Abstract:A fragment encoding melittin cDNA from Apis cerana cerana fused with glutathione S-transferase gene was inserted into the multiple cloning site of the pBacFastHTb to construct a recombinant donor plasmid, pBacHT-GSTAccM, which was transposed to the target bacmid in E. coli (DH10) by Tn7 transposition function. Then the recombinant baculovirus Bacmid-GSTAccM was transfected into Tn-5B1-4 cells of the cabbage looper, Trichoplusia ni, mediated by lipofectin. The expressed protein of about 34 kDa was detected by Western blotting and triple antibody sandwich ELISA, indicating that the recombinant protein is the fusion protein of GSTAccM. Thin layer scanning showed that the expression level of GSTAccM was about 7% of the total cell protein. Purified and recovered recombinant melittin of A. c. cerana showed bioactivity in activating rabbit platelets to aggregate.

Then Bacmid-GFPTAccM recombinant genome DNA was used to transfect Tn-5B1-4 cell of the cabbage looper, Trichoplusia ni, mediated by lipofectin. The expressed protein band of about 34 kD was determined by SDS-PAGE, and the thin layer scanning showed that the expression amount of GFPT-AccM fusion protein was about 3% of the total cell protein. Western blotting and the cytopathic effect of Tn cell after infection of Bacmid-GFPTAccM at different time proved that the fusion protein GFPT-AccM had been successfully expressed in Tn-5B1-4 cell.

Abstract:A cDNA encoding an α-l-fucosidase from Drosophila melanogaster was obtained from the recombinant plasmid named pGEM-DmFuca and inserted into the pBacHTeGFPT vector to construct the recombinant donor plasmid which was transposed to the target AcBacmid in Escherichia coli (DH10) by Tn7 transposition function. The AcBacmid-GFP-DmFuca plasmid was used to transfect Tn-5B1-4 cells of the Cabbage looper Trichoplusia ni. SDS–PAGE analysis revealed a band of about 80kDa. Using a polyclonal antiserum raised against α-l-fucosidase protein from D. melanogaster Western blotting analysis confirmed that the fusion protein eGFP-DmFuca has been successfully expressed in a biologically active form in Tn-5B1-4 cells. The recombinant protein, containing the histidine-tag motif, was purified using an affinity chromatography column. In vitro binding assays the purified eGFP-DmFuca interacts with α-l-fucose residues present on the micropyle of the D. melanogaster eggshell, confirming that the α-l-fucosidase is a good candidate as receptor involved in gamete interactions in fruit fly.

Abstract:Two cDNA fragments encoding prepromelittin of two vespoidea species,female Polistes hebraeus and Vespula maculifrons were obtained by restriction enzyme digestion from the recombinant plasmids and inserting into the multiple cloning sites of the pBacHTb to construct recombinant donor plasmids,which were used to transpose to the target Bacmids in E.coli(DH10) by Tn7 transposition function.Then these two recombinant Bacmids,Bacmid-PhPPM and Bacmid-VmPPM,were transfected into Tn-5B1-4 cells of the cabbage looper,Trichoplusia ni,mediated by Lipofectin.The expressed proteins detected by ELISA indicate that the recombinant Bacmids have been expressed in Tn-5B1-4 cells.

Abstract:The Illumina GA II high-throughput sequencing technology was used to establish a transcriptome library from the bulbs of Lycoris sprengeri. A total of 24 million short read with an average length of 90 nt were generated, which produced 98,150 unigenes with a mean length of 385 nt. A total of 45.9% of these unigenes (45,052) were annotated using BLAST (E-value <= 1.0E(-5)) against the nr, SwissProt, Kyoto encyclopedia of genes and genomes (KEGG) and Clusters of orthologous groups (COG) databases. The unigenes were annotated with gene ontology (GO) and COG. Of these, 2,173 unigenes were found to be related to carbohydrate metabolism, and 544 were involved in starch and sucrose metabolism.

Abstract:In insects, melanism, a fundamental pigmentation process, is of significant importance in evolutionary biology due to its complex genetic foundation. We investigated the role of the RNA-binding gene Musashi (msi) in melanism in Laodelphax striatellus, a Hemiptera species. We identified a single L. striatellus msi homolog, Lsmsi, encoding a 357 amino acid protein with 2 RNA recognition motifs. RNA interference-mediated knockdown of LsMsi resulted in complete body melanism and increased cuticular permeability. Additionally, we found the involvement of G protein-coupled receptor A42 and tyrosine hydroxylase (Th) in L. striatellus melanism. Knockdown of LsTh lightened the epidermis, showing dehydration signs, while LsA42 knockdown enhanced LsTh expression, leading to melanism. Surprisingly, Lsmsi knockdown decreased both LsA42 and LsTh expression, which was expected to cause whitening but resulted in melanism. Further, we found that Lsmsi influenced downstream genes like phenoloxidase homolog LsPo and dopa decarboxylase (Ddc) homolog LsDdc in the tyrosine-mediated melanism pathway. Extending to Nilaparvata lugens and Sogatella furcifera, we demonstrated the conserved role of msi in melanism among Delphacidae. Given MSI proteins' roles in cancer and tumors in vertebrates, our study is the first to link msi in insects to Delphacidae body color melanization via the tyrosine-mediated pathway, offering fresh perspectives on the genetic basis of insect melanism and msi functions.
An unusual myosuppressin from the blood-feeding bug Rhodnius prolixus

Abstract:The myosuppressin (MS) gene was cloned from a central nervous system (CNS) cDNA library of the hematophagous insect Rhodnius prolixus and is predicted to contain two introns and three exons. The mRNA transcribed from the myosuppressin gene encodes an 88 amino acid prepropeptide, which results in a mature decapeptide after post-translational modification. When compared with the myosuppressins isolated from other insects, the R. prolixus myosuppressin has a unique amino acid sequence (pQDIDHVFMRFamide), with isoleucine (I) in position 3 and methionine (M) in position 8. Reverse transcriptase (RT)-PCR shows that Rhopr-MS is expressed in the CNS and posterior midgut in R. prolixus and immunohistochemistry suggests that an RFamide-like peptide is present in endocrine-like cells in the midgut. Physiological assays using Rhopr-MS indicate that, despite the unusual M at position 8, it still retains myoinhibitory activity, inhibiting the frequency and reducing the amplitude of contractions in the anterior midgut and hindgut, and decreasing heart rate.
[Expression and Subcellular Location of NSm Protein of Tomato Spotted Wilt Virus in Plant and Insect Cells].

Abstract:OBJECTIVE:Expression and subcellular location of NSm protein of Tomato spotted wilt virus were studied using plant and insect cells.METHODS:First, the NSm gene, located on the ambisense M RNA segment of tomato spotted wilt virus, was cloned into the pCHF3 vector which includes a GFP gene. Agrobacterium-mediated transient expression from N. benthamiana leaves was used to study the location of NSm in plant cells. Second, to test whether plant-specific components were involved in tubule formation, the NSm gene was also expressed in a heterologous expression system, i. e., insect cells. T. ni (Tn) cells were infected with a recombinant baculovirus expressing the NSm gene.RESULTS:NSm-GFP fusion proteins diffused in the tobacco epidermal cells and were located at the edge of the cell walls. These proteins can also form discontinuous green fluorescent spots at the plasmodesmata, which were sometimes present in pairs between two neighboring cells. However, GFP proteins expressed alone distributed evenly around the cell wall and in the nucleus. In the entomic Tn cells, NSm proteins formed a large number of tubular structures extending from the surface.CONCLUSION:These findings suggest that NSm protein target the plasmodesmata specifically in plant cells, and they also could form tubular structures on the surface when expressed in entomic Tn cells.
Deciphering cellular heterogeneity in Spodoptera frugiperda midgut cell line through single cell RNA sequencing

Abstract:Using the 10x Genomics Chromium single-cell RNA sequencing (scRNA-seq) platform, we discovered unexpected heterogeneity in an established cell line developed from the midgut of the Fall armyworm, Spodoptera frugiperda, a major global pest. We analyzed the sequences of 18,794 cells and identified ten unique cellular clusters, including stem cells, enteroblasts, enterocytes and enteroendocrine cells, characterized by the expression of specific marker genes. Additionally, these studies addressed an important knowledge gap by investigating the expression of genes coding for respiratory and midgut membrane insecticide targets classified by the Insecticide Resistance Action Committee. Dual-fluorescence tagging method, fluorescence microscopy and fluorescence-activated cell sorting confirmed the expression of midgut cell type-specific genes. Stem cells were isolated from the heterogeneous population of SfMG-0617 cells. Our results, validated by KEGG and Gene Ontology analyses and supported by Monocle 3.0, advance the fields of midgut cellular biology and establish standards for scRNA-seq studies in non-model organisms.
An Unusual Alternative Splicing of Enbocin in Silkworm,Bombyx Mori

Abstract:Antimicrobial peptides are the most important substance for insect in defending themselves from infection of microorganism.In this study,three kinds of microorganisms,E.coli JM109,Agrobacterium LBA4404 and Bombyx mori nucleopolyhedrovirus(BmNPV),were used to infect silkworms and study the inductive expression of enbocin gene in silkworm hemocyte and fat body.The results showed that,in hemocyte,there was low level expression in all the treatments;and in fat body,there was very little difference in its expression level before and after BmNPV treatment,while a specific expression was observed after treatments with JM109 and LBA4404.This specific expression had a compensation effect to the normal expression of enbocin gene.Cloning and sequencing of the normal transcript and the specific transcript revealed that a change in the exon/intron splice site has occurred to the bacterium-induced specific transcript(GenBank accession number: FJ373019).The splice site change had allowed both exons of the transcript to include partial nucleotides of the intron,which created a premature stop codon in the spliced mRNA.Translation of this mRNA produced a truncated polypeptide that lacked the Cecropin domain in its C-terminus.

Abstract:Aminopeptidase H11, an integral membrane glycoprotein present only in the gut of Haemonchus contortus , could provide substantial protection as shown by 90% reduction in fecal egg counts, while its recombinant version expressed in E. coli induced little. To investigate the characteristics further, we amplified mRNA of H11 gene via reverse transcriptase polymerase chain reaction, followed by isolation of its 1,517-bp 5′-flanking region and determination of its genomic organization. The H11 gene contained 25 exons separated by 24 introns and spans 14,959 bp of genomic DNA. Analysis of the 1,517 bp 5′-flanking region of the H11 gene revealed a putative “TATA-less” promoter. Partial sequences of the last exon and its 3′-UTR of H11 isoform H11-4 were also identified upstream to the H11 gene with the same transcription orientation. The 1,517-bp 5′-flanking region and part of the first exon of the H11 gene were subcloned into the vector upstream of green fluorescence protein reporter gene and microinjected into the gonads of Caenorhabditis elegans . The transformed animals exhibited fluorescence in the distal intestine in the L4 larvae stage and adult worms. This study characterized gene structure of aminopeptidase H11, demonstrated different transcriptional pattern of its promoter region between free-living and blood-sucking nematode species, and highlights the utility of C. elegans as a heterologous system to study the biology roles of H11 isoforms.

Abstract:Ahead of Print article withdrawn by publisher. BACKGROUND Calciphylaxis in a nondialysis patient is a rare condition and is characterized by calcific deposition in tissue. We present a case of calciphylaxis in a nondialysis patient who was diagnosed by clinical presentation and skin biopsy and was treated with sodium thiosulfate with improvement of skin lesions. CASE A 43-year-old female with type 2 diabetes and atrial fibrillation taking oral anticoagulation medication presented with reddish drainage from the right buttock. On physical examination, a large perirectal abscess overlying necrosis was found. She also developed acute kidney injury with creatinine of 3.7 mg/dL at peak from 0.8 mg/dL at baseline. She received antibiotics intravenously and wound debridement. During hospitalization, she developed areas of numerous painful erythematous lesions with central dusky necrosis on bilateral lower extremities. Punch biopsy was done, which initially revealed small-vessel vasculitis. However, those lesions did not respond to steroid therapy. A second biopsy was done showing extensive fat necrosis and medial calcification of vessel walls consistent with calciphylaxis. She was treated with high-flow oxygen and sodium thiosulfate intralesionally and intravenously for 6 months. The lesions remarkably reduced in size and were less painful on follow-up. CONCLUSION High-dose oxygen and sodium thiosulfate could potentially be effective treatments for calciphylaxis in nondialysis patients. .
Complex RNA Secondary Structures Mediate Mutually Exclusive Splicing of Coleoptera <i>Dscam1</i>

Abstract:Mutually exclusive splicing is an important mechanism for expanding protein diversity. An extreme example is the Down syndrome cell adhesion molecular (Dscam1) gene of insects, containing four clusters of variable exons (exons 4, 6, 9, and 17), which potentially generates tens of thousands of protein isoforms through mutually exclusive splicing, of which regulatory mechanisms are still elusive. Here, we systematically analyzed the variable exon 4, 6, and 9 clusters of Dscam1 in Coleoptera species. Through comparative genomics and RNA secondary structure prediction, we found apparent evidence that the evolutionarily conserved RNA base pairing mediates mutually exclusive splicing in the Dscam1 exon 4 cluster. In contrast to the fly exon 6, most exon 6 selector sequences in Coleoptera species are partially located in the variable exon region. Besides, bidirectional RNA-RNA interactions are predicted to regulate the mutually exclusive splicing of variable exon 9 of Dscam1. Although the docking sites in exon 4 and 9 clusters are clade specific, the docking sites-selector base pairing is conserved in secondary structure level. In short, our result provided a mechanistic framework for the application of long-range RNA base pairings in regulating the mutually exclusive splicing of Coleoptera Dscam1.
Cloning of Precursors for Two MIH/VIH-Related Peptides in the Prawn, Macrobrachium Rosenbergii

Abstract:Two cDNA clones (634 and 1366 bp) encoding MIH/VIH (molt-inhibiting hormone/vitellogenesis-inhibiting hormone)-related peptides were isolated and sequenced from a Macrobrachium rosenbergii eyestalk ganglia cDNA library. The clones contain a 360 and 339 bp open-reading frame, and their conceptually translated peptides consist of a 41 and 34 amino acid signal peptide, respectively, and a 78 amino acid residue mature peptide hormone. The amino acid sequences of the peptides exhibit higher identities with other known MIHs and VIH (44–69%) than with CHHs (28–33%). This is the first report describing the cloning and sequencing of two MIH/VIH-related peptides in a single crustacean species. Transcription of these mRNAs was detected in the eyestalk ganglia, but not in the thoracic ganglia, hepatopancreas, gut, gill, heart, or muscle.
PXGS: a Poly-Transgene Expression System based on Mutually Exclusive Splicing of Dscam

Abstract:Biologists often need to investigate multiple genes simultaneously in an organism. However, it is currently not possible to express more than a few transgenes in an animal under conditional control. Here, we developed a technique based on the mutually exclusive splicing of the Down Syndrome Cell Adhesion Molecule1 (Dscam1) gene in Drosophila melanogaster to achieve simultaneous transgene expression of 12 genes at a time. We show that the hypervariable Dscam1 exon 4 region maintains its alternative splicing when placed in a UAS expression vector. Each of the twelve exon 4 alternates can be replaced with an exogenous gene of at least 10 kilobases and will express properly in vivo all under conditional genetic control. We demonstrate the expression of four different fluorophores placed in different exon 4 alternate positions in neural and non-neural cells in vivo. We validated the technique by rewiring Drosophila sensory neuron axons in vivo by simultaneously expressing several cell surface receptors within the neuron. This technology will also enable Drosophila melanogaster as a model system for synthetic biology research.

A Spatially Adaptive Gradient-Projection Algorithm to Remove Coding Artifacts of H.264

Abstract:In this paper, we propose a spatially adaptive gradient-projection algorithm for the H.264 video coding standard to remove coding artifacts using local statistics. A hybrid method combining a new weighted constrained least squares (WCLS) approach and the projection onto convex sets (POCS) approach is introduced, where weighting components are determined on the basis of the human visual system (HVS) and projection set is defined by the difference between adjacent pixels and the quantization index (QI). A new visual function is defined to determine the weighting matrices controlling the degree of global smoothness, and a projection set is used to obtain a solution satisfying local smoothing constraints, so that the coding artifacts such as blocking and ringing artifacts can be simultaneously removed. The experimental results show the capability and efficiency of the proposed algorithm.

Abstract:Rous sarcoma virus was shown to induce in chicken embryo fibroblasts (CEF) a 4.1-kilobase mRNA (designated CEF-147) encoding a 603-amino acid protein. Analysis of the protein sequence showed that it shared 59% amino acid identity with sheep prostaglandin G/H synthase, the enzyme that catalyzes the rate-limiting steps in the production of prostaglandins. Significant differences, at both the protein and mRNA levels, existed between the src oncogene product-inducible prostaglandin synthase and the protein isolated and cloned from sheep seminal vesicle, suggesting that the src-inducible prostaglandin synthase may be a new form of the enzyme. A distinguishing feature of src-inducible prostaglandin synthase mRNA is its low abundance in nonproliferating chicken embryo fibroblasts and its relatively high abundance in src-transformed cells. Additionally, the majority of the src-inducible prostaglandin synthase RNA present in nonproliferating cells was found to be nonfunctional because of the presence of an unspliced intron that separated the signal peptide from the remainder of the protein. Upon mitogenic stimulation, this intron was removed, resulting in the induction of fully-spliced CEF-147 mRNA.
Splicing of scorpion toxin gene BmKK2 in HEK 293T cells

Abstract:Using GFP as a reporter gene, splicing of scorpion toxin gene BmKK2 was investigated in cultured HEK 293T cells. The results of RT-PCR and western blotting showed that BmKK2's intron could be recognized and spliced in cultured HEK 293T cells. At the same time, a cryptic splicing site of BmKK2 gene was found at the 91st nucleotide site of the second exon, which is a typical form of alternative splicing. For the first time, alternative splicing would partially explain the diversity of scorpion toxins at the gene level. Moreover, replacing BmKK2's intron with BmP03's intron (an artificial BmKK2-BmP03 mosaic gene) did not affect the intron's recognition and splicing, but increased the expression of the toxin-GFP fusion protein by fluorescence imaging, which indicated that both introns may regulate the expression of toxin-GFP fusion protein. The artificial BmKK2-BmP03 mosaic gene was also spliced into two kinds of mRNA molecules, which showed that sequence of intron was not absolutely conserved. The results suggested that introns of scorpion toxin genes BmKK2 and BmP03 increase the diversity of scorpion toxins and regulate the expression of their genes.
Molecular characterization and cell-specific expression of an ion transport peptide in the tobacco hornworm, Manduca sexta

Abstract:The crustacean hyperglycemic hormone (CHH) peptides regulate diverse physiological processes from reproduction to metabolism and molting in arthropods. In insects, the ion transport peptides (ITP), also members of the CHH family, have only been implicated in ion transport. In this study, we sequenced a nucleotide fragment spanning the conserved A1/A2 region of the putative CHH/ITP gene. This fragment was amplified from larval cDNA of the tobacco hornworm, Manduca sexta and showed a high degree of sequence conservation with the same region from other insects and, to a lesser degree, with that of crustacean species, suggesting the presence of a Manduca-specific CHH/ITP mRNA (MasITP mRNA). CHH-like immunocytochemical analyses with two crustacean antisera (from Carcinus maenas and Cancer pagurus) identified the presence of CHH-like immunoreactivity in nervous tissue of all developmental stages, but not in the gut of M. sexta. Specifically, CHH-like peptides localized to paired type IA(2) neurosecretory cells of the pars lateralis of the brain (projecting ipsilaterallly to the corpora cardiaca-allata complex) and to neurosecretory cells and transverse nerves of the ventral nerve cord in larvae, pupae, and adults. The distribution of the putative MasITP peptide shifted during development in a manner consistent with metamorphic reorganization. A comparison of hemolymph equivalents of CHH detected by enzyme-linked immunosorbent assay with CHH-like immunoreactivity in transverse nerves provided evidence for the release of MasITP from the transverse nerves into the hemolymph at insect ecdysis. These data suggest the presence of an insect ITP in M. sexta and a role for this hormone during ecdysis.
Splice-site mutations cause Rrp6-mediated nuclear retention of the unspliced RNAs and transcriptional down-regulation of the splicing-defective genes

Abstract:Background: Eukaryotic cells have developed surveillance mechanisms to prevent the expression of aberrant transcripts. An early surveillance checkpoint acts at the transcription site and prevents the release of mRNAs that carry processing defects. The exosome subunit Rrp6 is required for this checkpoint in Saccharomyces cerevisiae, but it is not known whether Rrp6 also plays a role in mRNA surveillance in higher eukaryotes. Methodology/principal findings: We have developed an in vivo system to study nuclear mRNA surveillance in Drosophila melanogaster. We have produced S2 cells that express a human beta-globin gene with mutated splice sites in intron 2 (mut beta-globin). The transcripts encoded by the mut beta-globin gene are normally spliced at intron 1 but retain intron 2. The levels of the mut beta-globin transcripts are much lower than those of wild type (wt) ss-globin mRNAs transcribed from the same promoter. We have compared the expression of the mut and wt beta-globin genes to investigate the mechanisms that down-regulate the production of defective mRNAs. Both wt and mut beta-globin transcripts are processed at the 3', but the mut beta-globin transcripts are less efficiently cleaved than the wt transcripts. Moreover, the mut beta-globin transcripts are less efficiently released from the transcription site, as shown by FISH, and this defect is restored by depletion of Rrp6 by RNAi. Furthermore, transcription of the mut beta-globin gene is significantly impaired as revealed by ChIP experiments that measure the association of the RNA polymerase II with the transcribed genes. We have also shown that the mut beta-globin gene shows reduced levels of H3K4me3. Conclusions/significance: Our results show that there are at least two surveillance responses that operate cotranscriptionally in insect cells and probably in all metazoans. One response requires Rrp6 and results in the inefficient release of defective mRNAs from the transcription site. The other response acts at the transcription level and reduces the synthesis of the defective transcripts through a mechanism that involves histone modifications.
