Abstract:BackgroundThe development of antibiotic resistant bacteria stems from a number of factors, including inappropriate use of antibiotics in human and animal health and their prolonged use as growth promoters at sub-clinical doses in poultry and livestock production. We were interested in investigating plants that could be useful in protecting humans or animals against diarrhoea. We decided to work on extracts of nine plant species with good activity against Escherichia coli based on earlier work in the Phytomedicine Programme. Leaves of nine medicinal plant species with high antibacterial activity against Escherichia coli were extracted with acetone and their minimal inhibitory concentration (MIC) values determined using a microplate serial dilution technique against Gram-positive (Staphylococcus aureus, Enterococcus faecalis and Bacillus cereus) and Gram-negative (Escherichia coli, Salmonella Typhimurium and Pseudomonas aeruginosa) bacteria. Bioautography was used to determine the number of bioactive compounds in each extract. In vitro safety of the extracts was determined using the 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide reduction assay on Vero cells.ResultsThe extracts were active against all the pathogens with average MICs ranging from 0.02 to 0.52 mg/ml. As expected E. coli was relatively sensitive, while E. faecalis and S. Typhimurium were more resistant to the extracts (average MICs of 0.28 mg/ml and 0.22 mg/ml respectively). Cremaspora triflora and Maesa lanceolata leaf extracts had higher activity than the other extracts against Gram-positive and Gram-negative pathogens with mean MICs of 0.07 mg/ml and 0.09 mg/ml respectively. Extracts of Maesa lanceolata and Hypericum roeperianum had the highest total antibacterial activity (TAA) at 1417 and 963 ml/g respectively. All extracts with the exception of that of Maesa lanceolata, Elaeodendron croceum and Calpurnia aurea had relatively low cytotoxicity with LC50 > 20 μg/ml. Cremaspora triflora had the best selectivity index (SI) against S. aureus and E. coli of 2.87 and 1.15 respectively. Hypericum roeperianum had a SI of 1.10 against B. cereus. Bioautography revealed 1–6 visible antimicrobial compounds that were generally non-polar.ConclusionsThere was a weak positive, but statistically non-significant correlation between the potency of the extracts and their cytotoxicity (R = 0.45, ρ > 0.05). The activity of the extracts on the test bacteria was in some cases not correlated with cytotoxicity, as shown by selectivity indices >1. This means that cellular toxicity was probably not due to compounds with antibacterial activity. Some of the extracts had a good potential for therapeutic use against the bacterial pathogens or for application in treating diarhoea. It does not appear that activity against E. coli is a good predictor of activity against Gram-negative rather than Gram-positive bacteria. Further investigation is in progress on C. triflora and H. roeperianum, both of which had promising activities and potential safety based on cytotoxicity.

Health and economic loss due to these represent a rising burden worldwide which necessitates serious action at regional, national and global levels. Thus, alternative approaches to overcome this problem by using bioactive compounds from traditional medicinal plants are required. This study was designed to evaluate the in-vitro antibacterial activity of Punica granatum fruit peels, Nigella sativa seeds, and Echinops kebericho used in the traditional treatment of urinary tract infections. Methods: An experimental study was employed to evaluate the in vitro antibacterial activity of methanol and ethanol crude extract of Punica granatum fruit peels, Nigella sativa seeds, and Echinops kebericho roots of six dilutions (25, 50, 100, 125, 250, and 500) mg/mL. Disc diffusion and macro broth dilution methods were used to determine antimicrobial activity test and minimum inhibitory concentration respectively against E. coli, P. aeruginosa, K. pneumoniae, P. mirabilis , and S. aureus bacterias. Results: Antibacterial activities of ethanol and methanol crude extract of Punica granatum fruit peels against E. coli ATCC25922, P. aeruginosa ATCC27853, S. aureus ATCC25923, K. pneumoniae UK5099 and P. mirabilis UK5999 had highest inhibition zones among tested plants. All tested bacteria were highly sensitive to Punica granatum extract. The second most active plant extract in inhibiting the growth of tested bacteria was Nigella sativa while Echinops kebericho showed the smallest efficacy against tested bacteria. The inhibition zone diameter produced by the methanol extract of each screened plant had higher inhibition zones than ethanol extract. Conclusion: The crude extracts of Punica granatum fruit peels, Nigella sativa seeds, and Echinops kebericho roots have promising antibacterial activity against tested uropathogenic bacteria. The 66.7% of Pseudomonas aeruginosa ( P. aeruginosa ) strains showing resistance against Ciprofloxacin, Meropenem, Cotrimoxazole and K. pneumoniae strains were 100% resistant to Ceftriaxone, Ceftazidime, Ciprofloxacin, and Augmentin. 7 For E. coli high resistance rates were observed for ampicillin 83.81%, amoxicillin 75.79%, tetracycline 67.18%, trimethoprim-sulfamethoxazole 57.47%, and cephalothin 56.69% 8 and GP, Staphylococcus aureus ( S. aureus ) showed a high level of resistance for cotrimoxazole (53.3%), ampicillin (43%) and penicillin (36.7%). 9 Herbal traditional medicines are effective to combat bacterial resistance of uropathogens with high efficacy, and easy availability with minimal side effects, 10 this led the researcher to find new antimicrobial agents in particular from traditional medicinal plants. 11 One of the common traditional medicinal plants reported to have therapeutic potential for the management and cure of UTIs is Cranberry ( Vaccinium macrocarpon ). Abstract:The present study was isolate Bacterial pathogens form Urinary Tract Infection and identified the Bacterial pathogens from UTI patients. Determination of the antibiotic drug resistant pattern of the isolated pathogenic bacteria using standard antibiotic discs Ampicilin (25μg), Erithromycin (15μg), Chloramphenicol (10μg) Gentamicin (10μg) and Tetracycline (30 μg).The study was carried out, in vitro screening of ethanolic extracts of some medicinal plants against the bacterial pathogens Escherichia coli, Proteus vulgaris, Staphylococcus aureus and Pseudomonas aeruginosawere isolate from the UTI. When compared with standard antibiotic disc selected plants extracts were showed maximum zone of inhibition against all the pathogens. This investigation strongly recommends that phytochemical studies are required to determine the types of compounds responsible for the antibacterial effect of these medicinal plants. Abstract:Urinary tract infection is one of the most common bacterial disease found in all age groups and has become a major health problem in developing countries. UTI’s are treated with various antibiotics but extensive and inappropriate use of antibiotics promotes the development of antibiotic resistance among uropathogenic bacteria and also responsible for emerging multidrug resistant strains. The development of drug resistance among uropathogens made treatment of UTI is difficult which has led to the search of new source of antimicrobial agent that act against these uropathogenic bacteria. The ethnomedicinal plants are natural resource and known as traditional healer, which can use for successful therapy of various infectious diseases. In present study, we observed antibacterial potential of methanolic root extract of two medicinal plants and against urinary tract infection causing uropathogens. For this 120 urine samples of suspected patients were collected from different hospitals of Ujjain and processed in SRL laboratory Ujjain Center. It was seen that 72 samples were positive and showing significant bacteriuria and prevalence of UTI was 60%. The major causal bacterial agent was , and and their frequency of occurrence was 37.5%, 25%, 15.2%, 13.8% and 9.7%. The antibacterial effect of methanolic plant extracts of both and was determined against isolated uropathogens by disc diffusion method. The effectiveness of both and was observed and it was found that they have effective antibacterial activity against uropathogens so they can be used as alternative drug against uropathogens.
Antibacterial activity of Citrullus lanatus seed extract against clinical isolates of Klebsiella pneumoniae and Staphylococcus aureus

Abstract:As a result of increased emergence of bacterial resistance to the conventionally used antibiotics, it is necessary to search for alternatives from other sources like plant materials. This study was aimed at evaluating the in vitro antibacterial activity of Citrullus lanatus seed extract against isolates of Klebsiella pneumoniae from urine and Staphylococcus aureus from wound. The seeds were extracted using distilled water and methanol via cold maceration and then analysed for the presence of phytochemicals using standard methods. Tannins, steroids, cardiac glycosides and carbohydrates were present in both extracts. Only the aqueous extract contained alkaloids. The antibacterial activity of the two extracts was determined by agar well diffusion method. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined using broth dilution method. The aqueous extract had better activity than the methanol extract, having inhibition zones of 20 mm and 23 mm at 200 mg/mL and, 10 mm and 11 mm at 25 mg/mL against Klebsiella pneumoniae isolates respectively, and mean zone of inhibition between 27.50 mm to 29.33 mm at 200 mg/mL and, 16.50 mm to 17.33 mm at 25 mg/mL against Staphylococcus aureus isolates with MIC and MBC values of 0.195 mg/mL and 0.39 mg/mL against Klebsiella pneumoniae and, mean MIC and MBC values ranging from 0.30 mg/mL to 0.39 mg/mL and 0.78 mg/mL to 2.73 mg/mL against Staphylococcus aureus isolates. The results obtained reveal the potential use of Citrullus lanatus seeds in the development of standard antibiotics.
Effect of ethnomedicinal plants used in folklore medicine in Jordan as antibiotic resistant inhibitors on Escherichia coli

Abstract:Background: Escherichia coli occurs naturally in the human gut; however, certain strains that can cause infections, are becoming resistant to antibiotics. Multidrug-resistant E. coli that produce extended-spectrum beta lactamases (ESBLs), such as the CTX-M enzymes, have emerged within the community setting as an important cause of urinary tract infections (UTIs) and bloodstream infections may be associated with these community-onsets. This is the first report testing the antibiotic resistance-modifying activity of nineteen Jordanian plants against multidrug-resistant E. coli. Methods: The susceptibility of bacterial isolates to antibiotics was tested by determining their minimum inhibitory concentrations (MICs) using a broth microdilution method. Nineteen Jordanian plant extracts (Capparis spinosa L., Artemisia herba-alba Asso, Echinops polyceras Boiss., Gundelia tournefortii L, Varthemia iphionoides Boiss. & Blanche, Eruca sativa Mill., Euphorbia macroclada L., Hypericum trequetrifolium Turra, Achillea santolina L., Mentha longifolia Host, Origanum syriacum L., Phlomis brachydo(Boiss.) Zohary, Teucrium polium L., Anagyris foetida L., Trigonella foenum-graecum L., Thea sinensis L., Hibiscus sabdariffa L., Lepidium sativum L., Pimpinella anisum L.) were combined with antibiotics, from different classes, and the inhibitory effect of the combinations was estimated. Results: Methanolic extracts of the plant materials enhanced the inhibitory effects of chloramphenicol, neomycin, doxycycline, cephalexin and nalidixic acid against both the standard strain and to a lesser extent the resistant strain of E. coli. Two edible plant extracts (Gundelia tournefortii L. and Pimpinella anisum L.) generally enhanced activity against resistant strain. Some of the plant extracts like Origanum syriacum L.(Labiateae), Trigonella foenum- graecum L.(Leguminosae), Euphorbia macroclada (Euphorbiaceae) and Hibiscus sabdariffa (Malvaceae) did not enhance the activity of amoxicillin against both standard and resistant E. coli. On the other hand combinations of amoxicillin with other plant extracts used showed variable effect between standard and resistant strains. Plant extracts like Anagyris foetida (Leguminosae) and Lepidium sativum (Umbelliferae) reduced the activity of amoxicillin against the standard strain but enhanced the activity against resistant strains. Three edible plants; Gundelia tournefortii L. (Compositae) Eruca sativa Mill. (Cruciferae), and Origanum syriacum L. (Labiateae), enhanced activity of clarithromycin against the resistant E. coli strain. Conclusion: This study probably suggests possibility of concurrent use of these antibiotics and plant extracts in treating infections caused by E. coli or at least the concomitant administration may not impair the antimicrobial activity of these antibiotics.

Abstract:Urinary tract infections (UTIs) are becoming more common, requiring extensive protection from antimicrobials. The global expansion of multi-drug resistance uropathogens in the past decade emphasizes the necessity of newer antibiotic treatments and prevention strategies for UTIs. Medicinal plants have wide therapeutic applications in both the prevention and management of many ailments. Bacopa monnieri is a medicinal plant that is found in the warmer and wetlands regions of the world. It has been used in Ayurvedic systems for centuries. The present study aimed to investigate the antibacterial potential of the extract of B. monnieri leaves and its bioactive molecules against UTIs that are caused by Klebsiella pneumoniae and Proteus mirabilis. This in vitro experimental study was conducted by an agar well diffusion method to evaluate the antimicrobial effect of 80% methanol, 96% ethanol, and aqueous extracts of B. monnieri leaves on uropathogens. Then, further screening of their phytochemicals was carried out using standard methods. To validate the bioactive molecules and the microbe interactions, AutoDock Vina software was used for molecular docking with the Klebsiella pneumoniae fosfomycin resistance protein (5WEW) and the Zn-dependent receptor-binding domain of Proteus mirabilis MR/P fimbrial adhesin MrpH (6Y4F). Toxicity prediction and drug likeness were predicted using ProTox-II and Molinspiration, respectively. A molecular dynamics (MD) simulation was carried out to study the protein ligand complexes. The methanolic leaves extract of B. monnieri revealed a 22.3 mm ± 0.6 mm to 25.0 mm ± 0.5 mm inhibition zone, while ethanolic extract seemed to produce 19.3 mm ± 0.8 mm to 23.0 mm ± 0.4 mm inhibition zones against K. pneumoniae with the use of increasing concentrations. In the case of P. mirabilis activity, the methanolic extracts showed a 21.0 mm ± 0.8 mm to 24.0 mm ± 0.6 mm zone of inhibition and the ethanol extract produced a 17.0 mm ± 0.9 mm to 23.0 mm ± 0.7 mm inhibition zone with increasing concentrations. Carbohydrates, flavonoids, saponin, phenolic, and terpenoid were common phytoconstituents identified in B. monnieri extracts. Oroxindin showed the best interactions with the binding energies with 5WEW and 6Y4F, −7.8 kcal/mol and −7.6 kcal/mol, respectively. Oroxindin, a bioactive molecule, followed Lipinski's rule of five and exhibited stability in the MD simulation. The overall results suggest that Oroxindin from B. monnieri can be a potent inhibitor for the effective killing of K. pneumoniae and P. mirabilis. Additionally, its safety has been established, indicating its potential for future drug discovery and development in the treatment for UTIs.

Abstract:Development of cheap and reliable diagnostics is pivotal in the control of infectious diseases. Currently, one of the best strategies to tackle infectious disease is the use of natural products from plant origin, due to the perception that long term use of western medicine induces severe complications and also to stop the prevalence of antibacterial infection. This study aims at substantiating the traditional use of selected medicinal plants with antibacterial claim for possible lead for the development of more potent drugs in the treatment of bacterial infections. The preliminary phytochemical content, antioxidant and antibacterial potential of the selected medicinal plants were evaluated using standard procedures. Thirtheen plants were obtained through questionnaire administration: Garcinia kola, Costus afer, Vitellaria paradoxa, Pycnanthus angolensis, Cola acuminata Acanthospermum hipida, Aloe vera, Euadenia trifoliate, Microglossa abzelii, Moringa oleifera, Bryophyllum pinnatum, Baphia nitida and Afromonium melegueta. Thin layer chromatography (TLC) profiling of the various selected plants were carried out using two solvent system consisting of n-Hexane and ethyl acetate in varying ratio (8:2 and 7:3). Eight (8) plants, which showed good chromatographical profiling with the used solvent, were selected for further experiment (Garcinia kola, Vitellaria paradoxa, Acanthospermum hispidum, Aloe vera, Microglossa abzelii, Baphia nitida, Moringa oleifera and Bryophyllum pinnatum). These plants were subjected to qualitative phytochemistry and five plants (Moringa oleifera, Vitellaria paradoxa, Baphia nitida, Bryophyllum pinnatum and Garcinia kola) selected for quantitative phyto-screening for alkaloids, flavonoids, saponins and tannins. The antibacterial screening was carried out using Agar well diffusion and minimum inhibitory concentration (MIC) against the following isolate Staphylococcus aureus, Escherichia coli, Salmonella typhi and Bacillus subtilis. The antioxidant potential of the selected five plants were evaluated using in vitro (DPPH and FRAP) and the best plant was used for in vivo (SOD and MDA) analysis. The phytochemistry of the 13 plants revealed the presence or absence of Alkaloids, saponins, tannins, flavonoids, terpenoids, cardiac glycosides, carbohydrates and protein in the selected plants, and the best five were selected for quantitative evaluation showed they all have good quantity of alkaloids, flavonoids, saponins and tannins. The plants exhibited mild to strong antibacterial activities, having an MIC value ranging from 2.5 to >20 μg/mL when compared to ciprofloxacin (control) that is 5μg/mL. The plants showed good free radical scavenging activities and efficient ferric reducing power in a concentration dependent manner and a good enzyme activity of the superoxide dismutase (SOD) at a concentration of 100 μ/mL with a reduction in malondialdehyde (MDA) production as a marker of lipid peroxidation. The various plants have potentials for development of drugs for the treatment of infectious diseases; and a potential for further evaluation of their antibacterial claim.
In Vitro Antibacterial Activity, Preliminary Phytochemical Screening Profile, and In Vivo Toxicity of Seven Traditional Medicinal Plants in Ethiopia

Abstract:The threat of antibiotic-resistance calls for novel antibacterial agents. This study was aimed at screening medicinal plants for their antibacterial properties, phytochemical content and safety. Leaves of Allophylus abyssinicus (Hochst.) Radlk., Dicliptera laxata C.B.Clarke, Ligustrum vulgare L., Solanecio gigas (Vatke) c. Jeffrey and Gymnanthemum myrianthum (Hook.f.) H.Rob.; leaf and stem-bark of Olinia rochetiana A. Juss. and the seed of Cucurbita pepo L. were used. Chloroform and ethanol were used to extract G. myrianthum, D. laxata and O. rochetiana; ethyl acetate and methanol for the rest, and water for all. The extracts were tested against clinical/standard strains of Escherichia coli, Pseudomonas aeruginosa, Salmonella typhi and Staphylococcus aureus by the agar-diffusion method. The minimum inhibitory concentrations (MIC) and minimum bactericidal concentrations (MBC) were determined. Acute toxicity to mice was checked and preliminary phytochemical screening was done. Thirteen extracts, out of 24, were active (inhibition zone >7 mm) at differing levels (9.67±0.33-25.66±0.57 mm) against at least one bacterial strain. The MICs and MBCs were 1.95-15.6 mg/mL and 7.8-125 mg/mL respectively. The aqueous extract of S. gigas, methanol extracts of L. vulgare and A. abyssinicus, and ethanol extract of O. rochetiana leaf were the most active (MIC 1.95mg/ml) against S. aureus. Ethyl acetate extracts of A. abyssinicus, L. vulgare and S. gigas; aqueous of C. pepo, O. rochetiana and G. myrianthum; and all D. laxata had no antibacterial activity. P. aeruginosa was the least susceptible to any extract, although the methanol and aqueous extracts of S. gigas performed better against it. Preliminary phytochemical screening of selected extracts for phenols, flavonoids, tannins, steroids, terpenoids, steroidal glycosides, alkaloids, saponins, resins and glycosides showed positivity at least for four of these phytochemicals with glycoside and terpenoids in nearly all extracts and resin in none. The plants were not toxic to mice at 2000 mg/kg. Further consideration of S. gigas, L. vulgare, A. abyssinicus and O. rochetiana is recommended in light of their promising potential and safety.

Abstract:Background: With increasing bacterial resistance for antibiotic, there is an increasing interest in medicinal herbal as a new source of an antimicrobial agents and with less side effects than conventional drugs. Objective: Was to evaluate the potential of Syzygium aromaticum and rosemarinus officinalis extract on Proteus mirabilis isolates in vitro. Methods: Six pathogenic gram negative bacilli belonging to three different genus (E.coli, Proteus mirabilis, and Enterobacter aerogenes) and two isolates from each species were used in this study and four concentration (100%, 75%, 50%, 25% mg/ml ) from each plant extracts (aqueous and ethanolic of Syzygium aromaticum and rosemarinus officinalis extracts ) were prepared to be antibacterial agent. antibacterial activity was measured by using wells method and five antibiotics using to compare with the activity of antibacterial agent. Results: The result of the study shown that the ethanolic extracts of Syzygium aromaticum is a strong antibacterial agent (with diameter of inhibition zone 25 mm) against Proteus mirabilis. Enterobacter aerogenes with (15mm) of inhibition zon, E.coli did not show any inhibition zone also the aqueous and ethanolic of rosemarinus officinalis extracts and aqueous of Syzygium aromaticum extracts did not show any activity towards any gram negative bacilli also the results of the study show resistance isolates of E.coli and Proteus mirabilis toward several antibiotic while Enterobacter aerogenes is sensitive for more antibiotics that used in this study. Conclusions: Our study concluded that the extract of Syzygium aromaticum has high potential as antibacterial agent against UTI that caused by Proteus mirabilis, clove (syzygium aromaticum) can be used in pharmaceutical for the production of new synthetic agents. There is an increasing interest in phytochemicals as new sources of natural antioxidant and antimicrobial agents. The natural compounds which have biological activities are interest to pharmaceutical industries especially for treatment of human disease of microbial origin and several pathological disorders such as cancergenesis, coronary atherosclerosis, as well as in ageing processes.
Effect of certain bioactive plant extracts on clinical isolates of beta-lactamase producing methicillin resistant Staphylococcus aureus

Abstract:Ethanolic extracts and some fractions from 10 Indian medicinal plants, known for antibacterial activity, were investigated for their ability to inhibit clinical isolates of beta-lactamase producing methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-sensitive S. aureus (MSSA). Synergistic interaction of plant extracts with certain antibiotics was also evaluated. The MRSA test strains were found to be multi-drug resistant and also exhibited high level of resistance to common beta-lactam antibiotics. These strains produced beta-lactamases, which hydrolyze one or other beta-lactam antibiotics, tested. The extract of the plants from Camellia sinensis (leaves), Delonix regia (flowers), Holarrhena antidysenterica (bark), Lawsonia inermis (leaves), Punica granatum (rind), Terminalia chebula (fruits) and Terminalia belerica (fruits) showed a broad-spectrum of antibacterial activity with an inhibition zone size of 11 mm to 27 mm, against all the test bacteria. The extracts from the leaves of Ocimum sanctum showed better activity against the three MRSA strains. On the other hand, extracts from Allium sativum (bulb) and Citrus sinensis (rind) exhibited little or no activity, against MRSA strains. The antibacterial potency of crude extracts was determined in terms of minimum inhibitory concentration (MIC) by the tube dilution method. MIC values, of the plant extracts, ranged from 1.3 to 8.2 mg/ml, against the test bacteria. Further, the extracts from Punica granatum and Delonix regia were fractionated in benzene, acetone and methanol. Antibacterial activity was observed in acetone as well as in the methanol fractions. In vitro synergistic interaction of crude extracts from Camellia sinensis, Lawsonia inermis, Punica granatum, Terminalia chebula and Terminalia belerica was detected with tetracycline. Moreover, the extract from Camellia sinensis also showed synergism with ampicillin.TLC of the above extracts revealed the presence of major phytocompounds, like alkaloids, glycosides, flavonoids, phenols and saponins. TLC-bioautography indicated phenols and flavonoids as major active compounds.

Abstract:Abstract Background A biofilm is an extracellular polymeric substance (EPS) composed of polysaccharides, proteins, nucleic acids, and lipids that impede antibiotics and immune cells, thus providing a shielded environment for bacterial growth. Due to biofilm formation, some microbes can show up to 1000 fold increased resistance towards the antibiotics than the normal planktonic forms. The study was conducted to screen the crude extracts of medicinal plants used in Nepal for their in vitro antibiofilm activities. Methods Total phenolic and total flavonoid contents were determined by using a Folin-Ciocalteau reagent and aluminium trichloride method, respectively. Resazurin assay was used to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The initial antibiofilm activities and their inhibitory concentration (IC 50 ) values were determined by the microtiter based modified crystal violet staining method. Results Out of 25 different plant extracts were used for the study, methanolic extracts of 20 plants showed a biofilm inhibition activity against five different strong biofilm producing Escherichia coli strains. Calotropis gigantea exhibited inhibition against all five different E. coli strains with IC 50 values ranging from 299.7 ± 20.5 to 427.4 ± 2.7 μg/mL. Apart from that, Eclipta prostrata also showed biofilm formation inhibition, followed by Eupatorium adenophorum , Moringa oleifera , Ocimum tenuifolium, Oxalis lantifolia, Prunus persica, and Urtica parviflora . The extracts of C. gigantea, E. prostrata, Mangifera indica, O. tenuifolium, P. persica, and U. parviflora exhibited a moderate to poor MIC value ranging from 625 to 2500 μg/mL. The highest amount of phenolic content (TPC) was found in Acacia catechu followed by Morus alba , which was 38.9 and 25.1 mg gallic acid equivalents, respectively. The highest amount of flavonoid content was found in A. catechu followed by M. indica , which was 27.1 and 20.8 mg quercetin equivalents, respectively. Conclusion Extracts of C. gigantea , E. prostrata, P. persica , U. parviflora, and O. tenuifolium showed antibacterial as well as antibiofilm activity against pathogenic and strong biofilm producing E. coli . Thus, extracts or the pure compound from these medicinal plants could be used as antibiotics in the future.

Abstract:BackgroundThe extracts of the ten selected Sri Lankan medicinal plants have been traditionally used in the treatment of inflammatory mediated diseases. The extracts were investigated for anti-inflammatory and anti-oxidant potential in vitro to identify bio-active extracts for further chemical characterization.MethodsIn vitro anti-inflammatory activities of total ethanol extracts were investigated measuring the inhibitory activities of four pro-inflammatory enzymes, arachidonate-5- lipoxygenase (A5-LOX), hyaluronidase (HYL), xanthine oxidase (XO) and inducible nitric oxide (iNO) synthase. Cytotoxicity of extracts were determined by MTT assay. Oxidative burst inhibition (OBI) on human whole blood (WB) and isolated polymorphoneutrophils (PMNs) was carried out for a selected bio-active extract. Anti- oxidant activities of the extracts were determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, ferric reducing antioxidant power (FRAP), ferrous ion chelation (FIC) and oxygen radical absorbance capacity (ORAC) assays. Total polyphenol and total Flavonoid contents of the extracts were also determined. The most active plant extract was analysed using Gas chromatography-Mass spectrometry (GC-MS) and High Performance Liquid Chromatography (HPLC).ResultsThe ethanol bark extract of Flacourtia indica showed the highest A5-LOX (IC50: 22.75 ± 1.94 g/mL), XO (70.46 ± 0.18%; 250 μg/mL) and iNOs inhibitory activities on LPS- activated raw 264.7 macrophage cells (38.07 ± 0.93%; 500 μg/mL) with promising OBI both on WB (IC50: 47.64 2.32 μg/mL) and PMNs (IC50: 5.02 0.38 μg/mL). The highest HYL inhibitory activity was showed by the leaf extracts of Barathranthus nodiflorus (42.31 ± 2.00%; 500 μg/mL) and Diospyros ebenum (41.60 ± 1.18%; 500 μg/mL). The bark and leaf extracts of Callophyllum innophyllum (IC50: 6.99 ± 0.02 μg/mL) and Symplocus cochinchinesis (IC50: 9.85 ± 0.28 μg/mL) showed promising DPPH free radical scavenging activities. The GC-MS analysis of ethanol bark extract of F. indica showed the presence of two major bio-active compounds linoleic acid ethyl ester and hexadecanoic acid, ethyl ester (> 2% peak area). The HPLC analysis showed the presence polyphenolic compounds.ConclusionThe ethanol bark extract of F. indica can be identified as a potential candidate for the development of anti-inflammatory agents, which deserves further investigations. The bio-active plant extracts may be effectively used in the applications of cosmetic and health care industry.
ANTIBACTERIAL ACTIVITY OF BRYOPHYLLUM PINNATUM AGAINST CLINICAL ISOLATES OF ESCHERICHIA COLI AND KLEBSIELLA PNEUMONIA FROM URINARY TRACT INFECTION PATIENTS

Abstract:The emergence and spread of resistant strains of bacteria to routinely used antibiotics has made it imperative to continuously search for alternatives that can be used to cure infections, so as not to return to pre-antibiotic era. Plants are known to contain bioactive compounds that can be explored and used in the treatment of infection. The study investigated the antibacterial activity of ethanolic extracts of dried leaf extracts of Bryophyllum pinnatum against clinical isolates of Escherichia coli and Klebsiella pneumoniae from urinary tract infection patients attending Shehu Muhammad Kangiwa Medical Centre. The photochemical screening of the crude extracts of Bryophyllum pinnatum (leaf of life) carried out using standard procedures described by Trens and Jens 2002, revealed the presence of Saponins, Flavonoid, Terpenoids, Alkaloids, Phenol and Tannins . The antibacterial activity of the extracts was carried out using agar cup diffusion method. Four concentrations (200,100, 50 and 25mg/ml) of the extract were tested against Escherichia coli and Klebsiella pneumoniae. The zones of inhibition measured at 200,100, 50 and 25mg/ml were: 00,00,00 and 00 mm respectively. The extracts therefore showed no antibacterial activity against the isolates at all the four concentrations tested. The results of this study proofs that the ethanolic extract of B. pinnatum leaves has no effect on E. coli and K. pneumoniae isolated from patients with Urinary Tract Infection, and hence cannot be used to treat infections caused by these bacteria.
In vitro antibacterial activity of Morinda citrifolia extracts against eight pathogenic bacteria species

Abstract:Medical professionals continue to face a severe issue with the evolution of resistance to conventional antibiotics. The search for new novel compounds from plants has been proven to be the alternative solution. Morinda citrifolia is used traditionally for the treatment of infectious diseases. The present study investigates the antibacterial properties of M . citrifolia root, leaf, and fruit (fresh, dried, and fermented) extracts on three-gram-positive and five-gram-negative bacteria. The plant parts were processed and extracted in distilled water and ethanol (60%, 80%, and absolute (100%)). The antibacterial activities of the extracts were assessed in vitro using the agar well diffusion method, with Ciprofloxacin serving as the positive control. All the tests were conducted three times to obtain the average value of inhibition zones. Overall, root extracts showed the most significant antibacterial activity, followed by dried fruit, fermented fruit extract, fresh fruit, and the least leaf extract. Using one-way ANOVA and Tukey's post-hoc tests, the statistical analysis revealed significant differences in antibacterial activity among the extracts and solvent concentrations. The 100% ethanol extracts had significantly higher zones of inhibition compared to the other solvents. The most inhibitory activity was against Campylobacter spp. (21.33±1.80) for the 80% ethanol root extract. All the extracts of M . citrifolia were found to exhibit moderate antibacterial activity against all the bacteria pathogens. However, Enterococcus faecium , Campylobacter spp., and Bacillus cereus were most sensitive to all the plant extracts while Shigella spp. and Klebsiella spp. showed resistance to most extracts. This observed difference is significant for each strain extract depending on the bacteria strain and the type of solvent extract (p < 0.001). The findings indicate a promising antimicrobial potential of M . citrifolia extracts.

Abstract:This study determined the antioxidant and antibacterial activities of Thymus schimperi (Ts), Rhamnus prinoides (Rp), and Justicia schimperiana (Js) from Addis Ababa against MDR-Uropathogenic bacteria. Accordingly, Thymus schimperi had the highest total phenolic (TPC), flavonoid (TFC) and proanthocyanidin content. In Ts, the GC-MS analyses predicted 14 bioactive compounds. And among these, hexanedioic acid, bis(2-ethylhexyl) ester, thymol, and o-cymen-5-ol were the most predominant compounds, respectively. Six compounds were also predicted in Rp, where hexanedioic acid, bis(2-ethylhexyl) ester, β-D-glucopyranoside, methyl, and desulphosinigrin were the predominant, respectively. Whereas in the Js extract, five bioactive compounds were predicted, with hexanedioic acid, mono (2-ethylhexyl) ester, debrisoquine, and 8,11,14-heptadecatrienoate, methyl ester being predominant compounds, respectively. The extracts' TPC showed a strong negative correlation with the DPPH (2,2-diphenyl-1-picrylhydrazyl) assay (r = −0.999; p = 0.023). In addition, the TFC correlated significantly with the ABTS (2,2′-azino-di-(3-ethylbenzthiazoline sulfonic acid)) assay (r = 0.999; p = 0.032). Thymus schimperi showed the highest antibacterial activity against clinical isolates of Escherichia coli and Klebsiella pneumoniae ESBL at 1000 mg/mL, and Ts had the lowest MIC (4 mg/mL) among evaluated extracts against E. coli (ATCC25922). In conclusion, Ts and Rp possess higher predicted bioactive molecules, including antioxidant and antibacterial activities, which are potentially useful in treating urinary tract infections.

Monitoring in vitro antibacterial efficacy of 26 Indian spices against multidrug resistant urinary tract infecting bacteria

Abstract:Background: To screen methanolic extracts of 26 commonly used Indian spices against nine species of uropathogenic bacteria (Enterococcus faecalis, Staphylococcus aureus, Acinetobacter baumannii, Citrobacter freundii, Enterobacter aerogenes, Escherichia coli, Klebsiella pneumoniae, Proteus mirabilis, and Pseudomonas aeruginosa), isolated from clinical samples of a tertiary care hospital for antibacterial activity. Methods: Bacterial strains were subjected to antibiotic sensitivity testing by Kirby-Bauer's disc diffusion method. Monitoring antibacterial potentiality of spice extracts was done by the agar-well diffusion method with multidrug resistant (MDR) strains of nine uropathogens. Results: The Gram-positive (GP) bacteria E. faecalis and S. aureus were resistant to 16 of the 21 antibiotics used. Among the Gram-negative (GN) bacteria, resistant patterns were A. baumannii and E. aerogenes to 12, C. freundii to 14, E. coli to 12, K. pneumoniae to 10, P. mirabilis to 11, and P. aeruginosa to 15 antibiotics of the 18 antibiotics used. The most effective 15 spices, having at least 25-29 mm as the size of the zone of inhibition, were Allium cepa, Brassica juncea, Cinnamomum tamala, Cinnamomum zeylanicum, Coriandrum sativum, Cuminum cyminum, Curcuma longa, Mentha spicata, Murraya koenigii, Nigella sativa, Papaver somniferum, Piper nigrum, S. aromaticum, Trachyspermum ammi, and Trigonella foenum for at least one of the GP or GN MDR bacterial strains used. Moderate control capacity was registered by nine spices, Curcuma amada, Foeniculum vulgare, Illicium verum, Mentha spicata, Papaver somniferum, Syzygium aromaticum, Trachyspermum ammi, Trigonella foenum, and Zingiber officinale. However, the best two spices for controlling all the pathogens used were C. zeylanicum and C. longa, with the highest value of 29 mm as the inhibition zone size. Conclusion: The most effective and unique 16 spice plants recorded for the in vitro control of MDR uropathogens could further be pursued for the development of complementary and supplementary medicine against MDR bacteria.

Abstract:BACKGROUND: The extended-spectrum beta-lactamase (ESBL) -producing Escherichia coli isolates make many serious infections, especially urinary tract infections.OBJECTIVES: The purpose of this study was to determine the antibacterial activities of some natural plant extracts against ESBL-producing E. coli isolates, which harbor the TEM gene in urine samples of the patients who have urinary tract infections.MATERIALS AND METHODS: Evaluation has to be exactly determined for both methods of disk diffusion test and polymerase chain reaction (PCR), separately. We evaluated 120 strains of E. coli isolates from the urine culture of the patients in Boo-Ali Hospital (Zahedan, south-eastern Iran) who were suffering from urinary tract infections. The ESBL-producing E. coli isolates were evaluated by disk diffusion test and PCR through TEM gene detection. The minimal inhibitory concentration (MIC) of commonly used antibiotics including ceftazidime, ceftriaxon, amikacin, gentamicin and ciprofloxacin along with the MIC of the alcoholic extract of different natural plants including Myrtus communis L (Myrtaceae), Amaranthus retraflexus (Amaranthaceae), Cyminum cuminum L (Apiaceae), Marrubium vulgare (Laminaceae) and Peganum. harmala (Zygrophyllaceae) against the ESBL-producing E. coli isolates, which harbor the TEM genes, were determined using the microdulition method.RESULTS: Results of this study showed that in disk diffusion method, 80 samples of E. coli produced ESBLs. In PCR method, the TEM gene distribution in the isolated ESBL-producing organisms was 50 (41.6%). Amikacin was the most effective anti-bacterial agent and ciprofloxacin was the least effective against E. coli isolates. All the natural plant extracts mentioned above, especially P. harmala, were effective against the selected isolates of ESBL-producing E. coli. The most frequent ESBL rate producing E. coli isolates (32 out of 50) had MIC of 2.5 mg/mL in ethanol extract of P. harmala.CONCLUSIONS: The alcoholic extract of P. harmala was very effective against the selected ESBL-producing E. coli isolates harboring the TEM gene. Therefore, it could be suggested as an antibacterial agent in the future. More researches are necessary for detecting the mechanism of this plant's behavior and its pharmacological effects.

Abstract:In this study, the sensitivity and resistance of six types of bacterial isolates, Escherichia coli, pseudomonas aeruginosa, Staphylococcus aureus, Proteus vulgaris, Proteus mirabilis, and Klebsiella pneumoniae, against 10 antibiotics were tested for 10 antibiotics, Ciprofloxicine (CF) 25 g / ml Amoxicillin (AX) 50 g / ml, Ampicillin (Am) 50 µg / ml, Erythromycin (Ery) 15 µg / ml, Gentamycin ( GN) 25 g / ml, Rifampicin (RA) 50 µg / ml, Streptomycin (Sm) 25 µg / ml, Tetracyline (TE) 15 µg / ml Trimethprim (Tri) 10 µg / ml Nalidixic acid (Nal) 30 µg / ml. The bacterial isolates showed different resistance to most antibiotics. One isolate of each bacterium was selected for subsequent experiments. The aqueous and ethanol extracts of Citrus aurantium L fruit were prepared. The inhibitory effect of the prepared plant extracts on the selected bacteria was investigated by using the Agar diffiuision method and the Turbidity method. The prepared plant extracts showed a clear growth inhibitory activity, which was estimated by measuring the inhibition zone with a millimeter of studied bacteria. The results obtained showed that the value of the growth inhibitory activity increased with increasing the concentration of the extracts. Sub minimum inhibition concentration of plant extracts was used as a curing to remove antibiotic resistance of the bacterial species under study. Varying values for the percentage loss of antibiotic resistance were obtained as a result of the efficacy of the extracts, but some extracts did not show curing efficacy. The results of the curing experiments were supported by the characterization of the plasmid DNA on the agarose gel of the curing isolates. Electrophoresis showed the disappearance of the plasmid DNA bundles from the agarose gel for most of the bacteria under study compared to the non-curing isolates.

Abstract:Saudi Arabia flora has many medicinal plants which are traditionally used in inhibition of many human pathogens. Echinops macrochaetus (Camel thistle) and Xanthium spinosum (Spiny cocklebur) from the Asteraceae family and Lemongrass (Oymbopogon citrates) from the Gramineae family are one of the local plants which are quite popular but less studied scientfically. Hence in the present work, they were analysed for their medicinal efficacies , for which they were collected and extracted using either hot water or organic solvents (n-butanol, diethyl ether andethyl acetate). The antibacterial activities of the previous plants extracts were detected and the methanolic extract of the three tested plants gave excellent inhibition of the tested bacterial pathogen, Escherichia coli while the aqueous extracts recorded the lowest bacterial inhibition. The susceptibility of the tested bacteria to the three methanolic plant extracts and control antibiotic was tested using agar well diffusion method. The methanolic extract of Echinops recorded excellent antibacterial activity while Xanthium and Oymbopogon recorded moderate activities against the Gram negative Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Proteus mirabilis in addition to the Gram positive Staphylococcus aureus and Enterococcus faecalis. The mean antibacterial activities (bacterial index) was maximum for the methanol extract of Echinops (21.8 mm) followed by Xanthium (13.8 mm) and Oymbopogon (12.5 mm). Moreover, In vitro MTT test and Neutral Red assays were used to detect any antitumor activities of the three tested plant extracts. Echinops extract showed excellent antitumor activity against two cell lines, MCF-7 (breast cancer) and HepG2 (hepatocellular carcinoma) with no toxicity (recorded using Artimia salina for the assay). Also, using fluorescein diacetate microdilution method, minimal inhibitory concentrations (MICs) and the Fractional Inhibitory Concentrations of the methanolic extract of Echinops, ciprofloxacin and their mixture were calculated. FIC index ranged from 0.89-1.15 that means there is an additive effect between Echinops extract and ciprofloxacin. In conclusion, the results show that Echinops methanolic extract single or mixed with the antibiotic ciprofloxacin demonstrated excellent inhibitory activities for all tested urinary tract pathogenic bacteria.
