Abstract:Two strains of the cotton bollworm, Helicoverpa armigera (Hubner), one is a susceptible laboratory strain and the other is collected from Bt cotton fields, were used as the insect hosts to study the effect of Bt insecticidal protein on the development of the parasitoid, Microplitis mediator (Haliday). The results showed that the larval duration of the parasitoid was delayed, and the pupal weight, body weight of the newly emerged adult and adult longevity decreased significantly when the hosts larvae fed on diet containing Bt protoxin Cry1Ac at the concentrations of 0.5-8.0 μg/g in all time or from 12 h before parasitism till pupation of the parasitoid . Compared with the control, the larval weight and pupal weight of H. armigera decreased significantly when the larvae fed on diet containing cry1Ac. Pupation rate of H. armigera decreased significantly in the treatment with diet containing Cry1Ac at 4.0 μg/g. When H. armigera larvae fed on the transgenic cotton leaves expressing both Cry1A and CpTI proteins, the mortality of the laboratory strain was 48.5%-95.8% in 2-5 days after treatment, which was significantly higher than that of the field strain (30.9%-86.1%). It is concluded that feeding on diet containing Bt insecticidal protein, both strains of the cotton bollworm will have significantly negative effects on development of the parasitoid, M.mediator.
Effect of Bt Broccoli and Resistant Genotype of Plutella Xylostella (lepidoptera: Plutellidae) on Life History and Prey Acceptance of the Predator Coleomegilla Maculata (coleoptera: Coccinellidae)

Abstract:The ecological implications on biological control of insecticidal transgenic plants, which produce crystal (Cry) proteins derived from the soil bacterium Bacillus thuringiensis (Bt), remains a contentious issue and affects risk assessment decisions. In this study, we used a unique system of resistant insects, Bt plants and a predator to critically evaluate this issue. The effects of broccoli type (normal or expressing Cry1Ac protein) and insect genotype (susceptible or Cry1Ac-resistant) of Plutella xylostella L. (Lepidoptera: Plutellidae) were examined for their effects on the life history of the predator, Coleomegilla maculata DeGeer (Coleoptera: Coccinellidae) over two generations. Additional behavioral studies were conducted on prey choice. C. maculata could not discriminate between Bt-resistant and susceptible genotypes of P. xylostella, nor between Bt and normal broccoli plants with resistant genotypes of P. xylostella feeding on them. The larval and pupal period, adult weight and fecundity of each female were not significantly different when C. maculata larvae fed on different genotypes (Bt-resistant or susceptible) of insect prey larvae reared on Bt or non-Bt broccoli plants. The life-history parameters of the subsequent generation of C. maculata fed on Bt broccoli-reared resistant P. xylostella were also not significantly different from those on non-Bt broccoli. These results indicated that Cry1Ac did not harm the life history or prey acceptance of an important predator after two generations of exposure. Tri-Trophic Studies Using Cry1Ac-Resistant Plutella Xylostella Demonstrate No Adverse Effects of Cry1Ac on the Entomopathogenic Nematode, Heterorhabditis Bacteriophora

Abstract:The potential impacts on natural enemies of crops that produce insecticidal Cry proteins from Bacillus thuringiensis (Bt) are an important part of an environmental risk assessment. Entomopathogenic nematodes are important natural enemies of lepidopteran pests, and the effects of Bt crops on these nontarget organisms should be investigated to avoid disruption of their biological control function. The objective of this study was to investigate the effects of Cry1Ac-expressing transgenic Bt broccoli on the entomopathogenic nematode, Heterorhabditis bacteriophora Poinar (Rhabditida: Heterorhabditidae), under tri-trophic conditions. Using CrylAc-resistant Plutella xylostella L. (Lepidoptera: Plutellidae) larvae as hosts, we evaluated the potential impact of Cry1Ac-expressing Bt broccoli on several fitness parameters of H. bacteriophora. Virulence, reproductive potential, time of emergence, and preference of H. bacteriophora for the host (P. xylostella) were not significantly affected when CrylAc-resistant P. xylostella larvae were reared on leaves of Cry1Ac or non-Bt broccoli. Also the aforementioned parameters of the subsequent generation of H. bacteriophora did not differ between nematodes obtained from P. xylostella reared on CrylAc broccoli compared with those obtained from P. xylostella reared on non-Bt broccoli. To the best of our knowledge, the current study provides the first clear evidence that Cry1Ac does not affect important fitness parameters of H. bacteriophora.
Using Resistant Prey Demonstrates That Bt Plants Producing Cry1Ac, Cry2Ab, and Cry1F Have No Negative Effects on Geocoris Punctipes and Orius Insidiosus.

Abstract:Geocoris punctipes (Say) and Orius insidiosus (Say) are generalist predators found in a wide range of crops, including cotton (Gossypium hirsutum L.) and maize (Zea mays L.), where they provide important biological control services by feeding on an array of pests, including eggs and small larvae of caterpillars. A high percentage of cotton and maize in the United States and several other countries are transgenic cultivars that produce one or more of the insecticidal Cry proteins of Bacillus thuringiensis Berliner (Bt). Here we quantify effects of three Cry proteins on the life history of these predators over two generations when they are exposed to these Cry proteins indirectly through their prey. To eliminate the confounding prey quality effects that can be introduced by Bt-susceptible prey, we used Cry1Ac/Cry2Ab-resistant Trichoplusia ni (Hübner) and Cry1 F-resistant Spodoptera frugiperda (J.E. Smith) in a series of tri-trophic studies. Survival, development, adult mass, fecundity, and fertility were similar when predators consumed larvae feeding on Cry1Ac/Cry2Ab cotton or Cry1 F maize compared with prey feeding on isogenic or near-isogenic cotton or maize. Repeated exposure of the same initial cohort over a second generation also resulted in no differences in life-history traits when feeding on non-Bt- or Bt-fed prey. Enzyme-linked immunosorbent assay showed that predators were exposed to Bt Cry proteins from their prey and that these proteins became increasingly diluted as they moved up the food chain. Results show a clear lack of effect of three common and widespread Cry proteins on these two important predator species. The use of resistant insects to eliminate prey quality effects provides a robust and meaningful assessment of exposure and hazard.
Bt Crops Benefit Natural Enemies to Control Non-Target Pests.

Abstract:Crops producing insecticidal crystal (Cry) proteins from Bacillus thuringiensis (Bt) control important lepidopteran pests. However, pests such as aphids not susceptible to Cry proteins may require other integrated pest management (IPM) tactics, including biological control. We fed aphids on Bt and non-Bt plants and analyzed the Bt protein residue in aphids and compared the effects of Bt plants and a pyrethroid, lambda-cyhalothrin, on the performance of three natural enemies (predators: Coleomegilla maculata and Eupeodes americanus; parasitoid Aphidius colemani) of the green peach aphid, Myzus persicae. No Bt protein residues in aphids were detected and no significant differences were recorded in the performance of pyrethroid-resistant aphids that fed on Bt broccoli expressing Cry1Ab or Cry1C, or on non-Bt broccoli plants treated or not treated with the pyrethroid. This indicated the aphids were not affected by the Cry proteins or the pyrethroid, thus removing any effect of prey quality. Tri-trophic experiments demonstrated that no C. maculata and E. americanus survived consumption of pyrethroid-treated aphids and that ovipositional behavior of A. colemani was impaired when provided with pyrethroid-treated aphids. In contrast, natural enemies were not affected when fed aphids reared on Bt broccoli, thus demonstrating the safety of these Bt plants for IPM.
Eliminating host-mediated effects demonstrates Bt maize producing Cry1F has no adverse effects on the parasitoid Cotesia marginiventris

Abstract:The fall armyworm, Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae), is an important pest of maize in the United States and many tropical areas in the western hemisphere. In 2001, Herculex I ® (Cry1F) maize was commercially planted in the United States to control Lepidoptera, including S. frugiperda . In 2006, a population of S. frugiperda was discovered in Puerto Rico that had evolved resistance to Cry1F maize in the field, making it the first well-documented case of an insect with field resistance to a plant producing protein from Bacillus thuringiensis (Bt). Using this resistant population, we conducted tri-trophic studies with a natural enemy of S. frugiperda . By using resistant S. frugiperda , we were able to overcome possible prey-mediated effects and avoid concerns about potential differences in laboratory- or field-derived Bt resistance. We used the Cry1F-resistant S. frugiperda to evaluate effects of Cry1F on Cotesia marginiventris (Cresson) (Hymenoptera: Braconidae), a larval endoparasitoid of S. frugiperda, over five generations. Our results clearly demonstrate that Cry1F maize does not affect development, parasitism, survivorship, sex ratio, longevity or fecundity of C. marginiventris when they parasitize Cry1F maize-fed S. frugiperda. Furthermore, the level of Cry1F protein in the leaves was strongly diluted when transferred from Bt maize to S. frugiperda and was not detected in larvae, cocoons or adults of C. marginiventris. Our results refute previous reports of C. marginiventris being harmed by Bt proteins and suggest that such results were caused by prey-mediated effects due to using Bt-susceptible lepidopteran hosts.

Abstract:Bacillus thuringiensis (Bt) is the most widely used biopesticide worldwide and can produce several insecticidal crystal proteins and vegetative insecticidal proteins (Vips) at different growth stages. In our previous study, extracellular polysaccharides (EPSs) of Bt strain HD270 were found to enhance the insecticidal activity of Cry1Ac protoxin against Plutella xylostella (L.) and promote the binding of Cry1Ac to the intestinal brush border membrane vesicles (BBMVs). Whether the synergistic activity of Bt EPSs is common to other Cry1-type or Vip proteins is unclear, as is the potential synergistic mechanism. In this study, crude EPS-HD270 was found to increase the toxicity of Cry1-type toxins and Vip3Aa11 against different lepidopteran pests by approximately 2-fold. The purified EPS-HD270 also possessed synergistic activity against the toxicity of Cry1Ac and Vip3Aa11 against Spodoptera frugiperda (J.E. Smith) and Helicoverpa armigera (Hübner). Furthermore, we found that EPS-HD270 had a strong binding ability with Vip3Aa11 and promoted the binding of Vip3Aa11 to the BBMVs of H. armigera and S. frugiperda. Bt EPS-HD270 also protected Vip3Aa11 from proteolytic processing in larval midgut juice. Bt EPSs had universal synergistic effects on Cry1-type or Vip toxins against S. frugiperda and H. armigera. Bt EPS-HD270 exhibited synergistic activity with Vip3Aa through promotion of binding to BBMVs and protection from digestion by midgut protease. The results indicated that synergistic activity with Bt toxins was an important function of Bt EPSs, which was very different from other Bacillus spp.

Structure characteristics and function of a novel extracellular polysaccharide from Bacillus thuringiensis strain 4D19

Abstract:Bacillus thuringiensis (Bt) are entomopathogenic bacteria that produce different kinds of insecticidal proteins. However, studies on Bt exopolysaccharides are lacking. Here, we aimed to explore the characteristics and insecticidal synergism of EPS, an exopolysaccharide from Bt strain 4D19. The molecular weight of EPS-2 was 58.0 kDa, which consisted of mannose (44.2%), GlcN (35.5%), D-GalN (8.0%), glucose (5.5%), arabinose (5.1%), galactose (0.9%), Man-UA (0.3%) and Glc-UA (0.2%). The toxicity of insecticidal proteins against Plutella xylostella was increased by adding EPS. EPS-2 bound to Cry1Ac protoxin and promoted the binding of Cry1Ac protoxin to the gut membrane of P. xylostella, but did not bind to activated toxins. These results suggested that EPS-2 may bind to the protoxin C-terminal region to enhance insecticidal activity. Our findings indicated that Bt strains produce exopolysaccharide to enhance the toxicity of insecticidal crystal proteins, which could be applied in biopesticide research and product development.
Proteomics-based identification of midgut proteins correlated with Cry1Ac resistance in Plutella xylostella (L.)

Abstract:The diamondback moth, Plutella xylostella (L.), is a worldwide pest of cruciferous crops and can rapidly develop resistance to many chemical insecticides. Although insecticidal crystal proteins (i.e., Cry and Cyt toxins) derived from Bacillus thuringiensis (Bt) have been useful alternatives to chemical insecticides for the control of P. xylostella, resistance to Bt in field populations of P. xylostella has already been reported. A better understanding of the resistance mechanisms to Bt should be valuable in delaying resistance development. In this study, the mechanisms underlying P. xylostella resistance to Bt Cry1Ac toxin were investigated using two-dimensional differential in-gel electrophoresis (2D-DIGE) and ligand blotting for the first time. Comparative analyses of the constitutive expression of midgut proteins in Cry1Ac-susceptible and -resistant P. xylostella larvae revealed 31 differentially expressed proteins, 21 of which were identified by mass spectrometry. Of these identified proteins, the following fell into diverse eukaryotic orthologous group (KOG) subcategories may be involved in Cry1Ac resistance in P. xylostella: ATP-binding cassette (ABC) transporter subfamily G member 4 (ABCG4), trypsin, heat shock protein 70 (HSP70), vacuolar H(+)-ATPase, actin, glycosylphosphatidylinositol anchor attachment 1 protein (GAA1) and solute carrier family 30 member 1 (SLC30A1). Additionally, ligand blotting identified the following midgut proteins as Cry1Ac-binding proteins in Cry1Ac-susceptible P. xylostella larvae: ABC transporter subfamily C member 1 (ABCC1), solute carrier family 36 member 1 (SLC36A1), NADH dehydrogenase iron-sulfur protein 3 (NDUFS3), prohibitin and Rap1 GTPase-activating protein 1. Collectively, these proteomic results increase our understanding of the molecular resistance mechanisms to Bt Cry1Ac toxin in P. xylostella and also demonstrate that resistance to Bt Cry1Ac toxin is complex and multifaceted.
Effects of the pyrE deletion mutant from Bacillus thuringiensis on gut microbiota and immune response of Spodoptera exigua

Abstract:The gut microbiota is essential for the growth and development of insects, and the intestinal immune system plays a critical role in regulating the homeostasis of intestinal microorganisms and their interactions with pathogenic bacteria. Infection with Bacillus thuringiensis (Bt) can disrupt the gut microbiota of insects, but the regulatory factors governing the interaction between Bt and gut bacteria are not well understood. Uracil secreted by exogenous pathogenic bacteria can activate DUOX-mediated reactive oxygen species (ROS) production, which helps maintain intestinal microbial homeostasis and immune balance. To elucidate the regulatory genes involved in the interaction between Bt and gut microbiota, we investigate the effects of uracil derived from Bt on gut microbiota, and host immunity using a uracil deficient Bt strain (Bt GS57△ pyrE ) obtained by homologous recombination. We analyze the biological characteristics of the uracil deficient strain and found that the deletion of uracil in Bt GS57 strain changed the diversity of gut bacteria in Spodoptera exigua , as investigated using Illumina HiSeq sequencing. Furthermore, qRT-PCR analysis showed that compared with Bt GS57 (control), the expression of the SeDuox gene and the level of ROS were significantly decreased after feeding with Bt GS57△ pyrE . Adding uracil to Bt GS57△ pyrE restored the expression level of DUOX and ROS to a higher level. Additionally, we observed that PGRP-SA , attacin, defensin and ceropin genes were significant different in the midgut of S. exigua infected by Bt GS57 and Bt GS57△ pyrE , with a trend of increasing first and then decreasing. These results suggest that uracil regulates and activates the DUOX-ROS system, affects the expression of antimicrobial peptide genes, and disturb intestinal microbial homeostasis. We preliminarily speculate that uracil is a key factor in the interaction between Bt and gut microbiota, and these findings provide a theoretical basis for clarifying the interaction between Bt, host, and intestinal microorganisms, as well as for gaining new insights into the insecticidal mechanism of B. thuringiensis in insects.

Response of the Propylea japonica Microbiota to Treatment with Cry1B Protein

Abstract:Propylea japonica (Thunberg) (Coleoptera: Coccinellidae) is a dominant natural enemy of insect pests in farmland ecosystems. It also serves as an important non-target insect for environmental safety evaluations of transgenic crops. Widespread planting of transgenic crops may result in direct or indirect exposure of P. japonica to recombinant Bacillus thuringiensis (Bt) protein, which may in turn affect the biological performance of this natural enemy by affecting the P. japonica microflora. However, the effects of Bt proteins (such as Cry1B) on the P. japonica microbiota are currently unclear. Here, we used a high-throughput sequencing method to investigate differences in the P. japonica microbiota resulting from treatment with Cry1B compared to a sucrose control. The results demonstrated that the P. japonica microbiome was dominated by Firmicutes at the phylum level and by Staphylococcus at the genus level. Within-sample (α) diversity indices demonstrated a high degree of consistency between the microbial communities of P. japonica treated with the sucrose control and those treated with 0.25 or 0.5 mg/mL Cry1B. Furthermore, there were no significant differences in the abundance of any taxa after treatment with 0.25 mg/mL Cry1B for 24 or 48 h, and treatment with 0.5 mg/mL Cry1B for 24 or 48 h led to changes only in Staphylococcus, a member of the phylum Firmicutes. Treatment with a high Cry1B concentration (1.0 mg/mL) for 24 or 48 h caused significant changes in the abundance of specific taxa (e.g., Gemmatimonades, Patescibacteria, Thauera, and Microbacterium). However, compared with the control, most taxa remained unchanged. The statistically significant differences may have been due to the stimulatory effects of treatment with a high concentration of Cry1B. Overall, the results showed that Cry1B protein could alter endophytic bacterial community abundance, but not composition, in P. japonica. The effects of Bt proteins on endophytes and other parameters in non-target insects require further study. This study provides data support for the safety evaluation of transgenic plants.

Abstract:Bacillus thuringiensis is known as an entomopathogenic bacterium active against different agricultural pests. Recently, some novel functions have been explored, including plant growth-promoting activities. In our previous work, B. thuringiensis strains were found to effectively suppress Sclerotinia sclerotiorum by inducing systemic resistance (ISR) in Brassica campestris (L.). However, the mechanism by which exogenous signals from B. thuringiensis are perceived by plants is still not clear. Here, based on challenge-inoculation assays, we found that exopolysaccharides (EPSs) of B. thuringiensis strains could limit the spread of S. sclerotiorum by ISR in B. campestris. We purified and characterized the Bt 4F5 EPS, and the molecular weight was 66.2 kDa. The main monosaccharide units of Bt 4F5 EPS were determined to be mannose, GluA, glucose and galactose. Cellular defense response markers such as defense-related enzymes were induced after the 4F5 EPSs pretreatment in the leaves. Moreover, quantitative real-time PCR results indicated that the defense-related genes HEL and PDF1.2 were upregulated, suggesting an activation of the JA/ET signaling pathways by Bt 4F5 EPS. In addition, complete genome sequencing of the Bt 4F5 strain revealed twenty-one candidate genes involved in exopolysaccharide synthesis in chromosome. In , the EPSs of Bt strains play an important role in micro-associated molecular pattern (MAMP) perception during the process of Bt-triggered systemic resistance.

Abstract:Many insects, including the Plutella xylostella (L.), have developed varying degrees of resistance to many insecticides, including Bacillus thuringiensis (Bt) toxins, the bioinsecticides derived from Bt. The polycalin protein is one of the potential receptors for Bt toxins, and previous studies have confirmed that the Cry1Ac toxin can bind to the polycalin protein of P. xylostella, but whether polycalin is associated with the resistance of Bt toxins remains controversial. In this study, we compared the midgut of larvae from Cry1Ac-susceptible and -resistant strains, and found that the expression of the Pxpolycalin gene was largely reduced in the midgut of the resistant strains. Moreover, the spatial and temporal expression patterns of Pxpolycalin showed that it was mainly expressed in the larval stage and midgut tissue. However, genetic linkage experiments showed that the Pxpolycalin gene and its transcript level were not linked to Cry1Ac resistance, whereas both the PxABCC2 gene and its transcript levels were linked to Cry1Ac resistance. The larvae fed on a diet containing the Cry1Ac toxin showed no significant change in the expression of the Pxpolycalin gene in a short term. Furthermore, the knockout of polycalin and ATP-binding cassette transporter subfamily C2 (ABCC2) genes separately by CRISPR/Cas9 technology resulted in resistance to decreased susceptibility to Cry1Ac toxin. Our results provide new insights into the potential role of polycalin and ABCC2 proteins in Cry1Ac resistance and the mechanism underlying the resistance of insects to Bt toxins.

Bacillus thuringiensis Suppresses the Humoral Immune System to Overcome Defense Mechanism of Plutella xylostella

Abstract:Background: Plutella xylostella has become a notorious pest of cruciferous crops all over the world. Delta-endotoxins of Bacillus thuringiensis are widely used insecticidal proteins for controlling P. xylostella. However, the interaction mechanism of B. thuringiensis with the immune system of P. xylostella, at the genomic level, is still unclear. This study explored the immune response of P. xylostella to B. thuringiensis, at different time intervals, 6 h, 12 h, 18 h, 24 h, and 36 h, by using RNA-Sequencing (RNA-Seq) and RT-qPCR. Results: In total, 167 immunity-related genes were identified and placed into different families, including pattern recognition receptors (PRRs), signal modulators, immune pathways (Toll, IMD, and JAK/STAT), and immune effectors. It is worth mentioning that the analyses of the differentially expressed immunity-related genes revealed that most of the differentially expressed genes (DEGs) (87, 56, 76, 67, and 73 genes) were downregulated in P. xylostella following B. thuringiensis oral infection at 6 h, 12 h, 18 h, 24 h, and 36 h. Interestingly, our RNA-Seq analysis also revealed reduced expression of antimicrobial peptides, that play a vital role in the humoral immune system of P. xylostella. Conclusion: This study demonstrates that B. thuringiensis plays a novel role in controlling P. xylostella, by suppressing the immune system.

Resistance to Bacillus thuringiensis Cry1Ac toxin requires mutations in two Plutella xylostella ATP-binding cassette transporter paralogs

Abstract:The diamondback moth, Plutella xylostella, is a cosmopolitan pest and the first species to develop field resistance to toxins from the gram-positive bacterium Bacillus thuringiensis (Bt). Although previous work has suggested that mutations of ATP-binding cassette transporter subfamily C2 (ABCC2) or C3 (ABCC3) genes can confer Cry1Ac resistance, here we reveal that P. xylostella requires combined mutations in both PxABCC2 and PxABCC3 to achieve high-level Cry1Ac resistance, rather than simply a mutation of either gene. We identified natural mutations of PxABCC2 and PxABCC3 that concurrently occurred in a Cry1Ac-resistant strain (Cry1S1000) of P. xylostella, with a mutation (RA2) causing the mis-splicing of PxABCC2 and another mutation (RA3) leading to the premature termination of PxABCC3. Genetic linkage analysis showed that RA2 and RA3 were tightly linked to Cry1Ac resistance. Introgression of RA2 and RA3 enabled a susceptible strain (G88) of P. xylostella to obtain high resistance to Cry1Ac, confirming that these genes confer resistance. To further support the role of PxABCC2 and PxABCC3 in Cry1Ac resistance, frameshift mutations were introduced into PxABCC2 and PxABCC3 singly and in combination in the G88 strain with CRISPR/Cas9 mediated mutagenesis. Bioassays of CRISPR-based mutant strains, plus genetic complementation tests, demonstrated that the deletion of PxABCC2 or PxABCC3 alone provided < 4-fold tolerance to Cry1Ac, while disruption of both genes together conferred >8,000-fold resistance to Cry1Ac, suggesting the redundant/complementary roles of PxABCC2 and PxABCC3. This work advances our understanding of Bt resistance in P. xylostella by demonstrating mutations within both PxABCC2 and PxABCC3 genes are required for high-level Cry1Ac resistance.Bacillus thuringiensis (Bt) foliar sprays and transgenic crops expressing Bt toxins are used extensively to control insect pests, but the evolution of resistance limits their efficacy. Multiple studies have reported that ATP-binding cassette (ABC) transporters are important Bt receptors, and mutations in either ABCC2 or ABCC3 can lead to Cry1Ac-toxin resistance, although this process is not fully understood. In this study, we applied both forward and reverse genetic analyses to demonstrate that high-level Bt-Cry1Ac resistance in Plutella xylostella requires concurrent mutations in both PxABCC2 and PxABCC3. We identified inactivating mutations in these two genes from a Cry1Ac-resistant strain (Cry1S1000) of P. xylostella and conducted genetic linkage analysis, which supported the role that PxABCC2 and PxABCC3 were the causal genes of Cry1Ac resistance. We then knocked out PxABCC2 and PxABCC3 in a P. xylostella susceptible reference strain (G88) to confirm that high-level Cry1Ac resistance requires mutation of PxABCC2 and PxABCC3, rather than a mutation of either one gene. This finding expands our understanding of complex Bt resistance processes and may be relevant to Bt-Cry1Ac resistance in other lepidopteran insects.

Abstract:A variety of coordinated host-cell responses are activated as defense mechanisms against pore-forming toxins (PFTs). Bacillus thuringiensis (Bt) is a worldwide used biopesticide whose efficacy and precise application methods limits its use to replace synthetic pesticides in agricultural settings. Here, we analyzed the intestinal defense mechanisms of two lepidopteran insect pests after intoxication with sublethal dose of Bt PFTs to find out potential functional genes. We show that larval intestinal epithelium was initially damaged by the PFTs and that larval survival was observed after intestinal epithelium regeneration. Further analyses showed that the intestinal regeneration caused by Cry9A protein is regulated through c-Jun NH (2) terminal kinase (JNK) and Janus tyrosine kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathways. JAK/STAT signaling regulates intestinal regeneration through proliferation and differentiation of intestinal stem cells to defend three different Bt proteins including Cry9A, Cry1F or Vip3A in both insect pests, Chilo suppressalis and Spodoptera frugiperda . Consequently, a nano-biopesticide was designed to improve pesticidal efficacy based on the combination of Stat double stranded RNA (dsRNA)-nanoparticles and Bt strain. This formulation controlled insect pests with better effect suggesting its potential use to reduce the use of synthetic pesticides in agricultural settings for pest control. Bacillus thuringiensis (Bt) is the most successful bacteria to control insect pests through microbial pesticides and genetical modified crops. Its insecticidal proteins form pores on the midgut leading to target larvae death. However, the defense response of target insect pests is still unclear during this process. Here, we found that larvae intestine undergoes remodeling after intoxication with sublethal dose of Bt toxins. During the remodeling, the intestinal regeneration regulated by JNK and JAK/STAT signaling was induced after epithelial cells damaged by toxins. Surprisingly, this regulation is independent of EGFR pathway and unpaired gene that usually function in defense response in Drosophila . Finally, based on the findings, a mixture of dsRNA-nanoparticles and Bt bacterial strains was designed to improve pesticidal efficacy.

Abstract:A major beta-glucan binding proteins (beta-GBPs), 75.9 ku and 83.2 ku, were extracted from the hemolymph of Plutella xylostella larvae by using beta-1,3-glucan affinity precipitation method. The proteins were present in plasma but absent in haemocytes. The beta-GBPs had a specific affinity to beta-I, 3-glucan, a prophenoloxidase ( ProPO) trigger in P. xylostella hemolymph. The phenoloxidase ( PO) activity induced by the coexistence of both beta-GBPs and P-I 3-glucan was significantly higher than that by the beta-GBPs or beta-1 3-glucan alone. Moreover, the PO activities induced by mycelial lysates of four Zoophthora radicans isolates at the presence of beta-GBPs were much higher than that triggered by their protoplast lysates. The results indicate that the beta-GBPs may trigger the ProPO activity in P. xylostella hemolymph only if they specifically recognized beta-1 3-glucan from Z. radicans cell walls. This is a proof for a hypothesis that entomophthoralean protoplasts may evade from insect host immune defense. The protoplast or mycelial lysates of different Z. radicans isolates differed in ability to trigger the PO activities in the following order: ARSEF1342 > ARSEF2699 > F99101 > ARSEF1100. This trend was in accordance with that of their virulence to P. xylostella larvae, suggesting a possible relationship between host immune defense and fungal virulence.

Abstract:Concerns about resistance development to conventional insecticides in diamondback moth (DBM) Plutella xylostella (L.), the most destructive pest of Brassica vegetables, have stimulated interest in alternative pest management strategies. The toxicity of Bacillus thuringiensis subsp. aizawai (Bt GO33A) combined with chlorantraniliprole (Chl) has not been documented. Here, we examined single and combined toxicity of chlorantraniliprole and Bt to assess the levels of resistance in four DBM strains. Additionally, enzyme activities were tested in field-original highly resistant (FOH-DBM), Bt-resistant (Bt-DBM), chlorantraniliprole-resistant (CL-DBM), and Bt + chlorantraniliprole-resistant (BtC-DBM) strains. The Bt product had the highest toxicity to all four DBM strains followed by the mixture of insecticides (Bt + Chl) and chlorantraniliprole. Synergism between Bt and chlorantraniliprole was observed; the combination of Bt + (Bt + Chl) (1:1, LC50:LC50) was the most toxic, showing a synergistic effect against all four DBM strains with a poison ratio of 1.35, 1.29, 1.27, and 1.25. Glutathione S-transferase (GST) and carboxyl-esterase (CarE) activities showed positive correlations with chlorantraniliprole resistance, but no correlation was observed with resistance to Bt and Bt + Chl insecticides. Expression of genes coding for PxGST, CarE, AChE, and MFO using qRT-PCR showed that the PxGST and MFO were significantly overexpressed in Bt-DBM. However, AChE and CarE showed no difference in the four DBM strains. Mixtures of Bt with chlorantraniliprole exhibited synergistic effects and may aid the design of new combinations of pesticides to delay resistance in DBM strains substantially.

Abstract:Cadmium (Cd) pollution poses a serious threat to agricultural production and paddy field fauna. Crystalline proteins (e.g., Cry1Ab and Cry1Ac) are secreted by Bacillus thuringiensis, which can manage pests via a complicated toxic mechanism and have been widely used for pest control due to the commercialization of transgenic crops (e.g., cotton and rice) that expresses Bt insecticidal proteins. Nonetheless, studies on the effects of combined stress of Cd and Cry1Ab protein on field indicator species are limited. In the present study, we showed that spiders, Pirata subpiraticus, fed with Cd-containing flies+Cry1Ab had dramatically higher Cd accumulation than that in the spiders fed with Cd-containing flies (p < 0.05). In addition, the enrichment of Cd led to the activation of the protective mechanism by elevating the concentrations of glutathione peroxidase, glutathione S-transferase, and metallothionein in the spiders (p < 0.05). An in-depth transcriptome analysis revealed that the activities of ion metal binding proteins, transporters, and channels might play essential roles in the Cd accumulation process. More importantly, the higher Cd concentration in the combined Cd+Cry1Ab exposure prolonged developmental duration of P. subpiraticus, due to the down-regulated cuticle proteins (CPs) encoding genes involved in the molting process, which was regulated by a series of putative transcriptional factors such as ZBTB and zf-C2H2. Collectively, this integrated analysis illustrates that the combined Cd+Cry1Ab exposure increases the adverse effects of Cd stress on the growth, antioxidase, and CPs encoding genes of P. subpiraticus, thus providing a research basis and prospect for the rationality of transgenic Cry1Ab crops in the cultivation of heavy metal contaminated soil.
