Abstract:Antibiotic resistance is a global problem and bacterial biofilms contribute to its development. In this context, this study aimed to perform the synthesis and characterization of seven materials based on silica mesoporous nanoparticles functionalized with three types of fluoroquinolones, along with Cu2+ or Ag+ species to evaluate the antibacterial properties against Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, and Pseudomonas aeruginosa, including clinical and multi-drug-resistant strains of S. aureus and P. aeruginosa. In addition, in order to obtain an effective material to promote wound healing, a well-known proliferative agent, phenytoin sodium, was adsorbed onto one of the silver-functionalized materials. Furthermore, biofilm studies and the generation of reactive oxygen species (ROS) were also carried out to determine the antibacterial potential of the synthesized materials. In this sense, the Cu2+ materials showed antibacterial activity against S. aureus and E. coli, potentially due to increased ROS generation (up to 3 times), whereas the Ag+ materials exhibited a broader spectrum of activity, even inhibiting clinical strains of MRSA and P. aeruginosa. In particular, the Ag+ material with phenytoin sodium showed the ability to reduce biofilm development by up to 55% and inhibit bacterial growth in a "wound-like medium" by up to 89.33%.

Abstract:The rise of antibiotic resistance and the growing number of biofilm-related infections make bacterial infections a serious threat for global human health. Nanomedicine has entered into this scenario by bringing new alternatives to design and develop effective antimicrobial nanoweapons to fight against bacterial infection. Among them, mesoporous silica nanoparticles (MSNs) exhibit unique characteristics that make them ideal nanocarriers to load, protect and transport antimicrobial cargoes to the target bacteria and/or biofilm, and release them in response to certain stimuli. The combination of infection-targeting and stimuli-responsive drug delivery capabilities aims to increase the specificity and efficacy of antimicrobial treatment and prevent undesirable side effects, becoming a ground-breaking alternative to conventional antibiotic treatments. This review focuses on the scientific advances developed to date in MSNs for infection-targeted stimuli-responsive antimicrobials delivery. The targeting strategies for specific recognition of bacteria are detailed. Moreover, the possibility of incorporating anti-biofilm agents with MSNs aimed at promoting biofilm penetrability is overviewed. Finally, a comprehensive description of the different scientific approaches for the design and development of smart MSNs able to release the antimicrobial payloads at the infection site in response to internal or external stimuli is provided.

Abstract:The global increase in multidrug-resistant bacteria poses a challenge to public health and requires the development of new antibacterial materials. In this study, we examined the bactericidal properties of mesoporous silica-coated silver nanoparticles, varying the core sizes (28 nm and 51 nm). We also investigated gold nanoparticles (26 nm) coated with mesoporous silica coating as possible inert metal cores. To investigate the modification of antimicrobial activity after the surface charge change, we used silver nanoparticles with a 28 nm core coated with a mesoporous shell (16 nm) and functionalized with a terminal amine group. Furthermore, we developed a facile method to create Ag@mSiO2 doped films using polyurethane (IROGRAN) as a polymer matrix. The antibacterial effects of silver nanoparticles with different core sizes were analyzed against Gram-negative and Gram-positive bacteria relevant to the healthcare and food industry. The results demonstrated that gold nanoparticles were inert, while silver nanoparticles exhibited antibacterial effects against Gram-negative (Escherichia coli and Salmonella enterica subsp. enterica serovar Choleraesuis) and Gram-positive (Bacillus cereus) strains. In particular, the larger Ag@mSiO2 nanoparticles showed a minimum inhibitory concentration (MIC) and a minimum bactericidal concentration (MBC) of 18 µg/mL in the Salmonella strain. Furthermore, upon terminal amine functionalization, reversing the surface charge to positive values, there was a significant increase in the antibacterial activity of the NPs compared to their negative counterparts. Finally, the antimicrobial properties of the nanoparticle-doped polyurethane films revealed a substantial improvement in antibacterial efficacy. This study provides valuable information on the potential of mesoporous silica-coated silver nanoparticles and their applications in fighting multidrug-resistant bacteria, especially in the healthcare and food industries.

Abstract:The enormous influence of bacterial resistance to antibiotics has led researchers toward the development of various advanced antibacterial modalities. In this vein, nanotechnology-based devices have garnered interest owing to their excellent morphological as well as physicochemical features, resulting in augmented therapeutic efficacy. Herein, to overcome the multidrug resistance (MDR) in bacteria, we demonstrate the fabrication of a versatile design based on the copper-doped mesoporous silica nanoparticles (Cu-MSNs). Indeed, the impregnated Cu species in the siliceous frameworks of MSNs establish pH-responsive coordination interactions with the guest molecules, tetracycline (TET), which not only enhance their loading efficiency but also assist in their release in the acidic environment precisely. Subsequently, the ultrasmall silver nanoparticles-stabilized polyethyleneimine (PEI-SNP) layer is coated over Cu-MSNs. The released silver ions from the surface-deposited SNPs are capable of sensitizing the resistant strains through establishing the interactions with the biomembranes, and facilitate the generation of toxic free radicals, damaging the bacterial components. In addition to SNPs, Cu species impregnated in MSN frameworks synergistically act through the production of free radicals by participating in the Fenton-like reaction. Various physical characterization techniques for confirming the synthesis and successful surface modification of functional nanomaterials, as well as different antibacterial tests performed against MDR bacterial strains, are highly commendable. Remarkably, this versatile formulation has shown no significant toxic effects on normal mammalian fibroblast cells accounting for its high biocompatibility. Together, these biocompatible MSN-based trio-hybrids with synergistic efficacy and pH-responsive delivery of antibiotics potentially allow for efficient combat against MDR in bacteria.

Abstract:Nanoparticulate delivery of biocides has the potential to decrease levels of exposure to non-target organisms, and miminize long-term exposure that can promote the development of resistance. Silica nanoparticles are an ideal vehicle since they are inert, biocompatible, biodegradable, and thermally and chemically stable. Encapsulation of biocides within nanoparticulates can improve their stability and longevity and maximize the biocidal potential of hydrophobic volatile compounds. Herein, we have shown that the plant secondary metabolites allyl isothiocyanate and cinnamaldehyde demonstrated increased antimicrobial activity against Escherichia coli in planktonic form, when packaged into mesoporous silica nanoparticles. Furthermore, the biocide-loaded nanoparticles showed activity against Pseudomonas aeruginosa biofilms that have inherent resistance to antimicrobial agents. The delivery platform can also be expanded to traditional biocides and other non-conventional antimicrobial agents.

Abstract:Bacterial biofilms can initiate chronic infections that become difficult to eradicate. There is an unmet need for effective therapeutic strategies that control and inhibit the growth of these biofilms. Herein, light sensitive mesoporous silica nanoparticles (MSNs) with photothermal (PTT) and antimicrobial combined capabilities have been developed. These nanosystems have high therapeutic potential to affect the bacterial biofilm architecture and subsequently inhibit its growth. Nucleation of gold nanorods followed by the growth of a silica shell leads to a core@shell design (AuNR@MSN) with PTT properties. Incorporation of nitrosothiol groups (-SNO) with a heat liable linker, enables an enhanced nitric oxide release upon photothermal stimulation with near infrared radiation. Further loading of an antimicrobial molecule such as the levofloxacin (LEVO) antibiotic creates a unique nanoassembly with potential therapeutic efficacy against Staphylococcus aureus bacterial biofilms. A dispersion rate of the bacterial biofilm was evident when light stimuli is applied because impregnation of the nitrosothiol functionalized nanosystem with the antibiotic LEVO led to ca. 30% reduction but its illumination with near infrared (NIR) irradiation showed a biofilm reduction of ca. 90%, indicating that localized antimicrobial exposure and PTT improves the therapeutic efficacy. These findings envision the conception of near-infrared-activated nanoparticle carriers capable of combined therapy upon NIR irradiation, which enables photothermal therapy, together with the release of levofloxacin and nitric oxide to disrupt the integrity of bacterial biofilms and achieve a potent antimicrobial therapy.

Abstract:Tuberculosis remains today a major public health issue with a total of 9 million new cases and 2 million deaths annually. The lack of an effective vaccine and the increasing emergence of new strains of Mycobacterium tuberculosis (Mtb) highly resistant to antibiotics, anticipate a complicated scenario in the near future. The use of nanoparticles features as an alternative to antibiotics in tackling this problem due to their potential effectiveness in resistant bacterial strains. In this context, silver nanoparticles have demonstrated high bactericidal efficacy, although their use is limited by their relatively high toxicity, which calls for the design of nanocarriers that allow silver based nanoparticles to be safely delivered to the target cells or tissues. In this work mesoporous silica nanoparticles are used as carriers of silver based nanoparticles as antimycobacterial agent against Mtb. Two different synthetic approaches have been used to afford, on the one hand, a 2D hexagonal mesoporous silica nanosystem which contains silver bromide nanoparticles distributed all through the silica network and, on the other hand, a core@shell nanosystem with metallic silver nanoparticles as core and mesoporous silica shell in a radial mesoporous rearrangement. Both materials have demonstrated good antimycobacterial capacity in in vitro test using Mtb, being lower the minimum inhibitory concentration for the nanosystem which contains silver bromide. Therefore, the interaction of this material with the mycobacterial cell has been studied by cryo-electron microscopy, establishing a direct connection between the antimycobactericidal effect observed and the damage induced in the cell envelope.
Bioactive mesoporous silica nanostructures with anti-microbial and anti-biofilm properties

Abstract:The increasing rate of antibiotic resistant bacteria associated with nosocomial infections in severely ill patients has urged the need for new antibacterial therapies. Nanostructured materials represent emerging innovative approaches to controlled delivery of different antimicrobial drugs. Delivery systems encapsulating natural compounds with antibacterial effects, such as essential oils have shown a great potential. Herein we report the development of SiO2 mesoporous nanosystems loaded with eucalyptus (EUC), orange (ORA), and cinnamon (CIN) essential oils. These systems were characterized with respect to morphology (using scanning electron microscopy, SEM, and transmission electron microscopy, TEM), porosity (by BET and TEM analysis), chemical composition (by X-ray diffraction, XRD, and Fourier transform infrared spectrometry, FTIR) and loading capacity (by thermogravimetric analysis, TGA). The anti-bacterial and anti-adherence effects were tested against clinically relevant microbial species (Staphylococcus aureus ATCC 25923; Escherichia coli ATCC 25922; and Candida albicans ATCC 10231), while the biocompatibility was evaluated by in vitro tests with L929 mouse fibroblast cells.

Preparation and Study of the Antibacterial Applications and Oxidative Stress Induction of Copper Maleamate-Functionalized Mesoporous Silica Nanoparticles

Abstract:Mesoporous silica nanoparticles (MSNs) are an interesting class of nanomaterials with potential applications in different therapeutic areas and that have been extensively used as drug carriers in different fields of medicine. The present work is focused on the synthesis of MSNs containing a maleamato ligand (MSN-maleamic) and the subsequent coordination of copper(II) ions (MSN-maleamic-Cu) for the exploration of their potential application as antibacterial agents. The Cu-containing nanomaterials have been characterized by different techniques and the preliminary antibacterial effect of the supported maleamato-copper(II) complexes has been tested against two types of bacteria (Gram positive and Gram negative) in different assays to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The biological results showed a moderate antibacterial activity against Escherichia coli which motivated a more detailed study of the antibacterial mechanism of action of the synthesized maleamate-containing nanosystems and whose findings showed oxidative stress generation in bacterial cells. All the prepared nanomaterials were also tested as catalysts in the “solvent free” selective oxidation of benzyl alcohol, to observe if there is a potential correlation between the catalytic oxidation capacity of the materials and the observed oxidative stress in bacteria. This may help in the future, for a more accurate rational design of antibacterial nanosystems, based on their observed catalytic oxidation activity.

Abstract:In this manuscript, we propose a simple and versatile methodology to design nanosystems based on biocompatible and multicomponent mesoporous silica nanoparticles (MSNs) for infection management. This strategy relies on the combination of antibiotic molecules and antimicrobial metal ions into the same nanosystem, affording a significant improvement of the antibiofilm effect compared to that of nanosystems carrying only one of these agents. The multicomponent nanosystem is based on MSNs externally functionalized with a polyamine dendrimer (MSN-G3) that favors internalization inside the bacteria and allows the complexation of multiactive metal ions (MSN-G3-Mn+). Importantly, the selection of both the antibiotic and the cation may be done depending on clinical needs. Herein, levofloxacin and Zn2+ ion, chosen owing to both its antimicrobial and osteogenic capability, have been incorporated. This dual biological role of Zn2+ could have and adjuvant effect thought destroying the biofilm in combination with the antibiotic as well as aid to the repair and regeneration of lost bone tissue associated to osteolysis during infection process. The versatility of the nanosystem has been demonstrated incorporating Ag+ ions in a reference nanosystem. In vitro antimicrobial assays in planktonic and biofilm state show a high antimicrobial efficacy due to the combined action of levofloxacin and Zn2+, achieving an antimicrobial efficacy above 99% compared to the MSNs containing only one of the microbicide agents. In vitro cell cultures with MC3T3-E1 preosteoblasts reveal the osteogenic capability of the nanosystem, showing a positive effect on osteoblastic differentiation while preserving the cell viability.
[A case of asymptomatic posterior mediastinal pheochromocytoma diagnosed during the procedure of video-assisted thoracic surgery (VATS)].

Abstract:The patient was a 20-year-old male. On routine check-ups, a posterior mediastinal tumor was found radiologically and referred to our hospital for the operation. During the Video-Assisted Thoracic Surgery (VATS), sudden elevation of the blood pressure up to 270 mmHg was observed by the tumor palpation. At once we diagnosed as pheochromocytoma. Changing the VATS to usual thoracotomy the tumor was completely removed and histologically confirmed. After the operation the patient was uneventful and the value of norèpinephrine reduced to normal range from 2912 pg/ml. The posterior mediastinal pheochromocytoma is rare and this report is the 6 th case in Japan. Clinical characteristics were discussed in addition to the review of the literature.
Mesoporous Silica Nanoparticles Decorated with Polycationic Dendrimers for Infection Treatment

Abstract:This work aims to provide an effective and novel solution for the treatment of infection by using nanovehicles loaded with antibiotics capable of penetrating the bacterial wall, thus increasing the antimicrobial effectiveness. These nanosystems, named "nanoantibiotics", are composed of mesoporous silica nanoparticles (MSNs), which act as nanocarriers of an antimicrobial agent (levofloxacin, LEVO) localized inside the mesopores. To provide the nanosystem of bacterial membrane interaction capability, a polycationic dendrimer, concretely the poly(propyleneimine) dendrimer of third generation (G3), was covalently grafted to the external surface of the LEVO-loaded MSNs. After physicochemical characterization of this nanoantibiotic, the release kinetics of LEVO and the antimicrobial efficacy of each released dosage were evaluated. Besides, internalization studies of the MSNs functionalized with the G3 dendrimer were carried out, showing a high penetrability throughout Gram-negative bacterial membranes. This work evidences that the synergistic combination of polycationic dendrimers as bacterial membrane permeabilization agents with LEVO-loaded MSNs triggers an efficient antimicrobial effect on Gram-negative bacterial biofilm. These positive results open up very promising expectations for their potential application in new infection therapies.
The Influence of Silver-Containing Bionanomaterials Based on Humic Ligands on Biofilm Formation in Opportunistic Pathogens

Abstract:The uncontrolled use of antibiotics has led to a global problem of antimicrobial resistance. One of the main mechanisms of bacterial resistance is the formation of biofilms. In order to prevent the growth of antimicrobial resistance, it is crucial to develop new antibacterial agents that are capable of inhibiting the formation of biofilms. This makes this area of research highly relevant today. Promising candidates for these antibacterial agents are new bionanomaterials made from natural humic substances and silver nanoparticles. These substances have the potential to not only directly kill microorganisms but also penetrate biofilms and inhibit their formation. The goal of this study is to synthesize active pharmaceutical substances in the form of bionanomaterials, using ultradispersed silver nanoparticles in a matrix of coal humic substances, perform their characterization (NMR spectroscopy, TEM, and ICP-AES methods), and research their influence on biofilm formation in the most dangerous opportunistic pathogens (E. coli, Methicillin-resistant St. Aureus, K. pneumoniae, P. aeruginosa, St. aureus, A. baumannii, and K. Pneumonia). The results showed that all of the studied bionanomaterials had antibacterial activity against all of the opportunistic pathogens. Furthermore, they were found to have a suppressive effect on both pre-existing biofilms of these bacteria and their formation.
Cationic and Anionic Antimicrobial Agents Co-Templated Mesostructured Silica Nanocomposites with a Spiky Nanotopology and Enhanced Biofilm Inhibition Performance.

Abstract:Silica-based materials are usually used as delivery systems for antibacterial applications.In rare cases,bactericidal cationic surfactant templated silica composites have been reported as antimicrobial agents.However,their anti-bacterial efficacy is limited due to limited control in content and structure.Herein,we report a"dual active templating"strat-egy in the design of nanostructured silica composites with intrinsic antibacterial per-formance.This strategy uses cationic and anionic structural directing agents as dual templates,both with active antibacterial property.The cationic-anionic dual active templating strategy further contributes to antibacterial nanocomposites with a spiky surface.With controllable release of dual active antibacterial agents,the spiky nanocomposite displays enhanced anti-microbial and anti-biofilm properties toward Staphylococcus epidermidis.These findings pave a new avenue toward the designed synthesis of novel antibacterial nanocomposites with improved performance for diverse antibacterial applications.

Abstract:This research aims to improve antimicrobial materials based on functional silica nanoparticles. Three different methods were used in the study to create silica nanoparticles with other properties. The nanoparticles' morphological structures are porous, hollow, and filled with spherical forms. The surface of these nanoparticles was grafted with poly(1-vinyl-1,2,4-triazole) (PVTri). The morphological properties of nanocomposites were used for analysis. In contrast, thermal gravimetric analysis was used to characterize the thermal properties of nanocomposites (thermogravimetric analysis). The silica nanoparticles were evaluated for their in vitro antimicrobial activity against Escherichia coli, Staphylococcus aureus, and Saccharomyces cerevisiae using minimum inhibitory concentration measurement. Silica nanoparticles have different antifungal and antibacterial properties related to their structure. The cytotoxic effects of the silica nanoparticles on HaCaT cells were performed with an MTS assay. In this study, we observed that high doses of HSS and e-SiO2 decreased cell growth, while HSS and e-SiO2 composite with PVTri increased cell proliferation.

Abstract:Antimicrobial resistance (AMR), caused by microbial infections, has become a major contributor to morbid rates of mortality worldwide and a serious threat to public health. The exponential increase in resistant pathogen strains including Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) poses significant hurdles in the health sector due to their greater resistance to traditional treatments and medicines. Efforts to tackle infectious diseases caused by resistant microbes have prompted the development of novel antibacterial agents. Herein, we present selenium and copper oxide monometallic nanoparticles (Se-MMNPs and CuO-MMNPs), characterized using various techniques and evaluated for their antibacterial potential via disc diffusion, determination of minimum inhibitory concentration (MIC), antibiofilm, and killing kinetic action. Dynamic light scattering (DLS), scanning electron microscopy (SEM/EDX), and X-ray diffraction (XRD) techniques confirmed the size-distribution, spherical-shape, stability, elemental composition, and structural aspects of the synthesized nanoparticles. The MIC values of Se-MMNPs and CuO-MMNPs against S. aureus and E. coli were determined to be 125 μg/mL and 100 μg/mL, respectively. Time–kill kinetics studies revealed that CuO-MMNPs efficiently mitigate the growth of S. aureus and E. coli within 3 and 3.5 h while Se-MMNPs took 4 and 5 h, respectively. Moreover, CuO-MMNPs demonstrated better inhibition compared to Se-MMNPs. Overall, the proposed materials exhibited promising antibacterial activity against S. aureus and E. coli pathogens.

Abstract:Drug-resistant bacterial infections cause significant harm to public life, health, and property. Biofilm is characterized by overexpression of glutathione (GSH), hypoxia, and slight acidity, which is one of the main factors for the formation of bacterial resistance. Traditional antibiotic therapy gradually loses its efficacy against multi-drug-resistant (MDR) bacteria. Therefore, synergistic therapy, which regulates the biofilm microenvironment, is a promising strategy. A multifunctional nanoplatform, SnFe2O4-PBA/Ce6@ZIF-8 (SBC@ZIF-8), in which tin ferrite (SnFe2O4, denoted as SFO) as the core, loaded with 3-aminobenzeneboronic acid (PBA) and dihydroporphyrin e6 (Ce6), and finally coated with zeolite imidazole salt skeleton 8 (ZIF-8). The platform has a synergistic photothermal therapy (PTT)/photodynamic therapy (PDT) effect, which can effectively remove overexpressed GSH by glutathione peroxidase-like activity, reduce the antioxidant capacity of biofilm, and enhance PDT. The platform had excellent photothermal performance (photothermal conversion efficiency was 55.7 %) and photothermal stability. The inhibition rate of two MDR bacteria was more than 96 %, and the biofilm clearance rate was more than 90 % (150 μg/mL). In the animal model of MDR S. aureus infected wound, after 100 μL SBC@ZIF-8+NIR (150 μg/mL) treatment, the wound area of mice was reduced by 95 % and nearly healed. The serum biochemical indexes and H&E staining results were within the normal range, indicating that the platform could promote wound healing and had good biosafety. In this study, we designed and synthesized multifunctional nanoplatforms with good anti-drug-resistant bacteria effect and elucidated the molecular mechanism of its anti-drug-resistant bacteria. It lays a foundation for clinical application in treating wound infection and promoting wound healing.

Abstract:The sol–gel process emerges as an interesting strategy to obtain hybrid materials (organic–inorganic materials), it takes place under mild conditions and can produce nanoparticles with different shapes and sizes, leading to composite materials with multifunctional properties. In this paper will describe the synthesis and characterization of the mesoporous silica obtained by the sol–gel process as well as their potential applications as antimicrobial agents, biological markers, odontology, and others. Highly ordered mesoporous silica has gained attention from scientists because it exhibits large specific area and pore volume as well as low density. The preparation of mesoporous silica matrixes via the sol–gel process by using the surfactant cetyltrimethylammonium bromide and the copolymer Pluronic P-123 as structure-driving agents. The results revealed that samples V and VII behaved the most efficiently. Log10 CFU/mL decreased by more than two digits for Candida albicans, Staphylococcus aureus, and Streptococcus mutans and was statistically significant for all the tested microorganisms. Therefore, this study demonstrated that the functionalized mesoporous silica is a viable antimicrobial agent when it is incorporated into dental acrylic resin.
