Abstract:Vibrio harveyi is a causative agent of vibriosis in the large yellow croaker Pseudosciaena crocea and causes severe losses to the aquaculture industry in China. The vaccines based on the outer membrane proteins (OMPs) of the pathogens are considered to be the optimum intervention for this disease. In this study, two V. harveyi OMP genes, OmpK* and glyceraldehyde-3-phosphate dehydrogenase (GAPDH*), were cloned, sequenced, and characterized. The recombinant proteins (r-OmpK and r-GAPDH) were expressed by the prokaryotic expression vector pET-30a(+) and purified with nickel-nitrilotriacetic acid affinity chromatography. Western blots showed that rabbit antisera against purified r-OmpK and r-GAPDH specifically reacted with the native OMP of V. harveyi. Large yellow croakers were immunized with r-OmpK and r-GAPDH. Specific antibody titer assessed by enzyme-linked immunosorbent and phagocytosis assays demonstrated that specific and innate immunity was stimulated in response to the OMPs of V. harveyi. Challenge results indicated that vaccination of large yellow croakers with r-OmpK and r-GAPDH increased relative survival (37.7% and 40.0%, respectively) against wild V. harveyi.

Abstract:Aim: To investigate the immunoprotection of three recombinant proteins derived from the Vibrio harveyi outer membrane proteins (OMPs) OmpK, glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and their fusion OmpK-GAPDH as vaccine candidates from vibriosis of large yellow croaker (Pseudosciaena crocea).Methods: The ompK gene, of which the leader sequence was omitted, was fused with the gapdh gene. Three recombinant proteins r-OmpK, r-GAPDH and r-OmpK-GAPDH were expressed and purified. Western blots were carried out to detect the specificity of the antibodies raised against the recombinant proteins; Fish were immunized with recombinant proteins and challenged by native V. harveyi. The immunoresponse to the recombinant proteins were determined by ELISA and phagocytic activity assay.Conclusions: The fusion protein r-OmpK-GAPDH can afford greater protection against the wild V. harveyi than r-OmpK or r-GAPDH alone or their mixture in humoral and cellular immunity, indicating that OmpK and GAPDH could produce a synergistic immunoprotection against vibriosis of large yellow croaker (Pseudosciaena crocea) when fused into OmpK-GAPDH with a linker.Significance and Impact of the Study: It has been realized that a multi-component OMP antigen can induce a higher frequency of immune effectors than a single OMP. The results presented here bring forth a good suggestion for the subunit vaccine design based on the OMPs of gram-negative pathogens.
Immune response of large yellow croaker Larimichthys crocea towards a recombinant vaccine candidate targeting the parasitic ciliate Cryptocaryon irritans

Abstract:Cryptocaryon irritans , a parasitic ciliate, pose a major threat to marine teleost fish aquaculture. So far, no effective and safe control method is available. In this study, the protective efficacy of a recombinant truncated surface antigen of C. irritans (rCiSA32.6t) for large yellow croaker ( Larimichthys crocea ) against the parasite challenge with a sub-lethal dose of the infective theronts was evaluated by comparing the relative percent survivals (RPS), the specific antibody titers in sera, and the expression levels of the immune-related genes among the negative or adjuvant control fish, fish intraperitoneally immunized with rCiSA32.6t. The results showed that a RPS of 50.1% in rCiSA32.6t-immunized fish was achieved in comparison to negative control fish against C. irritans . A significant increase was noted in the antigen-specific immunoglobulin M (IgM) and immunoglobulin T (IgT) antibody levels in the sera of the rCiSA32.6t-vaccinated fish. Compared to the negative control fish, quantitative real-time PCR analysis indicated that the interleukin-1beta, IgT, and IgM heavy chain mRNA level in the fish head kidney, spleen, gill, and skin tissue were upregulated post-rCiSA32.6t immunization. This study indicates that the rCiSA32.6t can provide a high level of immune protection against C. irritans infection in grouper and is therefore pursued as a candidate C. irritans vaccine.

Abstract:Mesoporous silica nanoparticles (MSNs) have been used in the field of biomedicine as antigen carriers and adjuvants for protective antigens. In the present study, an oral nanovaccine against Vibrio alginolyticus was prepared employing MSNs as carriers. The uptake of the dihydrolipoamide dehydrogenase (DLDH) antigens in the intestine of large yellow croaker was evaluated using an immunohistochemistry assay. Additionally, the effects of the nanovaccine on the early immune response in large yellow croaker were investigated via oral vaccination. The presence of the antigens was detected in the mucosa and lamina propria of the foregut, midgut, and hindgut of large yellow croaker at 3 h following oral immunization. The expression levels of cytokines (i.e., lysozyme, IFN-γ, IFITM, TNF-α, IL-1β, IL-2, IL-4, IL-10, and IL-13) in the intestine, spleen, and head kidney tissues of large yellow croaker before and after the immune challenge were determined via RT-qPCR assay. The obtained results revealed that the expression levels of lysozyme, IFN-γ, IFITM, TNF-α, IL-1β, IL-2, IL-4, IL-10, and IL-13 in the intestine and head kidney of the vaccinated large yellow croaker, as well as the expression of lysozyme, IL-1β, and IL-10 in the spleen, exhibited time-dependent oscillation regulation patterns. Notably, the nanovaccine immunization could induce early (6 h) and high expression of IFN-γ in the spleen and kidney tissues after the bacterial infection. The current study supplements the available data on the early immune response to fish nanovaccines. It also provides a valuable theoretical basis for the future development of large yellow croaker oral vaccines.

Abstract:We investigated the effects of fish protein hydrolysate (FPH) on growth performance and humoral immune response of the large yellow croaker (Pseudosciaena crocea R.). One thousand and two hundred large yellow croakers [initial average weight: (162.75±23.85) g] were divided into four groups and reared in floating sea cages (3 m×3 m×3 m). The animals were fed with 4 diets: basal diet only (control) or diets supplemented with 5%, 10% and 15% (w/w) FPH. The results show that dietary FPH levels significantly influenced the growth and immunity of the large yellow croaker. Compared with the control group, total weight gain (TWG) in all treatment groups, relative weight gain (RWG) and specific growth rate (SGR) in fish fed with diets supplemented with 10% and 15% FPH were significantly increased (P<0.05). Similar results were observed in immune parameters [lysozyme activity, serum complements, immunoglobulin M (IgM)]. Lysozyme activity, complement C4 and IgM were also significantly increased (P<0.05) in fish fed with diets supplemented with 10% and 15% FPH, while complement C3 level was significantly increased (P<0.05) in all treatment groups. In general, with the supplementation of FPH, particularly at dose of 10%, the growth performance and immunity of the large yellow croaker can be improved effectively.

Abstract:Aeromonas veronii is a major pathogenic bacterium in humans and animals. When it causes outbreaks, there are enormous economic losses to the aquaculture industry. An effective live attenuated vaccine strain, AhisJ, was obtained in our previous studies by gene knockout in Aeromonas veronii TH0426 using the suicide vector pRE112. Here, we evaluated whether the live attenuated vaccine AhisJ was suitable for prevention of Aeromonas veronii infection by injection and immersion in loaches. Compared with that of the TH0426 wild-type strain, the virulence of the live vaccine was significantly weakened. Vaccine safety assessment results also indicated that 1 x 10(7) CFU/mL live vaccine was safe and did not induce clinical symptoms or obvious pathological changes. Additionally, after challenging loaches with Aeromonas veronii TH0426, the relative percent survival of the IN3 injection group was 65.66%, and that of the IM group was 50.78%. Our data show that the live attenuated vaccine Delta hisJ can improve the immune protection rate of loaches. Furthermore, increased enzyme activity parameters (SOD, LZM, ACP, and AKP) in the skin mucus, increased enzyme activity parameters (SOD, LZM, ACP, AKP, and GPx) in the serum, increased specific IgM antibodies and cytokine IL-1 beta contents in the serum, and increased cytokine (IL-15, pIgR, IL-1 beta, and TNF-alpha) expression in the liver and spleen were observed. These data are the first to indicate that the live attenuated vaccine Delta hisJ is suitable for the development of a safe and effective vaccine against Aeromonas veronii infection in loach aquaculture.

Isolation and characterization of probiotic Lysinibacillus species from the gastrointestinal tract of large yellow croaker (Larimichthys crocea)

Abstract:The objective of this study was to identify effective strategies for disease prevention and control that circumvent antimicrobial resistance (AMR) issues, thereby facilitating sustainable aquaculture practices. This research focused on isolating probiotic Bacillus strains from the intestinal tract of healthy large yellow croaker ( Larimichthys crocea ). Among the 11 strains tested, four demonstrated protease and lipase activities. Further testing identified one strain, Lysinibacillus sp. (LYD11), with strong inhibitory activity against Vibrio harveyi and Vibrio alginolyticus , two major pathogens of the large yellow croaker. The screened strains showed more than 45% co-aggregation with five pathogens and were capable of inhibiting pathogen colonization through competition, repulsion, and substitution adhesion of 20.56 to 48.45%, 40.37 to 75.62% and 55.51 to 85.30%, respectively. The hydrophobicity and self-aggregation rates of LYD11 were 82.73% and 49.47%, respectively, indicating a potent ability for adhesion and colonization within the fish intestine. LYD11 demonstrated robust growth under the NaCl 0.5–3.5% and pH range of 6–8 and was unaffected by the gastrointestinal environment. Safety evaluations at high concentrations in grouper, in both injected and fed groups, indicated no adverse effects, as evidenced by the absence of morbidity or mortality. Consequently, Lysinibacillus sp. LYD11 emerges as a potential probiotic candidate for the prevention and control of bacterial diseases in the large yellow croaker.

Safety of an Oral Somatostatin DNA Vaccine Using Attenuated Salmonella typhimurium as Carrier for Grass Carp

Abstract:The juvenile grass carps were fed with somatostatin DNA vaccines using attenuated Salmonella typhimurium as carrier(ZJ111/ZJ111/pcDNA3-SS,10~8,10~9,10~(10) cfu/mL) through oral route to evaluate the survival of fishes in comparison to S.typhimurium(10~9 cfu/mL) and PBS as controls.In another experimental setting,larger grass carps(body weight of about 150 g) fed with 1 mL of ZJ111/pcDNA3-SS(10~8 cfu/mL),S.typhimurium(10~8 cfu/mL) and PBS.The liver and spleen was taken 7 days post-fedding,for electronic microscopic analyses.Mortalities of juvenile grass carps fed with the strain ZJ111/pcDNA3-SS at 10~8,10~9 and 10~(10) cfu/mL were 0%,6.67% and 26.67% respectively.The morphology of cells of liver and spleen was normal from the fishes fed with ZJ111/pcDNA3-SS and PBS while there were apoptotic changes in the liver and spleen cells of fishes fed with wild type S.typhimurium.These results indicated that oral administration of ZJ111/pcDNA3-SS at the dose of 10~8 cfu/mL was relatively safe for the carps.
Construction and Characterization of an Aeromonas hydrophila Multi-Gene Deletion Strain and Evaluation of Its Potential as a Live-Attenuated Vaccine in Grass Carp

Abstract:Aeromonas hydrophila is an important pathogen that causes motile Aeromonas septicemia (MAS) in the aquaculture industry. Aerolysin, hemolysin, serine protease and enterotoxins are considered to be the major virulence factors of A. hydrophila. In this study, we constructed a five-gene (aerA, hly, ahp, alt and ast) deletion mutant strain (named Aeromonas hydrophila five-gene deletion strain, AHFGDS) to observe the biological characteristics and detect its potential as a live-attenuated vaccine candidate. AHFGDS displayed highly attenuated and showed increased susceptibility to fish blood and skin mucus killing, while the wild-type strain ZYAH72 was highly virulent. In zebrafish (Danio rerio), AHFGDS showed a 240-fold higher 50% lethal dose (LD50) than that of the wild-type strain. Immunization with AHFGDS by intracelomic injection or immersion routes both provided grass carp (Ctenopharyngodon idella) significant protection against the challenge of the strain ZYAH72 or J-1 and protected the fish organs from serious injury. Further agglutinating antibody titer test supported that AHFGDS could elicit a host-adaptive immune response. These results suggested the potential of AHFGDS to serve as a live-attenuated vaccine to control A. hydrophila infection in aquaculture.
Efficacy of a recombinant Lactobacillus plantarum Lp-pPG-Malt as an oral vaccine candidate against Aeromonas hydrophila infection in crucian carp

Abstract:Aeromonas hydrophila (A. hydrophila), a gram-negative bacterium, causes serious diseases with various clinical symptoms in farm raised fish. Thus, different ways to prevent and control A. hydrophila infection need to be explored, including a vaccine. In this study, we evaluated the protective efficacy of an oral vaccine prepared from the A. hydrophila TPS maltoporin (Malt) with Lactobacillus plantarum (L. plantarum) against A. hydrophila infection in crucian carp (Carassius auratus). For the in vivo experiment, the oral vaccine was administered to crucian carp by feeding them fish diets containing Lp-pPG-Malt, Lp-pPG and PBS for 28 days. The enzyme-linked immunosorbent assay (ELISA), leukocyte phagocytosis assay and real-time quantitative polymerase chain reaction (RT-qPCR) were performed to measure the protective efficacy of the Lp-pPG-Malt. ELISA and leukocyte phagocytosis assay confirmed that Lp-pPG-Malt significantly enhanced the IgM level and nonspecific immune response of crucian carp compared with the control groups (Lp-pPG and PBS). The RT-qPCR results showed that the Lp-pPG-Malt increased the relative expression of immune-related genes (IL-10, IL-1β, TNF-α, IFN-γ) of crucian carp in various tissues (liver, spleen, head kidney and hind intestine). Moreover, Lp-pPG-Malt significantly increased the relative percent survival of fish after intraperitoneal injection with A. hydrophila (55%) compared with the Lp-pPG and PBS groups (0%). These findings suggest that Lp-pPG-Malt can serve as an oral vaccine candidate for A. hydrophila infection and that Malt can be used as an effective antigen in crucian carp farming.
Development of a hyper-adhesive and attenuated Edwardsiella ictaluri strain as a novel immersion vaccine candidate in yellow catfish (Pelteobagrus fulvidraco)

Abstract:Edwardsiella ictaluri, a Gram-negative intracellular pathogen, is the causative agent of enteric septicemia in channel catfish, and catfish aquaculture in China suffers heavy economic losses due to E. ictaluri infection. Vaccination is an effective control measure for this disease. In this study, an attenuated E. ictaluri strain was acquired through deletion mutation of the T3SS protein eseJei, and the ΔeseJei strain fails to replicate in the epithelioma papillosum of carp cells. The type 1 fimbria plays a pivotal role in the adhesion of E. ictaluri, and it was found in this study that deletion of −245 to −50 nt upstream of fimA increases its adhesion to around five times that of the WT strain. A hyper-adhesive and highly attenuated double mutant (ΔeseJeiΔfimA-245--50 strain) was constructed, and it was used as a vaccine candidate in yellow catfish via bath immersion at a dosage of 1 × 105 CFU/mL. It was found that this vaccine candidate can stimulate protection when challenged with E. ictaluri HSN-1 at 5 × 107 CFU/mL (∼20 × LD50). The survival rate was 83.61% for the vaccinated group and 33.33% for the sham-vaccinated group. The RPS (relative percent of survival) of the vaccination trial reached 75.41%. In conclusion, the ΔeseJeiΔfimA-245--50 strain developed in this study can be used as a vaccine candidate. It excels in terms of ease of delivery (via bath immersion) and is highly efficient in stimulating protection against E. ictaluri infection.

Abstract:Visceral white spot disease (VWND) caused by Pseudomonas plecoglossicida poses a major threat to the sustainable development of large yellow croaker ( Larimichthys crocea ) aquaculture. Genome-wide association analysis (GWAS) and RNA-seq research indicated that LcCD82a play an important role in resistance to visceral white spot disease in L. crocea , but the molecular mechanism of LcCD82a response to P. plecoglossicida infection is still unclear. In this study, we cloned and validated the Open Reading Frame (ORF) sequence of LcCD82a and explored the expression profile of LcCD82a in various tissues of L.crocea . In addition, two different transcript variants ( LcCD82a-L and LcCD82a-S ) of LcCD82a were identified that exhibit alternative splicing patterns after P. plecoglossicida infection, which may be closely related to the immune regulation during pathogenetic process of VWND. In order to explore the function of LcCD82a , we purified the recombinant protein of LcCD82a-L and LcCD82a-S . The bacterial agglutination and apoptosis function analysis showed that LcCD82a may involve in extracellular bacterial recognition, agglutination, and at the same time participate in the process of antigen presentation and induction of cell apoptosis. Collectively, our studies demonstrate that LcCD82a plays a crucial role in regulating apoptosis and antimicrobial immunity.

Efficacy of a formalin-inactivated vaccine against Lactococcus garvieae infection in golden pompano Trachinotus ovatus

Abstract:Lactococcus garvieae is one of the primary pathogens causing the severe disease to golden pompano ( Trachinotus ovatus ). Vaccination is the most effective measure of preventing the occurrence and spread of infectious diseases. However, no commercially available vaccine protects T. ovatus against L. garvieae infection. In the present study, the formalin-inactivated L. garvieae vaccines, consisting of bacterin alone (aqueous vaccine) and combined with oil adjuvant MontanideTM ISA 763 A VG (vaccine+763A), were prepared and administered to T. ovatus by intraperitoneal injection. The protective efficacy was evaluated by testing the relative percent survival (RPS), serum enzyme activity, and expression levels of immune-related genes in the tissues of T. ovatus after immunization. The results indicated that the relative percentage survival (RPS) values of aqueous vaccine and vaccine+763A were 80% and 90.8% at 8 weeks post-vaccination. The level of antibody titer, lysozyme activities, superoxide dismutase, and catalase in the two vaccinated fish groups increased significantly compared to that in the control group. Moreover, the expression levels of immune-related genes IgM , TNF-α , IL-8 , MHCI-α , MHCII , CD4 , CD8α, and IL-1β in all the tested tissues from T. ovatus were significantly enhanced after inoculation with aqueous vaccine and vaccine+763A. These findings demonstrated that the developed vaccine combined with oil adjuvant ISA 763 A VG improved immune responses and provided good protective efficacy against L. garvieae infection in the golden pompano culture.

Evaluation of protective immune response of immersion inactivated vaccine against Singapore grouper iridovirus

Abstract:Singapore grouper iridovirus (SGIV) always causes high transmission efficiency and mortality in the larval and juvenile stages of grouper in aquaculture industry. Although inactivated virus and recombinant DNA vaccines administered via intraperitoneal injection have shown efficacy in protection against SGIV, their potential applications in field testing were limited due to the vaccine delivery methods. Here, we developed an immersion vaccine containing inactivated virus and Montanide IMS 1312 adjuvant (IMS 1312) and evaluated its protective efficacy against SGIV infection. Compared to the PBS group, fish vaccinated with immersion inactivated vaccine with or without IMS 1312 were significantly protected against SGIV, with a relative percent survival (RPS) of 57.69% and 38.47%, respectively. Furthermore, the transcripts of viral core genes were reduced, and the histopathological severity caused by SGIV were relatively mild in multiple tissues of the IMS+V group. The immersion vaccine activated the AKP and ACP activities and increased the mRNA levels of IFN and inflammation-associated genes. The transcriptome analysis showed that a total of 731 and 492 genes were significantly regulated in the spleen and kidney from the IMS+V group compared to the PBS group, respectively. Among them, 129 DEGs were co-regulated, and enriched in the KEGG pathways related to immune and cell proliferation, including MAPK signaling, JAK-STAT signaling and PI3K-Akt signaling pathways. Similarly, the DEGs specially regulated in the kidney and spleen upon vaccine immunization were significantly enriched in the KEGG pathways related to interferon and inflammation response.Together, our results elucidated that the immersion vaccine of inactivated SGIV with IMS 1312 induced a protective immune response of grouper against SGIV.
Oral Vaccination With Recombinant Pichia pastoris Expressing Iridovirus Major Capsid Protein Elicits Protective Immunity in Largemouth Bass (Micropterus salmoides)

Abstract:Largemouth bass iridovirus (LMBV) can cause high mortality and lead to heavy economic loss in the cultivation of largemouth bass, but there was no effective treatment. Here, the present study constructed a recombinant Pichia pastoris expressing LMBV major capsid protein ( MCPD ). The recombinant GS115-pW317- MCPD was then used to immunize largemouth bass via oral administration, and mucosal immune response mediated by immunoglobulins (Igs) was measured after oral immunization. Serum antibody levels were measured by ELISA, neutralizing antibody titers were determined by serum neutralization test (SNT), antigen presentation-related gene expressions were detected by RT-PCR, and the histopathological characteristics of immunized fish were assessed after challenging with 0.1 ml 10 7.19 TCID 50 /ml LMBV. The relative percentage survival (RPS) was also determined. Our results showed that the serum antibody titers of immunized fish were significantly higher than that of control groups (P < 0.05). IgT and IgM expressions in gut were increased significantly after vaccination with GS115-pW317- MCPD ; however, much stronger response in gut was observed as compared with gill. The expression levels of major histocompatibility complex ( MHC ) II, CD 8, and T-cell receptor ( TCR ) were significantly elevated in GS115-pW317- MCPD group (P < 0.05), while CD 4 and MHC I transcription levels remained unchanged after oral immunization ( P > 0.05). The RPS of fish orally immunized with 1.0 × 10 8 CFU/g GS115-pW317- MCPD was reached up to 41.6% after challenge with 0.1 ml 10 9.46 TCID 50 /ml LMBV. Moreover, orally immunizing with GS115-pW317- MCPD can relieve the pathological damage caused by LMBV. Therefore, GS115-pW317- MCPD showed a promising potential against LMBV.

Abstract:An 8‐week feeding experiment was conducted to investigate the effects of dietary glycyrrhizic acid (GA) on growth, survival and immune response of juvenile large yellow croaker, in seawater floating net cages. GA was supplemented into the basal diet to formulate four isonitrogenous and isoenergetic practical diets containing 0.00% (the control diet), 0.01%, 0.02% and 0.04% GA of dry weight, respectively. Triplicate groups of 60 fish were fed to apparent satiation by hand twice daily. The results showed that the specific growth rate, survival rate and feed efficiency ratio revealed no significant differences among dietary treatments (P > 0.05). The phagocytic index of head kidney macrophage was significantly increased by the supplementation of 0.04% dietary GA compared to the control group (P < 0.05). Fish fed 0.04% dietary GA also showed significantly higher serum lysozyme activity than fish fed the control diet and diet with 0.01% GA (P < 0.05). The cumulative mortality rate after natural infestation of parasites (protozoan, Cryptocaryon irritans Brown) showed no significant differences among dietary treatments. These results suggested that dietary glycyrrhizic acid improved certain non‐specific immunological parameters of juvenile large yellow croaker. However, GA was not able to protect juvenile large yellow croaker effectively from protozoan infection.

Abstract:An 8 weeks growth experiment was conducted to determine the effects of dietary vitamin C on the survival, growth, tissue ascorbic acid contents and immunity of large yellow croaker (Pseudosciaena crocea) with initial weight of 17.82±0.68 g. Seven practical diets were formulated to contain 0.1, 12.2, 23.8, 47.6, 89.7, 188.5 and 489.0 mg ascorbic acid equivalent kg−1 diet, supplied as l-ascorbyl-2-polyphosphate (LAPP). Each diet was fed to triplicate groups of fish in seawater floating cages (1.5×1.5×2.0 m), and each cage was stocked with 120 fish. Fish were fed twice daily (05:00 and 17:00) to apparent satiation for 8 weeks. The water temperature fluctuated from 19.5 to 25.5 °C, the salinity from 25 to 28‰ and dissolved oxygen content was approximately 7 mg l−1 during the experimental period. Results showed that the specific growth rate (SGR) (from 1.80 to 1.96% d−1) had an increasing trend with the increase of dietary vitamin C, but no significant difference was observed among dietary treatments. No gross deficiency signs were observed in any of the experimental fish. Survival rate, however, significantly increased with increasing dietary vitamin C (P<0.05). The vitamin C contents in liver and muscle correlated positively with the vitamin C in diets. The vitamin C requirement was estimated to be 28.2 mg kg−1 based on survival, and 87.0 mg kg−1 on liver content of vitamin C. The activities of serum lysozyme and alternative complement pathway (ACP), phagocytosis percentage (PP) and respiratory burst activity of head kidney significantly increased with increasing dietary vitamin C. The challenge experiment with Vibrio harveyi showed that fish fed the diets with supplementation of vitamin C had significantly lower cumulative mortality compared to the control group (66.7%), and the cumulative mortality (16.7%) in fish with 489.0 mg kg−1 ascorbic acid was significantly lower than that (41.7%) in fish with 23.8 mg kg−1 ascorbic acid. These results suggested that vitamin C significantly influenced the immune response and disease resistance of large yellow croaker.

Abstract:Bacterial septicemia represents a significant disease affecting cultured grass carp culture, with the primary etiological agent identified as the Gram-negative bacterium Aeromonas veronii. In response to an outbreak of septicemia in Guangzhou, we developed a formaldehyde-inactivated vaccine against an A. veronii strain designated AV-GZ21-2. This strain exhibited high pathogenicity in experimental infections across at all developmental stages of grass carp. Mortality rates for grass carp weighing 15 ± 5 g ranged from 16 % to 92 % at exposure temperatures of 19 °C-34 °C, respectively. The median lethal dose (LD50) for grass carp groups weighing 15 ± 5 g, 60 ± 10 g, 150 ± 30 g and 500 ± 50 g were determined to be 1.43, 2.52, 4.65 and 7.12 × 107(CFU/mL), respectively. We investigated the inactivated vaccine in conbination with aluminum hydroxide gel (AV-AHG), Montanide ISA201VG (AV-201VG), and white oil (AV-WO) adjuvants. This study aimed to optimize inactivation conditions and identify the adjuvant that elicits the most robust immune response. The AV-GZ21-2 inactivated bacterial solution (AV),when combined with various adjuvants, was capable of inducing a strong specific immune response in grass carp. The relative percent survival (RPS) following a lethal challenge with AV-GZ21-2 were 94 % for AV-AHG, 88 % for AV-201VG, 84 % for AV-WO and 78 % for AV alone. The minimum immunization dose of the AV-AHG vaccine was determined to be 6.0 × 107 CFU per fish, providing immunity for a duration of six months with an immune protection level exceeding 75 %. Furthermore, the AV-AHG vaccine demonstrated significant protective efficacy against various epidemic isolates of A. veronii. Consequently, we developed an inactivated vaccine targeting a highly pathogenic strain of A. veronii, incorporating an aluminum hydroxide gel adjuvant, which resulted in high immune protection and a duration of immunity exceeding six months. These findings suggest that the AV-AHG vaccine holds substantial potential for industrial application.

Abstract:Vaccine immunization is currently the only effective way to prevent and control the grass carp haemorrhagic disease, and the primary pathogen in these infections is grass carp reovirus genotype II (GCRV-II) for which there is no commercial vaccine. In this study, we evaluated the safety of the GCRV-II avirulent strain GD1108 which isolated in the early stage of the laboratory through continuously passed in grass carp. The immunogenicity and protective effects were evaluated after immunization by injection and immersion. The avirulent strain GD1108 could infect and replicate in the fish which did not revert to virulence after continuous passage. No adverse side effects were observed and the vaccine strain did not spread horizontally among fish. Two routes of immunization induced high serum antibody titers of OD450nm value were 0.79 and 0.76 and neutralization titers of 320 and 320 for the injection and immersion routes of inoculation, respectively. The expression of immune-related genes increased after immunization and the levels were statistically significant. Challenge of immunized fish with a virulent GCRV-II strain resulted in protection rates of 93.88% and 76.00% for the injection and immersion routes, respectively. The avirulent strain GD1108 revealed good safety and immunogenicity via two different inoculation routes. Although the injection route provided the best immune effect, two pathways provided protection against infection with virulent GCRV-II strains in various degrees. These results indicated that the avirulent strain GD1108 can be used for the development and application as live vaccine.
