Abstract:The enzyme L-phenylalanine ammonia-lyase (EC 4.3.1.5), which catalyzes the conversion of L-phenylalanine into trans-cinnamic acid, was adsorbed in the walls of asymmetric hollow fibers and covalently bound to the innerwall of small bore nylon tubing. Multi-tubular enzyme reactor cartridges were constructed both from hollow fibers and nylon tubes and the capacity of the reactors to degrade phenylalanine at physiological concentrations was evaluated in perfusion studies at different flow rates ranging from 30 to 80 ml/min. Hollow fiber enzyme reactors showed significantly higher activity than nylon tube reactors of commensurable dimensions and this finding is attributed to the difficulties in immobilizing L-phenylalanine ammonia-lyase via covalent linkages. The results suggest that extracorporeal use of such multi-tubular enzyme reactors offer a promising approach to deplete serum phenylalanine levels.

Abstract:Hepatic ischemia–reperfusion injury (HIRI) is the main cause of postoperative liver dysfunction and liver failure. Traditional separation of HIRI diagnosis and therapy confers several disadvantages, including the inability to visualize the therapeutic and asynchronous action. However, developing a versatile material with integrated diagnosis and treatment for HIRI remains a great challenge. Given that hypochlorous acid (HOCl) plays a crucial oxidative role in HIRI, we developed a single-component multifunctional fluorescent theranostic platform (MB-Gly) with a "three-in-one" molecular design incorporating a near-infrared fluorophore methylene blue, glycine and a HOCl-response unit, which could not only provide real-time visualization of HIRI but also boost targeted drug delivery. Using MB-Gly, we were able to achieve real-time and dynamic monitoring of HOCl during HIRI in hepatocytes and mouse livers and reduce the liver damage in hepatocytes and mice. RNA sequencing illustrated the therapeutic role of MB-Gly associated with changes in gene expression related to apoptosis, oxidative stress, metabolism and inflammation. To the best of our knowledge, this is the first multifunctional fluorescent theranostic system for HIRI reported to date. Our smart "three-in-one" approach shines light on the etiology and pathogenesis of HIRI, providing profound insights into the development of potential therapeutic targets.

Abstract:Liver fibrosis is a major stage in the development of liver disease, and it is important to investigate its pathogenesis for early intervention or even reversal. Recent studies have found that intestinal disease can aggravate liver fibrosis through the role of the "gut-liver axis". Hypoxia is considered to be a typical characteristic of many diseases including ulcerative colitis and liver fibrosis. However, there is no fluorescent probe for hypoxia detection to investigate the "gut-liver axis". Herein we design and synthesize a turn-on fluorescent probe termed Cy-AP, which displays high sensitivity and selectivity to hypoxia given by sodium dithionite (Na2S2O4) in vitro with near-infrared (NIR) emission (725 nm). The possible response mechanism of Cy-AP toward hypoxia is given and proved though HPLC, MS, and theory calculation. Moreover, on the basis of low cell cytotoxicity, the probe is used in visualizing hypoxia in four cell lines (HepG2, HCT116, HeLa, and MCF-7 cells) by fluorescence imaging, flow cytometry, and 3D imaging. Furthermore, due to its NIR emission, Cy-AP can monitor the hypoxia condition in vivo such as in liver fibrosis mice and ulcerative colitis mice models. In particular, the probe can validate the existence and mechanism of the "gut-liver axis" in vivo by monitoring hypoxia. To the best of our knowledge, this is the first work to give a strategy for studying the "gut-liver axis" by a NIR hypoxia-sensitive fluorescent probe, which will provide some powerful support for delaying the progression of liver fibrosis and thus promoting the treatment of liver disease.

Abstract:Hepatic ischemia-reperfusion injury (HIRI) is characterized by mitochondrial dysfunction and oxidative stress. Monitoring mitochondrial hydrogen peroxide (mtH 2 O 2 ) levels in real-time through super-resolution imaging is crucial for elucidating its distribution in live cells and its mechanism of action during HIRI. However, low-background fluorogenic probes have been overlooked in the context of super-resolution imaging. In this study, we developed a low-background fluorogenic probe ( Mito-WG ) with the potential for super-resolution morphology-correlated mitochondrial identification to track the fluctuates of mtH 2 O 2 in HIRI. Activation of the desirable fluorescence properties of the probe by mtH 2 O 2 was confirmed using structural illumination microscopy (SIM), enabling high-quality mitochondrial imaging with exceptional specificity and sensitivity. Fluctuations in mtH 2 O 2 levels were successfully observed in both cellular and rat models of HIRI. Furthermore, we associated the decline in mitochondrial redox homeostasis with accelerated mtH 2 O 2 production during HIRI, which triggered mitophagy deficiency and led to cell death. In conclusion, Mito-WG possesses excellent photophysical and low-background properties for SIM imaging, making it a promising tool for mtH 2 O 2 tracking in HIRI research and clinical diagnosis.

Abstract:Reversible NIR luminescent probes with negligible photocytotoxicity are required for long-term tracking of cycling hypoxia in vivo. However, almost all of the reported organic fluorescent hypoxia probes reported until now were irreversible. Here we report a reversible arylazo-conjugated fluorescent probe (HDSF) for cycling hypoxia imaging. HDSF displays an off-on fluorescence switch at 705 nm in normoxia-hypoxia cycles. Mass spectroscopic and theoretical studies confirm that the reversible sensing behavior is attributed to the two electron-withdrawing trifluoromethyl groups, which stabilizes the reduction intermediate phenylhydrazine and blocks the further reductive decomposition. Cycling hypoxia monitoring in cells and zebrafish embryos is realized by HDSF using confocal imaging. Moreover, hypoxic solid tumors are visualized and the ischemia-reperfusion process in mice is monitored in real-time. This work provides an effective strategy to construct organic fluorescent probes for cycling hypoxia imaging and paves the way for the study of cycling hypoxia biology. Most fluorescence probes to measure hypoxia are irreversible. Here, the authors develop a near infrared luminescent probe to measure hypoxia which is reversible and use the probe to monitor hypoxia-reoxygenation in several animal models.
A novel hepatocyte-targeting ratiometric fluorescent probe for imaging hydrogen peroxide in zebrafish

Abstract:Hydrogen peroxide (H2O2), as one of important reactive oxygen species (ROS), can be endogenously produced in the liver, and used as a biomarker for predicting drug-induced liver injury (DILI). So, it is important to construct an efficient method for monitoring H2O2 in the liver to assess DILI. Herein, a hepatocyte-targeted fluorescent probe (GCP) for ratiometric detection of H2O2 is developed. GCP consists of two main parts: coumarin-pyran-based fluorophore and galactose-based hepatocyte-targetable group. The probe itself shows near-infrared (NIR) emission owing to its extended p-conjugation. Meanwhile, a large emission shift from 706 to 482 nm (224 nm) is observed after the reaction with H2O2. Taking advantage of the spectroscopic property, GCP exhibits high sensitivity toward H2O2 with 19-fold fluorescence ratio enhancement and is highly selective over other ROS. Moreover, the probe possesses a good hepatocyte-targeting ability and is applied for ratiometric monitoring endogenous H2O2 in HepG2 cells. Furthermore, the probe can trace H2O2 level during DILI in HepG2 cells. Most importantly, GCP can locate in the liver of zebrafish and detect H2O2 level in the liver during DILI, which will be a potentially useful tool to assess DILI.

Abstract:Hepatic ischemia-reperfusion injury (HIRI) is defined as liver tissue damage and cell death caused by reperfusion during liver transplantation or hepatectomy. Oxidative stress is one of the important mechanisms of HIRI. Studies have shown that the incidence of HIRI is very high, however, the number of patients who can get timely and efficient treatment is small. The reason is not hard to explain that invasive ways of detection and lack of timely of diagnostic methods. Hence, a new detection method is urgently needed in clinic application. Reactive oxygen species (ROS), which are markers of oxidative stress in the liver, could be detected by optical imaging and offer timely and effective non-invasive diagnosis and monitoring. Optical imaging could become the most potential tool of diagnosis of HIRI in the future. In addition, optical technology can also be used in disease treatment. It found that optical therapy has the function of anti-oxidative stress. Consequently, it has possibility to treat HIRI caused by oxidative stress. In this review, we mainly summarized the application and prospect of optical techniques in oxidative stress-induced by HIRI.

Abstract:The redox homeostasis between hydrogen peroxide (H2O2) and sulfur dioxide (SO2) is necessary to maintain the normal physiological function of biological system. However, Due to the lack of fluorescent probes for the reversible dual detection of SO2 and H2O2, it is not clear which of the above signaling molecules predominates to cause redox state imbalance in biological systems. Herein, we designed a new muti-color reversible fluorescence probe Mito-BHY based on ICT mechanism. The probe has three different fluorescence signals (blue, green, and red) before and after reacting to SO2 and H2O2, and it can also reversibly detect SO2 and H2O2 through changes in the green and red fluorescence signals. The probe Mito-BHY has the characteristics of favorite response to SO2 and H2O2 with fast response speed, highly selectivity and biocompatibility, and can be used to monitor SO2 and H2O2 in the living cells and mice. Using Mito-BHY, we discovered that the content of H2O2 increased during cell redox imbalance and ischemia reperfusion in the mice, while the content of SO2 remained basically unchanged. This work will contribute to the understanding of the variation of SO2 and H2O2 levels, and is expected to provide an effective tool for pathological studies of redox imbalance.

Abstract:Hepatic ischemia-reperfusion injury (HIRI), which can severely impair liver function, has a potential relationship with reactive nitrogen species. Nitroxyl (HNO) has been discovered for its involvements in some biological functions and pharmacological activities. But till now there has been no knowledge on the role of HNO in HIRI process, mainly because accurately tracking its' fluctuation at molecular level in vivo is extremely difficult. Herein, we developed a responsive ratiometric near-infrared-II (NIR-II) nanoprobe with rare earth ions-doped nanoparticles (RENPs) as the luminophore, and a molecular trigger which can specifically react with HNO to regulate the NIR-II emission of RENPs. With this nanoprobe, we revealed the relationship between HNO and the HIRI process, and disclosed that HNO may be a product of stress reaction during this process. This work not only created a useful tool for visually tracking HNO in vivo but also provided the first-hand information about the role of HNO in HIRI.

Abstract:Drug-induced liver injury (DILI) not only poses a serious health threat to patients, but is also a major obstacle to the development of new drugs. The liver toxicity of many drugs is not detected until late in the development process or even after marketing, which significantly increases the cost of new drug development. This may be limited by the lack of tools to detect DILI. Studies have shown that abnormal changes in peroxynitrite (ONOO-) levels in the endoplasmic reticulum of liver cells may be an early manifestation of DILI. Therefore, the development of fluorescent probes to detect ONOO- in the endoplasmic reticulum may provide assistance in the detection of DILI and the screening of new drugs. Herein, we report a fluorescent probe, ER-N, capable of detecting ONOO- in the endoplasmic reticulum with high selectivity and sensitivity. The probe selects di(trifluoromethyl)benzene as the targeting group of the endoplasmic reticulum, which possesses excellent targeting to the endoplasmic reticulum, and provides a new design method for the establishment of other endoplasmic reticulum-targeting probes. In addition, the probe ER-N successfully realized the detection of endogenous and exogenous ONOO- in cells and zebrafish. More importantly, the probe ER-N was able to be used for tracing changes in the levels of ONOO- in the endoplasmic reticulum of cells and tissues under drug stimulation. Together, these results suggest that the probe ER-N has the potential to be a tool for DILI detection and new drug screening.

Abstract:AbstractReversible imaging probes that allow for the dynamic visualization of the redox cycle between hydroxyl radical (⋅OH) and hydrogen sulfide (H2S) are vital to probe the redox imbalance‐involved pathological process in vivo. Herein, we report a reversible ratiometric photoacoustic (PA) imaging nanoprobe (1‐PAIN) for the real‐time imaging of ⋅OH/H2S redox cycle in vivo.1‐PAIN displays a low PA ratio between 690 and 825 nm (PA690/PA825), which significantly increases by ≈5‐fold upon oxidation by ⋅OH, and is switched back to the initially low PA690/PA825value upon reduction by H2S.1‐PAIN could dynamically report on the hepatic ⋅OH production in mice during the lipopolysaccharide (LPS)‐induced liver inflammation process, and visualize hepatic H2S generation during theN‐acetyl cysteine (NAC)‐induced anti‐inflammation process.1‐PAIN can act as a useful tool to probe the redox state in living biology, beneficial for the study of redox imbalance‐related diseases.

Abstract:As the metabolic and detoxification center of the body, the liver is closely involved in many critical physiological processes, such as metabolism, immunity, digestion, storage of liver glycogen and synthesis of proteins. Normal liver function is essential for maintaining the health and stability of the body, and abnormal liver function can lead to a variety of liver diseases, such as non-alcoholic fatty liver, liver cancer, hepatitis, and liver fibrosis. Early diagnosis can buy valuable time for the treatment of liver disease. In the process of disease onset and progression, the fluctuation of biomarkers can directly reflect the health status of the liver, providing valuable opportunities and avenues for the diagnosis of liver disease and assessment of treatment efficacy. In recent years, a large number of fluorescent probes have been reported for the detection of biomarkers of liver diseases, which provides rich and valuable information for the study of pathological processes, drug development, diagnosis and treatment of diseases. The review systematically summarizes the recent progress of fluorescence probes in the detection of hepatopathy-related biomarkers in the past three years, including the design and recognition mechanism of fluorescence probes, the main types of liver diseases and typical biomarkers. Also, we deeply discussed the problems and shortcomings of the current fluorescent probes and furnished the direction and path for future development. In the future, long wavelength probes, multi-detection probes, quantitative analysis probes, and multi-modal probes will be popular trends. We expect that this review may facilitate the blossom of novel fluorescent probes for disease detection research and further provide meaningful guidance for the diagnosis and treatment of diseases.
