To begin with a semi-medical problem, the incidence of head lice in school children is not always given by local authorities, and even the data available (as that for Glasgow, in Table I) is almost certainly an underestimate, owing to rapid inspections which are all that can be done by school nurses in the time available. Donaldson (1970~ in a special detailed survey of Teesside, revealed levels up to 16 per cent in some districts; and my colleague, Maunder (1971) has given reasons for supposing that up to a million inhabitants of the U.K. may be infested with one or another forms of lice.
Effect of Enzymatic Hydrolysis on the Zinc Binding Capacity and in vitro Gastrointestinal Stability of Peptides Derived From Pumpkin (Cucurbita pepo L.) Seeds

Abstract:Zinc is a crucial micronutrient for maintaining body immune system and metabolism function. However, insufficient intake from diet may lead to zinc deficiency and impair normal body function. In addition, conventional zinc salts supplementation has the disadvantage of low bioavailability since the zinc ions may be easily chelated by dietary fiber or phytate commonly found in diets rich in plants, and form precipitates that cannot be absorbed. Therefore, the objective of the present study is to prepare pumpkin seed derived peptides and to evaluate the effect of structure and surface properties on the zinc binding behavior of the pumpkin seed protein hydrolysate (PSPH), as well as their gastrointestinal stability. Briefly, different PSPHs were prepared using enzymatic hydrolysis method with bromelain, papain, flavourzyme, alcalase, and pepsin. The particle size, zeta potential, surface hydrophobicity, degree of hydrolysis, ATR-FTIR spectra, and zinc binding capacity were determined. The representative samples were chosen to characterize the binding energy and surface morphology of PSPH-Zn. At last, the in vitro gastrointestinal stability of PSPH and PSPH-Zn were evaluated. Our results showed that peptides hydrolyzed by papain had the largest average molecular weight, smallest particle size, highest hydrophobicity, and the greatest zinc binding capacity. Zinc showed better gastrointestinal stability in PSPHs chelates than in its salt. Meanwhile, PSPH-Zn with higher zinc binding capacity showed better stability. The result of this study indicated pumpkin seed hydrolyzed by papain may be used as a potential source for zinc fortification. The findings in this study may provide important implications for developing plant-based zinc chelating peptides.

Abstract:The life history of Echinostoma paraensei, a new Brazilian species with 37 collar spines, has been completed experimentally. The planorbid snail Biomphalaria glabrata serves as first intermediate host, and intramolluscan stages also develop in Physa rivalis. Sporocysts usually develop in the heart. Rediae are of two types, distinguishable by large or small pharynges. Cercariae released 25 or more days postexposure develop into metacercariae in the pericardial sac and kidney of snails, sometimes in various tissues of snails infected with rediae. Adults develop in hamsters, mice, or rats after they are fed metacercariae, but not in pigeons, chicks, or ducklings. The natural final host is not known. In recent papers (Basch and Lie, 1966; Lie, 1966a, 1967; Lie et al., 1965) we described the destructive antagonism of larval echinostomes to trematode sporocysts developing in the same snail. Various echinostome species naturally transmitted by Biomphalaria glabrata (Say) in Brazil are being sought and evaluated to determine their efficiency in destroying Schistosoma mansoni Sambon. This paper describes the life history of a new species of echinostome encountered during our studies. MATERIALS AND METHODS Cercariae of this species appeared in collections of B. glabrata snails made in Belo Horizonte (Bairro Sao Domingos), Caratinga, and Bambui, all in the State of Minas Gerais, Brazil. Snails were shipped by air to San Francisco, California, where we studied the life histories of their trematodes. Cercariae were permitted to encyst in laboratory-bred B. glabrata, and metacercariae were fed to hamsters and albino rats and mice. Eggs from washed hamster stools produced miracidia, which were used principally to infect laboratory-raised albino B. glabrata of a strain obtained from the National Institutes of Health, Bethesda, Maryland. Infected snails were kept in clear plastic, 1-gal aquaria at 24 to 27 C and fed on red leaf lettuce (Lactuca sativa). Techniques for the study of the parasite were the same as in previous studies (Lie, 1963a; 1965; 1966b, c). All measurements are in microns. Eggs kept in distilled water in a petri dish at 28 C begin to hatch in 11 days. Exposure to light stimulates hatching, which usually occurs in the morning. Newly hatched miracidia swim rapidly in the water for several hours and are positively phototactic. They enter the snail host through its exposed parts while shedding their epidermal plates. Complete penetration takes about half an hour. Abstract:Pumpkin seed oil is a by-product, abundant in nutrients and bioactive components that promote several health benefits. This study aimed to compare chemical compositions, antioxidant, and pharmacological activities of pumpkin seed oils extracted from Cucurbita moschata Duch. Ex Poir. (PSO1) and Cucurbita moschata (Japanese pumpkin) (PSO2) by aqueous enzymatic extraction. An enzyme mixture consisting of pectinase, cellulase, and protease (1:1:1) was used in the enzymatic extraction process. Fatty acid composition of the oils was determined using fatty acid methyl ester/gas chromatographic-mass spectrometry. Antioxidant activity assays were measured by using stable free radical diphenylpicrylhydrazyl, radical cation 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonate, ferric reducing/antioxidant power, and ferric thiocyanate assay. Inhibition of enzymes involving skin aging and whitening process was investigated. Linoleic acid was a major component of all pumpkin seed oils. Additionally, there was also a significant amount of oleic acid, palmitic acid, and stearic acid detected. PSO2 possessed the highest antioxidant activities compared to PSO1 and commercial pumpkin seed oils (COM1 and COM2). Both PSO1 and PSO2 exhibited higher inhibitory effects on hyaluronidase, collagenase, and tyrosinase than the commercials. Therefore, aqueous enzymatic extraction could yield pumpkin seed oils with higher antioxidant, anti-aging, and whitening activities. This is beneficial for further pharmacological studies and can be used as a functional food for skin benefits.

Abstract:Pumpkin (Cucurbita maxima) seeds are a rich source of ingredients such as aminoacids, fatty acids, minerals, and phytochemicals exhibiting nutraceutical effects on human health. In this work, partially defatted pumpkin seeds flour, a by-product obtained during the manufacture of pumpkin seeds oil, was studied as an additive for common wheat flour. We explored the physico-chemical properties as well the content in amino acids of the partially defatted pumpkin seeds. The obtained results revealed that partially defatted pumpkin seeds are a good source of protein (42.75% d.m.), lipids (12.28% d.m.), total carbohydrates (37.4% d.m.), from which crude fiber (26.64% d.m.). This by-product presents a high mineral content (mg/100g): potassium (1290), magnesium (693), iron (87.8), zinc (11.5) and copper (2.49).The partially defatted pumpkin seeds proteins contain significant amounts of essential amino acids such as valine, histidine, isoleucine, leucine, threonine and methionine.
Physicochemical and gel properties of pumpkin seed protein: a comparative study

Abstract:Summary The physicochemical properties and heat‐induced gel properties of pumpkin seed protein isolates (PSPI) were explored and compared with soybean protein isolate (SPI) and pea protein isolates (PPI). These plant proteins were subjected to ultrasound, acid, or alkaline treatment (pH 3 or 11). The content of amino acids, and subunit composition were different in PSPI compared with SPI and PPI. Rheological and textural experiments showed that native PSPI had the highest gel strength among the three proteins. This was due to the high hydrophobicity of PSPI and the filling effect of insoluble particles in the protein solution. PSPI had a more pronounced improvement in gel properties after ultrasound or alkaline treatment compared with SPI and PPI, which was due to the increased surface hydrophobicity, increased solubility, and reduced particle size of the proteins. The acid treatment induced insoluble aggregates increased the protein particle size, and decreased the protein solubility, resulting in a negative effect on the PSPI protein gel. Therefore, it can be concluded that PSPI has better heat‐induced gelation than SPI and PPI and can be used as a substitute for SPI and PPI in the food industry.

Abstract:ABSTRACT: Response surface methodology was used to determine optimum conditions for extraction of protein from pumpkin seed meal. A central composite rotational design (CCRD) including independent variables such, pH and sodium trimetaphosphate (STMP) content was used. Maximum yield was obtained at a central point of CCRD, with STMP content and pH of 4% and 4.5, respectively. These conditions resulted in protein yield of 50.04 g of soluble protein from extract/100 g pumpkin seed. The optimization of pH and STMP content allowed obtaining a product with a high protein concentration (62.56 g 100 g-1), digestibility (62.03 g 100 g-1) and concentration of essential amino acids (27.26 g 100 g-1). Regarding the polyphenols concentration, phosphorylated protein concentrate from pumpkin seed and the pumpkin seed meal presented 13.11 g 100 g-1 and 23.19 g 100 g-1, respectively. The adequacy of the model was confirmed by extracting the protein under optimum values. These results help in designing the process of optimal protein extraction from pumpkin seeds.

Abstract:Nowadays, agricultural waste byproducts are exploited in the food industry rather than discarded. Pumpkin is one of the most significant vegetable crops that is widely consumed in farmland and certain urban regions. The current study was designed to measure the phytochemical constituents, food application, health benefits, and toxicity of pumpkin and pumpkin byproducts. Pumpkins and pumpkin byproducts (seeds, leaf, and skin/peel) can be utilized as functional ingredients. Different parts of the pumpkin contain bioactive compounds including carotenoids, lutein, zeaxanthin, vitamin E, ascorbic acid, phytosterols, selenium, and linoleic acid. Pumpkin is used in various food sectors as a functional food, including baking, beverages, meat, and dairy industries. Furthermore, the leaves and pulp of the pumpkin are used to produce soups, purees, jams, and pies. Different parts of pumpkins have several health benefits such as antidiabetic, antioxidant, anticancer, and anti-inflammatory effects. Therefore, this review paper elaborates on the pumpkins and pumpkin byproducts that can be used to develop food products and may be valuable against various diseases.

Abstract:Pumpkin peel is a main by-product produced during pumpkin consumption and processing, which is often disposed of as food waste. Therefore, it is necessary to develop ways to utilize pumpkin peel. In this study, the pectic polysaccharides were extracted from pumpkin peel, and their properties were evaluated. Results indicated that the extraction yield of pectin polysaccharides from Dongsheng pumpkin peel was the highest. Meanwhile, among the pumpkin peel pectic polysaccharides (PPs), Dongsheng PP exhibited the highest methylation degree (22.38%), acetylation degree (15.92%), molecular weight (6.26 × 105 Da) and initial apparent viscosity. Variety had little effect on the microstructure and functional groups of PPs. Compared with the other PP samples, Beibei PP (BBP) had the lowest molecular weight coupled with highest protein content, resulting in the highest emulsifying activity, emulsion stability and antioxidant activity. In addition, all the PPs had the ability to promote the growth of probiotics, and BBP and Banli PP (BLP) shown better capacities to promote the growth of Lactobacillus acidophilus and Bifidobacterium breve. Hence, it could be concluded that extraction pectin from pumpkin peel is a potential method for the utilization of pumpkin peel.

Abstract:Pumpkin polysaccharide was extracted with hot water. Then the protein in the polysaccharide was removed by Sevage method, and finally the pumpkin polysaccharide (P) was obtained by dialysis and column chromatography separation. The content of polysaccharide reached 96.8 %. The sugar chain in pumpkin polysaccharide had α-glycoside bond. The pumpkin polysaccharide could significantly increase the protein content, GSH-Px activity and SOD activity in serum, liver, spleen and kidney of mice, and also significantly reduce the MDA content in serum, liver, spleen and kidney of mice. Therefore, pumpkin polysaccharide has better antioxidant activity in vivo. This provides a basis for the development and utilization of antioxidant food.

Abstract:Pumpkin is a well-known vegetable, among the members of Cucurbitaceae family, due to its importance as pharma food. Keeping in view the antidiabetic and plasma lipids lowering potential of pumpkin, the present study was conducted to investigate that, which part of pumpkin (peel, flesh, and seeds), possess more bioactive compounds, exhibiting antihyperglycemic and antihyperlipidemic potential. Albino rats with 190-210 g body weight were divided into 11 groups. Five rats were included in each group; group A was negative control, group B was positive control, and groups C to K were diabetic rats fed with pumpkin peel, flesh, and seed powders. Diabetes was induced in rats with the help of alloxan monohydrate. During 28 days of experimental period, blood glucose level of different rat's groups was checked with the help of glucometer, at every 7 days interval and at the end of 28 days study, plasma lipids were checked with the help of commercial kits. A significant decrease in blood glucose level ( 128.33 ± 1.67 mg / dl ), TC ( 88.43 ± 0.66 mg / dl ), TG ( 69.79 ± 0.49 mg / dl ), and LDL-C ( 21.45 ± 0.08 mg / dl ) was recorded in rat groups fed with 15 g pumpkin seed powder, at the end of study. After pumpkin seeds, second significant antihyperglycemic and antihyperlipidemic effect was recorded in rat's groups fed with 15 g pumpkin peel powder. Pumpkin flesh powder effect in lowering blood glucose level and plasma lipids was less significant as compared to seeds and peel powder. As the dose of the pumpkin powders was increased from 5 to 10 and then 15 g, the blood glucose-lowering and plasma lipid-lowering effect became more significant. Similarly, as the experimental duration was expanded from first week to 28 days, this antihyperglycemic and antihyperlipidemic effect became more significant. These results were sufficient to conclude that pumpkin has high potential to be used in human diet to cope with noncommunicable diseases like diabetes and hypercholesterolemia.

Abstract:Pumpkin-seed protein isolates were treated with high-intensity ultrasound (HIU). The effects of different ultrasound powers at the same treating time on the physicochemical, structure and foaming properties of pumpkin-seed protein isolates (PSPIs) were investigated. The results showed that the ultrasound treatment transformed the PSPI aggregates into smaller aggregates with a more uniform distribution. When the pH value increased, the solubility and foaming ability of the protein also increased significantly. Increasing the ultrasound power also increased the surface hydrophobicity of the PSPIs and the total sulfhydryl contents, while the active sulfhydryl content decreased. The most significant effect on the total sulfhydryl content occurred when 500 W ultrasonic power was used. The ultrasound treatment also had a significant effect on the chromaticity and turbidity of the pumpkin-seed protein isolates. With an increase in ultrasonic power, scanning electron microscopy (SEM) showed that the sizes of particles decreased as their distribution increased. Moreover, ultrasound treatment was found to be beneficial for enhancing and improving foaming performance. Ultrasound modification affected the protein's physicochemical properties and structure, which contributed greatly to the corresponding functional and nutritional properties of the protein. It would be, therefore, useful to develop and utilize plant protein and to improve its added-value.
Quality assessment and variety classification of seed‐used pumpkin by‐products: Potential values to deep processing

Abstract:Seed‐used pumpkin (SUP) is known as a traditional popular crop, which is mainly processed for seeds. However, the by‐products (49 times the amount of seeds) were disposed directly into the field as waste. In this study, potential values of seed‐used pumpkins' by‐products (SUPBs, peel and pulp) as food resource were investigated. Physico‐chemical, nutritional, and aroma profile of ten varieties of SUPBs were characterized, and variety differences were also distinguished. Peel "a*" value, water, fructose, crude fat, sucrose, and Ca contents were the 6 characteristic indicators of SUPBs which screened through correlation analysis, principal component analysis (PCA), and PCA‐X model. Comprehensive evaluation of physico‐chemical, nutritional, and aroma profile, four varieties by‐products (Jf8#, Nf8#, Rbf#, and Rf9#) were always characterized into Cluster Ⅰ. Other varieties were classified into Cluster Ⅱ based on aroma profile. However, two varieties by‐products (Db1# and Xn1#) presented significant differences from others (Db2#, Db3#, Db4#, and Myxc2#) in physico‐chemical and nutritional indices, they were classified as Cluster III and IV, respectively. Db1# had the highest nutritional value of soluble solid (11.78 ºBx), pectin (1,166.15 mg/ 100 g), total carotenoid (19.57 mg/ 100 g), and total sugar (13.69 g/ 100 g). Among all the SUPBs, Db1# had a relatively higher nutritional value, which was suitable as food resource for deep processing.

Abstract:The identification and isolation of bioactive peptides from food sources is a very active research area considering their potential use in both functional food products and pharmaceuticals. In silico tools can be used to predict possible bioactivity of peptides and can aid experimental procedures by narrowing down high number of enzyme combinations for enzymatic hydrolysis of proteins. In this study, tomato seed, which is a valuable industrial waste, was investigated as a potential bioactive peptide source. Nine tomato (Solanum lycopersicum) seed proteins were chosen for in silico analysis to assess their potential ACE inhibitory, DPP-IV inhibitory and antioxidative peptides. After homology assessment, the profiles of potential biological activity of proteins and the simulated enzymatic hydrolysis with fifteen individual enzymes and two enzyme combinations were performed and the released peptides were evaluated. This study demonstrates that tomato seed proteins represent a remarkable source for the generation of bioactive peptides. According to the in silico results, it is predicted that enzymatic hydrolysis with pepsin and pepsin + trypsin combination may produce peptide fragments with high bioactivity. However, it should be noted that the resulting peptides and their activities were predicted under in silico conditions that might be different from experimental conditions.

Optimization of aqueous-assisted extraction of polysaccharides from pumpkin (Cucurbita moschata Duch) and their biological activities

Abstract:The pumpkin pulp contains a greater composition of edible polysaccharides and has reported with excellent biological applications. This research pertains to optimize the extraction of polysaccharides from the fleshy portion of the pumpkin using aqueous assisted extraction (AAE). The result showed that the optimal extraction condition of pumpkin polysaccharide was as follows: extraction temperature at 55 °C, pH 4.5, and enzyme concentration of 4000 µ/g for 80 min. Under the optimal extraction condition, the yield of pumpkin polysaccharide via AAE (15.4) was significantly higher. The biological activities of extracted polysaccharide including α-amylase inhibition (57.41% at 1000 µg/mL) and anti-inflammatory (50.41% at 25 µg/mL) activity increased significantly. Additionally, the antioxidant activities of extracted pumpkin polysaccharides including IC50 values of DPPH and ABTS were 59.87% and 58.74%, respectively. The pumpkin polysaccharide has maximum inhibitory effects against bacterial strains especially for Escherichia coli than that of fungal strains. It is suggested that the aqueous assisted extraction of is a cost-effective promising method to decrease the processing time as well as enhancing extracted polysaccharide yield - times.
Antioxidant, Antibacterial and Antiproliferative Activities of Pumpkin (cucurbit) Peel and Puree Extracts - an in Vitro Study.

Abstract:Natural resources right from the beginning of the human civilization has paved the way to human being to combat different challenges. The big challenge was to safe the human being from diseases and shortage of food. Plants helped the man in both areas very efficiently. No doubt when plants are used as food actually we are also taking lot of compounds of medicinal values in an excellent combination which naturally reduce the risk of diseases. Extraction and purification of several medicinally important compounds also gave the way to develop pharmaceutical industry in addition to its own therapeutic effects against different lethal diseases. Pumpkin is one of the several medicinal important vegetables used in different way on the behalf of its admirable power to combat different diseases. Antioxidant and biological studies showed very important results. A good coherence was found among extraction yield (10.52 to 18.45%), total phenolics (1.13 to 6.78 mg GAE/100g), total flavonoids (0.23 to 0.72mg CE/100g) and antioxidant potential (≻70%). Antibacterial assays of peel and puree extracts advocated good potential to stop the growth and division of pathogenic bacteria. Further biological activity study was carried out using MDBK cancer cell line. The growth inhibitory effect on cancer cell line using MTT assay showed methanol extracts of peel and puree both remained efficient to inhibit growth (≻35%) and cell division of cancer cells. Our results showed that extracts of pumpkin puree and its waste, peel, may be utilize to prepare functional food against pathogenic born diseases and most active compounds may also be extracted, concentrated and converted into tablets or suspension form for therapeutic purposes.
Extraction of pumpkin polysaccharide by complex enzyme method and its antioxidant research.

Abstract:[Objective] This paper aims to study a new method of extracting pumpkin polysaccharide from pumpkin. Single factor experiments were conducted to examine the effects of extracting time,temperature,the solid-liquid ratio and pH value on the extraction yield of polysaccharide from pumpkin. [Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. [Conclusion] This study provided basic data for research and application of Pumpkin polysaccharide.

Abstract:This study aimed to extract polysaccharides from pumpkin, characterize the structures of four of them, and evaluate their in vitro antioxidant and hypoglycemic activities. Additionally, an animal model of type 2 diabetes mellitus (T2DM) was established and used to determine their hypoglycemic and hypolipidemic effects in vivo , and the underlying mechanisms related to the regulation of gut microbiota. Water-extracted crude pumpkin polysaccharides (W-CPPs), water extraction and alcohol precipitation crude pumpkin polysaccharides (WA-CPPs), deproteinized pumpkin polysaccharides (DPPs), and refined pumpkin polysaccharides (RPPs) were sequentially extracted and purified from pumpkin powder by hot water extraction, water extraction, and alcohol precipitation, deproteinization and DEAE-52 cellulose gel column, respectively. The extraction and purification methods had significant influence on the extraction yield, physicochemical properties, and in vitro antioxidant and hypoglycemic activities. W-CCP and RPPs had a significant positive free radical-scavenging capacities and inhibitory activities on α-glucosidase and α-amylase. RPP-3 not only inhibited the uptake of glucose in Caco-2 monolayer but also promoted the excretion of glucose, while RPP-2 had no inhibitory effect. Animal experiment results showed that W-CPP treatment significantly improved the T2DM symptoms in mice, which included lowering of fasting blood glucose (FBG), reducing insulin resistance (IR), and lowering of blood lipid levels. It increased the diversity of intestinal flora and reduced the harmful flora of model mice, which included Clostridium, Thermoanaerobe, Symbiotic bacteria, Deinococcus, Vibrio haematococcus, Proteus gamma , and Corio . At the family level, W-CPP (1,200 mg/kg) treatment significantly reduced the abundance of Erysipelotrichaceae , and the Akkermanaceae of Verrucobacterium became a biomarker. Pumpkin polysaccharides reshaped the intestinal flora by reducing Erysipelotrichaceae and increasing Akkermansia abundance, thereby improving blood glucose and lipid metabolism in the T2DM mice. Our results suggest that W-CCP and RPP-3 possess strong antioxidant and hypoglycemic activities, and are potential candidates for food additives or natural medicines.

Abstract:Traditional pumpkin (Cucurbita moschata) and Japanese pumpkin (C. maxima) consist of natural polysaccharides. From a scientific basis, natural polysaccharides could be applied to improve hydration in the cosmetic field. The purified polysaccharide was extracted and deproteinized with the CaCl2 method. Japanese pumpkin showed the higher value of physicochemical properties including yield (12.96 ± 0.60%), total polysaccharide content (0.89 ± 0.04 mg/mL), swelling capacity (4.00 ± 0.00%), swelling index (1.04 ± 0.00%), solubility (126.67 ± 5.77%), viscosity (1.25 ± 0.00 cps), water capacity (0.93 ± 0.15 g/g) and oil absorption capacity (5.93 ± 0.06 g/g) than traditional pumpkin. Additionally, Japanese pumpkin (IC50 9.30 ± 0.58 μg/mL) provided higher antioxidant activity by DPPH assay than traditional pumpkin (IC50 9.98 ± 0.25 μg/mL). The evaluation of efficacy on skin hydration in fifteen Thai volunteers indicated that Japanese pumpkin showed non-skin irritation. An extract concentration of 0.05–0.1% showed a significantly increased effect in moisturizing ability according to concentration (p < 0.05). This result supported that it was safe and effective to use as a moisturizer for cosmetic products.
