Abstract:Identifying potent bacterial algicidal agents is essential for the development of effective, safe, and economically viable algaecides. Challenges in isolating and purifying these substances from complex secretions have impeded progress in this field. Metabolomics profiling, an efficient strategy for identifying metabolites, was pioneered in identifying bacterial algicidal substances in this study. Extracellular secretions from different generations of the algicidal bacterium Brevibacillus sp. were isolated for comprehensive analysis. Specifically, a higher algicidal efficacy was observed in the secretion from Generation 3 (G3) of Brevibacillus sp. compared to Generation 1 (G1). Subsequent metabolomics profiling comparing G3 and 1 revealed 83 significantly up-regulated metabolites, of which 9 were identified as potential algicidal candidates. Back-validation highlighted the potency of 4-acetamidobutanoic acid (4-ABC) and 8-hydroxyquinoline (8-HQL), which exhibited robust algicidal activity with 3d-EC 50 values of 6.40 mg/L and 92.90 μg/L, respectively. These substances disrupted photosynthetic activity in M. aeruginosa by ceasing electron transfer in PSII, like the impact exerted by Brevibacillus sp. secretion. These findings confirmed that 4-ABC and 8-HQL were the main algicidal components derived from Brevibacillus sp.. Thus, this study presents a streamlined strategy for identifying bacterial algicidal substances and unveils two novel and highly active algicidal substances. Environmental Implication Harmful cyanobacterial blooms (HCBs) pose significant environmental problems and health effects to humans and other organisms. The increasing frequency of HCBs has emerged as a pressing global concern. Bacterial-derived algicidal substances are expected to serve as effective, safe, and economically viable algaecides against HCBs. This study presents a streamlined strategy for identifying bacterial algicidal substances and unveils two novel substances (4-ABC and 8-HQL). These two substances demonstrate remarkable algicidal activity and disrupt the photosynthetic system in M. aeruginosa . They hold potential as prospective algaecides for addressing HCBs.

Abstract:Hazardous impacts caused by harmful algal blooms (HABs) have facilitated the development of novel effective approaches for controlling the frequent HABs. Recently, bacteria with algicidal effects, as a promising alternative tool for eliminating the HABs-forming species have received wide concerns. In the present study, a marine bacterial strain 6A1 with potent algicidal activity was successfully isolated from a bloom of Akashiwo sanguinea , and the characteristics related to its algicidal action were systematically investigated. Sequencing results of 16S rDNA suggested that this bacterium belongs to the genus Arenibacter in the family Flavobacteriaceae, while the phylogenetic analysis revealed that Arenibacter sp. strain 6A1 was the only algicidal bacterium in Arenibacter to date, according to the best of our knowledge. Arenibacter sp. strain 6A1 had an extraordinarily high algicidal performance when it was added to A. sanguinea culture with a volume fraction of 1.5% for 2 h. The algicidal performance of strain 6A1 was concentration-dependent and time-dependent. The best algicidal efficacy of strain 6A1 was seen during the stationary phase, while the resistance of A. sanguinea to strain 6A1 declined as the growth cycle progressed. Subsequently, the indirect algicidal mode of Arenibacter sp. strain 6A1 was verified and the algicidal compounds produced by strain 6A1 were proved to be heat-resistant. Further research showed that strain 6A1 caused a severe damage in photosynthesis, oxidative stress, and also damage in three HABs-forming algae to varying degrees. The greatest algicidal performance was found on unarmored dinoflagellate A. sanguinea , followed by raphidophyte Chattonella antiqua and armored dinoflagellate Alexandrium tamarense . The Arenibacter sp. strain 6A1 exhibited selective algicidal activity on a wide range of algal species, primarily targeting Pyrrophyta and Ochrophyta. The proposed bacteria (strain 6A1) can be a powerful and eco-friendly tool for emergency treatment of HABs caused by certain algal species in coastal water bodies.

Isolation of an algicide from a marine bacterium and its effects against the toxic dinoflagellate Alexandrium catenella and other harmful algal bloom species

Abstract:The aim of this study was to isolate and identify bacteria demonstrating an algicidal effect against Alexandrium catenella and to determine the activity and range of any algicide discovered. The morphological and biochemical attributes of an algicidal bacterium, isolate YS-3, and analysis of its 16S rRNA gene sequence revealed it to be a member of the genus Brachybacterium. This organism, designated Brachybacterium sp. YS-3, showed the greatest effect against A. catenella cells of all bacteria isolated, and is assumed to produce secondary metabolites. When 10% solutions of culture filtrates from this strain were applied to A. catenella cultures, over 90% of cells were killed within 9 h. Bioassay-guided isolation of the algicide involved led to the purification and identification of an active compound. Based on physicochemical and spectroscopic data, including nuclear magnetic resonance and mass analyses, this compound was identified as 1-acetyl-β-carboline. This algicide showed significant activity against A. catenella and a wide range of harmful algal bloom (HAB)-forming species. Taken together, our results suggest that Brachybacterium sp. YS-3 and its algicide represent promising candidates for use in HAB control.

Abstract:Abstract A study was conducted to elucidate the mechanism of donor‐pecific Mixed Lymphocyte Reaction (MLR and Cell Mediated Lymphotoxicity (CML) unresponsiveness in a renal transplant recipient with a long‐term well‐functioning kidney. The peripheral blood lymphocytes (PBL) of the recipient, who had not shown rejection since his transplantation 5 years previously, and those of his mother (donor), his father and two healthy third parties were examined. MLR, CML, semimicro MLR in a double chamber, interleukin‐2 (IL‐2) synthesis assay and limiting dilution assay were performed. This recipient showed donor‐pecific MLR and CML unresponsiveness. IL‐2 assay showed that the PBL of the recipient produced less IL‐2 against the donor than against the father and the third parties. The addition of exogenous recombinant IL‐2 (rIL‐2; Takeda Co.) to the priming MLR caused a recovery of CML against the donor. A limiting dilution assay indicated that cytotoxic T cell precursor (CTLp) frequencies against the donor and father did not differ. The suppressor assay in a double chamber indicated that the PBL of the recipient stimulated by the donor PBL had a non‐pecific suppressive effect on MLR, CML and IL‐2 synthesis of the PBL across the Major Histocompatibility Complex (MHC) barrier. This suppressive effect was abolished by OKT3 or OKT8 monoclonal antibody and complement. Thus, the recipient had donor‐pecific suppressor T cells that produced a humoral non‐pecific suppressive factor only when stimulated by the donor PBL, and this factor suppressed PLR and CML by inhibiting IL‐2 synthesis of the PBL.
Algicidal interaction between Paenibacillus polymyxa MEZ6 and microalgae

Abstract:Aims: Algicidal bacteria can be used for control of harmful algal bloom and extraction of algal bioproducts based on their algae-lysing activities. This work investigated the algae-lysing activity of a newly isolated algicidal bacterium, Paenibacillus polymyxa strain MEZ6 and its possible mechanisms. Methods and results: Algicidal bacteria were isolated from soil samples collected at the university campus. Co-inoculation tests identified that one isolate, MEZ6, can rapidly kill eukaryotic algae including Chlamydomonas reinhardtii, Tribonema minus, Haematococcus pluvialis, and Chlorella ellipsoidea. The strain was determined as Paenibacillus polymyxa MEZ6 based on 16S rRNA gene sequence and genome comparisons. The algicidal activity was detected in both living cells and cell-free supernatant of spent culture medium, suggesting cell-cell contact is not required for algicidal activity. Strain MEZ6 was less active towards cyanobacterial strains compared to algae. Genomic sequence and comparative proteomic analyses were performed to explore the possible algicidal mechanisms of the strain. Differentially expressed protein analysis identified a number of proteins related to polysaccharides degradation and antimicrobial secondary metabolite biosynthesis that may be involved in the algicidal activity of MEZ6. Conclusion: Paenibacillus polymyxa MEZ6 is a newly discovered gram-positive algicidal bacterial strain with high lytic activity towards several algal species. Significance and impact of the study: Our study extends the understanding of the versatile characters of Paenibacillus polymyxa and sheds new insights into its application in algae biotechnology.

Abstract:One strain of algicidal bacterium which can inhibit Harmful algal blooms (HABs), FDT5, was isolated from activated sludge and found to have good algicidal effects on Microcystis aeruginosa. It was revealed that: The FDT5 was a Gram-negative bacterium and identified as Ochrobactrum sp.; The greater the initial bacterial cell density, the faster the degradation of chlorophyll a.; The algicidal efficiency was evaluated at the most favorable conditions which were a temperature of 30-35 degrees C, a pH of 7.6 and complete darkness; The FDT5 strain lysed Microcystis aeruginosa not directly but by secreting metabolites which could withstand high temperatures and pressure.
Revealing the algicidal characteristics of Maribacter dokdonensis: An investigation into bacterial strain P4 isolated from Karenia mikimotoi bloom water

Abstract:Harmful algal blooms (HABs) are a global environmental concern, causing significant economic losses in fisheries and posing risks to human health. Algicidal bacteria have been suggested as a potential solution to control HABs, but their algicidal efficacy is influenced by various factors. This study aimed to characterize a novel algicidal bacterium, Maribacter dokdonensis (P4), isolated from a Karenia mikimotoi (Hong Kong strain, KMHK) HAB and assess the impact of P4 and KMHK's doses, growth phase, and algicidal mode and the axenicity of KMHK on P4's algicidal effect. Our results demonstrated that the algicidal effect of P4 was dose‐dependent, with the highest efficacy at a dose of 25% v/v. The study also determined that P4's algicidal effect was indirect, with the P4 culture and the supernatant, but not the bacterial cells, showing significant effects. The algicidal efficacy was higher when both P4 and KMHK were in the stationary phase. Furthermore, the P4 culture at the log phase could effectively kill KMHK cells at the stationary phase, with higher algicidal efficacy in the bacterial culture than that of the supernatant alone. Interestingly, P4's algicidal efficacy was significantly higher when co‐culturing with xenic KMHK (~90% efficacy at day 1) than that with the axenic KMHK (~50% efficacy at day 1), suggesting the presence of other bacteria could regulate P4's algicidal effect. The bacterial strain P4 also exhibited remarkable algicidal efficacy on four other dinoflagellate species, particularly the armored species. These results provide valuable insights into the algicidal effect of M. dokdonensis on K. mikimotoi and on their interactions.

Abstract:Alexandrium tamarense is a toxic harmful algal blooms (HABs) causing species, which poses great threat to human health and marine economy. In this study, we isolated an algicidal bacterium Altererythrobacter sp. LY02 towards to A. tamarense and later investigated the algicidal activity, algicidal mode, characteristics of algicidal active substance and algicidal procedure. The results indicated that the cell-free filtrate of strain LY02 showed high algicidal effect on algal growth, however, bacterial cells almost lost algicidal activity. The algicidal active substance was temperature- and pH-stability, and its molecular weight was less than 1000 Da, and was a non-proteinaceous material or non-polysaccharide, mid-polar substance. Under the algicidal effect of active substance, the morphology and structure of A. tamarense cells were seriously damaged as well as organelles. Our study confirmed that the algicidal active substance could be used as an excellent bio-agent for controlling HABs caused by A. tamarense.
Glucose Triggers the Cytotoxicity of Citrobacter Sp R1 Against Microcystis Aeruginosa

Abstract:Algicidal bacteria offer a promising option for killing Microcystis aeruginosa, one notorious cyanobacteria causing harmful algal blooms. In this study, Citrobacter sp. R1 presented high algicidal activity (81.6±2.2%, 72h) against M. aeruginosa when cultured using glucose, while it showed no algicidal activity (0±3.4%) when cultured using wheat bran, suggesting that appropriate carbon source is crucial for algicidal bacteria in killing M. aeruginosa. The underlying algicidal mechanism of strain R1 was explored by studying the effect of different carbon sources (glucose and wheat bran) on its key algicidal gene expression and total protein translation. While the glycogen synthase gene (glgA), cloned from strain R1 via transposon mutagenesis, was for the first time related to algicidal activity, its transcriptional level was not positively correlated with the algicidal activity of strain R1. We found that, the translation of total protein of strain R1 was relatively less when cultured with glucose, compared to growth with wheat bran. This indicated that the functional algicidal gene of strain R1 exerts its algicidal activity at protein translational level. These findings not only reveal the importance of appropriate carbon source for strain R1 for controlling M. aeruginosa, but also bring insights into its underlying algicidal mechanism.
Efficient inactivation of harmful algae K. mikimotoi by a novel algicidal bacterium via a rare direct contact pathway: Performances and mechanisms

Abstract:Harmful algal blooms (HABs) caused by Karenia mikimotoi have posed great threats to marine ecosystems, and algal inactivation by symbiotic bacteria has been recognized as environmental benign methods for controlling HABs. However, the identified algicidal bacteria for K. mikimotoi is limited and exclusively based on indirect algicidal pathways, which may cause secondary pollution due to releasing toxic algicidal agents. In this study, a novel strain of algicidal bacteria Tenacibaculum sp. GD3 was isolated from the phycosphere of K. mikimotoi. The bacterial strain GD3 could achieve 92.6 % of inactivation efficiency against K. mikimotoi within 8 h of co-culturing period, which outperformed those in existing literatures reported so far. The algicidal mechanisms were revealed to be a rare direct cell-to-cell contact pathway, and the GD3 could grow by utilizing metabolites from K. mikimotoi , exhibiting excellent bacterial adaptability in the phycosphere. Cell morphology changes were monitored by live cell imaging system combined with SEM and TEM observations, which showed that the GD3 was first attached to the algal cell membrane, followed by lipid peroxidation and lysis of membrane protein. Oxidative stress responses were induced as reveled by up-regulation of intracellular ROSs and antioxidant enzyme activity. Photosynthetic parameters including rETR max , F v /F m , YII and NPQ were reduced, and expression of functional genes involved in decomposition of chlorophyll and cell wall was significantly suppressed. Moreover, the intracellular release profile and acute toxicity assessment indicated that the GD3 could also detoxify the K. mikimotoi cultures and the released biomolecules would not cause adverse effect to marine environment. This study not only provides a novel algicidal bacterium against K. mikimotoi via a rare direct mode, but also helps to better understand the algicidal mechanisms at physiological and genetic level, thus moving forward the areas of HABs control by microbiological strategies.

Isolation and Algae-Lysing Characteristics of the Algicidal Bacterium B5.

Abstract:Water blooms have become a worldwide environmental problem. Recently, algicidal bacteria have attracted wide attention as possible agents for inhibiting algal water blooms. In this study, one strain of algicidal bacterium B5 was isolated from activated sludge. On the basis of analysis of its physiological characteristics and 16S rDNA gene sequence, it was identified as Bacillus fusiformis. Its algaelysing characteristics on Microcystis aeruginosa, Chlorella and Scenedesmus were tested. The results showed that: (1) the algicidal bacterium B5 is a Gram-negative bacterium. The 16S rDNA nucleotide sequence homology of strain B5 with 2 strains of B. fusiformis reached 99.86%, so B5 was identified as B. fusiformis; (2) the algal-lysing effects of the algicidal bacterium B5 on M. aeruginosa, Chlorella and Scenedesmus were pronounced. The initial bacterial and algal cell densities strongly influence the removal rates of chlorophyll-a. The greater the initial bacterial cell density, the faster the degradation of chlorophyll-a. The greater the initial algal cell density, the slower the degradation of chlorophyll-a. When the bacterial cell density was 3.6 fi 107 cells/ml, nearly 90% of chlorophyll-a was removed. When the chlorophyll-a concentration was less than 550 μg/L, about 70% was removed; (3) the strain B5 lysed algae by secreting metabolites and these metabolites could bear heat treatment.
Algicidal activity of a novel indigenous bacterial strain of Paracoccus homiensis against the harmful algal bloom species, Karenia mikimotoi

Abstract:Harmful algal blooms have deleterious effects on aquatic ecosystems and human health. The application of algicidal bacteria is a promising and environmentally friendly method of preventing and eradicating harmful algal blooms. In this study, a screen for algicidal agents against harmful algal blooms was used to identify an algicidal bacterial strain (strain O-4) isolated from a Karenia mikimotoi culture. Strain O-4 exhibited a strong inhibitory effect on harmful K. mikimotoi and was identified as Paracoccus homiensis via 16S rRNA gene sequence analysis. This strain killed K. mikimotoi by secreting active algicidal compounds, which were stable at temperatures of −80–121 °C but were sensitive to strongly acidic conditions (pH = 2). The algicidal properties of strain O-4 against K. mikimotoi were cell density- and time-dependent. No significant changes or negative effects were noted for two other Chlorophyta species, which highlighted the specificity of the studied algicidal substance. Finally, single-factor experiments revealed the optimum growth conditions of strain O-4 under different pH and temperature conditions. Therefore, strain O-4 has the potential to be used as a bio-agent for reducing the biomass of harmful K. mikimotoi blooms.

Abstract:The dinoflagellate Noctiluca scintillans is a harmful algal species that is globally distributed and poses a certain threat to marine ecosystems. Recent research has shown that the application of algicidal bacteria is a promising method to prevent and control such harmful algal blooms (HABs), given its advantages of safety and efficiency. In this study, a strain of algicidal bacterium LD-B6 with high efficiency against N. scintillans was isolated from the coastal waters of Lianyungang, China. 16S rDNA sequence analysis showed that the strain LD-B6 belongs to the genus Pseudoalteromonas . Furthermore, the algicidal effect of LD-B6 on N. scintillans was investigated. The results showed that strain LD-B6 exerted strong algicidal activity against N. scintillans . After 12 h of bacterial culture addition to algal cultures at a 2% final volume rate, the algicidal activity reached 90.5%, and the algicidal activity of LD-B6 was influenced by the density of N. scintillans . In addition, the algicidal bacterium LD-B6 was found to indirectly lyse algal cells by secreting extracellular compounds. These algicidal compounds were stable, indicating that they are not proteins. Importantly, strain LD-B6 was broadly general, showing varying degrees of lysing effects against five of the six algal species tested. On the basis of the described studies above, the algicidal powder was also initially developed. In summary, the isolated bacterial strain LD-B6 shows the potent algicidal capability to serve as a candidate algicidal bacterium against N. scintillans blooms.

Abstract:Raphidiopsis raciborskii ( R. raciborskii ) forms harmful cyanobacterial blooms globally, and poses a great threat to the safety of drinking water and public health. There is a great need to develop eco-friendly biological alternative measures to mitigate mass blooms of R. raciborskii . However, previous rare studies on algicidal microorganisms against R. raciborskii restricted this aim. Recently, an algicidal bacterium Streptomyces sp. HY (designated HY) was identified with flavones producing ability, and could remove up to 98.73 % of R. raciborskii biomass within 48 h by directly attacking the cyanobacterium and release of algicidal substances (i.e., flavonoids) with a inoculum ratio of 5 %. Algicidal rate of HY was enhanced by 88.05 %, 89.33 % under dark and light, and full-light conditions respectively, when compared with the dark condition. Its algicidal substances were stable in a broad range of temperature (−80–55 °C) and pH (3−11) conditions, and all treated groups exhibited ≈ 100 % algicidal rate at day 3. HY treatment disrupted the photosynthesis system and triggered serious oxidative stress resulting in severe morphological injury. Thereby, HY treatment significantly affected expression levels of several essential genes (i.e., psbA , psaB , rbcL , ftsZ , recA , grpE ), and simultaneously inhibited the biosynthesis and release of cylindrospermopsin. Yet, HY treatment didn't show any toxicity to zebrafish test embryos. Such results indicate that HY is a promising algicidal candidate strain to control global R. raciborskii blooms, and holds great promises for an effective biological measure to sustain water safety.

Optimized Culturing Conditions for an Algicidal Bacterium Pseudoalteromonas Sp. SP48 on Harmful Algal Blooms Caused by Alexandrium Tamarense

Abstract:AbstractBacteria play an important role in preventing algal blooms and reducing their harm to the environment. To improve the algicidal activity of Pseudoalteromonas SP48 which had an inhibition effect on dinoflagellate Alexandrium tamarense, its growth medium and fermentation conditions were optimized for this bacterium. In this study, we used two steps to establish the optimum conditions. First, the proper proportion of medium was selected based on an orthogonal design. Then, the fermentation conditions were further optimized through uniform design in an enlarged 5L bioreactor. To test the algicidal ability of Pseudoalteromonas SP48 under the optimum conditions, algal cell morphology was observed by transmission electron microscopy (TEM). After the orthogonal design, we found that the optimum medium was [0.7% (m/v) tryptone, 0.2% (m/v) soluble starch, 0.2% (m/v) sucrose, 0.1% (m/v) FeSO4, and 1.2% (m/v) K2HPO4] for Pseudoalteromonas SP48 growth. Based on these results, optimum fermentation conditions were further explored in a 5L fermentation cylinder using a uniform design; the influence of variables such as incubation time, carbon type, and rotation speed were tested. The optimal fermentation conditions were fermentation time (42 hr), tryptone (1.1%), seeding volume (1.4 × 1013 cells), and rotation speed (250 r/min). Under these established optimum conditions, the biomass of strain SP48 increased by 79.2% and its lethal dose 50% (LD50) decreased by 54.0%, respectively. The TEM results showed that compared with the control group, the cell wall and cell membrane of A. tamarense were significantly damaged, and the structure and shape of the organelles were destroyed by algicidal bacteria of Pseudoalteromonas SP48. Overall, our results demonstrate that the optimized culture conditions could significantly enhance the algicidal activity of Pseudoalteromonas SP48 against a harmful dinoflagellate, such as A. tamarense. It will effectively provide a scientific foundation for both production of algicidal substances and HABs control.

Abstract:Introduction: Harmful algal blooms (HABs) have been increasing in frequency and expanding their ranges on coastlines worldwide in recent decades. Algicidal bacteria play a pivotal role in eliminating HABs, yet the characteristics of bacterial communities and their algicidal activity during a Karenia longicanalis bloom remain poorly understood. Methods: In this study, we investigated bacterial communities using 16S rRNA sequencing during a K. longicanalis bloom to identify bacteria with high algicidal activity that could be isolated. Five sampling sites in Tongxin Bay, located in Lianjiang County, China, including TX1 to TX5, were selected based on the concentration of K. longicanalis cells. Results: Our 16S rRNA sequencing results revealed that the TX4 site was enriched with genera known to contain algicidal bacteria, such as Pseudoalteromonas and Alteromonas , which are members of the Gammaproteobacteria class, while Sulfitobacter , a member of the Alphaproteobacteria class, was enriched in the TX5 site. Among the 100 cultivable bacteria isolated from the 5 sampling sites, 6 exhibited an algicidal rate of over 80%, with FDHY-MQ5, isolated from the TX4 site, exhibiting an algicidal rate of approximately 100% against Karenia mikimotoi after 48 hours of challenge with 2% (v/v) bacterial volume (OD600=4.5) concentration. Our 16S rRNA sequencing result showed FDHY-MQ5 was a member of the Pseudoalteromonas genus, and this bacterium also demonstrated high algicidal activity against Heterosigma akashiwo and Alexandrium tamarense . Discussion: Our findings shed light on the changes in bacterial community structure and the algicidal behavior of bacteria towards algae during a K. longicanalis bloom, providing a research basis for a better understanding of HAB management.

Abstract:Algae blooms present an environmental problem worldwide. In response to the outbreak of harmful algal blooms in cyanobacteria, the role of biological control has drawn wide attention, particularly for algicidal bacteria. The mechanism underlying algicidal activity was determined in our study. Algae-lysing bacteria used were separated from water and sediment collected from the Fenhe scenic spot of Taiyuan. Genetic and molecular identification was conducted by polymerase chain reaction amplification based on 16S rDNA gene. These bacterial strains were identified as Raoultella planticola and Aeromonas sp. The algae-lysing characteristics were evaluated on Microcystis aeruginosa. For the two algicidal bacteria, the high inoculation ratio (>8%) of bacteria strains contributed to the lytic effect. M. aeruginosa could be completely removed by these strains at different cell ages. However, the time used decreased with an increase in cell age. The removal rate was increased while M. aeruginosa was in the lag and logarithmic phases. The earlier bacteria strains could be inoculated, the sooner all algae could be removed. Both algicidal substances were protein, which could destroy the photosynthetic systems and break the cell of M. aeruginosa. The algicidal bacteria strain has important theoretical and practical significance for economic and feasible algae removal and provides good germplasm resources and technical support for the control of cyanobacterial bloom.

Abstract:Chaetomorpha valida , filamentous green tide algae, poses a significant threat to both aquatic ecosystems and aquaculture. Vibrio alginolyticus Y20 is a new algicidal bacterium with an algicidal effect on C. valida . This study aimed to investigate the physiological and molecular responses of C. valida to exposure to V. alginolyticus Y20. The inhibitory effect of V. alginolyticus Y20 on C. valida was content dependent, with the lowest inhibitory content being 3 × 10 5 CFU mL −1 . The microscopic results revealed that C. valida experienced severe morphological damage under the influence of V. alginolyticus Y20, with a dispersion of intracellular pigments. V. alginolyticus Y20 caused the decrease in chlorophyll-a content and Fv / Fm in C. valida . At the molecular level, V. alginolyticus Y20 downregulated the expression of genes related to photosynthetic pigment synthesis, light capture, and electron transport. Furthermore, V. alginolyticus Y20 induced oxidative damage to algal cells. The production of reactive oxygen species significantly increased after 11 days of exposure. Malondialdehyde content significantly increased, and the cell membranes were severely damaged by lipid peroxidation. The content of superoxide dismutase and peroxidase also markedly increased, whereas catalase content decreased significantly. Additionally, peroxisomes were inhibited due to the downregulation of PEX expression, leading to irreversible oxidative damage to algal cells. Our findings provided a new theoretical basis for exploring the interaction between algicidal bacteria and green tide algae at the molecular level.

Abstract:Summary In recent decades,harmful algal blooms(HABs),which are natural phenomena that occur across all the world, have posed threats to decrease dissolved oxygen and have had a negative effect on fisheries, aquaculture,drinking water,tourism and human health.It is increasingly urgent to develop useful strategies for predicting and reducing the negative impacts of HABs.Actually,many methods have been devoted to controlling HABs,including copper sulfate,hydrogen peroxide,ozonization,ultrasonication,collection of algae from water surface using nets,iron powder and magnets,centrifugal separation,or ultraviolet radiation.As an effective and environment friendly strategy to control harmful algal bloom outbreaks,biologic control methods such as using algicidal bacteria against M.aeruginosa have been more and more attractive.It has been demonstrated that algicidal bacteria could serve as potential ways in reducing the impacts of HABs. In this study,a Citrobacter sp.N10 with algicidal activity against the toxic algae M.aeruginosa was isolated and identified from a lake of Zhejiang University using liquid infection technology and 1 6S rDNA sequence analysis.The growth and algicidal characters of Citrobacter sp.N10 were then studied in order to offer some useful information for HABs controlling. The results showed that the bacterium was gram-negative and its colonies were smooth, convex, low moisture,translucent or opaque and gray,and its colony surfaces were glossy.Sequence analysis of 1 6S rDNA demonstrated that strain N10 belonged to Citrobacter sp.When inoculation time was 2 to 12 h,N10 grew in logarithmic-growth period,and when inoculation time was beyond 12 h,N10 was in stationary phase.Strain N10 showed a high lysis of M.aeruginosa with an algicidal rate of 86.55% within 24 h.The algicidal rate of N10 increased with increasing incubation time and reached 97.08% within 72 h.pH value had the most significant effect on the growth of strain N10,followed by NaCl concentration and culture temperature,while these three conditions had different effects on algicidal activities of strain N10 against M.aeruginosa .A kind of substance against M.aeruginosa was secreted by strain N10,and the substance was sensitive to high temperature(121 ℃) but resistant to protease K.Relatively speaking,the thalli of strain N10 had hardly algicidal effect against M. aeruginosa .Transmission electron microscope was subsequently used to investigate algal cells??structures after 3 d with treatment of strain N10 cell-free culture filtrate(CCF).The results indicated that the cell membranes and cell walls of M.aeruginosa were broken,while the gas vesicles and some particulate matters such as the cytoplasm phosphate particles and blue bodies disappeared.In addition,the cellular ultrastructure of the photosynthetic lamellae was also found to be disordered and disappeared. In conclusion,the research on the characteristics of N10 against M.aeruginosa can provide a tool in controlling the bloom of M.aeruginosa .The findings are of theoretical significance and potential in reducing the impacts of HABs.

Abstract:We describe the isolation of Bacillus subtilis strain JA and demonstrate that this bacterium exhibited strong algicidal effects on the algae Alexandrium minutum with an inhibition rate exceeding 80 % within 48 h. B. subtilis JA significantly reduced the photosynthetic efficiency of A. minutum and caused extensive morphological damage to the algae. Genomic analysis of B. subtilis JA demonstrated that a putative AI-2 type quorum sensing (QS) gene (LuxS) is present in its genome cluster, which is regulate pheromone biosynthesis. Interestingly, the exogenous addition of a QS-oligopeptide (ComX-pheromone) improved the algicidal efficiency of B. subtilis JA, thus indicating that the algicidal activity of this bacterium is potentially regulated by QS. Collectively, our data describe a potential antialgal bacterium and speculated that its behavior can be modulated by QS signal. B. subtilis JA may therefore represent a valuable tool for the development of novel chemical-ecological methods with which to control harmful algae.
