Abstract:Background: Researchers are always searching for chemicals with antioxidant properties. The cell wall polysaccharide of Sargassum angustifolium consists of alginic acid. To use this polysaccharide and investigate on its various effects, it must first be isolated from brown algae and the operational parameters of extraction need to be optimized to reach the maximum antioxidant effect. Materials and methods: The effects of changing the parameters (temperature, time, and power of ultrasonic waves) used in the extraction of alginic acid were discussed based on changes in the antioxidant effect. After that, M/G (β-D-mannuronic acid to α-L-guluronic acid ratio) was measured using 1H NMR spectra, and the antioxidant activity of the extracted alginic acid was examined using the DPPH method. Results: The highest antioxidant effect was observed in sample No. 4 with an extraction temperature of 65°C, extraction duration of 25 min, and 640 W of ultrasonication power, whereas the lowest antioxidant effect was observed in sample No. 7 at 45°C, 20 min, and 480 W. Conclusion: Structural changes due to increasing temperature may cause less activity, whereas the optimum temperature was 65°C. There was no direct relationship between M/G and IC50, and it should be examined along with molecular weight. Increasing the intensity of the waves increased the antioxidant activity. The extraction yield can be increased by increasing A/W (Gram of algal sample per mL of solvent). It appears that the extraction under optimum conditions enhances the MM block epimer, which may lead to an increase in the antioxidant activity.
Optimization of alginate extraction conditions from the brown seaweed Dictyota mertensii using a central composite design

Abstract:This work extracts alginate from Dictyota mertensii, a brown macroalga found on the north coast of northeastern Brazil. A Central Composite Design (CCD) using Response Surface Methodology (RSM) was used to optimize the process of extracting the alginate present in the brown alga Dictyota mertensii, considering the factors of time, temperature, and concentration Na2CO3. Extraction yield ( η ), sulfate content, intrinsic viscosity ([ η ]), viscosimetric molar mass (Mv), color parameters (a*, b* e L*), M/G ratio and inhibitory concentration IC50 are model response variables. The Dictyota mertensii algae has its chemical composition expressed. Response surfaces are plotted from the variables studied, a quadratic model was predicted, the influence of factors is analyzed, and response variables are subjected to analysis of variance (ANOVA) and F-test at 95 % confidence. The predicted conditions are experimentally validated. The antioxidant potential was evaluated at seven concentrations (0.2–5.0 mg/mL) based on the stable free radical 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging. The maximum extraction yield was 29.28 ± 0.02 %, and sulfate content ranged from 3.50 ± 0,00 % to 12.33 ± 0.29 %. The maximum values of [ η ] and Mv were 5.84 ± 0.06 dL/g and 278.00 ± 2.66 kDa, respectively. The alginate showed a dark brown color and M/G ratio < 1, indicating that the extracted alginate provides rigid and compact gels, which allows its application in films, encapsulations, and protecting probiotics. The maximum antioxidant capacity and the IC50 were 54.93 ± 0.27 % and 0.51 ± 0.07 mg/mL, respectively, indicating the material obtained as a moderate natural antioxidant.

Optimizing the fucoidan extraction using Box-Behnken Design and its potential bioactivity

Abstract:Fucoidan is a sulfated polysaccharide that occurs naturally in the cell wall of brown seaweeds and has substantial biological efficacy. Optimizing the extraction of fucoidan from different brown seaweeds was the primary goal of this research. The optimization of fucoidan extraction was applied on the brown macroalga Turbinaria turbinata using a Box-Behnken Design (BBD) to inspect the impacts of different pH (3, 5, 7), temperature (70, 80, 90 °C) and extraction duration (60, 120, 180 min) on both the yield and sulfate content of fucoidan. The optimized parameters recorded to maximize the fucoidan yield and its sulfate content were a pH of 3.44 and a temperature of 82.26 °C for 60 min. The optimal conditions obtained from BBD were used for fucoidan extraction from T. turbinata, Sargassum cinereum, Padina pavonica, and Dictyota dichotoma. The highest average of fucoidan yield was derived from P. pavonica (40.76 ± 4.04 % DW). FTIR, 1H NMR, and HPLC were used to characterize extracted fucoidan. The extracted fucoidan's Physical characteristics, biochemical composition, antioxidant potential, antitumor effect against breast cancer cells (MCF-7), and antimicrobial and anticoagulant activity were assessed. The extracted fucoidan from D. dichotoma, followed by that extracted from S. cinereum, which had the highest sulphate content, depicted the highest antioxidant, anticancer, and anticoagulant activities. Fucoidan has demonstrated a strong antimicrobial action against some pathogenic microorganisms; Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Klebsiella pneumonia, and Candida albicans. The anticoagulant properties of fucoidan from D. dichotoma were stronger than those of fucoidan from S. cinereum, T. turbinata, and P. pavonica due to its higher sulphate content. These findings could be used for various biomedical applications to improve the pharmaceutical industry.
Improvement of Alginate Extraction from Brown Seaweed (Laminaria digitata L.) and Valorization of Its Remaining Ethanolic Fraction

Abstract:This study aimed to improve the conventional procedure of alginate isolation from the brown seaweed (Laminaria digitata L.) biomass and investigate the possibility of further valorization of the ethanolic fraction representing the byproduct after the degreasing and depigmentation of biomass. The acid treatment of biomass supported by ultrasound was modeled and optimized regarding the alginate yield using a response surface methodology based on the Box–Behnken design. A treatment time of 30 min, a liquid-to-solid ratio of 30 mL/g, and a treatment temperature of 47 °C were proposed as optimal conditions under which the alginate yield related to the mass of dry biomass was 30.9%. The use of ultrasonic radiation significantly reduced the time required for the acid treatment of biomass by about 4 to 24 times compared to other available conventional procedures. The isolated alginate had an M/G ratio of 1.08, which indicates a greater presence of M-blocks in its structure and the possibility of forming a soft and elastic hydrogel with its use. The chemical composition of the ethanolic fraction including total antioxidant content (293 mg gallic acid equivalent/g dry weight), total flavonoid content (14.9 mg rutin equivalent/g dry weight), contents of macroelements (the highest content of sodium, 106.59 mg/g dry weight), and microelement content (the highest content of boron, 198.84 mg/g dry weight) was determined, and the identification of bioactive compounds was carried out. The results of ultra high-performance liquid chromatography–electrospray ionization–tandem mass spectrometry analysis confirmed the presence of 48 compounds, of which 41 compounds were identified as sugar alcohol, phenolic compounds, and lipids. According to the 2,2-diphenyl-1-picrylhydrazyl assay, the radical scavenging activity of the ethanolic fraction (the half-maximal inhibitory concentration of 42.84 ± 0.81 μg/mL) indicated its strong activity, which was almost the same as in the case of the positive control, synthetic antioxidant butylhydroxytoluene (the half-maximal inhibitory concentration of 36.61 ± 0.79 μg/mL). Gram-positive bacteria (Staphylococcus aureus, Enterococcus faecalis, and Bacillus cereus) were more sensitive to the ethanolic fraction compared to Gram-negative bacteria (Escherichia coli, Pseudomonas aeruginosa, and Shigella sonnei). The obtained results indicated the possibility of the further use of the ethanolic fraction as a fertilizer for plant growth in different species and antifouling agents, applicable in aquaculture.

Abstract:Alginate is a naturally occurring polysaccharide polymer having very distinct and useful characteristics feasible across multiple sectors including agriculture, food, textile and pharmaceuticals serves as an emulsifier, film former, thickener, stabilizer, viscosifier, gelling agent, and therapeutic agent. The seaweed Sargassum wightii (Phaeophyceae) sourced from the Gulf of Mannar was utilized to produce sodium alginate. The seaweed underwent treated with five different acids: H2SO4, HCl, CH3COOH, HNO3, and C6H8O7 to achieve the high-quality alginate, with sulfuric acid yielding the highest at 401±13.11 mg/g. The FTIR and 1H NMR spectroscopic studies were elucidated the chemical properties of extracted alginates. The FTIR results revealed the presence of -OH, O-C-O, C-OH, and C-H groups in the extracted alginate samples. 1H NMR spectroscopic studies revealed the composition of mannuronic and glucuronic acid residues, with acetic acid accounting for the high M/G ratio of 0.85. Additionally, the DLS spectroscopy was employed to determine the particle size, polydispersity index and scattering index of alginate samples, and TGA analyses revealed that extracted alginate samples exhibit good thermal stability, with citric acid representing the highest.

Abstract:Marine environment is a major potential source of functional materials, including polysaccharides, vitamins, enzymes, oils, antioxidants and peptides. All these materials are extracted from different marine living organisms including microbes, plants and animals. Among these seaweeds or marine macroalgae are one of the important sources and they are a part of staple diet from time immemorial in the orient as they are nutritionally rich materials. Those species that adapted to these pressures will expand their living boundaries and higher potential of row material availability in industry like pharmaceutical market, textile, fertilizer and for animal and human consumption. The present study concerns about the specific brown seaweeds, which is suitable material for alginate and growing abundantly at seacoast of Gujarat. Out of many species of seaweeds growing on the coastline of Veraval, four species viz., Sargassum tenerrimum, Dictyota dichotoma, Spathoglossum asperum, Iyengaria stellata were selected for alginate extraction. The focus of this study is to utilize natural resources as alternatives and sustainability of human health to use sodium alginate as novel polymer and which is also biodegradable. It may be associated to other biologically active molecules and has a wide range of physicochemical and biochemical properties. Because of amazing properties, alginate and its salts are used in drug delivery system.

Abstract:The incorporation of natural fillers such as seaweed may potentially enhance the properties of biopolymer films. In this study, we investigated the effect of seaweed powder as a bio-filler in alginate-based films at different concentrations (10, 30, and 50%, w/w alginate) and particle sizes (100 and 200 μm) on the mechanical, barrier, antioxidant, and antimicrobial properties of alginate which are essential for food packaging applications. Initially, mechanical properties of the alginate films prepared at different temperatures were evaluated to find the optimal temperature for preparing alginate solution. The addition of seaweed powder did not have any positive effect on the mechanical properties of the alginate films. However, the barrier (water vapor transmission rate) and antioxidant properties were improved with the addition of seaweed filler regardless of concentration. In addition, selected films were prepared in plasma-activated water (PAW). The mechanical properties (tensile strength, but not elongation at break) of the films prepared with PAW improved compared to the films prepared in distilled water, while a significant decrease was observed when incorporated with the seaweed filler. The films prepared in PAW also showed improved barrier properties compared to those prepared in distilled water. The antimicrobial activity of the alginate-seaweed film-forming solution was in general more pronounced when prepared with PAW and stored at 10 °C, particularly at the highest concentration of the film-forming solution (83.3% v/v). A more pronounced inhibitory effect was observed on the Gram-positive S. aureus than on the Gram-negative E. coli, which has been attributed to the different composition and structure of the respective cell walls. This study has demonstrated the potential of seaweed filler in combination with PAW towards enhanced functionality and bioactivity of alginate films for potential food packaging applications.

Sodium alginate extracted algae are gaining attention as alternative biopolymer materials. This research aimed to extract and characterize the natrium alginate from three brown algae obtained from Sanur Beach, Denpasar, Bali, Indonesia. Alginate extraction was carried out using acid hydrolysis by Na2CO3 with further purification using isopropyl alcohol. Physicochemical characterization was carried out using FT-IR. The results showed that Sargassum aquifolium and Turbinaria ornata had higher yields (%) than Padina australis (P 0.05) in water content, protein content, fat content and pH of the three brown algae. Overall, these three types of brown algae have values ??that are close to the sodium alginate standard set by the Food Chemical Codex (FCC), except for the results of Padina australis in the yield parameter. The functional group Na which is characteristic of sodium alginate is present in all alginate isomers, but there are some functional groups that are still not available in the extraction process in this study based on FT-IR analysis. These findings indicate that sodium alginate extracts from various types of brown algae have specific characteristics as defined by the Food Chemical Codex (FCC) and show great potential to be used as biopolymer materials, especially in the biomedical field.
Ultrasound-assisted extraction of polysaccharides from brown alga (Sargassum angustifolium): structural characterization, antioxidant, and antitumor activities

Abstract:In this study, crude polysaccharide of Sargassum angustifolium (PSA) was extracted with an extraction efficiency of 7.14% and a molecular weight of 1831.55 kDa. Then two fractions PSA-1 and PSA-2 with yields of 38.12% and 61.07% and molecular weights of 809.2 and 644.1 kDa were obtained by a DEAE column. Based on the results, fucose (40.41%), glucose (19.30%) and galactose (27.13%) were identified as the dominant monosaccharides in polysaccharides. The IC 50 value of DPPH free radical scavenging by PSA, PSA-1 and PSA-2 was 0.32, 0.24 and 0.19 mg/mL, respectively. Also, the IC 50 value of ABTS radical scavenging by PSA, PSA-1 and PSA-2 was estimated at about 0.24, 0.17 and 0.14 mg/mL, respectively. According to the results, Fraction PSA-2 had the highest antioxidant activity. In this study, the ability of all fractions to inhibit Hela cancer cells was determined, and the most significant effect was observed by fraction PSA-2. The inhibition rate of HeLa cells by PSA and PSA-1 and PSA-2, at a concentration of 0.4 mg/mL was about 40.56% and 51.70% and 70.81%, respectively. Therefore, S. angustifolium polysaccharide, especially fraction PSA-2, can be introduced as a natural compound with anticancer and antioxidant activity for food and pharmaceutical applications.

Abstract:Alginate, a promising biopolymer in the food, biomedical, pharmaceutical, and electronic materials industries, is characterized by its biodegradability, biocompatibility, low toxicity, and gel-forming properties. It is most abundantly found in brown algae. However, conventional dilute acid and alkali extraction methods face limitations in commercialization due to their long processing time, low throughput, and high solvent requirements. In this study, a microwave-assisted extraction (MAE) process for sodium alginate was designed to improve extraction efficiency. The solid/liquid ratio, extraction temperature, and extraction solvent concentration were major variables affecting sodium alginate extraction from Undaria pinnatifida (sea mustard). They were then statistically optimized using response surface methodology. Under optimal conditions (13.27 g/L, 91.86 °C, 2.51% (w/v), and 15 min), the yield was 38.41%, which was 93.43% of the theoretical content of sodium alginate in Undaria pinnatifida. Our work has confirmed the productivity and industrial feasibility of the efficient extraction of sodium alginate from marine biomass, and we hope that it will serve as an encouraging case for the application of biopolymers as one of the desirable options for alternative petrochemicals to construct a sustainable society.

Characterization and gelling properties of a bioactive extract from Ascophyllum nodosum obtained using a chemical-free approach

Abstract:The bioactivity and gelling properties of a carbohydrate-rich algal extract obtained from locally harvested Ascophyllum nodosum seaweed using a chemical-free approach were investigated for its potential interest in food applications. Physicochemical characterisation and compositional analysis of the extract, using FTIR, biochemical methods and monosaccharide analysis, confirmed the presence of alginates and fucoidans, although the main polysaccharide present in it was laminarin. Significant amounts of phenolic compounds (~9 mg phloroglucinol/100 mg sample) were also detected. As a result, the extract exhibited good antioxidant activity. It also showed promising prebiotic potential, promoting the growth of beneficial Lactobacillus sp. and Bifidobacteria sp. when compared with commercial prebiotics, but not that of pathogenic bacteria such as E. coli or P. aeruginosa. The gelling properties of the raw extract were explored to optimize hydrogel bead formation by external gelation in CaCl2 solutions. This was enhanced at neutral to alkaline pHs and high extract and CaCl2 concentrations. The mechanical strength, nano- and microstructure of the hydrogel beads prepared under optimised conditions were determined using compression tests, synchrotron small- and wide-angle X-ray scattering (SAXS/WAXS) and scanning electron microscopy (SEM). It was concluded that the raw algal extract at neutral pH had potential for use as a gelling agent, although further enrichment with alginate improved the mechanical properties of the obtained gels. The advantages and disadvantages of applying the non-purified algal extract in comparison with purified carbohydrates are discussed.
Antioxidant and cytotoxic activities of sulfated polysaccharides from five different edible seaweeds

Abstract:In recent times, there has been a growing interest in the exploration of antioxidants and global trend toward the usage of seaweeds in the food industries. The low molecular weight up to 14 kDa sulfated polysaccharides of seaweeds (Portieria hornemannii, Spyridia hypnoides, Asparagopsis taxiformis, Centroceras clavulatum and Padina pavonica) were evaluated for in vitro antioxidant activities and cytotoxic assay using HeLa cell line and also characterized by FTIR. The high yield (7.74% alga dry wt.) of sulfated polysaccharide was observed in P. hornemannii followed by S. hypnoides (0.69%), C. clavulaum (0.55%) and A. taxiformis (0.17%). In the brown seaweed P. pavonica, the sulfated polysaccharide yield was 2.07%. High amount of sulfate was recorded in the polysaccharide of A. taxiformis followed by C. clavulaum, P. pavonica, S. hypnoides and P. hornemannii as indicative for bioactivity. The FTIR spectroscopic analysis supports the sulfated polysaccharides of S. hypnoides, C. clavulatum and A. taxiformis are similar to agar polymer whereas the spectral characteristics of P. hornemannii have similarities to carrageenan. The higher DPPH activity and reducing power were recorded in the polysaccharide of brown seaweed P. pavonica than the red seaweeds as follows: DPPH activities: S. hypnoides > A. taxiformis > C. clavulatum > P. hornimanii; Reducing power: A. taxiformis > P. hornimanii > S. hypnoides > C. clavulatum. The polysaccharide fractions contain up to 14 kDa from red seaweeds P. hornemannii and S. hypnoides followed by brown seaweed P. pavonica exhibit cytotoxic activity in HeLa cancer cell line (and are similar to structural properties of carrageenan extracted from P. hornemannii). The low molecular weight agar like polymer of S. hypnoides and alginate like brown seaweed P. pavonica showing better in vitro antioxidant activities that are capable of exhibiting cytotoxicity against HeLa cell line can be taken up further in-depth investigation for nutraceutical study.

Abstract:Microalgae have been reported in many studies with high biomass, high carbohydrate content, fast growth rate and multiple bio-activities, while Dictyosphaerium sp. was rarely studied. Polysaccharide prove to be remarkable in many biological activities. To optimize the effects of the ultrasonic-assisted extraction parameters on the yield of polysaccharide from Dictyosphaerium sp., a single-factor experiment and response surface methodology (RSM) with a central composite rotatable design were employed. Four independent variables were investigated: X1 (ratio of raw microalgal powder to water), X2 (extraction temperature), X3 (extraction time), X4 (extraction power). The optimal conditions obtained via single-factor experiment were: X1 (1:25), X2 (50 °C), X3 (50 min), X4 (500 W), while according to the analysis of RSM, the optimum conditions were: X1 (1:26), X2 (58 °C), X3 (55 min), X4 (525 W). The refined polysaccharide were further prepared and characterized using SEM, FT-IR, XRD and TGA/DSC. Five kinds of antioxidant assays were carried out and the results indicated that the refined polysaccharide had strong scavenging ability to DPPH· and OH·. This study provides theoretical and experimental basis for the development and utilization of microalgal resources.

Abstract:Alginates are biopolymers usually obtained from brown seaweed, brown algae (Ochrophyta, Phaeophyceae), and bacteria (Azatobacter vineland and Pseudomonas species) belonging to the family of polycationic copolymers. They are biocompatible, biodegradable, non-antigenic, and non-toxic biopolymer with molecular mass ranges from 32,000-40,000 g/mol in commercial grades. These can be used as edible films or coatings in food industries and also some natural or chemical additives could be incorporated into them to modify their functional, mechanical, nutritional as well as organoleptic properties. Due to their high viscosity and extraordinary shear-thinning effect, they can be used as dietary fibers, thickening, gelling and stabilizing agents. Commercial alginates have vast applications in the fields of biomedical engineering, biotechnology, environmental contaminants treatments, food processing, and pharmaceuticals. Alginates can be used in wound dressings, bone regeneration, neovascularization, protein delivery, cell delivery, theranostic agents, oral drug delivery, controlled release systems, raft formulations, immobilization of biological agents and treatment of environmental contaminants. Various carrier systems can be formulated by the use of alginates like hydrogel, tablets, microcapsules, films, matrices, microspheres, liposomes, nanoparticles, beads, cochleate, floating and supersaturated drug delivery systems. This review presents a broad range of promising applications of alginates, and it can be a great interest to scientists and industries engaged in exploring its hidden potential.

Abstract:Seaweeds of the genera Laminaria, Gelidium, and Porphyra have been used in both food and non-food industries due to their unique properties and characteristic biological activity. This study assesses the antioxidant activity and immunomodulatory properties of the acidic polysaccharides extracted from Laminaria ochroleuca, Porphyra umbilicalis, and Gelidium corneum collected in the Atlantic coast of Tarifa (Cadiz, Spain) The proliferation of murine cell line RAW 264 decreased with increasing concentration of polysaccharides of the three algal species. The highest both antioxidant (25.69 μmol TE g−1 DW) and immunomodulatory activities were observed in the sulfated polysaccharides of L. ochroleuca compared to that of P. umbilicalis and G. corneum. Sulfated polysaccharides of L. ochroleuca presented high potential anticancer activity in cell lines of human colon cancer HTC-116 (IC50 = 0.44 mg mL−1), human malignant melanoma G-361 (IC50 = 5.42 mg mL−1), breast adenocarcinoma human MCF-7 (IC50 = 8.32 mg mL−1), and human leukemia U-937 (IC50 = 3.72 mg mL−1). It is concluded that metabolites of L. ochroleuca can offer significant advantages for the pharmaceutical industry, particularly when macrophage activation is required.

Abstract:Brown macroalgae represent one of the most proliferative groups of living organisms in aquatic environments. Due to their abundance, they often cause problems in aquatic and terrestrial ecosystems, resulting in health problems in humans and the death of various aquatic species. To resolve this, the application of Sargassum has been sought in different research areas, such as food, pharmaceuticals, and cosmetics, since Sargassum is an easy target for study and simple to obtain. In addition, its high content of biocompounds, such as polysaccharides, phenols, and amino acids, among others, has attracted attention. One of the valuable components of brown macroalgae is their polysaccharides, which present interesting bioactivities, such as antiviral, antimicrobial, and antitumoral, among others. There is a wide variety of methods of extraction currently used to obtain these polysaccharides, such as supercritical fluid extraction (SFE), pressurized liquid extraction (PLE), subcritical water extraction (SCWE), ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), and microwave-assisted extraction (MAE). Therefore, this work covers the most current information on the methods of extraction, as well as the purification used to obtain a polysaccharide from Sargassum that is able to be utilized as alginates, fucoidans, and laminarins. In addition, a compilation of bioactivities involving brown algae polysaccharides in in vivo and in vitro studies is also presented, along with challenges in the research and marketing of Sargassum-based products that are commercially available.

Abstract:Alginate (Alg) polymers have received much attention due to the mild conditions required for gel formation and their good bio-acceptability. However, due to limited interactions with cells, many drugs, and biomolecules, chemically modified alginates are of great interest. Sulfated alginate (S-Alg) is a promising heparin-mimetic that continues to be investigated both as a drug molecule and as a component of biomaterials. Herein, the S-Alg literature of the past five years (2017-2023) is reviewed. Several methods used to synthesize S-Alg are described, with a focus on new advances in characterization and stereoselectivity. Material fabrication is another focus and spans bulk materials, particles, scaffolds, coatings, and part of multicomponent biomaterials. The new application of S-Alg as an antitumor agent is highlighted together with studies evaluating safety and biodistribution. The high binding affinity of S-Alg for various drugs and heparin-binding proteins is exploited extensively in biomaterial design to tune the encapsulation and release of these agents and this aspect is covered in detail. Recommondations include publishing key material properties to allow reproducibility, careful selection of appropriate sulfation strategies, the use of cross-linking strategies other than ionic cross-linking for material fabrication, and more detailed toxicity and biodistribution studies to inform future work.
Extraction and characterisation of sodium alginate from the Southern African seaweed Ecklonia maxima

Abstract:Ecklonia maxima is an endemic South African seaweed with numerous commercially valuable compounds, including sodium alginate. This polysaccharide contains 1,4-linked β- d -mannuronic (M) and α- l -guluronic acid (G) residues, and has enormous health and food applications and benefits for biomedicine. In this study, acid extraction of sodium alginate from Ecklonia maxima was performed, and its chemical and physical properties were compared with commercial sodium alginate. A yield of 58.7 % (w/w of dry alga) sodium alginate was recovered from the seaweed. As expected, Fourier-transform infrared spectroscopy and nuclear magnetic resonance spectroscopic analyses of sodium alginate revealed the presence of M and G residues in the polysaccharide. Circular dichroism (CD) spectroscopy revealed an M/G ratio of 1.86 from the extracted sodium alginate. The viscosity average molecular weight of sodium alginate was 429 kDa using the Mark–Houwink–Sakurada equation. Three-dimensional conformational analysis by Congo red and CD spectroscopy suggested that sodium alginate exhibits a triple-helix conformation in solution. Furthermore, X-ray powder diffraction analysis confirmed that the alginate has an amorphous structure. Sodium alginate extracted from E. maxima found on the coasts of Southern Africa exhibits a high mannuronic content and high viscosity, potentially beneficial for drug delivery and biomedical applications.
