Abstract:The global pandemic caused by SARS-CoV-2 has lasted two and a half years and the infections caused by the viral contamination are still occurring. Developing efficient disinfection technology is crucial for the current epidemic or infectious diseases caused by other pathogenic microorganisms. Gas plasma can efficiently inactivate different microorganisms, therefore, in this study, a combination of water spray and plasma-activated air was established for the disinfection of pathogenic microorganisms. The combined treatment efficiently inactivated the Omicron-pseudovirus through caused the nitration modification of the spike proteins and also the pathogenic bacteria. The combined treatment was improved with a funnel-shaped nozzle to form a temporary relatively sealed environment for the treatment of the contaminated area. The improved device could efficiently inactivate the Omicron-pseudovirus and bacteria on the surface of different materials including quartz, metal, leather, plastic, and cardboard and the particle size of the water spray did not affect the inactivation effects. This study supplied a disinfection strategy based on plasma-activated air for the inactivation of contaminated pathogenic microorganisms.

Abstract:Abstract Skin antiseptics have important implications for public health and medicine. Although conventional antiseptics have considerable antimicrobial activity, skin toxicity and the development of resistance are common problems. Plasma-treated water has sterilization and tissue-regenerative effects. Therefore, the aim of this study was to identify whether plasma-activated water (PAW) manufactured by our microwave plasma system can be used as a novel antiseptic solution for skin protection. PAW was produced by dissolving reactive nitrogen oxide gas using microwave plasma in deionized water. The antibacterial effects of PAW against Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Bacillus cereus, and Salmonella typhimurium and effective concentrations were investigated by a solid agar plate assay. The factors mediating the effects of PAW were evaluated by the addition of reactive species scavengers. Cytotoxicity and cell viability assays were performed to examine the protective effect of PAW on normal skin cells. PAW exhibited excellent sterilization and no toxicity in normal skin cells. Experiments also confirmed the potential of PAW as a sanitizer for SARS-CoV-2. Our findings support the use of PAW as an effective skin disinfectant with good safety in the current situation of a global pandemic.

Abstract:Coronavirus disease 2019 (COVID-19) has become a worldwide public health emergency, and the high transmission of SARS-CoV-2 variants has raised serious concerns. Efficient disinfection methods are crucial for the prevention of viral transmission. Herein, pulse power-driven cold atmospheric plasma (CAP), a novel sterilization strategy, was found to potently inactivate SARS-CoV-2-like coronavirus GX_P2V, six strains of major epidemic SARS-CoV-2 variants and even swine coronavirus PEDV and SADS-CoV within 300 s (with inhibition rate more than 99%). We identified four dominant short-lived reactive species, ONOO-, 1O2, O2- and·OH, generated in response to CAP and distinguished their roles in the inactivation of GX_P2V and SARS-CoV-2 spike protein receptor binding domain (RBD), which is responsible for recognition and binding to human angiotensin-converting enzyme 2 (hACE2). Our study provides detailed evidence of a novel surface disinfection strategy for SARS-CoV-2 and other coronaviruses.

Abstract:Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a highly transmissible virus that has precipitated a worldwide pandemic of coronavirus disease since 2019. Developing an effective disinfection strategy is crucial to prevent the risk of surface cross-contamination by SARS-CoV-2. This study employed pseudovirus and the receptor-binding domain (RBD) protein of SARS-CoV-2 as models to investigate the spike protein inactivation process and its underlying mechanisms using a novel nonthermal technology . Cold plasma combined with 222 nm ultraviolet (CP+UV) treatment was applied to accelerate the generation of reactive species and enhance sterilization efficiency. The results indicated that the binding activity of RBD protein was completely inhibited at specific concentrations (0.01–0.05 mg/cm 2 ) with corresponding treatment times of 15–30 s. The mechanism potentially involves the reactive species generated by CP+UV, which react with the spike protein RBD of SARS-CoV-2, leading to the loss of SARS-CoV-2 infectivity by causing damage to the β-sheet structure and chemical bonds in the RBD protein. Validated by a biosafety level 3 (BSL3) laboratory, the CP+UV treatment for 30 s could completely inactivate SARS-CoV-2 with a concentration of 19054 ± 1112 TCID 50 /cm 2 . Therefore, this study potentially provides a novel disinfection strategy for the inactivation of SARS-CoV-2 on surface cross-contamination.

Abstract:Disinfection of hazardous microorganisms that may challenge environmental safety is a crucial issue for economic and public health. Here, we explore the potential of a novel electrochemical disinfectant named plasma activated water (PAW), which was generated by nonthermal plasma, for inactivating Staphylococcus aureus (S. aureus). Meanwhile, the influence of bovine serum albumin (BSA) on the PAW disinfection efficacy was investigated. In the presence of BSA, PAW treatments achieved a reduction of S. aureus ranging from 2.1 to 5.5 Log, when without BSA it reached 7 Log. The sterilization efficacy depended on the PAW treatment time of S. aureus and plasma activation time for PAW generation. The results of electron spin resonance spectra showed the concentrations of hydroxyl radical (OH center dot) and nitric oxide radical (NO center dot) in water activated by plasma for 10 min (10-PAW) were higher than those in water activated by plasma for 5 min (5-PAW). Additionally, the physiological analysis of S. aureus demonstrated that the integrity of cell membrane, membrane potential, and intracellular pH homeostasis as well as DNA structure were damaged by PAW, and the molecule structure and chemical bonds of S. aureus were also altered due to PAW. Thus, PAW can be a promising chemical-free and environmentally friendly electrochemical disinfectant for application in the medical and food industries.

Abstract:Cold atmospheric plasma (CAP) displays antimicrobial, antitumor, and antiviral properties, while the underlying mechanism is seldom clearly elucidated. In this work, we employed CAP with air-feeding gas to directly inactivate SARS-CoV-2. The results indicate that the typical SARS-CoV-2 morphological spikes disappeared after plasma treatment and the proteosomes of SRAS-CoV-2 were modified. In addition, we also evaluated the safety of the air plasma device in simulating daily life environments through rat experiments. We evaluated rats' daily physiological behavior, body weight, food consumption, organ histopathology, blood biochemical indicators, and so on. These results demonstrate air plasma device is a safe and effective mean prevents virus transmissions and infections.

Abstract:Abstract Background In 2020, a new coronavirus, SARS-CoV-2, quickly spread worldwide within a few months. Although coronaviruses typically infect the upper or lower respiratory tract, the virus RNA can be detected in plasma. The risk of transmitting coronavirus via transfusion of blood products remains. As more asymptomatic infections are identified in COVID-19 cases, blood safety has become particularly important. Methylene blue (MB) photochemical technology has been proven to inactivate lipid-enveloped viruses with high efficiency and safety. The present study aimed to investigate the SARS-CoV-2 inactivation effects of MB in plasma. Methods The SARS-CoV-2 virus strain was isolated from Zhejiang University. The live virus was harvested from cultured VERO-E6 cells, and mixed with MB in plasma. The MB final concentrations were 0, 1, 2, and 4 μM. The “BX-1 AIDS treatment instrument” was used at room temperature, the illumination adjusted to 55,000 ± 0.5 million Lux, and the plasma was irradiated for 0, 2, 5, 10, 20, and 40 mins using light at a single wavelength of 630 nm. Virus load changes were measured using quantitative reverse transcription- PCR. Results BX-1 could effectively eliminate SARS-CoV-2 within 2 mins in plasma, and the virus titer declined to 4.5 log10 TCID50 (median tissue culture infectious dose)/mL. Conclusion BX-1 is based on MB photochemical technology, which was designed to inactivate HIV-1 virus in plasma. It was proven to be safe and reliable in clinical trials of HIV treatment. In this study, we showed that BX-1 could also be applied to inactivate SARS-CoV-2. During the current outbreak, this technique it has great potential for ensuring the safety of blood transfusions, for plasma transfusion therapy in recovering patients, and for preparing inactivated vaccines.

Abstract:Efficient disinfection of pathogens is a critical concern for environmental disinfection and clinical anti-infective treatment. Plasma-activated water (PAW) is a promising alternative to chemical disinfectants and antibiotics for its strong disinfection ability and not inducing any acute toxicity. Previous plasma sources are commonly placed near or fully in contact with water as possible for more efficient activation, but the risk of electrode corrosion and metal particle contamination of water threatens the safety and stability of PAW. In this work, plasma-activated gas (PAG) rich in high-valence NOx is generated by a hybrid plasma configuration and introduced into water for off-site PAW production. It is found that plasma-generated O3 dominates the gas-phase reactions for the formation of high-valence NOx. With the time-evolution of O3 concentration, the gaseous NO3 radicals are produced behind N2O5 formation, but will be decomposed before N2O5 quenching. By decoupling the roles of gaseous NO3, N2O5, and O3 in the water activation, results show that short-lived aqueous species induced by gaseous NO3 radicals play the most crucial role in PAW disinfection, and the acidic environment induced by N2O5 is also beneficial for microbial inactivation. Moreover, SEM photographs and biomacromolecule leakage assays demonstrate that PAW disrupts the cell membranes of bacteria and thus achieves inactivation. In real-life applications, an integrated device for off-site PAW production with a yield of 2 L/h and a bactericidal efficiency of >99.9 % is developed. The PAW of 50 mL produced in 3 min using this device is more effective in disinfection than 0.5 % NaClO and 3 % H2O2 with the same bacterial contact time. Overall, this work provides new avenues for efficient PAW production and deepens insights into the fundamental chemical processes that govern the reactive chemistry in PAW for environmental and biomedical applications.
Isolation and characterization of proteic allergens in refined peanut oil

Abstract:Allergic reactions to peanut oil are very much debated, even if the responsibility of peanut oil has been evoked in several cases of adverse reactions, including death related to severe asthma. The aim of the present study was to investigate the presence of allergenic proteins in peanut oil. Proteins were extracted from commercial refined peanut oil, with a relative content in the order of 0.1–0.2 μg per g of oil, and molecular sizes ranging from 14 up to 76 kDa in SDS‐PAGE. Eight protein bands were systematically observed in crude, neutralized and refined oils, with a molecular mass ranging from ≈ 14 to 76 kDa, including one at 18 kDa which was identified by Western blot performed with serum from two allergic patients. The protein extract gave positive IgE‐RIA with patient sera, positive in vitro leucocyte histamine release tests and positive skin‐prick tests in allergic patients. The allergenic protein was purified by HPLC and [125I] iodide‐labelled. It had an isoelectric point at 4.5 in isoelectrofocusing. In conclusion, we have demonstrated the presence of allergenic proteins in crude and refined peanut oil. These proteins are the same size as two allergens previously described in peanut protein extracts.
Reactive radical-driven bacterial inactivation by hydrogen-peroxide-enhanced plasma-activated-water

Abstract:The combined effects of plasma activated water (PAW) and hydrogen peroxide (H 2 O 2 ), PAW/HP, in sterilization were investigated in this study. To assess the synergistic effects of PAW/HP, S. aureus was selected as the test microorganism to determine the inactivation efficacy. Also, the DNA/RNA and proteins released by the bacterial suspensions under different conditions were examined to confirm membrane integrity. Additionally, the intracellular pH (pH i ) of S. aureus was measured in our study. Electron spin resonance spectroscopy (ESR) was employed to identify the presence of radicals. Finally, the oxidation reduction potential (ORP), conductivity and pH were measured. Our results revealed that the inactivation efficacy of PAW/HP is much greater than that of PAW, while increased H 2 O 2 concentration result in higher inactivation potential. More importantly, as compared with PAW, the much stronger intensity ESR signals and higher ORP in PAW/HP suggests that the inactivation mechanism of the synergistic effects of PAW/HP: more reactive oxygen species (ROS) and reactive nitrogen species (RNS), especially OH and NO radicals, are generated in PAW combined with H 2 O 2 resulting in more deaths of the bacteria.
Strong alkaline electrolyzed water efficiently inactivates SARS-CoV-2, other viruses, and Gram-negative bacteria

Abstract:Air spaces and material surfaces in a pathogen-contaminated environment can often be a source of infection to humans, and disinfection has become a common intervention focused on reducing the contamination levels. In this study, we examined the efficacy of SAIW, a unique electrolyzed water with chlorine-free, high pH, high concentration of dissolved hydrogen, and low oxygen reduction potential, for the inactivation of several viruses and bacteria. Infectivity assays revealed that initial viral titers of enveloped and non-enveloped viruses, including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), influenza A virus, herpes simplex virus type 1, human coronavirus, feline calicivirus, and canine parvovirus, were reduced by 2.9- to 5.5-log10 within 30 s of SAIW exposure. Similarly, the culturability of three Gram-negative bacteria (Escherichia coli, Salmonella, and Legionella) dropped down by 1.9- to 4.9-log10 within 30 s of SAIW treatment. Mechanistically, treatment with SAIW was found to significantly decrease the binding and subsequent entry efficiencies of SARS-CoV-2 on Vero cells. Finally, we showed that this chlorine-free electrolytic ion water had no acute inhalation toxicity in mice, demonstrating that SAIW holds promise for a safer antiviral and antibacterial disinfectant.

Abstract:Cold atmospheric plasma (CAP) has gaining potential, and very effective to curb or deactivate the various microorganisms such as bacteria and virus. Lately, the major outbreak SARS-CoV-2 infection has affected humanity largely with added complexity of its ability to mutate to variants such as Omicron. We have earlier shown the effectiveness of CAP on SARS-CoV-2 spike protein and, in this study, we have evaluated the effectiveness of CAP to deactivate Omicron. We studied the ability of the binding of Angiotensin converting Enzyme Protein (ACE2) protein to the plasma treated spike S1-S2 protein and spike Receptor binding domain (RBD) using Cold atmospheric plasma direct treatment as well as Plasma activated water (PAW). Results have shown the binding efficiency of Omicron spike protein to ACE2 decrease with increase in treatment time with both direct treatment and PAW as evidenced using spectroscopic techniques. The reactive species (RONS) play a major role in the efficient deactivation of the binding of ACE2 to the Omicron spike protein. Correspondingly, the comparison between the efficiency between direct treatment and PAW has also been discussed.

Abstract:A direct current atmospheric pressure cold plasma microjet (PMJ) was used to produce plasma activated water (PAW) in two generation modes above (PAW‐A) and beneath (PAW‐B) the water surface. The physicochemical characteristics and biological effects of them were measured respectively. Results showed that PAW‐B exhibited stronger disinfection efficiency than PAW‐A, which was associated with ORP and conductivity, but not pH value, temperature, H2O2, NO3− or NO2−. Furthermore, the cell membrane integrity and membrane potential of S. aureus were destroyed more severely by PAW‐B. More importantly, a significant increase of intracellular ROS was induced by PAW‐B, which was supposed to contribute most to the sterilization. This work provides valuable guidance for the production and wide applications of PAW.

Abstract:Non-thermal plasma is a promising bactericidal method due to the existence of ultraviolet, high energy electrons, reactive oxygen species, and reactive nitrogen species in plasma. Recently, the plasma activated water (PAW) has attracted more and more attention. In our study, PAW was applied to four different bacteria: Streptococcus mutans (S. mutans), Staphylococcus aureus (S. aureus), Escherichia coli (E. coli) and Candida albicans (C. albicans). The inactivation effect of PAW was observed through colony counting method and fluorescence staining. The reactive oxygen species in the PAW was investigated by optic emission spectrometer and Hydrogen Peroxide Test Kit. Effective inactivation under PAW treatment of S. mutans, S. aureus, E. coli was achieved within 20 min, while weak inactivation effect to C. albicans. In addition, reactive oxygen species such as atomic oxygen and hydrogen peroxide were detected in the PAW. Our study provides an experimental basis in the prevention of oral diseases and sterilization of medical tubular equipment by PAW.
In situ generation of cold atmospheric plasma-activated mist and its biocidal activity against surrogate viruses for COVID-19

Abstract:Aims: To provide an alternative to ultra violet light and vapourized hydrogen peroxide to enhance decontamination of surfaces as part of the response to the COVID-19 pandemic. Methods and results: We developed an indirect method for in situ delivery of cold plasma and evaluated the anti-viral activity of plasma-activated mist (PAM) using bacteriophages phi6, MS2, and phiX174, surrogates for SARS-CoV-2. Exposure to ambient air atmospheric pressure derived PAM caused a 1.71 log10 PFU ml-1 reduction in phi6 titer within 5 min and a 7.4 log10 PFU ml-1 reduction after 10 min when the the PAM source was at 5 and 10 cm. With MS2 and phiX174, a 3.1 and 1.26 log10 PFU ml-1 reduction was achieved, respectively, after 30 min. The rate of killing was increased with longer exposure times but decreased when the PAM source was further away. Trace amounts of reactive species, hydrogen peroxide and nitrite were produced in the PAM, and the anti-viral activity was probably attributable to these and their secondary reactive species. Conclusions: PAM exhibits virucidal activity against surrogate viruses for COVID-19, which is time and distance from the plasma source dependent.

Abstract:Abstract Coronavirus disease 2019 (COVID-19) is an infectious viral disease caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) that emerged at the end of 2019. SARS-CoV-2 can be transmitted through droplets, aerosols, and fomites. Disinfectants such as alcohol, quaternary ammonium salts, and chlorine-releasing agents, including hypochlorous acid, are used to prevent the spread of SARS-CoV-2 infection. In the present study, we investigated the efficacy of ionless hypochlorous acid water (HOCl) in suspension and by spraying to inactivate SARS-CoV-2. The virucidal efficacy of HOCl solution in tests against SARS-CoV-2 was evaluated as 50% tissue culture infectious dose. Although the presence of organic compounds influenced the virucidal efficacy, HOCl treatment for 20 s was significantly effective to inactivate Wuhan and Delta strains in the suspension test. HOCl atomization for several hours significantly reduced the SARS-CoV-2 attached to plastic plates. These results indicate that HOCl solution with elimination containing NaCl and other ions may have high virucidal efficacy against SARS-CoV-2. This study provides important information about the virucidal efficacy and use of HOCl solution.

Abstract:Plasma‐activated water could efficiently inactivate the oral pathogenic bacteria, providing a potent strategy for bacteria eradication to reduce the incidence of oral diseases. Oral diseases stemming from oral pathogenic bacteria pose a significant global health concern, and current methods for managing these pathogens have limitations. Plasma‐activated water (PAW), containing various reactive species, emerges as a promising disinfectant with impressive inactivation capabilities. In this study, PAW prepared by mixed‐mode plasma‐activated gases was applied to inactivate oral pathogenic bacteria, including Streptococcus mutans and Porphyromonas gingivalis. The PAW could reduce more than 6.1‐log10 planktonic bacteria and 4.1‐log10 bacteria within biofilm, respectively, and PAW treatment of planktonic bacteria effectively inhibited biofilm formation. Compared to chlorhexidine, PAW exhibited superior inactivation effects in both planktonic bacteria and biofilm. This study presented a potent strategy for bacteria eradication to reduce the incidence of oral diseases.

Abstract:This study aimed to compare the SARS-CoV-2-inactivation activity and virucidal mechanisms of ozonated water (OW) with those of slightly acidic electrolyzed water (SAEW) and 70% ethanol (EtOH). SARS-CoV-2-inactivation activity was evaluated in a virus solution containing 1%, 20% or 40% fetal bovine serum (FBS) with OW, SAEW or EtOH at a virus-to-test solution ratio of 1:9, 1:19 or 1:99 for a reaction time of 20 s. EtOH showed the strongest virucidal activity, followed by SAEW and OW. Even though EtOH potently inactivated the virus despite the 40% FBS concentration, virus inactivation by OW and SAEW decreased in proportion to the increase in FBS concentration. Nevertheless, OW and SAEW showed potent virucidal activity with 40% FBS at a virus-to-test solution ratio of 1:99. Real-time PCR targeting the viral genome revealed that cycle threshold values in the OW and SAEW groups were significantly higher than those in the control group, suggesting that OW and SAEW disrupted the viral genome. Western blotting analysis targeting the recombinant viral spike protein S1 subunit showed a change in the specific band into a ladder upon treatment with OW and SAEW. OW and SAEW may cause conformational changes in the S1 subunit of the SARS-CoV-2 spike protein.
