Abstract:Prior research has shown that some personality traits are associated with cognitive outcomes and may confirm risk or protection against cognitive decline. The present study expands on previous work to examine the association between a more comprehensive set of psychological characteristics and cognitive performance in a diverse cohort of older adults. We also examine whether controlling for brain atrophy influences the association between psychological characteristics and cognitive function. A total of 157 older adults completed a battery of psychological questionnaires (Openness to Experience, Conscientiousness, Agreeableness, Neuroticism, Extraversion, positive affect, negative affect—sadness, negative affect—anger, sense of purpose, loneliness, grit, and self-efficacy). Cognitive outcomes were measured across multiple domains: episodic memory, semantic memory, executive function, and spatial ability. Baseline brain (MRI) variables included gray matter, hippocampus, and total white matter hyperintensity volume. Parallel process, multilevel models yielded intercept (individual cognitive domain scores) and linear slope (global cognitive change) random effects for the cognitive outcomes. Positive affect (β = 0.013, SE = 0.005, p = .004) and Openness (β = 0.018, SE = 0.007, p = .009) were associated with less cognitive change, independent of baseline brain variables and covariates. Greater sadness predicted more cognitive decline when controlling for covariates, but not brain atrophy. A variety of psychological characteristics were associated with the cross-sectional measures of cognition. This study highlights the important impact of positive and negative affect on reducing or enhancing the risk of longitudinal cognitive decline. Abstract:Introduction: Personality traits and neuropsychiatric symptoms such as neuroticism and depression share genetic overlap and have both been identified as risks factors for development of aging-related neurocognitive decline and Alzheimer's disease (AD). This study aimed to examine revised personality factors derived from the Temperament and Character Inventory, previously shown to be associated with psychiatric disorders, as predictors of neuropsychiatric, cognitive, and brain trajectories of participants from a population-based aging study. Methods: Mixed-effect linear regression analyses were conducted on data for the full sample ( N max = 1,286), and a healthy subsample not converting to AD-dementia during 25-year follow-up ( N max = 1,145), complemented with Cox proportional regression models to determine risk factors for conversion to clinical AD. Results: Two personality factors, Closeness to Experience (CE: avoidance of new stimuli, high anxiety, pessimistic anticipation, low reward seeking) and Tendence to Liabilities (TL: inability to change, low autonomy, unaware of the value of their existence) were associated with higher levels of depressive symptoms, stress (CE), sleep disturbance (TL), as well as greater decline in memory, vocabulary and verbal fluency in the full sample. Higher CE was additionally associated with greater memory decline across 25 years in the healthy subsample, and faster right hippocampal volume reduction across 8 years in a neuroimaging subsample ( N = 216). Most, but not all, personality-cognition associations persisted after controlling for diabetes, hypertension and cardiovascular disease. Concerning risks for conversion to AD, higher age, and APOE -ε4, but none of the personality measures, were significant predictors. Conclusion: The results indicate that personality traits associated with psychiatric symptoms predict accelerated age-related neurocognitive declines even in the absence of neurodegenerative disease. The attenuation of some personality effects on cognition after adjustment for health indicators suggests that those effects may be partly mediated by somatic health. Taken together, the results further emphasize the importance of personality traits in neurocognitive aging and underscore the need for an integrative (biopsychosocial) perspective of normal and pathological age-related cognitive decline.

Abstract:Research suggests that personality traits are associated with mild cognitive impairment (MCI), dementia, and mortality risk, but the timing of when traits are most important in the progression to dementia and the extent to which they are associated with years of cognitive health span are unclear. This project applied secondary data analysis to the Rush Memory and Aging Project (N = 1954; baseline Mage = 80 years; 74% female) over up to 23 annual assessments. Multistate survival modeling examined the extent to which conscientiousness, neuroticism, and extraversion, assessed using the NEO Five Factor Inventory, were associated with transitions between cognitive status categories and death. Additionally, multinomial regression models estimated cognitive health span and total survival based on standard deviation units of personality traits. Adjusting for demographics, depressive symptoms, and apolipoprotein (APOE) ε4, personality traits were most important in the transition from no cognitive impairment (NCI) to MCI. For instance, higher conscientiousness was associated with a decreased risk of transitioning from NCI to MCI, hazard ratio (HR) = 0.78, 95% CI [0.72, 0.85] and higher neuroticism was associated with an increased risk of transitioning from NCI to MCI, HR = 1.12, 95% CI [1.04, 1.21]. Additional significant and nonsignificant results are discussed in the context of the existing literature. While personality traits were not associated with total longevity, individuals higher in conscientiousness and extraversion, and lower in neuroticism, had more years of cognitive health span, particularly female participants. These findings provide novel understanding of the simultaneous associations between personality traits and transitions between cognitive status categories and death, as well as cognitive health span and total longevity. Morphological study of sinus flow in the confluence of sinuses

Abstract:The confluence of sinuses (CS; torcular herophili) is represented by the junction of the superior sagittal (SSS), straight (SS), occipital (OS), and two transverse sinuses (TS). The objective of this study was to interpret sinus flow around the CS by morphological investigation of the sinuses. This study is based on visual examination of dural venous sinuses in the region of the CS in 31 adult cadavers. In the inflow zone, we examined the direction of SSS and SS flow. In the communication zone, we examined the extent to which outflow sinuses communicate with other sinuses. In the outflow zone, we used the diameters of outflow sinuses to determine anatomical dominance. The SSS entered the CS via the right TS in 16 cases (51.6%) and via the center of the CS in 14 cases (45.2%). The SS entered via the center of the CS in 18 cases (58.1%) and via the left TS in 11 cases (35.5%). Outflow sinuses communicated freely in 26 cases (83.8%) and communicated partially in five cases (16.2%). Partial communication was the result of a septate CS. In terms of outflow, the right TS was dominant in 11 cases (35.5%), and in 18 cases (58.1%), outflow was symmetrical. The direction of SSS inflow was different from that of SS inflow, and partial communication was observed in five cases (16.1%). Therefore, the presence of a septum may be considered an anatomical factor, with implications in diagnosis or in the sacrifice of the outflow sinus of the CS. Clin. Fibrinogen in relation to degree and composition of coronary plaque on intravascular ultrasound in patients undergoing coronary angiography

Abstract:Objective The aim of this study was to provide additional insight into the role of fibrinogen in coronary artery disease by investigating the associations between plasma fibrinogen with both degree and composition of coronary atherosclerosis as determined by virtual histology-intravascular ultrasound. Patients and methods In 581 patients undergoing coronary angiography for acute coronary syndrome (ACS) or stable angina pectoris, preprocedural blood samples were drawn for fibrinogen, C-reactive protein (CRP), interleukin-6, and plasminogen activator inhibitor-1 measurements, and virtual histology-intravascular ultrasound of a nonculprit coronary artery was performed. The degree [plaque volume, plaque burden (PB), and lesions with PB≥70%] and the composition of coronary atherosclerotic plaque (fibrous, fibrofatty, dense calcium, necrotic core tissue, and thin-cap fibroatheroma lesions) were assessed. Results Fibrinogen showed a tendency toward a positive association with PB [&bgr; (95% CI): 2.55 (−0.52–5.61) increase in PB per ln(g/l) fibrinogen, P=0.09], which was driven significantly by an association in the ACS subgroup [&bgr; (95% CI): 4.11 (0.01–8.21) increase in PB per ln(g/l) fibrinogen, P=0.049]. Fibrinogen was also related to the presence of lesions with PB 70% or more in both the full cohort [OR (95% CI): 2.27 (1.17–4.43), P=0.016] and ACS patients [OR (95% CI): 2.92 (1.17–7.29), P=0.022]. All associations were independent of established cardiovascular risk factors, but not CRP. Interleukin-6 and plasminogen activator inhibitor-1 did not provide incremental value to fibrinogen when examining the associations with degree of atherosclerosis. Substantial associations with plaque composition were absent. Conclusion Fibrinogen is associated with degree of coronary atherosclerosis, especially in ACS patients. However, whether this association is independent of CRP might be questioned and needs further investigation.

Differences regarding the five-factor personality model in patients with subjective cognitive decline and mild cognitive impairment

Abstract:Personality and dementia are connected in different ways. A broad knowledge about personality and prodromal stages of dementia might be helpful to identify dementia as early as possible. Hence, personality differences between three cognitively impaired groups on the basis of patients' self-assessments of personality traits and connections between personality and cognitive functioning were examined via a cross-sectional study. The sample consisted of cognitively impaired patients (N = 133), aged 50 and older, who came to a memory clinic due to cognitive complaints. The test procedure encompassed a cognitive screening, the Neuropsychological Test Battery Vienna (NTBV), and self-assessment questionnaires such as the Big Five Plus One Persönlichkeitsinventar (B5PO). While patients with subjective cognitive decline (SCD) did not differ from those with non-amnestic mild cognitive impairment (naMCI) concerning the different personality traits, patients with amnestic mild cognitive impairment (aMCI) showed significantly lower scores for extraversion (p < 0.05), openness (p < 0.001), and empathy (p < 0.001) than patients with SCD as well as patients with naMCI. Thus, cognitively impaired groups mainly differ concerning personality traits depending on whether they do show memory decline or not.

Abstract:Abstract Background Five-factor model (FFM) personality traits have been associated consistently with risk of Alzheimer’s disease and related dementias (ADRD). Less is known about how these traits are associated with functioning in specific domains of cognitive function in older adulthood. Methods Participants ( N = 2865) were drawn from the 2016 Harmonized Cognitive Assessment Protocol sub-study of the Health and Retirement Study (HRS). Participants completed a battery of cognitive tasks that measured performance in five domains: Memory (eight tasks), speed-attention-executive (five tasks), visuospatial ability (three tasks), fluency (one task), and numeric reasoning (one task). Participants completed an FFM personality measure as part of the regular HRS assessment in either 2014 or 2016. Linear regression was used to examine the association between the traits and each cognitive task and composite scores for the five domains, controlling for age, sex, race, ethnicity, and education. We also tested whether the associations were moderated by these sociodemographic factors or mental status. Results Neuroticism was associated with worse performance on all of the cognitive tasks. Conscientiousness was associated with better performance across all five cognitive domains, although not necessarily with every task. Openness and Agreeableness were associated with better performance in all domains, except for numeric reasoning. Extraversion was associated with better speed-attention-executive and fluency. There was no robust evidence that the association between personality and cognition was moderated by sociodemographic characteristics or global cognitive function. Conclusions Personality traits have pervasive associations with functioning across five cognitive domains. Consistent with the literature on personality and risk of ADRD, Neuroticism and Conscientiousness were associated with cognitive performance in the expected direction in all domains. Extraversion was the only trait that showed domain-specific associations. The present research supports models of personality and health in the context of cognition and suggests that personality is associated with intermediate markers of cognitive health.

Abstract:Cognitive reserve explains differential susceptibility of cognitive performance to neuropathology. We investigated whether certain personality traits underlie cognitive reserve and are accordingly associated with better cognition and less cognitive decline in the presence of age-related brain changes. We included healthy adults aged 19-80 years for cross-sectional (N=399) and longitudinal (N=273, mean follow-up time=5 years, SD=0.7 years) analyses. Assessment of the BIG5 personality traits openness, conscientiousness, extraversion, agreeableness, and neuroticism was questionnaire-based. Each cognitive domain (perceptual speed, memory, fluid reasoning, vocabulary) was measured with up to six tasks. Cognitive domain-specific brain status variables were obtained by combining 77 structural brain measures into single scores using elastic net regularization. These brain status variables explained up to 43.1% of the variance in cognitive performance. We found that higher openness was associated with higher fluid reasoning and better vocabulary after controlling for brain status, age, and sex. Further, lower brain status was associated with a greater decline in perceptual speed only in individuals with low openness. We conclude that high openness benefits cognitive reserve.

Abstract:Extensive evidence has been found for the associations between personality traits and health. However, it remains unknown whether the relationships between personality and health show differential patterns across different life stages. The current research examined how the associations between the levels of and changes in the Big Five personality traits and different types of health outcomes (self-rated, physical, and physiological health outcomes) differ across ages over the life span (Sample 1, age range: 15-100) and during the aging process (Sample 2, age range: 50-109) in particular. Using data from the two large longitudinal studies-the Household, Income, and Labor Dynamics in Australia Survey and the Health and Retirement Study, we observed three important patterns. First, levels of and changes in personality traits were significantly associated with health across different life phases, and these effects were observed even in very old ages. Second, overall, the prospective relations between personality traits/changes in personality traits and health outcomes increased in strength in middle adulthood and/or early stages of late adulthood; however, the strength of their connections diminished in very old ages. Finally, there were some trait-specific and health outcome-specific patterns in the age-differential associations between personality and health. Findings from the present study contribute to enhancing our understanding of the personality-health link from a developmental perspective and provide critical information for the design and implementation of screening and interventions targeting health promotion. Abstract:Personality traits are broad constructs composed of nuances, operationalized by personality items, that can provide a more granular understanding of personality associations with health outcomes. This study examined the associations between personality nuances and incident dementia and evaluated whether nuances associations replicate across two samples. Health and Retirement Study (HRS, N = 11,400) participants were assessed in 2006/2008, and the English Longitudinal Study of Ageing (ELSA, N = 7,453) participants were assessed in 2010/2011 on personality and covariates. Dementia incidence was tracked for 14 years in the HRS and 8 years in ELSA. In both HRS and ELSA, higher neuroticism domain and nuances (particularly nervous and worry) were related to a higher risk of incident dementia, whereas higher conscientiousness domain and nuances (particularly responsibility and organization) were associated with a lower risk of dementia. To a lesser extent, higher extraversion (active), openness (broad-minded, curious, and imaginative), and agreeableness (helpful, warm, caring, and sympathetic) nuances were associated with a lower risk of dementia, with replicable effects across the two samples. A poly-nuance score, aggregating the effects of personality items, was associated with an increased risk of incident dementia in the HRS and ELSA, with effect sizes slightly stronger than those of the personality domains. Clinical, behavioral, psychological, and genetic covariates partially accounted for these associations. The present study provides novel and replicable evidence for specific personality characteristics associated with the risk of incident dementia.

Abstract:Objectives: Recent research has linked psychological (personality) factors and specific genetic risk polymorphisms to performance on neurocognitive phenotypes. We examined whether episodic or semantic memory performance is associated with (a) three personality traits (i.e. neuroticism, extraversion, and openness to experience), (b) two neurodegenerative-related polymorphisms (i.e. Apolipoprotein E (APOE; rs7412; rs429358), Clusterin (CLU; rs11136000)), and (c) cross-domain risk interactions (magnification effects). Methods: Linear growth models were examined to test independent associations between personality traits and declarative memory performance, and potential interaction effects with APOE and CLU genetic risk. Normal older adults (n = 282) with personality and genetic data from the Victoria Longitudinal Study were included at baseline and for up to 14 years of follow-up. Results: First, we observed that higher openness to experience levels were associated with better episodic and semantic memory. Second, three significant gene × personality interactions were associated with poorer memory performance at baseline. These synergistic effects are: (a) APOE allelic risk (ε4+) carriers with lower openness to experience levels, (b) CLU (no risk: T/T) homozygotes with higher extraversion levels, and (c) CLU (no risk: T/T) homozygotes with lower neuroticism levels. Conclusions: Specific neurodegenerative-related genetic polymorphisms (i.e. APOE and CLU) moderate and magnify the risk contributed by selected personality trait levels (i.e. openness to experience, extraversion) on declarative memory performance in non-demented aging. Future research could target interactions of other personality traits and genetic polymorphisms in different clinical populations to predict other neurocognitive deficits or transitions to cognitive impairment and dementia.

Abstract:Nerve conduction studies are normal in small fibre neuropathy and special methods such as skin biopsies or quantitative sensory testing are required for diagnosis. In skin biopsies, nerve fibres are stained immunohistochemically and loss of distal nerve endings can be quantified directly. Assessment of thermal detection thresholds is used to evaluate the function of the sensory thermal pathways, but cannot discriminate between central and peripheral lesions. Small fibre neuropathy is often associated with potentially treatable diseases, and treatment of neuropathic pain may be required.
Personality traits and brain health: a large prospective cohort study

Abstract:Personality has recently emerged as a critical determinant for multiple health outcomes. However, the evidence is less established for brain health, and the underlying mechanisms remain unclear. Here, utilizing data of 298,259 participants from the UK Biobank, five personality traits, including warmth, diligence, nervousness, sociability and curiosity, were constructed, and their relationships with brain disorders were examined with Cox regression and Mendelian randomization analyses. The results revealed consistent deleterious roles of nervousness, while the protective roles of warmth, diligence, sociability and curiosity in brain disorders were emphasized. Neuroimaging analyses highlighted the associations of personality traits with critical brain regions including the frontal cortex, temporal cortex and thalamus. Exploratory analyses revealed the mediating effects of neutrophil and high-density lipoprotein, indicating the contribution of inflammation and lipid metabolism to the associations between personality and brain health. This study provides a foundation for personality-oriented interventions in brain health, and it is necessary to validate our findings in other populations.

Abstract:BackgroundA recent study yielded first evidence that personality plays an important role in explaining the influence of a prominent APOE polymorphism on cognitive decline and Alzheimer’s disease (AD) in elderly humans. Adding to this, two earlier studies examined this polymorphism in the context of individual differences in temperament traits in young humans with mixed results. In general, research linking the prominent APOE ϵ2, ϵ3 and ϵ4 variants and human personality is of special interest, because an influence of this gene and its prominent polymorphism on personality in young adulthood could be of diagnostic value to predict AD and its development in later years.ResultsIn the present study N = 531 participants provided buccal swabs and filled in a self-report inventory measuring the Five Factor Model of Personality. No association between common genetic variations of the APOE gene (in detail the genotypes ϵ3/ϵ3, ϵ2/ϵ3 and ϵ3/ϵ4) and personality could be observed. The remaining genotypes, including the high risk constellation ϵ4/ϵ4 for AD, were too seldom to be tested.ConclusionsIn sum, the present study yielded no evidence for a direct link between common genetic variants of the APOE gene and personality in young adulthood.

Testing of significant changes in big-five personality factors over time in the presence and absence of memory impairment and life-related stress

Abstract:This study tests whether memory impairment and perceived increases in stress due to life changes independently contribute to similar personality changes, such as increased neuroticism and decreased conscientiousness. Longitudinal data from 12,912 participants aged 50+ from the Health and Retirement Study (2006-2020) were analyzed using Latent Growth Curve Models (LGCMs). Six LGCMs were simultaneously estimated to examine how changes in personality across three data points spanning 8 years are predicted by both the classification of memory impairment (MI) statuses assessed via the Telephone Interview for Cognitive Status and estimated increases in life stress. Decreases were observed in all Big Five personality factors (B = - 0.240/- 0.510, SE = 0.085/0.089) over the 8-year period. No significant differences in personality changes were found between MI cases and non-MI cases. Increased life stress was significantly associated with a rise in neuroticism (B = 0.587, SE = 0.094) and reductions in conscientiousness, extraversion, agreeableness, and openness (B ranges from - 0.223 to - 0.497). The findings emphasize adaptive/developmental shifts in personality in response to life stress rather than neuropathological ones related to memory impairment. Clinical assessment of personality changes in mid-to-late life should first consider psychological maladaptation to situational threats and challenges.

Abstract:Mounting evidence suggests considerable diversity in brain aging trajectories, primarily arising from the complex interplay between age, genetic, and environmental risk factors, leading to distinct patterns of micro- and macro-cerebral aging. The underlying mechanisms of such effects still remain unclear. We conducted a comprehensive association analysis between cerebral structural measures and prevalent risk factors, using data from 36,969 UK Biobank subjects aged 44-81. Participants were assessed for brain volume, white matter diffusivity, Apolipoprotein E (APOE) genotypes, polygenic risk scores, lifestyles, and socioeconomic status. We examined genetic and environmental effects and their interactions with age and sex, and identified 726 signals, with education, alcohol, and smoking affecting most brain regions. Our analysis revealed negative age-APOE-epsilon 4 and positive age-APOE-epsilon 2 interaction effects, respectively, especially in females on the volume of amygdala, positive age-sex-APOE-epsilon 4 interaction on the cerebellar volume, positive age-excessive-alcohol interaction effect on the mean diffusivity of the splenium of the corpus callosum, positive age-healthy-diet interaction effect on the paracentral volume, and negative APOE-epsilon 4-moderate-alcohol interaction effects on the axial diffusivity of the superior fronto-occipital fasciculus. These findings highlight the need of considering age, sex, genetic, and environmental joint effects in elucidating normal or abnormal brain aging.

Abstract:Abstract Background and Objectives This study aimed to critically examine current knowledge on the role of personality in successful aging, considering McAdams’ (Hooker & McAdams, 2003; McAdams, 2015) three-level, six-foci model of personality. The aim was also to pinpoint knowledge gaps that research should address in the future. Research Design and Methods We carried out a scoping review on the literature on successful aging and personality, following the five-step framework proposed by Arksey and O’Malley (2005) and further expanded by Levac et al. (2010). Results Research into the role of personality in successful aging has grown significantly over the last 20 years. However, the increasing number of publications on the topic was primarily accounted for by studies focused on layer 1 and particularly layer 2 of McAdams’ model of personality, with layer 3 being scarcely addressed. Research that included more than one layer of personality was rare. Discussion and Implications Our study pinpointed gaps that should be considered in future research in this area. These gaps were related to advancing towards agreed-upon definitions of successful aging and personality, broadening the scope of research on this topic, and integrating personality dimensions on research on successful aging.

Abstract:Objective: Associations have been found between five-factor model (FFM) personality traits and risk of developing specific predementia syndromes such as subjective cognitive decline (SCD) and mild cognitive impairment (MCI). The aims of this study were to: 1) Compare baseline FFM traits between participants who transitioned from healthy cognition or SCD to amnestic MCI (aMCI) versus non-amnestic MCI (naMCI); and 2) Determine the relationship between FFM traits and risk of transition between predementia cognitive states. Methods: Participants were 562 older adults from the Einstein Aging Study, 378 of which had at least one follow-up assessment. Baseline data collected included levels of FFM personality traits, anxiety and depressive symptoms, medical history, performance on a cognitive battery, and demographics. Follow-up cognitive diagnoses were also recorded. Results: Mann–Whitney U tests revealed no differences in baseline levels of FFM personality traits between participants who developed aMCI compared to those who developed naMCI. A four-state multistate Markov model revealed that higher levels of conscientiousness were protective against developing SCD while higher levels of neuroticism resulted in an increased risk of developing SCD. Further, higher levels of extraversion were protective against developing naMCI. Conclusions: FFM personality traits may be useful in improving predictions of who is at greatest risk for developing specific predementia syndromes. Information on these personality traits could enrich clinical trials by permitting trials to target individuals who are at greatest risk for developing specific forms of cognitive impairment. These results should be replicated in future studies with larger sample sizes and younger participants.

Abstract:Introduction: Personal views of aging (VoA) reflect individuals' perceptions, attitudes, and expectations regarding their aging selves. The present cross-sectional study was aimed at examining whether personality traits, as defined by the Big Five model, are associated with different VoA concepts related to both subjective age and awareness of age-related gains and losses in midlife and older age. Materials and methods: A sample of 224 participants aged 46–85 years reported their felt age and completed the Awareness of Age-Related Change (AARC) questionnaire, assessing perceptions of age-related gains (AARC-Gains) and losses (AARC-Losses) in various functioning domains, as well as the short version of the Big Five Inventory. Results: Linear regression models showed that Openness contributed to explain youthful subjective age. Extraversion explained higher AARC-Gains scores, whereas Emotional Stability, along with younger chronological age and perceiving better self-rated health, contributed to explaining lower AARC-Losses scores. Discussion: These findings confirm the relationship between personality traits and personal VoA. They further suggest that such an association varies depending on the VoA measure considered. They underscore the importance of considering personality among those individual characteristics capable of shaping personal VoA, with implications for the development of tailored interventions and the understanding of the underlying mechanisms linking personal VoA to health and longevity outcomes in midlife and older age.
