. The 34 cultivars were further evaluated by natural inoculation in three different locations in 2011 and 2012, respectively. Finally, only three varieties, Kanyakumari 29, Madurai 25, and Vietnam 160, which all came from South-east Asia, had the consistent disease incidence less than 10%in different locations and years. To verify the resistance of the three varieties against RBSDVD, we carried out the interval sowing inoculation experiment in the three different locations in 2012. In the interval sowing experiment, the disease incidence of Kanyakumari 29 was less than 12%, whereas Madurai 25 and Vietnam 160 less than 9%. In addition, in the artificial field inoculation and artificial room inoculation experiments, the disease incidence of the three varieties was less than 9%. The above natural and artificial inoculation experiments showed that Kanyakumari 29, Madurai 25, and Vietnam 160 performed stable and high RBSDVD resistance. Taken together, the RBSDVD evaluation system combining field with artificial infestation identifications used in this study is accurate and reliable. This system could be used to widely screen RBSDVD resistance resources. The construction of RBSDVD evaluation system and the RBSDVD resistance re-sources identified in this study will provide the useful tools and materials for the identification of RBSDVD resistance gene(s) and development of RBSDVD resistance varieties.
Morphological and Quality Characterization of Grape Berry and Rachis in Response to Postharvest 1-Methylcyclopropene and Elevated Oxygen and Carbon Dioxide Atmospheres

Abstract:This research studied the morph'ological characterization and quality attributes of 'Kyoho' and 'Yongyou NO.1' (Vitis vinifera L. x Vitis labrusca L.) grape berry and rachis in response to postharvest treatment with 1-methylcyclopropene (1-MCP) alone or in combination with elevated 80% O-2 (H-O-2)/20% CO2 (H-CO2). Results indicated that the integrated application of exogenous 1-MCP alone and combined with H-O-2/H-CO2 significantly prevented the rachis browning and chlorophyll degradation, maintained the cellular microstructure integrity and promoted esters and terpenes synthesis. Additionally, the transcriptional expression of genes involved in ethylene biosynthesis was sharply downregulated by 1-MCP treatment in both cultivar rachis and berries. And genes expression related to softening was also downregulated by 1-MCP alone and plus elevated O-2/CO2 atmospheres treatment. Particularly, the combinatorial treatment of 1-MCP and H-O-2 effectively impeded berry abscission and alcohols accumulation; whereas 1-MCP with H-CO2 treatment maintained the membrane permeability in berries. Nevertheless, 1-MCP alone or in combination with elevated atmospheres did not significantly affect total soluble solids and titratable acidity and did not harm sensory quality in both 'Kyoho' and 'Yongyou NO.1' cultivars after 32 days of storage.
Histological and transcriptomic insights into the interaction between grapevine and Colletotrichum viniferum

Abstract:Introduction: Grape is of high economic value. Colletotrichum viniferum, a pathogen causing grape ripe rot and leaf spot, threatens grape production and quality. Methods: This study investigates the interplay between C. viniferum by Cytological study and transcriptome sequencing. Results: Different grapevine germplasms, V. vinifera cv. Thompson Seedless (TS), V. labrusca accession Beaumont (B) and V. piasezkii Liuba-8 (LB-8) were classified as highly sensitive, moderate resistant and resistant to C. viniferum, respectively. Cytological study analysis reveals distinct differences between susceptible and resistant grapes post-inoculation, including faster pathogen development, longer germination tubes, normal appressoria of C. viniferum and absence of white secretions in the susceptible host grapevine. To understand the pathogenic mechanisms of C. viniferum, transcriptome sequencing was performed on the susceptible grapevine "TS" identifying 236 differentially expressed C. viniferum genes. These included 56 effectors, 36 carbohydrate genes, 5 P450 genes, and 10 genes involved in secondary metabolism. Fungal effectors are known as pivotal pathogenic factors that modulate plant immunity and affect disease development. Agrobacterium-mediated transient transformation in Nicotiana benthamiana screened 10 effectors (CvA13877, CvA01508, CvA05621, CvA00229, CvA07043, CvA05569, CvA12648, CvA02698, CvA14071 and CvA10999) that inhibited INF1 (infestans 1, P. infestans PAMP elicitor) induced cell death and 2 effectors (CvA02641 and CvA11478) that induced cell death. Additionally, transcriptome analysis of "TS" in response to C. viniferum identified differentially expressed grape genes related to plant hormone signaling (TGA, PR1, ETR, and ERF1/2), resveratrol biosynthesis genes (STS), phenylpropanoid biosynthesis genes (PAL and COMT), photosynthetic antenna proteins (Lhca and Lhcb), transcription factors (WRKY, NAC, MYB, ERF, GATA, bHLH and SBP), ROS (reactive oxygen species) clearance genes (CAT, GSH, POD and SOD), and disease-related genes (LRR, RPS2 and GST). Discussion: This study highlights the potential functional diversity of C. viniferum effectors. Our findings lay a foundation for further research of infection mechanisms in Colletotrichum and identification of disease response targets in grape.
The electrophoretical activity of the buccal epithelium cells nuclei in newborns of smoking mothers.

Abstract:The electrophoretical activity of nuclei (EMN) occurs in all cells of the human body. The EMN is variable and depends on the particular stage of human ontogenesis. The factors that modify the ontogenesis may influence the electrophoretical activity of nuclei. The aim of the study was to compare the index of electrophoretical activity of nuclei (index EMN) derived from the buccal epithelium of 40-week newborns (n = 26) from healthy, single pregnancies of smoking mothers with newborns (n = 31) of non-smoking mothers. The cells of the buccal epithelium were analysed using light microscope in the environment of alternating field (voltage 20-30 V; intensity 0.1 mA; frequency 1-2 Hz) generated by Biotest processor. In each group we estimated the number of cells with mobile nuclei and cells with motionless nuclei per 100 cells being observed. The result of this research was that there is not activity of nuclei in the group of newborns of smoking mothers (index EMN = 0 with p < 0.05). The absence of activity of nuclei makes it impossible to use the EMN method for estimation of biological age in the group of newborns of smoking mothers.
Isolation and Identification of Pear Ring Rot Fungus and Resistance Evaluation of Different Pear Varieties

Abstract:Botryosphaeria dothidea is a significant plant pathogen responsible for causing ulcers, wilt, and fruit decay across a wide range of host plants. One notable fungal disease attributed to B. dothidea is pear tree ring rot, which currently ranks among the most severe diseases affecting pear trees in China. This pathogen primarily targets branches and fruits, occasionally impacting leaves as well, leading to tree weakening, fruit rot, and leaf drop. The annual repercussions of this disease severely affect both the yield and quality of pear fruits, thereby impeding the healthy development of the pear industry. Recent studies have indicated that other species within the B. dothidea complex can also induce pear ring rot; however, specific physiological strains of B. dothidea remain unreported. Consequently, this study collected tissues from pear trees infected with ring rot from orchards located in Liaoning, Hebei, Shandong, and other regions throughout China. Through morphological characterization combined with pathogenicity assessments and DNA sequence comparisons involving partial internal transcribed spacer (ITS), translation elongation factor (TEF), and β-tubulin (TUB) genes, 21 strains belonging to the Botryosphaeria spp. were identified. These 21 strains served as research subjects for inoculating dormant annual branches from 30 germplasm resources of pear trees in vitro. The results demonstrated that all tested strains could induce lesions on the branches which were characterized by dark brown spots. Furthermore, inoculation experiments involving these 21 strains were conducted to evaluate the resistance levels of various pear varieties against ring rot disease. The resistance was assessed by inoculating different isolates onto distinct pear varieties; this approach established the criteria for evaluating resistance while minimizing identification errors stemming from the variable responses exhibited by certain varieties towards individual strains. Ultimately, this study aims to provide a theoretical foundation for effective prevention and treatment strategies against pear ring rot.
Transcriptional profiling of ESTs responsive to Rhizobium vitis from 'Tamnara' grapevines (Vitis sp.)

Abstract:Genes related with defense responses were screened from the cDNA library constructed with Rhizobium vitis-inoculated or salicylic acid (SA)-treated 'Tamnara' grapevine (Vitis sp.) leaves. Among 13,728 expressed sequence tags (ESTs) from 'Tamnara' grapevine upon R. vitis inoculation and SA treatment, 6776 unigenes containing 1915 contigs and 4860 singletons were obtained. In gene ontology analysis, there were about 3200 clones related with biological process, 3555 with molecular function, and 3354 with cellular component genes. Proteins of secretory organ (35%), plasma membrane (30%), endoplasmic reticulum (20%), and vacuole (11%) were predicted. Photosynthesis-related genes and defense-related genes were most abundant. Among ESTs, 199 resistance-related ones were mapped to the genome of Vitis vinifera L. with three markers, GLP1-12, MHD98, and MHD145, which are known to be linked to resistance against powdery mildew. Approximately, 120 simple sequence repeats (SSRs) detected in cDNAs could be used as EST-derived SSR markers in disease resistant grape breeding.
Research to Fungicide Sensitivity of Colletotrichum spp. Isolated from Apple Fruits in Cheongsong, Korea

Abstract:Apple grower in the Cheongsong region, Korea has reported the increased loss of apple yield due to severe bitter rot incidence. We noticed that this indience is because the Colletotrichum population has developed resistance to commonly used fungicides. We isolated 39 Colletotrichum isolates from 13 orchards in Cheongsong, and all the isolated Colletotrichum species were identified as C. siamense or C. fructicola . These 39 strains were tested for mycelial growth and conidial germination against 12 fungicides. trifloxystrobin (30–55% in recommended concentrations) was shown lower inhibitory effect on mycelial growth. However, the inhibition of conidial germination was shown higher than mycelial growth (62–100%). Kresoxim-methyl was shown lower inhibitory effect on mycelial growth (29–55%). conidial germination inhibitory effect was shown 51% to 96%. dithianon was shown diversity response to inhibition of mycelial growth (43–100%). tebuconazole was shown high inhibitory effect on mycelial growth (84–100%) and conidial germination inhibitory effect was shown to be 64 to 100%. metconazole has been found to display with high inhibitory effect on mycelial growth (79–100%) and conidial germination (70–80%). fluazinam was shown to possess high inhibitory effect on mycelial growth (87–100%) and conidial germination (94–100%). This study provides basic information for the effective management of apple bitter rot.
Evaluation of screening methods for anthracnose fruit rot resistance in chilli (Capsicum spp.)

Abstract:Anthracnose fruit rot caused by Colletotrichum spp. is a serious production constraint causing severe marketable yield loss in chilli. Field evaluation of chilli accessions for resistance to Colletotrichum spp. depends on various factors affecting disease expression such as edaphic conditions, temperature, rainfall, humidity and other variables that are difficult to control, therefore considered less accurate. Also, high chances of cross contamination with different Colletotrichum species leads to inconclusive assays for specific pathogen species and isolate. To identify a stable and reliable screening method, various chilli accessions were subjected to in vitro pin-prick and non-wounding spray methods using a specific pathogen isolates. When chilli accessions were screened against C. gloeosporioides isolate ‘IHRCg-1’, the in vitro pin- prick method showed positive correlation with the non- wounding spray method, except in the accession PBC80. The change in bioassay influenced the disease reaction pattern in the accession PBC 80, probably the pin pricks break the basal cuticle defense mechanism that was retained in spray inoculation method indicating varied resistance pattern. However, in the accession PBC 81 stable resistance pattern was observed against isolates of both species viz., C. truncatum ‘IIHR Ct-1’ and C. gloeosporioides ‘IIHR Cg-1’ and in the accession PBC 80 against C. truncatum ‘IIHR Ct-1’ in both the inoculation methods that depicted the expression of resistance genes during both methods of inoculation. Based on disease development pattern, the red ripe chilli expressed a variant reaction to infection by C. truncatum and C. gloeosporioides. The peak anthracnose infection at 10 DAI and 14 DAI is an accurate duration to record ‘IIHR Cg-1’ and ‘IIHR Ct-1’ infection, respectively on chilli ripe fruit for assaying the resistance.

Abstract:Paenibacillus polymyxa HT16 was evaluated for its ability to reduce white rot disease caused by Coniella diplodiella. HT16 showed significant inhibitory effects on C. diplodiella in vitro, reducing white rot disease in 'Kyoho' and 'Thompson Seedless' table grape varieties by more than 40% after inoculation. Liquid cultures were more effective than washed cell suspensions or cell-free supernatants, and a higher content of, and longer incubation time with, strain HT16 were more effective in reducing disease incidence. The spore germination rate and mycelial morphology of C. diplodiella were adversely affected by HT16. These results suggested that HT16 may be a promising biocontrol agent against pre-harvest and post-harvest decay in table grapes.
A method for phenotypic evaluation of grapevine resistance in relation to phenological development

Abstract:Fungus-resistant grapevine cultivars, so called PIWIs, are characterized by increased resistance to powdery mildew and downy mildew. However, in order to maintain the durability of resistance in these new grape cultivars, targeted fungicide treatments are recommended. For ideal schedule of these treatments, it is necessary to recognize the most sensitive organs of the grape. This study introduces a method for phenotypic evaluation of Plasmopara viticola resistance in grape clusters under controlled and standardized conditions during phenological development over the entire season. The approach was validated with the traditional cultivar Pinot Noir and the PIWIs Cabernet Cortis ( Rpv3.3 , Rpv10 ), Solaris ( Rpv3.3 , Rpv10 ) and Souvignier Gris ( Rpv3.2 ). All cultivars were susceptible during the early stages of development up to flowering, and resistance levels increased as phenological development progressed. Cabernet Cortis and Solaris clusters were susceptible until fruit development (BBCH 71–73) when they became almost completely resistant. No differences between Souvignier Gris and Pinot Noir were detected until berries were pea-sized (BBCH 75) when P. viticola resistance of Souvignier Gris clusters increased significantly. Ontogenetic resistance in Pinot Noir was detected at berry touch (BBCH 77–79) and clusters of this cultivar were almost completely resistant at the beginning of ripening (BBCH 81–83). These results indicate that the approach presented is suitable for determining the resistance of grape cultivars at different stages of development. Consequently, in the future, fungicide applications can be adjusted more precisely to the resistance level of a grape cultivar during the growing season.

Abstract:The aim of this study was to screen the potential of locally grown sugarcane genotypes for red rot resistance and activity of native microbial strains against the pathogen. Field trials were conducted with 70 genotypes for consecutive years and results showed that only two genotypes viz., SSRI-1 and CO-0238 showed resistant behavior towards red rot of sugarcane. The red rot pathogen Colletotrichum falcatum Went was characterized and pathogenicity tests on two susceptible genotypes (NSG-59 and CPSG-2923) showed high virulence of SUCF04 isolate to develop severe disease lesions. The native rhizospheric microbiota was screened for microbial consortia exhibiting fine antifungal activity against the highly virulent pathogenic strain. The antagonism assay exposed that 10 bacterial isolates out of 46 showed great potential for antifungal activity. The selected bacterial isolates revealed 68–99% pathogen inhibition during the assay. The fungal strains with biological control potential inhibited the pathogen growth by 20–80% and a group of three strains with more than 50% antifungal activity were characterized. The molecular characterization of these microbes revealed that the isolates were belonging to Bacillus subtilus, Pseudomonas putida, Pseudomonas fluorescence, Trichoderma harzianum and several other important taxa. This study revealed that only two sugarcane genotypes were found as resistant against red rot pathogen, while most of the genotypes showed susceptible to moderately susceptible response. Moreover, the native residential microbiota associated with sugarcane exhibited great antifungal potential and can be utilized for disease protection and improved crop productivity. © 2021 Friends Science Publishers

Abstract:Colletotrichum spp. are notorious phytopathogens that cause anthracnose rot disease in jujube fruits, have a complex lifestyle (with necrotrophic, hemibiotrophic, quiescent, and endophytic phases), they spread rapidly mainly through the production of spores and are increasingly becoming resistant to various fungicides which makes them difficult to control. This study investigated the inhibition of spore germination and mycelial growth of Colletotrichum spp. to suppress anthracnose rot disease in jujube fruits through the activity of cell wall degrading enzymes, and improvement of jujube fruit characteristics and yield via the production of indole-3-acetic acid (IAA) and gibberellin acid (GA3) by Bacillus velezensis CE 100. The bacterial culture broth exhibited a strong chitinase, β-1,3-glucanase and protease and the crude fraction suppressed the germination of spores by more than 95.0 % at a concentration of 100 µL/mL, and substantially reduced mycelial growth of Colletotrichum spp. The microscopic analysis revealed that the treatments degraded the cell walls and caused severe structural distortion of the spores and hyphae of Colletotrichum spp. Field application of B. velezensis CE 100 on the jujube tree crown and in the rhizosphere lowered the rate of anthracnose disease incidence by 2.8-fold and disease severity index (DSI) by 4.4-fold, compared to the control. In addition, B. velezensis CE 100 produced IAA (up to 15.6 mg/mL) and GA3 (up to 1.3 mg/mL), and the field application of the bacterial culture on the jujube tree crown and in the rhizosphere consequently improved the average fruit characteristics (fruit length, fruit diameter and fresh weight by 1.2-, 1.4- and 1.5-fold), chlorophyll index (photosynthetic rate by 1.1-fold), and fruit yield (by 2.1-fold) of jujube, compared to the control group. Thus, the inoculation of B. velezensis CE 100 has the potential effectiveness as a bio-fungicide against anthracnose rot disease in jujube fruits, and as a bio-fertilizer to improve fruit characteristics and yield.

Abstract:The effectiveness of the yeast Rhodotorula glutinis was evaluated in the field experiment for control of the gray rot disease on strawberry caused by the fungus Botrytis cinerea. The results showed that all treatments of R. glutinis yeast led to a significant superiority in inducing systemic strawberry resistance and an increase in vegetative, productivity and quality indicators of three strawberry cultivars, compared to the Prado fungicide treatment and the pathogenic fungus treatment only. Treatment of filtrate and yeast cells (R. glutinis - Cs +F with B. cinerea) achieved the highest activity of the enzymes that induce plant resistance, which included peroxidase, chitinase and β-glucanase, the enzymatic activity was 2.25, 2.31 and 1.62 units/ml for Albion cultivar, with significant superiority compared to other treatments. The treatment of (R. glutinis - Cs +F) in the presence of the pathogenic fungus recorded the highest vegetative parameters, which included plant height and shoot weight in Albion cultivar, reaching 27.33 cm and 14.66 g, respectively. All treatments of R. glutinis yeast led to a significant superiority in affecting the severity of infection for all studied strawberry cultivars under the infection conditions with the pathogenic fungus B. cinerea, the Albion cultivar achieved the highest decrease in the infection severity reached 17.37% in (R. glutinis - Cs +F with B. cinerea ), while it had the least effect on the severity of infection in the Festival cultivar, reached 95.28% in the treatment of the pathogenic fungus. The yield of strawberry fruits in Albion cultivar was superior compared to other cultivars, as the plant yield reached 68.92 g/plant in the treatment of (R. glutinis-Cs +F with B. cinerea), while the lowest yield was 16.69 g/plant in the Festival cultivar with the pathogenic fungus treatment only. The treatment of yeast filtrate and cells with B. cinerea achieved the lowest infection intensity, the highest hardness and the highest vitamin C content of strawberry fruits after 7 days of harvest, compared to the Prado fungicide treatment and the pathogenic fungus treatment only.

Abstract:Wild European Grapevine (Vitis sylvestris), the ancestor of domesticated grapevine harbours resistance factors blocking the spread of Black Rot, an emerging disease in viticulture, suppressing the appearance of symptoms. Black Rot is a grapevine disease caused by the ascomycete Phyllosticta ampelicida. Neglected so far, this is developing into a pertinent problem in organic viticulture as resistant varieties are still lacking. Here, we follow cellular details of the infection process in the susceptible vinifera variety Müller‐Thurgau and screen the ancestral European wild grapevine (V. vinifera sylvestris) for resistance to Black Rot. Using a standardized infection assay, we follow fungal development using LTSEM and quantify key stages on different hosts using fluorescence microscopy. There is considerable variation in susceptibility, which is associated with more rapid leaf maturation. Hyphal growth on different carbon sources shows a preference for pectins over starch, cellulose or xylans. In the resistant sylvestris genotypes Ketsch 16 and Ketsch 18 we find that neither spore attachment nor appressorium formation, but hyphal elongation is significantly inhibited as compared to Müller‐Thurgau. Moreover, defence‐related oxidative burst and accumulation of phenolic compounds is stimulated in the resistant genotypes. We arrive at a model, where more rapid maturation of the cell wall in these sylvestris genotypes sequesters pectins as major food source and thus block hyphal elongation. This paves the way for introgression of genetic factors responsible for cell wall maturation into V. vinifera to develop Black Rot‐resistant varieties of grapevine.

Characterization and Biocontrol Efficacy of Bacillus velezensis GYUN-1190 against Apple Bitter Rot

Abstract:The application of synthetic fungicides has resulted in environmental pollution and adverse effects on non-target species. To reduce the use of agrochemicals, crop disease management requires microbial biological control agents. Bacillus-related genera produce secondary metabolites to control fungal pathogens. Bacillus velezensis GYUN-1190, isolated from soil, showed antagonistic activity against Colletotrichum fructicola, the apple anthracnose pathogen. Volatile organic compounds and culture filtrate (CF) from GYUN-1190 inhibited C. fructicola growth in vitro, by 80.9% and 30.25%, respectively. The CF of GYUN-1190 inhibited pathogen spore germination more than cell suspensions at 10 8 cfu/ml. Furthermore, GYUN-1190 CF is effective in inhibiting C. fructicola mycelial growth in vitro, and it suppresses apple fruit bitter rot more effectively than GYUN-1190 cell suspensions and pyraclostrobin in planta. The mycelial growth of C. fructicola was completely inhibited 48 h after immersion into the CF, in compared with positive controls and GYUN-1190 cell suspensions. The genetic mechanism underlying the biocontrol features of GYUN-1190 was defined using its whole-genome sequence, which was closely compared to similar strains. It consisted of 4,240,653 bp with 45.9% GC content, with 4,142 coding sequences, 87 tRNA, and 28 rRNA genes. The genomic investigation found 14 putative secondary metabolite biosynthetic gene clusters. The investigation suggests that B. velezensis GYUN-1190 might be more effective than chemical fungicides and could address its potential as a biological control agent.

Abstract:Black rot (Guignardia bidwellii) and downy mildew (Plasmopara viticola) are two major grapevine diseases against which the development of efficient biocontrol solutions is required in a context of sustainable viticulture. This study aimed at evaluating and comparing the efficacy and modes of action of bacterial culture supernatants from Bacillus velezensis Buz14 and B. ginsengihumi S38. Both biocontrol agents (BCA) were previously demonstrated as highly effective against Botrytis cinerea in grapevines. In semi-controlled conditions, both supernatants provided significant protection against black rot and downy mildew. They exhibited antibiosis against the pathogens by significantly decreasing G. bidwellii mycelial growth, but also the release and motility of P. viticola zoospores. They also significantly induced grapevine defences, as stilbene production. The LB medium, used for the bacterial cultures, also showed partial effects against both pathogens and induced plant defences. This is discussed in terms of choice of experimental controls when studying the biological activity of BCA supernatants. Thus, we identified two bacterial culture supernatants as new potential biocontrol products exhibiting multi-spectrum antagonist activity against different grapevine key pathogens and having a dual mode of action.
A rapid and efficient Agrobacterium-mediated transient transformation system in grape berries

Abstract:Transient transformation is extremely useful for rapid in vivo assessment of gene function, especially for fruit-related genes. Grape berry, while an important fruit crop, is recalcitrant to transient transformation, due to the high turgor pressure in its mesocarp cells that limits the ability of Agrobacterium to penetrate into the tissue. It is urgent to establish a simple transient transformation system for rapid analysis of gene function. In this study, different injection methods, grape genotypes, and developmental stages were tested in order to develop a rapid and efficient Agrobacterium -mediated transient transformation methodology for grape berries. Two injection methods, namely punch injection and direct injection, were evaluated using the β-glucuronidase ( GUS ) gene and by x-gluc tissue staining and 4-methylumbelliferyl-β-D-glucuronide fluorescence analysis. The results indicated that there were no significant differences on transformation effects between the two methods, but the latter was more suitable because of its simplicity and convenience. Six grape cultivars ('Hanxiangmi', 'Moldova', 'Zijixin', 'Jumeigui', 'Shine-Muscat', and 'A17') were tested for transient transformation. 'Hanxiangmi', 'Moldova', and 'Zijixin' grape berries were not suitable for agroinfiltration due to frequently fruit cracking, browning, and formation of scar skin. The fruit integrity rates of 'Jumeigui', 'Shine-Muscat', and 'A17' berries were all above 80%, and GUS activity was detected in the berries of the three cultivars 3–14 days after injection with the Agrobacterium culture, while higher GUS activities were observed in the 'Jumeigui' berries. The levels of GUS activity in injected berries at 7–8 weeks after full blooming (WAFB) were more than twice at 6 WAFB. In subsequent assays, the over-expression of MYB transcription factor VvMYB44 via transient transformation accelerated the anthocyanin accumulation and fruit coloring through raising the expression levels of VvLAR1 , VvUFGT , VvLDOX , VvANS , and VvDFR , which verified the effectiveness of this transformation system. These experiments finally identified the reliable grape cultivars and suitable operational approach for transient transformation and further indicated that this Agrobacterium -mediated transient transformation system was efficient and suitable for the elucidation of gene function in grape berries.

Abstract:Anthracnose leaf spot and ripe rot disease, caused by Colletotrichum gloeosporioides in India, is of economic importance. The objective of the investigation was to isolate and characterise bacterial antagonists from the grape berry surface. Nine bacteria out of the 46 that were isolated significantly inhibited C. gloeosporioides in a dual culture assay. Six of these nine microorganisms demonstrated pathogen inhibition by producing extracellular volatile and non-volatile compounds. Bacteria SB5, SB13, and SB15 were chosen for the polyhouse experiment because they demonstrated the best disease control in a detached leaf assay. At 1 × 10 6 CFU/ml of bacterial concentration, both curative and preventive trials showed significant disease inhibition. Based on morphological and molecular characterisation the bacteria were identified as Bacillus species. All three strains could be effectively used for the biological control of C. gloeosporioides .
