Abstract:Silybum marianum L. is a therapeutic plant belonging to the family Asteraceae, which has exhibited silymarin, a principal component used to cure various physiochemical disorders. The study appraised the phytochemical analysis, antioxidant activity and chemical analysis of an extract from the seed, stem and leaves. Qualitative and quantitative phytochemical analysis was evaluated by the Folin–Ciocalteu reagent method and aluminum chloride colorimetric method, respectively. While the antioxidant activity was determined by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and acetate buffer in ferric chloride (FRAP) assay, respectively, the chemical profile was evaluated by Gas Chromatography-Mass Spectrometry (GC-MS) assay. The study outcomes identified that alkaloids, glycosides, flavonoids, terpenoids, steroids and catcholic tannins were present in seed, stem and leaves extracts except for saponins and Gallic tannins. Whereas, phenols were absent only in seed extract. Quantitative analysis revealed the presence of phenols and flavonoids in appreciable amounts of 21.79 (GAE/g), 129.66 (QE/g) and 17.29 (GAE/g), 114.29 (QE/g) from the leaves and stem extract, respectively. Similarly, all extracts expressed reasonable DPPH inhibition (IC50) and FRAP reducing power such as 75.98, 72.39 and 63.21% and 46.60, 51.40 and 41.30 mmol/g from the seeds, stem and leaves extract, respectively. Additionally, chemical analysis revealed the existence of 6, 8 and 9 chemical compounds from the seeds, stem and leaves extract, respectively, corresponding to 99.95, 99.96 and 98.89% of the whole extract. The chemical compound, Dibutyl phthalate was reported from all extracts while, Hexadecanoic acid, methyl ester and Silane, (1,1-dimethylethyl), dimethyl (phenylmethoxy) were reported only from the seed and leaves extract. Moreover, Methyl stearate was also a major compound reported from all extracts except for seed extract. It is demonstrable that extracts from different parts of S. marianum possess significant antioxidant activity, as well as valuable chemical compounds accountable for therapeutic effects that might be incorporated as an alternative to synthetic chemical agents.

Abstract:Salinity is an important challenge around the world, effecting all physiological and biochemical processes of plants. It seems that seed priming can diminish the negative impacts of salinity. To study the effects of hydro-priming and inoculation with Piriformospora indica (Pi) and Azotobacter chroococcum (Az) on physio-biochemical traits, flavonolignans and fatty acids composition of milk thistle under saline conditions, a greenhouse experiment was carried out. Our results indicated that under salinity, seed priming, especially Pi, improved physio-biochemical properties in milk thistle. Under 120 mM NaCl, inoculation with Pi increased membrane stability index (MSI) and relative water content (RWC) (by 21.86 and 33.43%, respectively). However, peroxidase (POX) (5.57- and 5.68-fold in roots and leaves, respectively), superoxide dismutase (SOD) (4.74- and 4.44-fold in roots and leaves, respectively), catalase (CAT) (6.90- and 8.50-fold in roots and leaves, respectively) and ascorbate peroxidase (APX) (5.61- and 5.68-fold in roots and leaves, respectively) activities increased with increasing salinity. Contrary to salinity, hydro-priming with Az and Pi positively altered all these traits. The highest content of the osmolytes, adenosine triphosphate (ATP) content and rubisco activity were recorded in Pi treatments under 120 mM NaCl. Stearic acid (20.24%), oleic acid (21.06%) and palmitic acid (10.48%) increased, but oil content (3.81%), linolenic and linoleic acid content (22.21 and 15.07%, respectively) decreased under saline conditions. Inoculations of Pi positively altered all these traits. The present study indicated that seed priming with Pi under 120 mM NaCl resulted in maximum silychristin, taxidolin, silydianin, isosilybin, silybin and silymarin of milk thistle seeds.
Effect of Field Trial on Silymarin Complex Composition and Antioxidant Assessment of Milk Thistle (Silybum marianum L. Gaertner)

Abstract:Plant foods are receiving increasing attention as a valuable source of health beneficial compounds. Understanding the impact of growing conditions on the quality of milk thistle is critical for determining appropriate agro-ecological and agro-economic parameters for its production and, subsequently, food products rich in health-beneficial compounds. For this purpose, a randomized milk thistle cultivation trial was established in the experimental field of Agritec Plant Research Plc. in Šumperk, Czech Republic, and carried out for two subsequent growing seasons in 2020 and 2021. The milk thistle achenes, variety Mirel, were sown in the row width of 12.5, 25 and 37 cm; and the qualitative parameters of each field trial such as achenes yields, silymarin complex determination and also antioxidant assessment (total phenolic content, total flavonoids content, DPPH and ABTS radical scavenging activity) were evaluated. The environmental impact of the extraction process was reduced by using pressurized liquid extraction with 60% EtOH (v/v). The weather conditions during the trial as well as the row spacing of milk thistle sowing were revealed to have a significant influence on the evaluated parameters (p ≤ 0.05). The highest yields of evaluated parameters were obtained for the growing season 2021 and the row spacing of 37 cm.

Abstract:Silymarin is a bioflavonoid complex extract derived from dry seeds of Milk thistle [(Silybum marianum(L.) Gaemrnt. (Fam. Asteraceae/Compositaceae)] whose hepatoprotective effect has clinically been proved. Low toxicity, favorable pharmacokinetics, powerful antioxidant, detoxifying, preventive, protective and regenerative effects and side effects similar to placebo make silymarin extremely attractive and safe for therapeutic use. The medicinal properties of silymarin and its main component silibinin have been studied in the treatment of Alzheimer's disease, Parkinson's disease, sepsis, burns, osteoporosis, diabetes, cholestasis and hypercholesterolemia. Owing to its apoptotic effect, without cytotoxic effects, silymarin possesses potential applications in the treatment of various cancers. Silymarin is being examined as a neuro-, nephro- and cardio-protective in the damage of different etiologies due to its strong antioxidant potentials. Furthermore, it has fetoprotective (against the influence of alcohol) and prolactin effects and is safe to be used during pregnancy and lactation. Finally, the cosmetics industry is examining the antioxidant and UV-protective effects of silymarin. Further clinical studies and scientific evidence that silymarin and silibinin are effective in the therapy of various pathologies are indispensable in order to confirm their different flavonolignan pharmacological effects.
Phytotherapeutic properties of milk thistle seeds: An overview

Abstract:Milk thistle ( Silybum marianum , Asteraceae) seeds have been used for over 2000 years as remedy for several diseases especially for liver and still widely used. The active constituents of milk thistle seed are three flavonolignans viz. silibinin, silychristin, and silidianin collectively known as silymarin extracted from milk thistle seeds, available commercially as standardized extract. Milk thistle seed extract (silymarin) and its constituents (mainly silibinin) act as antioxidant and hepatoprotective; effective in treating toxin poisoning, hepatitis, cirrhosis, fibrosis of liver; stimulate liver regeneration. However, human studies regarding management of alcoholic cirrhosis and hepatitis are equivocal. Milk thistle seed has anti-inflammatory, immunomodulatory, lipid and biliary effects. It also has antiviral, antitumor and other therapeutic properties. Milk thistle preparations are safe, well tolerated and cause no serious side effects except mild gastrointestinal and allergic reactions. Milk thistle seed is a very promising herbal drug. More research is warranted to substantiate its broad ranging phytotherapeutic effects.

Abstract:The oldest remedies identified to mankind are herbal medicines. India is recognized worldwide for its Ayurvedic treatment. India has rich history of using many plants for medicinal purposes. Remedial plants are cooperating extremely dynamic position in customary drugs for the action of a variety of illness. However a key obstacle, which has hindered the promotion in use of alternative medicines in the developed countries, is no evidence of documentation and absence of stringent quality control measures. There is a demand for the evidence of every investigate effort execute on conventional remedies in the appearance of certification. The purpose of current review is to make accessible up-to-date information on, botany, morphology, ecological biodiversity, therapeutic uses, phytochemistry and pharmacological activities on diverse parts of Silybum marianum (L.) Gaertn (S. marianum). This review was assembled using technical literature from electronic search engine such as Springer link, Bio Med Central, Pub Med, Scopus, Science Direct, Scielo, Medline and Science domain. Supplementary texts were obtained from books, book chapters, dissertations, websites and other scientific publications. S. marianum a member of the Asteraceae family, is a tall herb with large prickly white veined green leaves and a reddish-purple flower that ends in sharp spines. It is native of the Mediterranean region and which has also spread in East Asia, Europe, Australia and America. Confident chemical constituents were exposed cognate as silybin A, silybin B, isosilybin A, isosilybin B, silychristin, silydianin, apigenin 7-O-β-(2′′- O-α-rhamnosyl)galacturonide, kaempferol 3-O-α-rhamnoside-7-O-β-galacturonide, apigenin 7-O-β-glucuronide, apigenin 7-O-β-glucoside, apigenin 7-O-β-galactoside, kaempferol-3-O-α-rhamnoside, kaempferol, taxifolin and quercetin. The plant is exclusively used as anti-diabetic, hepatoprotective, hypocholesterolaemic, anti-hypertensive, anti-inflammatory, anti-cancer, and as an anti-oxidant. Seeds of the plant are also used as an anti-spasmodic, neuroprotective, anti-viral, immunomodulant, cardioprotective, demulcent and anti-haemorrhagic. The plant is also serves as a galactagogue, agent that induces milk secretion and used in the treatment of uterine disorders. The plant is employed in dissimilar conventional schemes of remedy in the cure of different illness.

Abstract:Advancements in the field of research and development have paved toward the use of herbal agents to cure and control health maladies. Milk thistle ( Silybum marianum L .) is one of the herbs that possess pharmacological and therapeutic attributes. Various bioactive components in milk thistle have a key role in pathological conditions like blood-related disorders, hepatic ailments, arthritis, ulcerative colitis, and carcinomas. Among various bioactive components, silymarin is an active agent which isa standardized mixture of flavonolignans (70%–80%) along with silybin, silydianin, and silychristin, with silybin being the main active chemical component. Silymarin has been used medicinally to treat liver disorders, including acute and chronic viral hepatitis, toxin/drug-induced hepatitis, cirrhosis, and alcoholic liver diseases. Traditionally, the plant was employed to increase human reproduction ability, possess reactive oxygen species (ROS) scavenging, antioxidant, anti-inflammatory and immunomodulatory properties, has UV-induced skin damage protection, maintains the menstrual cycle, lessen viral infection, decrease fibrosis, and blood glucose as well as improve functions of the liver, pancreas, and reproductive cells. This review summarizes the basic as well as advanced therapeutic effects of silymarin which will ultimately lead researchers in developinphytopharmaceutical-based complementary medicine.

Abstract:Abstract Cadmium (Cd +2 ) is a potentially toxic element that inhibits growth and development of several species of plants along with Silybum marianum (L.) Gaertn which is an essential medicinal plant belonging to family Asteraceae. The exogenous application with 0.25μM Salicylic acid (SA) and 10μM hydrogen peroxide (H 2 O 2 ) to ameliorate harmful effects of cadmium (500 μM) on milk thistle were studied that were grown at two different ecological zones of Balochistan province of Pakistan i.e. Quetta (Qta) and Turbat (Tbt). The design of experiment was Randomized Complete Block Design (RCBD) with three replicates. Application of SA and H 2 O 2 , priming (P), Foliar spray (FS) and combinational treatments (P+FS) were highly helpful in alleviating the negative role of cadmium toxicity. The essential nutrients i.e. nitrate (NO 3 - ), calcium (Ca 2+ ) and potassium (K) were affected by Cd +2 induced toxicity however, the substantial role of SA and H 2 O 2 widely helped to reduce the cadmium stress and boosted up the plant nutrients content. In a nutshell, exogenous treatments of SA and H 2 O 2 enhanced the yield potential along with highest silymarin contents in milk thistle seeds which is of prime significance for its medicinal importance in treatment of liver diseases. The data obtained in this study highly recommend the priming and foliar spray of SA and H 2 O 2 on milk thistle plants, as the best solution to alleviative the cadmium toxicity which will ultimately leads to better growth and yield of the plants.

Abstract:Milk thistle's extract by an organic solvent is known as silymarin, which is a mixture of about 75% flavolignan and trace content of flavonoid (mainly taxifolin), other ingredients involve fatty acids and polyphenolic compounds. The major flavolignans are silybin, isosilybin, silydianin, and silychristin, quantitatively silybin is considered as the major compound followed by isosilybin, and both are represent approximately 60% of silymarin. This study was designed to determine the concentration of silybin in different parts of the plant and to study the effect of extraction methods, solvents and the time of collection on silybin concentration, which have been done by HPLC instrument. The results show that the higher yield of silybin is obtained from the seed using ethanol in the soxhlet apparatus and that all other parts of the plant which are produced in huge amount by the plant also contain a good concentration of silybin when compared with seeds, So they can be used as an alternative source for silybin production.

Abstract:Silybum marianum (L.) Gaertn, a herbaceous plant with a long history in traditional medicine for the treatment of hepatobiliary diseases, particularly in Europe, which has attracted attention for its remarkable therapeutic effect. This review systematically summarizes the research progress in the botany, phytochemistry, pharmacology, comprehensive utilization and synthetic biology of S . marianum . Up to now, more than 20 types of flavonolignan components have been isolated from S . marianum . In addition, the rearch on fatty acids and triterpenoids is also constantly improving. Among them, silybin is the most active compound in flavonolignans components. Its pharmacological effects in vivo and in vitro include anti-inflammatory, antioxidant, anti-tumour, hypoglycaemic, neuroprotective and immunoregulatory properties. The use of coniferyl alcohol and taxifolin as substrates to produce silybin and isosilybin under the action of enzyme catalysis is the commonly used biosynthetic pathway of silymarin, which provides support for a comprehensive analysis of the synthetic pathway of silymarin. In addition to medicinal use, the extracts of plants also have broad application prospects in the production of food, healthcare products, cosmetics and other aspects. In addition, the chemical composition, pharmacological mechanism and synthetic biology of S. marianum need to be further studied, which is very important for its clinical efficacy and resource development.

Abstract:Extracts prepared from the seeds of the medicinal plant milk thistle [Silybum marianum (L.) Gaertn. (Asteraceae)] are widely used as dietary supplements due to anti-inflammatory, antitumor, and hepatoprotective effects. Called silymarin, the main components of lipophilic extracts of milk thistle seeds are flavonoids and flavonolignans including silybin A, silybin B, isosilybin A, isosilybin B, silydianin, silychristin, taxifolin, and 2,3-dehydrosilybins. The aim of this study was to develop a method based on UHPLC-MS/MS for the chemical authentication and standardization of milk thistle silymarin. Validation included the method of standard addition to account for the lack of a blank matrix. Potential matrix effects were investigated by analyzing silymarin standards dissolved only in the initial UHPLC mobile phase. Measurements of six flavonolignans and taxifolin in the milk thistle extract using UHPLC-MS/MS with standard addition or external standard calibration produced similar results for all analytes except silydianin and 2,3-dehydrosilybin B, which showed significant peak enhancement during negative ion electrospray due to botanical matrix effects. The UHPLC-MS/MS-based method of standard addition requires <10 min per injection and is suitable for the standardization of silymarin from milk thistle in support of preclinical and clinical studies of safety and efficacy.

Abstract:Milk thistle is one of the most popular ingredients in the liver protection products market. Silymarin is the main component of milk thistle and contains multiple isomers. There have been few studies focusing on the compositional ratios of silymarin isomers. In this study, we developed an HPLC method for the separation and quantification of silymarin isomers, thereby elucidating their compositional ratios. Through the analysis of more than 40 milk thistle extract products on the market, we found that the ratios, specifically Ratio 1 (the silybin B content to the silybin A content, SBNB/SBNA) and Ratio 2 (the sum of the contents of silybin B and isosilybin B to the sum of the contents of silybin A and isosilybin A, (SBNB + IBNB)/(SBNA + IBNA)), are highly consistent across milk thistle extracts, averaging approximately 1.58 and 1.28, respectively. Furthermore, such ratios were verified in milk thistle seed samples. This study introduces significant findings concerning the stable ratios among silymarin isomers in milk thistle extracts and seeds, thereby offering an innovative approach for quality assurance of milk thistle extracts.

Abstract:Grains are major farm animals' diet ingredients, and one of the main concerns is when are mycotoxin (MyT) contaminated, compromising animals' health, performance, and product safety. Among the natural phytocompounds that are being used to prevent MyT damage, silymarin (SIL), an extract from the seed of the milk thistle (MT) is a suitable candidate. This review aims to examine the scientific evidence concerning the anti-MyT toxicity effects of MT/SIL in poultry and livestock. In vitro and in vivo studies (n = 27) showed that MT whole plant, seed, or SIL-standardized extract had positive effects on animal health, performance, and restoring the hepatic activity, with a reduction of organ lesions caused by MyT. Furthermore, showed utility for combating MyT-immunodepression, improving intestinal health, and limiting the excretion of toxins residues in food of animal origin, although in some cases, MT/SIL supplementation does not produce appreciable effects. The use of MT in animal nutrition can be useful since the bioactive compounds, also if present in variable amounts, can help the animals to counteract the effects of MyT. The use of the phytoextract of SIL, due to its cost, can be useful if it reported the specific bioactive compounds, recognize for their pharmacological activities.

Abstract:based this therapeutic attempt on the supposition that the infusion of insulin microdoses at a continuous basal rate could represent an effective desensitization method. This supposition was inaccurate because the patient's immunological profile was unchanged after 1 yr on CSII. We think that CSII is able to maintain the mast cells in chronically degranulated insulin-infusion areas and so to block the immunologic reaction liable for allergic manifestations even when greater amounts of insulin (premeal boluses) are delivered. In conclusion, CSII could be useful in the management of insulin allergy when usual therapeutic schemes are unsuccessful.

Abstract:BACKGROUND: Recent years have seen an explosion of scientific papers that deal with drugs from the fruits of milk thistle and its active substances silymarin (standardized mixture of flavonolignanes), thus justifying an updated systematic review.METHODS: Electronic databases identified silymarin, silibinin, silicristin or milk thistle as descriptors in >700 papers (34% published in last 5 years; 92% dealt with animal pharmacological). Only papers adequately reporting on experimental conditions, dosing, variables tested and statistics were analysed.RESULTS: Silymarin was found to modify specifically the functions related to various transporters and receptors located in the cell membranes; that is, organic anion uptake transporter peptides (OATP), ABC transporters (P-gp), bile salt export pump, as well as TNF-alpha-dependent and possibly selectin-dependent phenomena. In the cytoplasm, some antioxidant properties and the inhibition of the lipoxygenase pathway seem quite selective and could concur to the antitoxic effects. Some effects like the inhibition of inducible nitric-oxide synthase, of nuclear factor kappa B, and reduction of collagen synthesis are indicative of DNA/RNA-mediated effects. Several studies using 'in vitro' and 'in vivo' cancer models suggest a potential of silymarin in such diseases. Topical and systemic silymarin has skin protective properties against UV-induced damage in epidermis and causes an up-regulation of tumour-suppressor genes p53- and p21CIP1. There were no data on hepatic viral replication, viremia or spontaneous tumours in the data examined.CONCLUSIONS: Data presented here do not solve the question about the complex mechanism(s) of action of the medicinal herbal drug silymarin. Silymarin may be a natural multi-functional and multi-target drug.

Abstract:Abstract Silymarin ( Silybum marianum ; SM), popularly known as milk thistle, is an extract that has been used for many centuries to treat liver diseases. In recent years, several studies have shown that SM is not only just another antioxidant but also a multifunctional compound that exhibits several beneficial properties for use in the treatment and prevention of different types of pathologies and disorders. This review aims at demonstrating the main protective activities of SM in diseases, such as cancer, diabetes, hepatitis, non-alcoholic fatty liver disease, alcoholic liver disease, hepatitis C virus, hepatitis B virus, metabolic syndrome, depression, cardiovascular diseases and thalassemia, in addition to its photoprotective activity in in vitro tests and preclinical studies. Its main functions include antioxidant and anti-inflammatory effects, and it acts as modulator of signaling pathways. It has been suggested that SM presents great multifunctional potential and is capable of achieving promising results in different types of research. However, caution is still needed regarding its indiscriminate use in humans as there are only a few clinical studies relating to the adequate dose and the actual efficacy of this extract in different types of diseases.

Gaertn. can be a winter annual or a biennial medicinal plant. This study was focused on milk thistle as a weed in the sustainable farming system with the crop rotation of maize for grain, common pea for grain, durum wheat and milk thistle. The assessment of the occurrence of Milk thistle in sustainable crop rotation was conducted at the Experimental Base of Faculty of Agrobiology and Food Resources, Slovak University of Agriculture in Nitra in the years 2013–2015. An actual weed infestation of maize, common pea and durum wheat stands with milk thistle was evaluated before preemergence application of herbicides, in the spring time. Second screening of actual weed infestation of all stands in sustainable crop rotation with milk thistle was done before crops harvest. Screening of each field was made on 1 m2 area with three replications. The three randomly established sample quadrants were situated minimally 10 m from field margin and apart from each other, respectively. The level of infestation was evaluated according to average density of weeds per square meter. Obtained data were statistically analyzed by software Statistica 7.0 by Analysis of variance (ANOVA), LSD test (P = 0.05). According to statistical analyses, in the year 2014 maize stands was infested with the highest and highly significant amount of Silybum marianum (7.8 plants per m2). Stands of pea for grain in the second year after milk thistle cropping were infested only with 1.23 plants per m2. Durum wheat stands were infested with 0.96 milk thistle plants per m2. The originality of this paper is in testing the introduction of a new perspective crops in sustainable crop rotation. Milk thistle not infestation fields within the reporting unit. Dominant weed species in canopy of Silybum marianum are: Echinochloa crus galli, Chenopodium album, Amaranthus spp. (retroflexus, powelli), Atriplex spp. (patula, acuminate, prostrata), Avena fatua , etc. All dominant weed species belongs to the group of annual early and late spring weeds. Next very dangerous group are perennial weeds Cirsium arvense, Convolvulus arvensis and Sonchus arvensis . Keywords : Sylibum marianum L. Abstract:Liver disorders are one of the common recent problems affects on the human health. These disorders due to many environmental polluted sources. Many herbal, medicinal and pharmaceutical plants and their extracts are widely studied by many researchers. Silybum marianum got a bright reputation in relieve the liver diseases, and that might be for the potent silymarin mixture. Mechanism of action for silymarin conducted mainly to the antiradical and anticarcinogenic roles. Ethyl acetate (100mg/kg bw) and ethanol seed extracts for S. marianum (100mg/kg bw) were tested against the injection (i.p.) by carbon tetrachloride (2 ml/kg bw) the inducer of liver damage. Their activity were compared with standard hepatic drug hepaticum (100mg/kg bw) for 10 days. Ethanolic extract showed the most significantly decrease in the liver enzymes. For the oxidative experiments, ethyl acetate showed the most increase for glutathione level and the risk factor HDL/LDL significantly. Hepaticum was the most powerful group for the significant decreasing for malondialdehyde and fucosidase activity. Some equal improvements were noticed in the histopathological studies for the protective groups.
