Abstract:The design and orderly layered co-immobilization of multiple enzymes on resin particles remain challenging. Herein, the SpyTag/SpyCatcher binding pair was fused to the N-terminus of an alcohol dehydrogenase (ADH) and an aldo-keto reductase (AKR), respectively. A non-canonical amino acid (ncAA), i.e. p-azido-L-phenylalanine (p-AzF), as the anchor for covalent bonding enzymes, was genetically inserted into pre-selected sites in the AKR and ADH. Employing the two bioorthogonal counterparts of SpyTag/SpyCatcher and azido-alkynyl for the immobilization of AKR and ADH enabled the sequential dual-enzyme coating on porous microspheres. The ordered dual-enzyme reactor was subsequently used to asymmetrically synthesize (S)-1-(2-chlorophenyl) ethanol from the corresponding prochiral ketone, enabling the in situ regeneration of NADPH. The reactor exhibited a high catalytic conversion of 74% and good reproducibility, retaining 80% of its initial activity after six cycles. The product had an enantiomeric excess (e.e.) of 99.9% that was maintained in each cycle. Additionally, the double-layer immobilization method significantly increased the enzyme loading capacity, approximately 1.7 times greater than that of the traditional single-layer immobilization. More importantly, it simultaneously achieves both the purification and immobilization of multiple enzymes on carriers, thus providing a convenient approach to facilitate cascade biocatalysis.

Abstract:Sequential immobilization of multiple enzymes can not merely improve the enzymatic activity and enzyme stability but also facilitate the recovery for reuse, well imitating the natural enzyme complexes. Here, we developed a novel approach of preparing sequential multienzyme nanomachineries based on chemical crosslinking and protein assembly, and then characterized the nanodevice. In the preparation process, KgBDH-SC (SpyCatcher-fused (2R,3R)-2,3-butanediol hydrogenase) was firstly immobilized onto the functionalized silicon dioxide nanoparticles with glutaraldehyde, and then FDH-ST (SpyTag-fused formate hydrogenase) was immobilized by the spontaneous protein reaction between SpyTag and SpyCatcher to achieve the sequential co-immobilized enzymes FDH-ST-SC-Kg2, which can asymmetrically reduce 2-hydroxyacetophenone to (R)-1-phenyl-1,2-ethanediol by cofactor regeneration. In comparison to the mixture of free enzymes, the sequential co-immobilized enzymes created under the design conditions performed around 50% activity recovery. Furthermore, FDH-ST-SC-Kg2 presented wider pH tolerance and greater organic solvents tolerance than the mixture of free enzymes as well as retained 51.7% of overall activity after being recycled eight times. Finally, the sequential co-immobilized enzyme enabled the biosynthesis of (R)-1-phenyl-1,2-ethanediol from 80 mM 2-hydroxyacetophenone in the phosphate buffer reaction system, with 87.4% yield and over 99% optical purity.

Abstract:Two non-canonical amino acids (ncAAs) with bio-orthogonal reactive groups, namely, p-azido-L-phenylalanine (p-AzF) and p-propargyloxy-L-phenylalanine (p-PaF), were genetically inserted into an aldo-keto reductase (AKR) and an alcohol dehydrogenase (ADH), respectively, at two preselected sites for each enzyme. The variants were expressed in the genome recoded bacterium Escherichia coli MG1655. Supernatants of the individual cell lysates were subsequently mixed to produce orderly combi-crosslinked enzymes (O-CLEs) of AKR and ADH by co-polymerization of the two variants through their reactive bio-orthogonal groups. The O-CLEs exhibited high catalytic efficiency (Kcat/KM=106±0.342 S−1·mM−1) that was 4.3 times that of site-specific combi-crosslinked enzymes (S-CLEs), and 28.0 times that of random combi-cross-linked enzyme aggregates (combi-CLEAs) in the asymmetric cascade synthesis of (R)-1-(2-chlorophenyl) ethanol. The S-CLEs and CLEAs were produced using dibenzocycloocta-4a,6a-diene-5,11-diyne (DBA) and glutaraldehyde as the crosslinking agent, respectively. Moreover, high activities and selectivity (ee >99.99%) were maintained at high substrate concentrations in prolonged operation.

Abstract:The field of enzyme cascades in limited microscale or nanoscale environments has undergone a quick growth and attracted increasing interests in the field of rapid development of systems chemistry. In this study, alcohol dehydrogenase (ADH), lactate dehydrogenase (LDH), and mesoporous silica nanoparticles (MSN) immobilized nicotinamide adenine dinucleotide (NAD+) were successfully immobilized on the zeolitic imidazolate frameworks (ZIFs). This immobilized product was named ZIF@ADH/NAD-MSN/LDH, and the effect of the multi-enzyme cascade was studied by measuring the catalytic synthesis of lactic acid. The loading efficiency of the enzyme in the in-situ co-immobilization method reached 92.65%. The synthesis rate of lactic acid was increased to 70.10%, which was about 2.82 times that of the free enzyme under the optimal conditions (40 °C, pH = 8). Additionally, ZIF@ADH/NAD-MSN/LDH had experimental stability (71.67% relative activity after four experiments) and storage stability (93.45% relative activity after three weeks of storage at 4 °C; 76.89% relative activity after incubation in acetonitrile-aqueous solution for 1 h; 27.42% relative activity after incubation in 15% N, N-Dimethylformamide (DMF) solution for 1 h). In summary, in this paper, the cyclic regeneration of coenzymes was achieved, and the reaction efficiency of the multi-enzyme biocatalytic cascade was improved due to the reduction of substrate diffusion.

Abstract:A fully automated method has been developed for segmentation of four different structures in the neonatal brain: white matter (WM), central gray matter (CEGM), cortical gray matter (COGM), and cerebrospinal fluid (CSF). The segmentation algorithm is based on information from T2-weighted (T2-w) and inversion recovery (IR) scans. The method uses a K nearest neighbor (KNN) classification technique with features derived from spatial information and voxel intensities. Probabilistic segmentations of each tissue type were generated. By applying thresholds on these probability maps, binary segmentations were obtained. These final segmentations were evaluated by comparison with a gold standard. The sensitivity, specificity, and Dice similarity index (SI) were calculated for quantitative validation of the results. High sensitivity and specificity with respect to the gold standard were reached: sensitivity >0.82 and specificity >0.9 for all tissue types. Tissue volumes were calculated from the binary and probabilistic segmentations. The probabilistic segmentation volumes of all tissue types accurately estimated the gold standard volumes. The KNN approach offers valuable ways for neonatal brain segmentation. The probabilistic outcomes provide a useful tool for accurate volume measurements. The described method is based on routine diagnostic magnetic resonance imaging (MRI) and is suitable for large population studies.

Abstract:Chiral alcohols are not only important building blocks of various bioactive natural compounds and pharmaceuticals, but can serve as synthetic precursors for other valuable organic chemicals, thus the synthesis of these products is of great importance. Bio-catalysis represents one effective way to obtain these molecules, however, the weak stability and high cost of enzymes often hinder its broad application. In this work, we designed a biological nanoreactor by embedding alcohol dehydrogenase (ADH) and glucose dehydrogenase (GDH) in metal-organic-framework ZIF-8. The biocatalyst ADH&GDH@ZIF-8 could be applied to the asymmetric reduction of a series of ketones to give chiral alcohols in high yields (up to 99 %) and with excellent enantioselectivities (>99 %). In addition, the heterogeneous biocatalyst could be recycled and reused at least four times with slight activity decline. Moreover, E. coli containing ADH and GDH was immobilized by ZIF-8 to form biocatalyst E. coli@ZIF-8, which also exhibits good catalytic behaviours. Finally, the chiral alcohols are further converted to marketed drugs (R)-Fendiline, (S)-Rivastigmine and NPS R-568 respectively.

Abstract:Abstract Co-immobilization of cells and enzymes is often essential for the cascade biocatalytic processes of industrial-scale feasibility but remains a vast challenge. Herein, we create a facile co-immobilization platform integrating enzymes and cells in covalent organic frameworks (COFs) to realize the highly efficient cascade of inulinase and E. coli for bioconversion of natural products. Enzymes can be uniformly immobilized in the COF armor, which coats on the cell surface to produce cascade biocatalysts with high efficiency, stability and recyclability. Furthermore, this one-pot in situ synthesis process facilitates a gram-scale fabrication of enzyme-cell biocatalysts, which can generate a continuous-flow device conversing inulin to D-allulose, achieving space-time yield of 161.28 g L −1 d −1 and high stability (remaining >90% initial catalytic efficiency after 7 days of continuous reaction). The created platform is applied for various cells (e.g., E. coli , Yeast) and enzymes, demonstrating excellent universality. This study paves a pathway to break the bottleneck of extra- and intracellular catalysis, creates a high-performance and customizable platform for enzyme-cell cascade biomanufacturing, and expands the scope of biocatalysis process intensification.

Abstract:Integrating nanozymes with natural enzymes to form cascade reactions is one of the most promising ways to develop biocatalysts with versatile performance; however, the applicability of the cascade is typically hampered by the instability of enzymes and the hindrance of mass transfer in the host environment. Utilizing amorphous ZIF-90 (aZIF-90) as a host material, herein, we have reported a one-pot way to encapsulate glucose oxidase (GOx) and magnetic nanoparticles (MNP) to form GOx/MNP@aZIF-90. We reasoned that the amorphous structure of ZIF-90 not only provides a protected environment to confine the cascade reaction but also generates mesopores and internal voids to improve the performance of the enzymatic cascade. The catalytic activity of aZIF-90 was almost 4 times higher than that of crystalline composites, and the residual activity was higher than 80% after being stored for 9 days. This is the first time that GOx and MNP were simultaneously confined in aZIF-90 with mesopores, which suggested that an amorphous metal-organic framework is promising in the development of an enzymatic cascade.

Abstract:The integration of multimodal chemical/biocatalysis technology has provided a broad prospect in the efficiency of cascade reactions for a long time. Inspired by the intricate structure and superior efficiency of natural multi-enzyme systems, it is attractive for developing the next generation of high-performance orderly enzyme cascade biological platforms, combined with the catalase-like activity and immobilization properties of nanozymes. In our simple and convenient design, a biomimetic multi-enzyme hybrid nanocluster with excellent catalytic activity was synthesized by in-situ adsorption and immobilization of biological enzyme on nanozyme Fe 3 O 4 @OA. The combination of nanozyme Fe 3 O 4 @OA with single enzyme (concatenated alditol oxidase, catAldO) or double enzymes (catAldO and concatenated dihydroxy-acid hydrolyase, catDHAD) enjoys outstanding synergistic biocatalytic ability. Nanozyme Fe 3 O 4 @OA alternatives catalase to catalyze H 2 O 2 cycle and regenerate O 2 , which implements the internal circulation of O 2 → H 2 O 2 → O 2 system and accelerates the conversion of glycerol. It realized the high efficiency synergistic catalysis of nanozyme and biological enzyme successfully. In contrast to simple biocatalyst mixtures in solution, the prepared multienzyme hybrid resulted in the obvious improvements in activity for tandem reactions. The cascade bioactivity of the multienzyme complex was enhanced by the synergistic effect of the immobilization of nanozyme Fe 3 O 4 @OA, the opening of substrate channel between the enzyme and nanozyme, and the effective reduction of the transport distance among intermediate metabolites. In this study, it highlights the superiority of synergistic bioreactor integrated with nanoclusters contain Fe 3 O 4 @OA nanozyme and biological enzyme, realizing rapid recovery and multiple reusability of nanoclusters, and suggesting great application potential in multi-enzyme synergistic cascade catalysis.

Abstract:The encapsulated enzyme system by metal–organic frameworks exhibited great potential in pharmaceuticals, enzyme fuel cells, and biocatalysis. However, the enzymatic activity and overall catalytic efficiency are severely hampered due to their enzyme leaching and causal enzyme location. Herein, for the first time, horseradish peroxidase (HRP) and glucose oxidase (GOx) used as model enzymes were cross-linked by rationally designed DNA scaffold network, and the multienzyme system has been successfully encapsulated into zeolite imidazolate framework-8 (ZIF-8). The prepared encapsulation multienzyme can efficiently suppress enzyme leaching from the MOFs skeleton, and the leaching amount was more than 10-times lower than the traditional MOFs encapsulation strategy. The well-tailored proximity and colocalization of cascade enzymes by the DNA scaffold network obviously improved the overall catalytic efficiency, enzymatic activity, and kinetic performance. The K m and k cat/K m for encapsulation multienzyme was 13.8 mM and 3.47 s−1 mM−1, which was 0.27-fold lower and 9.9-fold higher than those of free enzymes, respectively, indicating a better substrate affinity. Furthermore, the multienzyme system exhibited excellent stability and reusability compared with free enzymes and control groups, which could preserve 95% initial activity at 50 °C after 75 min incubation and more than 87% of the original activity after 14 cycles. Finally, the immobilized multienzyme exhibited enhanced performance to detect low concentration of glucose (0.4 µM) with excellent selectivity. The outcome indicated that the encapsulated multienzyme could be extended for broad practical applications in biotechnology, biocatalysis, and biomedical engineering.
Biocatalytic cascades driven by enzymes encapsulated in metal–organic framework nanoparticles

Abstract:Biocatalytic transformations in cells, such as enzyme cascades, involve complex networks proceeding in spatially confined microenvironments. Here, inspired by nature, we demonstrate effective biocatalytic cascades by the encapsulation of two or three enzymes, or enzyme/cofactor components, in zeolitic imidazolate framework-8 metal–organic framework nanoparticles (ZIF8-NMOFs) that act as nanoreactors. The integration of the two-enzyme system (glucose oxidase and horseradish peroxidase) or three-enzyme system (β-galactosidase, glucose oxidase and horseradish peroxidase) in the NMOFs leads to 7.5-fold and 5.3-fold enhancements in the activity of the catalytic cascades, respectively, compared with the bulk mixture of the catalysts in solution. In addition, the encapsulation of alcohol dehydrogenase, NAD+–polymer and lactate dehydrogenase in the NMOFs yields a coupled biocatalytic cascade involving coupled NAD+-dependent enzymes, leading to the catalytic reduction of pyruvic acid to lactic acid by ethanol.

Abstract:Nanozymes are a class of nanomaterials with enzyme-like activities that have been extensively studied for their possibilities and superiorities over natural enzymes. Intelligent design and fabrication of a nanozyme platform with highly efficient and stable tandem enzyme characteristics is necessary. Herein, a nanozyme cascade promoting bioplatform based on protein/bimetallic nanoparticles attached to ZIF-derived multilayer N-doped carbon nanosheets is proposed. Due to the carbon-based large surface area and solitary pairs of nitrogen electrons, the metal could be riveted to the carbon-based surface and reduced in situ to nanoparticles. Simultaneously, the addition of BSA has made it possible to establish a biological platform. The obtained BSA-PtAu@CNS has a linear range of 2 × 10-6 to 2 × 10-3 M for H2O2 and apparent dynamic Km of 0.005 × 10-3 M, which shows stronger affinity with H2O2 than HRP. The nanohybrid could adapt to severe reaction environment and enable prominent tandem dual-enzyme activity for glucose sensing. As a result, the cascade nanozyme bioplatform based on BSA-PtAu@CNS is accomplished and provides insights for subsequent biocompatible nanomaterials for use in medical and diagnostic applications.

Abstract:Abstract Biocatalytic transformations in living organisms, such as multi-enzyme catalytic cascades, proceed in different cellular membrane-compartmentalized organelles with high efficiency. Nevertheless, it remains challenging to mimicking biocatalytic cascade processes in natural systems. Herein, we demonstrate that multi-shelled metal-organic frameworks (MOFs) can be used as a hierarchical scaffold to spatially organize enzymes on nanoscale to enhance cascade catalytic efficiency. Encapsulating multi-enzymes with multi-shelled MOFs by epitaxial shell-by-shell overgrowth leads to 5.8~13.5-fold enhancements in catalytic efficiencies compared with free enzymes in solution. Importantly, multi-shelled MOFs can act as a multi-spatial-compartmental nanoreactor that allows physically compartmentalize multiple enzymes in a single MOF nanoparticle for operating incompatible tandem biocatalytic reaction in one pot. Additionally, we use nanoscale Fourier transform infrared (nano-FTIR) spectroscopy to resolve nanoscale heterogeneity of vibrational activity associated to enzymes encapsulated in multi-shelled MOFs. Furthermore, multi-shelled MOFs enable facile control of multi-enzyme positions according to specific tandem reaction routes, in which close positioning of enzyme-1-loaded and enzyme-2-loaded shells along the inner-to-outer shells could effectively facilitate mass transportation to promote efficient tandem biocatalytic reaction. This work is anticipated to shed new light on designing efficient multi-enzyme catalytic cascades to encourage applications in many chemical and pharmaceutical industrial processes.

Abstract:Protein immobilization is critical to utilize their unique functions in diverse applications. Herein, we report that orthogonal peptide-protein chemistry enabled multilayer construction can facilitate the incorporation of various folded structural domains, including calmodulin in different states, affibody, and dihydrofolate reductase (DHFR). An extended conformation is found to be the most advantageous for steady film growth. The resulting protein thin films exhibit sensitive and selective responsive behaviors to biosignals, such as Ca2+, trifluoperazine, and nicotinamide adenine dinucleotide phosphate (NADPH), and fully maintain the catalytic activity of DHFR. The approach is applicable to different substrates such as hydrophobic gold and hydrophilic silica microparticles. The DHFR enzyme can be immobilized onto silica microparticles with tunable amounts. The multilayer setup exhibits a synergistic enhancement of DHFR activity with increasing numbers of bilayers and also makes the embedded DHFR more resilient to lyophilization. Therefore, this is a convenient and versatile method for protein immobilization with potential benefits of synergistic enhancement in enzyme performance and resilience.

Abstract:Utilizing covalent organic frameworks (COFs) as a hypotoxic and porous scaffold to encapsulate enzyme (enzyme@COF) has inspired numerous interests at the intersection of chemistry, materials, and biological science. In this study, we report a convenient scheme for one-step, aqueous-phase synthesis of highly crystalline enzyme@COF biocatalysts. This facile approach relies on an ionic liquid (2 μL of imidazolium ionic liquid)-mediated dynamic polymerization mechanism, which can facilitate the in situ assembly of enzyme@COF under mild conditions. This green strategy is adaptive to synthesize different biocatalysts with highly crystalline COF "exoskeleton", as well evidenced by the low-dose cryo-EM and other characterizations. Attributing to the rigorous sieving effect of crystalline COF pore, the hosted lipase shows non-native selectivity for aliphatic acid hydrolysis. In addition, the highly crystalline linkage affords COF "exoskeleton" with higher photocatalytic activity for in situ production of H2O2, enabling us to construct a self-cascading photo-enzyme coupled reactor for pollutants degradation, with a 2.63-fold degradation rate as the poorly crystalline photo-enzyme reactor. This work showcases the great potentials of employing green and trace amounts of ionic liquid for one-step synthesis of crystalline enzyme@COF biocatalysts, and emphasizes the feasibility of diversifying enzyme functions by integrating the reticular chemistry of a COF.

Abstract:Plasma membranes of lymphocytes obtained from pig mesenteric lymph nodes were reconstituted after solubilization with bile salts. The proportion by weight of immunoglobulin in the reconstituted membrane was no greater than about 5-10% of that in the original membrane. Possible reasons for the low reincorporation of immunoglobulin are discussed.

Abstract:Encapsulating enzymes within metal-organic frameworks has enhanced their structural stability and interface tunability for catalysis. However, the small apertures of the frameworks restrict their effectiveness to small organic molecules. Herein, we present a green strategy directed by visible linker micelles for the aqueous synthesis of MAF-6 that enables enzymes for the catalytic asymmetric synthesis of chiral molecules. Due to the large pore aperture (7.6 Å), double the aperture size of benchmark ZIF-8 (3.4 Å), MAF-6 allows encapsulated enzyme BCL to access larger substrates and do so faster. Through the optimization of surfactants' effect during synthesis, BCL@MAF-6-SDS (SDS = sodium dodecyl sulfate) displayed a catalytic efficiency (Kcat/Km) that was 420 times greater than that of BCL@ZIF-8. This biocomposite efficiently catalyzed the synthesis of drug precursor molecules with 94-99% enantioselectivity and nearly quantitative yields. These findings represent a deeper understanding of de novo synthetic encapsulation of enzyme in MOFs, thereby unfolding the great potential of enzyme@MAF catalysts for asymmetric synthesis of organics and pharmaceuticals.

Abstract:In recent decades, enzyme-based biocatalytic systems have garnered increasing interest in industrial and applied research for catalysis and organic chemistry. Many enzymatic reactions have been applied to sustainable and environmentally friendly production processes, particularly in the pharmaceutical, fine chemicals, and flavor/fragrance industries. However, only a fraction of the enzymes available has been stepped up towards industrial-scale manufacturing due to low enzyme stability and challenging separation, recovery, and reusability. In this context, immobilization and co-immobilization in robust support materials have emerged as valuable strategies to overcome these inadequacies by facilitating repeated or continuous batch operations and downstream processes. To further reduce separations, it can be advantageous to use multiple enzymes at once in one pot. Enzyme co-immobilization enables biocatalytic synergism and reusability, boosting process efficiency and cost-effectiveness. Several studies on multi-enzyme immobilization and co-localization propose kinetic advantages of the enhanced turnover number for multiple enzymes. This review spotlights recent progress in developing versatile biocatalytic cascade systems by multi-enzyme co-immobilization on environmentally friendly biopolymers and nanostructured materials and their application scope in the chemical and biotechnological industries. After a succinct overview of carrier-based and carrier-free immobilization/co-immobilizations, co-immobilization of enzymes on a range of biopolymer and nanomaterials-based supports is thoroughly compiled with contemporary and state-of-the-art examples. This study provides a new horizon in developing effective and innovative multi-enzymatic systems with new possibilities to fully harness the adventure of biocatalytic systems.

Abstract:The strategies based on enzyme immobilization into/on metal-organic frameworks (MOFs) have been widely developed, but they still face some challenges in practical applications, such as limited recycle times, low loading efficiency. In this work, aspartate aminotransferase (AspAT), branched-chain aminotransferase (BCAT), polyvi-nyl pyrrolidone/aspartate (PA), cobalt ions and sodium 2-amino terephthalate were one-pot mixed in water at room temperature, and self-assembled to encapsulate the enzymes into cobalt-amino terephthalate framework (CAF) in 15 min. Compared with free enzymes, the biocomposite (A&B&PA@CAF) exhibited superior perfor-mances such as high encapsulation efficiency (55%), reusability (>= 12), and resistance capacity to harsh con-ditions. Meanwhile, the activity recovery (AR) value (59.4%) of the biocomposite without adding polyvinyl pyrrolidone and aspartate was almost half of that (108.2%) of A&B&PA@CAF, and this enhancement was further demonstrated to be from Asp action via 1H-NMR and UV-Vis. To investigate the general applicability of this approach, the other aminases were also de novo immobilized with all their AR values reached more than 117%, even 180% for omega-transaminase. This study provided a sustainable and green approach for clean production of the significant chiral amine compounds.
