Abstract:The study presented the effects of modulating the hydrophobic interaction and disulfide bond on the properties of myofibrillar protein (MP) emulsion gels at high temperature (95 degrees C) and the differentiation on the contri-bution of non-covalent (hydrophobic interaction) and covalent intermolecular interactions (disulfide bond) to the properties of interfacial protein films were also determined. The hydrophobic interactions among MP were modulated by the addition of octenyl succinic anhydride (OSA), and the disulfide bonds were modulated by the SH/SS exchange reactions mediated by GSH. The results showed that the MP emulsion gel properties at 95 degrees C were improved by modulating the hydrophobic interaction or disulfide bonds, and the dynamic interfacial adsorption of MP and dissipation quartz crystal microbalance experiments showed the interfacial adsorption pattens of protein were also changed. In addition, the hydrophobic interactions putted emphasis on improving the gel matrix, whereas the disulfide bonds focused on increasing the stiffness of interfacial protein films.

Abstract:The self-assembly adsorption of function protein on crystal surfaces, as a common phenomenon, broadly takes place in many applications of biosensors, biocapsules and bioMEMS/bioNEMS. To systematically investigate the different adsorption characteristic of the same function protein on two different crystal surfaces under the identical environment, a hybrid surface composing silica and discontinuous Gold Nano Film (GNF) was fabricated by Physical Vapor Deposition (PVD) and ultrasonic cleaning method, where the dynamic process of the self-assembly adsorption of Bovine Serum Albumin (BSA) was in situ observed by Atomic Force Microscope (AFM). The variations on the junction area of the two different surfaces were studied in the aqueous solution before and after injecting BSA with 0.05 mg/ml concentration. It was found that silica, compared with same hydrophilic GNF, took on a fairly weak adsorption force. The results indicated that the adsorption strength of BSA on the hydrophilic crystal surface was determined not only by hydrophilic property, but also other interaction forces, like Van der Waals and so on. Moreover, observed under the contact mode of AFM, BSA adsorbed on GNF had great tendency to forming a ridge-like topography. These results may be helpful in the application of immunosensors and other areas.
The adsorption characteristics of 2D fibril and 3D hydrogel aggregates at the O/W interface combining molecular dynamics simulation

Abstract:The 2D/3D microstructures and hydrophobic domains of interfacial proteins play important roles in interfacial adsorption and emulsification stability. The effects of spatial conformation, crystal structure and micro-structures on interfacial adsorption were investigated combining molecular dynamics simulation. The results showed that with the increase of pHs, the α-helix contents and surface hydrophobicity in the emulsified aggregates decreased significantly, and the β-sheet contents increased significantly, in which the formation of interfacial protein membrane caused molecular re-arrangement. The α-helix contents and surface hydrophobicity in 2D fibril aggregates were significantly higher than those in 3D hydrogel aggregates, but the β-sheet result was opposite along with different hydrophobic amino acid sequences. The peak intensity and crystallinity of interfacial proteins in hydrogel aggregation mode were higher than those in fibril aggregation mode, and the transformation of crystal structure can affect the interfacial microstructure. The interfacial protein membrane is mainly composed of fibrous rod-like aggregates, among which 2D fibrous network structure is formed at pH 3.0 and 3D multilayer network structure is formed at pH 9.0. Due to the differences in spatial conformation, crystal structure and characteristic micro-structure caused by emulsification and adsorption, the interfacial adsorption rate of 2D fibril aggregates is higher, and the ability of 3D hydrogel aggregates to reduce interfacial tension is greater.

Abstract:The influence of surfactants (Span 80, Span 60, PGPR and sodium caseinate) on the stability of W/O emulsions stabilized by diacylglycerol (DAG)-based solid Lipid nanoparticles (SLN) was investigated. DAG-based SLN showed phase separation at high NaCl concentration and acidic pH although the SLN-stabilized W/O emulsion was minimally affected. Span 80 increased the hydrophobicity whereas Span 60 increased the hydrophilicity of SLN. With concentration close to CMC , both Span surfactants caused detachment of SLN from interface and reduced emulsion stability. Nonetheless, addition of a small amount (0.1 wt%) of Span surfactants increased the interfacial modulus. The co-adsorption of SLN and PGPR at the oil-water interface promoted the formation of denser network structure and higher interfacial elasticity, which enhanced the emulsion rigidity and freeze-thaw stability. Sodium caseinate (NaCAS) showed intense competition with particles for adsorption at the interface and caused emulsion destabilization. Consequently, the NaCl release rate was directly influenced by the emulsion stability and the interaction between surfactants with SLN. The results have important implications for applying DAG nanoparticles as stabilizers in W/O emulsions with tailoring interfacial rigidity and water-soluble component release behaviors.

Abstract:The behavior at the air/water interface and the structures of Longmuir-Blodgett monolayers at different surface pressures of rod-coil molecules, which consist of a Y-shaped rigid aromatic segment containing peripheral tetradecyloxy groups and a flexible poly(ethylene oxide) (PEO) chain with 17, 21, 34, or 45 repeating ethylene oxide units (Y17, Y21, Y34, and Y45), were investigated. For the Y21 and Y34 molecules, AFM images revealed two kinds of cylindrical nanoarchitectures formed upon compression. The nanostructured films were further investigated by UV/Vis and FTIR spectroscopy. The formation of the cylindrical nanoarchitectures was due to different tilting angles offered by the mismatch of the cross-sectional areas of the PEO chain and the benzene ring with attached alkyl chains, and the different PEO contents of the molecules. The multiple pi,pi stacking and hydrophobic interactions provide exceptional stability of the nanostructures and allow them to be preserved in the course of flipping. For the shortest PEO chain of the Y17 molecule, spontaneous aggregation occurred. The Y45 molecule revealed the formation of 2D circular domains caused by entanglement of the longest PEO chains and coiling at the air/water interface. In addition, an interesting vortical morphology was obtained for the Y21 molecule upon deposition of the film onto a mica substrate, which indicates that the substrate chemistry also has an effect on the morphologies during the film-transfer process.

Abstract:Controlling the assembly of polyelectrolytes and surfactant at liquid-liquid interfaces offers new ways to fabricate soft materials with specific physical properties. However, little is known of the relationships between the kinetics of interfacial assembly, structural and rheological properties of such interfaces. We studied the kinetics at water-oil interface of the assembly of a positively charged biopolymer, chitosan, with an anionic fatty acid using a multi-scale approach. The growth kinetics of the membrane was followed by interfacial rheometry and space- and time- resolved dynamic light scattering. This set of techniques revealed that the interfacial complexation was a multi-step process. At short time-scale, the interface was fluid and made of heterogeneous patches. At a 'gelation' time, the surface elastic modulus and the correlation between speckles increased sharply meaning that the patches percolated. Confocal and electron microscopy confirmed this picture, and revealed that the basic brick of the membrane was sub-micrometric aggregates of chitosan/ fatty acid.

Abstract:Particle film (particulate film or granular film) based emulsions have attracted great attention because they are widely used in oil, paper, food, cosmetics, pharmaceuticals and other fields. This paper summarizes the characteristics of film-forming particles on the oil-water interface such as density, size and wettability, and expounds the adsorption and diffusion of particles on the oil-water interface. The structures of particle film are highlighted, including the distribution and orientation of particles perpendicular to the interface, the arrangement of particles on the oil-water interface and the spatial structures of film. The influencing faCtors of the state of particles on the oil-water interface as well as the particle film structures are summed up, then the nature of them is analyzed from the aspects of energy and mechanics. The mechanism of particle film stabilizing emulsion is elucidated from the structures of particle film and the interfacial viscoelasticity. The particle film hinders the collision and coalescence between the dispersed droplets, which is the basis of stable emulsions. Meanwhile, strengthened interfacial viscoelasticity makes sure the particle film will not easily collapse during dispersed droplets' movement, collision and flocculation. The mechanism of solid particles stabilizing emulsion as emulsifiers provides a theoretical basis for not only the preparation of stable emulsions but also the demulsification of emulsions, which is of important realistic significance. At last, we propose prospects of future development and possible solutions of the study on the mechanism of particle film stabilizing emulsion.
Effect of Polycation in Subphase on Aggregation Structure of Monolayer and Langmuir-Blodgett Film of Anionic Amphiphile

Abstract:The aggregation structure of the monolayers prepared from an anionic amphiphile (2C14SNa) and 2C14S/poly(ethylenimine) (PEI) complex at the subphase temperature of 293 K was investigated on the basis of electron microscopic observations. The aggregation structure and surface properties of the Langmuir-Blodgett (LB) films prepared from those monolayers were also investigated by means of X-ray diffraction. Fourier transform infrared attenuated total reflection (FT-IR-ATR) spectroscopy and X-ray photoelectron spectroscopy (XPS). 2C14SNa monolayer was crystallized by compression on the water surface (compressing crystallized monolayer). For crystallization of the 2C14SNa monolayer, higher surface pressure was required because of the strong repulsion among the hydrophilic groups of the 2C14SNa molecules. The formation of polyion complex between amphiphile and PEI effectively affected the aggregation states of the monolayer on the water surface. Owing to the formation of polyion complex, the repulsive force among the hydrophilic group of anionic amphiphiles was effectively reduced and the monolayer could be crystallized without compression. Also, the transfer ratio of the monolayer onto the substrate approached unity by the addition of PEI into water subphase. Wide-angle X-ray diffraction, FT-IR, and XPS results indicated the existence of an ultrathin polymer layer between the 2C14S monolayers.
Understanding Protein-Interface Interactions of a Fusion Protein at Silicone Oil-Water Interface Probed by Sum Frequency Generation Vibrational Spectroscopy

Abstract:Protein adsorbed at the silicone oil-water interface can undergo a conformational change that has the potential to induce protein aggregation on storage. Characterization of the protein structures at interface is therefore critical for understanding the protein-interface interactions. In this article, we have applied sum frequency generation (SFG) spectroscopy for studying the secondary structures of a fusion protein at interface and the surfactant effect on protein adsorption to silicone oil-water interface. SFG and chiral SFG spectra from adsorbed protein in the amide I region were analyzed. The presence of beta-sheet vibrational band at 1635 cm-1 implies the protein secondary structure was likely perturbed when protein adsorbed at silicone oil interface. The time-dependent SFG study showed a significant reduction in the SFG signal of preadsorbed protein when polysorbate 20 was introduced, suggesting surfactant has stronger interaction with the interface leading to desorption of protein from the interface. In the preadsorbed surfactant and a mixture of protein/polysorbate 20, SFG analysis confirmed that surfactant can dramatically prevent the protein adsorption to silicone oil surface. This study has demonstrated the potential of SFG for providing the detailed molecular level understanding of protein conformation at interface and assessing the influence of surfactant on protein adsorption behavior.
Comparison of interfacial interactions between bovine serum albumin with water- and oil-soluble surfactants: an interfacial rheological study

Abstract:The interfacial interactions between bovine serum albumin (BSA) with a water-soluble surfactant polyoxyethylene sorbitan monooleate (Tween 80) and an oil-soluble surfactant sorbitan monooleate (Span 80), respectively, were studied using the interfacial dilatational rheological experiments. The BSA molecules were irreversibly adsorbed at interface and formed a network with high dilatational modulus. The increasing interfacial pressure induced the transformation of BSA molecules from a compact globular to an unfolded flexible conformation, which enhanced the relaxation process. Tween 80 formed hydrophilic complexes with BSA in the aqueous phase, significantly slowing down the dynamic adsorption. In addition, Tween 80 played a dominant role at the interface due to its higher interfacial activity, thus inhibiting the interfacial performance of BSA. As BSA and Span 80 diffused towards the interface from different phases, they adsorbed rapidly at the interface and formed a mixed adsorption layer, thus exhibiting synergistic effects in decreasing the interfacial tension. However, the alternate arrangement of BSA and Span 80 at the interface disrupted the inter-protein interactions, resulting in a significant decrease in the dilatational modulus of the interfacial film. The obtained results showed that Span 80 from the oil phase and Tween 80 from the aqueous phase interacted with the protein in different ways, and thus presented distinct dynamic adsorption processes and interfacial performance. The present work provided a theoretical foundation for their practical application in food, cosmetic, and pharmaceutical industries.

Oil-water interfacial behaviour of different caseins and stability of emulsions: Effect of micelle content and caseins concentrations

Abstract:This study aimed to investigate the interfacial behaviour of caseins in different micelle content and its effect on the stability of emulsions, including micellar casein concentrate (MCN), calcium caseinate (CaC) and sodium caseinate (NaC). Results revealed that at high protein concentrations (0.5 %-2.5 %), MCN, CaC and NaC exhibited similar interfacial behaviour as well as unfolding rate constants (k 1 ) of 3.11-3.41 × 10-4 (s-1), 2.96-3.35 × 10-4 (s-1) and 2.75-3.27 × 10-4 (s-1), respectively. The interfacial layer formed was dominated by non-micelles, and microscopic images revealed the thickness of the interfacial layer to be 10-20 nm. By contrast, at low concentrations, the differences in the slope of E-π curves and k 1 indicated that the micelle content of casein affects protein interfacial behaviour and properties and that micellar casein is involved in the formation of the interfacial layer. The formation of large numbers of droplets during emulsion preparation results in a similar low concentration environment. Cryo-TEM showed adsorption of micellar casein in all three casein-stabilised emulsions, and the amount of adsorption was proportional to the micelle content. NaC has faster adsorption and rearrangement rates due to fewer micelles and more non-micelles, so that NaC forms smaller droplets and more stable emulsions than those formed by MCN and CaC within the range of 0.5 % to 2.0 %.

Abstract:4-[4(-(4(-decyloxy phenylazo) naphthloxy] butyl trimethylammonium bromide can form a stable monolayer on the subphase of an aqueous sodium dextran sulfate solution. Infrared transmission, reflection-absorption and ultraviolet-visible spectra have been measured for the one- and multi-layer LB films to study the molecular orientation and aggregation in the films. It was found that the title compound does not form molecular aggregation in the LB films. The alkyl tail is nearly perpendicular to the substrate surface and adopts largely trans-zigzag conformation.

Abstract:The Langmuir-Blodgett (LB) technology is an excellent method for the preparation of ultrathin films with a controlled thickness and well-defined molecular order. The materials of choice for constructing the LB films cover a wide range from low molecular weight amphiphiles to performed polymers and biological compounds. At the same time, with the combination of materials science and life science, it is more important and valuable to study the structure of LB films made by protein and lipid and their relation. Then in order to obtain good biological LB films, available film-forming conditions must be obtained. Therefore, it is important and essential to study the behavior and states of bovine serum albumin-phosphatidylcholine (BSA-PC) monolayer at air-water interface. The target of using BSA-PC is to form a measure of preparing biological molecular LB films so that we can simulate biological environment and make further investigation on interactive mechanism and application of biological molecular films. Abstract:In response to the problem of complex interaction between oil and water in the oil-water interface, especially heavy oil and water, this study investigated the effects of complex surfactants on the interaction of two phases and their aggregation characteristics by molecular dynamics simulation. The results showed that increasing the content of sodium lauryl polyether carboxylate (AEC-9Na) was beneficial to the coordination between it and alkyl glycoside (APG-10), improved the interfacial activity, and enhanced the interfacial stability of the composite system, and the best effect was achieved when AEC-9Na:APG-10 = 8:2. The thickness of the oil and water film on the oil-water interface was irregular. When the concentration of AEC-9Na was lower than that of APG-10, the total thickness of the interfacial film (ttotal) first increased. When the content of AEC-9Na is higher, a large number of sodium ions were adsorbed near the -COO- group of AEC-9Na, which will polarize out of the hydration layer structure and attract water molecules from the second hydration layer on the heavy oil surface to the first hydration layer through electrostatic interaction. Then, the thickness of the interface film was compressed, and the interface film was reduced. When the ratio increased to 10:0, the oil and water phase competed to adsorb surfactant molecules, and the headgroup tended to lay on the interface. Moreover, the hydrophilicity of the surfactant layer was weakened, and the thickness of the water film decreased. The distribution of surfactant was looser than 8:2, the light components of heavy oil molecules (saturated and aromatic hydrocarbons) entered the gap between surfactants in large quantities, and the hydrophobic tail chain tended to be laid on the oil-water interface. The oleophilicity of the surfactant layer increased, and the thickness of the oil film remarkably increased, so the total thickness of the interface film increased again.

Interfacial multilayer self-assembly of protein and polysaccharides: Ultrasonic regulation, stability and application in delivery lutein

Abstract:In this study, the layer-by-layer adsorption behavior of sodium caseinate, pectin, and chitosan on the oil-water interface was illustrated using multi-frequency ultrasound. We investigated the impact of ultrasound on various factors, such as particle size, zeta potential, and interfacial protein/polysaccharide concentration. It was observed that ultrasound has significantly decreased droplet size and increased the surface area at the interface, hence promoting the adsorption of protein/polysaccharide. In the sonicated multilayer emulsion, the concentrations of interface proteins, pectin, and chitosan increased to 84.82 %, 90.49 %, and 83.31 %, respectively. The findings of the study indicated that the application of ultrasonic treatment had a significant impact on the emulsion's surface charge and the prevention of droplet aggregation. As a result, the stability of the emulsion system, including its resistance to salt, temperature, and storage conditions, has been significantly improved. Moreover, the emulsion showed an increase in the retention rate of lutein by 21.88 % after a high-temperature water bath and by 19.35 % after UV irradiation. Certainly, the multilayer emulsion treated with ultrasound demonstrated a superior and prolonged releasing behavior. These findings demonstrated the suitability of the ultrasound treatment for the preparation of emulsions to deliver bioactive compounds.

Abstract:Biopolymer electrostatic complexes are popular Pickering stabilizers whose structures greatly affect their interfacial properties. This study comprehensively demonstrated the interfacial adsorption and assembly of dissolved octenyl succinic anhydride (OSA) starch (OSA-D)/chitosan (CS) electrostatic complexes with different structures through complementary characterization methods. We found that compared with single-component systems, OSA-D/CS complexes exhibited significantly increased wetting stability and adsorption rate to the interface, which was reinforced by molecular dynamics simulations. Their soft structures and the entanglement of molecular chains led to the formation of thick and highly viscoelastic multilayer adsorbed films, which greatly resisted deformation against shearing forces. The adsorption and assembly of the complexes were strongly influenced by OSA-D/CS ratios and pH, which could be related to the different interfacial interaction strengths. Overall, the electrostatic complexation, structural characteristics, and interfacial properties of OSA-D/CS complexes were well related, thereby providing valuable information for the regulation of controlled interfaces and bulk system properties.

Abstract:Lamellar liquid crystals comprising oil, water and surfactant(s) were formulated and analysed in order to examine how these materials responded to the inclusion of inorganic nanoparticles, in terms of their structural and rheological characteristics. Lamellar phases were formed from mixtures of water, para-xylene and Triton X-100, and analysis was performed via small-angle neutron scattering (SANS), polarising light microscopy (PLM), and amplitude and viscosity sweeps. The partial replacement of Triton X-100 with oleic acid appeared to cause an increase in bilayer thickness, attributed to less efficient packing of the different molecules. Addition of oleic acid also appeared to cause both a loss in lamellar repeat ordering, attributed to heterogeneity of the bilayers, and a rise in long range order, potentially caused by the stiffer bilayers. Adding silica nanoparticles of different size and surface chemistry caused a stiffening of the samples at the expense of a longer-range lamellar repeat order. This strengthening is attributed to aggregation at the domain boundaries, and it was found that hydrophobic particles tended to form stronger aggregates while for larger particles (20 nm as opposed to 10 nm) aggregation was apparently reversible. These results give a more comprehensive understanding of how to reliably control the structural and rheological properties of lamellar liquid crystals, and emphasise the importance of the size and surface chemistry of any inclusions, for applications in cosmetics, drug delivery, and microfluidics.

Abstract:Dissipative particle dynamics simulations are applied to investigate the monolayer and multilayer film formations on different solid substrates by physical deposition. The influences of the polymer concentration, the polymer chain length, the solvent quality, and the interactions between the polymer solution and the solid substrate surface on the film formation dynamics and the mechanism are studied in detail. The results are analyzed in terms of the thickness and the shape of the deposited film, the kinetics of phase separation in the polymer solution, and the contact angle formed between the polymer aggregations and the substrate surface. Moreover, we suggest two strategies, designing a deposition process analogous to "chemical titration" and physically blocking interlayer diffusion by a simple crosslinked network barrier, to deposit the compact monolayer and multilayer films with better quality, respectively.
