Abstract:The complex coacervation of oppositely charged polyelectrolytes is an important issue, which is relevant to many biological and industrial applications. While various biomolecules have been observed to form hierarchical multiphase structures in cells, its mechanism is still not fully understood. Here, we theoretically study the complex coacervation between the polyanion C and polycations A and B in solution and focus on the influence of charge sequence along the polyions on the multiphase coacervation. The electrostatic free energy is calculated with random phase approximation, and the phase diagrams are constructed by using the convex hull algorithm. It is revealed that the large asymmetry of charge patterns between A and B chains may induce the multiphase separation, driving the formation of two condensed phases, AC coacervate and BC coacervate, coexisting with a dilute phase. On the basis of our result, we propose a good criterion to determine if multiphase separation occurs or not. Furthermore, we analyze the effect of charge sequence of polyanion C as well as the addition of salt on the multiphase coacervation. This work provides insights into the underlying physics of sequence-dependent electrostatic interactions and the design of complex coacervates of polyelectrolyte mixtures.

Abstract:The liquid-liquid phase separation of biopolymers in living cells contains multiple interactions and occurs in a dynamic environment. Resolving the regulation mechanism is still a challenge. In this work, we designed a series of peptides (XXLY) 6 SSSGSS and studied their complexation and coacervation behavior with single-stranded oligonucleotides. The “X” and “Y” are varied to combine known amounts of charged and non-charged amino acids, together with the introduction of secondary structures and pH responsiveness. Results show that the electrostatic interaction, which is described as charge density, controls both the strength of complexation and the degree of chain relaxation, and thus determines the growth and size of the coacervates. The hydrophobic interaction is prominent when the charges are neutralized. Interestingly, the secondary structures of peptides exhibit profound effect on the morphology of the phases, such as solid phase to liquid phase transition. Our study gains insight into the phase separation under physiological conditions. It is also helpful to create coacervates with desirable structures and functions.

Abstract:Polyelectrolyte (PE) hydrogels are ubiquitous in nature.Tailoringthe interactions between PE gels and salt provides versatile implementsto overcome their critical drawbacks such as weak and brittle nature.Adding salts to PE gels introduces both counterions and co-ions, andthe variations of PE chain conformations and their macroscopic propertieshave been mainly attributed to the interplay between PE charged groupsand their counterions, yet the role of co-ions is generally ignored.This work demonstrates that by changing co-ion species with largeratomic sizes, monovalent haloids are capable of triggering the phaseseparation of common poly(acrylic acid) single network (PAAc-SN) hydrogelsinto polymer-sparse and polymer-rich regions, accompanied by remarkableenhancement in strength and toughness. The capabilities of co-ionsinducing phase separation of PAAc-SN gels follow a reverse Hofmeisterseries: Cl- < Br- < I-. Due to the enhanced polymer-polymer interactionsand viscoelastic energy dissipation, the phase-separated PAAc-SN gelsexhibit exceptional mechanical properties, with fracture stress andtearing energy of 96.7-folds and 1636-folds larger than those of theas-prepared gels, respectively. The largest fracture stress and tearingenergy reach 8.8 & PLUSMN; 1.3 MPa and 29.8 & PLUSMN; 2.4 kJ/m(2), exceeding most state-of-the-art PE hydrogels. A combination ofFTIR, H-1 NMR, and all-atom molecular dynamics simulationsreveals that the reverse Hofmeister series-aided phase separationoriginates from changes of network chain conformation through thefollowing influences rooted in the larger size of co-ions: (i) enhancinghydrophobic polymer-polymer interactions; (ii) acceleratingthe ion pair formation between carboxyl groups and their sodium counterions.These results not only provide a new method for hydrogel strengtheningand toughening but also emphasize the significant role of co-ionsin tuning the properties of PE gels.
Structure and Dynamics of Hybrid Colloid-Polyelectrolyte Coacervates: Insights from Molecular Simulations

Abstract:Electrostatic interactions in polymeric systems are responsible for a wide range of liquid-liquid phase transitions that are of importance for biology and materials science. Such transitions are referred to as complex coacervation, and recent studies have sought to understand the underlying physics and chemistry. Most theoretical and simulation efforts to date have focused on oppositely charged linear polyelectrolytes, which adopt nearly ideal-coil conformations in the condensed phase. However, when one of the coacervate components is a globular protein, a better model of complexation should replace one of the species with a spherical charged particle or colloid. In this work, we perform coarse-grained simulations of colloid-polyelectrolyte coacervation using a spherical model for the colloid. Simulation results indicate that the electroneutral cell of the resulting (hybrid) coacervates consists of a polyelectrolyte layer adsorbed on the colloid. Power laws for the structure and the density of the condensed phase, which are extracted from simulations, are found to be consistent with the adsorption-based scaling theory of coacervation. The coacervates remain amorphous (disordered) at a moderate colloid charge, $Q$, while an intra-coacervate colloidal crystal is formed above a certain threshold, at $Q > Q^{*}$. In the disordered coacervate, if $Q$ is sufficiently low, colloids diffuse as neutral non-sticky nanoparticles in the semidilute polymer solution. For higher $Q$, adsorption is strong and colloids become effectively sticky. Our findings are relevant for the coacervation of polyelectrolytes with proteins, spherical micelles of ionic surfactants, and solid organic or inorganic nanoparticles.

Abstract:It is well-known that the phase behavior and physicochemical and adhesive properties of complex coacervates are readily tuneable with the salt concentration of the medium. For toxicity reasons, however, the maximum applicable salt concentration in biomedical applications is typically low. Consequently, other strategies must be implemented in order to optimize the properties of the resulting complex coacervates. In this work, the effect of the charge density of a strong polyanion on the properties of complex coacervates was studied. To control this charge density, statistical anionic/charge-neutral hydrophilic copolymers were synthesized by means of an elegant protection/deprotection strategy and subsequently complexed with a strong polycation. The resulting complexes were observed to have an increasing water content as well as faster relaxation dynamics, with either increasing salt concentration or decreasing charge density. Time-salt and time-salt-charge density superpositions could be performed and showed that the relaxation mechanism of the complex coacervates remained unchanged. When the charge density was decreased, lower salt concentration complexes became suitable for viscoelastic adhesion with improved injectability. Such complex coacervates are promising candidates for injectable biomedical adhesives.

Abstract:Polyelectrolyte complex coacervates represent a wide class of materials with applications ranging from coatings and adhesives to pharmaceutical technologies. They also underpin multiple biological processes, which are only now beginning to be deciphered. The means by which molecular-scale architecture propagates into macroscopic structure, thermodynamics, and dynamics in complex coacervates is of central concern in physics, chemistry, biology, and materials science. How does polyion charge sequence dictate thermodynamic behavior? How does one tailor rheology or interfacial tension using macromolecular architecture? What emergent functionality from polymer complex coacervates has biological consequences? Recent developments in coacervate science shed light on many of these issues and raise exciting new challenges for the close integration of theory, simulations, and experiment.

Abstract:Polymers with electric charge, known as polyelectrolytes, are well known to form complex coacervates, which have vital implications in various biological processes and beyond. While significant advancements have been made in comprehending the molecular interactions that complex coacervation, the interactions that the coacervates against coalescence present an intricate experimental challenge and remain a subject of ongoing investigation. In a recent experimental study, polydiallyldimethylammonium chloride polycationic (PDDA) and anionic adenosine triphosphate (ATP) coacervates have been shown to stabilize upon transferring them to deionized water. Here, we perform molecular dynamics simulations of PDDA-ATP coacervates both in supernatant and in DI water, to understand the ion dynamics and structure within stable coacervates. We produced and analyzed an aggregated sum of 63 simulation data of PDDA-ATP coacervates in explicit water when they are in supernatant and deionized (DI) water. We found that discarding the supernatant and transferring the coacervates to DI water causes an immediate ejection of a significant amount (more than 50%) of small ions ( and ) from the coacervates to the bulk solution. Subsequently, the DI water environment alters the ionic density profiles in coacervates and the surface ion dynamics. We calculated a notable slowdown for the coacervate ions when they were transferred to the DI water. These results suggest that the initial ejection of the ions from the coacervates in DI water potentially brings the outer layer of the coacervates to a physically bound state that prevents or slows down the further mobility of ions.

Abstract:The liquid-liquid phase separation of a polyelectrolyte solution containing one type of negatively and two types of positively charged polymers with different charge densities is studied theoretically by random phase approximation (RPA). It is predicted that multicoacervate phases could coexist, driven purely by electrostatic correlations. The asymmetry of the linear charge density could induce an effective immiscibility between two positively charged polyelectrolytes, leading to the multiphase separation. Adding salt will induce the disappearance of the dilute phase, forming two coexisting complex phases, instead of fusion between coacervates. Raising temperature could either induce a two coexisting complex phase, or a dilute phase coexisting with a coacervate phase, depending on the bulk concentration. Our predictions are in good agreement with experiments and provide insights in the further designing of the multiphase coacervation system.

Abstract:Polyelectrolyte complexes are omnipresent both in nature and in the technological world, including nucleotide condensates, biological marine adhesives, food stabilizers, encapsulants, and carriers for gene therapy. However, the true phase behavior of complexes, resulting from associative phase separation of oppositely charged polyelectrolytes, remains poorly understood. Here, we rely on complementary experimental and simulation approaches to create a complete quantitative description of the phase behavior of polyelectrolyte complexes that represents a significant advance in our understanding of the underlying physics of polyelectrolyte complexation. Experiments employing multiple approaches with model polyelectrolytesoppositely charged polypeptides poly(l-lysine) and poly(d,l-glutamic acid) of matched chain lengthsled to phase diagrams with compositions of the complex and the supernatant that were in excellent agreement with simulation results. Contrary to the widely accepted theory for complexation, we found preferential partitioning of salt ions into the supernatant phase. Additionally, the salt partitioning into the supernatant phase was found to initially increase and then decrease on increasing the salt concentrations, manifesting as a distinct minimum in the salt partition coefficients. These trends were shown by simulations to be strongly influenced by the excluded volume interactions in the complex phase, which were not accounted for in their entirety in earlier theories. We believe the comprehensive data we present will be conducive to the development of an accurate physical theory for polyelectrolyte complexation with predictive capabilities.

Abstract:Interfacial tension is a crucial property for a charged polymer system undergoing liquid-liquid phase separation as it governs the structure and morphology of condensate phases. While the interface problem of simple two-phase coacervation has been well documented, little light has been shed so far on the multiphase separation process. In this study, we theoretically investigate the interfacial properties in multiphase coacervation driven by the asymmetry of the linear charge density for different polycations. We calculate the density profiles of all species at these interfaces as well as the variations of interfacial tensions with salt concentration. The results indicate that in a three-phase coacervation system, direct contact of the dilute phase with the most condensed phase is unstable, so a middle layer consisting of a less condensed phase must appear. Then, a core-shell structure is formed, which is independent of the presence or absence of salt ions. Our study is in good agreement with the previous literature and provides deeper insight into understanding the spatial structure of multiphase coacervates.
The Effect of Ion Pairs on Coacervate-Driven Self-Assembly of Block Polyelectrolytes

Abstract:The incorporation of oppositely charged polyelectrolytes into a block copolymer system can lead to formation of microphase separated nanostructures driven by the electrostatic complex between two oppositely charged blocks. It is a theoretical challenge to build an appropriate model to handle such coacervate-driven self-assembly, which should capture the strong electrostatic correlations for highly charged polymers. In this paper, we develop the self-consistent field theory considering the ion paring effect to predict the phase behavior of block polyelectrolytes. In our model, two types of ion pairs, the binding between two oppositely charged monomers and the binding between charged monomers and counterions, are included. Their strength of formation is controlled by two parameters Kaa and Kac, respectively. We give a detailed analysis about how the binding strength Kac and Kaa and salt concentration affect the self-assembled nanostructure of diblock polyelectrolyte systems. The results show that the binding between two oppositely charged blocks provides driven force for microphase separation, while the binding between charged monomers and counterions competes with the polyion pairing and thus suppresses the microphase separation. The addition of salt has a shielding effect on the charges of polymers, which is a disadvantage to microphase separation. The phase diagrams as a function of polymer concentration and salt concentration at different situations are constructed, and the influence of Kaa, Kac, and charged block composition fa is analyzed in depth. The obtained phase diagrams are in good agreement with currently existing experimental and theoretical results.
Assessing the Structure and Equilibrium Conditions of Complex Coacervate Core Micelles by Varying Their Shell Composition and Medium Ionic Strength

Abstract:Complex coacervates result from an associative phase separation commonly involving oppositely charged polyelectrolytes. When this associative interaction occurs between charged-neutral diblock copolymers and oppositely charged homopolymers, a nanometric aggregate called a complex coacervate core micelle, C3M, is formed. Recent studies have addressed the issue of their thermodynamic or kinetic stability but without a clear consensus. To further investigate this issue, we have studied C3Ms formed by the combination of poly(diallyldimethylammonium) and copolymer poly(acrylamide)-<i>b</i>-poly(acrylate) using different preparation protocols. Dynamic light scattering and small-angle X-ray scattering measurements suggest that these structures are in an equilibrium condition because the aggregates do not vary with different preparation protocols or upon aging. In addition, their stability and structures are critically dependent on several parameters such as the density of neutral blocks in their shell and the ionic strength of the medium. Decreasing the amount of copolymer in the system and, hence, the density of neutral blocks in the shell results in an increase in the aggregate size because of the core growth, although their globular shape is retained. On the other hand, larger clusters of micelles were formed at higher ionic strengths. Partially replacing 77% of the copolymer with a homopolymer of the same charge or increasing the ionic strength of the system (above 100 mmol L<sup>-1</sup> NaCl) leads to a metastable state, after which phase separation is eventually observed. SAXS analyses reveal that this phase separation above a certain salt concentration occurs due to the coagulation of individual micelles that seem to retain their individual globular structures. Overall, these results confirm earlier claims that equilibrium C3Ms are achieved close to 1:1 charge stoichiometry but also reveal that these conditions may vary at different shell densities or higher ionic strengths, which constitute vital information for envisioning future applications of C3Ms.

Organic versus Hybrid Coacervate Complexes : Co-Assembly and Adsorption Properties

Abstract:We report the co-assembly and adsorption properties of coacervate complexes made from polyelectrolyte-neutral block copolymers and oppositely charged nanocolloids. The nanocolloids put under scrutiny were ionic surfactant micelles and highly charged 7 nm cerium oxide (CeO2) nanoparticles. Static and dynamic light scattering was used to investigate the microstructure and stability of the organic and hybrid complexes. The microstructure of the CeO2-based complexes was resolved using cryogenic transmission electronic microscopy (Cryo-TEM), and it revealed that the cores were clusters made from densely packed nanoparticles. In the concentration range of interest, c = 10-4 - 1 wt. %, the surfactant-based complexes were shown to exhibit a critical association concentration (cac) whereas the nanoparticle-polymer hybrids did not. The adsorption properties of the same complexes were investigated above the cac by stagnation point adsorption reflectometry. The adsorbed amount was measured as a function of time for polymers and complexes using anionically charged silica and hydrophobic poly(styrene) substrates. It was found that all complexes adsorbed readily on both types of substrates up to a level of 1 - 2 mg m-2 at stationary state. Upon rinsing however, the adsorbed layer was removed for the surfactant-based systems, but not for the cerium oxide clusters. As for the solution properties, these finding were interpreted in terms of a critical association concentrations which are very different for organic and hybrid complexes.

Abstract:We use dissipative particle dynamics to study polyelectrolyte complexation in dilute solutions under conditions of either chain-length or concentration asymmetry between the polycation and the polyanion, characterized by the macromolecular charge ratio of the minor component to the major component. Our results show that generally the systems form clusters carrying net macromolecular charges. The stoichiometry of these net-charged macromolecular clusters depends on the overall charge asymmetry, and clusters with certain stoichiometry can dominate the cluster population under a wide range of chain-length and concentration asymmetry. The morphology and stability of these net-charged clusters are closely related to their stoichiometry. When the charge ratio reaches a threshold value, the polyions condense into a single large coacervate cluster, signaling the onset of macroscopic coacervation. The threshold value is less for concentration asymmetry systems than for chain-length asymmetry systems at the same salt concentration. This threshold value decreases with added salt, as salt ions cause merging of small clusters into larger ones, eventually leading to the dominance of a single large cluster. This “salting-out” phenomenon is followed by a “salting-in” behavior, as further addition of salt dissolves the large cluster. The preferred size and composition of the net-charged clusters observed in the simulation highlight the significant role of length and concentration asymmetry on polyelectrolyte complex coacervation and hint at the possibility of a microstructured liquid.

Abstract:We demonstrated recently that polyelectrolytes with cationic moieties along the chain and a single anionic head are able to form physical hydrogels due to the reversible nature of the head-to-body ionic bond. Here we generate a variety of such polyelectrolytes with various cationic moieties and counterion combinations starting from a common polymeric platform. We show that the rheological properties (shear modulus, critical strain) of the final hydrogels can be modulated over three orders of magnitude depending on the cation/anion pair. Our data fit remarkably well within a scaling model involving a supramolecular head-to-tail single file between cross-links, akin to the behaviour of pine-processionary caterpillar. This model allows the quantitative measure of the amount of counterion condensation from standard rheology procedure.

Abstract:The wetting behavior of complex coacervates underpins their use in many emerging applications of surface science, particularly wet adhesives and coatings. Many factors dictate if a coacervate phase will condense on a solid surface, including solution conditions, the nature of the polymer--substrate interaction, and the underlying supernatant--coacervate bulk phase behavior. In this work, we use a simple inhomogeneous mean-field theory to study the wetting behavior of complex coacervates on solid surfaces both off-coexistence (wetting transitions) and on-coexistence (contact angles). We focus on the effects of salt concentration, the polycation/polyanion surface affinity, and the applied electrostatic potential on the wettability. We find that the coacervate generally wets the surface via a first order wetting transition with second order transitions possible above a surface critical point. Applying an electrostatic potential to a solid surface always improves the surface wettability when the polycation/polyanion--substrate interaction is symmetric. For asymmetric surface affinity, the wettability has a nonmonotonic dependence with the applied potential. We use simple scaling and thermodynamic arguments to explain our results.

Small Amphiphile-Based Coacervation

Abstract:Coacervation plays an important role in the molecular assembly towards soft materials with a diversity of function (e. g., underwater adhesives of mussels and membraneless organelles). Coacervation is observed when one homogenous solution spontaneously separates into two immiscible liquid phases of low and high solute concentration, also known as liquid-liquid phase separation (LLPS), which enables spatiotemporally local concentration of specific molecules. LLPS is a common physical phenomenon in aqueous solutions of polyelectrolytes, surfactants and biomolecules, which has been extensively explored for applications in the fields of environmental remediation, cosmetic formulation, protein purification, extractive fermentation and pharmaceutical microencapsulation. This review summarizes the development of LLPS with low molecular weight amphiphiles to construct simple and complex coacervates using conventional surfactants and novel amphiphiles such as azobenzene-derivatives and peptides. We also highlight the applications of these amphiphile coacervates in the extraction of biomolecules, construction of protocell models and drug delivery.

Abstract:Coacervation, or liquid-liquid phase separation (LLPS) of biomacromolecules, is increasingly recognized to play an important role both intracellularly and in the extracellular space. Central questions that remain to be addressed are the links between the material properties of coacervates (condensates) and both the primary and the secondary structures of their constitutive building blocks. Short LLPS-prone peptides, such as GY23 variants explored in this study, are ideal model systems to investigate these links because simple sequence modifications and the chemical environment strongly affect the viscoelastic properties of coacervates. Herein, a systematic investigation of the structure/property relationships of peptide coacervates was conducted using GY23 variants, combining biophysical characterization (plate rheology and surface force apparatus, SFA) with secondary structure investigations by infrared (IR) and circular dichroism (CD) spectroscopy. Mutating specific residues into either more hydrophobic or more hydrophilic residues strongly regulates the viscoelastic properties of GY23 coacervates. Furthermore, the ionic strength and kosmotropic characteristics (Hofmeister series) of the buffer in which LLPS is induced also significantly impact the properties of formed coacervates. Structural investigations by CD and IR indicate a direct correlation between variations in properties induced by endogenous (peptide sequence) or exogenous (ionic strength, kosmotropic characteristics, aging) factors and the β-sheet content within coacervates. These findings provide valuable insights to rationally design short peptide coacervates with programmable materials properties that are increasingly used in biomedical applications.
Complexation Behavior of Oppositely Charged Polyelectrolytes: Effect of Charge Distribution

Abstract:Complexation behavior of oppositely charged polyelectrolytes in a solution is investigated using a combination of computer simulations and experiments, focusing on the influence of polyelectrolyte charge distributions along the chains on the structure of the polyelectrolyte complexes. The simulations are performed using Monte Carlo with the replica-exchange algorithm for three model systems where each system is composed of a mixture of two types of oppositely charged model polyelectrolyte chains (EGEG)5/(KGKG)5, (EEGG)5/(KKGG)5, and (EEGG)5/(KGKG)5, in a solution including explicit solvent molecules. Among the three model systems, only the charge distributions along the chains are not identical. Thermodynamic quantities are calculated as a function of temperature (or ionic strength), and the microscopic structures of complexes are examined. It is found that the three systems have different transition temperatures, and form complexes with different sizes, structures, and densities at a given temperature. Complex microscopic structures with an alternating arrangement of one monolayer of E/K monomers and one monolayer of G monomers, with one bilayer of E and K monomers and one bilayer of G monomers, and with a mixture of monolayer and bilayer of E/K monomers in a box shape and a trilayer of G monomers inside the box are obtained for the three mixture systems, respectively. The experiments are carried out for three systems where each is composed of a mixture of two types of oppositely charged peptide chains. Each peptide chain is composed of Lysine (K) and glycine (G) or glutamate (E) and G, in solution, and the chain length and amino acid sequences, and hence the charge distribution, are precisely controlled, and all of them are identical with those for the corresponding model chain. The complexation behavior and complex structures are characterized through laser light scattering and atomic force microscopy measurements. The order of the apparent weight-averaged molar mass and the order of density of complexes observed from the three experimental systems are qualitatively in agreement with those predicted from the simulations.

Abstract:Gels are key materials in biological systems such as tissues and may control biocondensate formation and structure. To further understand the effects of elastic environments on biomacromolecular assembly, we have investigated the phase behavior and radii of complex coacervate droplets in polyacrylamide (PAM) networks as a function of gel modulus. Poly-l-lysine (PLL) and sodium hyaluronate (HA) complex coacervate phases were prepared in PAM gels with moduli varying from 0.035 to 15.0 kPa. The size of the complex coacervate droplets is reported from bright-field microscopy and confocal fluorescence microscopy. Overall, the complex coacervate droplet volume decreases inversely with the modulus. Fluorescence microscopy is also used to determine the phase behavior and concentration of fluorescently tagged HA in the complex coacervate phases as a function of ionic strength (100-270 mM). We find that the critical ionic strength and complex coacervate stability are nonmonotonic as a function of the network modulus and that the local gel concentration can be used to control phase behavior and complex coacervate droplet size scale. By understanding how elastic environments influence simple electrostatic assembly, we can further understand how biomacromolecules exist in complex, crowded, and elastic cellular environments.
