Abstract:Abstract The hair structure of the Lutrinae has been analysed to see if the different species could be identified using hair characteristics and also to determine if the hair characteristics have an adaptive value or are influenced by the phylogeny of the group. The macroscopic (colour, morphology, length, width) and microscopic (cross-section, medulla, cuticle) characteristics of the primary hairs (PHs), which are more species-specific, and also the length and width of the secondary hairs of each of the 13 known otter species were examined. The PHs of the different species showed many similar characteristics, but also some significant divergences. It was not possible to identify every species confidently because of great similarities in medulla and cuticle structure. Apparently, the macroscopic and microscopic hair structure in the Lutrinae has been influenced by both phylogenetic relationships and adaptive pressures. An influence of the climate and of the association to water could be recognized.

Abstract:Guard hairs were collected from four live domesticated ungulate species and shed guard hairs of six wild ungulate species from zoo captive animals from five individuals each. Photographic reference was prepared showing analytic features of hair characteristics. Study results were analysed and cuticle and medulla patterns were identified along with pigmentation features from the literature available for wild and domestic ungulates from India and abroad. Clear and easily distinguishable morphological characters of hair medulla and cuticle were used in the present study. Scat analysis of big cats used in this study is easy, speedy and efficient which can be used in routine investigations related to wildlife, crime forensics as well as human animal conflicts by studying carnivore feeding habits. In a majority of the animal species, the distal part of the hair showed maximum variation from the rest of the hair portions. The cuticle scales were imbricate in all tested animals. Scale position in almost all the tested animals was transversal except in goat (proximal part and medial part) and mouse deer (Distal part). Majority of the species showed smooth margins at proximal and medial part. Whereas the distal part scale margin was crenate and rippled in appearance the proximal part and medial part of hair of the majority of sampled animals showed a regular wave -type scale pattern whereas the distal part of hair showed irregular wave-type scale pattern in dominance. The composition of the medulla was multicellular in all the sampled deer species. Only the cow calf’s hair medulla was unicellular and uniseriate in appearance. A comparison of the hair of the domestic pig with that of the wild boar and gaur hair with that of cow calf and buffalo calf hair was made for the first time in the present study. Similarly goat hair morphology can also be differentiated from other cervids in this study. Medulla and cuticle characters in combination with each other can help differentiate wild ungulate species from the domestic ones since these wild ungulate species are frequently involved in hunting crime investigations. Therefore, the photographic reference presented in this study can be used in wildlife forensic science as well as predator diet analysis as an appropriate reference for prey species identification.

Abstract:Weak degree of hair differentiation and microstructure of hair and whiskers of Otohylomys megalotis are similar to those in Hylomys suillus and Neotetracus sinensis: there are spear-shaped lead hairs and zigzag guard hairs of three orders, downy hairs are missing. In O. megalotis: 1) hairs are longer than those of other gymnures; 2) thin convoluted hair bases bound up contributing to the formation of the inert layer of air near the surface of the skin, improves the thermal insulation properties of hair in the absence of downy hairs; 3) strength in thinnest areas of the shaft (at its base and excesses) is provided by thickening of its cuticular scales, the special interconnection between cuticle and cortex, and cruciform layout of medulla discs in these places; 4) the pineal cuticular ornament of hair bases is characteristic of all three species of gymnures and resembles that of other insectivores, as well as of some marsupials and carnivores that reflects similar hair adaptations to the habitats; 5) for the first time discovered specialized pyramidal medulla of vibrissae, stiffening a shaft that is necessary for effective transfer of mechanical impulses to nerve cells of vibrissae follicle and functioning of whiskers as a tactile organ; 6) a long proboscis with well-developed nasal vibrissae and also numerous long whiskers on muzzle, neck, wrists and forearms are important and effective for the O. megalotis orientation in complex terrain karst habitats.
A Descriptive Study of Hair Morphology of Three Carnivore families (Canidae, Ursidae and Herpestidae)

Abstract:Aim: To study the morphological characteristics of dorsal guard hair of five carnivoran species native to Tamil Nadu for the creation of an identification key for application in wildlife forensics. Study Design: The experiment was designed to observe the physical characters of each hair strand and measure widely used morphological parameters from native species of the state. The results were compared with previous findings to validate the differences and similarities with our study. Place and Duration of Study: Advanced Institute for Wildlife Conservation (AIWC), Tamil Nadu Forest Department, Vandalur, Chennai, Tamil Nadu, between August 2020 and September 2021. Methodology: Hair samples were collected for golden jackal, Indian fox, dhole, sloth bear and Indian grey mongoose. Hair characters such as scale pattern, scale distance and scale margin, medulla structure, cortex thickness, medulla thickness, medullary index and cross-sectional structure were observed. Results: The hair length of golden jackal, Indian fox, dhole, sloth bear and Indian grey mongoose were as follows 44.94 ± 2.24 mm, 29.70 ± 1.51 mm, 29.60 ± 0.97 mm, 83.83 ± 2.93 mm, 54.60 ± 2.03. The cuticular characters showed few variations compared to previous studies. The medullary structure was wide medulla with vacuoles for all three canid species while sloth bear has a narrow simple unbroken medulla and Indian grey mongoose had wide medulla with lattice. The medullary index for golden jackal, Indian fox, dhole, sloth bear and Indian grey mongoose were 0.73 ± 0.05 (S.D.), 0.77 ± 0.071 (S.D.), 0.66 ± 0.06 (S.D.), 0.18 ± 0.08 (S.D.) and 0.63 ± 0.05 (S.D.) respectively. Conclusion: The morphometric characters of hair showed variation in scale pattern, scale margin, scale distance and medullary index when compared to the previous studies. The research was successful in creating a taxonomic key for identifying the five mammalian species from their guard hair.

Abstract:Mammalian hair, as a key feature related to endothermy, physical protection, and camouflage, has a long geohistorical record that dates back even to the Mesozoic. However, the limited information on the taphonomy of hair hinders the understanding of its evolutionary significance in deep time. To address this, we conducted taphonomic experiments to simulate the transformation process of black hair. The changes in the morphological structure, microscopic characteristics, and chemical composition of hair during the simulated fossilization process were documented by scanning electron microscopy, Fourier transform infrared (FTIR) spectroscopy, and Raman spectroscopy. Our results revealed that damage to the morphology of hair began at a mild temperature (150–175°C), and hair transformed into a fluid-like mass at a relatively high temperature (200°C). In addition, the morphological structure of melanosomes had relatively greater preservation potential than keratins did, and the microscopic features of melanosomes were persistently stable. The Raman and FTIR spectra indicated that the characteristic melanin and keratin peaks changed as the temperature increased. Although the chemical signals were not lost completely, the variations in the characteristic amide bands in the FTIR spectra indicate that the keratins severely degraded at high temperatures (>200°C), whereas the characteristic melanin bands in the Raman spectra suggested that melanin signals were still recognizable, even at high temperatures (>200°C). These results were confirmed by principal component analysis (PCA) and principal component analysis linear discriminant analysis (PCA–LDA). In addition, the Raman signals of all the samples in the simulated experiments were distinguished from the signals of common geological carbonaceous materials (humic acid), which suggested that Raman spectroscopy can be used to detect melanin in fossils.

Abstract:Mylodon darwinii is an extinct terrestrial species of sloth with a wide fossil record in South America, the record of the skin and hairs found from "La Cueva del Milodón", Chile. More than a century after the first studies on its hair, we aimed to redescribe the morphological characteristics of the hairs of M. darwinii by applying modern protocols of trichology and microscopy. Hair patterns were also compared with those of two other extinct species of sloths available in the literature. Hairs from mummified specimen housed in Museo de La Plata (Argentina) were analyzed through light and scanning electron microscopy to identify their cuticular and medullary patterns. Guard hairs lack medulla and have a wavy cuticle with irregular scale orientation, while underhairs have an amorphous cuticle pattern and no medulla. Hence, the former ones help in the diagnosis of the hair morphology of the species, while the latter ones do not. Differences found in the final product of each microscopy technique are discussed and should be considered in future research. The outcomes presented herein are a step further toward the understanding of evolutionary processes within Xenarthra, providing important data for such a poorly studied group that has only been recorded in Central and South America.

Abstract:Abstract The hair cycling pattern differs between humans and mice: for the former it is mosaic, while the latter show a wave pattern. We focused on the presence or absence of synchronization of the hair cycle based on the distribution of Merkel cells. The three-dimensional distribution and number of Merkel cells in the skin at various sites were determined in guinea pigs and rats. Using ethylenediaminetetraacetic acid, various types of hair follicles were obtained from the facial, abdominal, nipple, back, tail, perianal and plantar skin. Epidermal sheets containing numerous hair follicles were immunostained using monoclonal murine antibody CK20 which is generally accepted to be a specific immunocytochemical marker for Merkel cells of various species. Merkel cells were absent in the trunk skin of guinea pigs and rats, in complete contrast to the situation in humans. In vibrissa and other hair follicles within specialized sensitive areas, Merkel cells were found in varying numbers. Some differences in their distribution and number were observed between guinea pigs and rats. The absence of Merkel cells in the hair follicles of the trunk skin of rats and guinea pigs implies that Merkel cells are not essential for the induction and promotion of hair growth but have some different functional significance in hair follicles.

The mammary hair of Monodelphis domestica and homology of the mammary pilosebacous unit

Abstract:The unitary mammary gland is a synapomorphy of therian mammals and is thought to have evolved from the pilosebaceous organ in the mammalian stem lineage from which the lactogenic patch of monotremes is also derived. One of the key lines of evidence for the homology of the nipple and the lactogenic patch is that marsupials have retained a transient hair associated with developing mammary glands. However, these structures have not been documented since the early 20th-century drawings of Ernst Bresslau. In this study, we examine the developing mammary organs of Monodelphis domestica and document the presence of mammary hairs in 12-week-old females, as well as their absence after 18 weeks of age. Histochemical staining for cystine confirms the structures as keratinized hairs. Milk ducts of both juvenile and adult nipples show a division between KRT18+ luminal epithelium and KRT14+ ACTA2+ myoepithelium. These patterns match those in eutherians and suggest a conserved ductal morphology and mechanism of milk expulsion. Finally, PTHLH, a peptide hormone which promotes homeotic transformation of hairy skin into hairless nipples in the mouse, was detected in the Monodelphis milk duct during the mammary hair stage, suggesting that the mutual exclusivity of "hairless nipple" and "hair" organ identity is derived in eutherian mammals. These results reveal shared characteristics of the M. domestica nipple with both the eutherian nipple and the pilosebaceous organ, consistent with the evolutionary derivation of the mammary gland from an ancestral hair organ via developmental individualization of pilosebaceous and mammary identities.
Guard hair micro-morphology of four non-human primates in Shasha Forest Reserve, Osun State, Nigeria

Abstract:Mammalian guard hairs have been used for their identification and have been proved useful in wildlife population surveys and trafficking. The qualitative and quantitative features of the dorsal guard hairs of four non-human primates (NHPs) from Shasha Forest Reserve were studied using standard procedures to determine their differences. The NHPs were mona (MM), putty-nosed (PNM), and white-throated (WTM) monkeys, and red-capped mangabey (RCM). The qualitative features determined were medulla pattern, and structure, scale margin distance and type, and scale pattern. The quantitative values studied were scale length and width, shaft diameter (μm), medulla diameter, and medulla index and fraction. Continuous medulla pattern was the only qualitative feature common to all the species. The MM hair had the highest recorded values for all morphological characteristics except shaft diameter and scale width in which PNM had the highest value of 323.00 ± 58.37 and 630.55 ± 213.95μm respectively. The medullary diameter, index and fraction, and shaft diameter were highly significant (P<0.001) among all the species. Post-hoc comparison showed that the medullary diameter, index, and fraction of hairs of the MM was significantly different (P≤0.001) from the other three NPHs. The shaft diameter of the MM was also significantly different from that of the PNM (P≤0.001), and WTM (P≤0.004). The scale width of RCM differed from PNM (P≤0.01), and WTM (P≤0.05). These empirically established morphological differences in guard hairs of the four NHPs in SFR would be useful in verifying their habitat occupancy and forensic evidence in case of illegal trafficking.
Characterization of Secondary Structure of Pig Hair Fiber Using Fourier-transform Infrared Spectroscopy

Abstract:<span>Hair or bristle is one of the key by-products of humane slaughter of pigs. In the present study, Fourier-transform infrared (FTIR) spectroscopy was used for examining protein secondary structure of hair fiber from different breeds of pigs (Ghungroo, Ninag Megha, Hampshire, and Duroc). Prominent amide I (1635–1648 cm<sup>−1</sup>) and amide II (1517–1528 cm <sup>−1</sup>) bands could be observed in the spectra. Differences in presence/absence of certain FTIR bands between breeds suggests influence of genetic background in the structure of hair fiber. Deconvolution resolved secondary structure of protein in amide I region of spectra into corresponding α-helix (38.0 ± 2.9%), β-sheets (32.2 ± 2.0%), β-turns (19.4 ± 3.1%) and unorganized structural components (10.3 ± 2.5%). Heating of fibers from 80°C to 120°C resulted in changes in amide regions and α-helix to β-sheet ratio of amide I band. Further, correlations were calculated between area under the curve of various FTIR bands and tensile properties of the fiber. Area of FTIR band at 1635 cm <sup>−1</sup> was positively correlated with tenacity, initial modulus, extensibility, and work of rupture (r = 0.30 to 0.39). In nutshell, the study reveals subtle differences in secondary structure of hair with respect to breed or temperature treatment and suggests relation between FTIR spectral characteristics and tensile properties.</span>

Abstract:The thermoregulation system in animals removes body heat in hot temperatures and retains body heat in cold temperatures. The better the animal removes heat, the worse the animal retains heat and visa versa. It is the balance between these two conflicting goals that determines the mammal's size, heart rate and amount of hair. The rat's loss of tail hair and human's loss of its body hair are responses to these conflicting thermoregulation needs as these animals evolved to larger size over time.

Abstract:Humans have a characteristic distribution of hair across the body. Visible, relatively long and thick terminal hair fibres are present on the scalp and eyebrows in childhood, and are stimulated to grow on other parts of the body, such as the beard and armpits, by hormones during puberty. The short and fine vellus hairs, in contrast, are not readily visible and cover most of the body, including the face. Here we report quantification of the timing and characteristics of hair follicle development in human embryogenesis, from gestational weeks 8 to 19, and compare this to mouse hair follicle development. We find that human hair follicles develop first on the head, where we identify several distinct initiation sites, followed by the torso. Although terminal and vellus hair follicles have clear differences in the adult, both hair types initially develop from placodes and dermal condensates of similar size. Once their development is initiated, we find that human hair follicles grow and mature at the same rate, regardless of anatomical location, but have different density at different body sites. These findings suggest that regional hair differences in human skin, such as the distinction between scalp and forehead, are largely caused by processes acting after the initial hair follicle morphogenesis. Efforts to understand the evolution of human hairlessness should, therefore, focus on genetic and cellular events that take place after hair follicle morphogenesis. Finally, we compared human skin appendages, including eccrine sweat glands, with those in mouse. We found that molecular markers, such as EDA, EDAR, SOX2 and WNT pathway components, are broadly similar in expression between both species, although specific differences do exist. Together with comparison of morphology and gene expression, these results support the use of embryonic mouse primary hair follicles as a model for human hair follicle development.

Abstract:Animal hairs, like other natural fibers, display excellent mechanical properties, especially, the tensile toughness and fracture resistance. Several structure-mechanics models have attributed mechanical superiority of hair to its unique nanocomposite structure which consists of intermediate filaments and matrix. However, the contribution of fibrils and their associated interfaces on the mechanical properties of animal hairs remains unclear. Herein, using the small- and wide-angle X-ray scattering, and an ultrahigh-speed microcamera system, it is confirmed that the conformation and fibrils (which represent both nanofibrils and microfibrils) of the keratin channel endow tensile toughness and fracture resistance to camel hairs. During the stretching process, an α-β transition occurred at the secondary structure level, leading to the formation of a tensile plateau, which improves the toughness compared with the structure without a conformation transition. Meanwhile, fibrils further toughened the camel hairs and resisted their crack propagation through confined fibrillar slippage, splitting, and pulling. These structure-property relations in natural hairs can inspire damage-tolerant polymer fiber design.
Known and Unknown Features of Hair Cuticle Structure: A Brief Review

Abstract:The cuticle is the outermost layer of overlapping flattened cells of hair and has been subjected to many years of study to understand its structure and how it develops in the follicle. The essential function of the cuticle with its tough inelastic protein content is to protect the inner cortex that provides the elastic properties of hair. Progress in our knowledge of hair came from studies with the electron microscope, initially transmission electron microscopy (TEM) for internal structure and later the scanning electron microscope (SEM) for cuticle surface shape and for investigating changes caused by various environmental influences such as cosmetic treatments and industrial processing of wool. Other physical techniques have been successfully applied in conjunction with proteomics. The outstanding internal features of the cuticle cells are the internal layers consisting of keratin filament proteins and the keratin-associated proteins. The stability and physical toughness of the cuticle cell is partly accounted for by the high content of disulphide crosslinking. The material between the cells that holds them tightly together, the cell membrane complex, consists of a layer of lipid on both sides of a central protein layer. The lipid contains 18-methyleicosanoic acid that is part of the hydrophobic lipid surface of hair. For the past decade there have been aspects that remained unanswered because they are difficult to study. Some of these are discussed in this brief review with suggestions for experimental approaches to shed more light.

The distribution of peroxide regulating enzymes in the canine eye.

Abstract:The retina and rod outer segments (ROS), rich in unsaturated lipids, are highly susceptible to autoxidation via free radical mechanisms. In the canine eye, four enzymes, i.e.; p-phenylenediamine (PPD) peroxidase, glutathione peroxidases, catalase and superoxide dismutase which are involved in the production or degradation of peroxides, were measured. All enzymes were present in the retina, ciliary body and iris, but differed in concentration. Superoxide dismutase showed high activity in the ROS. In the retinal pigment epithelium (RPE) a peroxidase utilizing PPD as cosubstrate and superoxide dismutase were both present. The former enzyme was found equally distributed between soluble and insoluble forms. A pigment granule fraction from the RPE only contained PPD-peroxidase. The compartmentalization of peroxide regulating enzymes in the eye is striking. Whereas the retina seems well protected against superoxide free radicals and hydrogen peroxide by virtue of superoxide dismutase, peroxidases and catalase activities, the ROS are only protected by superoxide dismutase. Therefore, after phagocytosis of the ROS, any peroxidized lipids, organic peroxides, or unconverted superoxide radicals contained within the ROS, must be detoxified by a specific RPE peroxidase and superoxide dismutase. Hydrogen peroxide or other peroxidized compounds apparently are not degraded by glutathione peroxidases or catalase, since these enzymes were not measurable in our RPE preparations. The role of antioxidative enzymes in the eye, which retard peroxide and free radical information, appears to be specific and regional.

Abstract:COL17A1 immunofluorescence signals. Therefore, we diagnosed the patient with JEB-nH. Non-Herlitz junctional epidermolysis bullosa is an autosomal recessive, non-lethal variant of junctional EB caused by a deficiency of COL17A1, LAMA3, LAMB3 and LAMC2. Because the most characteristic symptoms of JEB-nH are late onset, clinical diagnosis was difficult and genetic testing was indispensable for a proper diagnosis in this patient. Late-onset nonscarring alopecia is a characteristic finding in JEB-nH. It was speculated from a mouse study that the cell-matrix attachment through COL17A1 is important to maintain the stemness of the hair stem cells located in the hair-bulge area. Close observation of hair changes in this patient will provide a good opportunity to investigate hair stem cell loss in human JEB-nH.

Abstract:AbstractHuman hair has three main regions, the medulla, the cortex and the cuticle. An existing model for the cortex suggests that the α-keratin- based intermediate filaments (IFs) align with the hair’s axis, but are orientationally disordered in-plane. We found that there is a new region in the cortex near the cuticle’s boundary in which the IFs are aligned with the hair’s axis, but additionally, they are orientationally ordered in-plane due to the presence of the cuticle/hair boundary. Further into the cortex, the IF arrangement becomes disordered, eventually losing all in-plane orientation. We also find that in the cuticle, a key diffraction feature is absent, indicating the presence of the β-keratin rather than that of the α-keratin phase. This is direct structural evidence that the cuticle contains β-keratin sheets. This work highlights the importance of using a sub-micron x-ray beam to unravel the structures of poorly ordered, multi-phase systems.

Fundamental hair follicle biology and fine fibre production in animals

Abstract:Hair 'fine' fibre is an important commercial product of farmed and certain wild animal species. The fibre is produced in follicles embedded in skin. These have properties in common with other tissues of the integument and have importance in determining yield and quality of fibre. Means of understanding and improving these characteristics are informed by knowledge of integumental and follicle biology. This paper reviews contemporary information that identifies the major fibre-producing species and their production characteristic. It surveys knowledge describing fundamental biology of the integument and considers information derived for the hair follicle from studies on a number of species including genetically modified mice. It identifies the composition of the follicle and describes components and interrelationships between epidermal hair-fibre producing epidermis and fibroblast- and connective tissue-containing dermis. The structure of different primary and secondary follicle types, and associated structures, are described. Focus is given to the alterations in anatomy and in behaviour from active to inactive state, which occurs during the hair follicle cycle. Information is provided on the anatomical substructures (hair medulla, cortex, cuticles and supporting sheaths and dermal papilla), cellular and extracellular composition, and adhesion and chemical signalling systems, which regulate development from the early embryo to post-natal state and subsequent cycling. Such signalling involves the dermis and its specialist fibroblasts, which secrete signalling molecules, which along with those from local epidermis and systemic sources, largely determine structure and function of epidermal cells. Such chemical signalling typically includes endocrine-, paracrine-, autocrine- and juxtacrine-acting molecules and interactions with their receptors located on cell membranes or intracellularly with transduction of message mediated by transcription factors at gene level. Important hormones and growth factors and inhibitors regulating morphogenic and/or mitogenic activity are identified. These mediate mechanisms associated with presence or absence in skin and development of patterning for primary or secondary follicles. Reference is made to deposition of individual keratins and keratin-associated proteins in follicle sub-structures and to fibre properties such as length, diameter, medullation, crimp and lustre. Pre- and post-natal regulation of pigmentation by melanocytes is reviewed. Brief attention is given to genomic and non-genomic variation and impact on the phenotypes expressed and the role of regulatory gene products as potential molecular markers for selection of superior animals. The importance of nutrients in providing substrates for follicular structures and enzymes and in molecules facilitating gene expression is also considered.

Abstract:<section class="article-section__content"><h3 class="article-section__sub-title section1"> Background</h3><p>Porcine ear skin is used in studies of percutaneous penetration as a substitute for human skin. The objective of the present study was to determine the structure of the hair follicles on the dorsal area of porcine ear skin and make a morphometric comparison with the hair follicles of human skin.</p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Materials and Methods</h3><p>Sections of frozen biopsies were cut vertically to the skin surface in longitudinal sections using a cryotome and were investigated using microscopy. For each hair follicle, various parameters were determined.</p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Results</h3><p>The follicular density in porcine ear skin varies according to the area studied, and the length of most of the follicles was approximately 1458 ± 286 μm. The size of the follicular orifice was also determined in a total of 305 follicles. It showed a diameter of roughly 113 ± 43 μm.</p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Conclusion</h3><p>The results showed a very good similarity between human and pig hair follicles. Therefore, porcine ear skin can be considered as a very suitable model of human skin in dermal and especially follicular penetration studies.</p></section>

Abstract:Wool is the critical textile raw material which is produced by the hair follicle of sheep. Therefore, it has important implications to investigate the molecular mechanism governing hair follicle development. Due to high cellular heterogeneity as well as the insufficient cellular, molecular, and spatial characterization of hair follicles on sheep, the molecular mechanisms involved in hair follicle development and wool curvature of sheep remains largely unknown. Single-cell RNA sequencing (scRNA-seq) technologies have made it possible to comprehensively dissect the cellular composition of complex skin tissues and unveil the differentiation and spatial signatures of epidermal and hair follicle development. However, such studies are lacking so far in sheep. Here, single-cell suspensions from the curly wool and straight wool lambskins were prepared for unbiased scRNA-seq. Based on UAMP dimension reduction analysis, we identified 19 distinct cell populations from 15,830 single-cell transcriptomes and characterized their cellular identity according to specific gene expression profiles. Furthermore, novel marker gene was applied in identifying dermal papilla cells isolated in vitro . By using pseudotime ordering analysis, we constructed the matrix cell lineage differentiation trajectory and revealed the dynamic gene expression profiles of matrix progenitors' commitment to the hair shaft and inner root sheath (IRS) cells. Meanwhile, intercellular communication between mesenchymal and epithelial cells was inferred based on CellChat and the prior knowledge of ligand–receptor pairs. As a result, strong intercellular communication and associated signaling pathways were revealed. Besides, to clarify the molecular mechanism of wool curvature, differentially expressed genes in specific cells between straight wool and curly wool were identified and analyzed. Our findings here provided an unbiased and systematic view of the molecular anatomy of sheep hair follicle comprising 19 clusters; revealed the differentiation, spatial signatures, and intercellular communication underlying sheep hair follicle development; and at the same time revealed the potential molecular mechanism of wool curvature, which will give important new insights into the biology of the sheep hair follicle and has implications for sheep breeding.
